To genuinely understand how complex biological structures function, we must integrate knowledge of their dynamic behavior and of their molecular machinery. The combined use of light or laser microscopy and electron microscopy has become increasingly important to our understanding of the structure and function of cells and tissues at the molecular level. Such a combination of two or more different microscopy techniques, preferably with different spatial- and temporal-resolution limits, is often referred to as ‘correlative microscopy’. Correlative imaging allows researchers to gain additional novel structure–function information, and such information provides a greater degree of confidence about the structures of interest because observations from one method can be compared to those from the other method(s). This is the strength of correlative (or ‘combined’) microscopy, especially when it is combined with combinatorial or non-combinatorial labeling approaches. In this topical review, we provide a brief historical perspective of correlative microscopy and an in-depth overview of correlative sample-preparation and imaging methods presently available, including future perspectives on the trend towards integrative microscopy and microanalysis.  相似文献

The topographic distribution of dopamine (DA) uptake, choline uptake, choline acetyltransferase (ChAT) activity and GABA uptake within the striata of weaver mutant mice and control mice was determined. Uptake of [3H]dopamine, [3H]choline and [14C]GABA, as well as ChAT activity were determined in samples prepared from the dorsolateral, dorsomedial, ventrolateral and ventromedial portions of the striatum. In 45–60 day old control mice, dopamine uptake was homogeneously distributed throughout the striatum. On the other hand, striata from weaver mice exhibited an uneven distribution with the ventral aspects having greater uptake activity than the dorsal regions. Thus, although the ventral portion of the striatum is less severely affected than the dorsal portion, all areas of the striatum exhibited significantly reduced uptake rates. In 9 and 12 month old mice, choline uptake was higher in lateral than medial zones of the striatum of both genotypes and no differences were observed between genotypes. GABA uptake was higher in the ventral striatum than in the dorsal striatum but again no differences were found between weaver and control mice. The results of this study indicate that the entire weaver striatum is severely deficient in its ability to recapture dopamine and thus is functionally compromised. The results also indicate that the striatal cholinergic and GABAergic interneurons are not directly or indirectly affected by the weaver gene.Special ïssue dedicated to Dr. Morris H. The scavenger receptor class B type I (SR-BI) plays an important role in mediating selective uptake of high-density lipoprotein (HDL)-derived cholesterol and cholesteryl ester in liver and steroidogenic tissues. The molecular mechanism by which this receptor mediates selective cholesteryl ester uptake remains still enigmatic. We applied ultrasensitive fluorescence microscopy to visualize the intracellular transport routes of HDL particles taken up via SR-BI in a Chinese hamster ovarian cell line. Although diffusion of the receptor bound particles on the cell surface is slow, internalization is accompanied by a dramatic increase in the mobility of the particles. HDL particles are endocytosed as clusters and actively transported to the perinuclear region of the cell. Costaining with organelle markers confirmed the involvement of an acidic compartment and the Golgi apparatus in the uptake process; finally, resecretion of the HDL particles was observed.  相似文献

Diffuse optical imaging (DOI) techniques provide a wide‐field or macro assessment of the functional tumor state and have shown substantial promise for monitoring treatment efficacy in cancer. Conversely, intravital microscopy provides a high‐resolution view of the tumor state and has played a key role in characterizing treatment response in the preclinical setting. There has been little prior work in investigating how the macro and micro spatial scales can be combined to develop a more comprehensive and translational view of treatment response. To address this, a new multiscale preclinical imaging technique called diffuse and nonlinear imaging (DNI) was developed. DNI combines multiphoton microscopy with spatial frequency domain imaging (SFDI) to provide multiscale data sets of tumor microvascular architecture coregistered within wide‐field hemodynamic maps. A novel method was developed to match the imaging depths of both modalities by utilizing informed SFDI spatial frequency selection. An in vivo DNI study of murine mammary tumors revealed multiscale relationships between tumor oxygen saturation and microvessel diameter, and tumor oxygen saturation and microvessel length (|Pearson's ρ| ≥ 0.5, P < 0.05). Going forward, DNI will be uniquely enabling for the investigation of multiscale relationships in tumors during treatment.   相似文献

