The occurrence and distribution of Dictyostelid cellular slime molds in southern California were investigated by means of a clonal isolation technique that permitted quantitative sampling of populations. Thirteen species were isolated. These include (in approximate order of decreasing frequency): Dictyostelium rosarium Raper and Cavender, D. mucoroides Brefeld, D. sphaerocephalum (Oud.) Sacc. and March., D. aureum Olive, Polysphondylium pallidum Olive, D. minutum Raper, D. giganteum Singh, D. purpureum Olive, P. violaceum Brefeld, D. mucoroides variant, Acytostelium leptosomum Raper and Quinlan, D. polycephalum Raper, and a new species of Dictyostelium. D. rosarium and D. aureum, both rarely reported in the literature, were common and widespread. Trends in habitat preference of the Dictyostelids encountered are discussed. Selected taxonomic details are provided for D. rosarium, D. aureum and members of the “D. mucoroides complex“—D. mucoroides, D. sphaerocephalum, D. giganteum, and a D. mucoroides variant.  相似文献

Sixteen species of cellular slime molds were isolated from Southeast Asian forest soils. Ten of these, Dictyostelium mucoroides Brefeld, D. purpureum Olive, D. polycephalum Raper, D. lacteum van Tieghem, D. rhizopodium Raper and Fennell, D. lavandulum Raper and Fennell, D. vinaceo-fuscum Raper and Fennell, D. coeruleo-stipes Raper and Fennell, Polysphondylium violaceum Brefeld, and P. pallidum Olive have been previously described and are well-recognized species occurring in other parts of the world. Two, one in the genus Dictyostelium and one belonging to the family Guttulinaceae, are considered species by the author but have not been formally described. Four are described in this paper as new: Dictyostelium intermedium, D. multi-stipes, D. bifurcatum, and Acytostelium subglobosum. A new variety papilloideum of D. lacteum is also described. One other discovery of special interest is an isolate of Polysphondylium violaceum which produces abundant macrocysts, now known to be the sexual stage in the life cycle of cellular slime molds.  相似文献

The distribution of eleven dictyostelid cellular slime molds found in Norway was studied. They were divided into four groups according to their dominance. In decreasing order the groups were: (1) two varieties of Dictyostelium mucoroides , (2) D. aureostipes and Polysphondylium violaceum , (3) D. fasciculatum, D. minutum, P. pallidum and Dictyostelium sp. 1, and (4) Dictyostelium sp. 2 and 3 and Acytostelium lep–tosomum. Differences in distribution related to altitudes and latitudes, climates and dominant tree species were observed for some species.  相似文献

Discadenine,3-(3-amino-3-carboxypropyl)-N6-delta 2-isopentenyladenine, which inhibits spore germination, was previously found in Dictyostelium discoideum. Studies on the distribution of discadenine in different species of cellular slime molds by high-pressure liquid chromatography showed that discadenine is present in D. discoideum, Dictyostelium purpureum, and Dictyostelium mucoroides, but not in Dictyostelium minutum, Polysphondylium violaceum, or Polysphondylium pallidum. Discadenine synthetase, which is involved in biosynthesis of discadenine with N6-delta 2-isopentenyladenine as substrate, was only detected in cells of the former three species. In addition, discadenine inhibited spore germination only in these three species. These results clearly demonstrate that discadenine is produced as an inhibitor of spore germination in the species of cellular slime molds in which the acrasin is cyclic adenosine 5'-monophosphate (AMP). This means that there is a structural and biochemical correlation between the spore germination inhibitor and the acrasin, since 5'-AMP, a direct precursor in discadenine biosynthesis, can be derived from cyclic AMP by hydrolysis with cyclic AMP phosphodiesterase.  相似文献

Abstract: A skull and mandible of the new species Dicerorhinus gwebinensis sp. nov. of Rhinocerotidae (Mammalia, Perissodactyla) is described. The material is collected from the upper part of the Irrawaddy sediments (Plio‐Pleistocene) in central Myanmar. D. gwebinensis sp. nov. is morphologically more similar to the extant species D. sumatrensis (Sumatran rhinoceros) than to other species of the genus but differs from D. sumatrensis in having the comparatively shorter nasal, the more concave dorsal profile of the skull, the more elevated occiput and presence of molar crista in M3/. This is the first discovery of Dicerorhinus in the upper Miocene to lower Pleistocene of the Indian subcontinent and Mainland Southeast Asia, and fills the chronological and geographical gap of this lineage in Asia. The Dicerorhinus clade probably migrated into Southeast Asia from East Asia by the Pliocene or early Pleistocene. This hypothesis is supported by the scarcity or absence of this clade in the Neogene mammalian fauna of the Indian Subcontinent.  相似文献

