Four new square-planar Ni(II) and Cu(II) complexes with [N2O2] binding system were synthesized by metal-directed condensation of 2,6-diformyl-4-methylphenol with benzoyl or acetylacetone mono-S-methylisothiosemicarbazone. Only mononuclear “one-armed” complexes were obtained as a consequence of the different reactivity of the two carbonyl groups in the hydroxydialdehyde. The complexes were characterized by elemental analysis, EI MS and UV-Vis spectroscopy. The structural assignment was confirmed by X-ray diffraction analysis and NMR spectroscopy, for the Ni(II) complexes, and by ESR spectroscopy and magnetic measurements, for the Cu(II) complexes.  相似文献

Three coordination compounds of formula {M(bmim)2Cl2} were synthetised (M?=?Co, Zn, and Hg) and fully characterised. Each complex incorporates 1-benzyl-2-methylimidazole (bmim) as ligand. The coordination polyhedron around the metal center for all complexes has a quasi-regular tetragonal geometry. Density functional theory calculations were carried out on the title compounds and as well on hypothetical complexes (Cu, Ni), in order to elucidate their electronic and molecular structure. The calculations reproduced the Co, Zn, and Hg experimental structures and could predict stable complexes in the case of Ni(II) and Cu(II) ions. The carbonic anhydrase (CA, EC 4.2.1.1) inhibitory effects of the three complexes were investigated. Only compound {Hg(bmim)2Cl2} (3) exhibited a modest inhibitory effect against hCA I, probably due to the affinity of Hg(II) for His residues at the entrance of the active site cavity.  相似文献

Methimazole (MeimzH) is an anti-thyroid drug and the first choice for patients with Grave’s disease. Two new copper(II) complexes of this drug: [Cu(MeimzH)2(NO3)2].0.5H2O and [Cu(MeimzH)2(H2O)2](NO3)2·H2O were synthesized and characterized by elemental analysis, dissolution behavior, thermogravimetric analysis and UV-vis, diffuse reflectance, FTIR and EPR spectroscopies. As it is known that copper(II) cation can act as an inhibitor of alkaline phosphatase (ALP), the inhibitory effect of methimazole and its copper(II) complexes on ALP activity has also been investigated.  相似文献

1‐phenyl‐3‐methyl‐4‐benzoyl‐5‐pyrazolone 4‐ethyl‐thiosemicarbazone (HL) and its copper(II), vanadium(V) and nickel(II) complexes: [Cu(L)(Cl)]·C2H5OH·( 1 ), [Cu(L)2]·H2O ( 2 ), [Cu(L)(Br)]·H2O·CH3OH ( 3 ), [Cu(L)(NO3)]·2C2H5OH ( 4 ), [VO2(L)]·2H2O ( 5 ), [Ni(L)2]·H2O ( 6 ), were synthesized and characterized. The ligand has been characterized by elemental analyses, IR, 1H NMR and 13C NMR spectroscopy. The tridentate nature of the ligand is evident from the IR spectra. The copper(II), vanadium(V) and nickel(II) complexes have been characterized by different physico‐chemical techniques such as molar conductivity, magnetic susceptibility measurements and electronic, infrared and electron paramagnetic resonance spectral studies. The structures of the ligand and its copper(II) ( 2 , 4 ), and vanadium(V) ( 5 ) complexes have been determined by single‐crystal X‐ray diffraction. The composition of the coordination polyhedron of the central atom in 2 , 4 and 5 is different. The tetrahedral coordination geometry of Cu was found in complex 2 while in complex 4 , it is square planar, in complex 5 the coordination polyhedron of the central ion is distorted square pyramid. The in vitro antibacterial activity of the complexes against Escherichia coli, Salmonella abony, Staphylococcus aureus, Bacillus cereus and the antifungal activity against Candida albicans strains was higher for the metal complexes than for free ligand. The effect of the free ligand and its metal complexes on the proliferation of HL‐60 cells was tested.  相似文献

