The Iberian hare Lepus granatensis is a common and abundant species throughout the Iberian Peninsula; however, studies documenting its ecology are scarce. Between August 1996 and September 1997, after a long drought, we studied the effects of a widespread flood on the Iberian hare ecology in ecotone pastures of Doñana National Park. Hares were captured using vertical nets through part of 1996 and 1997. Fourteen adult hares (nine females and five males) were tagged with radiocollars and located by triangulation on a daily basis at least once a week. Habitat use was estimated using a geographical information system. The average home range size was 28 ha for males and 24 ha for females. No significant differences were observed between sexes. Significant differences were observed in the use of scrub areas and in dry pastures. During the dry and wet seasons, males and females were most often located in the pastures of the ecotone, rush stands and scrub. The dense bracken (Pteridium aquilinum) and seaside bulrush (Scirpus maritimus) communities were avoided. These results are the first on the spatial ecology of this species in its native habitat.  相似文献

A quantitative hydrogeomorphic approach was applied to an extensive survey of temporary wetlands in the Doñana National Park (SW Spain) in search for quantitative thresholds for wetland classification. Twenty freshwater ponds on the aeolian sand mantle and 46 sites on silty-clay substrate, located in the southern marshland, were surveyed during the heaviest rainy period so far recorded (October 1995–September 1997). On average, temporary ponds showed higher water depth, longer flooding period, lower conductivity (<0.5 mS cm?1), lower pH (6.7), lower phosphate concentration (0.4 μM) and a more balanced proportion of Ca2+/Na+ than temporary marshes. During floods, marshland sites exhibited higher water transparency, pH (9.5), alkalinity (3.5 meq l?1), conductivity (8.2 mS cm?1), phosphate concentration (0.7 μM), Na+ and Ca2+ concentrations (97.2 and 3.5 meq l?1, respectively) than ponds. Study sites were significantly segregated (ANOSIM test: R = 0.88, < 0.01, n = 92) in relation to water depth and conductivity. A conductivity of 1.6 mS cm?1 is proposed as a threshold between marshland sites and ponds during floods. Marshland sites were further segregated into two groups (ANOSIM test: R = 0.777, P < 0.01, n = 23) according to the Na+/Ca2+ ratio (in meq l?1) at a threshold value of 25. An ordination by PCA showed that five variables grouped 81.4% of the total variance in two axes. The first PCA axis (60.7% of variance) separated temporary wetlands into ponds and marshland sites according to variables related to substrate and hydrology (Na+/Ca2+ ratio, conductivity, water depth and flooding period). Other variables (e.g., water transparency, alkalinity, pH, submersed macrophyte biomass, phosphate, nitrate and planktonic chlorophyll concentrations) did not produce a significant segregation between marshland and pond sites during floods. Further discrimination within each wetland type was thus not achieved.  相似文献

Due to the invasive character of the exotic Argentine ant (Linepithema humile), its use of aphids in trees, and the ecological importance of the Doñana National Park (Spain) that is invaded by this species, we designed a study to analyze the extent of the problem with native species of arboreal ants. By searching for de visu the species that inhabited 182 cork trees, we found out that the structure of the community of native arboreal ants has been greatly influenced by interspecific competition. The introduced species L. humile and the native species Crematogaster scutellaris and Lasius brunneus are dominant, while Camponotus lateralis and Camponotus truncatus are subordinate species associated with C. scutellaris. The distribution of the species in the trees depends on these relationships. Species richness is determined by tree size, thus, when a tree is large enough all native species may appear together. However, in areas colonized by L. humile, this is the only species occupying the tree, regardless of tree size. L. humile is displacing native arboreal ant species as shown by the fact that from 1992 to 2000 the exotic species occupied 23 new cork trees (of the 182 studied) previously inhabited by native species.  相似文献

Changes in the vegetation of stabilized sand dunes of Doñana National Park in the last four centuries have been analysed, combining ecological and historical sources of information.Quantitative vegetation data have been obtained from 5 inventories of a 15 km long transect, consisting of 34 points where nearby woody plants have been recorded. Inventories were taken in 1636, 1647, 1652, 1682 and 1859. The analysis of these data, combined with climatic and management information on the area, reveals that until 1636 a mediterranean forest dominated byQuercus suber, existed; it was used as a game preserve. From 1636 to 1682 vegetation underwent profound changes with a marked drop in trees and mature scrub. The vegetation was replaced by a pioneer scrub species. This rapid change was brought about by the introduction of cattle and pigs, timber and cork exploitation, and the use of prescribed fire. The climatic crisis that arose circa 1700 led to erosion of the sandy soils, which made the vegetation change irreversibly. Livestock pressure hampered natural forest regeneration until 1737 whenPinus pinea plantations started. From then onwards little change in vegetation is known to have occurred.It is suggested that the main steps of change in Doñana vegetation history may have occurred in many other areas of the Mediterranean Basin.  相似文献

