Summary In a 35-year-old calcareous beech forest stand five beech trees (Fagus sylvatica L.) were felled every 2 weeks, and xylem sap was obtained by means of water displacement from the lowest trunk sections, each 100 cm in length. From mid-October 1988 to mid-October 1989 a total of 130 trees were investigated. The seasonal variations of the Ca, Mg, K and Mn contents, as well as those of pH, show four characteristic phases. Additionally, distribution of the mineral contents along the trunk was studied in four trees. The seasonal increase and decrease of xylem sap mineral contents along the trunk is shown for the characteristic phenophases. The Ca, Mg, K, and Mn contents of xylem saps were determined by means of atomicabsorptionspectrophotometry.  相似文献

Four-year-old citrus trees ( Citrus unshiu Marcovitch) were fed via the roots with (15NH4)2sO4 or K15NO3 as a nitrogen source. Nitrogenous compounds and their isotopic abundances in fine roots and xylem sap from trunks were assayed in order to obtain information on the species of nitrogen released by the root system into the ascending xyiem stream.
Arginine, asparagine, nitrate and proline in xylem sap accounted for 48, 21, 13 and 10%, respectively, of the total nitrogenous constituents tested in the sap. However, in the trees fed with labelled ammonium the main nitrogenous compound labelled with 15N in the xylem sap was asparagine and glutamine, which accounted for 79% and 18%, respectively, of total labelled nitrogen. In the xylem sap of trees fed with labelled nitrate, nitrate accounted for 94% of total labelled nitrogen. Nitrate and asparagine followed by glutamine showed the highest ratios of isotopic abundance in xylem sap as compared to fine roots. Proline and arginine had much lower ratios. These results indicate that nitrate, asparagine and glutamine are the main nitrogenous compounds released by the roots to the xylem stream, whereas arginine and proline are released into the xylern vessels by the trunk tissues. Furthermore, nitrate and asparagine are probably in steady movement upward in the trunk xylem, whereas glutamine is more easily taken up by the trunk tissues than nitrate and asparagine.  相似文献

Xylem sap was extracted from cut branches of stone-fruit and pip-fruit trees at different times of the year, and analysed for nitrogen and carbohydrate content. Growth of Stereum purpureum was measured in the different sap samples in vitro. Nitrogen and carbohydrate levels in xylem sap from peach trees were highest in late winter and early spring, and these samples supported most growth of S. purpureum. At full bloom the levels of nitrogen and carbohydrate in xylem sap from stone-fruits (peach, nectarine, plum, cherry) were several times greater than in sap from pip-fruits (apple, pear), and the growth of S. purpureum in sap from stone-fruits was approximately ten times that in sap from pip-fruits. The results suggest that seasonal variations in susceptibility of stone-fruits to silver-leaf disease, and the greater susceptibility of stone-fruits than of pip-fruits, are due to differential ability of their xylem saps to support growth of S. purpureum.  相似文献

Xylem sap composition of spruce is influenced by several factors, such as the sampled organ, the sampling period, the availability of soil nutrients, and the soil water potential. Based on literature data and ongoing investigations carried out with adult trees, we present an overview on the main factors influencing xylem sap concentrations of Norway spruce. Direct measurements of nutrient fluxes in the xylem sap are then used to suggest a general scheme of mineral element cycling within adult trees. In Norway spruce (Picea abies Karst.), nutrient concentration in the xylem sap was higher in twigs and fine roots compared to the bottom of the trunk, the highest concentrations beeing observed in spring during the shoot elongation. Xylem sap concentrations were higher in spruce growing at nutrient rich sites than at poor sites. The combination of twig and trunk xylem sap analysis, together with xylem flow measurements in the trunk during the course of a vegetation period allowed the quantification of mineral fluxes via xylem sap flow in the trunk and twigs. These results were compared to gross mineral uptake measurements at the same site. Ca flux in the trunk xylem sap was lower than the gross uptake of Ca. Mg flux in trunk sap was approximately equivalent to Mg gross uptake whereas P and K fluxes in trunk sap were much higher than the gross uptake. Fluxes of Ca, Mg, K and P in the twig sap were much higher than that in trunk sap. Data suggest that internal cycling is responsible for a large part of the nutrient fluxes in the xylem sap of the crown. Xylem sap composition thus appears to be a tool which can complement other sources of information on mineral uptake and cycling in adult spruce  相似文献