The pyrazolone derivative N-(1-phenyl-3-methyl-4-propyl-pyrazolone-5)-salicylidene hydrazone (H2L) and its copper(II) complex [Cu2L2CH3OH]·2CH3OH have been both synthesized and characterized by elemental analyses, IR spectroscopy, X-ray crystallography, theoretical calculation and pharmacological testing. It’s found that the Cu(II) complex possesses more powerful anticancer effectivity than that of the ligand. In order to make its anticancer principium clearly, we investigate their structures. In ligand there are several coordination spots, such as N, O atoms, which are close to biological environment. The crystallographic structural analysis of the complex reveals that the two Cu centers display two different coordination patterns. O1, O2, N3, and N4 from the ligand take part in the coordination with Cu atoms, resulting in the formation of the double-nuclear complex. The pharmacological testing results show that the coordination effect improves the antitumor activity of the ligand. The calculated Fukui function for H2L and its deprotonated form L2− predicts that the most probable reactive sites for electrophilic attack are oxygen atoms. The result is agreement well with the experimental data of the crystal structure analyses.  相似文献

Schiff base ligand (L) was obtained by condensation reaction between 4-aminopyrimidin-2(1H)-one (cytosine) with 2-hydroxybenzaldehyde. The synthesized Schiff base was used for complexation with Cu(II) and Fe(II) ions used by a molar (2 : 1 mmol ration) in methanol solvent. The structural features of ligand, Cu(II), and Fe(II) metal complexes were determined by standard spectroscopic methods (FT-IR, elemental analysis, proton and carbon NMR spectra, UV/VIS, and mass spectroscopy, magnetic susceptibility, thermal analysis, and powder X-ray diffraction). The synthesized compounds (Schiff base and its metal complexes) were screened in terms of their anti-proliferative activities in U118 and T98G human glioblastoma cell lines alone or in combination with electroporation (EP). Moreover, the human HDF (human dermal fibroblast) cell lines was used to check the bio-compatibility of the compounds. Anti-proliferative activities of all compounds were ascertained using an MTT assay. The complexes exhibited a good anti-proliferative effect on U118 and T98G glioblastoma cell lines. In addition, these compounds had a negligible cytotoxic effect on the fibroblast HDF cell lines. The use of compounds in combination with EP significantly decreased the IC50 values compared to the use of compounds alone (p<0.05). These results show that newly synthesized Cu(II) and Fe(II) complexes can be developed for use in the treatment of chemotherapy-resistant U118 and T98G glioblastoma cells and that treatment with lower doses can be provided when used in combination with EP.  相似文献

The complexation of cyclohexanespiro-5-(2,4-dithiohydantoin), L, with copper and nickel was studied by means of experimental and theoretical methods. The Cu(I) and Ni(II) complexes were synthesized and characterized using 13C CPMAS NMR, IR and FAB-MS. Reduction of Cu(II) ions and the formation of Cu(I) complexes with dithiohydantoin was proved. Various coordination modes were investigated on the basis of calculated (DFT-GIAO) shielding constants of the free ligand and model structures of the complexes. General trends in the changes of spectroscopic parameters (NMR chemical shifts, vibrational modes) upon different types of coordination were outlined. Dimeric structures for the Cu(I) and Ni(II) complexes were proposed in which the ligands were coordinated in N3^S4- and N3^S2-bridging ways, respectively, acting as monoanions. The results demonstrate that the combined experimental (13C CPMAS NMR, IR) and theoretical (DFT) approach can be used to characterize the molecular structure of solid complexes for which crystallographic data are not available.  相似文献

A novel emissive tetra-naphthylmethylene pendant-armed macrocyclic ligand and a series of complexes with monovalent and divalent metal ions have been synthesized. Solid compounds have been isolated as mononuclear (Co(II), Cu(II) and Zn(II)) or dinuclear (Co(II), Ni(II), Cu(II), Zn(II), Cd(II) and Ag(I)), complexes, depending on the counterions used. The chemical and photophysical properties of the free ligand, the protonation behavior and its metal complexes have been investigated in solution. UV-Vis spectroscopy has revealed a 1:1 binding stoichiometry for Cu(II), Zn(II), Cd(II), Ni(II) and Co(II), and 2:1 molar ratio for Ag(I). In chloroform, the free ligand presents two emission bands related to the monomer naphthalene emission and a red-shifted band attibutable to an exciplex due to a charge transfer from the nitrogen lone electron pair to the excited chromophore. Upon protonation of the free amines or due to metal complexation, the exciplex band disappears. The crystal structure of [Ag2L(NO3)2] is also reported. The structure reveals that both metal ions are into the macrocyclic cavity in a distorted square plane {AgN3O} environment. Each Ag(I) atom interacts with two neighbouring amine nitrogen atoms, one pyridine nitrogen and one oxygen atom from a monodentate nitrate ion.  相似文献