Different lines of cell suspension cultures of Taxus × media Rehd. and Taxus floridana Nutt. were cryopreserved with a two-step freezing method using a simple and inexpensive freezing container instead of a programmable freezer. Four to seven days old suspension cell cultures were precultured in growth medium supplemented with 0.5 M mannitol for 2 d. The medium was then replaced with cryoprotectant solution (1 M sucrose, 0.5 M glycerol and 0.5 M dimethylsulfoxide) and the cells incubated on ice for 1 h. Before being plunged into liquid nitrogen, cells were frozen with a cooling rate of approximately −1 °C per min to −80 °C. The highest post-thaw cell viability was 90 %. The recovery was line dependent. The cryopreservation procedure did not alter the nuclear DNA content of the cell lines. The results indicate that cryopreservation of Taxus cell suspension cultures using inexpensive freezing container is possible.  相似文献

The presence of β-lactamases in Y. enterocolitica has been reported to vary with serovars, biovars and geographical origin of the isolates. An understanding of the β-lactamases in other related species is important for an overall perception of antibiotic resistance in yersiniae. Manufacture of nitroorganic explosives generates toxic wastes leading to contamination of soils and waters, especially groundwater. For that reason bacteria living in environments highly contaminated with 2,4,6-trinitrotoluene (TNT) and other nitroorganic compounds were investigated for their capacity for TNT degradation. One isolate, Raoultella terrigena strain HB, removed TNT at concentrations between 10 and 100 mg l−1 completely from culture supernatants under optimum aerobic conditions within several hours. Only low concentrations of nutrient supplements were needed for the cometabolic transformation process. Radioactivity measurements with ring-labelled 14C–TNT detected about 10–20% of the initial radioactivity in the culture supernatant and the residual 80–90% as water-insoluble organic compounds in the cellular pellet. HPLC analysis identified aminodinitrotoluenes (2-ADNT, 4-ADNT) and diaminonitrotoluenes (2,4-DANT) as the metabolites which remained soluble in the culture medium and azoxy-dimers as the main products in the cell extracts. Hence, the new isolate could be useful for the removal of TNT from contaminated waters.  相似文献

Two extracellular enzymes (MsP1 and MsP2) capable of efficient β-carotene degradation were purified from culture supernatants of the basidiomycete Marasmius scorodonius (garlic mushroom). Under native conditions, the enzymes exhibited molecular masses of ~150 and ~120 kDa, respectively. SDS-PAGE and mass spectrometric data suggested a composition of two identical subunits for both enzymes. Biochemical characterisation of the purified proteins showed isoelectric points of 3.7 and 3.5, and the presence of heme groups in the active enzymes. Partial amino acid sequences were derived from N-terminal Edman degradation and from mass spectrometric ab initio sequencing of internal peptides. cDNAs of 1,604 to 1,923 bp, containing open reading frames (ORF) of 508 to 513 amino acids, respectively, were cloned from a cDNA library of M. scorodonius. These data suggest glycosylation degrees of ~23% for MsP1 and 8% for MsP2. Databank homology searches revealed sequence homologies of MsP1 and MsP2 to unusual peroxidases of the fungi Thanatephorus cucumeris (DyP) and Termitomyces albuminosus (TAP).  相似文献

South American camelids comprise the guanaco (Lama guanicoe) and the vicuña (Vicugna vicugna), which are wild species, and the domestic llama (Lama glama) and alpaca (Lama pacos). This paper presents the first complete mitochondrial (mt) genome of the guanaco and the mt coding sequence of the vicuña. The guanaco mtDNA is 16,649 nt long and its composition and organization are similar to the mitochondrial genome of other mammals. Excluding the control region, comparison of the complete guanaco and vicuña mtDNA showed 4.4% sequence divergence. Nucleotide differences in peptide coding genes varied from 1.9% in ATP6 to 6.4% in Cyt b. These values are compatible with the close relatedness of both species identified by other authors. Based on the differences between the control region sequence here reported and that previously described, we also discuss the occurrence of NUMTs in the genome of South American camelids.  相似文献

The mating system and seed variation of Acacia hybrid (A. mangium × A. auriculiformis) were studied using allozymes and random amplified polymorphic DNA (RAPD) markers, respectively. Multi-locus outcrossing rate estimations indicated that the hybrid was predominantly outcrossed (mean±s.e. t m = 0.86±0.01). Seed variation was investigated using 35 polymorphic RAPD fragments. An analysis of molecular variance (AMOVA) revealed the highest genetic variation among seeds within a pod (66%–70%), followed by among pods within inflorescence (29%–37%), and the least variation among inflorescences within tree (<1%). In addition, two to four RAPD profiles could be detected among seeds within pod. Therefore, the results suggest that a maximum of four seeds per pod could be sampled for the establishment of a mapping population for further studies.  相似文献

