The physiological parameters of microcuttings, namely leaf area, weight (wt) and position on the stock shoot, had significant effects on root vigour, microshoot growth and incidence of shoot tip necrosis (STN) in cultures of grape (Vitis vinifera L.) ‘Arka Neelamani’. Single-node leafy cuttings cultured in MS medium containing 1 μM IAA and 0.1 μM GA3 generally rooted first and subsequently sprouted into a single shoot. Small leafed cuttings exhibited slow root emergence, poor quality roots, early sprouting and weak shoot growth. Large leafed cuttings on the other hand, showed early rooting, vigorous roots, delayed sprouting and healthier shoots. Significant correlations were observed between fresh root wt per plantlet at 1 month in culture and wt of lamina, total wt and leaf area of the cuttings in that order. A significant correlation also existed between wt of roots and height or wt of the sprout that developed. The study suggests that the weight or area of the leaf governed the root growth in a microcutting. STN was observed in some plantlets particularly those derived from large leafed cuttings. Such cuttings showed vigorous roots and delayed but fast sprout growth that ended in STN. Plantlets showing STN had less Ca++ and Mg++ in the shoot tissue than in the shoots of normal plantlets while the roots showed similar Ca++ or higher Mg++ contents. The presence of leaf in microcuttings of grape cvs. Arka Neelamani and Thompson Seedless promoted rooting in vitro (MS, 1 μM IAA, 0.1 μM GA3, 3% sucrose) but the effect varied depending on the number of leaves and position of the leaf on the cutting. Single node cuttings with a full-length lower internode and a lamina at top (LAT) showed earlier rooting and more root and shoot growth than cuttings with lamina positioned at the middle (LAM), while cuttings with a leaf at the base (LAB) of the cutting and full-length upper internode exhibited a lower percent rooting and sprouting, poor root and shoot growth, and low survival. Partial or complete removal of the upper internodal segment in LAB cuttings improved rooting and sprouting suggesting the possible operation of an inhibitory effect by the upper internode. Retaining an upper leaf in LAB cuttings (LAB+UL) resulted in necrosis of the upper leaf often followed by the lower one. The extent of necrotic damage was influenced by the leaf area and position or age of the cutting on the stock shoot. Retaining the lower internode in LAB and LAB+UL cuttings which held the node–leaf junction away from the medium, or reducing the concentration of MS medium helped significantly in improving the survival and performance of these cuttings. The difference in reaction between LAB and LAT cuttings was attributable mainly to the difference in the sensitivity of the stem part that came in contact with the medium. Removal of the leaf in LAB cuttings reduced this sensitivity. The majority of the LAB and LAB+UL cuttings, as well as non-rooting or delayed rooting LAT and LAM cuttings, exhibited high purple pigmentation of leaf, petiole and stem. Two-leafed cuttings in vitro showed poor survival, less rooting and low plantlet output compared to single-leafed cuttings. 长白落叶松 (Larixolgensis)扦插繁殖的试验结果表明 ,嫩枝扦插效果好于硬枝扦插 ,并且母树年龄是影响其扦插生根的重要因子。母树年龄为 2年生时 ,生根率达 95 %以上。本文在此试验的基础上 ,对嫩枝和硬枝插穗中内源植物激素水平进行了测定。结果发现嫩枝插穗中IAA、CTKs的含量要高于硬枝插穗中IAA、CTKs的含量 ,并且随母树年龄的增加而降低 ;相反 ,嫩枝插穗中ABA含量要低于硬枝插穗中ABA的含量 ,并且随着母树年龄的增加而增加。实验结果表明 :IAA、ABA的含量和扦插生根有密切关系。IAA促进生根 ,ABA抑制生根。IAA/ABA比值可以用来衡量长白落叶松的生根能力  相似文献

3种植物生长调节剂对树头菜扦插生根的影响   总被引:2,自引:0,他引:2  
利用植物生长调节剂不同种类及质量浓度处理树头菜插穗,了解其扦插生根机制并筛选出适宜扦插繁殖的生长调节剂及浓度配比,为其扩大繁殖和生产提供科学依据。以树头菜一年生枝条为材料,采用3种植物生长调节剂(IAA、NAA、ABT-1)的3种不同质量浓度进行正交试验,清水处理(CK)作为对照,扦插3个月后对其芽生长量及生根性状(生根率、生根量、最长根长、平均根长等)进行测定并综合分析。本次试验筛选出的树头菜扦插最优方案为A1B2C2,即ABT-1质量浓度为50 mg·L-1、NAA质量浓度为200 mg·L-1、IAA质量浓度为200 mg·L-1,该处理生根率(83.33%)、平均根长(24.03 mm)、最长根长(46.13 mm)、平均根数(24条)、根系效果指数(19.06)为CK的2.27、2.97、3.03、4.50、12.79倍,根系发育较好。不同植物生长调节剂在适宜的浓度下混合利用可以显著增加树头菜的生根能力,进而加快生长并提高其扦插生根率。  相似文献

