A strain designated as S85T was isolated from a seaweed collected from coastal area of Chuuk State in Micronesia. The strain was gram-negative, rod-shaped, and non-motile and formed yellow colonies on the SWY agar (0.2 % yeast extract and 1.5 % agar in seawater) and Marine agar 2216. The strain grew at pH 5–9 (optimum, pH 8), at 15–40 °C (optimum, 25–28 °C), and with 1–9 % (w/v) NaCl (optimum, 3 %). The phylogenetic analysis based on 16S rRNA gene sequence showed that strain S85T was related to Lutibacter litoralis CL-TF09T and Maritimimonas rapanae A31T with 91.4 % and with 90.5 % similarity, respectively. The dominant fatty acids were iso-C15:0, iso-C15:0 3-OH and iso-C17:0 3-OH, C16:0 3-OH and summed feature 3 (C16:1 ω7c and/or iso-C15:0 2-OH). The major isoprenoid quinone was MK-6. The DNA G+C content of the type strain was 34.6 mol %. The major polar lipids were phosphatidylethanolamine, an unknown glycolipid and two unknown polar lipids. Based on this polyphasic taxonomic data, strain S85T stands for a novel species of a new genus, and we propose the name Ochrovirga pacifica gen. nov., sp. nov. Strain JLT2015T was isolated from surface seawater of the Southeastern Pacific. The strain was Gram-negative, aerobic, motile by gliding, and rod shaped. The dominant fatty acids were C18:1ω7c, C16:0, and C16:1ω7c. The major respiratory ubiquinone was Q-10, and the predominant polyamine pattern was spermidine. The components of the polar lipid profile were phosphatidylethanolamine, phosphatidylglycerol, diphosphatidylglycerol, and sphingoglycolipid. The DNA G+C content was 64.2 %. Phylogenetic analysis based on 16S rRNA gene sequence revealed strain JLT2015T belonged to belong to the family Sphingomonadaceae, exhibiting 94.7 % 16S rRNA gene sequence similarity with Novosphingobium pentaromativorans. On the basis of the taxonomic data presented, together with phylogenetic and genetic characteristics, strain JLT2015T is considered to represent a novel genus, for which the name Pacificamonas flava gen. nov., sp. nov. is proposed. The type strain of Pacificamonas flava is JLT2015T (=LMG27364T = CGMCC1.12401T).  相似文献

A novel heterotrophic, yellow pigmented, aerobic, Gram-negative, nonmotile, oxidase- and catalase-positive bacterium KMM 3938T was isolated from sea water collected in the Sea of Japan, Russia. The strain grew at mesophilic temperature range, and required the presence of NaCl for growth. 16S rRNA gene sequence analysis revealed that strain KMM 3938T is a member of the family Flavobacteriaceae. The predominant fatty acids were C13:0 iso, C14:0 iso, C15:0 iso, C15:0, C15:1Δ6, 3OH-C15:0:3 iso, and 3OH-C15:0. The G + C content of the DNA of KMM 3938T was 32.4 mol%. On the basis of phenotypic, chemotaxonomic, genotypic, and phylogenetic characteristics, the novel bacterium was assigned to the genus Polaribacter as Polaribacter butkevichii sp. nov. The type strain is KMM 3938T (= KCTC 12100T = CCUG 48005T). The GenBank accession number for Polaribacter butkevichii KMM 3938T is AY189722.  相似文献

A novel hyperthermophilic bacterium was isolated from pink filamentous streamers (pink filaments) occurring in the upper outflow channel (temperature, 82 to 88°C) of Octopus Spring in Yellowstone National Park, Wyo. The gram-negative cells grew at low salinity at temperatures up to 89°C in the neutral to alkaline pH range. Depending on the culture conditions, the organisms occurred as single motile rods, as aggregates, or as long filaments that formed streamer-like cell masses. The novel isolate grew chemolithoautotrophically with hydrogen, thiosulfate, and elemental sulfur as electron donors and oxygen as the electron acceptor. Alternatively, under aerobic conditions, formate and formamide served as sole energy and carbon sources. The novel isolate had a 16S rRNA sequence closely related to the 16S rRNA sequence obtained from uncultivated pink filaments. It represents a new genus in the order Aquificales, the type species of which we name Thermocrinis ruber (type strain, OC 1/4 [= DSM 12173]).  相似文献

