Prediction of the fate of hydrophobic organic contaminants in soils is complicated by the competing processes of sorption and biodegradation. To test the hypothesis that sorbed naphthalene is unavailable to degradative microorganisms, we developed a simple kinetic method to examine the rates and extents of naphthalene degradation in soil-free and soil-containing systems in a comparison of two bacterial species. The method is predicated on the first-order dependence of the initial mineralization rate on the naphthalene concentration when the latter is below the Michaelis-Menten half-saturation constant (Km) for naphthalene for the organism under study. Rates and extents of mineralization were estimated by nonlinear regression analysis of data by using both a simple first-order model and a three-parameter, coupled degradation-desorption model described for the first time here. Bioavailability assays with two bacterial species (Pseudomonas putida ATCC 17484 and a gram-negative soil isolate, designated NP-Alk) gave dramatically different results. For NP-Alk, sorption limited both the rate and extent of naphthalene mineralization, in accordance with values predicted on the basis of the equilibrium aqueous-phase naphthalene concentrations. For strain 17484, both the rates and extents of naphthalene mineralization exceeded the predicted values and resulted in enhanced rates of naphthalene desorption from the soils. We conclude that there are important organism-specific properties which make generalizations regarding the bioavailability of sorbed substrates inappropriate.  相似文献

Phenanthrene biodegradation was investigated at different soil water contents [0.11, 0.22, 0.33, 0.44 g H2O (g soil)?1] to determine the effects of water availability on biodegradation rate. A subsurface horizon of Kennebec silty loam soil was used in this study. [9-14C] phenanthrene was dissolved in a mixture of organic contaminants that consisted of 76% decane, 6% ρ-xylene, 6% phenanthrene, 6% pristane, and 6% naphthalene, and then added to the soil. The highest rate of mineralization, in which 0.23% of the [9-14C] phenanthrene degraded to 14CO2 after 66 days of incubation, was observed at the soil water content of 0.44 g H2O/g dry soil. Most of the 14C remained in the soil as the parent compound or as nonextractable compounds by acetonitrile at the highest water content. Concentrations of nonextractable compounds increased with water content, but residual extractable phenanthrene decreased significantly with increasing water content, which presumably indicates that bio-transformation occurred. The mineralization analysis of radiolabeled 9th carbon in phenanthrene underestimated phenanthrene biodegradation. The strong adsorption and low solubility of phenanthrene contributed to the low mineralization of phenanthrene 9th carbon. The other components were subject to higher biological and abiotic dissipation processes with increasing soil water content.  相似文献

Summary The mineralization of 14C-labelled naphthalene was studied in pristine and oil-contaminated soil slurry (30% solids) under denitrifying conditions using a range of concentrations from below to above the aqueous phase saturation level. Results from sorption-desorption experiments indicated that naphthalene desorption was highly irreversible and decreased with an increase in the soil organic content, thus influencing the availability for microbial consumption. Under denitrifying conditions, the mineralization of naphthalene to CO2 occurred in parallel with the consumption of nitrate and an increase in pH from 7.0 to 8.6. When the initial substrate concentration was 50 ppm (i.e. close to the aqueous phase saturation level), about 90% of the total naphthalene was mineralized within 50 days, and a maximum mineralization rate of 1.3 ppm day–1 was achieved after a lag period of approx. 18 days. When added at concentrations higher than the aqueous phase saturation level (200 and 500 ppm), similar mineralization rates (1.8 ppm day–1) occurred until about 50 ppm of the naphthalene was mineralized. After that the mineralization rates decreased logarithmically to a minimum of 0.24 ppm day–1 for the rest of the 160 days of the experiments. For both of these higher concentrations, the reaction kinetics were independent of the concentration, indicating that desorption of the substrate governs the mineralization rate. Other results indicated that pre-exposure of soil to oil contamination did not improve the degradation rates nor reduce the lag periods. 农业土壤和黑碳(BC)两种不同的吸附剂吸附苯酚平衡后分离,每组一部分不做处理,另一部分通过加入无酚灭菌溶液脱附平衡后分离,制备得到在不同吸附位点上吸附有苯酚的两类不同类型的4种吸附苯酚的吸附剂,研究了在不同Pseudomonasputida ATCC 11172菌密度条件下吸附在这4种吸附剂上的苯酚的脱附行为.结果表明,土壤及BC对苯酚的吸附均呈现明显的非线性,可用Freundlich模型描述.吸附态的苯酚能否被微生物利用取决于微生物及吸附剂的性质,BC具有发达的微孔结构,微孔小于假单胞菌细胞尺寸,导致假单胞菌无法直接利用吸附在BC上的苯酚;土壤基本无微孔结构,微生物较易与吸附的苯酚发生表面接触,直接利用吸附态苯酚.BC和土壤上的吸附态苯酚的脱附行为能用三元位点模型很好地描述,模型计算结果表明BC上的苯酚脱附主要受慢速脱附和极慢速脱附控制,微生物降解速率受脱附控制,降解可加速BC上的慢速脱附和极慢速脱附;土壤上的苯酚脱附主要受快速脱附控制,微生物降解不受脱附速率限制,对土壤上的脱附行为基本无影响.  相似文献

