Changes in chicken embryo thymus after partial decerebration (including the hypophysis) and after hypophyseal or thymic allograft were investigated. Chicken embryos were partially decerebrated at 36–40 h of incubation and on day 12 received a hypophysis or a thymus allograft from 18-day-old donor embryos. The thymuses of normal, sham-operated and partially decerebrate embryos were collected on day 12 and 18. The thymuses of the grafted embryos were collected on day 18. The samples were examined with histological method and tested for the anti-PCNA and anti-CD3 immune-reactions. After partial decerebration, the thymic cortical and medullary compartments diminished markedly in size. Anti-PCNA and anti-CD3 revealed a reduced immunereaction, verified also by statistical analysis. In hypophyseal or grafted embryos, the thymic morphological compartments improved, the anti-PCNA and anti-CD3 immune-reactions recovered much better after the thymic graft, probably due to the thymic growth factors and also by an emigration of thymocytes from the same grafted thymus.Key words: hypophysectomy, hypophyseal and thymic allograft, chicken embryonal thymus, PCNA, CD3 markers.  相似文献

In human embryos the hypophyseal sac (Rathke's pouch) originates at the roof of the mouth until stage 15 as a broad rim. As the mandibular arch and the maxillary swelling enhance the mesodermal masses in forming the early palatal shelves the rim is reduced to a cleft of about 0.2 mm in broadness in stage 17. From stage 18 up to stage 23 there is a prominent papilla in the midline of the mouth's roof which later on may become recanalized. The different SEM-aspects of the pharyngeal hypophysis are demonstrated.  相似文献

The development of the intermediate lobe of the hypophysis was studied in the embryonic C3H mouse; at least four glands from embryos of every gestational day from 15 to 19 were examined. In the 16 day-old embryo prospective secretory cells proliferate at the centre of the intermediate lobe anlage. At the same stage cylindrical cells bordering the hypophyseal cleft begin to reorganize into marginal cells. By the end of fetal life marginal cells are well differentiated. In the 17 day-old embryo a few granular inclusions appear in some centrally located cells. Secretory cells increase in number during the following two embryonic days. Some of these cells contain polymorphic populations of granular and vesicular inclusions by gestational day 19. The possibility of a dual formation of secretory inclusions is discussed. The result implies that the onset of granule-formation by these cells is not contemporaneous with the start of production of melanophore-expanding substances, the presence of which has been detected by earlier biological assays.  相似文献

Pituitary sections from 15 to 21 day-old rat foetuses have been studied with the immunofluorescence technique, using antibodies anti alpha-MSH, anti beta-MSH and anti beta (1-24) ACTH. The first ACTH cells appear on day 17 of pregnancy in the pars distalis of the hypophysis and only on day 18 in the pars intermedia. beta-msh cells have been observed on day 16 in the pars anterior and on day 17 in the pars intermedia, while alpha-MSH cells appear only on day 18 and exclusively in the pars intermedia. The cytodifferentiation of the beta-MSH and ACTH cells occurs in the pars intermedia with about a 24 hours delay in comparison to that of the pars distalis. The first revealed cells are always located in the posterior half of the pituitary gland. The corticostimulating activity of the hypophysis has been tested with the fluorescence intensity of the corticotrophs, the adrenal weight, the adrenal content in corticosterone and the plasma corticosterone level. The fluorescence of the corticotrophs increases on day 18, shows a maximum on day 19 and decreases until term. The adrenal weight rises regularly between day 16 to day 20, thereafer the increase subsides. Adrenal and plasma corticosterone concentrations reach a peak on day 19 of pregnancy. These data suggest that hypophyseal corticostimulating activity is very high between days 18 and 19 and decreases between days 19 and 21.  相似文献

Differentiation of the thyroid in the hypophysectomized chick embryo   总被引:1,自引:0,他引:1  
The influence of hypophyseal hormones on the normal development of the thyroid gland was investigated by decapitating chick embryos at a time before the hypophysis has begun to form. Thyroids from decapitated embryos were compared with thyroids of normal embryos from shortly after decapitation to the time when the embryo should hatch. The thyroids were examined for ultrastructural differences and for accumulation of thyroglobulin. Thyroxine levels in the blood were also examined. It was concluded that the thyroids in decapitated embryos develop normally up to about 12 days of development, by which time morphogenesis and differentiation of the gland is essentially complete. The rates of thyroid growth and accumulation of thyroglobulin are diminished after 12 days of development. The amount of thyroxine in the blood is decreased by about one-half. Thyroxine was found in the yolk of unincubated eggs at levels sufficient to sustain blood thyroxine levels throughout embryonic development. We conclude that hormones that are synthesized by the embryonic hypophysis affect thyroid development only through their influence on generalized growth and metabolic activity.  相似文献