The highly Mo efficient winter wheat cultivar 97003 yielded more than 90% and the low Mo efficient winter wheat cultivar 97014 less than 50% under Mo deficient conditions when compared to the Mo fertilizer treatment. The mechanism of Mo efficiency, molybdenum uptake and distribution in plant parts during all growth stages, was studied with these two cultivars when grown in an acid yellow-brown earth with no Mo (CK) and added Mo (+Mo) treatments. The results showed that accumulation of Mo and dry matter in shoots of cultivar 97003 was significantly higher than those of cultivar 97014 under CK through the entire growth period. Most of Mo was found accumulated in shoots after the stem elongation stage. Only low amount of Mo was accumulated during the cold winter until stem elongation stage where severe symptoms occurred in cultivar 97014 without Mo supply, while the Mo concentration of cultivar 97003 was significantly higher, thus improved its cold hardiness. Molybdenum concentrations in spikes and seeds were very low pointing to a low Mo mobility even under Mo sufficiency. However, much more Mo was distributed in the upper leaves at stem elongation stage, in spikes in heading stage, in seeds in maturity in cultivar 97003 than in cultivar 97014 under conditions of Mo deficiency. In the efficient cultivar, the Mo distribution ratios to the upper leaves and spikes were even higher without Mo supply, suggesting that a higher phloem mobility and thus a more efficient use of Mo under Mo deficiency stress. The ability of Mo uptake and phloem-mobility are discussed and it is suggested to be the important physiological basis of Mo efficiency.  相似文献

High-resolution imaging of autophagy has been used intensively in cell culture studies, but so far it has been difficult to visualize this process in detail in whole animal models. In this study we present a versatile method for high-resolution imaging of microbial infection in zebrafish larvae by injecting pathogens into the tail fin. This allows visualization of autophagic compartments by light and electron microscopy, which makes it possible to correlate images acquired by the 2 techniques. Using this method we have studied the autophagy response against Mycobacterium marinum infection. We show that mycobacteria during the progress of infection are frequently associated with GFP-Lc3-positive vesicles, and that 2 types of GFP-Lc3-positive vesicles were observed. The majority of these vesicles were approximately 1 μm in size and in close vicinity of bacteria, and a smaller number of GFP-Lc3-positive vesicles was larger in size and were observed to contain bacteria. Quantitative data showed that these larger vesicles occurred significantly more in leukocytes than in other cell types, and that approximately 70% of these vesicles were positive for a lysosomal marker. Using electron microscopy, it was found that approximately 5% of intracellular bacteria were present in autophagic vacuoles and that the remaining intracellular bacteria were present in phagosomes, lysosomes, free inside the cytoplasm or occurred as large aggregates. Based on correlation of light and electron microscopy images, it was shown that GFP-Lc3-positive vesicles displayed autophagic morphology. This study provides a new approach for injection of pathogens into the tail fin, which allows combined light and electron microscopy imaging in vivo and opens new research directions for studying autophagy process related to infectious diseases.  相似文献

In this study, the mechanism of the internalization and the cellular distribution of 59 fluorescein conjugated PS-ODN (FITC-ODN) after transfection with different mixed lipidic vesicles/oligo complexes (lipoplexes) have been investigated. Mixed lipidic vesicles were prepared with one of the most used cationic lipid (DOTAP) and different amounts of a cholic acid (UDCA) to release the oligo into HaCaT cells. Using flow cytometry, the cellular uptake of the oligo was studied with and without different inhibitors able to block selectively the different pathways involved in the internalization mechanism. The intracellular distribution of the oligo was analyzed by confocal laser scanning microscopy (CLSM), treating the cells with the lipoplexes and directly observing without any fixing procedure. To better carry out the colocalization studies, fluorescent-labeled markers, specific for the different cellular compartments, were coincubated with 59 fluorescein-conjugated 29-mer phosphorotioate oligonucleotide (FITC-ODN). The different lipidic vesicles affect the internalization mechanism of FITC-ODN. After using the inhibitors, the uptake of complexes involved a different internalization mechanism. The live CLSM analysis demonstrated that, after 1 hour from the complex incubation, the oligo was transferred into cells and localized into the endosomes; after 24 hours, the oligo was intracellularly localized close to the nuclear structure in a punctuate pattern. However, the results from fusion experiments showed also a binding of a quite low amount of oligo with the cell membranes.  相似文献

