The antioxidant activities of water extract, methanol extract, ethyl acetate, and n-BuOH fractions of methanol extract from Lysimachia foenum-graecum Hance were investigated in this study. Various methods, such as the total antioxidant capacity measured by phosphomolybdenum method, scavenging activities towards 1,1-diphenyl-2-picryhydrazyl (DPPH) radical, superoxide anion radical, and hydroxyl radical, were established in in vitro systems. The amounts of total phenolics and total flavonoids in the extracts and fractions were also determined by spectrophotometric methods. The results showed that all the extracts and fractions exhibited antioxidant and radical-scavenging activities at different magnitudes of potency. The decreasing order of antioxidant and radical-scavenging activities among the extracts assayed through all the four methods were found to be ethyl acetate fraction>n-BuOH fraction>methanol extract>water extract. This similar order of the amounts of total phenolics and total flavonoids shows that the extent of antioxidant and radical-scavenging activities is in accordance with the amounts of phenolics and flavonoids present in extracts and fractions. The extracts of L. foenum-graecum Hance might be valuable antioxidant natural sources and seemed to be applicable in both healthy medicine and food industry.  相似文献

Citrus limonoids have shown to inhibit the growth of cancer in colon, lung, mouth, stomach and breast in animal and cell culture studies. For the first time in the present study, an attempt has been made to isolate antioxidant fractions and five limonoids from red Mexican grapefruit seeds. Defatted seed powder was successively extracted with hexane, ethyl acetate (EtOAc), acetone, methanol (MeOH) and MeOH/water and the extracts were concentrated under vacuum. Radical scavenging activity of 1,1-diphenyl-2-picrylhydrazyl (DPPH) and total phenolic content were also measured for comparison with the antioxidant capacity in the phosphomolybdenum method for the above extracts. Acetone and MeOH extracts, respectively, showed the highest (85.7%) and lowest (53.3%) radical scavenging activity, at 500 ppm. The total phenolic contents were found to be highest in the acetone extract (15.94%) followed by the MeOH extract (5.92%), ethyl acetate extract (5.54%) and water extract (5.26%). Antioxidant capacity of the extracts as equivalents to ascorbic acid (micromol/g of the extract) was in the order, EtOAc extract > acetone extract > water extract > methanol extract. Furthermore, the EtOAC and acetone extracts were loaded onto silica gel columns to obtain four limonoid aglycons. MeOH fraction was loaded onto a dowex-50 and sepabeads resin column to obtain a limonoid glucoside. The purity of the isolated five compounds was analyzed by HPLC using a C18 column and UV detection at 210 nm. Finally, the structures of the compounds were identified as obacunone, nomilin, limonin, deacetylnomilin (DAN) and limonin-17-beta-D-glucopyranoside (LG) using 1H and 13C NMR studies.  相似文献

In the present study, ethyl acetate, butanol and aqueous fractions derived from total methanol extract of Butea monosperma flowers were evaluated for radical scavenging activities using different in vitro models like reducing power assay, scavenging of 2,2 diphenyl-1-picrylhydrazyl (DPPH) radical, nitric oxide radical, superoxide anion radical, hydroxyl radical and inhibition of erythrocyte hemolysis using 2, 2' azo-bis (amidinopropane) dihydrochloride (AAPH). Methanol extract along with its ethyl acetate and butanol fractions showed potent free radical scavenging activity, whereas aqueous fraction was found to be devoid of any radical scavenging properties. The observed activity could be due to the higher phenolic content in the extracts (16.1, 25.29, and 17.74% w/w in methanol extract, ethyl acetate and butanol fractions respectively). HPTLC fingerprint profile of the ethyl acetate and butanol fractions were developed which would serve as reference standard for quality control of the extracts.  相似文献

以水、甲醇、乙醇和乙酸乙酯为溶剂,对罗汉果干果进行提取,分别采用磷钼酸铵体系、邻苯三酚自氧化体系、Fenton反应体系和卵黄脂质过氧化体系测定各种提取物的总抗氧化性能、超氧阴离子自由基和羟基自由基清除性能及其抗脂质过氧化作用。结果表明,四种溶剂提取物均具较强的抗氧化性和活性氧自由基清除性能,其能力的大小顺序为:乙酸乙酯提取物>水提物>甲醇提取物>乙醇提取物。  相似文献