Dictyostelid cellular slime molds (dictyostelids) associated with grassland ecosystems of the central and western United States were investigated at nine sites that included examples of the three major ecological types of grasslands (tall grass, mixed grass and short grass) generally recognized for the region. Samples of soil/humus collected from each site were examined with the Cavender method of isolating dictyostelids. For each of those six sites with well developed gallery forests present, an additional set of forest soil/humus samples was collected. A more intensive sampling effort was carried out at one site (Konza LTER) to assess the possible effects of burning and grazing on dictyostelid diversity and density. Twelve species of dictyostelids were recovered from grassland sites, whereas gallery forest sites yielded only nine species. Four cosmopolitan species (Dictyostelium giganteum, D. mucoroides, D. sphaerocephalum and Polysphondylium pallidum) were represented by the greatest densities of clones, with D. sphaerocephalum particularly common. The general pattern across all sites was that both species richness and density of dictyostelids decreased with decreasing precipitation. Samples collected from ungrazed grassland plots yielded higher numbers of both species and clones as compared to grazed plots, and the general pattern was for both values to increase as the interval between fires increased. For numbers of clones this correlation was statistically significant.  相似文献

Aggregative amoebae of Dictyostelium discoideum, D. mucoroides, D. purpureum, and D. rosarium react chemotactically to cyclic AMP (cAMP). We measured the chemotactic activity of 14 cAMP analogs and found that these four species have a similar sensitivity to chemical modifications of cAMP; this suggests that the cAMP receptor is identical in all of these species. Besides the induction of a chemotactic response, cAMP analogs also may delay or prevent cell aggregation. cAMP analogs like N1-O-cAMP, 2'-H-cAMP, and 5'NH-cAMP are chemotactically nearly as active as cAMP and induced no, or only a short, delay of cell aggregation. Other cAMP derivatives, such as 6-Cl-cPMP and 8-Br-cAMP, are chemotactically active only at high concentrations and delayed cell aggregation for several hours. Still other cAMP analogs, which do not induce a chemotactic reaction in D. mucoroides, D. purpureum, and D. rosarium, either prevented cell aggregation [cAMPS(S), cAMPS(R), and 3'-NH-cAMP[ or had no effect on cell aggregation [cAMPN(CH3)2(S) and cAMPN(CH3)2(R)]. cAMP analog 3'-NH-cAMP prevented cell aggregation by the inhibition of chemotaxis, whereas cell locomotion was not affected. Although we cannot provide a satisfactory explantation for these observations, our data suggest that occupation and activation of the cAMP receptors do not always induced a chemotactic response.  相似文献

Previous studies have shown that Dictyostelium discoideum spore coat proteins are found in prespore cells, which are localized to the posterior region of migrating slugs, and in the coats of mature spores. Prespore vesicles, identified by morphology and by staining with anti-D. mucoroides spore serum, are also localized in the posterior region of migrating slugs. Using antisera specific to the spore coat proteins, we show that the spore coat proteins are packaged in prespore vesicles. They are present in the vesicles as a complex which can be dissociated by denaturation. The anti-D. mucoroides spore serum reacts with at least five proteins in whole spore extracts including the spore coat proteins SP96 and SP70.  相似文献

Spores of all strains of Dictyostelium discoideum tested in this study germinated after a heat shock of 45 C for 30 min. Whereas the strains differed in their rates of germination, the rate for each strain was constant. A correlation existed between the rate of germination and the rate of vegetative growth when spores were inoculated into bacterial streaks. Heat shock clearly increased spore germination in D. purpureum, but the response was less dramatic than in D. discoideum. Enhancement also occurred in D. rosarium, but only in media containing peptone. Strains of D. mucoroides gave varied responses, and these could be divided into those which required mutrients for spore germination and those which did not. The spores of Polysphondylium pallidum were resistant to mild heat (45 C), but were not activated; peptone was required for germination. In contrast, the microcysts of this species were heat-labile and required no added nutrients for excystment.  相似文献

