The objective of this article is to study the effect of 5-aminolevulinic acid (ALA) and enhanced chlorophyll content, antioxidative enzymes and photosynthesis rate by foliar application of ALA. We evaluated three concentrations (control-distilled water, T1-50 mg l−1, T2-150 mg l−1, T3-250 mg l−1) of ALA and seven cultivars, “Sanchidaye” (Sa-1), “Lichuandasuomian” (Li-1), “Aijiaohuang” (Ai-1), “Qingyou” No. 4 (Qi-1), “Aikang” No. 5 (Ak-1), “Hanxiao” (Ha-1) and “Shulv” (Sl-1). “Ak-1” showed strongest response of POD (peroxidase) enzyme activity (0.4 U g−1 min−1) in 250 mg l−1 ALA solution. The highest CAT (catalase) activity (0.8 U g−1 min−1) after administration of 250 mg l−1 ALA was observed in “Li-1”. Meanwhile, highest (1.42 mg l−1) total chlorophyll content was also observed in “Ak-1”, when leaves were treated in 50 mg l−1 ALA, “Li-1” and “Ai-1” showed strongest response of specific activity of superoxide dismutase (SOD) in 50 mg l−1 and 50 mg l−1 ALA. Two hundred and fifty milligram per milliliter of ALA-treatment significantly improved the net photosynthetic rate.  相似文献

Shoots of rose (hybrid tea) cv. “First Prize” were induced to flower in vitro on Murashige and Skoog (MS) medium containing various sucrose concentrations (15, 30 or 45 g l−1) and different phytohormone combinations of different cytokinins [N6-benzyladenine (BA); thidiazuron (TDZ) and zeatin] with α-naphthaleneacetic acid (NAA). Results indicate that sucrose is the key factor in floral morphogenesis while cytokinin increases the flowering percentage and helps the normal development of floral buds. From the three cytokinins that were used, BA and zeatin were considered to be more suitable as inductive flowering agents than TDZ. Reduced inorganic and organic salt concentration in MS media had a positive effect on in vitro flowering. The morphology of shoots bearing floral buds varied with different cytokinin treatments. The highest percentage (45%) of flowering was obtained on MS medium supplemented with 3.0 mg l−1 BA, 0.1 mg l−1 NAA and 30 g l−1 sucrose.  相似文献

The influence of 5-aminolevulinic acid (ALA) on the growth, photosynthesis, and fruit quality of gibberellin-induced seedless 2-year-old “Delaware“ grapevine was examined. The result of soil treatment (0.1–10 mg l−1) and foliar spray treatment (30–300 mg l−1) with ALA after flowering was significant growth improvement, up to 55% in the lateral shoot, and up to 38% in leaf area. Optimal doses were 1 mg l−1 soil treatment and 100 mg l−1 foliar treatment. The photosynthetic rate of plants treated with ALA increased by a significant 9.2%–22.5%. Relative to the control, based on measurements of the 5th leaf of each shoot in the ripening period, the optimum concentrations for growth and photosynthesis enhancement in grapevines were 100 mg l−1 (foliar treatment) and 1 mg l−1 (soil treatment). Total plant weight at harvest increased a significant 13% at the optimal treatment doses. In terms of fruit quality, the cluster fresh weight increased a significant 44.9%–53% and fruit colour showed a tendency to become darker in all plants treated with ALA. The °Brix value in the plant treated with 100 mg l−1 ALA was a significant 2.7% higher than that of the control. We consider that leaf area and photosynthetic rates during cultivation are important for the acquisition of photoassimilates and that these are likely to be causally associated with improvement of total dry weight and advance of fruit quality. In addition, a possibility of advancing the harvest time of grapes by ALA treatment was shown.  相似文献