The effect of microbial inoculation on the mineralization of naphthalene in a bioslurry treatment was evaluated in soil slurry microcosms. Inoculation by Pseudomonas putida G7 carrying the naphthalene dioxygenase (nahA) gene resulted in rapid mineralization of naphthalene, whereas indigenous microorganisms in the PAH-contaminated soil required a 28 h adaptation period before significant mineralization occurred. The number of nahA-like gene copies increased in both the inoculated and non-inoculated soil as mineralization proceeded, indicating selection towards naphthalene dioxygenase producing bacteria in the microbial community. In addition, 16S rRNA analysis by denaturing gradient gel electrophoresis (DGGE) analysis showed that significant selection occurred in the microbial community as a result of biodegradation. However, the indigenous soil bacteria were not able to compete with the P. putida G7 inoculum adapted to naphthalene biodegradation, even though the soil microbial community slightly suppressed naphthalene mineralization by P. putida G7.  相似文献

Successful remediation of soils co-contaminated with organics and metals may require a combination of technologies. This research addresses the organic component within co-contaminated sites. It is well known that metal contaminants in soil can partially or completely inhibit normal heterotrophic microbial activity and hence prevent in situ biodegradation of organics. Previous work has shown that a rhamnolipid biosurfactant can complex metals such as lead and cadmium. It has also been demonstrated, in pure culture, that rhamnolipid can mitigate metal inhibition during the degradation of naphthalene. The goal of this study was to investigate whether rhamnolipid could reduce the toxicity of a model metal, cadmium, to indigenous soil populations in two different soils, Brazito and Gila, during the mineralization of phenanthrene. Results show that cadmium inhibited phenanthrene mineralization in both soils at bioavailable cadmium concentrations as low as 27 µM. This inhibition was reduced by the addition of rhamnolipid. Since rhamnolipid is degraded by soil populations, a rhamnolipid pulsing strategy was used to maintain a constant level of rhamnolipid in the system. Using this strategy, phenanthrene mineralization levels comparable to the control (0 mM Cd/0 mM rhamnolipid) were achieved in the presence of toxic cadmium concentrations. This research demonstrates that pulsed application of rhamnolipid may allow bioremediation of organic contaminants in sites that are co-contaminated with organics and metals.  相似文献

Excited triplet states of organic chromophores, in particular aromatic ketones, are capable of inducing oxidation of a variety of organic compounds. These reactions probably play an important role in the degradation of organic contaminants in sunlit natural waters. The effect of dissolved natural organic matter (DOM) on the oxidation rate of twenty-two aquatic organic contaminants, including phenols, anilines, phenylurea and s-triazine herbicides, and some pharmaceuticals, was investigated using photoexcited benzophenone-4-carboxylate (CBBP) as the oxidant. For about half of the studied compounds, a decrease in depletion rate was observed in the presence of Suwannee River fulvic acid, used as a reference DOM. Also, depletion rates decreased with increasing DOM concentration, as verified for five selected compounds. Such an inhibitory effect of DOM on oxidation is attributed to its antioxidant properties, whereby oxidation intermediates of the contaminants are supposed to be reduced back to their parent compounds. The presented screening study shows that DOM may be a relevant factor for inhibiting the oxidation of many organic contaminants in surface waters and possibly in engineered water treatment systems.  相似文献