A novel Gram-negative, orange-pigmented bacterial strain JLT2008T was isolated from the surface seawater of the Western Pacific and subjected to a polyphasic taxonomic study. Phylogenetic analysis based on 16S rRNA gene sequences revealed that strain JLT2008T belonged to the genus Erythrobacter, sharing the highest similarity (96.6 %) with Erythrobacter gangjinensis K7-2T and the lowest similarity (94.9 %) with Erythrobacter litoralis DSM 8509T. Strain JLT2008T did not contain bacteriochlorophyll a, and the predominant respiratory lipoquinone was ubiquinone-10. The major fatty acids were C18:1 ω7c, C16:0, C16:1 ω7c/C16:1 ω6c. The prominent polar lipids were sphingoglycolipid, phosphatidylethanolamine, and phosphatidylglycerol. The genomic G + C content was 60.1 mol %. Based on the polyphasic taxonomic data, a novel species within the genus Erythrobacter, and with the name Erythrobacter westpacificensis sp. nov., is proposed. A new aerobic, Gram-negative, chemo-organotrophic, euryhaline bacterium, designated strain JL2009T, was isolated from surface water of the Caribbean Sea. The strain formed flaxen colonies on marine ager 2216 (MA; Difco) medium. Cells were dimorphic, with stalks or a polar flagellum, and reproduction occurred by means of binary fission. Growth occurred at 15–45 °C (optimum at 35 °C), 0–15 % (w/v) NaCl (optimum at 1–10 %) and pH 5–9 (optimum at pH 7–8). Phylogenetic analysis based on 16S rRNA gene sequences showed that the strain formed a distinct evolutionary lineage within the family Hyphomonadaceae. Strain JL2009T was most closely related to Maricaulis parjimensis MCS 25T (92.2 % DNA sequence similarity), Woodsholea maritime CM2243T (90.9 %), and Oceanicaulis alexandrii C116-18T (90.9 %). The major respiratory quinone was Q-10. The dominant cellular fatty acids were summed feature 8 (C18:1 ω7c), C18:0 and 11-methyl C18:1 ω7c. The polar lipid pattern indicated the presence of phospholipid, phosphatidyl glycerol and glycolipids. The G + C content of the genomic DNA was 70.5 mol%. On the basis of the chemotaxonomic and phenotypic characteristics and the phylogenetic evidence, strain JL2009T represents a novel genus and species in the family Hyphomonadaceae, for which the name Euryhalocaulis caribicus gen. nov., sp. nov. is proposed. A new acetotrophic marine methane-producing bacterium that was isolated from the methane-evolving sediments of a marine canyon is described. Exponential phase cultures grown with sodium acetate contained irregularly shaped cocci that aggregated in the early stationary phase and finally differentiated into communal cysts that released individual cocci when ruptured or transferred to fresh medium. The irregularly shaped cocci (1.9 ± 0.2 mm in diameter) were gram negative and occurred singly or in pairs. Cells were nonmotile, but possessed a single fimbria-like structure. Micrographs of thin sections showed a monolayered cell wall approximately 10 nm thick that consisted of protein subunits. The cells in aggregates were separated by visible septation. The communal cysts contained several single cocci encased in a common envelope. An amorphous form of the communal cyst that had incomplete septation and internal membrane-like vesicles was also present in late exponential phase cultures. Sodium acetate, methanol, methylamine, dimethylamine, and trimethylamine were substrates for growth and methanogenesis; H2-CO2 (80:20) and sodium formate were not. The optimal growth temperature was 35 to 40°C. The optimal pH range was 6.5 to 7.0. Both NaCl and Mg2+ were required for growth, with maximum growth rates at 0.2 M NaCl and 0.05 M MgSO4. The DNA base composition was 41 ± 1% guanine plus cytosine. Methanosarcina acetivorans is the proposed species. C2A is the type strain (DSM 2834, ATCC 35395).  相似文献
