目的克隆并构建耐甲氧西林金黄色葡萄球菌(MRSA)青霉素结合蛋白2a(PBP2a)全长及转肽酶区的原核表达质粒。方法登录基因文库查找获得mecA基因的编码序列,应用PCR技术扩增获得DNA片段,将此基因片段插入PET-32a载体,同时酶切鉴定阳性克隆,DNA序列测定验证序列正确性。结果 PCR扩增获得了mecA基因全长及转肽酶区DNA片段,成功插入到原核表达载体PET32a,双酶切鉴定及DNA序列测定证实插入片段正确。结论成功构建了PBP2a全长及转肽酶区片段表达质粒,为该蛋白的纯化表达和疫苗研究奠定了基础。  相似文献

mecA基因PCR扩增法检测耐甲氧西林金黄色葡萄球菌   总被引:9,自引:2,他引:9  
目的 应用mecA基因PCR扩增法检测耐甲氧西林金黄色葡萄球菌(methicillin resistant staphylococcus aureus,MRSA)。方法 临床分离的70株金黄色葡萄球菌,应用mecA基因PCR扩增法鉴定MRSA,并与苯唑西林纸片扩散法进行比较。结果 70株金黄色葡萄球菌用PCR扩增法和纸片扩散法有6株鉴定有差异,4株。mecA基因阳性而纸片扩散法鉴定为敏感,1株mecA基因阳性纸片扩散法鉴定为临界耐药,1株mecA基因阴性却表现为苯唑西林耐药,2种方法符合率为91.43%。结论 mecA基因PCR扩增法可以准确、快速判定MRSA,特别是对隐匿型或低水平耐药菌株的检出有重要的价值。  相似文献

耐甲氧西林金黄色葡萄球菌(MRSA)的产生是由甲氧西林敏感的金黄色葡萄球菌(MSSA)获得外源性的SCCmec所致。MRSA菌株可以产生一种新的青霉素结合蛋白PBP2a,PBP2a降低了与β-内酰胺类抗生素的亲合力,从而对β-内酰胺类抗生素产生耐药性。PBP2a由mecA基因编码,mecA基因存在于葡萄球菌盒式染色体(Staphylococcal cassette chromosome mec,SCCmec)中,SCCmec是一种可移动的遗传元件,该元件还携带除mecA基因外的其他抗菌药物的耐药基因,造成多重耐药(Multidrug-resistance,MDR)。SCCmec目前主要分为8型,其中又分为若干亚型。SCCmec的基因型与MRSA的流行背景有关,不同地区的SCCmec基因分型分布可能不同。  相似文献

近年来,耐甲氧西林金黄色葡萄球菌在全世界各地感染率和分离率不断提高,已成为目前院内感染的重要病原菌之一。运用有效、可靠、廉价的分子分型方法对分析耐甲氧西林金黄色葡萄球菌的流行病学特征及来源,对制定控制院感及流行的措施非常重要的。本研究概述了各种分子分型方法的原理及比较,如SCCmec分型、脉冲场凝胶电泳分型、多位点序列分型、葡萄球菌A蛋白分型和毒力因子分型等。脉冲场凝胶电泳仍然是暴发流行中MRSA分子分型的金标准,而其他分型方法更适合用于检测菌株的变异和建立国际监测。  相似文献

耐甲氧西林葡萄球菌感染的调查与分析   总被引:7,自引:0,他引:7  
目的调查住院患者耐甲氧西林葡萄球菌(methicillin-resistant staphylococci,MRS)的感染情况,以便采取有效的防治措施。方法收集临床分离菌株,用mecA基因PCR扩增法鉴定MRS,并结合临床资料对本院MRS感染进行回顾性调查研究。结果住院患者标本中共收集到68株葡萄球菌,其中MRS 38株(阳性率为55.9%)。MRS感染主要多发于年龄>60岁,男性,合并多种疾病患者,科室分布以呼吸内科、泌尿外科及ICU病房为主。药敏结果显示MRS对万古霉素、利奈唑烷及喹努普汀的敏感率为100%,对替考拉宁敏感率为94.7%,对利福平的敏感率为57.9%,其余抗菌药物敏感率均<30%。结论及时了解本院患者MRS感染分布及耐药情况,有助于我们采取相应的监测及防治措施。  相似文献