In an effort to better understand the biological efficacy of the tridentate aroyl hydrazone Cu(II) complexes, three Cu(II) complexes of acetylpyridine benzoyl hydrazone (HL), [Cu(L)(NO3) (H2O)]·H2O (C1), [Cu(L)2] (C2) and [Cu(L)(HL)]·(NO3)(Sas) (C3) (Sas = salicylic acid) were synthesized and characterized. X-ray crystal structures and infrared (IR) spectra of the complexes reveal that the L ligand of C1 and C2 are predominantly in the enolate resonance form, while one L ligand in C3 is represented enolate resonance form and the other HL ligand exhibits keto resonance form. All Cu(II) complexes showed significantly more anticancer activity than the ligand alone. Interestingly, the Cu complexes where the ligand/metal ratio was 1:1 (C1) rather than 2:1 (C2 and C3) had higher antitumor efficacy. Moreover, the 1:1 Cu/ligand complex, C1, promotes A549 cell apoptosis possibly through the intrinsic reactive oxygen species (ROS) mediated mitochondrial pathway, accompanied by the regulation of Bcl-2 family proteins.  相似文献

The new complex compounds [RuLCl(p‐cymene)] ? 3H2O and [NiL2(H2O)2] ? 3H2O (L: 1‐{4‐[(2‐hydroxy‐3‐methoxybenzylidene)amino]phenyl}ethanone) were prepared and characterized using FT‐IR, 1H‐ and 13C‐NMR, mass spectroscopy, TGA, elemental analysis, X‐ray powder diffraction and magnetic moment techniques. Octahedral geometry for new Ni(II) and Ru(II) complexes was proposed. Thermal decomposition confirmed the existence of lattice and coordinated water molecule in the complexes. To determine the antioxidant properties of Schiff base ligand and its Ni(II), Ru(II) metal complexes, FRAP, CUPRAC, ABTS and DPPH methods of antioxidant assays were used. Moreover, enzyme inhibition of complexes was evaluated against carbonic anhydrase I and II isoenzymes (CA I and CA II) and acetylcholinesterase (AChE). For CA I and CA II, the best inhibition enzymes, was the Ni(II) complex with 62.98±18.41, 86.17±23.62 Ki values, whereas this inhibition effect showed ligand with 24.53±2.66 Ki value for the AChE enzyme.  相似文献

Novel calix[4]pyrrole bearing vic-dioxime ligand (LH2) of the general formula, R1R2C2N2O2H2 (where, R1 = C6H5- and R2 = C39H50N5-) has been synthesized by the reaction of anti-chlorophenylglyoxime with 3-aminophenylcalix[4]pyrrole at room temperature. The mononuclear Cu(II), Ni(II) and Co(II)complexes of this vic-dioxime ligand were prepared and their structures were confirmed by elemental analysis, FT-IR, TGA and magnetic susceptibility measurements; the HMBC, DEPT, 1H and 13C NMR spectra of the LH2 ligand were also reported. The electrochemical property of the complexes was investigated in DMSO by cyclic voltammetry at 200 mV s−1 scan rate. The cyclic voltammetric measurements clearly indicated that Co(LH)2·2H2O complex differs from the Ni(LH)2 and Cu(LH)2 complexes upon the exhibition of quasi-reversible one-electron transfer reduction process in the negative region instead of an irreversible process.  相似文献

Abstract

In this work, we have synthesized a few novel mononuclear complexes of Cu(II), Co(II), Ni(II) and Zn(II) using a pyrazolone-derived Schiff base ligand. They were characterized by spectroscopic and analytical methods. The elemental analyses, UV-Vis, magnetic moment values and molar conductance of the complexes reveal that the complexes adopt an octahedral arrangement around the central metal ions. The interaction of complexes with CT-DNA was studied by absorption spectral titration and viscosity measurements. The observed data show that the complexes bind with CT-DNA via an intercalation mode. Efficient pUC18 DNA cleavage ability of the synthesized compounds was explored by gel electrophoresis. The antimicrobial activity of these compounds against a set of bacterial and fungal strains reveals that the complexes exhibit better activity than the free ligand. Moreover, all the complexes were evaluated against two cancer (HeLa and HepG2) and one normal (NHDF) cell lines. The data were compared with cisplatin. Anti–inflammatory activity has been experimentally validated which proves that theoretical predictions concur with the experimental results. In addition, molecular docking studies have been performed to consider the nature of binding mode and binding affinity of these compounds with DNA (1BNA) and protein (3hb5). These studies reveal that the mode of binding is intercalation and the complexes have higher binding energy scores than the free ligand.  相似文献