The influence of branches and mature leaves on the rooting and subsequent development of cuttings was examined, using Cotinus coggygria cv. Royal Purple. A model system was developed, whereby branched cuttings could be harvested from stock hedges and manipulated to alter leaf area, the number of actively‐growing, lateral branches and thus the source: sink ratio for photoassimilates. Highest percentage rooting ((80%) was promoted by retention of branches and a full leaf area. Reducing leaf area resulted in a lower rooting percentage (44%); however, greatest reductions in rooting were associated with the removal of lateral branches ((22%). Applying exogenous auxin (indole‐3‐butyric acid) at the excision point where branches had been removed significantly improved rooting potential, but did not fully substitute for the presence of branches with active shoot tips. Negative effects associated with removing a proportion of mature leaves appeared to relate to alterations in carbon balance rather than an influence on the supply of endogenous auxin to the potential rooting zone. The use of branched cuttings accelerated root and shoot development and resulted in a finished plant being produced more rapidly than is achieved from conventional, non‐branched cuttings. The results presented indicate a means for improving the efficiency of production of Cotinus coggygria, which may be applicable to a wider range of ornamental plants.  相似文献

In order to improve vegetative propagation of a difficult to root Cotinus coggygria the stock plants were subjected to: etiolation, shading and spraying with IBA, combined with the application of two commercially available rooting powders. The IBA treatment was more suitable for rooting of C. coggygria cuttings than the NAA application and it enhanced rhizogenesis regardless of the form of auxin application (foliar application to a stock plant or a rooting powder used directly on cuttings) and the amount of light provided to stock plants. Etiolation did not improve rhizogenesis in stem cuttings, however, reduction of light intensity by 50% and 96% of the ambient prior to harvest of cuttings affected rooting positively. Positive effects of shading can be ascribed to changes in shoot anatomy, i.e. a weaker sclerenchyma development. Synergistic effect of shading and foliar auxin application can result from the increase in leaf blade area and/or thinner lower epiderm. Enhanced rooting in cuttings from shoots grown out under reduced light intensity was accompanied by decrease in the contents of total soluble sugars, soluble proteins and free ABA and by increase in total chlorophyll, free amino acids, polyphenolic acids and free IAA contents.  相似文献

To determine the contribution of polar auxin transport (PAT) to auxin accumulation and to adventitious root (AR) formation in the stem base of Petunia hybrida shoot tip cuttings, the level of indole-3-acetic acid (IAA) was monitored in non-treated cuttings and cuttings treated with the auxin transport blocker naphthylphthalamic acid (NPA) and was complemented with precise anatomical studies. The temporal course of carbohydrates, amino acids and activities of controlling enzymes was also investigated. Analysis of initial spatial IAA distribution in the cuttings revealed that approximately 40 and 10 % of the total IAA pool was present in the leaves and the stem base as rooting zone, respectively. A negative correlation existed between leaf size and IAA concentration. After excision of cuttings, IAA showed an early increase in the stem base with two peaks at 2 and 24 h post excision and, thereafter, a decline to low levels. This was mirrored by the expression pattern of the auxin-responsive GH3 gene. NPA treatment completely suppressed the 24-h peak of IAA and severely inhibited root formation. It also reduced activities of cell wall and vacuolar invertases in the early phase of AR formation and inhibited the rise of activities of glucose-6-phosphate dehydrogenase and phosphofructokinase during later stages. We propose a model in which spontaneous AR formation in Petunia cuttings is dependent on PAT and on the resulting 24-h peak of IAA in the rooting zone, where it induces early cellular events and also stimulates sink establishment. Subsequent root development stimulates glycolysis and the pentose phosphate pathway.  相似文献