了解我院患者耐甲氧西林金黄色葡萄球菌(MRSA)的分子流行病学特点,为临床抗感染治疗提供依据。收集2007年1月~2008年9月我院分离的耐甲氧西林金黄色葡萄球菌共54株,采用PCR进行SCCmec基因分型、葡萄球菌A蛋白(SPA)分型,并检测杀白细胞毒素(PVL)基因,同时应用脉冲场凝胶电泳(PFGE)进行同源性分析。54株MRSA菌株SCCmec基因分型为SCCmecⅡ型17株,SCCmecⅢ型33株,SCCmecⅣ型2株,SCCmecⅤ型2株;SPA基因分型将28株归属为t030,9株为t002,8株为t037,5株为t570,2株为t437,t163和t796各1株;PVL毒素检测只有2株SCCmecⅣ型菌株阳性;PFGE证实院内MRSA感染主要为2种克隆株传播,同时还有其他型别出现。本院MRSA流行传播的SCCmec基因型主要以Ⅲ型占优势,同时发现有携带PVL毒素的CA-MRSA分离株流行,应引起密切关注。  相似文献

耐甲氧西林金黄色葡萄球菌(MRSA)是引起医院感染的多重耐药菌,其有效的治疗药物为万古霉素。近年已发现对万古霉素耐受的金黄色葡萄球菌,金黄色葡萄球菌一旦对万古霉素耐药,临床将面临无药可供选择的局面。由于其所造成治疗上的困难,对其耐药机制的深入研究和该菌准确、及时的检出对于寻找新的治疗靶位和防止其播散有着极其重要的意义。本文就mecA耐药决定子、调节基因、染色体上的辅助基因对耐甲氧西林金黄色葡萄球菌耐药表达的影响及其表型和基因检测方法作一综述。  相似文献

摘要:目的 了解临床分离的耐甲氧西林金黄色葡萄球菌(MRSA)耐消毒剂基因携带状况及其对消毒剂抗性水平。方法 采用聚合酶链反应(PCR)法和体外抗菌试验方法进行实验室检测。结果 10株临床分离的MRSA中,检出4株携带qacA/B基因,检出率为40.0%。含氯消毒剂对4株qacA/B基因阳性MRSA的MIC值均高于标准菌株。戊二醛消毒剂对2株MRSA基因阳性MRSA的MIC值和1株MRSA基因阳性MRSA的MBC值高于标准菌株,其他均与标准株相同。结论 临床分离的MRSA qacA/B基因阳性率较高,携带qacA/B基因阳性的MRSA对含氯消毒剂有产生抗性的趋势。  相似文献

目的评价实时荧光PCR(RT-PCR)用于检测呼吸道感染耐甲氧西林金黄色葡萄球菌(MRSA)中的方法学特性,为临床快速诊断呼吸道MRSA定植或感染提供可靠循证依据。方法通过RT-PCR技术对痰液标本中金黄色葡萄球菌(SA)特异性核酸酶编码基因nuc和MRSA的特异性耐药基因mec A进行定量检测,并同时以细菌培养、鉴定和MRSA耐药表型确诊实验作为参考方法,评价其灵敏度、特异度、阴阳性预测能力;用10倍梯度稀释法配制已知标准菌株菌悬液评估RT-PCR的最低检出限。结果 RT-PCR法较细菌培养法对MRSA检测的总符合率为97.5%,检测SA的敏感度为97.2%、特异度为98.2%、阳性预测值为92.1%、阴性预测值为99.4%,检测甲氧西林耐药性的敏感度为100%、特异度为97.7%、阳性预测值为85.2%、阴性预测值为100%;RT-PCR对nuc基因和mec A基因的最低检出限均为103/m L。结论RT-PCR法与细菌培养法对于检测MRSA差异无统计学意义,且灵敏度优于后者,是用于临床快速排除性筛查呼吸道感染MRSA的较佳检测方法。  相似文献

Staphylococcus aureus is a highly successful human pathogen responsible for a wide range of infections. This study provides insights into the virulence, pathogenicity, and antimicrobial resistance determinants of methicillin‐susceptible and methicillin‐resistant S. aureus (MSSA; MRSA) recovered from non‐healthcare environments. Three environmental MSSA and three environmental MRSA are selected for proteomic profiling using isobaric tag for relative and absolute quantitation tandem mass spectrometry (iTRAQ MS/MS). Gene Ontology annotation and Kyoto Encyclopedia of Genes and Genomes pathway annotation are applied to interpret the functions of the proteins detected. 792 proteins are identified in MSSA and MRSA. Comparative analysis of MRSA and MSSA reveals that 8 of out 792 proteins are upregulated and 156 are downregulated. Proteins that have differences in abundance are predominantly involved in catalytic and binding activity. Among 164 differently abundant proteins, 29 are involved in pathogenesis, antimicrobial resistance, stress response, mismatch repair, and cell wall synthesis. Twenty‐two proteins associated with pathogenicity including SPA, SBI, CLFA, and DLT are upregulated in MRSA. Moreover, the upregulated pathogenic protein ENTC2 in MSSA is determined to be a super antigen, potentially capable of triggering toxic shock syndrome in the host. Enhanced pathogenicity, antimicrobial resistance, and stress response are observed in MRSA compared to MSSA.  相似文献

