The backcross progenies of the barley–wheat hybrids Hordeum vulgare L. (2n = 14) × Triticum aestivum L. (2n= 42) and two alloplasmic lines derived from them were studied using microsatellite markers of barley and wheat. The F1 hybrids and first backcross plants BC1 contained the genetic material of both cultivated barley and the cultivars of common wheat involved in developing of these hybrid genotypes. The genomes of BC3, BC4, and alloplasmic lines contained no microsatellite markers of the cultivated barley, whereas chromosomes of each homeologous group of common wheat were identified. In chromosomes of backcross progenies BC3, BC4, and alloplasmic lines yielded by backcrosses of hybrids and various common wheat cultivars, microsatellite markers of the parental wheat cultivars were shown to undergo recombination.  相似文献

The fertility characteristics expressed during morphogenesis in first-generation self-pollinated backcrossed progenies (BC1) obtained from amphiploid barley–wheat hybrids [Hordeum geniculatum All. (2n= 28) ×Triticum aestivum L. (2n= 42)] (2n = 70) backcrossed with common wheat were studied. It was found that, in the case of self-pollination of BC1 plants, karyotype stabilization leads to the formation of alloplasmic euploid (2n = 42), telocentric substitution (2n = 40 + 2t), and telocentric addition (2n = 42 + t), (2n = 42 + 2t) plant forms, which may serve as the sources of the respective alloplasmic lines of common wheat. That the expression of fertility characters in BC1F8plants was shown to depend on growth conditions. The main mechanism of hybrid incompatibility of BC1F1–BC1F8plants was expressed as grass-clump dwarfism.  相似文献

Genomic in situ hybridization (GISH) has been used to study characteristics of the formation of alloplasmic lines detected among self-pollinated backcrossed progeny (BC1F5–BC1F8) of barley–wheat amphiploids [Hordeum geniculatum All. (2n = 28) × Triticum aestivum L. (2n = 42)] (2n = 70). The chromosome material of the wild barley H. geniculatum has been shown to contribute to these lines. For example, fifth-generation plants (BC1F5) had genotypes (2n= 42w + 2g), (2n = 42w + 1g + 1tg), and (2n = 41w + 1g), where w is common wheat chromosomes, g is barley (H. geniculatum) chromosomes, and tg is the telocentric chromosome of wild barley. Beginning from theBC1F6 generation, alloplasmic telocentric addition lines (2n= 42 + 2tg) and (2n = 42 + 1tg) appear. This lines has been found cytogenetically unstable. The progeny of each of these cytological types include not only the (2n= 42 + 2tg) and (2n = 42 + 1tg) addition plants, but also plants with the monosomic (2n = 41 + 1tg) and the disomic (2n = 40 + 2tg) substitutions, as well as the (2n = 41 + 2tg) plants, which lack one wheat chromosome and have two telocentric barley chromosomes. It has been demonstrated that the selection for well-filled grains favors the segregation of telocentric addition lines (2n = 42 + 2tg) and (2n = 42 + 1tg).  相似文献

We studied some features of the development of self-fertile 42-chromosome lines on the base of self-pollination progeny of 46-chromosome plants obtained by backcrossing of barley-wheat hybrids Hordeum marinum subsp. gussoneanum Hudson (= H. geniculatum All.) (2n = 28) × Triticum aestivum L. (2n = 42). The stabilization of karyotypes, resulting in 42-chromosome plants of the wheat type was generally completed by generation BC1F10. The plants of all self-pollination progenies, including BC1F10, showed some phenotypic traits characteristic of wild barley. Plants of BC1F10 with the chromosome sets 2n = 42 and 2n = 42 + t were analyzed by RAPD with a set of 115 primers. Fragments of the wild barley genome were detected in RAPD patterns with 19 primers. Cross-hybridization confirmed that these fragments belonged to the wild barley genome. We raised four phenotypically different 42-chromosome lines from grains obtained from plants of generation BC1F10, and these lines proved to be cytogenetically stable and self-fertile when grown in the field.  相似文献

Two novel delphinidin 3-(tri or di)-glycoside-7-glycosides were isolated from the violet–blue flowers of Vinca minor L. and V. major L. (Family: Apocynaceae), and determined to be delphinidin 3-O-[2-O-(β-xylopyranosyl)-6-O-(α-rhamnopyranosyl)-β-galactopyranoside]-7-O-(α-rhamnopyranoside) [= delpphinidin 3-(2G-xylosylrobinobioside)-7-rhamnoside] as major floral anthocyanin of V. minor and delphinidin 3-O-[6-O-(α-rhamnopyranosyl)-β-galactopyranoside]-7-O-(α-rhamnopyranoside) [= delpphinidin 3-robinobioside-7-rhamnoside] as major floral anthocyanin of V. major by chemical and spectroscopic methods. In addition, chlorogenic acid and kaempferol 3-O-[6-O-(α-rhamnopyranosyl)-β-galactopyranoside]-7-O-(α-rhamnopyranoside) [= kaempferol 3-robinobioside-7-rhamnoside (robinin)] were identified in these flowers. In this paper, the relation between the structure of floral anthocyanins and classification of Vinca species was discussed.  相似文献