土壤中溶解性有机质(DOM)是生物活性和物理化学反应活性都很活跃的有机组分,主要通过疏水吸附、分配、氢键、电荷转移、共价键、范德华力等多种作用与有机污染物结合,提高溶液中有机污染物的溶解度,改变土壤中有机污染物的吸附-解吸、迁移-转化等环境行为.DOM对有机污染物的吸附-解吸、迁移-转化过程的影响有双重性:一方面,DOM与有机污染物在土壤表面的共吸附可增加土壤对有机污染物的吸附容量,促进有机污染物在土壤中的吸持;另一方面,DOM对有机污染物的增溶作用,有利于土壤中有机污染物的解吸,提高移动性.作为光敏剂,DOM能提高土壤中有机物的光解反应速率.在一定条件下,DOM也可影响土壤中有机污染物的水解过程.DOM对土壤中有机污染物环境行为的影响与DOM和有机污染物的性质及其相互作用的介质条件密切相关.  相似文献

Abstract: Exploiting microorganisms for remediation of waste sites is a promising alternative to groundwater pumping and above ground treatment. The objective of in situ bioremediation is to stimulate the growth of indigenous or introduced microorganisms in regions of subsurface contamination, and thus to provide direct contact between microorganisms and the dissolved and sorbed contaminants for biotransformation. Subsurface microorganisms detected at a former manufactured gas plant site contaminated with coal tars mineralized significant amounts of naphthalene (8–43%) and phenanthrene (3–31%) in sediment-water microcosms incubated for 4 weeks under aerobic conditions. Evidence was obtained for naphthalene mineralization (8–13%) in the absence of oxygen in field samples. These data suggest that biodegradation of these compounds is occurring at the site, and the prospects are good for enhancing this biodegradation. Additional batch studies demonstrated that sorption of naphthalene onto aquifer materials reduced the extent and rate of biodegradation, indicating that desorption rate was controlling the biodegradation performance.  相似文献

An Arthrobacter strain mineralized naphthalene and n-hexadecane dissolved in 2,2,4,4,6,8,8-heptamethylnonane. The extent of mineralization increased with greater volumes of solvent. Measurements under aseptic conditions of the partitioning of naphthalene into the aqueous phase from the solid phase or from heptamethylnonane showed that the rates were rapid and did not limit mineralization. The rate of mineralization of hexadecane was rapid, although partitioning of the compound into aqueous solution was not detected. The Arthrobacter sp. grown in media with or without heptamethylnonane did not excrete products that increased the aqueous solubility of naphthalene and hexadecane. Measurements of the number of cells in the aqueous phase showed that the Arthrobacter sp. attached to the heptamethylnonane-water interface, but attachment was evident even without a substrate in the heptamethylnonane. Tests with small inocula of the Arthrobacter sp. demonstrated that at least a portion of naphthalene or hexadecane dissolved in heptamethylnonane was degraded by cells attached to the solvent-water interface. The cells did not adhere in the presence of 0.1% Triton X-100. The surfactant prevented mineralization of the hexadecane initially dissolved in heptamethylnonane, but it increased the rate and extent of mineralization of naphthalene initially dissolved in heptamethylnonane. The data show that organic solvents into which hydrophobic compounds partition affect the biodegradation of those compounds and that attachment of microorganisms to the organic solvent-water interface may be important in the transformation.  相似文献