Shoot proliferation and rooting of three cranberry (Vaccinium macrocarpon Ait.) cultivars Bergman, Pilgrim, and Stevens were obtained in vitro on a modified nutrient medium containing zeatin following a one-step procedure. Bergman and Stevens differed in terms of shoot height, leaf number per shoot, rooting frequency, root number per explant, and root length; this was manifested with various concentrations of zeatin. Shoots proliferated and roots developed best when nodal segments were cultured in the medium supplemented with very low concentration of zeatin (2–4 μM). Such zeatin-induced tissue culture (TC) shoots of Bergman, Pilgrim, and Stevens were rooted ex vitro and compared with those propagated by conventional softwood cuttings (SC) for growth and morphology over four growth seasons. Significant interactions for leaf number per upright were observed among the treatments. The cultivars differed in terms of runner number per plant, upright length, number of leaves per upright, and shoot vigor. The propagation method had an effect on morphology of cranberry plants. The TC plants produced more runners and uprights with more leaves per upright than the conventional cuttings. This increase in vegetative growth of in vitro-derived plants over stem cuttings varied among genotypes. In vitro culture on zeatin-containing nutrient medium apparently induces the juvenile branching characteristics that favored enhanced vegetative growth with more shoots and leaf production.  相似文献

The basal cut end of coppice shoot cuttings of Pongamia pinnata was treated for 24 h with 0 (water treated control) or 5.0 mmol/L of KMnO4, KCI, and KH2PO4 or 2.5 mmol/L of K2HPO4 and K2SO4. Inorganic salts of P, S, Cl and Mn significantly influenced IAA ionization and adventitious rhizogenesis. P and S salts had lower IAA ionization potential, but more pronounced effect on adventitious rhizogenesis than Cl and Mn salts. The linear regression analysis also established negative correlations between salt induced IAA ionization with various characteristics of adventitious rhizogenesis such as sprouting (r = -0.83, p < 0.05), rooting (r = -0.82, p < 0.05), root number (r = -0.95, p < 0.01), and root length (r = -0.80, p < 0.1). The implication of IAA ionization in adventitious rhizogenesis has been discussed and the possible role of inorganic salts therein suggested.  相似文献

Photoperiod controls the initiation and development of roots on cuttings of Bryophyllum tubiflorum. Root initiation occurred when either the mother plant or the cuttings were exposed subsequently to SD conditions. Cuttings from LD plants exposed to LD did not root at all even after 4 weeks showing that short days are necessary for rooting of cuttings of this plant. The short day requirement can be substituted by IAA or IBA as roots were initiated in cuttings from LD plants when treated with the auxins under LD conditions. GA stimulates rooting of cuttings under SD conditions but does not replace the SD requirement. It failed to induce rooting under LD conditions.  相似文献

Adventitious root formation in stem cuttings of mung bean was enhanced by ethrel, which had an additive effect when employed simultaneously with indolebutyric acid (IBA). Abscisic acid (ABA) did not influence the number of roots per cutting whereas gibberellic acid (GA3) and kinetin were without effect on rooting at lower concentrations but were inhibitory at higher concentrations. Nevertheless, all three of these chemicals showed synergistic interactions with IBA and/or indol-3-ylacetic acid (IAA) and thereby significantly promoted root formation. A localised application of morphactin to the epicotyl of cuttings totally inhibited root production irrespective of which of the foregoing growth regulators were suppliedvia the hypocotyl. Morphactin application also prevented root formation in cuttings treated with vitamin D2. The various growth regulators employed had differing effects on growth of roots but there was no simple relationship between their effects on root formation and subsequent root growth.  相似文献

BACKGROUND AND AIMS: The aim of this study was to determine the role of nitrogen- and storage-affected carbohydrate availability in rooting of pelargonium cuttings, focusing on the environmental conditions of stock plant cultivation at low latitudes, transport of cuttings, and rooting under the low light that prevails during the winter rooting period in Central European greenhouses. METHODS: Carbohydrate partitioning in high-light-adapted cuttings of the cultivar 'Isabell' was studied in relation to survival and adventitious root formation under low light. Effects of a graduated supply of mineral nitrogen to stock plants and of cutting storage were examined. KEY RESULTS: Nitrogen deficiency raised starch levels in excised cuttings, whereas the concentrations of glucose and total sugars in leaves and the basal stem were positively correlated with internal total nitrogen (Nt). Storage reduced starch to trace levels in all leaves, but sugar levels were only reduced in tissues of non-nitrogen deficient cuttings. Sugars accumulated in the leaf lamina of stored cuttings during the rooting period, whereas carbohydrates were simultaneously exhausted in all other cutting parts including the petioles, thereby promoting leaf senescence. The positive correlation between initial Nt and root number disappeared after storage. Irrespectively of storage, higher pre-rooting leaf glucose promoted subsequent sugar accumulation in the basal stem and final root number. The positive relationships between initial sugar levels in the stems with cutting survival and in leaves with root formation under low light were confirmed in a sample survey with 21 cultivars provided from different sources at low latitudes. CONCLUSIONS: The results indicate that adventitious rooting of pelargonium cuttings can be limited by the initial amount of nitrogen reserves. However, this relationship reveals only small plasticity and is superimposed by a predominant effect of carbohydrate availability that depends on the initial leaf sugar levels, when high-light adaptation and low current light conditions impair net carbon assimilation.  相似文献

