Although the statutory goals for chemical regulation are consistent with the precau tionary principle, the current U.S. regulatory program governing synthetic chemicals generally adopts little precautionary controls for the manufacture of most chemicals. For the vast majority of chemicals in use, current law places the burden of producing scientific evidence on the regulatory agency, which actually may serve to discourage companies from testing the safety of their chemicals, since the results could then be used against them in regulatory proceedings. By contrast, for a small subset of chemicals — new chemicals that belong to suspect categories — regulatory controls are quite precautionary. The result of this schizophrenic approach to chemical regulation is a regulatory system that is characterized by the absence of preventative regulation for most existing chemicals, an inequitable barrier to entry for newer safer chemicals, and a lack of information upon which to understand the safety of most chemicals in the U.S. Informal reforms of the current regulatory program are already underway to provide a more consistent and precautionary approach to chemical regulation, although to fully advance the dual goals of regulatory consistency and precaution in the regulation of chemicals, legislative action is necessary. 相似文献

The effect of 3 insecticides (chlordane, dieldrin and carbaryl) and one herbicide (2,4-D-fluid) was studied on the induction of ouabain-resistant mutants in Chinese hamster V79 cells at concentrations approaching the Environmental Protection Agency (EPA) tolerance limits. The kinetics and dose range for cytotoxicity were determined by colony formation assay. Results showed that these compounds enhanced the number of ouabain-resistant mutants and acted as weak mutagens. 相似文献

Using probabilistic analysis may be very useful for risk management in developing countries, where information, resources, and technical expertise are often scarce. Currently, most regulatory agencies recommend using deterministic approaches for the analysis of problems relating to decision-making. However, this approach does not incorporate uncertainty in the variables, nor the propagation of uncertainty through the different processes in which they are involved. The complexity of the problem is therefore arbitrarily reduced, and valuable information that could be useful for proposing realistic policies is not considered. This article compares the results of a deterministic analysis with those of a probabilistic one for regulating arsenic in Chile, and differences are established for public policy as a result of building uncertainty into the analysis. It is concluded that the use of a deterministic approach can lead to higher risks than necessary and that probabilistic results can help the regulator negotiate stricter standards. Alternatively, the regulator may end up imposing much higher costs to sources than originally expected as these will be forced to use expensive technology to comply consistently with a given standard. 相似文献

Regulation of nuclear receptor activities by two human papillomavirus type 18 oncoproteins,E6 and E7
The human papillomavirus (HPV) E6 and E7 oncoproteins are two major proteins that remain expressing in HPV-associated human cancers. The high-risk HPVs synthesize E6 and E7 oncoproteins to alter the function of cellular regulatory proteins, such as p53 and retinoblastoma gene product, respectively. In this study, we demonstrated that HPV-18 E6 and E7 proteins were able to directly interact with some nuclear receptors (NRs), such as thyroid receptor, androgen receptor, and estrogen receptor (ER), whether or not appropriate hormones were present. The functional roles of these two oncoproteins in NRs depended on the cell type (including ligand), promoter context, and NR type. These two oncoproteins regulated ER functions through ER's AF-1, AF-2, or both. Hence, our results provide new insights into the mechanisms controlling the proliferation and immortalization of HPV infected cells by these two oncoproteins mediating through their regulatory functions in NR systems. 相似文献

Rabbit zygotes and embryos were exposed to hypertonic sucrose in phosphate-buffered saline (SPBS). In experiment one, 144 zygotes shrank to 32-36% of their initial volume in 1.0 M SPBS within 30 min. Neither hypertonic treatment with 0.5 M or 1.0 M SPBS nor micropuncture of the zona pellucida after shrinkage affected embryo development into blastocysts in vitro (88%, 83%, and 82%, respectively), compared to that of the controls (93%, P greater than .05). In experiment two, 252 two- to four-cell- and 177 morula-stage embryos were exposed to isotonic PBS control or 0.5 M, 1.0 M, or 1.5 M SPBS for 30, 60, 90, 120, and 150 min before transfer to PBS (290 mOsm). Embryo development was significantly reduced (P less than .05) when embryos were exposed in 0.5 M and 1.0 M SPBS for more than 60 min or in 1.5 M SPBS for more than 30 min. In experiment 3, morulae exposed for 60 min to 0.5 M or 1.0 M SPBS shrank to 37-39% or 32-35% of their initial volume and then expanded to 87-94% or 81-90% of their initial volume, respectively, after being returned to isotonic PBS for 60 min, but embryos in 1.5 M SPBS had erratic osmotic behavior. In experiment four, 192 two- to four-cell embryos exposed to 0.5 M SPBS for 0, 30, and 60 min before transfer to oviducts of recipients resulted in the production of 39%, 42% and 31% young, respectively (P greater than .05).(ABSTRACT TRUNCATED AT 250 WORDS) 相似文献

