在15 ℃~35 ℃的温度范围内比较测定了玫烟色拟青霉(Paecilomyces fumosoroseus) 6株野生菌的菌落生长、产孢量及孢子活力.结果显示,3个指标均以25 ℃左右最好,但同一温度下菌株间或同一菌株在不同温度下的差异显著, 菌株Pfr116和Pfr6206具有相对稳定优良的生长、产孢及活孢率性状.对不同浓度多菌灵对6株野生菌的菌落生长和单孢菌落形成的影响测定表明,菌株间存在较大差异.不同多菌灵浓度对单孢菌落形成的抑制率观察值可用逻辑斯蒂模型较好地拟合(r2≥0.90).用所获参数估计的最低禁菌浓度MIC值显示,Pfr4205和Pfr116对多菌灵表现为低抗性(MIC≤20.0 μg·ml-1);Pfr153、Pfr612和Pfr2175属于低水平中抗,MIC值仅略大于20 μg·ml-1;而Pfr6206的MIC值高达93.5 μg·ml-1,接近MIC>100 μg·ml-1的高抗标准.因此,Pfr6206为适温性和抗多菌灵特性俱佳的菌株,可作为抗多菌灵和适应不同季节或地区的害虫生防菌剂的候选菌株.  相似文献

采用有机磷固体培养基和无机磷固体培养基从沉积物中分离出具有解磷能力的菌株,通过平板划线分离纯化后得到6株磷细菌,其中2株为有机P细菌(编号为OP1、OP2),4株为无机磷细菌(编号分别为NOP1、NOP2、NOP3、NOP4).测定发现,OP1、OP2和NOP3溶磷能力较强,NOP4解磷能力较微弱,而NOP1及NOP2在分离纯化后失去了解磷能力;菌株OP1及OP2具有较强的分解有机磷卵磷脂的能力,接种OP1、OP2菌株的培养液中水溶性磷含量分别比对照增加了38.53和64.53倍;接种NOP3菌株的培养液中磷含量比对照增加了54.06倍.  相似文献

通过对不同化工厂废水处理池的活性污泥进行富集、不同浓度银离子驯化后,分离得到30株银离子抗性细菌,最大硝酸银耐受量可达80 mg·ml-1.对这些细菌进行质粒抽提与鉴定,质粒检出率为76.67%.40 mmol·L-1的苯甲酸钠对HAg4细菌质粒的消除率可达98.75%;而350 μg· ml-1的吖啶橙对HAg4细菌质粒的消除率只有77.78%.细菌质粒与银离子抗性密切相关.  相似文献

为了揭示油松-根系微生物的互作关系及其对油松林分稳定性的影响,本研究选择陕西省黄龙县天然和人工油松林,采集油松根际与非根际土壤,测定非根际土壤化学特性,分离纯化根际土壤解磷(有机磷和无机磷)细菌,通过DNA基因测序鉴定解磷细菌,并测定解磷细菌的解磷能力。结果表明: 天然油松林非根际土壤中全碳(TC)、全氮(TN)含量以及C/N、N/P极显著高于人工油松林。2种油松根际土壤中共鉴定出20属65种解磷细菌,其中以芽孢杆菌属、链霉菌属和假单胞菌属为优势菌群;天然油松林根际解磷细菌多样性、丰富度和均匀度指数均高于人工油松林,而优势度指数低于人工油松林。链霉菌属与非根际土壤TC、TN和C/N、N/P呈正相关,而芽孢杆菌属和假单胞菌属与非根际土壤硝态氮、铵态氮、有效磷及全磷含量呈正相关。2种油松林根际土壤不同解磷细菌的解磷能力存在差异,其中天然和人工油松林根际共有的解磷细菌为假单胞菌Pseudomonas sp.34-5,其对磷酸钙的解磷能力最高,为11.40 μg·mL-1;天然油松林根际独有的解磷细菌蕈状芽胞杆菌BF1-5对卵磷脂的解磷能力最高,为4.58 μg·mL-1。该林区2种油松林根际解磷细菌菌群结构存在显著差异。与人工油松林相比,天然油松林根际土壤解磷细菌群落多样性更丰富且分布更均匀,解磷能力普遍高于人工林。  相似文献