为探明有鳞大花亚组杜鹃扦插生根的最佳IBA浓度和扦插时间,该研究以江西杜鹃、百合花杜鹃为材料,分别采用腐叶土+河沙(1:1)、泥炭+珍珠岩+蛭石(3:1:1)基质,开展了4个IBA浓度和4个扦插时间的生根试验.结果表明:IBA浓度对除老叶留存数外的所有指标有显著影响,其中100 mg·L-1 IBA处理生根率、新梢长最大,腐烂率最低,其它指标也表现良好,为最佳生根浓度;50 mg·L-1 IBA处理根幅、新梢率最大,但不定根数最少,效果其次;200 mg·L-1 IBA处理促进根系生长,但生根率较低、特别是显著抑制新梢发育;对照处理生根效果最差.扦插时间对所有生根指标均有显著影响,早春(04-18)木质化硬枝扦插除老叶留存数较差外,其它指标均表现极佳,为最适扦插时间;秋季(10-19)半木质-木质化过渡枝扦插效果其次;夏季(06-21)嫩枝及(08-16)半木质化枝生根效果极差,不宜进行扦插育苗.物种、基质对生根指标也有显著影响,百合花杜鹃扦插生根能力强于江西杜鹃,泥炭+珍珠岩+蛭石(3:1:1)基质生根效果优于腐叶土+河沙(1:1).该研究结果首次发现早春新梢萌发前采用木质化硬枝扦插可以显著提高两种杜鹃的生根效果,为该亚组杜鹃的扦插育苗提供了科学依据.  相似文献

MM. 106 apple rootstock plants grown in a polythene tunnel show greater apical dominance and a higher propensity to root as cuttings than plants grown in the field. Experiments were conducted to test the hypothesis that the growth habit and rooting behaviour of polythene tunnel plants were caused by increased concentrations of idole-3yl-acetic acid. Cuttings taken from field-grown plants which had been sprayed with IAA showed increased rooting. In shoots of both field-grown and polythene tunnel-grown plants endogenous IAA levels were highest in the upper shoot region and declined progressively with distance from the apex. Plants grown in the polythene tunnel, however, did not contain significantly higher IAA levels than field plants. Lignosulfonates (LIGNs), low-cost by-products from the paper industry, are already commercialized as fertilizers; they stimulate both vegetative and reproductive growths and fructification. LIGNs have been tested in in vitro cultures and here too, they improve shoot growth and vigor, and rooting of various plant materials. This study aimed at to extend the in vitro application of LIGNs at different developmental stages in order to increase the productivity of systems generating vitroplants. The present results showed the beneficial effects of various LIGN applications on growth of a tropical orchid, Phalaenopsis, multiplication of Saintpaulia ionantha and rooting of poplar and Sequoiadendron sempervirens shoot cuttings. One of the most interesting observations was the stimulating effect of Ca-chelated LIGN on growth of Phalaenopsis and on rooting of Sequoiadendron. The significant and reproducible effects of LIGNs at several steps of micropropagation of different plant materials represent a potential tool to improve quality without embarrassing side-effects.  相似文献