Chabaudacuaria n. g. is erected, as monotypic, for C. multispinosa (Pérez Vigueras, 1938) n. comb. (Spirurida: Acuariidae) [syns Cheilospirura multispinosa Pérez Vigueras, 1938; Acuaria multispinosa (Pérez Vigueras, 1938) Yamaguti, 1961]. This species, a parasite of various ardeid birds, is redescribed by means of light and scanning electron microscopy based on material from great blue herons Ardea herodias L. and yellow-crowned night herons Nyctanassa violacea (L.) in Florida, USA. Chabaudacuaria n. g. resembles Acuaria Bremser, 1811, Cheilospirura Diesing, 1861, Skrjabinocerca Schikhobalova, 1930 and Xenocordon Mawson, 1982 in its straight cordons which do not anastomose. However, it can be distinguished from them by the didelphic-prodelphic uterus and the absence of caudal alae in the males. In the pattern of its cordons (consisting of a row of plates and a longitudinal ridge) and the absence of an area rugosa, the new genus is similar to Chevreuxia Seurat, 1918, Syncuaria Gil??bert, 1927, Aviculariella Wehr, 1931, Skrjabinocara Kurashvili, 1940, Decorataria Sobolev, 1949 and Desportesius Chabaud & Campana, 1949, which are characterised by anastomosing cordons. The didelphic-prodelphic female reproductive system of Chabaudacuaria is intermediate between the didelphic-amphidelphic uterus of Chevreuxia and the monodelphic-prodelphic uterus of Syncuaria, Aviculariella, Skrjabinocara, Desportesius and Decorataria. Therefore, the straight and non-anastomosing cordons are considered to be autapomorphic for Chabaudacuaria.  相似文献

Summary Chloroplast and mitochondrial compartments of a parental line of wheat (Triticum aestivum L., cv. Moisson) and its anther-derived doubled haploid lines have been analyzed and compared on the basis of their DNA restriction patterns. The results obtained show that no noticeable difference can be detected between doubled haploid lines and parental line at the level of ctDNA and mtDNA organization. It may be concluded that in vitro culture by itself does not systematically generate a cytoplasmic variation in germ cells.  相似文献

Summary The phenology and build-up of spikelet number under 10 h day-length were studied in five wheat lines: the multispikelet line Noa, the regular line Mara, the F1 hybrid between them and monosomics 2D of Mara and of this hybrid (lacking the 2D chromosome of Mara). Noa had a longer spike development phase, a higher initial number of spikelet primordia and a slower rate of spikelet production than Mara. The F1 hybrid was similar to Noa in its high initial number of spikelets and to Mara in its high rate of spikelet production. This hybrid had a shorter spikelet phase than both parents. Deletion of one dose of the Mara 2D chromosome from either Mara or the F1 hybrid caused a reduction in the rate of spikelet production and an increase in the duration of the spikelet phase. These effects were due to the reduced dosage of the 2D chromosome. However, in the F1 hybrid this deletion also caused an increase in the spike development phase — an indication that Noa carries on its 2D chromosome a recessive gene for late heading date which acts on the spike development phase. This gene of Noa is independent of the day-length sensitive gene ppd, and is different from Noas dominant gene for large initial number of spikelets.  相似文献

The pattern of homoeologous metaphase I (MI) pairing has been fully characterized in durum wheat × Aegilops cylindrica hybrids (2n = 4x = 28, ABCcDc) by an in situ hybridization procedure that has permitted individual discrimination of every wheat and wild constituent genome. One of the three hybrid genotypes examined carried the ph1c mutation. In all cases, MI associations between chromosomes of both species represented around two-third of total. Main results from the analysis are as follows (a) the A genome chromosomes are involved in wheat–wild MI pairing more frequently than the B genome partners, irrespective of the alien genome considered; (b) both durum wheat genomes pair preferentially with the Dc genome of jointed goatgrass. These findings are discussed in relation to the potential of genetic transference between wheat crops and this weedy relative. It can also be highlighted that inactivation of Ph1 provoked a relatively higher promotion of MI associations involving B genome.  相似文献

A study is made of the seasonal and daily variations in the concentrations of pollen ofOlea europaea L. over three consecutive years (1993–1995) in the atmosphere of Jaén (southern Spain). A Burkard volumetric spore trap was used for sampling. The results show that the highest concentrations of airborne olive pollen occur during May and the first 2 weeks of June, when levels often exceed 500 grains/m3 and occasionally reach nearly 5000 grains/m3 (the levels of allergenic pollen in the atmosphere of Jaén are among the highest in Europe). Over the 3-year study period a significant seasonal variation was detected, not only in the development of the principal pollination period, but also in the value of the maximum pollen concentrations recorded.  相似文献