利用盆栽试验研究两种不同配比的控释复合肥CRFA(4%树脂包膜,N∶P2O5∶K2O为14∶14∶14)和CRFB(4%树脂包膜,N∶P2O5∶K2O为20∶8∶10)及普通复合肥CCF(N∶P2O5∶K2O为15∶15∶15)对杭白菊营养吸收和生长效应的影响.结果表明:普通复合肥CCF1(每盆6 g氮素用量的CCF)和CCF2(每盆3 g氮素用量的CCF)施入土壤后30 d,土壤中碱解氮、有效磷、有效钾含量分别为163.29和145.26 mg·kg-1、180.39和163.13 mg·kg-1、300.08 和213.15 mg·kg-1,而后迅速下降.控释复合肥养分释放较慢,其土壤碱解氮含量在施肥后缓慢升高,在施肥后60 d达到高峰,此时CRFA1(每盆6 g氮素用量的CRFA)、CRFB1(每盆6 g氮素用量的CRFB)、CRFA2(每盆3 g氮素用量的CRFA)、CRFB2(每盆3 g氮素用量的CRFB)分别为129.51、138.65、118.36、126.31 mg·kg-1;CRFA1和CRFA2处理土壤有效磷含量与CCF处理变化趋势基本一致,施肥后30 d分别达到169.54和133.46 mg·kg-1,CRFB1和CRFB2处理在施肥后60 d左右达到释放高峰,含量分别为137.13和84.68 mg·kg-1,然后缓慢下降.两种不同养分配比的控释复合肥处理植株叶面积、叶面积系数、分枝数、开花率、每株花数、鲜花直径等农艺性状均明显优于等氮素用量的普通复合肥处理,其中CRFB控释效果优于CRFA,其更符合杭白菊对养分的需求,且在本试验条件下,CRFB2处理产量最高.  相似文献

采用在根内生成有色铜沉淀的方法研究大豆(Glycine max)初生根凯氏带对铜离子的通透性。用真空泵抽取浓度为200 μmol·L–1 的CuSO4溶液进入根中, 然后在重力作用下从根基部灌注400 μmol·L–1的K4[Fe(CN)6]溶液, 两种物质在根内相遇即可产生棕色的Cu2[Fe(CN)6]沉淀, 根据沉淀的位置来确定铜离子所经过的途径。结果表明: Cu2+可以穿过内皮层凯氏带, 在木质部导管壁以及凯氏带至木质部之间的细胞壁处产生棕色沉淀, 侧根发生的部位也产生了大量的沉淀; 当抽取K4[Fe(CN)6]溶液后再灌注CuSO4溶液, 发现Cu2+仍然可以穿过凯氏带, 并在凯氏带外侧以及外皮层细胞的细胞壁处产生棕色沉淀。研究结果证明凯氏带并不是一个可以完全阻止离子进出的完美屏障。  相似文献

采用48 h急性毒性试验研究了DDT对多刺裸腹溞的48小时LC50值,采用生命表试验方法研究了暴露于不同浓度(1、8、16、24、32和40 μg·L-1)DDT溶液中的多刺裸腹溞的生命表统计学参数.结果表明:DDT对多刺裸腹溞48小时LC50值为324 μg·L-1;浓度在1~40 μg·L-1的 DDT对多刺裸腹溞的生命期望、净生殖率、世代时间和总繁殖率均没有显著影响(P>0.05),但对种群内禀增长率(rm)影响显著 (P<0.05).与空白对照组相比,浓度为8、16和40 μg·L-1的DDT显著地提高了多刺裸腹溞种群rm.在使用多刺裸腹溞的生命表统计学参数监测亚致死浓度的DDT的生态影响时,rm是较敏感的指标.  相似文献