Summary Nitrate reductase (EC 1.6.6.2) activity (NRA), as measured by an in vivo assay, is present in needle leaves and mycorrhizal fine root tips of adult Norway spruce [Picea abies (L.) Karst.] in at least equal amounts on a fresh weight basis, in both adult and 5-year-old trees. NRA could also be demonstrated in trunk wood of deroted trees after fertilization with 5 mM , exhibiting a longitudinal profile in the trunk. Inducibility in needles can more efficiently be achieved by NO2 (100 g·m-3) than by 5 mM nitrate, which is effective only in root-amputated trees. A remarkably high level of needle-NRA in unfertilized trees, which are characterized by a very low level of nitrate in the xylem sap, suggests that NRA in spruce needles may in part be constitutive. Organic-N is a major nitrogen source for the needles even in root-amputated trees, indicating pronounced exchange processes between ray parenchyma and trunk xylem, which in turn are modified by the nitrogen source fed to the trunk stump. Intact trees exhibit a very similar amino acid composition of the xylem sap, regardless of whether or has been fed. The amino acid pattern of the needles is not thrown out of balance by flooding with and , which occurs in fertilized derooted trees. This indicates a distinct potential for homoeostasis of nitrogen entrance-metabolism (i.e. NRA and glutamine synthetase activity) in the needles. In the ectomycorrhiza/fine root-system (EMC), marked differences in NRA were observed depending on root-tip diameter and along the longitudinal profile of the fine roots. EMC-nitrate reductase is strongly enhanced by . Needle-NRA exhibits a circannual rhythm. An early summer maximum is followed by a December minimum. This activity pattern matches well the transitory increase of soluble nitrogen in spring and the total protein maximum in winter. In an indirect way assimilatory NRA may well contribute to nitrogen overfertilization (by consumption of NOX) as one possible cause of the contemporary decline of spruce populations.  相似文献

The principal forms of amino nitrogen transported in xylem were studied in nodulated and non-nodulated peanut (Arachis hypogaea L.). In symbiotic plants, asparagine and the nonprotein amino acid, 4-methyleneglutamine, were identified as the major components of xylem exudate collected from root systems decapitated below the lowest nodule or above the nodulated zone. Sap bleeding from detached nodules carried 80% of its nitrogen as asparagine and less than 1% as 4-methyleneglutamine. Pulse-feeding nodulated roots with 15N2 gas showed asparagine to be the principal nitrogen product exported from N2-fixing nodules. Maintaining root systems in an N2-deficient (argon:oxygen, 80:20, v/v) atmosphere for 3 days greatly depleted asparagine levels in nodules. 4-Methyleneglutamine represented 73% of the total amino nitrogen in the xylem sap of non-nodulated plants grown on nitrogen-free nutrients, but relative levels of this compound decreased and asparagine increased when nitrate was supplied. The presence of 4-methyleneglutamine in xylem exudate did not appear to be associated with either N2 fixation or nitrate assimilation, and an origin from cotyledon nitrogen was suggested from study of changes in amount of the compound in tissue amino acid pools and in root bleeding xylem sap following germination. Changes in xylem sap composition were studied in nodulated plants receiving a range of levels of 15N-nitrate, and a 15N dilution technique was used to determine the proportions of accumulated plant nitrogen derived from N2 or fed nitrate. The abundance of asparagine in xylem sap and the ratio of asparagine:nitrate fell, while the ratio of nitrate:total amino acid rose as plants derived less of their organic nitrogen from N2. Assays based on xylem sap composition are suggested as a means of determining the relative extents to which N2 and nitrate are being used in peanuts.  相似文献