To test the capacity of the epithelial component of the chick embryo thymus to induce tolerance to major histocompatibility complex (MHC) antigens, pre-colonized thymic rudiments were grafted into chick embryonic recipients. Semi-allogeneic or allogeneic transplantations were done between two lines of chickens histocompatible at the MHC locus. Approximately 10% of these thymic chimeras hatched and were studied 3 mo after hatching. Thymic grafts were not rejected by the allogeneic host. The tolerance of chimeric chickens to thymus donor MHC antigens was tested by using a skin graft rejection test and a graft-vs-host (GvH) assay. Chimeric chickens that received an MHC-incompatible thymic graft during the embryonic life tolerated skin graft with the MHC haplotype of the thymus donor. Nevertheless, the lymphocytes within the thymic graft, the host thymus, and the blood were tolerant to the host MHC antigens but were alloreactive in GvH reaction for the MHC antigens of the thymic graft type. These results suggest that the epithelial component of the thymus when taken before the starting of the colonization by hemopoietic precursors and grafted into an early chick embryonic host can induce a tolerance for the MHC determinants involved in allograft rejection but not in the GvH reaction.  相似文献

Previous studies in our laboratory have demonstrated that the presence of the thymus is essential for rapid and stable tolerance induction in allotransplant models. We now report an attempt to induce tolerance to kidney allografts by transplanting donor thymic grafts simultaneously with the kidney in thymectomized recipients. Recipients were thymectomized 3 wk before receiving an organ and/or tissues from a class I-mismatched donor. Recipients received 1) a kidney allograft alone, 2) a composite allogeneic thymokidney (kidney with vascularized autologous thymic tissue under its capsule), or 3) separate kidney and thymic grafts from the same donor. All recipients received a 12-day course of cyclosporine. Thymectomized animals receiving a kidney allograft alone or receiving separate thymic and kidney grafts had unstable renal function due to severe rejection with the persistence of anti-donor cytotoxic T cell reactivity. In contrast, recipients of composite thymokidney grafts had stable renal function with no evidence of rejection histologically and donor-specific unresponsiveness. By postoperative day 14, the thymic tissue in the thymokidney contained recipient-type dendritic cells. By postoperative day 60, recipient-type class I positive thymocytes appeared in the thymic medulla, indicating thymopoiesis. T cells were both recipient and donor MHC-restricted. These data demonstrate that the presence of vascularized-donor thymic tissue induces rapid and stable tolerance to class I-disparate kidney allografts in thymectomized recipients. To our knowledge, this is the first evidence of functional vascularized thymic grafts permitting transplantation tolerance to be induced in a large animal model.  相似文献

Ovaries from 10- to 18-day-old chick embryos hypophysectomized by partial decapitation were cultured in vitro and their estradiol secretion was compared to that of ovaries from control embryos. The production of estradiol was not less in the decapitated than in the control embryos at 15-18 days, neither per ovary nor on the basis of ovarian weight. However, the difference was significant at 10-11 days. These results suggest that the hypophysis controls estradiol secretion by the chick embryo ovary in the early stages, but not in the later ones.  相似文献

Experiments involving sequential transplantations of the chick embryonic thymus at E9 to E12 into a first 3-day host quail embryo and then into a second chick host allowed demonstration of the cyclic periodicity of hemopoietic cell seeding of the embryonic thymus. After a first wave of colonization occurring between E6.5 and E8, the thymus becomes refractory to hemopoietic cell entry for about 4 days. It resumes its capacity to be seeded by a second wave of blood-borne stem cells at E12. After a second period of non receptivity starting at E14, a third wave of incoming cells reaches the thymus around E18. Therefore, with a slightly different periodicity, the same cyclic mechanism regulates the renewal of lymphocytes in chick and quail embryos. Quail hemopoietic cells were immunostained in the chimeric thymuses, with a species specific monoclonal antibody (anti-MB1) which recognizes a common surface antigenic determinant on all endothelial and blood cells of the quail (except erythrocytes). Two steps could thus be distinguished in the seeding process. When the thymus becomes receptive for hemopoietic cells, the latter first accumulate in the intrathymic blood vessels before penetrating massively in the thymic parenchyma. The quail chick-chimera system combined with the use of a species- and cell-type-specific antibody provides a unique tool for studying thymic colonization by lymphocyte precursors.  相似文献