Somatic embryogenesis in cotton (Gossypium hirsutum L.) is accelerated when the plant regeneration medium is supplemented with haemoglobin (erythrogen). In cotton SVPR 2 lines, a higher frequency of embryoid formation was observed when the medium contained 400 mg/l haemoglobin. Fresh weight of the callus, rate of embryoid induction, number of embryoids formed and the percentage of plant regeneration from somatic embryos were increased. Among the two different cultivars tested, MCU 11 showed no response to the presence of haemoglobin when compared to SVPR 2, and embryogenic callus formation was completely absent in the former. Medium containing MS salts, 100 mg/l myo-inositol , 0.3 mg/l thiamine-HCL, 0.3 mg/l Picloram (PIC), 0.1 mg/l kinetin and 400 mg/l haemoglobin effected a better response with respect to embryogenic callus induction. After 8 weeks of culture, a high frequency of embryoid induction was observed on medium containing MS basal salts, 100 mg/l myo-inositol, 0.3 mg/l PIC , 0.1 mg/l isopentenyl adenine, 1.0 g/l NH4NO3 and 400 mg/l haemoglobin. Plant regeneration was observed in 75.8% of the mature somatic embryos, and whole plant regeneration was achieved within 6–7 months of culture. The regenerated plantlets were fertile and similar to in vivo-grown, seed-derived plants except that they were phenotypically smaller. A positive influence of haemoglobin was observed at concentrations up to 400 mg/l at all stages of somatic embryogenesis. The increase in the levels of antioxidant enzyme activities, for example superoxide dismutase and peroxidase, indicated the presence of excess oxygen uptake and the stressed condition of the plant tissues that arose from haemoglobin supplementation. In cells of Zea mays (root hairs, coleoptiles) and Riccia fluitans (rhizoids, thalli) intracellular Ca2+ and pH have been measured with double-barrelled microelectrodes. Free Ca2+ activities of 109–187 nM (Riccia rhizoids), 94–160 nM (Riccia thalli), 145–231 nM (Zea root hairs), 84–143 nM (Zea coleoptiles) were found, and therefore identified as cytoplasmic. In a few cases (Riccia rhizoids), free Ca2+ was in the lower millimolar range (2.3±0.8 mM). A change in external Ca2+ from 0.1 to 10 mM caused an initial and short transient increase in cytoplasmic free Ca2+ which finally levelled off at about 0.2 pCa unit below the control, whereas in the presence of cyanide the Ca2+ activity returned to the control level. It is suggested that this behaviour is indicative of active cellular Ca2+ regulation, and since it is energy-dependent, may involve a Ca2+-ATPase. Acidification of the cytoplasmic pH and alkalinization of the vacuolar pH lead to a simultaneous increase in cytoplasmic free Ca2+, while alkalinization of pHc decreased the Ca2+ activity. Since this is true for such remote organisms as Riccia and Zea, it may be concluded that regulation of cytoplasmic pH and free Ca2+ are interrelated. We found a potent platelet aggregation inhibitor in the culture broth of Streptomyces strain M-193. The inhibitor was purified as white crystals and investigated; it was identified as staurosporine. It strongly inhibited the platelet aggregation induced by collagen or ADP with IC50 values of 3.4 μm, and 11.6 μm, respectively. The inhibitor had no effect on thrombin-induced platelet aggregation or membrane stabilization against heat-induced hemolysis.  相似文献

Abstract

This paper presents an interdisciplinary approach to crop improvement that links physiology with plant breeding and simulation modelling to enhance the selection of high‐yielding, drought‐tolerant varieties. In a series of field experiments in Queensland, Australia, we found that the yield of CIMMYT wheat line SeriM82 ranged from 6% to 28% greater than the current cultivar Hartog. Physiological studies on the adaptive traits revealed that SeriM82 had a narrower root architecture and extracted more soil moisture, particularly deep in the profile. Results of a simulation analysis of these adaptive root traits with the cropping system model APSIM for a range of rain‐fed environments in southern Queensland indicated a mean relative yield benefit of 14.5% in water‐deficit seasons. Furthermore, each additional millimetre of water extracted during grain filling generated an extra 55 kg ha?1 of grain yield. Further root studies of a large number of wheat genotypes revealed that wheat root architecture is closely linked to the angle of seminal roots at the seedling stage – a trait which is suitable for large‐scale and cost‐effective screening programmes. Overall, our results suggest that an interdisciplinary approach to crop improvement is likely to enhance the rate of yield improvement in rain‐fed crops.  相似文献

Summary Differences in levels of resistance toSeptoria tritici blotch were observed in plants with a specific height-reducing gene. When the gene Rht 2 was present either as an isoline or in the progeny, a higher degree of resistance was found. The most susceptible plants were observed in populations carrying the Rht 1 gene. Associations, as determined by phenotypic correlations, were detected betweenSeptoria tritici blotch and tall stature, late heading, and maturity. Plants having short stature, early heading, early maturity, and acceptable levels of resistance were identified in the F2 population whenRht 2 was present. Results of this study indicated that wheat breeders must select the appropriate dwarfing source that may confer resistance and grow large F2 populations, in order to increase the probability of obtaining desired genotypes.  相似文献