The work was conducted with the purpose to evaluate antioxidant activity of Parmelia saxatilis (PS) by different analytical methods. Water and methanol were used as solvents and antioxidative effects were measured by a ferric thiocyanate method (FTC) and thiobarbituric acid test (TBA). The antioxidant activity increased with the increasing amount of extracts (from 50 to 250 μg) added to linoleic acid emulsion. The methanol extract of PS exhibited high antioxidative activity that was not significantly (P < 0.05) different from α-tocopherol, while aqueous extracts of PS showed low antioxidative activity. Similar trends of antioxidant activity were observed using either the FTC or TBA methods. Antioxidant activity, reducing power, free radical scavenging (DPPH·), superoxide anion radical scavenging, metal chelating and hydrogen peroxide scavenging activities of PS extracts showed dose dependence and increased with concentration of PS extract. The results obtained in the present study indicate that the PS might be a potential source of natural antioxidant.  相似文献

Various solvent extracts of the lichen Usnea ghattensis showed good antioxidant activity. A methanol extract prevented lipid peroxidation by 87% followed by 65% in Trolox at 20 μg/ml. It also showed superoxide anion scavenging activity and free radical scavenging activity 56% and 73%, respectively. The known antioxidants butylated hydroxytoluene (BHT), butylated hydroxyanisol (BHA) and quercetin at similar concentrations showed superoxide anion scavenging activity of 68, 59 and 47% and free radical scavenging activity 83, 77 and 69%, respectively. In addition, these extracts were inhibitory against Bacillus licheniformis, Bacillus megaterium, Bacillus subtilis and Staphylococcus aureus with MIC values of 5–10 μg/ml.Received after revisions 10 May 2005  相似文献

用清除二苯代苦味酰基(DPPH)自由基、2,2′-连氨-(3-乙基苯并噻唑啉-6-磺酸)二氨盐(ABTS)自由基和铁离子还原/抗氧化能力(FRAP)测定法对洛阳和信阳产景天三七(Sedum aizoon)体外总抗氧化活性进行评价,并与阳性对照丁基羟基茴香醚(BHA)、没食子酸丙酯(PG)和二丁基羟基甲苯(BHT)比较。发现洛阳和信阳景天三七有较好的体外抗氧化活性。洛阳和信阳产乙酸乙酯部位活性最强,正丁醇部位活性次之。在6个提取部位中,信阳产景天三七乙酸乙酯部位活性最强,其清除DPPH、ABTS自由基能力强于阳性对照BHA和BHT,还原Fe3+的能力大于阳性对照BHT,洛阳产景天三七乙酸乙酯部位和正丁醇部位活性次之。  相似文献

本文评价了香露兜叶的乙醇提取物以及石油醚、乙酸乙酯、正丁醇和水相组分的抗氧化活性。在体外测定了提取物和组分的总抗氧化活性,二苯代苦味肼基(DPPH)自由基清除活性,超氧阴离子自由基清除活性和铁离子还原能力,并利用分光光度法测定了其总酚和总黄酮的含量。结果表明,所有的提取物和组分均表现出抗氧化和自由基清除活性。抗氧化活性的大小如下:正丁醇部分>乙酸乙酯部分>乙醇提取物>水部分>石油醚部分。总酚含量的顺序和其相似,说明提取物和组分中的酚类化合物使其具有抗氧化活性。香露兜叶提取物可能会成为有价值的天然抗氧化资源,并将在保健品和食品中得到应用。  相似文献

Phenolic glucosides having radical scavenging activity were examined from the fraction eluted with 20% methanol on Amberlite XAD-2 resin applied to lemon (Citrus limon) juice by using reversed phase chromatography. Four phenolic glucosides were identified as 1-feruloyl-beta-D-glucopyranoside, 1-sinapoyl-beta-D-glucopyranoside, 6,8-di-C-glucosylapigenin and 6,8-di-C-glucosyldiosmetin by (1)H-NMR, (13)C-NMR, and MS analyses. They exhibited radical scavenging activity for 1,1-diphenyl-2-picrylhydrazyl (DPPH) and superoxide, although the activity was low in comparison with eriocitrin, a potent antioxidant in lemon fruit, and the eriodictyol of its aglycone. The phenolic compounds in lemon juice were examined for their suppressive effect on the expression of blood adhesion molecules by measuring the expression of intercellular adhesion molecule-1 (ICAM-1) in human umbilical vein endothelial cells (HUVECs) induced by necrosis factor-alpha (TNF-alpha). 6,8-Di-C-glucosylapigenin, apigenin, and diosmentin of the flavones were found to significantly suppress the expression of ICAM-1 at 10 muM (P<0.05). The phenolic glucosides isolated in this study were contained in comparative abundance in daidai (Citrus aurantium) and niihime (Citrus unshiu x Citrus tachibana) among the sour citrus juices.  相似文献