Summary In the pancreas of Scyliorhinus stellaris large islets are usually found around small ducts, the inner surface of which is covered by elongated epithelial cells; thus the endocrine cells are never exposed directly to the lumen of the duct. Sometimes, single islet cells or small groups of endocrine elements are also incorporated into acini. Using correlative light and electron microscopy, eight islet cell types were identified:Only B-cells (type I) display a positive reaction with pseudoisocyanin and aldehyde-fuchsin staining. This cell type contains numerous small secretory granules (Ø280 nm). Type II- and III-cells possess large granules stainable with orange G and azocarmine and show strong luminescence with dark-field microscopy. Type II-cells have spherical (Ø700 nm), type III-cells spherical to elongated granules (Ø450 × 750 nm). Type II-cells are possibly analogous to A-cells, while type III-cells resemble mammalian enterochromaffin cells. Type IV- cells contain granules (Ø540 nm) of high electron density showing a positive reaction to the Hellman-Hellerström silver impregnation and a negative reaction to Grimelius' silver impregnation; they are most probably analogous to D-cells of other species. Type VI-cells exhibit smaller granules (Ø250 × 500 nm), oval to elongated in shape. Type VI-cells contain small spherical granules (Ø310 nm). Type VII-cells possess two kinds of large granules interspersed in the cytoplasm; one type is spherical and electron dense (Ø650 nm), the other spherical and less electron dense (Ø900 nm). Type VIII-cells have small granules curved in shape and show moderate electron density (Ø100 nm). Grimelius-positive secretory granules were not only found in cell types II and III, but also in types V, VI, and VII. Skin tissue, in addition to its specific use in dermal research, provides an excellent model for developing the techniques of vibrational microscopy and imaging for biomedical applications. In addition to permitting characterization of various regions of skin, the relative paucity of major biological constituents in the stratum corneum (the outermost layer of skin), permits us to image, with microscopic resolution, conformational alterations and concentration variations in both the lipid and protein components. Thus we are able to monitor the effects of exogenous materials such as models for drug delivery agents (liposomes) and permeation enhancers (DMSO) on stratum corneum lipid organization and protein structure. In addition, we are able to monitor protein conformational changes in single corneocytes. The current article demonstrates these procedures, ranging from direct univariate measures of lipid chain conformational disorder, to factor analysis which permits us to image conformational differences between liposomes that have permeated through the stratum corneum from those which have remained on the surface in a reservoir outside the skin.  相似文献

Treatment of mammalian muscle with the divalent cation ionophore A23187 causes the release of Ca2+ from the sarcoplasmic reticulum and allows the ultrastructural changes of the mitochondria during Ca2+-uptake to be demonstrated in situ. Electron micrographs reveal that the mitochondria swell dramatically during uptake, before contracting again when the accumulated Ca2+ is released once more into the cytoplasm. When maximally swollen, the mitochondria are apparently subdivided and internal "septa" are formed. The ultrastructural details concerning these internal membranous structures are shown in detail and their significance is discussed.  相似文献

