Summary Complex segregation analysis of plasma -L-fucosidase in 45 British families provides evidence for an additive major gene causing low activities of fucosidase. There was no significant evidence of polygenic heritability or common family environment.  相似文献

Measurements of wing-beat frequency (WBF) have been used to characterize flight muscle metabolic rate in Drosophila melanogaster during tethered flight. Progeny of crosses between 17 X-chromosome substitution lines and three null-activity stocks have been studied in order to determine the effect on flight metabolism of sharply reduced activity of -glycerophosphate dehydrogenase (GPDH). It was found that flies with an approximate 50% reduction in GPDH activity have a metabolic rate that is, in most cases, indistinguishable from that of wild-type flies and, in the most extreme cases, reduced by only 4%. These results demonstrate that Gpdh is not a major gene for flight metabolism, in the quantitative genetic sense of the term. These results are in agreement with the Kacser and Burns (1973, 1979, 1981) theory of flux, which postulates that the activity of an enzyme embedded in a multienzyme pathway can sometimes vary from wild-type to very low levels (perhaps 5–10% wild type) with no significant effect on flux through the total pathway.This research was supported by several grants to JWC: NSF Grant 8211667, a grant from the Graduate School, University of Minnesota, and a Research Career Development Award from the NIH.  相似文献

Vitamin E is a lipophilic antioxidant that has been inversely associated with certain chronic diseases; however, the biological processes regulated by this vitamin have not been fully elucidated. The objective of the present study was to examine the association between the most biologically active and abundant form of vitamin E in the circulation, α-tocopherol, and the plasma proteome. Subjects were from the Toronto Nutrigenomics and Health Study and included men and women (n=1,022) who completed a general health and lifestyle questionnaire and 196-item food frequency questionnaire, and provided a fasting blood sample. Plasma α-tocopherol concentrations were measured by high-performance liquid chromatography and 54 plasma proteins were assayed by a mass spectrometry-based multiple reaction monitoring method. Analysis of covariance was used to compare mean concentrations of plasma proteins across tertiles of α-tocopherol. Plasma concentrations of apolipoprotein C-III, fibrinogen alpha, beta, and gamma chains, fibronectin and fibrinopeptide A were significantly and positively associated with plasma α-tocopherol, while intermediate levels of α-tocopherol were significantly associated with higher levels of alpha-1B-glycoprotein (all P<.0009). These findings show that circulating levels of α-tocopherol are significantly associated with specific plasma proteins and suggest novel physiological effects of vitamin E.  相似文献

α-L-Fucosidase (EC 3.2.1.51) activity was studied in different reproductive organs, seminal plasma and spermatozoa of the bull. The highest specific activity of α-L-fucosidase was found in the epididymis. Gel filtration at pH 7.0 revealed two α-L-fucosidases (α-L-fucosidase I and α-L-fucosidase II) in most reproductive tissues, but seminal plasma, spermatozoa and epididymal cauda contained only form I. Fractionation at basic pH (pH 8.5) resulted in the elution of α-L-fucosidase as form II. Some differences were encountered in pH profiles and thermal stabilities of the two enzyme forms and they showed additional polymorphism after chromatofocusing. The comparison of enzyme profiles after fractionations suggests that cauda epididymidis is the main source of the seminal plasma activity in the bull.  相似文献

Summary 4-Methylumbelliferyl neuraminidase activity was studied in fibroblasts, leukocytes, and frozen tissues from adult patients with -galactosidase-neuraminidase deficiency and specific clinical manifestations. This enzyme was almost completely deficient in fibroblasts, but the residual activity was relatively high (20% of the control mean) in the leukocytes from the patients. The frozen liver from one patient showed the enzyme activity as high as controls.This enzyme consisted of two components, freeze-labile and freeze-stable, and it was demonstrated that only the labile enzyme was deficient in fibroblasts and leukocytes. The apparently normal activity of neuraminidase in frozen autopsy tissues of a patient may be explained by the loss of the labile component in control tissues after a long-term freezing. The neuraminidase activity was variable in parents and no definite conclusion was drawn on the hereditary nature of the disease.  相似文献

Summary Transportand metabolism of -naphthaleneacetic acid -naphthaleneacetic acid, and -decalylacetic acid, all labelled with 14C in the carboxyl, group, were studied. Only -naphthaleneacetic acid is transported in a polar way. Most of the radioactivity in the tissue is in a low molecular form, either free or as immobilization products. The immobilization of -naphthaleneacetic acid is similar to that of -naphthaleneacetic acid. Immobilization of -decalylacetic acid is typically different. Bioassays showed -naphthaleneacetic acid as the sole biologically active component. It is concluded that stereo requirements necessary for biological activity are also required for polar auxin transport. It is further concluded that the observed specificity of the transport system is not related to the formation of immobilization products.  相似文献