The effects of indole-3-acetic acid (IAA) and four IAA conjugates, indoleacetylalanine (IAAla), indoleacetylaspartic acid (IAAsp), indoleacetylglycine (IAGly), and indoleacetylphenylalanine (IAPhe), on growth and morphogenesis in tomato leaf discs in vitro were examined. Free IAA stimulated root initiation in the absence of cytokinin and stimulated callus growth in the presence of 0.89 M benzylaminopurine (BAP). Free IAA also inhibited shoot initiation obtained with 8.9 M BAP. The activities of the IAA conjugates depended on the conjugating amino acid, the concentration of the conjugate, and the response being measured. IAAsp had little or no activity in promoting root initiation or callus growth or in inhibiting shoots, while IAPhe was similarly inactive except at the highest concentration tested (100 M). IAAla and IAGly were both very active in inhibiting shoots and promoting callus growth, but were much less active in stimulating rooting, except at 100 M, at which concentration they were as effective as free IAA. Thin-layer chromatography of the IAA conjugates revealed that IAAla, IAGly and IAPhe were largely stable to autoclaving, but that IAAsp underwent some hydrolysis to products identical with free IAA and aspartic acid. Pretreatment of seedlings with IAA, IAAla or IAGly altered the subsequent shoot initiation response from leaf discs on media with and without IAA.  相似文献

Propagation by softwood canes and cuttings is preferred as a practical system for vegetative reproduction of many ornamental plant species, despite the advances in tissue culture techniques. Dracaena purplecompacta L. is a species that has a high demand for exports. Conversely, coconut water (CW) is a rich supplement that naturally contains plant growth regulators such as indole acetic acid (IAA). The objective of this work was to evaluate the potential of CW extracts containing natural IAA, on adventitious root development in vegetative propagation of ornamental plant canes of D. purplecompacta L. Five different concentrations (28, 57, 143, 286, 571 μM of natural IAA) of CW extracts were tested. Another set of treatment was carried out with the same concentrations of authentic IAA hormone for comparison purpose. The 143-μM IAA CW extract recorded the best root induction and development. It was found that the root expression was faster (5 weeks) with the use of the novel method. In the conventional method, the canes are propagated by quick dip application of commercial product containing artificial hormone IAA and placing them on coir fiber dust beds. It takes up to 6 weeks for the canes to develop adventitious roots to the desired level. Steeping canes in 143-μM IAA CW extract improved rooting in D. purplecompacta L., and it was comparable to the application of 143-μM authentic IAA. The study indicates that adventitious root development, shoot development, and leaf emergence of D. purplecompacta L. is promoted by IAA CW extracts.  相似文献

Propagation through vegetative cuttings is a widely used technique that may bias estimates of genetic and environmental effects on plant growth. Leafy stem cuttings from 210 genotypes from eight populations of Salix pulchra were rooted and raised at three levels of nitrogen availability. Cuttings showed a complex suite of responses to vegetative propagation. Population and/or genotypic variation in response to vegetative propagation was observed in (1) retention of leaves during rooting, (2) date that cuttings resumed shoot growth after rooting, and (3) the frequency of cuttings that remained shoot dormant throughout the experiment. Nitrogen treatments also caused different responses to vegetative propagation, influencing date that cuttings resumed shoot growth and frequency of shoot dormancy. Because each of these responses had a direct effect on final plant size, I concluded that final size was a product of both differences among genotypes and treatments in plant growth rate, as well as genotype- and treatment-specific responses to vegetative propagation. This study shows that plant growth experiments can be designed to quantify responses to vegetative propagation and statistically remove these artifacts of propagation from estimates of genetic and environmental effects on plant growth.  相似文献

The relationship between shoot growth and rooting was examined in two, 'difficult-to root' amenity trees, Syringa vulgaris L. cv. Charles Joly and Corylus avellana L. cv. Aurea. A range of treatments reflecting severity of pruning was imposed on field-grown stock prior to bud break. To minimise variation due to the numbers of buds that developed under different treatments, bud number was restricted to 30 per plant. Leafy cuttings were harvested at different stages of the active growth phase of each species. With Syringa, rooting decreased with later harvests, but loss of rooting potential was delayed in cuttings collected from the most severe pruning treatment. Rooting potential was associated with the extent of post-excision shoot growth on the cutting but regression analyses indicated that this relationship could not entirely explain the loss of rooting with time, nor the effects due to pruning. Similarly, in Corylus rooting was promoted by severe pruning, but the relationship between apical growth on the cutting and rooting was weaker than in Syringa, and only at the last harvest did growth play a critical role in determining rooting. Another unusual factor of the last harvest of Corylus was a bimodal distribution of roots per cutting, with very few rooted cuttings having less than five roots. This implies that, for this harvest at least, the potential of an individual cutting to root is probably not limited by the number of potential rooting sites.  相似文献