Thymosin α1 (Tα1), a 28-amino acid N α -acetylated peptide, has a powerful general immunostimulating activity. Although biosynthesis is an attractive means of large-scale manufacture, to date, Tα1 can only be chemosynthesized because of two obstacles to its biosynthesis: the difficulties in expressing small peptides and obtaining N α -acetylation. Graptolites from the Jaeger collection at the Museum für Naturkunde (Berlin, Germany) provide important information on structural details of Silurian (Wenlock–Ludlow) retiolitids as well as for the biostratigraphic and biogeographic distribution of these magnificent graptolites. Species of the genera Cometograptus, Spinograptus and Plectograptus are described from isolated glacial boulder material, collected in northern Germany and from shale specimens found in the Lower Graptolite Shale of Thuringia. The biostratigraphic placement of material derived from glacial erratic boulders, however, is far from being precise. The fauna associated with the neotype of Plectograptus macilentus in the ‘Unterer Graptolithenschiefer’ of Thuringia is discussed and illustrated. Cometograptus alfeisenacki from the Cyrtograptus lundgreni Biozone is recognized as a new species. The genus is discovered for the first time in North German glacial erratic boulders.  相似文献

Background  

Carotenoids are a group of C40 isoprenoid molecules that play diverse biological and ecological roles in plants. Tomato is an important vegetable in human diet and provides the vitamin A precursor β-carotene. Genes encoding enzymes involved in carotenoid biosynthetic pathway have been cloned. However, regulation of genes involved in carotenoid biosynthetic pathway and accumulation of specific carotenoid in chromoplasts are not well understood. One of the approaches to understand regulation of carotenoid metabolism is to characterize the promoters of genes encoding proteins involved in carotenoid metabolism. Lycopene β-cyclase is one of the crucial enzymes in carotenoid biosynthesis pathway in plants. Its activity is required for synthesis of both α-and β-carotenes that are further converted into other carotenoids such as lutein, zeaxanthin, etc. This study describes the isolation and characterization of chromoplast-specific Lycopene β-cyclase (CYC-B) promoter from a green fruited S. habrochaites genotype EC520061.  相似文献

ALS1 and ALS3 encode cell-surface associated glycoproteins that are considered to be important for Candida albicans biofilm formation. The main goal of the present study was to monitor ALS1 and ALS3 gene expression during C. albicans biofilm formation (on silicone) under continuous flow conditions, using the Centers for Disease Control biofilm reactor (CDC reactor). For ALS1, we found few changes in gene expression until later stages of biofilm formation (72 and 96 h) when this gene appeared to be downregulated relative to the gene expression level in the start culture. We observed an induction of ALS3 gene expression in the initial stages of biofilm formation (0.5, 1, and 6 h), whereas at later stages, this gene was also downregulated relative to the gene expression level in the start culture. We also found that biofilms of an als3/als3 deletion mutant contained less filaments at several time points (1, 6, 24, and 48 h), although filamentation as such was not affected in this strain. Together, our data indicate an important role for ALS3 in the early phases of biofilm formation in the CDC reactor, probably related to adhesion of filaments, while the role of ALS1 is less clear.  相似文献

Ciliated protists are important predators of bacteria in many aquatic habitats, including sediments. Since, many biochemical transformations within the nitrogen cycle are performed by bacteria, ciliates could have an indirect impact on the nitrogen cycle through selective grazing on nitrogen-transforming bacteria. As a case study, we examined ciliate grazing on nitrifying bacteria of the genera Nitrosomonas and Nitrospira. All experiments were designed as in vitro-experiments with cultures of different bacteria and ciliate species. The nitrifying bacteria used in our experiments were Nitrosomonas europaea [Winogradsky 1892] and Nitrospira moscoviensis [Ehrich 2001]. The ciliates comprised of four species that are known as efficient bacterivores and common members of the protist community in aquatic systems: Paramecium aurelia [Müller 1773], Euplotes octocarinatus [Carter 1972], Tetrahymena pyriformis [Ehrenberg 1830] and Cyclidium glaucoma [Müller 1786]. Our experimental approach, using a combination of DAPI and FISH staining, was successful in allowing the observation of ingestion of specific bacteria and their detection within ciliate food vacuoles. However, the ciliates in this study showed no significant selective grazing. No food preferences for a any bacterial taxon or any size class or morphotype were detected. Correlation with time between ciliate abundance and bacterial abundance or biovolume, using log transformed growth rates of ciliates and bacteria, showed no significant results. On the bacterial side, neither an active defence mechanism of the nitrifying bacteria against ciliate grazing, such as changes in morphology, nor competition for resources were observed. These results suggest that in our in vitro-experiments grazing by ciliates has no influence on abundance and growth of nitrifying bacteria and nitrification.  相似文献