金黄色葡萄球菌femB基因与甲氧西林高水平耐药密切相关,可能成为开发抗MRSA药物的新靶位.以金葡菌基因组DNA为模板,PCR扩增femB全长基因,所得片段与pGM-T载体连接并转化感受态大肠杆菌DH5α,阳性克隆以PCR、双酶切及测序鉴定.将鉴定正确的目的片段定向克隆到pGEX-4T-1表达载体中,转化至大肠杆菌BL21后经IPTG诱导表达GST/FemB融合蛋白;采用SDS-PAGE及Western blot对融合蛋白进行鉴定.结果显示,重组质粒在宿主菌中获得了高效表达,融合蛋白相对分子质量为75 kD,该融合蛋白可与抗GST-tag抗体特异结合;表明femB基因的原核表达系统构建成功,为进一步研究其生物学功能奠定了基础.  相似文献

Totarol is a diterpene compound extracted from the totara tree. Totarol and eight other diterpenes were found to potentiate methicillin, one reducing the minimum inhibitory concentration of methicillin against resistant Staphylococcus aureus 256-fold. Totarol did not inhibit the synthesis of DNA or peptidoglycan in S. aureus, but reduced the respiration rate by 70%. Under potentiation conditions, diterpenes had only a slight effect on the respiration rate, but had a significant effect on expression of PBP 2a. We conclude that the primary staphylococcal target for totarol is the respiratory chain, but that potentiation of methicillin by diterpenes is by interference with PBP 2a expression.  相似文献

Aims: Methicillin‐resistant Staphylococcus aureus (MRSA) ST398 has recently been described as a zoonotic agent. Its transmission between animals seems to be a pivotal factor in its emergence and dissemination. This experimental trial was performed to describe MRSA ST398 contamination and transmission in pigs after a low dose inoculation. Methods and results: Twelve specific pathogen‐free (SPF) pigs were randomly divided between two separate pens. Three pigs in each pen received a nasal inoculation of 2 × 104 colony‐forming units per animal, and three naïve pigs were left in contact with them. Every 2 days and at necropsy, different samples were screened for MRSA. It was detected in nasal swabs from five inoculated and three naïve contact pigs, as early as 1 day after inoculation. MRSA was also found in environmental wipes but never in faecal samples. At necropsy, MRSA was detected in the lymph nodes of two contact pigs and in the tonsils and lymph nodes of three inoculated pigs. Twelve other SPF pigs were included as negative control in a separate room. Conclusion: This experiment showed that inoculation of a low dose of MRSA ST398 could lead to the horizontal transmission of the bacterium between pigs, the contamination of mandibular lymph nodes and the contamination of the environment without faecal carriage. Significance and Impact of the Study: The minimal inoculated dose via nasal route to observe transmission of MRSA ST398 between pigs is equal or lower to 2 × 104 colony‐forming units per animal, and faecal excretion seems not to be a necessary condition for horizontal transmission.  相似文献

The present study evaluated the antibacterial activity and the synergy of the sanguisorbigenin (SGB) from the dried root of Sanguisorba officinalis L. combined with β-lactam antibiotics against methicillin-resistant Staphylococcus aureus. A total of six strains of reference strain and clinical isolates were used to determine the antibacterial activity using a broth microdilution assay, and the synergistic effects were determined using a checkerboard assay. To analyse the mechanism of synergy, we conducted the level of penicillin-binding protein 2a by western blot. In addition, quantitative RT-PCR was performed to analyse the mecA gene expression. The minimal inhibitory concentration values of SGB against six strains of S. aureus were in the range of 12·5–50 μg ml−1, and there were synergy, or partial synergy effects when SGB was combined with antibiotics. Furthermore, when treated with SGB, the level of penicillin-binding protein 2a and the expression of the mecA gene was reduced significantly. In conclusion, this study demonstrated that SGB is a potential natural antibacterial agent against methicillin-resistant S. aureus that represents a considerable burden on the healthcare system worldwide, and may an exceptionally modulator of β-lactam antibiotics.  相似文献