Hydrophilic spore proteins were extracted from Nosema sp. and Nosema trichoplusiae. These proteins were subjected to electrophoretic analysis. The resulting electrophoretic spectra were found to be unstable when (1) two genera of hosts were used for spore propagation, (2) hosts were reared at a variety of temperatures, (3) protein was extracted from spores stored for different periods of time, or (4) spore incubation period was varied. Comparison of the major bands obtained from spore protein of the isolates indicated no overlap in relative migration values. Although variation in spectra was observed, the use of major band patterns indicate electrophoretic analysis of hydrophilic spore protein can provide characters useful in the separation and identification of microsporidan isolates. Nosema sp. and Nosema trichoplusiae are not considered to be closely related phylogenetically.  相似文献

Some Syrian hamster cell lines persistently infected with lymphocytic choriomeningitis virus (LCMV) do not produce extracellular virus particles but do contain intracytoplasmic infectious material. The proteins of these cells were labeled with [35S]methionine or with [3H]glucosamine and [3H]mannose, and immunoprecipitates were prepared with anti-LCMV sera. A substantial amount of the LCMV nucleocapsid protein (molecular weight about 58,000) was detected, along with GP-C, the precursor of the virion glycoproteins GP-1 and GP-2. GP-1 and GP-2 themselves were not detected. A new method of transferring proteins electrophoretically from sodium dodecyl sulfate-polyacrylamide gels to diazotized paper in high yield revealed several additional LCMV proteins present specifically in the persistently infected cells, at apparent molecular weights (X10(3] of 112, 107, 103, 89, 71 (probably GP-C), 58 (nucleocapsid protein), 42 to 47 (probably GP-1), and 40 (possibly GP-2). By iodinating intact cells with I3, GP-1 but not GP-2 or GP-C was revealed on the surfaces of the persistently infected cells, whereas both GP-1 and GP-C were found on the surfaces of acutely infected cells. The absence of GP-C from the plasma membrane of the persistently infected cells might be related to defective maturation of the virus in these cells. Cytoplasmic viral nucleoprotein complexes were labeled with [3H]uridine in the presence or absence of actinomycin D, purified partially by sedimentation in D2O-sucrose gradients, and adsorbed to fixed Staphylococus aureus cells in the presence of anti-LCMV immunoglobulin G. Several discrete species of viral RNA were released from the immune complexes with sodium dodecyl sulfate. Some were appreciably smaller than the 31S and 23S species of standard LCMV virions, indicating that defective interfering viral RNAs are probably present in the persistently infected cells. Ribosomal 28S and 18S RNAs, labeled only in the absence of actinomycin D, were coprecipitated with anti-LCMV serum but not with control serum, indicating their association with LCMV nucleoproteins in the cells.  相似文献

Ganoderma sp. is an airborne fungal spore type known to trigger respiratory allergy symptoms in sensitive patients. Aiming to reduce the risk for allergic individuals, we analysed fungal spore circulation in Szczecin, Poland, and its dependence on meteorological conditions. Statistical models for the airborne spore concentrations of Ganoderma sp.—one of the most abundant fungal taxa in the area—were developed. Aerobiological sampling was conducted over 2004–2008 using a volumetric Lanzoni trap. Simultaneously, the following meteorological parameters were recorded: daily level of precipitation, maximum and average wind speed, relative humidity and maximum, minimum, average and dew point temperatures. These data were used as the explaining variables. Due to the non-linearity and non-normality of the data set, the applied modelling techniques were artificial neural networks (ANN) and mutlivariate regression trees (MRT). The obtained classification and MRT models predicted threshold conditions above which Ganoderma sp. appeared in the air. It turned out that dew point temperature was the main factor influencing the presence or absence of Ganoderma sp. spores. Further analysis of spore seasons revealed that the airborne fungal spore concentration depended only slightly on meteorological factors.  相似文献