通过开顶式气室(OTCs)研究了OTC对照(自然CO2浓度约342 μmol·mol-1,O3浓度约30 nmol·mol-1)、高浓度CO2(550 μmol·mol-1)、高浓度O3(浓度为80 nmol·mol-1)及其交互作用(CO2 550 μmol ·mol-1,O3 80 nmol·mol-1)对春小麦不同发育时期生物量、总酚量、黄酮含量及成熟期产量性状的影响.结果表明:CO2浓度增加条件下,春小麦生物量和产量性状都显著高于OTC对照(P<0.05);而O3浓度升高条件下,小麦生物量降低,株高、穗长、穗粒质量及千粒重也显著低于对照;CO2和O3交互作用下各项指标处于二者之间.说明CO2可以缓解O3对小麦的负效应,而O3对CO2的正效应具有削弱作用,但二者的作用并非简单的叠加.CO2、O3浓度增加及其交互作用显著增加了春小麦叶片中的总酚含量,其中两者交互作用的效应更大,但在小麦生长后期,总酚含量增加量比对照有所降低.在小麦生长前期,各处理总黄酮含量均低于对照;而在成熟期,各处理都显著高于对照.  相似文献

施磷对稻田土壤及田面水磷浓度影响的模拟   总被引:15,自引:0,他引:15  
通过施用不同剂量磷肥稻田土壤淹水培养试验,研究了施磷对稻田土壤及田面水磷浓度的影响.结果表明,土壤速效磷(Olsen-P)浓度在施磷后迅速下降,60 d后趋于稳定.随施磷量的增加,土壤速效磷和缓效磷库量均递增,Olsen-P与施磷量呈正相关关系(y=21.49+0.086x),表明该土壤有很高的固磷潜力.施磷后田面水中全磷浓度呈先迅速上升后又缓慢下降趋势,施磷120 d后,田面水中全磷浓度与施磷量呈指数相关关系(y=0.3 72e0.0022x),施磷量在400~800 kg·hm-2之间田面水全磷浓度加速增长,如果施磷量达到或超过800 kg·hm-2,则磷容易进入田面水并导致流失,低于该施磷量时,则磷进入田面水中的量较少.利用分段回归模型模拟土壤Olsen-P与水面全磷关系,预测出导致田面水中磷激增的土壤Olsen-P浓度“突变点”为82.7 mg·kg-1,即施磷量为712 kg·hm-2.因此,土壤Olsen-P浓度可作为预测田面水中磷损失程度的指标.  相似文献

磷尾矿土壤中解磷细菌的筛选及解磷能力的测定   总被引:2,自引:0,他引:2  
以贵州瓮福磷尾矿土壤为原料,从中分离纯化得到具有较高解磷能力的细菌。利用溶磷圈试验筛选出具有明显溶磷圈的细菌,通过形态学特征、生理生化试验、16S rDNA基因序列分析及系统发育树对其进行初步鉴定。再以磷酸三钙为唯一磷源对筛选所得菌株进行液态培养,探索不同菌株解磷能力与培养液pH值之间的关系,并通过钼锑抗比色法测定3株细菌的最大解磷能力。从分离纯化得到的4株细菌筛选出具有明显溶磷圈的细菌PSB1、PSB3和PSB4,初步鉴定菌株PSB1为普城沙雷氏菌(Serratia plymuthica),PSB3为嗜麦芽窄食单胞菌(Stenotrophomonas maltophilia),PSB4为泡囊短波单胞菌(Brevundimonas vesicularis),最大解磷能力分别为148.87μg/mL、153.84μg/mL和146.76μg/mL。与国内已报道的文献相比,实验筛选所得的3株细菌都是具有较高解磷能力的菌种。菌株PSB1和PSB4培养液中pH值与其解磷能力存在显著的负相关性,而菌株PSB3不存在,其解磷机理还需进一步研究。  相似文献