The objective of the present work was selection of cultivar and suitable medium for regenerating shoots from leaf segments of non-heading Chinese cabbage. We evaluated six types of supplemented media with 2.0, 5.0 and 10.0 mg l−1 6-BA; 1.0 and 2.0 mg l−1 TDZ; 0.1, 0.3, 0.5, 0.8 and 1.0 mg l−1NAA; 3.0, 5.0 and 7.5 mg l−1AgNO3; 0.01 mg l−1 2–4, D and 4.0 mg l−1 KT for shoot regeneration and six cultivars “Sanchidaye”, “Liuchuandasuomian”, “Qingyou 4”, “Liangbaiye”, “AiKang 5” and “Hanxiao F3”, furthermore for root formation three types of supplemented media with 0.2, 0.3, 0.5 mg l−1 NAA, and for survival rate two types of base media: turf + vermiculite + manure (1:2:0.2) and soil + vermiculite (1:2). Culturing leaf segments on MS medium supplemented with 2 mg l−1 TDZ; 0.5 mg l−1 NAA and 7.5 mg l−1 AgNO3 gave the highest number of shoots per leaf segment (66) while roots were best formed on the medium supplemented with 0.2 mg l−1 NAA. Survival rate was highest (61.6%) in the turf: vermiculite: manure (1:2:0.2) medium. The highest percentage of responding leaf segments, number of shoots per leaf segment, rooting percentage and survival rate were observed in “Liuchuandasuomian”. The plantlets were transferred to the soil and grown into mature plants in pots. These results could be used for preliminary selections of cultivars to transfer disease resistance (Bt) gene through agrobacterium in non-heading Chinese cabbage.  相似文献

The effects of silicate and glucose on growth and eicosapentaenoic acid (EPA) production by the diatom Nitzschia laevis were studied. By alternately altering the concentrations of silicate (2.7–64 mg l−1) and glucose (1–40 g l−1) in the medium, the highest cell dry weight (ca. 5.5 g l−1) was obtained at 20 g l−1 glucose and 32 mg l−1 silicate, while the highest specific growth rate (ca. 0.65 day−1) was obtained at a relatively low glucose concentration (5 g l−1) and high silicate concentrations (32–64 mg l−1). At glucose levels of 5 and 20 g l−1, EPA content was higher with lower silicate concentrations (2.7 and 16 mg l−1 silicate, respectively), while at a silicate level of 16 mg l−1, higher glucose concentrations (20–40 g l−1) facilitated EPA formation. The highest EPA yield (131 mg l−1) was obtained at 20 g l−1 glucose and 32 mg l−1 silicate, while the highest EPA productivity (15.1 mg l−1 day−1) was obtained at 20 g l−1 glucose and 64 mg l−1 silicate. In view of antibiotics being detected in surface waters, experiments were conducted to determine the impacts of tetracycline on planktonic bacteria in wetland and river waters. The minimum inhibitory concentration (MIC) method is often used to measure for resistance or susceptibility of microbes to antibiotics with typical concentrations of antibiotics being mg L−1. Moreover, there is the belief that antibiotics in the lower μg L−1 range are unlikely to affect bacteria. We examined this assumption by measuring the effects of a broad range of tetracycline concentrations on bacterial protein production by the incorporation of l-[4,5-3H]leucine method. Tetracycline significantly (P < 0.05) inhibited production in river water bacteria at a “free” concentration of 5 μg L−1, but the inhibition was significant only at 1000 μg L−1 in wetland water. The data indicate that planktonic bacteria can be very sensitive to tetracycline at extremely low concentrations and that microbial production is seriously affected.  相似文献

Highly efficient (>90%) protocols were developed for in vitro regeneration from de-embryonated cotyledon explants of peanut (Arachis hypogaea L.). Phytohormone combinations and concentrations, explant source and orientation, period of incubation and the response of genotypes were examined for optimization of the regeneration efficiency. Adventitious shoot primordia could be induced from de-embryonated cotyledon explants when (1) the proximal end of the explant was kept in contact with the shoot induction medium-I supplemented with 5 mg l−1 BAP + 2 mg l−1 2,4-D for 4 weeks, or (2) the distal end was kept in contact with shoot induction medium-II supplemented with only BAP at 20 mg l−1 for the first 2 weeks, followed by subculture in the same medium containing 15 mg l−1 BAP for the next 2 weeks. Orientation of placing the explant on the above media was critical for in vitro regeneration. The factors affecting the morphogenic responses like, repetitive organogenesis, shoot elongation, in vitro flowering and rhizogenesis were examined. Shoot bud formation was genotype independent. Histological studies showed multicellular origin of adventitious shoot primordia. The protocols gave healthy and fertile plants within 4 months.  相似文献

