The internal transcribed spacer (ITS) region of nuclear ribosomal DNA, trnL and trnL-F genes of Cardamine glechomifolia Levl. (family Brassicaceae) were sequenced and analyzed with the sequence of related Cardamine species retrieved from NCBI GenBank to detect pattern of evolutionary differentiation. All trees resulting from combined sequence analyses data of ITS, trnL and trnL-F gene resolve that C. glechomifolia – an endemic species to South Korea clade with Cardamine microzyga (100% bootstrap support). The evolutionary history was inferred using the Maximum Parsimony method. The consistency index is (0.588235), the retention index is (0.687500), and the composite index is 0.519622 (0.404412) for all sites and parsimony-informative sites (in parentheses). The result of the analysis using Maximum Parsimony was found congruence with Maximum Likelihood method and in Baseyan analysis.  相似文献

Nasturtium officinale (2n=4x=32),N. microphyllum (2n=8x=64) andN. ×sterile (2n=6x=48) have been investigated by isozyme analyses to study evolutionary processes withinNasturtium. Four additional species assumed to be involved in the formation of the octoploidN. microphyllum (Rorippa amphibia, R. sylvestris, R. palustris, andCardamine amara) were also examined. A total of 641 individuals were analyzed for six isozyme systems (alcohol dehydrogenase, aspartate aminotransferase, glutamate dehydrogenase, leucine aminopeptidase, malate dehydrogenase, and phosphoglucoisomerase). Eleven gene complexes coding for 43 allozymes were detected. Fifteen alleles were observed inN. officinale, twelve of them being fixed. All alleles fixed inN. officinale were also present and fixed inN. microphyllum. Seven additional fixed alleles were observed inN. microphyllum. The presence of these seven alleles inRorippa taxa provide evidence for an allopolyploid origin ofN. microphyllum withN. officinale and aRorippa taxon involved.C. amara is not a parent species ofN. microphyllum. N. ×sterile showed a fixed banding pattern which was identical to that ofN. microphyllum. It is argued thatN. ×sterile is a hybrid betweenN. officinale andN. microphyllum. Human impact has played a major role in the evolution ofN. ×sterile. The formation and persistence of the hybrid were influenced by introducingN. officinale into the natural distribution area ofN. microphyllum and by creating ditches and ponds where due to its vegetative capabilitiesN.×sterile could establish.  相似文献

The phylogenetic relationships of nine genera in four tribes of the family Brassicaceae were estimated from the sequences of the internal transcribed spacer region (ITS) of the 18S–25S nuclear ribosomal DNA. The entire ITS region of 16 accessions belonging to 10 species of seven genera was sequenced. Eight published sequences of Brassicaceae were also used. A total of 27 sequences were included in this study; four of them were found to be pseudogenes. Both the neighbor-joining and the parsimony trees suggest that the nine genera can be divided into three groups: (1) Arabidopsis,Cardaminopsis,Capsella, and Lepidium; (2) Rorippa and Cardamine; and (3) Brassica, Sinapis, and Raphanus. In contradiction to the proposal that Cardaminopsis and Arabidopsis be put into an expanded tribe Arabideae, our data show that these two genera are more closely related to Capsella and Lepidium (tribe Lepidieae) than to Rorippa and Cardamine (tribe Arabideae). Further, our data show that within the tribe Brassiceae, Raphanus is more closely related to B. nigra than to the B. oleracea/B. rapa clade. This result is in agreement with the nuclear data obtained in several studies, but is in conflict with the RFLP data of mitochondrial and chloroplast DNA. As pointed out by previous authors, it is possible that Raphanus is a hybrid between the B. nigra and B. oleracea/B. rapa lineages with the latter as the maternal parent.  相似文献