用压力室灌流挤压法结合原子吸收分光光度计测定了胡杨、沙枣、柽柳、梭梭和花棒等5种荒漠优势植物组织以及细胞内和质外体溶液中K+、Na+含量,并用TPS-1型光合蒸腾测定系统和露点微伏压计测定了叶片(同化枝)的蒸腾速率和组织渗透势,以分析荒漠植物离子吸收特点与其适应性的关系。结果表明:5种植物叶片(同化枝)中K+含量差异较小,但Na+含量却有极显著差异,其中梭梭Na+含量最高、胡杨和柽柳次之、花棒和沙枣相对较低,且梭梭和柽柳的根系和组织细胞膜对Na+也具有更高的透性。另外,实验结果还显示组织Na+含量与组织渗透势和蒸腾失水率均呈显著负相关,即Na+的吸收、积累可能在渗透调节和减少水分散失中具有重要作用。由此可见,梭梭和柽柳能够通过大量吸收和积累无机离子来降低渗透势、增强吸水力,同时减少蒸腾失水,具有很强的荒漠环境适应能力;而胡杨蒸腾耗水量较大、花棒和沙枣生理吸水的动力不足,与梭梭和柽柳相比,其荒漠环境适应能力相对较弱。  相似文献

Summary— The in situ distribution of phosphorus in perichromatin granules (PCGs), and in the surrounding nucleoplasm was investigated in rat liver cells by means of electron spectroscopic imaging of unstained preparations. A 2–3 nm fibril containing high concentration of phosphorus was found to be the main substructural feature of the PCGs revealed in the maps of phosphorus. This fibril is folded within the PCG with no apparent order. Fibrils of similar diameter and phosphorus content were also found in both the halo surrounding the PCG and dispersed in the nucleoplasm. Some of such fibrils are in continuity with those occurring within PCGs. Sometimes these fibrils are grouped forming a stalk connecting the PCG to chromatin. Some stalked PCGs are U-shaped or kidneyshaped, resembling Balbiani ring granules in the process of formation as observed in Chironomus salivary gland cell nuclei. The external fibrils are interpreted as perichromatin fibrils considered to be precursors of PCGs.  相似文献

The net uptake, distribution and assimilation of NO 3 were studied in pea plants subjected to either long-term continuous Cd treatment for 10 d (10 or 50 M Cd) or short-term treatment (72 h) with 50 M Cd. In the latter treatment, the effects of transferring the plants to a Cd-free nutrient solution for a 'recovery period' of 96 h were also studied. All these treatments were compared with 'controls', plants which received no Cd. In both experiments, the reduction in fresh weight was associated with a decrease in the content (%) of shoot and root water and in transpiration rate as Cd concentration increased. The concentration of 3 in the shoots and sap decreased dramatically and net 3 uptake was severely inhibited, effects associated with a loss of shoot nitrate reductase (NR) activity. In the short-term Cd treatment, net 3 uptake was almost completely inhibited after 24 h, but recovered after the transfer of plants to a Cd-free nutrient solution. Similarly, a dramatic decrease in the shoot NR activity was observed. The uptake, distribution and tissue partitioning of K was also studied, which is considered to be the major counterion of 3 . Potassium uptake was similarly affected by Cd, as inferred from the ratio 3 /K uptake, which was ca. 10. The ratio K/ 3 tissue content increased in the shoot concomitantly to Cd in both long-term and short-term metal supply. These parameters showed a tendency of K similar to that observed for 3 , although its relative tissue distribution was not affected by Cd.  相似文献

Summary The distribution of an antigenic material specific for the cervicovaginal epithelium (CVA) was studied in the vaginal epithelium of the adult mouse with immunofluorescence and immunoferritin techniques. The antigen localization has been correlated to the fine structure of the vaginal epithelium in various states of functional activity. The antigen distribution in adult ovariectomized mice and in ovariectomized mice treated with estradiol was compared with that in normal cycling mice. CVA was found to be associated with the exterior of the cell membrane of vaginal epithelium cells, being part of the glycocalyx.Two cell types, mucous or keratinized, are derived from the germinative cell layer of the vaginal epithelium, depending on the hormonal environment. Mucous cells with morphological features that characterize cells about to cornify have been demonstrated. Fluorescence as well as ferritin particles, indicating the presence of antigen-antibody complexes, were seen within the mucous droplets of the mucous cells. The CVA production is apparently connected with vaginal mucus formation. The CVA distribution in the adult vaginal epithelium is discussed in relation to the distribution demonstrated earlier in the cervicovaginal epithelium of neonatal mice.This investigation was supported by grants from the Norwegian Cancer Society (Landsforeningen mot Kreft) and the Norwegian Research Council.  相似文献