Yellow lupin (Lupinus luteus) cv. Juno seedlings exposed to increasing concentrations of Pb2+ (50–350 mg l−1) were analysed in respect to its effect on the degradation of lipids, the content of antioxidants (ascorbate, α-tocopherol) and the activity of the ascorbate glutathione cycle enzymes (dehydroascorbate reductase DAR; EC 1.8.5.1 and glutathione reductase GR; EC 1.6.4.2). Lipid peroxidation, expressed as the content of TBArm (thiobarbituric acid reactive metabolites), increased only at 50 and 100 mg l−1 Pb2+, whereas at higher lead concentrations it decreased as compared with the control. The level of free fatty acids was not significantly affected as a result of Pb2+ exposure, except for 300 mg l−1 Pb2+. The content of α-tocopherol increased significantly at the range of concentrations between 150 and 300 mg l−1 and at these concentrations lipid peroxidation was inhibited. Ascorbic acid (AA) and dehydroascorbic acid (DHA) content increased considerably in roots treated with 100 and 150 mg l−1 Pb2+. In general the activity of DHAR and GR was stimulated by lead, however at higher Pb2+ concentrations (300 and 350 mg l−1) GR revealed lower enzymatic activity. Our results suggest that in lead-treated roots lipids might be protected against reactive oxygen species (ROS) by lipid-soluble molecules of α-tocopherol and this direct defense seems to be supported by AA as well as the enzymes: DHAR and GR.  相似文献

Four antibiotics were evaluated for their effects on eliminating the hypervirulent Agrobacterium tumefaciens strain C58C1 ATHV RifR (pEHA101)/p35-gus-intron from walnut somatic embryos and on the production of secondary somatic embryos and the transformed somatic embryos. Exposure to 100–1000 mg l−1 of ampicillin, carbenicillin or cefotaxime respectively for up to 60 days did not eliminate the A. tumefaciens while timentin at 500–1000 mg l−1 eradicated it from somatic embryos. One-hour acidified medium treatments and the addition of 100 mg l−1 kanamycin to 500 mg l−1 ampicillin, carbenicillin, cefotaxime or timentin were of little help in eliminating the Agrobacterium. All four antibiotics reduced somatic embryo production, carbenicillin minimally and cefotaxime maximally, especially at higher concentrations, in comparison with antibiotic-free medium. Putative transformed embryos were selected for continued proliferation on a 100 mg l−1 kanamycin-containing medium. Histochemical assessments indicated that more gus-positive somatic embryos, particularly fully gus-positive embryos, regenerated from timentin-containing medium than from other antibiotic-containing media under equivalent conditions. A procedure for in vitro culture of the parasitic flowering plant western hemlock dwarf mistletoe, Arceuthobium tsugense (Rosend.) G.N. Jones subsp. tsugense, is described. A factorial experiment evaluated the effects of media (Harvey's medium (HM) and modified White's medium (WM)), temperatures (15 °C and 20 °C), presence or absence of light, and plant growth regulators (the auxin 2,4-dichlorophenoxyacetic acid (2,4-D) and the cytokinin 6-benzylaminopurine (BAP) at varying concentrations (0.001 mg l−1 to 1 mg l−1)). Seed explants germinated in less than one week in culture and produced radicles. Optimal conditions for radicle elongation were WM at 20 °C in the presence of light and without plant growth regulators. Some of the radicles split at the tip to yield callus while others swelled to become spherical holdfasts. Holdfasts were also produced at the tips of radicles, and callus arose from split holdfasts. Factors that promoted holdfast production were Harvey's medium, light, and 2,4-D at 1 mg l−1. Callus development from split radicles and split holdfasts was optimal on WM with 0.5 mg l−1 2,4-D and 1 mg l−1 BAP at 20 °C in the dark. Eichhornia crassipes (Mart.) has strong ability to remove Cu2+ from copper-contaminated water. Physiological responses in E. crassipes exposed to known concentrations of Cu2+ were examined in this study, and demonstrated that E. crassipes could accumulate 314 mg kg−1 dry weight of Cu when exposed to 5 mg l−1 of Cu2+ for periods up to 14 d. However, there were marked changes in physiology of the plant commencing at Cu2+ concentrations of 1 mg l−1. Results of this study showed that E. crassipes could tolerate moderate concentrations (i.e. 0.5 mg l−1) of Cu2+, without significant changes in photosynthetic pigment concentrations, while high concentrations (i.e. 5 and 10 mg l−1) of Cu2+ resulted in substantial loss in pigment concentrations. Increases in malondiadehyde (MDA) content were also demonstrated in plant exposure to high Cu2+ concentrations. Soluble protein content increased to a level slightly higher than the control at <0.5 mg l−1 of Cu2+, but then decreased with exposure to >1 mg l−1 of Cu2+. Our results suggest that E. crassipes has a substantial capacity to accumulate copper when cultivated at moderate concentrations of Cu2+, without marked changes in its physiology. The findings indicate that E. crassipes is a promising possibility for phytoremediation of moderately Cu-contaminated water bodies. Handling editor: S. M. Thomaz  相似文献