Macrocystis (Lessoniaceae) displays an antitropical distribution, occurring in temperate subtidal regions along western North America in the northern hemisphere and throughout the southern hemisphere. We used the noncoding rDNA internal transcribed spacer regions (ITS1 and ITS2) to examine relatedness among (1) Macrocystis and several genera of Laminariales, (2) four species of Macrocystis ( M. integrifolia Bory from the northern hemisphere, M. angustifolia Bory and M. laevis Hay from the southern hemisphere, and M. pyrifera [L.] C. Ag. from both hemispheres), and (3) multiple clones of several individuals. Of the taxa included in our phylogenetic analysis, the elk kelp, Pelagophycus porra (Lem.) Setch., was the sister taxon to Macrocystis spp. Macrocystis individuals from the southern hemisphere (representing three species) formed a strongly to moderately supported clade, respectively, when the ITS1 and ITS2 sequences were analyzed separately. No distinction was detected between the two species in the northern hemisphere. Thus, Macrocystis may be a monospecific genus ( M. pyrifera ). A northern-hemisphere-to-southern-hemisphere pattern of dispersal was inferred, because northern-hemisphere individuals were more diverse and displayed paraphyletic clades, whereas southern-hemisphere individuals were less diverse and formed a monophyletic clade. High intraindividual variation in ITS1 sequences was observed in one individual from Santa Catalina Island (CA), suggesting very recent and rapid mixing of genotypes from areas to the north and Baja California (Mexico) or introgressive hybridization with Pelagophycus.  相似文献

The systematics ofScleromitrula andVerpatinia of the family Sclerotiniaceae is reevaluated on the basis of morphological, cultural and molecular criteria.Scleromitrula shiraiana, Verpatinia species andCiborinia candolleana share gross morphological, microanatomical and cultural features which clearly distinguish them from the closely relatedCiborinia andRutstroemia species. Phylogenetic analyses of sequences from the internal transcribed spacer region (ITS1, ITS2, and the 5.8S gene) of nuclear ribosomal DNA demonstrate that the stipitate-capitate specimens ofScleromitrula andVerpatinia species plus the stipitate-cupulateCiborinia candolleana constitute a monophyletic clade separate from a clade including the type species ofCiborinia. Scleromitrula is emended to includeS. shiraiana, the new speciesS. rubicola, C. candolleana, and specimens formerly assigned toVerpatinia. The polyphyly ofSarcostemma sensu latu, suspected on morphological and karyological reasons is confirmed by analysis of thetrnL-F spacer region. With the exception ofCynanchum montevidense (subgen.Mellichampia, Sundell 1981), Old World and New World taxa belong to different clades.Sarcostemma sensu stricto andPlatykeleba show a strong affinity to Old WorldCynanchum; the affinities ofOxystelma cannot be determined at present.Philibertia andBlepharodon are sister groups, both are sister toFunastrum, andPentacyphus is basal to the New World clade.  相似文献

We diagnose new subfossil specimens belonging to the extinct palaeopropithecid, Mesopropithecus,from several caves at the Ankarana massif in northern Madagascar. They include three partial crania with mandibles, and the first vertebrae, ribs, clavicle, pelvis, ulna, and hand and foot bones of Mesopropithecusever found. Several other postcranial elements are known for this northern variant of Mesopropithecusand for previously described species from central and southern Madagascar. The new materials cannot be accommodated in either currently recognized species of this genus and are,hereby, given the name Mesopropithecus dolichobrachion. M. dolichobrachionis the second new species of extinct subfossil lemur to be found in the caves of Ankarana. It is unlike the two previously described species of this genus primarily in its larger size, different limb proportions, and aspects of its long bone morphology. Most notable is the fact that this is the only species of Mesopropithecuswith the forelimb longer than the hind limb— hence the name, the “long-armed” Mesopropithecus.Various characteristics place M. dolichobrachionphenetically closest (of species belonging to Mesopropithecus)to Babakotia radofilai, Archaeoindris,and Palaeopropithecus.If these similarities are derived specializations rather than primitive for palaeopropithecids, a new generic name will be required.  相似文献