The reaction of the tripod ligand hydrotris(2-mercapto-1-imidazolyl)borate Tmxylyl with zinc(II) perchlorate in methanol afforded the mononuclear complex of the type [Tmxylyl-Zn(mimxylyl)]ClO4 (1). Whereas under the same conditions, the reaction with copper(II) perchlorate gives rise to the simultaneous formation of the dinuclear copper(I) complex [TmxylylCu]2 (2). The chemical formulae of the complexes have been characterized by elemental chemical analysis, IR-NMR spectroscopies, and single crystal X-ray methods. In complex 1, the zinc(II) atom displays a distorted tetrahedral environment. While in complex 2, the Tmxylyl ligand bridges the two copper(I) atoms in an asymmetric manner with trigonal geometry. The inverted conformation of the ligand Tmxylyl at the boron center, allows the B-H units to be directed towards the copper centers. The greater reactivity of the borohydride groups towards metal centers enhances the reduction of Cu(II) to Cu(I). The obtained kinetic results for the methylation reactions of 1 and 2 indicate that these bound thione complexes are less suitable to electrophilic attack than the thiolate ligand.  相似文献

Cobalt(II), nickel(II), copper(II) and zinc(II) complexes with 2-acetylthiophene benzoylhydrazone have been synthesized and characterized by elemental analyses, magnetic susceptibility measurements, electronic, IR, NMR and ESR spectral techniques. The molecular structures of ligand and its copper(II) complex have been determined by single crystal X-ray diffraction technique. The Cu(II) complex possesses a CuN2O2 chromophore with a considerable delocalization of charge. The structure of the complex is stabilized by intermolecular π–π stacking and C–H?π interactions. Hatbh acts as a monobasic bidentate ligand in all the complexes bonding through a deprotonated C–O and >CN groups. Electronic spectral studies indicate an octahedral geometry for the Ni(II) complex while square planar geometry for the Co(II) and Cu(II) complexes. ESR spectrum of the Cu(II) complex exhibits a square planar geometry in solid and in DMSO solution. The trend g|| > g > 2.0023 indicates the presence of an unpaired electron in the dx2-y2 orbital of Cu(II). The electro-chemical study of Cu(II) complex reveals a metal based reversible redox behavior. The Ni(II) complex shows exothermic multi-step decomposition pattern of the bonded ligand. The ligand and its most of the metal complexes show appreciable corrosion inhibition properties for mild steel in 1 M HCl medium. [Co(atbh)2] complex exhibited the greatest impact on corrosion inhibition among the other compounds.  相似文献

The Consistent Force Field (CFF) method of conformational analysis was applied to ionic copper complexes with naturally occurring and allo-isomers of isoleucine and threonine, namely, [Cu(Thr)2H?2]?2, (Cu(Ile)]+, and (Cu(Thr)]+ Hydration stabilization by means of a hydration shell model, as well as the effect of ligated water, were also computed. The difference in stability between the complexes of naturally occurring and allo-isoleucine is in accordance with experimental data. The theoretical results on [Cu(Thr)]+ complexes suggest a possible role of specific intramolecular interactions in determining the observed stability difference.  相似文献

In the aqueous solution of copper(II) ions, bidentate L-α-alaninehydroxamic acid (CH3CH(NH2)-CONHOHHL) binds cupric ion forming of monodimeric and bis(L-α-alaninehydroxamato)copper(II) complexes. These complexes were studied by potentiometric, ESR and spectrophotometric methods.The ESR studies provide important evidence for the formation of different Cu(II) complexes with L-α-alaninehydroxamic acid, depending on pH. The ESR spectra can be used to follow the appearance of the individual complexes, to estimate the coordination sphere around Cu(II) and to observe the equilibria between different complexes.The solution electronic spectra are reported. The experimental curve was resolved into precise- positioned absorption bands by Gaussian analysis for the bis(L-α-alaninehydroxamato)copper(II) species. These data were used in a weak tetragonal ligand field model to calculate ligand field parameters.The distribution and the relevant stability constants of species present in aqueous solutions were obtained by analytical potentiometry.  相似文献

