A new theoretical model is presented which describes dynamics of interaction between the plasmic membrane, cell cortex and cytoplasmic liquid. Analysis of the equation system describing this interaction has shown that besides the uniform solution (corresponding to a flat cell surface) there exists the stable autowave solution (corresponding to standing or running harmonic waves along the cell surface). The latter solution can be related to the experimentally observed waves moving along the cell surface.  相似文献

The in depth-mining of the proteome necessitates the comprehensive analysis of proteins in individual subcellular compartments to uncover interesting patterns of protein expression that include assessment of protein location, trafficking and of post-translational modifications that are location specific. One of the compartments of substantial interest from a diagnostic and therapeutic point of view is the plasma membrane which contains intrinsic membrane proteins and other proteins expressed on the cell surface. Technologies are currently available for the comprehensive profiling of the cell surface proteome that rely on protein tagging of intact cells. Studies are emerging that point to unexpected patterns of expression of specific proteins on the cell surface, with a common occurrence of proteins previously considered to occur predominantly in other compartments, notably the endoplasmic reticulum. The profiling of the cell surface and plasma membrane proteomes will likely provide novel insights and uncover disease related alterations.  相似文献

Fertilization of eggs and effects of many growth factors on the membrane receptors of the cultured somatic cells induce similar changes in the plasma membrane transport properties, which determine changes in Ca2+ and H+ concentrations in the cytoplasm. The data are discussed which favour the concept that Ca2+ and H+ are secondary messengers of growth-stimulating influences and control many intracellular processes related to cell multiplication.  相似文献

A double mutant strain, Bacillus subtilis div IV B1 dal trp, has been constructed which grows as filamentous cells and minicells and requires d-alanine for growth. Removal of d-alanine from a growing population of cells resulted in cell bulging 25% of the cell length from one cell pole, followed by cell lysis. Microorganisms, or microbes, can function as threatening pathogens that cause disease in humans, animals, and plants; however, they also act as litter decomposers in natural ecosystems. As the outermost barrier and interface with the environment, the microbial cell surface is crucial for cell-to-cell communication and is a potential target of chemotherapeutic agents. Surface ultrastructures of microbial cells have typically been observed using scanning electron microscopy (SEM) and atomic force microscopy (AFM). Owing to its characteristics of low-temperature specimen preparation and superb resolution (down to 1 nm), cryo-field emission SEM has revealed paired rodlets, referred to as hydrophobins, on the cell walls of bacteria and fungi. Recent technological advances in AFM have enabled high-speed live cell imaging in liquid at the nanoscale level, leading to clear visualization of cell-drug interactions. Platinum-carbon replicas from freeze-fractured fungal spores have been observed using transmission electron microscopy, revealing hydrophobins with varying dimensions. In addition, AFM has been used to resolve bacteriophages in their free state and during infection of bacterial cells. Various microscopy techniques with enhanced spatial resolution, imaging speed, and versatile specimen preparation are being used to document cellular structures and events, thus addressing unanswered biological questions.  相似文献

The present study investigated the mechanism underlying alterations of cell surface sugar chains of Jurkat cells by inducing apoptosis with etoposide, an inhibitor of topoisomerase II. Within 3[emsp4 ]h of etoposide treatment, flowcytometric analysis revealed a decrease in Maackia amurensis agglutinin recognized 2,3-linked sialic acid moieties and an increase in Ricinus communis agglutinin recognized galactose. The results suggested that asialo-sugar chains on glycoconjugates were rapidly induced on the etoposide-treated cell surface. To clarify the desialylation mechanism, we studied 2,3-sialyltransferase mRNA expression and the activity of sialidase on the cell surface during etoposide-induced apoptosis. The expression of hST3Gal III and hST3Gal IV mRNAs were down-regulated and sialidase activity on the cell surface increased threefold within 2[emsp4 ]h of etoposide treatment. Moreover, the decrease in 2,3-linked sialic acid levels was significantly suppressed in the presence of 2,3-dehydro-2-deoxy-N-acetylneuraminic acid, an inhibitor of sialidase. These results suggested that activation or exposure of sialidase on the cell surface was induced by etoposide treatment and was the main cause of the decrease in sialic acids.  相似文献

Summary Acids like hydrogen fluoride, hydrazoic and fluoroacetic have been shown to prevent the germination of ascospores of N. tetrasperma when dormant spores are treated. On the other hand, propionate, cysteine and others are ineffective when used in this way. When activated ascospores were treated, much lower concentrations of the acids were sufficient to poison the spores. As in other systems, these substances are most effective at a pH below their pKa.The kinetics of uptake of fluoride by dormant ascospores were studied and shown to be very different from those reported for cations. However, P32 was not absorbed by dormant ascospores, even at pH 1.5.Respiratory inhibition by azide and fluoroacetate occurred immediately after the spores were activated, but in the case of 5-nitro-2-furfuryl methyl ether no effect was observed until just before germination occurred.These results suggest that a permeability barrier exists in the dormant ascospore which disappears upon germination. Intracellular antibodies (intrabodies) constitute a potent tool to neutralize the function of target proteins inside specific cell compartments (cytosol, nucleus, mitochondria and ER). The intrabody technology is an attractive alternative to the generation of gene-targeted knockout animals and complements or replaces knockdown techniques such as antisense-RNA, RNAi and RNA aptamers. This article focuses on intrabodies targeted to the ER. Intracellular anti-bodies expressed and retained inside the ER (ER intrabodies) are shown to be highly efficient in blocking the translocation of secreted and cell surface molecules from the ER to the cell surface.The advantage of ER intrabodies over cytoplasmic intrabodies is that they are correctly folded and easier to select. A particular advantage of the intrabody technology over existing ones is the possibility of inhibiting selectively post-translational modifications of proteins.The main applications of ER intrabodies so far have been (i) inactivation of oncogenic receptors and (ii) functional inhibition of virus envelope proteins and virus-receptor molecules on the surface of host cells.In cancer research, the number of in vivo mouse models for evaluation of the therapeutic potential of intrabodies is increasing.In the future, endosomal localized receptors involved in bacterial and viral infections, intracellular oncogenic receptors and enzymes involved in glycosylation of tumour antigens might be new targets for ER intrabodies.  相似文献