Colletotrichum cereale is an ascomycete inhabitant of cool-season Pooideae grasses. The fungus has increased in frequency over the past decade as a destructive pathogen of Poa annua and Agrostis stolonifera turfgrass. Colletotrichum cereale exists as two lineages, designated clades A and B, but little is known about the distribution of these clades in natural environments, or what role these subdivisions may play in the trajectory of disease outbreaks. In this study, our objective was to determine the frequency of C. cereale clades A and B. To rapidly discriminate between the two C. cereale clades, a real-time PCR assay was developed based on the Apn2 gene. A collection of 700 C. cereale pathogens and endophytes from twenty Pooideae grass genera were genotyped. 87% of the collection was identifed as part of clade A, 11.7% as part of clade B, and 1.3% was a mixture. Colletotrichum cereale from turfgrass hosts in North America were most commonly members of clade A (78%). The overabundance of clade A in turfgrass isolates was directly attributable to the dominance of this lineage from southern sampling sites, irrespective of host. In contrast, 111 C. cereale turfgrass isolates collected from northern sampling sites were evenly distributed between clades A and B. Only 28% of C. cereale from A. stolonifera at northern sampling sites were part of clade A. These data show that environmental factors such as geographic location and host identity likely played a role in the distribution of the major C. cereale clades in North American turfgrass.  相似文献

Bactrocera caudata is a pest of pumpkin flower. Specimens of B. caudata from the northern hemisphere (mainland Asia) and southern hemisphere (Indonesia) were analysed using the partial DNA sequences of the nuclear 28S rRNA and internal transcribed spacer region 2 (ITS-2) genes, and the mitochondrial cytochrome c oxidase subunit I (COI), cytochrome c oxidase subunit II (COII) and 16S rRNA genes. The COI, COII, 16S rDNA and concatenated COI+COII+16S and COI+COII+16S+28S+ITS-2 nucleotide sequences revealed that B. caudata from the northern hemisphere (Peninsular Malaysia, East Malaysia, Thailand) was distinctly different from the southern hemisphere (Indonesia: Java, Bali and Lombok), without common haplotype between them. Phylogenetic analysis revealed two distinct clades (northern and southern hemispheres), indicating distinct genetic lineage. The uncorrected ‘p’ distance for the concatenated COI+COII+16S nucleotide sequences between the taxa from the northern and southern hemispheres (‘p’ = 4.46-4.94%) was several folds higher than the ‘p’ distance for the taxa in the northern hemisphere (‘p’ = 0.00-0.77%) and the southern hemisphere (‘p’ = 0.00%). This distinct difference was also reflected by concatenated COI+COII+16S+28S+ITS-2 nucleotide sequences with an uncorrected ''p'' distance of 2.34-2.69% between the taxa of northern and southern hemispheres. In accordance with the type locality the Indonesian taxa belong to the nominal species. Thus the taxa from the northern hemisphere, if they were to constitute a cryptic species of the B. caudata species complex based on molecular data, need to be formally described as a new species. The Thailand and Malaysian B. caudata populations in the northern hemisphere showed distinct genetic structure and phylogeographic pattern.  相似文献

Two new species ofCarex (Cyperaceae) are described from southern South America.Carex transandina occurs in naturally-deposited rock talus and scree in southwestern Chile andC. austroamericana occurs in the Patagonian steppe in Chile and Argentina. In the past, specimens ofC. austroamericana have been erroneously referred toC. andina var.subabscondita Kük., and it is shown here that the latter entity is conspecific withC. nelmesiana Barros. The taxonomy ofC. nelmesiana is presented and its relationship to other members ofCarex sect.Junciformes (Boeckl.) Kük. is discussed.  相似文献

The results of a pilot DNA sequencing study of cycads conducted at the new molecular systematics laboratory at Fairchild Tropical Garden are presented and assessed with reference to previous phylogenetic analyses and classification schemes based on morphology and anatomy. Two DNA regions were sequenced and analyzed for variation, an intron in the trriL gene in the chloroplast genome (trriL intron) and the internal transcribed spacer region between the 5.8S and 26S ribosomal DNA subunits (ITS2). The trnL intron proved to be relatively conservative among cycad genera, while the ITS2 region contained higher levels of variation. Parsimony analysis of the sequences suggests a number of relationships, some of which were inferred by previous morphological studies, some of which are new. The sequences ofCycas are the most divergent among cycads, suggesting the longest isolation.Dioon is relatively isolated from the other genera and contains two major clades.Stangeria does not appear closely related toBowenia but does seem to have a weak affinity withZamia andMicrocycas. Lepidozamia is more closely related toEncephalartos than toMacrozamia. Sequence variation among the species ofCeratozamia is low.Microcycas andZamia are closely related.  相似文献