Summary Ovule perforation technique and media components (plant growth regulators andl-glutamine) were tested on in vitro growth of immature (<3 mm) embryos of “Springcrest” and “Earligrande” peaches. Ovule perforation was 2 to 4 times more effective in promoting embryo growth than leaving ovules intact.l-Glutamine (400 mg·liter−1) promoted an increase in growth but could not be used with indole-acetic acid plus kinetin because an antagonistic effect on embryo growth occurred. This study investigated the factors affecting in vitro flowering of Perilla frutescens. The shoots regenerated from cotyledonary and hypocotyl explants cultured on Murashige and Skoog (MS) medium supplemented with benzyladenine (BA) and indole-3-acetic acid, each at 0.5 mg l−1, were excised and transferred to MS medium containing 30 g l−1 of sucrose, 8.25 g l−1 of ammonium nitrate, and 1.0 mg l−1 of BA. After 40 d of culture, 86.2% of shoots flowered and most of which self-fertilized in vitro and produced mature fruits with viable seeds. These seeds were germinated and plants were grown to maturity and flowered in soil under greenhouse conditions. The in vitro flowering system reported in this study may facilitate rapid breeding of P. frutescens and offers a model system for studying the physiological mechanism of flowering.  相似文献

The effects of antibiotics commonly used in Agrobacterium-mediated transformation were studied on Pinus pinaster tissues. Embryogenic tissue growth from three embryogenic lines and adventitious bud induction from cotyledons from three open-pollinated seed families were analysed. Cefotaxizme, carbenicillin and timentin commonly used for Agrobacterium elimination, at concentrations of 200–400 mg l −1 did not inhibit the embryogenic tissue growth on filter paper nor as clumps. Adventitious bud induction and bud number were significantly reduced for one of the tested families when using 400 mg l−1 cefotaxime or timentin. The selection agent kanamycin significantly inhibited growth of embryogenic tissue on filter paper in all the embryogenic lines␣and concentrations tested (20–50 mg l−1). Kanamycin also inhibited growth of embryogenic clumps after two subcultures at 5–50 mg l−1. In␣cotyledons, kanamycin inhibited adventitious bud␣formation in the three seed families used, regardless of the concentrations tested (5–25 mg l−1). There was a significant effect of the seed family on the bud induction and the number of adventitious buds produced. From the results obtained, we propose the use of timentin to eliminate Agrobacterium in transformation experiments, at concentrations of 400 mg l−1 for embryogenic tissues and of 300 mg l−1 for cotyledons. For selection of transformed tissues carrying the kanamycin resistance gene, kanamycin should be used at 20 mg l−1 for embryogenic tissues on filter paper, at 5 mg l−1 when clumps are in direct contact with the selection medium, and bellow 5 mg l−1 for adventitious bud induction.  相似文献