Summary The relative hydraulic conductivity (k) of xylem and resistance (R) to water flow through trunk, primary roots and branches in Picea abies trees growing under contrasting light conditions were investigated. The xylem permeability to water was measured by forcing 10 mM water solution of KC1 through excised wood specimens. From the values of k, the sapwood transverse area and the length of conducting segments, R of the whole trunk, branches and roots was calculated. The relative conductivity of xylem in open-grown trees exceeded that of shade-grown trees by 1.4–3.1 times, while k was closely correlated with the hydraulically effective radius (R e) of the largest tracheids (R 2 was 0.85–0.94 for open- and 0.51–0.79 for shade-grown trees). Because of both a low k and a smaller sapwood area in shade-grown trees the resistance to water movement through their trunk, roots and branches was many times higher. The distribution of R between single segments of the water-conducting pathway differed considerably in trees from different sites. At high water status the largest share of the total resistance in open- as well as shade-grown trees resides in the apical part of the trunk. The contribution of the branches to total xylem resistance is supposed to increase with developing water deficit.  相似文献

Summary The nitrogenous compounds in the xylem (bleeding) sap of lines of field-grownPhaseolus vulgaris L., known to vary in N2 fixation and yield, were measured during growth with and without N fertilizer. Forty nine-67% of the total sap N was in the form of nitrate in fertilized plants, with low amounts of the ureides, allantoin and allantoic acid (4–12%). Ureides contributed between 17 and 38% to the total sap N of non-fertilized plants, with nitrate generally comprising less than 40%. Among the nine lines grown without fertilizer there were significant differences in the % of the sap N as ureides and also in the total mol N ml–1 sap.The ASN/GLN ratio (mol/mol) was greater in sap collected from the two parental lines given fertilizer compared with non-fertilized plants. However the actual ratio varied between the parental lines. Together the two amides generally comprised between 43–62% of the amino-N in sap samples from the two parental lines with or without N fertilizer.For each N treatment (fertilized or non-fertilized) there were no obvious differences in sap composition between the high N2 fixing lines and the low N2 fixing lines. However there was generally a positive relationship between the rate of N translocation (total N concn. ml–1 sap x rate of exudation) and the ranking of the lines on the basis of higher N2 fixation rates (acetylene reduction), which was to a large extent independent of the source of N available to the plant.  相似文献

Removal of fruits and treatment with SADH (succinic acid 2,2-dimethylhydrazide) were used to change the balance between sources and sinks for photosynthates in Malus domestica‘Golden Delicious’. Sorbitol and sugar content were measured in bleeding sap, in xylem sap prepared by suction, and in 80% methanol extracts of the roots. Concentration as well as total amount of sorbitol in bleeding sap sampled in July and September were lower in fruiting than in defruited trees, and so was the total amount of sorbitol in xylem sap from the trunk. SADH treatment tended to reduce the sorbitol content. Sorbitol in root extracts, expressed as percentage of methanol (80%) insoluble root dry matter, was highest in fruiting trees; but root dry matter was here less than half of that in defruited trees. Sorbitol content in xylem sap as well as sorbitol + sugar percentage of roots showed a distinct maximum in late winter followed by a heavy decrease during spring. It is suggested that sorbitol in xylem sap during the growth season represents a return transport from the roots, and that the level of sorbitol in this return transport reflects, to a certain degree, the ratio between leaf area and assimilate demand by the tree.  相似文献

Xylem sap of sinker (tap) root, cluster feeding roots, lateral roots and from an age series of main stem extensions of 6-year trees of Banksia prionotes was collected and analyzed for principal organic and inorganic solutes. During the phase of root uptake activity in winter and spring, cluster roots were principal xylem donors of malate, phosphate, chloride, sodium, potassium and amino acid N whereas other parts of the root served as major sources to the shoot of other cations, nitrate and sulphate. Sinker root xylem sap was at all times less concentrated in solutes than that of lateral roots into which cluster roots were voiding exported solutes. Phosphate was abstracted from xylem by stem tissue during winter and it and a range of other solutes released back to xylem immediately prior to extension growth of the shoot in summer. Phloem sap collected from mid regions of stems was unusually low in potassium and phosphate relative to chloride and sulphate in comparison with phloem sap of other species, and its low potassium: sodium ratio relative to xylem indicated poor discrimination against sodium during phloem loading. Data are discussed in relation to the asynchronous seasonal cycles of nutrient uptake and shoot growth.  相似文献