Sequences of the internal transcribed spacers (ITS) of nuclear ribosomal DNA were determined for 15 species ofIllicium (Illiciaceae) to examine phylogenetic relationships. The ITS trees show a major dichotomy between the two North American species (I. floridanum andI. parviflorum) and the remaining east Asian species. This suggests that the existing division between two sections (sect.Illicium and sect.Cymbostemon) ofIllicium based on tepal characters in unnatural. The ITS phylogeny shows congruence with palynology: of the species examined, the three species (I. angustisepalum, I. anisatum andI. fargesii) from sect.Illicium that possess trizonocolpate pollen consistently form a clade, although nesting within a clade consisting of the species of sect.Cymbostemon, which generally have trisyncolpate pollen. The low ITS sequence divergence and the close relationship among east Asian species suggest a recent diversification of this group of species or an unusual slowdown of sequence mutations.  相似文献

New sequences have been produced of the ITS rDNA region of twenty-nine species ofParmeliaceae, and analysed by neighbour-joining maximum likelihood distance analysis, and maximum parsimony analysis. Twelve monophyletic groups have been identified, most of which correspond to proposed generic segregated fromParmelias. lat.ParmeliaandParmelina,Melanelia, andPuncteliaappear as independent monophyletic groups; the type species of the two latter genera were, however, not included in the analysis. The parsimony analysis does not supportXanthoparmeliaas a monophyletic group separate fromNeofuscelia, andRimelia reticulatais nested withinParmotrema. Parmelinopsis horrescensis more closely related toHypotrachyna revolutathan toParmelina. Genetic distance values suggest thatFlavoparmelia caperatais closer toParmotremathan to other taxa included, and thatPlatismatia glaucaandHypogymnia tubulosaare closer toParmelias. lat. than to thePhysciaceae. However, there is no conclusive support for any of the latter groupings in the parsimony analysis. We conclude that the ITS region contains valuable information for studying generic delimitations within the familyParmeliaceae, but our study suggests that this region will not conclusively resolve within-family relationships in this group.  相似文献

Based on the study of herbarium material, a critical taxonomic account has been provided onCardamine sect.Macrophyllum (Cruciferae) from the Asian territory of the Soviet Union. Different morphological characters were examined but special attention was paid to flowers, nectar glands, pollen grains, fruits and seeds, followed by the anatomical studies of fruits and seeds. 8 species have been recognized which are arranged in 2 subsections underCardamine sect.Macrophyllum viz. 1.Macrophyllae (C. leucantha, C. tomentella, C. macrophylla andC densiflora) and 2.Prorepentae (C. schinziana, C. yezoensis, C. amareaformis andC. prorepens). Phylogenetic trend also has been traced out suggesting preliminary centres of divergency and polymorphism of the treated taxa.  相似文献

The Japanese golden chanterelle commonly identified as Cantharellus cibarius was sampled in a broad range of forest vegetation. A total of 90 fresh and 11 herbarium specimens were examined microscopically, subjected to sequencing analysis of their nuclear ribosomal RNA (rDNA) and tef-1 genes, and their characteristics were compared with those of European C. cibarius. Based on morphological and ecological characteristics, basidioma samples from Japan were divided into four species. While specimens of Cantharellus sp. 4 from Hokkaido Island were included in the European C. cibarius clade phylogenetically, the other three species formed three unique clades. Among these, Cantharellus anzutake sp. nov. is sister to the clade of C. cibarius and was widely sampled from the northern limit of Honshu Island to the southern limit of Kumejima Island in Ryukyu Islands. Although C. anzutake was morphologically similar to C. cibarius, the two species were phylogenetically distinct. Other morphologically similar but genetically distinct chanterelle species from India exhibited macroscopic and microscopic differences compared with C. anzutake.  相似文献