Antioxidant potential of leaves of three different species of Annona was studied by using different in vitro models eg., 1,1-diphenyl-2-picryl hydrazyl (DPPH), 2,2-azinobis-(3-ethylbenzothizoline-6-sulphonate) (ABTS), nitric oxide, superoxide, hydroxy radical and lipid peroxidation. The ethanolic extract of A. muricata at 500 microg/ml showed maximum scavenging activity (90.05%) of ABTS radical cation followed by the scavenging of hydroxyl radical (85.88%) and nitric oxide (72.60%) at the same concentration. However, the extract showed only moderate lipid peroxidation inhibition activity. In contrast, the extract of A. reticulata showed better activity in quenching DPPH (89.37%) and superoxide radical (80.88%) respectively. A.squamosa extract exhibited least inhibition in all in vitro antioxidant models excepting hydroxyl radical (79.79%). These findings suggest that the extracts of A. muricata possess potent in vitro antioxidant activity as compared to leaves of A. squamosa and A. reticulata suggesting its role as an effective free radical scavenger, augmenting its therapeutic  相似文献

In vitro antioxidant activities of three selected Indian red seaweeds - viz., Euchema kappaphycus, Gracilaria edulis and Acanthophora spicifera were evaluated. Total phenolic content and reducing power of crude methanol extract were determined. The antioxidant activities of total methanol extract and five different solvent fractions (viz., petroleum ether (PE), ethyl acetate (EA), dichloromethane (DCM), butanol (BuOH) and aqueous) were also evaluated. EA fraction of A. spicifera exhibited higher total antioxidant activity (32.01 mg ascorbic acid equivalent/g extract) among all the fractions. Higher phenolic content (16.26 mg gallic acid equivalent/g extract) was noticed in PE fraction of G. edulis. Reducing power of crude methanol extract increased with increasing concentration of the extract. Reducing power and hydroxyl radical scavenging activity of E. kappaphycus were higher compared to standard antioxidant (alpha-tocopherol). The total phenol content of all the seaweeds was significantly different (P<0.05). In vitro antioxidant activities of methanol extracts of all the three seaweeds exhibited dose dependency; and increased with increasing concentration of the extract.  相似文献

以乙酸乙酯和甲醇为提取剂,采用索氏提取法,剩余残渣采用热水浸提,最终得到桦褐孔菌不同极性提取物,对其DPPH自由基、羟基自由基及超氧阴离子自由基清除活性作用进行了研究,确定桦褐孔菌的抗氧化能力,为深入研究和开发桦褐孔菌功能性食品奠定理论基础。实验结果表明桦褐孔菌具有较好的抗氧化活性,其中乙酸乙酯提取物的DPPH自由基清除率、羟基自由基清除率和超氧阴离子自由基清除率均高于其他两组分及BHT,桦褐孔菌提取物有望成为功能性食品组分中合成抗氧化剂的天然替代品。  相似文献

采用70%丙酮溶液对荸荠皮中抗氧化物质进行提取,得到红棕色浸膏。通过定性及定量方法分析了荸荠皮提取物中可能存在的具有抗氧化活性的物质;采用DPPH自由基法测定了荸荠皮提取物对DPPH自由基的清除能力。结果显示,荸荠皮提取物中含有多酚类和黄酮类等化合物,其多酚含量为3.31%(w/w,以干物质计)。DPPH自由基法显示,荸荠皮提取物具有一定的清除DPPH自由基能力,其清除能力与提取物浓度之间显示出良好的剂量-效应关系。该提取物(IC50值为130.37 ppm)对DPPH自由基清除能力略低于BHT(IC50值为94.16 ppm)。  相似文献