Dwarf french beans, Phaseolus vulgaris L., were grown with or without inoculation with rhizobia (strain 3644), and with or without a combined nitrogen source (nitrate or ammonium ions). The distribution of radioactivity into products of dark 14CO2 assimilation was studied in roots or nodules from these plants. A detailed study was also made of the distribution and rates of excretion of nitrogen in xylem bleeding sap in 28 day old plants grown on the various sources of nitrogen. Whereas detached nodules accumulated radioactive glycine, serine and glutamate when incubated with 14CO2, bleeding sap from plants root fed 14CO2 contained low levels of radioactivity in these compounds but higher levels in allantoin. Chemical analysis showed allantoin to be the major compound transported in the xylem of nodulated plants, whether or not they were fed on combined nitrogen. Experiments were conducted to characterize the distribution of N compounds in the xylem sap of nodulated and nonnodulated soybean plants through development and to determine the effects of exogenous N on the distribution of N compounds in the xylem. Xylem sap was collected from nodulated and nonnodulated greenhouse-grown soybean plants (Glycine max [L.] Merr. “Ransom”) from the vegetative phase to the pod-filling phase. The sum of the nitrogen in the amino acid, nitrate, ureide (allantoic acid and allantoin), and ammonium fractions of the sap from both types of plants agreed closely with total N as assayed by a Kjeldahl technique. Sap from nodulated plants supplied with N-free nutrient solution contained seasonal averages of 78 and 20% of the total N as ureide-N and amino acid-N, respectively. Sap from nonnodulated plants supplied with a 20 millimolar KNO3 nutrient solution contained seasonal averages of 6, 36, and 58% of total N as ureide-N, amino acid-N, and nitrate-N, respectively. Allantoic acid was the predominant ureide in the xylem sap and asparagine was the predominant amino acid. When well nodulated plants were supplied with 20 millimolar KNO3, beginning at 65 days, C2H2 reduction (N2 fixation) decreased relative to nontreated plants and there was a concomitant decrease in the ureide content of the sap. A positive correlation (r = 0.89) was found between the ureide levels in xylem sap and nodule dry weights when either exogenous nitrate-N or urea-N was supplied at 10 and 20 millimolar concentrations to inoculated plants. The results demonstrate that ureides play a dominant role in N transport in nodulated soybeans and that the synthesis of ureides is largely dependent upon nodulation and N2 fixation.  相似文献

The use of the relative ureide content of xylem sap [(ureide-N/total N) × 100] as an indicator of N2 fixation in soybeans (Merr.) was examined under greenhouse conditions. Acetylene treatments to inhibit N2 fixation were imposed upon the root systems of plants totally dependent upon N2 fixation as their source of N and of plants dependent upon both N2 fixation and uptake of exogenous nitrate. Significant decreases in the total N concentration of xylem sap from plants of the former type were observed, but no significant decrease was observed in the total N concentration of sap from the latter type of plants. In both types of plants, acetylene treatment caused significant decreases in the relative ureide content of xylem sap. The results provided further support for a link between the presence of ureides in the xylem and the occurrence of N2 fixation in soybeans. The relative ureide content of xylem sap from plants totally dependent upon N2 fixation was shown to be insensitive to changes in the exudation rate and total N concentration of xylem sap brought about by diurnal changes in environmental factors. There was little evidence of soybean cultivars or nodulating strains affecting the relative ureide content of xylem sap. `Ransom' soybeans nodulated with Rhizobium japonicum strain USDA 110 were grown under conditions to obtain plants exhibiting a wide range of dependency upon N2 fixation. The relative ureide content of xylem sap was shown to indicate reliably the N2 fixation of these plants during vegetative growth using a 15N method to measure N2 fixation activity. The use of the relative ureide content of xylem sap for quantification of N2 fixation in soybeans should be evaluated further.  相似文献