Chick embryos were hypophysectomized by partial decapitation at the stage of 42 h of incubation and grafted with a hypophysis from a 12-days-old donor embryo on the chorio-allantoic membrane at 9 1/2 days. Two days later, their ovary was removed for organ culture and its oestradiol secretion rate was compared to that of the ovary of hypophysectomized, non grafted control embryos. The oestradiol secretion rate in the grafted embryos was almost twice that in the hypophysioprivic embryos and in the range of that in normal embryos. This result suggests that the hypophysis controls oestradiol secretion of the ovary in the 11 1/2-days-old chick embryo.  相似文献

The appearance of C-type virus particles in thymus cells of Swiss mouse embryos, 11.5 to 15.5 days post-conception age (PCA), was studied with the electron microscope. In thymic rudiments of all specimens examined, virus particles were seen in epithelial cytoplasm, budding from epithelial cell surfaces and in extracellular spaces. Lymphoid cells were first seen in thymic rudiments of 13.5 days PCA, and did not display virus particles at this stage. At 14.5 days PCA, thymic lymphocytes had localized plasmalemmal thickenings of high electron-density which were adjacent to extracellular virus particles. Viruses appeared to be penetrating thymic lymphocytes by viropexis in embryos of 15.5 days PCA. At this stage, many lymphocytes also had cytoplasmic virus-containing vesicles and viral buds at their surfaces. These observations suggest the possibility that, in embryos, C-type viruses are transmitted horizontally from thymic epithelium to early populations of thymic lymphocytes.  相似文献

A significant increase in ovarian volume and in the numbers of germ cells was observed in rabbit foetuses hypophysectomised by decapitation on day 19 and killed on day 28 when compared with unoperated females of the same litter. Extirpation of the thymus, during its differentiation as a lymphoid organ, in the early larval life of the leopard frog was attended by a marked impairment of allograft immunity. In cases in which thymectomy was most successful—i.e., in which no thymic tissue was found in the larvae on later autopsy—the survival of allogeneic grafts was greatly prolonged. When thymectomy was less successful—i.e., when fragments ranging widely in size were found in the larvae at autopsy—the results were variable: the larvae either rejected allografts more slowly than control larvae or responded to them at the same rate as the control larvae. The “less successful” cases of thymectomy most likely represented instances of the regeneration of thymic tissue.  相似文献

Male and female rats were given oestradiol benzoate (1 mg s.c. twice a week for 3 weeks) and/or sodium nitroprusside (SN), a donor of nitric oxide (NO), which was administered in their food in amounts of 0.2 or 0.6 mg/rat/day. Neither oestradiol-induced hypertrophy of the hypophysis, nor the serum prolactin (PRL) level, was affected by the simultaneous administration of SN. The PRL content of the hypophysis rose after oestradiol in the males, but the increase was again uninfluenced by the simultaneous administration of SN and the cAMP content of the hypophysis--raised after oestradiol--was likewise unaffected. The amount of cGMP in the hypophysis after oestradiol rose only in males. Both the serum and the hypophyseal prolactin level were found to be correlated to the cAMP and the cGMP content of the hypophysis. It was found that the simultaneous administration of SN together with oestradiol slightly reduced the increase in the cGMP content of the hypophysis elicited with oestradiol treatment only.  相似文献

The mitogen effect on migration of eosinophils and monocytes was studied in embryonic chickens. On the 13th embryonic day, chickens were injected with mitogens, such as concanavalin A (Con A), phytohemagglutinin-P (PHA-P), and lipopolysaccharide (LPS), into the allantoic cavity, and the mitogenic effect was estimated from the relative frequencies of eosinophils and monocytes by enumerating the number of oxidase positive cells (OPC) in the spleen, thymus, and bursa of Fabricius. Splenic frequencies of OPC increased in the embryos treated with mitogens. Similar influences were also detected in the thymic OPC. Higher responses were seen on the 18th embryonic day in the number of splenic OPC when embryos were treated with Con A or PHA-P than with LPS. These findings suggest that Con A and PHA-P are preferential OPC accumulation promoters. However, bursal frequencies of OPC in the cortex were low after mitogenic stimulations when compared with controls, although appreciable responses were detected in the bursal medulla after LPS stimulation. These results suggest that the migration pattern in the population of eosinophils and monocytes is affected not only by T cell mitogens but is also derived from LPS stimulation.  相似文献