Dissolved organic matter (DOM) interaction with pesticides was examined studying the ability of DOM to desorb 8 pesticides previously sorbed to soil. DOM was originating from municipal waste composts at two maturity degrees, recovered at 20°C and by hot-pressurised subcritical water. Pesticide desorption depended on their previous sorption on soil. When sorption was low (KOC ≤ 50, sulcotrione, metalaxyl), water was more efficient than DOM for desorption. On the contrary, when sorption was high (KOC ≥ 2000, trifluraline), little effect of DOM was observed. For the moderately sorbed pesticides, DOM favoured pesticide desorption compared to water. For the lowest sorbed pesticides (KOC ≤ 100), hysteresis was increased with larger proportions of DOM extracted with subcritical-water. Dissolved organic matter extracted from fresh-immature compost had larger capacity to mobilize the sorbed pesticides than the DOM from the mature compost. The pesticide desorption resulted from the positive and competitive interactions between pesticide, DOM and soil surfaces. These interactions were modelled considering separate partitioning coefficients. A general equation allowed the deduction of specific coefficients describing interactions in solution between pesticides and the non-sorbed fraction of DOM remaining in solution. This fraction was supposed to contain the most hydrophilic fraction of DOM and was able to interact with the most polar pesticide (amitrol). When pesticide hydrophobicity increased, the partitioning between pesticide and DOM decreased. Modelling the three-phase system (liquid, DOM and solid phases) pointed out that the solid phase played the most important role on pesticide behaviour through the sorption process of DOM and pesticides.  相似文献

This research addressed the effect of mineral and organic soil constituents on the fate of organic compounds in soils. Specifically, it sought to determine how the associations between organic chemicals and different soil constituents affect their subsequent biodegradation in soil. Four 14C-labeled surfactants were aseptically adsorbed to montmorillonite, kaolinite, illite, sand, and humic acids. These complexes were mixed with a woodlot soil, and 14CO2 production was measured over time. The mineralization data were fitted to various production models by nonlinear regression, and a mixed (3/2)-order model was found to most accurately describe the mineralization patterns. Different mineralization patterns were observed as a function of the chemical and soil constituents. Surfactants that had been preadsorbed to sand or kaolinite usually showed similar mineralization kinetics to the control treatments, in which the surfactants were added to the soil as an aqueous solution. Surfactants that had been bound to illite or montmorillonite were typically degraded to lesser extents than the other forms, while surfactant-humic acid complexes were degraded more slowly than the other forms. The desorption coefficients (Kd) of the soil constituent-bound surfactants were negatively correlated with the initial rates of degradation (k1) and estimates of 14CO2 yield (Po) as well as actual total yields of 14CO2. However, there was no relationship between Kd and second-stage zero-order rates of mineralization (ko). Microbial community characteristics (biomass and activity) were not correlated with any of the mineralization kinetic parameters. Overall, this study showed that environmental form had a profound effect on the ultimate fate of biodegradable chemicals in soil. This form is defined by the physicochemical characteristics of the chemical, the composition and mineralogy of the soil, and the mode of entry of the chemical into the soil environment.  相似文献

The transport and transformation of dissolved organic matter (DOM) and dissolved inorganic nitrogen (DIN) through the soil profile impact down-gradient ecosystems and are increasingly recognized as important factors affecting the balance between accumulation and mineralization of subsoil organic matter. Using zero tension and tension lysimeters at three soil depths (20, 40, 60 cm) in paired forest and maize/soybean land uses, we compared dissolved organic C (DOC), dissolved organic N (DON) and DIN concentrations as well as DOM properties including hydrophilic-C (HPI-C), UV absorption (SUVA254), humification index and C/N ratio. Soil moisture data collected at lysimeter locations suggest zero tension lysimeters sampled relatively rapid hydrologic flowpaths that included downward saturated flow through the soil matrix and/or rapid macropore flow that is not in equilibrium with bulk soil solution whereas tension lysimeters sampled relatively immobile soil matrix solution during unsaturated conditions. The effect of land use on DOC and DON concentrations was largely limited to the most shallow (20 cm) sampling depth where DOC concentrations were greater in the forest (only zero tension lysimeters) and DON concentrations were greater in the cropland (both lysimeter types). In contrast to DOC and DON concentrations, the effect of land use on DOM properties persisted to the deepest sampling depth (60 cm), suggesting that DOM in the cropland was more decomposed regardless of lysimeter type. DOC concentrations and DOM properties differed between lysimeter types only in the forest at 20 cm where soil solutions collected with zero tension lysimeters had greater DOC concentrations, greater SUVA254, greater humification index and lower HPI-C. Our data highlight the importance of considering DOM quality in addition to DOC quantity, and indicate long-term cultivation reduced the delivery of relatively less decomposed DOM to all soil depths.  相似文献