采用甲醇、乙醇、异丙醇对樟芝发酵液和菌丝体进行提取,对其不同提取物进行还原力,DPPH、羟基自由基、超氧自由基、过氧化氢清除能力,亚铁离子螯合能力等抗氧化能力的测定。比较发酵液提取物和菌丝体提取物的抗氧化效果,比较不同提取剂提取物的抗氧化效果。结果显示:(1)发酵液提取物还原力最高的为异丙醇提取物,亚铁离子螯合能力最强的为乙醇提取物;DPPH清除能力乙醇和异丙醇提取物最高都达到97.88%;羟基自由基清除能力最高的为乙醇提取物,达到84.22%;3种提取物的超氧自由基清除能力都较高,具有最高清除率的异丙醇提取物达到94.89%;过氧化氢清除能力最高的为乙醇提取物的97.53%。(2)菌丝体提取物还原力最高和亚铁离子螯合能力最强的均为乙醇提取物;DPPH清除能力最高的为乙醇提取物的89.95%;3种提取物的羟基自由基清除能力普遍偏低,最高的仅为异丙醇提取物的37.28%;超氧自由基清除能力则普遍较高,最高为甲醇提取物的90.05%;过氧化氢清除能力最高的为乙醇和异丙醇提取物,最高清除率均为95.06%。抗氧化性能的研究比较,为进一步开发利用樟芝提供了依据。  相似文献

Free radical scavenging activity of the extracts of petals (bud, early stage, full blooming and ending stage), leaf, stem, root and seeds of Mogami-benibana (safflower, Carthamus tinctorius Linne), the contents of the major active components of carthamin and polyphenols, and neuroprotective effect of the petal extracts and carthamin in the brain of mice and rats were examined. Water extracts of Mogami-benibana petals scavenged superoxide, hydroxyl and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals and singlet oxygen. The scavenging activities of the extract of safflower petals with various colors showed the order of orange, yellow and white from high to low. This order is consistent with the contents of carthamin, which is a pigment of orange color and is found highest in orange petals and lowest in white petals. There was also a relationship between DPPH radical scavenging activity and carthamin content in the petal extracts of safflower. The neuroprotective effects were examined in cellular and animal models. Mogami-benibana petal extract inhibited glutamate-induced C6 glia cell death, significantly decreased the formation of malondialdehyde in mouse cerebrum, and inhibited the increase in thiobarbituric acid reactive substances and 8-hydroxy-2′-deoxyguanosine (8-OHdG) in the cerebral cortex of rats subjected to an injection of FeCl3 solution into the sensory motor cortex. Carthamin showed similar effects in inhibiting 8-OHdG by the petal extract in rats. These results suggest that the petal extract of Mogami-benibana has free radical scavenging activity and neuroprotective effect and carthamin is one of the major active components. Antioxidant activity of defatted methanol extract of D. malabarica bark was studied for its free radical scavenging property on different in vitro models e.g. 1,1-diphenyl-2-picryl hydrazyl (DPPH), nitric oxide, superoxide, hydroxyl radical and lipid peroxide radical model. The extract showed good dose-dependent free radical scavenging property in all the models except in hydroxyl radical inhibition assay. IC50 values were found to be 9.16, 13.21, 25.27 and 17.33 microg/ml respectively in DPPH, nitric oxide, superoxide and lipid peroxidation inhibition assays. In hydroxyl radical inhibition assay 1000 microg/ml extract showed only 10% inhibition with respect to the control. Measurement of total phenolic compounds by Folin-Ciocalteu's phenol reagent indicated that 1 mg of the extract contained 120.7 microg equivalent of pyrocatechol. The results indicate that the antioxidant property of the extract may be due to the high content of phenolic compounds. However, the underlying mechanism may not involve hydroxyl radical scavenging property.  相似文献