The molecular mechanism of the unique cis to trans isomerization of unsaturated fatty acids in the solvent-tolerant bacterium Pseudomonas putida S12 was studied. For this purpose, the carbon isotope fractionation of the cistrans isomerase was estimated. In resting cell experiments, addition of 3-nitrotoluene for activation of the cistrans isomerase resulted in the conversion of the cis-unsaturated fatty acids into the corresponding trans isomers. For the conversion of C16:1 cis to its corresponding trans isomer, a significant fractionation was measured. The intensity of this fractionation strongly depended on the rate of cistrans isomerization and the added concentration of 3-nitrotoluene, respectively. The presence of a significant fractionation provides additional indication for a transition from the sp2 carbon linkage of the cis-double bond to an intermediate sp3 within an enzyme–substrate complex. The sp2 linkage is reconstituted after rotation to the trans configuration has occurred. As cytochrome c plays a major role in the catabolism of Cti polypeptide, these findings favour a mechanism for the enzyme in which electrophilic iron (Fe3+), provided by a heme domain, removes an electron of the cis double bond thereby transferring the sp2 linkage into sp3.  相似文献

The photoswitching behaviour of the green fluorescent protein (GFP) chromophore and its analogs opens up exciting horizons for the engineering and development of molecular devices for high sensitivity in vivo studies. In this work we present the synthesis and photophysical study of four GFP chromophore analogs belonging to butenolide and pyrrolinone classes. These chromophores possess an intriguing photoinduced cistrans isomerization mechanism. Stereochemical structural assignment was unambiguously performed by 1D Nuclear Overhauser Effect NMR measurements. The spectroscopic properties of both cis and trans isomers were studied, and photoconversion quantum yield for cistrans isomerization was assessed to be in the 0.1–0.4 range. Finally, the 3JC,H coupling constant in the 13C–C=C–H motif was in excellent agreement with theoretical DFT calculations, thus providing a further confirmation of cistrans photoisomerization of the structurally analog GFP chromophore.  相似文献

The two freshwater sponges Ephydatia fluviatilis and Ephydatia mülleri belong to the widespread Spongillidae family. Their morphological tracts are very similar and can be distinguished mainly on the basis of their gemmuloscleres. However, when gemmules are absent it is essential to have an unambiguous species attribution for a population genetic study based on fresh tissues and historical collections. This article reports a simple Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) analysis, applied to DNA extracted from both gemmules and fresh tissues in order to discriminate between the two congeneric E. fluviatilis and E. mülleri. Such a biomolecular method is based on the discriminative enzymes’ digestion of each of the three amplified fragments 5.8S-ITS2-28S, D3 domain of the 28S subunit and COI. Two restriction enzymes were tested for a 620–642 bp fragment of 5.8S-ITS2-28S and for a 342 bp fragment of the D3 domain of the 28S, one restriction enzyme was tested for a 681 bp fragment codifying for COI. Obtained digestion patterns were diagnostic for each of the two species, providing a relatively simple, fast and cheap method for species attribution compared to sequencing. Handling editor: C. Tea waste (TW) was investigated as a new supplement for substrate mixtures in Ganoderma lucidum cultivation. The effects of sawdust (S) based substrates supplemented with TW at the various levels (75S:25TW, 80S:20TW, 85S:15TW, and 90S:10TW) and Ganoderma lucidum strains on yield, biological efficiency (BE) and the chemical composition of fruiting bodies were determined in solid-state fermentation. Significant differences were found among substrates regarding yield and BE, while yield and BE of the strains were not different. The substrate formulations producing highest yield and BE were 80S:20TW (87.98 g/kg substrate and 34.90%) and 75S:25TW (82.30 g/kg substrate and 31%). Yield and BE of substrates containing TW were generally higher than that of the control (80sawdust:18wheat bran:1sucrose:1 CaCO3). Nitrogen, potassium, iron, and manganese contents and C:N ratios of substrates were strongly correlated with yield. BE showed positive and significant correlations with potassium, iron and manganese. Moisture content, potassium, magnesium, calcium, iron, and zinc contents of the fruiting bodies were affected by both strain and substrate. It was concluded that TW can be used as a supplement for substrate preparation in G. lucidum cultivation.  相似文献