Tetradecenyl succinic acid (TSA) is the major component of a detergent builder (C12-C14 alkenyl succinic acid), which is inherently biodegradable. 14C-TSA was dosed as a component of sewage sludge into a soil with a history of sludge amendment at final added concentrations of 1.5 and 30 mg (kg soil)-1. In addition, it was dosed to the soil in an aqueous solution to a final added concentration of 30 mg (kg soil)-1. Dose and form were found to have a pronouced effect on the mineralization kinetics. When dosed in a realistic form and concentration (i.e. 1.5 mg (kg soil)-1 as a component of sludge), TSA was mineralized at its highest rate and to its greatest extent, and the mineralization half-life was 2.4 days. When dosed at 30 mg (kg soil)-1 as a component of sludge, mineralization began immediately, and the half-life was 23 days. In contrast, when dosed at this concentration in aqueous solution, the onset of mineralization was preceded by a 13 day lag period and the mineralization half-life was 69 days. Primary biodegradation and mineralization rates of TSA were very similar. Approximately, half the radioactivity was evolved as 14CO2, while the remaining radioactivity became non-extractable, having presumably been incorporated into biomass or natural soil organic matter (humics). This study demonstrated that TSA is effectively removed from sludge-amended soils as a result of biodegradation. Furthermore, it showed the effect that dose form and concentration have on the biodegradation kinetics and the importance of dosing a chemical not only at a relevant concentration but also in the environmental form in which it enters the soil environment.  相似文献

The dissolved organic matter (DOM) is the term used for organic components of natural origin present in the soil solution and is probably the most available C-source that primes microbial activity in subsoils. Contrasting effects of organic C components on pesticide degradation have been reported; however, most studies have used model organic compounds with compositions and concentrations which differ substantially from those found in the environment. Degradation of atrazine (AT) by Chelatobacter heintzii SalB was monitored in liquid batch assays in the absence or presence of well-defined model C compounds (glucose, gluconate and citrate) as model DOM (mDOM) or complex, less-defined, environmental DOM solutions (eDOM: isolated humic substances, soil and plant residue extracts) at environmentally relevant concentrations. Glucose significantly increased AT degradation rate by more than a factor of 8 at and above 2.5?mg C L(?-?1). Optical density measurements showed that this stimulation is related to microbial growth. Gluconate and citrate had no effects unless at non-relevant concentrations (1,000?mg DOC L(?-?1)) at which stimulations (gluconate) or inhibitions (citrate) were found. The effects of eDOM added at 10?mg DOC L(?-?1) on AT degradation were generally small. The AT degradation time was reduced by factors 1.4-1.9 in the presence of humic acids and eDOM from soils amended with plant residues; however, no effects were found for fulvic acids or eDOM from a soil leachate solution or extracted from unamended peat or forest soil. In conclusion, DOM supplied as both mDOM and eDOM did not inhibit AT degradation at environmentally relevant concentrations, and stimulation can be found for selected DOM samples and this is partly related to its effect on growth.  相似文献