An intraspecific comparison of -glycerophosphate (-GPDH: E.C.1.1.1.8) and alcohol dehydrogenase (ADH: E.C.1.1.1.1) enzyme activity levels was carried out in Drosophila melanogaster. The results indicate that (1) -GPDH is a relatively conservative and ADH a relatively variable enzyme system with regard to structurally determined activity variation but that (2) the conservative nature of -GPDH activity variation does not extend to the intra-genotypic level. The results are consistent with the view that different kinds of selective pressures are being exerted on the enzyme's structural and modifier gene loci.  相似文献

Abstract

The crystal-state preferred conformations of two tripeptides, one tetrapeptide, and one pen- tapeptide, each containing a single residue of the chiral, Cα,α-disubstituted glycine Cα-methyl, Cα-benzylglycine [(αMe)Phe], have been determined by X-ray diffraction. The tripeptides are Z-L-(αMe)Phe-(Aib)2-OH dihydrate and Z-Aib-D-(αMe)Phe-Aib-OtBu, the tetrapeptide is Z-(Aib)2-D-(αMe)Phe-Aib-OtBu, and the pentapeptide is pBrBz-(Aib)2-DL-(αMe)Phe-(Aib)2-OtBu. While the two tripeptides are folded in a β-bend conformation, two such conformations are consecutively formed by the tetrapeptide. The pentapeptide adopts a regular 310-helix promoted by three consecutive β-bends. This study confirms the strong propensity of short peptides containing Cα-methylated α-aminoacids to fold into β-bends and 310-helical structures. Since Aib is achiral, the handedness of the observed bends and helices is dictated by the presence of the (αMe)Phe residue. In general, we have found that the relationship between (αMe)Phe chirality and helix handedness is opposite to that exhibited by protein aminoacids. A comparison with the preferred conformation of other extensively investigated Cα-methylated aminoacids is made.  相似文献

Summary The variant -L-fucosidase genotype, phenotypically observed as a low plasma level of -L-fucosidase in healthy individuals, can modify the plasma expression of the primary genetic defect in mucolipidosis III. Three patients with clinical, radiological, and biochemical features of mucolipidosis III, had a normal plasma level of -L-fucosidase activity. The thermostability curves for the plasma -L-fucosidase from these patients and the plasma -L-fucosidase of the parents of one of them, are discussed to support the suggestion that they are homozygous for the autosomal recessive variant -L-fucosidase trait. From a practical point of view, these observation warn against the use of the plasma -L-fucosidase assay as a screening test for mucolipidoses.  相似文献

This study was conducted to examine the relationship between the concentration of zinc and neutral α-glucosidase (NAG) with semen quality. Semen samples from 75 male partners of couples who were attending the Obstetrics and Gynecology Department were analyzed for semen quality. Based on sperm count, the subjects were divided into three groups. Zinc and neutral α-glucosidase activity were estimated in seminal plasma. Results showed that mean the α-glucosidase activity was lowest among the azoospermic group with respect to oligozoospermic and normozoospermic groups. Mean zinc levels were also lower among azoospermics compared to oligozoospermic and normospermic groups. The difference was statistically significant (p<0.05). A significant positive correlation was observed between zinc levels and sperm count (r=0.29, p<0.05) and zinc and α-glucosidase activity (r=0.31, p<0.05) in seminal plasma. These results suggest that zinc and neutral α-glucosidase seem to play an important role in human reproduction.  相似文献

The combined thermal load created by exercise and a hot environment is associated with an exaggerated core temperature response. The elevated core temperature is believed to increase the total stress of the exercise. Increased stress during exercise has been associated with increased levels of cortisol. The association of cortisol with increased inflammatory responses following exercise in the heat is equivocal. Thus, the purpose of the current investigation was to explore the relationship between increases in rectal temperature (Tre) and TNFα and cortisol. To induce Tre changes, 8 male subjects (mean±SD, age=23.6±2 yr, VO2max=52.8±3.7 mL/kg/min, BMI=24.2±1.9) participated in two 40 min trials of cycle ergometry at 65% of VO2peak immersed to chest level in cool (25 °C) and warm (38.5 °C) water. Tre was monitored throughout each trial, with blood samples taken immediately pre and post of each trial. Neither cortisol nor TNFα changed significantly during exercise in the cool water; however, in the warm trial, both cortisol and TNFα significantly increased (p<0.004). Concordance correlations (Rc) between Δ cortisol and Δ TNFα indicated a strong but non-significant correlation (Rc=0.833, p=0.135). In conclusion, changes in core temperature may be impacting the relationship between exercise induced changes in cortisol and TNFα. Therefore, acute moderate-intensity exercise (40 min or less) in warm water impacts the stress and inflammatory response. Understanding this is important because exercise load may need to be adjusted in warm and hot environments to avoid the negative effects of elevated stress and inflammation response.  相似文献