The aim of the present study was to evaluate the effect of hypophysectomy on cell proliferation in the left ovary and the left testis of 8- to 14-day-old chick embryos. Hypophysectomy was performed by the partial decapitation technique. At 44-46 h of incubation, chick embryo heads were sectioned at the mesencephalic level and the prosencephalic region removed. Embryos were further incubated until 8-14 days of development. Cell division was evaluated by bromodeoxyuridine (BrdU) incorporation and by counting the total number of somatic and germ cells in the gonads. The ovary displayed an exponential increase in the number of somatic and germ cells and a higher rate of BrdU incorporation compared to the testis. BrdU incorporation was reduced in the ovary of hypophysectomized embryos at 9-14 days of incubation, while in the testis, the reduction was significant at 14 days of development. Changes in the total number of somatic and germ cells further suggest that the absence of hypophysis affects the growth of the ovary earlier than the growth of the testis. Reduction in the number of somatic and germ cells after hypophysectomy in the ovary was reversed by a hypophyseal graft on the chorioallantoic membrane. The adenohypophysis regulates, probably through gonadotropic hormones, proliferation of somatic and germ cells in the gonads during chick embryo development.  相似文献

Experiments on adult and old rats have shown age-related decrease of the CRF incretion in the hypothalamus, weakening of its response to dexamethasone, as well as a decrease of receptor corticosterone binding. Against this background in the hypophysis of old rats sensitivity ox CRF and serotonin increased, in stress the same content of ACTH as in adult rats secreted, it more markedly reacted to dexamethasone, and receptor of corticosterone binding increased in hypophysis. In old age, the role of hypophyseal link in control of the adrenocortical function is assumed to increase in stress.  相似文献

For the present study we used the classic model of early partial decapitation (DCx) of chicken embryos (Fugo, J. Exp. Zool., 85: 271-298, 1940; Betz, Gen. Comp. Endocrinol., 9: 172-186, 1967) in an attempt to analyze the neuroendocrine immune relationships during ontogeny. The elimination of the prosencephalon in chickens at 33-38 hr of incubation induced profound structural, histochemical, and morphometric changes in the embryonic development of the thymus gland. These included a greater development of the cortex than of the medulla, an increased mitotic index, high numbers of pyknotic cells, and enlarged connective tissue trabeculae containing numerous large lymphoid cells; hypertrophied reticular-epithelial cells; delayed appearance of medullary epithelial cysts; and intrathymic granulopoiesis. Furthermore, preliminary radioimmunoassays revealed a sharp increase in the values of circulating thymic hormones, mainly thymosin beta 4 in 17-day-old embryos. The results are discussed with regard to the possible role of prolactin, thyroxine, testosterone, and thymic hormones in the ontogenic development of the chicken thymus.  相似文献

In the present investigation the localization and activity of alkaline, neutral, and acid hydrolases of the thymus were studied during development of rats and mice and of various adult species using histochemical methods. If different procedures of tissue pretreatment were employed, several inhibition effects and morphological as well as enzyme histochemical artifacts occurred dependent on the mode of tissue pretreatment. After embedding in glycol methacrylate, sections of the thymus showed a better structural preservation than cryostat sections but were accompanied by a drastic decrease of activity and low localization quality of the final reaction products especially in the case of protease studies with 4-methoxy-2-naphthylamine peptides as substrates. Smears of thymic cells facilitated the allocation of enzymes to mobile or fixed cells in the stroma of the thymus. The perivascular localization of aminopeptidase M could only be shown with combined techniques. In comparison, primarily the proteases yielded information on the thymic stroma and in this context especially on the epithelial reticular cells and the stroma proper but also on thymocytes (lymphocytes) and enabled a species-dependent subdivision of the thymic reticulum already in the light microscope. Enzyme histochemically the development of the rat and mouse thymus could be subdivided into an early period and perinatal (pre- and postnatal) period of functional differentiation. Morphological (proliferation of cortical lymphocytes) and enzyme histochemical changes (disappearance of dipeptidylpeptidase IV, significant loss of alkaline phosphatase activity and beginning activity increase of aminopeptidase M) occurred primarily at the transition from the early to the prenatal period. During the postnatal phase, a significant activation of lysosomal enzymes in the thymic medulla and general enzymatic differentiation of the cortical epithelial reticular cells were found. Species differences and species similarities for the respective enzymes and their localization as well as for the thymic cells were noticed for adult rats, mice, guinea-pigs, hamsters, and marmoset monkeys. Differences were true especially for the thymocytes; less species differences were seen for the epithelial reticular cells; capsular and perivascular connective tissue and the macrophages behaved rather similarly. Species-independently certain medullary epithelial reticular cells showed high and typically localized alkaline phosphatase activities and species-dependently also high activities of neutral hydrolases.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献