Three hours after the absorption of selenite by the roots ofPhaseolus vulgaris a major part of the Se accumulates in the roots while the fraction conveyed towards the aerial organs is unevenly distributed. Uptake and distribution are enhanced when plants are kept in darkness or when the nutritive solution is supplied with glucose, suggesting an active transport of selenite. However, when roots are soaked in a low temperature solution or supplied with metabolic inhibitors, the level of Se decreased by about 35% in the roots and in the aerial organs. So, part of the selenite enters the roots by diffusion, while another part is metabolically linked. Sugar translocated from the leaves is implicated as a co-transport-substance for selenite. Antibiotics, such as gramicidin, synthetised by micro-organisms, can also increase the level of Se within the bean plant.  相似文献

Summary The fine structure of the absorptive cells in human small-intestinal biopsies cultured for 6, 24, and 48 h was analyzed qualitatively and quantitatively. The findings show generally good preservation of the cultured absorptive cells and a normal distribution, size, and relative volume of their cell organelles, but there was a systematic decrease in the apical cell surface and an increase in the number of apical vesicles and tubules after culturing. Since the apical vesicles and tubules are thought to have a function in the transport of cell-coat material from the Golgi apparatus to the cell surface, these findings raise the question of whether a delayed transport or extrusion of cell surface material occurs.The diminished relative volume of the mitochondria and the increased signs of autophagy in some poorly preserved absorptive cells, are assumed to be an adaption to less favourable culture conditions.The authors acknowledge the help of Dr. A.S. The apical uptake of 64CuCl2 was investigated in human differentiated intestinal Caco-2 cells grown on permeable supports. At pH 6.0 in the apical compartment, the uptake of copper was linear over the first 6 min and between 10 and 80 M CuCl2 exhibited non-saturable transport kinetics. In addition, copper uptake was energy-independent, affected by the valency state of copper, preferring Cu(II) over Cu(I), and not influenced by high (10 mM) extracellular calcium. The intracellular distribution of copper was investigated by FPLC at different times of uptake (`pulse') and of `chase'. Intracellular copper initially bound predominantly to low molecular weight components (i.e., glutathione), and subsequently shifted to higher molecular weight components such as metallothionein and Cu,Zn superoxide dismutase.  相似文献

Uptake by roots from contaminated soil is one of the key steps in the entry of radiocaesium into the food chain. We have measured the uptake by roots of radiocaesium and its transfer to shoots of a heathland grass, sheep fescue (Festuca ovina L.) from two contrasting agricultural soils, a sandy podzol and a clayey calcareous soil. A culture device which keeps the roots separate from the soil was used thus allowing rhizosphere soil to be obtained easily and enhancing the effect of root action. Biomass production and 137Cs in shoots and roots were recorded. Cs adsorption was studied on both the initial, nonrhizosphere soil and on rhizosphere soil in dilute soil suspension. Cs desorption was measured by resuspending subsamples of contaminated soil in solutions containing various concentrations of stable Cs. The proportion of Cs fixed, i.e. not readily desorbable, was calculated by comparison of the adsorption and desorption isotherms. Uptake by roots varied considerably between soils and did not appear to be diffusion limited. Root-to-shoot transfer did not differ for the two soils studied. Root action considerably enhanced Cs adsorption on the soils, particularly the in sandy podzol with a low Cs affinity. The value of Kd was increased by up to an order of magnitude. A large proportion of adsorbed Cs was found to be fixed, the Kd was up to seven times greater on desorption than adsorption, indicating that up to 80% of adsorbed Cs was not readily exchangeable. Root action had little effect on the fixed fraction. This revised version was published online in June 2006 with corrections to the Cover Date.  相似文献