三种柑橘类果皮提取物对铜绿微囊藻生长的影响   总被引:1,自引:0,他引:1  
通过分析测定生物量、叶绿素a含量以及叶绿素荧光参数,研究了3种柑橘类果皮甲醇提取液对铜绿微囊藻生长的影响。结果表明,3种提取液都能有效抑制铜绿微囊藻的生长、叶绿素a合成与光合系统Ⅱ(PSⅡ)活性,并且抑制效果随着作用浓度增加而增强。3种提取液抑制作用强弱的顺序为:蜜橘〉西柚〉脐橙。当蜜橘皮提取液浓度大于1.10g/L时,抑藻效果显著(P〈0.05),培养9d后对铜绿微囊藻生长的抑制率达到86.4%,且在实验期间抑制作用没有减弱。当脐橙皮与西柚皮提取液的浓度大于3.31g/L时,抑藻效果显著(P〈0.05),但培养5d后抑制作用开始减弱。据此推测,3种柑橘类果皮提取液中存在一类或几类物质,能够抑制铜绿微囊藻的叶绿素a合成,降低PSⅡ活性,从而降低其光合作用效率,导致铜绿微囊藻的生长受到抑制。且这类物质能自然降解,随着作用时间的延长,其抑藻效果也逐渐消失。  相似文献

Cellular damage caused by reactive oxygen species has been implicated in several diseases and hence antioxidants have significant importance in human health. Cold water, hot water and methanolic extract of Pleurotus squarrosulus were evaluated for antioxidant activity against hydroxyl radical, DPPH (1,1-Diphenyl-2-picrylhydrazyl) radical, superoxide radical, nitric oxide (NO) scavenging, reducing power, ferrous ion chelating ability and beta-carotenellinoleic acid assay. Total phenol, flavonoid, beta-carotene and lycopene content were also determined. Hot water extract showed significant antioxidant activity in all the test systems. Hydroxyl radical scavenging activity of all the extracts has been significant compared to positive control. Hot water extract has been found to have higher phenolic, total flavonoid, beta-carotene and lycopene content than cold water and methanolic extract of the mushroom. Results of this study showed that, hot water extract has maximum antioxidant property and may be utilized as a promising source of therapeutics.  相似文献

The suitability of citrus peels, generated as a by-product of the juice industry, as a source of antioxidants was investigated. Citrus peel powder was prepared by lyophilizing 70% ethanol extract from citrus peels. Extraction was carried out at room temperature (20 degrees C) for 72 h. The extract was subjected to gamma-irradiation treatment (20 kGy). The aqueous solutions of citrus peel powder were examined for color characteristics and antioxidant potential in terms of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging, beta-carotene bleaching and nitrite scavenging activities. There were significant changes in Hunter color values due to irradiation. The a*- and b*-values decreased due to radiation treatment. DPPH radical scavenging, beta-carotene bleaching and nitrite scavenging activities were not affected by irradiation treatment. Nitrite scavenging activity was the highest in the extract at pH 1.2 followed by pH 4.2 and 6.0. These functional properties of the aqueous solution were found to be stable in heat treatment. It could significantly improve oxidative stability of lipids in fish meat system. Based on these results there may be opportunities to use citrus peel powder as a functional component in the food processing industry with gamma irradiation treatment improving its color characteristics without adversely influencing the functional properties.  相似文献

Spermacoce verticillata (L.) G. Mey. is commonly used in the folk medicine by various cultures to manage common diseases. Herein, the chemical and biological profiles of S. verticillata were studied in order to provide a comprehensive characterization of bioactive compounds and also to highlight the therapeutic properties. The in vitro antioxidant activity using free-radical scavenging, phosphomolybdenum, ferrous-ion chelating and reducing power assays, and the inhibitory activity against key enzymes such as acetylcholinesterase (AChE) and butyrylcholinesterase (BChE), tyrosinase, α-amylase and α-glucosidase of S. verticillata extracts (dichloromethane, ethyl acetate, methanol and water) were investigated. The highest total phenolic and flavonoid content were observed in the methanolic and aqueous extracts. Exhaustive 2DNMR investigation has revealed the presence of rutin, ursolic and oleanoic acids. The methanolic extract, followed by aqueous extract have showed remarkable free radical quenching and reducing ability, while the dichloromethane extract was the best source of metal chelators. The tested extracts showed notable inhibitory activity against cholinesterases (AChE: 1.63–4.99 mg GALAE/g extract and BChE: 12.40–15.48 mg GALAE/g extract) and tyrosinase (60.85–159.64 mg KAE/g extract). No inhibitory activity was displayed by ethyl acetate and aqueous extracts against BChE and tyrosinase, respectively. All the tested extracts showed modest α-amylase inhibitory activity, while only the ethyl acetate and aqueous extracts were potent against α-glycosidase. This study further validates the use of S. verticillata in the traditional medicine, while advocating for further investigation for phytomedicine development.  相似文献