Coral–algal symbioses are essential for the survival of corals. Algal endosymbionts, specifically the dinoflagellate genus Symbiodinium, are divided into several genetic clades. The composition of Symbiodinium within corals plays an important role in the tolerance and/or sensitivity of host corals to local environments, due to individual Symbiodinium-specific physiological characteristics. While the majority of gamete-spawning corals acquire Symbiodinium from the surrounding environment, little is known about whether corals specifically select or randomly acquire Symbiodinium from the environmental population. In the present study, we compared the Symbiodinium clade composition of newly recruited Acropora corals with that of the environmental pool (water column, sediments, and adult colonies). More than 90 % of recruits harbored clades A and/or D until 6 months after settlement, despite the Symbiodinium environmental pool being mainly composed of clade C (mainly ITS1 type C2), and to a lesser extent clades A and D. In addition, the environmentally dominant type C2 Symbiodinium was not detected in Acropora recruits, while a few recruits harbored ITS1 types C1 or C15. Therefore, the clade composition of recruits may not reflect the abundance/density of Symbiodinium populations in the environment. Some members of clades A and D are known to exhibit tolerance to a wide range of environments. ITS1 type C1 also exhibits greater tolerance to thermal stress compared to ITS1 type C2. These tolerance characteristics of certain Symbiodinium may be vital for the initial survival of Acropora recruits, even if these Symbiodinium are rare in the environment.  相似文献

This review highlights evolutionary processes in the Brassicaceae which cause, accompany or are a consequence of biological invasions. Case studies in Capsella and Diplotaxis address the genetics of colonising species. The greatest colonising ability in Capsella bursa-pastoris is associated with polyploidy, predominant selfing, and high genetic diversity. Success of colonial populations seems to be due to the introduction of preadapted genotypes. Colonising species in Diplotaxis reveal contrasting evolutionary patterns. Genetic attributes in D. muralis include annuality, polyploidy, and predominant selfing. Very different from Capsella, D. muralis is nearly devoid of genetic diversity as revealed by molecular markers. In contrast to D. muralis, the colonising D. tenuifolia is perennial, diploid, self-incompatible, and displays high genetic diversity. Hybrid speciation, establishment of the hybrids in man-made habitats, stabilisation of their reproductive system, and reproductive isolation from the parent populations were analysed in Cardamine. The Nasturtium example highlights the importance of hybridisation for the evolution of invasiveness. The last case study concentrates on the evolutionary consequences of hybridisation between native and invading Rorippa species. Introgressive hybridisation between the invasive Rorippa austriaca and native Rorippa species is common and widespread in central Europe, and interspecific gene transfer has led to the formation of a new invasive genotype. Each successful invasion presents new aspects and sound case studies are needed in order to understand the ecology and evolution of the colonisation process and to enable us to assess the evolutionary consequences of biological invasions. Carex species are dominant and abundant plants in boreal and arctic landscapes, typically covering large wetland areas. Most of these vegetation-characteristic species are from Carex sections Phacocystis and Vesicariae, frequently growing together, but also forming monodominant stands. Here we study these species in a phylogenetic framework to infer whether this co-occurrence pattern results from convergent evolution. In both sections, we observed a Northern clade consisting only of arctic to boreal species, a Mixed clade of northern and more southerly distributed species and a Southern grade of species mainly from temperate or further southern zones. The species of the Northern clades of both sections that are the focus of the study may be of similar young age and are rather equally diversified in terms of molecular divergence and morphology, suggesting a replicate adaptive radiation in boreal to arctic habitats. Morphological characters possibly linked with functional importance are not significantly different between the respective clades of the two sections, whereas reproductive structures may be phylogenetically constrained. The evolution of salt tolerance and ongoing diversification in the Northern clade of sect. Phacocystis suggests that the observed replicate adaptive radiation might be a transitional state in the diversification of species and may explain why such radiations are so rarely documented.  相似文献