A cDNA encoding an α-l-fucosidase from Drosophila melanogaster was obtained from the recombinant plasmid named pGEM-DmFuca and inserted into the pBacHTeGFPT vector to construct the recombinant donor plasmid which was transposed to the target AcBacmid in Escherichia coli (DH10) by Tn7 transposition function. The AcBacmid-GFP-DmFuca plasmid was used to transfect Tn-5B1-4 cells of the Cabbage looper Trichoplusia ni. SDS-PAGE analysis revealed a band of about 80 kDa. Using a polyclonal antiserum raised against α-l-fucosidase protein from D. melanogaster Western blotting analysis confirmed that the fusion protein eGFP-DmFuca has been successfully expressed in a biologically active form in Tn-5B1-4 cells. The recombinant protein, containing the histidine-tag motif, was purified using an affinity chromatography column. In vitro binding assays the purified eGFP-DmFuca interacts with α-l-fucose residues present on the micropyle of the D. melanogaster eggshell, confirming that the α-l-fucosidase is a good candidate as receptor involved in gamete interactions in fruit fly.  相似文献

Background

Evidences suggest that β3 -adrenoceptor (β3-AR) plays an important role in heart failure (HF), although no data is reported indicating how these effects may change with the increasing age. Pulmonary congestion and edema are the major life-threatening complications associated with HF. The purpose of this study is to explore the relationship between the anti-β3-AR autoantibody and the expression of β3-AR in the lungs and heart for both aged patients and rats with HF.

Methods

Synthetic β3-AR peptides served as the target antigens in ELISA were used to screen the anti-β3-AR autoantibody in aged patients and rats. Two aged rat models were constructed based on aortic banding and sham-operation. The expression of β3-AR mRNA and protein in the lung and heart was measured in intervention and non-intervention groups by Western blot analysis at the baseline, 5th, 7th, 9th and 11th week, respectively.

Results

The frequency and titer of anti-β3-AR autoantibody in aged patients and rats with HF were higher than those in the control group (p<0.05). The expression of β3-AR mRNA and protein in pulmonary tissues decreased continually from the 7th week (p<0.05), followed by HF observed during the 9th week. The expression of β3-AR in myocardial tissues continued to increase after the 9th week (p<0.05), and the expression of both β3-AR mRNA and protein in the BRL group [HF group with BRL37344 (4-[-[2-hydroxy-(3-chlorophenyl)ethyl-amino] phenoxyacetic acid) (a β3-AR agonist) injection] was positively correlated with BRL37344 when compared with non-BRL group (HF group without BRL37344 injection) (p<0.05).

Conclusion

Anti-β3-AR autoantibody was detected in aged patients and rats with HF. The expression of β3-AR mRNA and protein in pulmonary tissues decreased continually, and began earlier than in the heart, but its expression in myocardial tissues increased continually and could be further promoted by β3-AR agonist.  相似文献

Abstract

Sixteen volunteers have been studied during 3–4 control nights and eight of these subjects again during four successive sleeps on 30‐h “days”;. The experiments took place in a comfortable environment provided by an isolation chamber. Rectal temperature and the sleep EEG were measured throughout. The relationship between sleep stages, particularly SWS and REM sleep, and short‐term changes in rectal temperature has been investigated during both protocols. Care was taken to correct for or remove those temperature changes that could be attributed to circadian rhythmicity or the effects of loss of masking due to being awake. Results showed that there was a small but significant effect of sleep stages, with SWS producing a fall and REM sleep a rise in rectal temperature after a delay of about 30–48 minutes. It is concluded that such spontaneous changes in sleeping subjects accord with the results of other studies which indicate that thermoregulatory reflexes to hot or cold stimuli alter in different sleep stages.  相似文献

Cross-cultural studies have revealed broad quantitative associations between subsistence practice and demographic parameters for preindustrial populations. One explanation is that variationin the availability of suitable weaning foods influenced the frequency and duration of breastfeeding and thus the length of interbirth intervals and the probability of child survival (the “weaning food availability” hypothesis). We examine the available data on weaning age variation in preindustrial populations and report results of a cross-cultural test of the predictions that weaning occurred earlier in agricultural and pastoral populations because dairy and cereal production increased the availability of easily digestible, nutrientrich foods appropriate for weaning. We found that, contrary to predictions, supplementation with liquid foods other than breast milk was delayed in agricultural populations relative to less agriculturally dependent ones and complementary feeding with solid foods was delayed in pastoral populations relative to those less dependent on herding. Although the duration of breastfeeding was longer in populations dependent on hunting, there was no qualitative evidence that such populations lacked foods appropriate for weaning. The patterns observed suggest that the relationships between demography and subsistence observed among preindustrial societies cannot be explained by the “weaning food availability” hypothesis. We discuss the implications for understanding the mechanisms underlying prehistoric human demography, subsistence shifts, allocation to parenting and mating effort, and the evolutionary implications of tradeoffs between diet and disease. Diana Smay is a graduate student in the Department of Anthropology, Emory University. Her research interests include bioarchaeology, paleoepidemiology, the evolution of disease, and the behavioral determinants of paleodemography.