Cellular slime molds are one of only three types of eukaryotes known to contain circular nuclear plasmids. Unlike the 2-microns circle in Saccharomyces, different strains of Dictyostelium can carry different, nonhomologous plasmids. Covalently closed, circular DNA plasmids have been identified in D. discoideum, D. mucoroides, D. giganteum, and D. purpureum. These plasmids range in size from 1.3-27 kb and in copy number from 50-300 molecules per cell. Plasmids have been identified in approximately one-fifth of all isolates examined. The organization of their DNA in nucleosomes establishes their presence in the nucleus. We have successfully cotransformed endogenous Dictyostelium plasmids into D. discoideum using the G418 resistance shuttle vector B10S. Transformants carrying D. discoideum plasmids are recovered at much higher frequency than those carrying plasmids from the other Dictyostelium species. We have constructed recombinant plasmids based on the D. discoideum plasmid Ddp2 and the G418 resistance gene. With these extrachromosomal vectors, transformed cells are recovered at frequencies of up to 10(-4) per input cell, the vectors are stably maintained at high copy number in the absence of selection, and the vectors can be used to introduce foreign DNA sequences into D. discoideum cells.  相似文献

A Burkard spore trap was used to monitor daily fungal spore counts in the atmosphere of Melbourne (Australia) between October 1991 and December 1994. Annual total spore counts varied widely (range 345 770 in 1994 to 1 106 037 in 1992), of which approximately half were identified asCladosporium sp. and only about 1%Alternaria sp. Highest daily total spore counts were recorded late-summer through to mid-winter, probably corresponding to senescence of annual grasses and leaf fall. Spore counts were negatively correlated with rainfall (P<0.05), significantly correlated with average temperature (P<0.001), and showed a highly significant linear relationship with cumulative temperature throughout the year.  相似文献

The fine structure of the mature spore of a haplosporidan found in the mussel Mytilus galloprovincialis L. is described. The mature spore (10 × 5 µm) is operculate and characterised by the presence of epispore cytoplasm that is prolongated into two bipolar extensions or 'tails' of 80–100 µm in length. The extensions are supported by bundles of microtubule-like structures which are not found in continuity with the wall. Due to the presence of episporal extensions and because the present species differs in some morphological characters from other haplosporidans assigned to the genus Minchinia Labbé, the parasite of the mussel has been placed in this genus as Minchinia sp.  相似文献

Over hundred specimens of Desmoscolex were found in samples from Yonge Reef, Lizard Island and Nymphe Island (Great Barrier Reef, Australia). In the present paper four new species are described: D. brevisetosus sp.n., characterised by the absence of subventral setae on the 2nd main ring and the presence of short subdorsal setae on the 16th main ring: D. dimorphus sp.n. characterized by the presence of sexual dimorphism (absence of sub-ventral setae on the 14th main ring, short subdorsal setae on the 16th main ring and smaller amphids in females); D. geraerti sp.n. and D. nymphianus sp.n. with the typical setal pattern and elongated laterally shifted subdorsal setae on the 13th, 16th and 17th main ring but the former species characterized by large bipartite amphids, cephalated spicules and trough-shaped gubernaculum; the latter species by the smaller rounded amphids with their posterior margin coinciding with that of the amphidial pore and by the short straight spicules.  相似文献