New amphiphilic block surfactants ABA based on a central segment of polycaprolactone with different molecular composition were evaluated in the bioremediation of naphthalene in water by Sphingomonas paucimobilis and compared with sodium dodecyl sulphate as reference surfactant (SDS). Also the biodegradation of the new surfactants by bacteria, S. paucimobilis and a mixture of bacteria (Pseudomonas aureginosa, Bacillus subtilis, Bacillus amyloliquefaciens and Bacillus megaterium) was studied by indirect impedance technique and carbon dioxide determination. All the bacteria biodegraded in solution and micellar phase the central segment of PCL with mineralization rates in the range of 0.024–0.036 mg of CO2 per day.S. paucimobilis biodegraded naphthalene in the presence of the new surfactants and GC analysis demonstrated that conversion to products started immediately after inoculum. In all the experiments, except for SDS, at 140 h of incubation time, the remaining naphthalene concentration was about 10% of the initial concentration. In contrast, the production of CO2 was delayed 4–7 days and values around 75% of naphthalene mineralization degree were achieved in three weeks. The addition of PCL-surfactants, in solution and in micellar phase, not interfered in the naphthalene mineralization. These results have shown promising potential of these biodegradable PCL-surfactants in surfactant-enhanced remediation (SER) technology for removing residual organics from contaminated groundwater and soils.  相似文献

Bioavailability of pesticides sorbed to soils is an important determinant of their environmental fate and impact. Mineralization of sorbed atrazine was studied in soil and clay slurries, and a desorption-biodegradation-mineralization (DBM) model was developed to quantitatively evaluate the bioavailability of sorbed atrazine. Three atrazine-degrading bacteria that utilized atrazine as a sole N source (Pseudomonas sp. strain ADP, Agrobacterium radiobacter strain J14a, and Ralstonia sp. strain M91-3) were used in the bioavailability assays. Assays involved establishing sorption equilibrium in sterile soil slurries, inoculating the system with organisms, and measuring the CO2 production over time. Sorption and desorption isotherm analyses were performed to evaluate distribution coefficients and desorption parameters, which consisted of three desorption site fractions and desorption rate coefficients. Atrazine sorption isotherms were linear for mineral and organic soils but displayed some nonlinearity for K-saturated montmorillonite. The desorption profiles were well described by the three-site desorption model. In many instances, the mineralization of atrazine was accurately predicted by the DBM model, which accounts for the extents and rates of sorption/desorption processes and assumes biodegradation of liquid-phase, but not sorbed, atrazine. However, for the Houghton muck soil, which manifested the highest sorbed atrazine concentrations, enhanced mineralization rates, i.e., greater than those expected on the basis of aqueous-phase atrazine concentration, were observed. Even the assumption of instantaneous desorption could not account for the elevated rates. A plausible explanation for enhanced bioavailability is that bacteria access the localized regions where atrazine is sorbed and that the concentrations found support higher mineralization rates than predicted on the basis of aqueous-phase concentrations. Characteristics of high sorbed-phase concentration, chemotaxis, and attachment of cells to soil particles seem to contribute to the bioavailability of soil-sorbed atrazine.  相似文献

DOM(Dissolved organic matter)是土壤微生物呼吸的重要底物,凋落物淋溶的DOM对土壤碳矿化具有重要影响。选择中亚热带地区具有代表性的杉木(Cunninghamia lanceolata)和米槠(Castanopsis carlesii)凋落叶作为研究对象,通过两个月的短期室内培养,把不同凋落叶浸提出的DOM添加到培养瓶中,定期测定土壤碳矿化速率,计算土壤碳累积矿化量,探讨两种等浓度等量DOM添加对土壤碳矿化的影响,并分析DOM化学性质在土壤碳矿化过程中的重要性。结果表明:米槠凋落叶浸提得到的DOC(Dissolved organic carbon)和DON(Dissolved organic nitrogen)浓度均显著高于杉木凋落叶的(P0.05),而杉木凋落叶浸提得到的DOM的UV吸收值(SUVA_(254))和HIX(Humification index)均显著低于米槠凋落叶的(P0.01)。添加等浓度等量杉木和米槠凋落叶DOM到土壤中均显著增加了土壤碳矿化速率,在第1天内分别比对照高198%和168%,3d后下降到61.8%和44.1%,14d后基本处于平稳状态,表明外源有机物添加对土壤碳矿化的前期影响较大。培养过程中,添加杉木和米槠凋落叶DOM的土壤碳矿化累积量均能采用双因素指数模型进行拟合(r~2=0.99),但添加两者凋落叶DOM后土壤碳矿化累积量没有显著差异。  相似文献