Phospholemman (PLM) is a 72-amino-acid phosphoprotein that is a major substrate for cAMP-dependent protein kinase, protein kinase C, and NIMA kinase. In lipid bilayers, PLM forms ion channels selective for Cl-, K+, and taurine. Effluxes of these abundant intracellular osmolytes play an important role in the control of dynamic cell volume changes in many cell types. We measured swelling-activated ion currents and regulatory volume decrease (RVD) in human embryonic kidney cells stably overexpressing canine cardiac PLM. In response to swelling, two clonal cell lines overexpressing PLM had increased swelling-activated ion current densities and faster and more extensive RVD. A third clonal cell line overexpressing mutant PLM showed reduced ion current densities and a diminished RVD response. These results suggest a role for PLM in the regulation of cell volume, perhaps as a modulator of an endogenous swelling-activated signal transduction pathway or possibly by participating directly in swelling-induced osmolyte efflux. 相似文献

Neotropical floodplain lakes provide an excellent opportunity to examine the regulation and stability of fish assemblages. At low water, when lakes are separated, fish are concentrated in the lakes and are presumably subject to strong interspecific interactions that can shape assemblage structure. At high water, when the lakes and river channels become broadly interconnected, ample potential exists for alteration of assemblage structure because eggs, larvae, and older fish may undergo spatial reshuffling among waterbodies. Twenty lakes of the Orinoco River floodplain in Venezuela were surveyed in the early and late dry seasons of two consecutive years. Marked differences in assemblage structure among lakes were established in the early dry season. Changes in assemblage structure during the dry season were large and detectable even at the ordinal level; they reflected a strong reduction of visually oriented fish relative to fish with adaptations to low light. Changes were similar in the two years and were apparently due to species-specific differences in mortality, which was high over the dry season. The annual flood is a strong natural fluctuation that greatly modifies assemblage properties during the wet season. Nevertheless, the potential for alteration of assemblage structure by reshuffling was not realized: assemblage properties early and late in the dry season were similar in the two years, indicating a regular and predictable annual cycle of change in assemblage structure. This regularity can be explained by mechanisms of regulation which appear to be linked to piscivory and the optical environment. In contrast with the prevailing views on neotropical fish assemblages, there seems to be a strong deterministic component to assemblage structure and dynamics in Orinoco floodplain lakes. 相似文献

Classic plant breeding has increased the beauty and utility of ornamental plants, but biotechnology can offer completely new traits for plants used in homes and gardens. The creation of blue petal color in carnations and roses are examples where biotechnology has created novelty that conventional hybridization cannot match. However, all innovations have benefits and risks, and future commercialization of transgenic ornamental plants raises complex questions about potential negative impacts to managed landscapes and natural ecosystems. Predictive ecological risk assessment is a process that uses current knowledge to estimate future environmental harms or benefits arising from direct or indirect exposure to a genetically-modified (GM) plant, its genes, or gene products. This article considers GM ornamental plants in the context of current ecological risk assessment principles, research results, and current regulatory frameworks. The use of ecological risk assessment by government agencies to support decision-making is reviewed in the context of ornamental plants. Government risk assessments have usually emphasized the potential for pollen-mediated gene flow, weediness in managed areas, invasion of natural areas, and direct harm to nontarget organisms. Some of the major challenges for predictive risk assessment include characterizing gene flow over time and space, plant fitness in changing environments, and impacts to nontarget organisms, communities and ecosystems. The lack of baseline information about the ecology and biodiversity of urban areas, gardens, and natural ecosystems limits the ability to predict potential hazards, identify exposure pathways, and design hypothesis-driven research. The legacy of introduced ornamental plants as invasive species generates special concern about future invasions, especially for GM plants that exhibit increased stress tolerance or adaptability. While ecological risk assessments are a valuable tool and have helped harmonize regulation of GM plants, they do not define the acceptable level of risk or uncertainty. An in vitro receptor binding assay, using filtration to separate bound from free [125I]insulin, was developed and used to characterize insulin receptors on membranes isolated from specific areas of rat brain. The kinetic and equilibrium binding properties of central receptors were similar to those of hepatic receptors. The binding profiles in all tissues were complex and were consistent with binding in multiple steps or to multiple sites. Similar binding properties were found among receptors in olfactory tubercle/bulb, cerebral cortex, hippocampus, striatum, hypothalamus, and cerebellum. High affinity [125I]insulin binding sites (KD = 3-11 nM) were distributed evenly between membranes isolated from P1 and P2 fractions of these brain areas, with the exception of the olfactory tubercle in which binding to P2 membranes was four-fold greater (Bmax = 150 fmol/mg protein). One difference between insulin receptors in brain and peripheral target tissues, however, was observed. Following exposure to 0.17 microM insulin for 3 h at 37 degrees C, the number of specific [125I]insulin binding sites on adipocytes decreased by 40%, while the number of binding sites on minces of cerebral cortex/olfactory tubercle remained constant. The results suggest that although the binding characteristics of central and peripheral insulin receptors are similar, these receptors do not appear to be regulated in the same manner. 相似文献

microRNA(miRNA)在人类恶性肿瘤的发生发展过程中起着重要作用。近期研究表明,miRNA通过结合特定靶标参与调控肝细胞癌(hepatocellularcarcinoma,HCC)的发生,可作为辅助生物标志物用于指导肝细胞癌的诊断和治疗,并为有效地监控和预防肝病提供了新途径。寻找miRNA靶标,阐明miRNA参与肝癌发生的调控机理,有利于肝癌的临床靶向基因治疗。通过总结miRNA在肝细胞癌中的调控机制及临床应用的研究进展,为寻找肝细胞癌早期诊断的生物标志物及介入治疗的靶点提供了参考。 相似文献