Spontaneous bleeding of sugar-rich sap from cambial-deep incisions in the bark of trunks was demonstrated for Eucalyptus globulus and other eucalypts across a range of localities and seasonal conditions in south-west Australia. High levels of sucrose and raffinose (up to 31% w/v total sugars) were present in the exudates, and upward and downward gradients in exudate sugar concentrations were recorded between samples obtained at different heights up trunks of E. globulus. The data indicated a phloem origin for the exudates, with source:sink pressure gradients driving translocation. Concentration ratios of sugars to amino acids were consistently lower in exudate from upper (distal) than basal regions of trunks, suggesting preferential partitioning of nitrogen upwards towards the trunk apex. A comparison of phloem and xylem sap composition from one plantation over a season showed nitrate in xylem but not phloem and substantial amounts of sodium, and high concentrations of chloride and sulphate relative to phosphate in xylem and phloem. Phloem sap sampled across a range of 29 contrasting plantations of E. globulus at peak stress (autumn) showed great inter-site variability in concentrations of amino acids, sulphur, sodium and certain trace elements and in C:N and Na:K ratios of sap. Carbon isotope ratios (δ13C) were strongly correlated with sugar concentrations of the sap samples from these and other plantations. Use of sap compositional attributes of phloem and δ13C values of translocated carbon is suggested for assessing the current nutritional condition and water status of E. globulus plantings. Nodulation, nitrogen (N2) fixation and xylem sap composition were examined in sand cultured plants of Bambara groundnut (Vigna subterranea L.) and Kersting's bean (Macrotyloma geocarpum L.) inoculated with Bradyrhizobium strain CB756 and supplied via the roots for a 4 week period from the third week onwards with different levels of (15N)-nitrate (0–15 mM). The separate contributions of nitrate and N2 to plant nitrogen were measured by isotope dilution. Increasing levels of nitrate inhibited nodule growth (measured as dry matter or nodule N) of both species parallel with decreased dependence on symbiotically-fixed N. Specific nodule activity (N2 fixed g nodule dry−1 d−1 of nodules) was reduced progressively with time in V. subterranea at higher (5 or 15 mM) levels of NO3, but this was not so for M. geocarpum. Root xylem bleeding sap of both species showed ureides (allantoin and allantoic acid) as predominant (>90%) solutes of nitrogen when plants were relying solely on atmospheric N. Levels of ureide and glutamine decreased and those of asparagine and nitrate in xylem increased with increasing level of applied nitrate. Relative levels of xylem ureide-N were positively correlated (R2=0.842 for M. geocarpum and 0.556 for V. subterranea), and the ratio of asparagine to glutamine in xylem exudate negatively correlated (R2=0.955 for M. geocarpum and 0.736 for V. subterranea) with plant reliance on nitrogen fixation. The data indicate that xylem sap analyses might be useful for indirect field assays of nitrogen fixation by the species and that Kersting's bean might offer some potential as a symbiosis in which N2 fixation is relatively tolerant of soil N.  相似文献

It is well established that nitrate is a potent inhibitor of nodulation and nitrogen fixation in legumes. The objective of this study was to demonstrate the relative insensitivity of these processes to nitrate with Calopogonium mucunoides, a tropical South American perennial legume, native to the cerrado (savannah) region. It was found that nodule number was reduced by about half in the presence of high levels of nitrate (15 mM) but nodule growth (total nodule mass per plant) and nitrogen fixation (acetylene reduction activity and xylem sap ureide levels) were not affected. Other sources of N (ammonium and urea) were also without effect at these concentrations. At even higher concentrations (30 mM), nitrate did promote significant inhibition (ca. 50%) of acetylene reduction activity, but no significant reduction in xylem sap ureides was found. The extraordinary insensitivity of nodulation and N2 fixation of C. mucunoides to nitrate suggests that this species should be useful in studies aimed at elucidating the mechanisms of nitrate inhibition of these processes.  相似文献