通过原位进行了对照(CK)、低氮(LN,50kgN.hm-2.a-1)、中氮(MN,100kgN.hm-2.a-1)和高氮(HN,150kgN.hm-2.a-1)处理,研究了川西南天然常绿阔叶林凋落物分解及养分释放对模拟N沉降的响应.结果表明:凋落物分解95%需要4.72~6.33年,分解率最高的为CK,最低的为HN.经过365d,各处理的分解率均低于CK,仅HN与CK间差异显著(P<0.05);C残留率均高于CK;N和K残留率均显著高于CK(P<0.05);P残留率均高于CK,仅LN与CK间差异显著(P<0.05).各处理凋落物的C/N升高3.9%~23.7%.凋落物分解过程中N元素的迁移模式为富集-释放,C、P和K元素则表现为直接释放.N沉降对凋落物中养分元素的释放及木质素和纤维素的降解均具有抑制作用.随着处理时间的延长,N沉降对川西南常绿阔叶林凋落物分解的影响从正效应转向负效应,且负效应随沉降浓度的增加而加强.  相似文献

The quantitative analysis of laboratory grown organisms (Plectonema boryanum and Staphylococcus aureus) revealed that a typical in vivo polyphosphate body (PPB) contains O (4.3 x 10(-8) microg), C (1.2 x 10(-8) microg), P (6.7 x 10(-9) microg), Mg (1.3 x 10(-9) microg), Ca (6.7 x 10(-10) microg), K (6.7 x 10(-10) microg), Fe (6.0 x 10(-10) microg), S (5.4 x 10(-10) microg) and Al (5.9 x 10(-10) microg). Quantitative X-ray analysis of samples from nature showed PPB contain O (1.63 x 10(-8) microg), C (4.75 x 10(-9) microg), P (2.50 x 10(-9) microg), Mg (5.0 x 10(-10) microg), Ca (2.50 x 10(-10) microg), K (2.50 x 10(-10) microg), Fe (2.25 x 10(-10) microg) and S (2.0 x 10(-10) microg). The mass of an average polyphosphate body was 6.7 x 10(-8) microg for P. boryanum, 2.5 x 10(-8) microg for S. aureus and for microbes from the natural environment 6.3 x 10(-8) microg. The results indicate that the PPB may have other unknown functions in addition to essential element storage, acting as a detoxification method by sequestering heavy metals and providing a homeostasis system in the cell.  相似文献

In the research, the single-and dual effects of phosphate-solubilizing bacteria (PSB) (B0, Pseudomonas sp. FA1, and Bacillus simplex UT1) and silicon (Si) (0, 150, 300, and 600 mg kg−1 used as silicic acid) on P uptake by sorghum (Sorghum bicolor L.) plant fertilized with soluble or insoluble P (rock phosphate—RP) were studied via a perlite-potted experiment. Moreover, the effects of various treatments on morphological (shoot and root dry weight), nutritional (the uptake of Si and K) and physiological parameters (activity of catalase, superoxide dismutase, and peroxidase enzymes) of this plant were also measured. When grown in RP-fertilized medium compared with those grown in soluble P-fertilized medium, both shoot biomass and root biomass of sorghum plants were noticeably diminished. The PSB strains and Si levels independently improved all the aforementioned parameters. Use of Si and PSB strains to sorghum plants grown in soluble P or insoluble P medium significantly augmented P use efficiency. Silicon not only augmented the uptake of P from sparingly soluble-P source (RP), but also augmented uptake of P from water-soluble P source. Both B. simplex UT1 and Pseudomonas sp. FA1 indicated a meaningful betterment in sorghum plant dry matter and uptake of P (and K and Si) under both soluble and insoluble P fertilization conditions with Pseudomonas sp. FA1 being more efficacious than B. simplex UT1. But, the dual use of the PSB with Si resulted in the greatest increase in sorghum plant P uptake and other measured growth indices. Application of 600 mg Si kg−1 and Pseudomonas sp. FA1 significantly augmented the P shoot concentration of sorghum plant fertilized with RP to an sufficient level (> 0.3%) in the range of P-fertilized sorghum plants. Therefore, in addition to PSB utilization, Si should be considered as soil amendment in agricultural soils inadequate in plant-available Si as a means of sustainable agriculture with respect to possible savings of scarce P resources.