The aim of the study was to examine the possibility of propagating in vitro four of the most common cultivars in Tuscany (central Italy): Terom, Violetto di Toscana, Chiusure and Empolese. The first three belong to the “Violetti” group, while cv Empolese belongs to the “Romaneschi” group. Explants were cultured on an induction medium (IM), which is a modified MS medium consisting of nitrate concentrations reduced by one quarter, 0.8 mg L−1 6-benzylaminopurine (BA) and 0.2 mg L−1 3-indole butyric acid (IBA). Explants were then transferred to a proliferation medium (PM) consisting of the same basal medium together with 0.03 mg L−1 BA and 0.05 mg L−1 gibberellic acid (GA3). A rooting double-phase was then established. The pre-rooting medium (PRM), consisting of a basal MS medium with half strength nitrate concentrations, 0.5 mg L−1 indole-3-acetic acid (IAA) and 1 mg L−1 paclobutrazol (PBZ) was used for two weeks. Over the next four weeks, a rooting medium (MR) was used, consisting of a basal MS medium with 2 mg L−1 β-cyclodextrin and 2 mg L−1 α-naphthaleneacetic acid sodium salt (NAA). The cv Empolese provided the highest number of proliferated explants and rooted plantlets using the method described.  相似文献

An investigation into the role of carbohydrates in flowering of the endangered species Kniphofia leucocephala Baijnath. (Asphodelaceae) in vitro revealed that a carbohydrate source is essential for the induction of inflorescences. Both the concentration and type of sugar influenced the percentage of flowering, with 60 g l−1 fructose and 10 μM N6-benzyladenine (BA) inducing the best flowering response. A high percentage of flowering was also observed with 60 g l−1 glucose and 10 μM BA. The optimal concentration of sucrose for flower induction was 30 g l−1, beyond which the flowering percentage declined, but could be partially restored by increasing the BA concentration. Although overall plant growth declined at high sugar concentrations, there does not appear to be any significant correlations between various growth parameters and flowering percentage, suggesting that sugars play a direct role in floral transition in vitro.  相似文献

Summary This study was conducted to examine the effect of biotin and thiamine concentrations on callus growth and somatic embryogenesis of date palm (Phoenix dactylifera L.). Embryogenic callus derived from offshoot tip explants was cultured on hormone-free MS medium containing biotin at 0, 0.1, 1, or 2 mg l−1 combined with thiamine at 0.1, 0.5, 2, or 5 mg l−1. Embryogenic callus weight, number of resultant embryos, and embryo length were significantly influenced by thiamine and biotin concentration. The optimum callus growth treatment consisted of 0.5 mg l−1 thiamine and 2 mg l−1 biotin. This treatment also gave the highest number of embryos. Embryo elongation was greatest at 0.5 or 2 mg l−1 thiamine combined with 1 mg l−1 biotin. Embryos from all treatments germinated and regenerants exhibited normal growth in soil. This study provides an insight into the importance of optimizing various culture medium components to overcome in vitro recalcitrace of date palm.  相似文献