Summary Nitrate reductase activity (NRA), nitrate content and biomass components of leaflets, leaf stalks, old stem, current-year stem and roots of ash trees (Fraxinus excelsior L.) growing in their natural habitats were investigated. In addition, NRA, total nitrogen and nitrate concentration were analyzed in the leaves and roots of ash trees from four different field sites. The highest NRA per gram biomass and also per total compartment biomass was found in the leaflets, even though root biomass was much higher than total leaflet biomass. The highest nitrate concentrations were found in the leaf stalks. Correlations between nitrate availability in the soil and NRA in leaves were not significant due to high variability of the actual soil nitrate concentrations. The seasonal variation in foliar NRA, nitrate concentration and total nitrogen concentration is much smaller in F. excelsior than reported for herbaceous species and is mainly caused by changes in the actual soil nitrate availability and by senescence of the leaves.  相似文献

Nitrate assimilation was examined in two cultivars (Banner Winterand Herz Freya) of Vicia faba L. supplied with a range of nitrateconcentrations. The distribution between root and shoot wasassessed. The cultivars showed responses to increased applied nitrateconcentration. Total plant dry weight and carbon content remainedconstant while shoot: root dry weight ratio, total plant nitrogen,total plant leaf area and specific leaf area (SLA) all increased.The proportion of total plant nitrate and nitrate reductase(NR) activity found in the shoot of both cultivars increasedwith applied nitrate concentrations as did NO3: Kjeldahl-Nratios of xylem sap. The cultivars differed in that a greaterproportion of total plant NR activity occurred in the shootof cv. Herz Freya at all applied nitrate concentrations, andits xylem sap NO3: Kjeldahl-N ratio and SLA were consistentlygreater. It is concluded that the distribution of nitrate assimilationbetween root and shoot of V. faba varies both with cultivarand with external nitrate concentration. Seasonal variation was followed in the content of total a-aminoacids, arginine, calcium, total carbohydrate, ß-galactosidaseactivity, magnesium, nitrate, phosphatase activity, phosphateand sulphate in vacuum-extracted xylem sap from Actinidia chinensisvar. hispida, the Chinese gooseberry or kiwifruit. There wasa marked increase in the concentration of most sap componentsjust prior to leaf emergence followed by a rapid decrease afterthe leaves had expanded. Experiments with excised extensionshoots showed that much of this spring-time increase in concentrationsof sap components was due to mobilization of nutrients withinthe shoot itself. Sap from the trunk and the older brancheschanged less in composition than did sap from the younger partsof the plant. The amplitude and direction of trends in concentrationof sap from the different parts of the plant varied with nutrientand with time. Analysis of vacuum-extracted xylem sap collectedduring periods of rapid transpiration from early summer onwardsgives a reliable indication of the composition of the transpirationstream. A novel technique for the physico‐chemical analysis of xylem sap by underwater access to the sapwood of trees is described. In situ measurements of dissolved oxygen in the sapwood are performed by combining this technique with a novel optical method for oxygen detection. In early spring, the oxygen concentration of the sapwood of Betula pendula was in the range of 80–230 µmol O2 L?1, corresponding to an oxygen deficit of 40–75% of air saturation. Oxygen concentration maxima and minima occurred early in the morning and in the afternoon, respectively, whereas xylem sap temperatures showed the reverse pattern. In the sapwood, hypoxia increased from the beginning of bud break until frondescence, when a deficit of 86% of air saturation marked the upper limit of oxygen depletion. There seemed to be no relationship between daily variations of oxygen concentration and xylem sap pressure. In summer, sap flow was a major determinant for the diurnal variation of dissolved oxygen concentration. Oxygen supply to the sapwood was determined by both radial influx into the trunk through intercellular gas spaces and transport of dissolved oxygen via xylem sap flow. Radial influx seemed to be favoured during night‐time, when the trunk was warmer than ambient air. During daytime, the hypoxia of the sapwood rose and increased sharply in the evening, when sap flow velocity approximated zero. High temperature in the sapwood enhanced the respiratory oxygen consumption of the wood parenchyma while the supply of dissolved oxygen via the transpiration stream became ineffective.  相似文献