Two novel bolaamphiphile based dicarboxylic ligands L1H2 and L2H2 are synthesized by desymmetrizing aromatic anhydrides. The corresponding Cu(II) complexes [Cu(L1) · EtOH]2 (1), [Cu(L2) · (CH3CN)]2 (2) are synthesized and characterized. The crystal structure obtained for (1) and (2) indicates that they are new class of tetralactone type macrocyclic Cu(II) chelate complexes with paddle wheel Cu2-acetate cage structure. The 1:1, Cu(II) and ligand ratio leads into formation of a novel binuclear Cu(II) tetracarboxylate complexes. The macrocyclic chelate ring size in compounds 1 and 2 was altered from [15] membered to [19] membered by introducing phthalyl and diphenyl head groups as discussed in detail. The single crystal X-ray structure shows the Cu(II)?Cu(II) distance 2.613(13) Å for 1 and 2.626(13) Å for 2, the corresponding room temperature EPR spectra recorded for powdered polycrystalline samples indicate the existence of Cu(II)?Cu(II) dimeric system.  相似文献

The bidentate ligand benzylacetylacetone was used to synthesize the Cu(II) complexes 1 and 2 without and with 4,4-bipyridine ligand, respectively. The complexes were characterized by analytical and spectroscopic studies. The mononuclear complex [Cu(C10H9O2)2] (1) has been synthesized by the reaction of copper acetate with the ligand whereas the tetranuclear complex [Cu4(4,4-bpy)4(C10H9O2)4(C2H3O2)4] (2) has been synthesized by the reaction of copper acetate with the ligand followed by the addition of 4,4-bipyridine. The X-ray analysis shows that the complex 1 has square planar geometry and the complex 2 has square pyramidal geometry around the metal centers. The thermogravimetric studies showed that the complexes undergo decomposition in multiple steps.  相似文献

2,6-Bis(1-triazolo)pyridine (1) was synthesized and characterized via IR and NMR. The regiochemistry of the compound was confirmed via single crystal X-ray analysis of the hydrochloride salt. A series of insoluble complexes of the general formulae M(1)2(X)2 or M(1)(X)2 [M=Co(II), Ni(II), Cu(II); X=ClO4, BF4, Cl] were prepared and their structures interpreted in light of infrared spectra and composition analysis. The results suggest that first row transition metals are not chelated by 1, but rather form extended coordination polymeric networks. A second family of complexes was prepared using 2,6-bis(1-imidazolo)pyridine to support this interpretation.  相似文献

Two novel copper (II) substituted thiosemicarbazone Schiff base complexes [Cu(L1)(µ-SCN)]n(NO3)2 (1) and [Cu2(µ-SCN)(SCN)(L2)2](NO3) (2) have been synthesized by condensing substituted thiosemicarbazides like 4-methyl-3-thiosemicarbazide or 4-ethyl-3-thiosemicarbazide with 2-acetylpyridine. Both the metal complexes 1 and 2 are characterized using different spectroscopic techniques like IR, UV-Vis, ESR spectroscopy followed by elemental analysis, cyclic voltammetric measurement and single crystal X-ray structure analysis. X-ray crystal structure analysis reveal that complex 1 is polymeric while complex 2 is dimeric in nature. The coordination geometry around Cu(II) are square pyramidal in which thiosemicarbazone Schiff base ligand coordinate to the central Cu(II) atom in tridentate fashion. The prominent interaction patterns of 1 and 2 with CT-DNA were examined by employing electronic absorption and emission spectral titrations, cyclic voltammetry and viscosity measurements. All the results show that CT-DNA binds with both copper (II) complexes 1 and 2. Furthermore, protein binding ability in vitro of complexes 1 and 2 with both BSA and HSA were carried out using multispectroscopic techniques and a static quenching pattern was observed in both cases. Molecular docking study was employed to ascertain the exact mechanism of action of 1 and 2 with DNA and protein molecules (BSA and HSA). In vitro cytotoxicity activity of complexes 1 and 2 toward AGS and A549 was evaluated using MTT assay which demonstrates that both complexes 1 and 2 have superior prospectus to act as anticancer agents.