Biological activity of alloferon (AF), peptide, consisting of 13 a. a. isolated from hemolymph of experimentally infected blow fly Calliphora vicina was studied. AF in concentrations form 1 x 10(-5) to 250 microg/ml was added into the culture medium of the target cell lines K562, J-96, P388DI, Hep22a and 3T3B-SV40. First two days the peptide in concentrations 1 x 10(-5) and 1 x 10(-3) microg/ml in most cases stimulated the cell proliferative activity and suppressed the cell growth when applied in concentrations 10 and 100 microg/ml. Trend in growth modulating effect was dependent on duration of AF treatment. The peptide did not expressed cytotoxic effect with the exception of destruction of P388D1 cells that was registered after 96 h incubation in the medium initially contained 100 microg/ml AF. Simultaneously, cytotoxic and growth modulating effects of doxorubicin and cytosinarabinoside, as well as hybrid molecules, AF--cytosinarabinoside (cytal) and AF--doxorubicin (doxal) have been studied. Doxorubicin and cytosinarabinoside expressed greater growth inhibition effect compared to the hybrid molecules and AF itself. The results obtained with mass cell cultures were supported by experiments where P388D1 cells clonogenic capacity was tested. Besides, it has been demonstrated that AF rapidly penetrates into cytoplasm of J-96 cells and concentrates mainly into and around the cell nuclei.  相似文献

We examined gender differences in growth hormone (GH) secretion during rest and exercise. Eighteen subjects (9 women and 9 men) were tested on two occasions each [resting condition (R) and exercise condition (Ex)]. Blood was sampled at 10-min intervals from 0600 to 1200 and was assayed for GH by chemiluminescence. At R, women had a 3.69-fold greater mean calculated mass of GH secreted per burst compared with men (5.4 +/- 1.0 vs. 1.7 +/- 0.4 microg/l, respectively) and higher basal (interpulse) GH secretion rates, which resulted in greater GH production rates and serum GH area under the curve (AUC; 1,107 +/- 194 vs. 595 +/- 146 microg x l(-1) x min, women vs. men; P = 0.04). Compared with R, Ex resulted in greater mean mass of GH secreted per burst, greater mean GH secretory burst amplitude, and greater GH AUC (1,196 +/- 211 vs. 506 +/- 90 microg x l(-1) x min, Ex vs. R, respectively; P < 0.001). During Ex, women attained maximal serum GH concentrations significantly earlier than men (24 vs. 32 min after initiation of Ex, respectively; P = 0.004). Despite this temporal disparity, both genders had similar maximal serum GH concentrations. The change in AUC (adjusted for unequal baselines) was similar for men and women (593 +/- 201 vs. 811 +/- 268 microg x l(-1) x min), but there were significant gender-by-condition interactive effects on GH secretory burst mass, pulsatile GH production rate, and maximal serum GH concentration. We conclude that, although women exhibit greater absolute GH secretion rates than men both at rest and during exercise, exercise evokes a similar incremental GH response in men and women. Thus the magnitude of the incremental secretory GH response is not gender dependent.  相似文献