The genus Calyptogena (Bivalvia: Vesicomyidae) comprises highlyspecialized bivalves living in symbiosis with sulphur-oxidizingbacteria in reducing habitats. In this study, the genus is revisedusing shell and anatomical features. The work is based on typematerial, as well as on the extensive collection of vesicomyidsobtained during twelve expeditions to the Pacific and IndianOceans. Nine Recent species are ascribed to the genus Calyptogena,four of which are new: C. pacifica Dall, 1891, C. fausta Okutani,Fujikura & Hashimoto, 1993, C. rectimargo Scarlato, 1981,C. valdiviae (Thiele & Jaeckel, 1931), C. gallardoi Sellanes& Krylova, 2005, C. goffrediae n. sp., C. starobogatovin. sp., C. makranensis n. sp. and C. costaricana n. sp. Thecharacteristic features of Calyptogena are: shell up to 90 mmin length, elongate-elliptical or elongate; presence of escutcheon;presence of broad posterior ramus (3b) of right subumbonal cardinaltooth as well as right posterior nymphal ridge; absence of pallialsinus as a result of attachment of intersiphonal septal retractorimmediately adjacent to ventral surface of posterior adductor;absence of processes on inner vulva of inhalant siphon; presenceof inner demibranch only, with descending and ascending lamellaewith interlamellar septa not divided into separate tubes. Themost closely related taxa to Calyptogena are probably the genusIsorropodon Sturany, 1896, and the group of species representedby ‘Calyptogenaphaseoliformis Métivier,Okutani & Ohta, 1986. These groups have several charactersin common, namely absence of pallial sinus, presence of singleinner pair of demibranchs and absence of processes on innervulva of inhalant siphon. The worldwide distribution of thegenus Calyptogena suggests that methane seeps at continentalmargins are the major dispersal routes and that speciation waspromoted by geographical isolation. Recent species diversityand fossil records indicate that the genus originated in thePacific Ocean. Sufficient data to discuss the distribution atspecies level exist only for C. pacifica, which has a remarkablynarrow bathymetric range. Published studies on the physiologyof C. pacifica suggest that adaptation to a specific geochemicalenvironment has led to coexisting vesicomyid genera. The bacteria-containinggill of C. pacifica and other Calyptogena species is one ofthe most specialized in the family Vesicomyidae and may reflectthese ecological adaptations. Diversity of arbuscular mycorrhizal fungi (AMF) in 27-year long-term NP-fertilization plots under a maize cropping system in Thailand was studied through spore morphological characterization. The plots received 0–0, 60–60, 120–120 and 180–180 kg N-P2O5 ha–1 year–1 as ammonium sulfate and triple superphosphate. The plots were sampled monthly for one year, the AMF spores were counted and morphotyped, and taxa were identified after morphotyping and monospecific pot culture. Spore number g–1 soil, relative spore abundance and Shannon-Wiener indexes were calculated. Sixteen putative taxa were recorded from the field of which nine sporulated on maize roots in pot culture. The long-term fertilization caused decreases in AMF total spore numbers and variation in species diversity depended on sampling time. Effects of fertilization on spore number and also relative spore abundance varied with species and sampling time. Among the nine species sporulating under maize, only Acaulospora sp.1 showed no change (P > 0.003 after Bonferroni correction) in spore number with fertilization in the field; and was therefore classified as an AMF species insensitive to fertilization. Spores of Entrophospora schenckii, Glomus mosseae, Glomus sp.1, Glomus geosporum-like and Scutellospora fulgida, though they decreased in absolute numbers in response to fertilization, showed no change (P > 0.003 after Bonferroni correction) in relative abundance; these species were classified as AMF species slightly sensitive to fertilization. Three unidentified species of Glomus, though they decreased in absolute numbers in response to fertilization, showed decreases (P < 0.003 after Bonferroni correction) in relative abundance; these species were classified as AMF species highly sensitive to fertilization.  相似文献

The present study was conceived to elucidate the potential importance of the D1 turnover-mediated repair mechanism in UV-B tolerance of the photosynthetic apparatus in microalgae. To this end, the lab-identified UV-B sensitive and tolerant species of Chlorophyte and Chromophyte algae was used to examine photosynthetic response to UV-B exposure in the presence vs. the absence of streptomycin, an inhibitor of chloroplast protein synthesis. Measurements of photosynthetic O2 evolution capacity and chlorophyll fluorescence parameters (Fv/Fm, ΦPSII) illustrated species-specific UV-B sensitivity of the photosynthetic apparatus. Addition of the inhibitor streptomycin caused significant enhancements of UV-B-caused depression of photosynthesis in UV-B tolerant species, while little effect was observed in the sensitive species. In the tolerant species, recovery from UV-B induced 20 percnt; decline in Fv/Fm reached completion within 2 hours, much faster than that in the sensitive species. Immunoblotting revealed that exposure to UV-B radiation caused substantial degradation of the D1 protein in the sensitive Heterococcus brevicellularis, which was little enhanced by addition of the inhibitor. The same UV-B exposure lead to less D1 degradation in the tolerant Scenedesmus sp., which was significantly enhanced by addition of the inhibitor. This study shows that UV-B tolerance of the photosynthetic apparatus in microalgae was associated with a strong capacity for recovery from the UV-B-induced damage and this capacity related to the D1 turnover-mediated repair cycle, and largely determined UV-B tolerance of the photosynthetic apparatus in these organisms.  相似文献