Physiological effects of exposure to silver (AgCln n−1; 250 μg Ag l−1 or 1000 μg Ag l−1) in seawater fish were investigated using adult starry flounders. While all fish survived up to 10 days in 250 μg Ag l−1, flounders started to die after day 4 in 1000 μg l−1. Dose-dependent increases in plasma and hepatic silver concentrations showed that silver was available for uptake. There were minimal negative effects on hematological parameters, acid-base status, and blood gases. Plasma ammonia showed a pronounced (three- to four-fold), but transient increase in flounders exposed to either 250 μg Ag l−1 or 1000 μg Ag l−1. Whole body ammonia and acid equivalent efflux measurements indicated that ammonia retention was due to a combination of stimulated production and inhibited excretion. In the 1000-μg Ag l−1 group there was a similar transient increase in plasma [magnesium], which was restored by day 4. In contrast, plasma chloride and sodium levels increased gradually towards the point when fish began to die. At 250 μg Ag l−1, the Na+/K+-ATPase activity of the intestine was unaffected but there was a two-fold increase in branchial Na+/K+-ATPase activity. The latter effect was interpreted as compensation for an elevated chloride and sodium load. The increases in plasma chloride and sodium concentrations were accompanied by a marked suppression of drinking, thereby indicating that acute silver toxicity was likely caused by a combination of elevated electrolyte concentrations and dehydration. Summary A procedure for the regeneration of complete plantlets of Tylophora indica from cultured leaf callus via somatic embryogenesis is described. Callus induction from leaf explants was on Murashige and Skoog (MS) medium with different concentrations of 2,4-dichlorophenoxyacetic acid (2.4-D; 0.03–3 mg l−1; 0.0–13.56 μM) and kinetin (Kn; 0.01 mg l−1; 0.05 μM). The best response for callus induction was obtained on MS medium containing 2 mg l−1 (9.04 μM) 2.4-D and 0.01 mg l−1 (0.05 μM) Kn. After two subeultures on the same medium the embryogenic callus was transferred to MS medium with different concentrations of the cytokinin, 6-benzyladenine (0.5–3 mg l−1; 2.22–13.32 μM) and 2-isopentenyladenine (2ip; 0.53 mg l−1; 2.46–14.76 μM) along with 0.01 mg l−1 (0.05 μM) indole-3-butyric acid (IBA) for somatic embryo development and maturation. MS medium with 2 mg l−1 (9.84 μM) 2ip produced the maximum number of mature somatic embryos. The mature embryos were bipolar and on transfer to MS basal medium produced complete plantlets. After hardening the regenerants were planted in the Gudalur forests of Western Ghats. Total DNA was extracted from 14 regenerants and the mother plant. Random amplified polymorphic, DNA (RAPD) analysis was carried out using 20 arbitrary oligonucleotides. The amplification products were monomorphic among all the plants revealing the genetic homogeneity and true-to-type nature of the regenerants.  相似文献

To determine the sources and sinks of atmospherically deposited Pb at a forested watershed (Plastic Lake) in central Ontario, Canada, Pb pools and fluxes through upland, wetland and lake compartments were measured during 2002/2003 and compared with previous measurements taken between 1989 and 1991. In 2002/2003, annual bulk deposition of Pb was 0.49 mg m−2 compared with 1.90–1.30 mg m−2 in 1989–1991. Annual Pb concentrations in stream water draining the upland part of the catchment were very low (0.04 μg l−1) and were approximately half those measured in 1989–1991 (0.11–0.08 μg l−1). Leaching losses in stream water were small and mass balance estimates indicate almost complete retention (>95%) of atmospherically deposited Pb in upland soils. In contrast, annual Pb concentrations in stream water draining a wetland were between 0.38 and 0.77 μg l−1, with the highest concentration occurring in 2002/2003 and mass balance calculations indicate that the wetland is a net source of Pb in all measured years. Lead concentrations in the lake outflow were low and the average Pb concentration measured in 2002/2003 (0.09 μg l−1) was approximately half the value recorded in 1989–1991 (0.19 μg l−1 both years). Annual mass balance estimates indicate that the lake retained between 2.47 mg m−2 (1989/1990) and 1.42 mg m−2 (2002/2003) and that in 2002/2003 68% of the Pb input to the lake is derived from the terrestrial catchment. These estimates are higher than sediment core records, which indicate around 18 mg m−2 Pb was retained in sediment during the 1990s. Nevertheless, Pb concentrations decrease with sediment depth and 206Pb/207Pb concentrations increase with depth, a pattern also observed in mineral soils that reflects the substantial contribution of anthropogenic Pb to the watershed. Lead isotope data from soil and sediment indicate a recent anthropogenic Pb signal (206Pb/207Pb ∼ 1.185) in upper soils and sediments and an older anthropogenic signal (206Pb/207Pb ∼ 1.20) in deeper soil and sediment. Lead isotope data in sediment and vegetation indicate that practically all the Pb cycled in the forest at Plastic Lake is anthropogenic in origin.  相似文献