通过人工冰冻和电导率的测定,对黄土丘陵沟壑区引种载培的美国黄松、班克松和乡土树种油松的抗寒性进行了鉴定,并探讨其抗寒机理.结果表明,班克松的抗寒性比油松强,而美国黄松的抗寒性比油松稍弱.班克松的束缚水/自由水比值高达7.0,组织中ABA含量高达164.3 μg·g-1 FW;但可溶性糖和K+含量较低,分别为12.0%和2 450 μg·g-1 DW.油松则是可溶性糖、K+和ABA含量都较高,分别为18.68%、4 538 μg·g-1 DW和95.8 μg·g-1 FW;束缚水/自由水比值较低,为2.58.美国黄松的可溶性糖含量较高,18.05%;但束缚水/自由水比值、K+和ABA含量都较低,分别为2.18、2 275 μg·g-1 DW和63.3 μg·g-1 FW,可能是其抗寒性较弱的内在原因.班克松较低的叶绿素含量和较高的类胡萝卜素/叶绿素比值对其抗寒性也有贡献.说明3种树种虽然都是抗寒树种,但其内在机理仍有差异.  相似文献

In the adult animal, ANG-(1-7) may counterbalance some effects of ANG II. Its effects in the fetus are unknown. Basal ANG-(1-7), ANG I, ANG II, and renin concentrations were measured in plasma from ovine fetuses and their mothers (n = 10) at 111 days of gestation. In the fetus, concentrations of ANG I, ANG-(1-7), and ANG II were 86 +/- 21, 13 +/- 2, and 14 +/- 2 fmol/ml, respectively. In the ewe, concentrations of ANG I were significantly lower (20 +/- 4 fmol/ml, P < 0.05) as were concentrations of ANG-(1-7) (2.9 +/- 0.6 fmol/ml), whereas ANG II concentrations were not different (10 +/- 1 fmol/ml). Plasma renin concentrations were higher in the fetus (4.8 +/- 1.1 pmol ANG I x ml(-1) x h(-1)) than in the ewe (0.9 +/- 0.2 pmol x ml(-1) x h(-1), P < 0.05). Infusion of ANG-(1-7) (approximately 9 microg/h) for a 3-day period caused a significant increase in plasma concentrations of ANG-(1-7) reaching a maximum of 448 +/- 146 fmol/ml on day 3 of infusion. Plasma levels of ANG I and II as well as renin were unchanged by the infusion. Urine flow rate, glomerular filtration rate, and fetal arterial blood pressure did not change and were not different than values in fetuses receiving a saline infusion for 3 days (n = 5). However, the osmolality of amniotic and allantoic fluid was significantly higher in fetuses that received ANG-(1-7). Also, compared with the saline-infused animals, mRNA expression levels of renin, the AT(1) receptor, and AT(2) receptor were elevated in kidneys of fetuses that received infusions of ANG-(1-7). Infusion of an ANG-(1-7) antagonist ([D-Ala(7)]-ANG-(1-7), 20 microg/h) for 3 days had no effect on fetal blood pressure or renal function. In conclusion, although infusion of ANG-(1-7) did not affect fetal urine flow rate, glomerular filtration rate, or blood pressure, changes in fetal fluids and gene expression indicate that ANG-(1-7) may play a role in the fetal kidney.  相似文献

采用室内模拟方法,研究了重金属镉对黄颡鱼[Pelteobagrus fulvidraco (Richardson)]鳃线粒体结构和能量代谢的影响及其作用机理.结果表明,50 μg·L-1组黄颡鱼鳃线粒体结构未受损,且各项测定指标与对照组无显著差异(P>0.05);500 μg·L-1组黄颡鱼鳃线粒体严重受损,除LD、MDA、ADP、AMP和血浆K+含量显著高于对照组外,其余检测指标均显著低于对照组(P<0.05).高浓度镉短期暴露将降低鱼鳃线粒体SOD活性而导致线粒体氧化损伤,同时抑制PFK和ATP酶活性, 影响鳃的能量供应和利用,最终降低鳃血浆渗透压和离子浓度调节能力可能是其毒性机理之一.  相似文献
