Stanniocalcin (STC)1 is the mammalian homologue of STC which was originally identified as a calcium/phosphate-regulating hormone in bony fishes. STC1 is a homodimeric phosphoglycoprotein with few if any identified unique motifs in its structure with the exception of CAG repeats in the 5'-untranslated region. In contrast to fish STC which is expressed mainly in the corpuscles of Stannius, STC1 is expressed in a wide variety of tissues, but unexpectedly is not detected in the circulation under normal circumstances. Thus, STC1 may play an autocrine/paracrine rather than a classic endocrine role in mammals. Consistent with this, pleiotropic effects of STC1 have been postulated in physiological and measured in pathological situations. There is much current interest in identifying a specific STC1 receptor and putative signaling pathways to which it may be coupled. In this regard, STC1 may regulate intracellular calcium and/or phosphate (Pi) levels. In the skeletal system, for example, Pi uptake in bone-forming osteoblasts via a direct effect of STC1 on expression of the NaPi transporter Pit1 may contribute to bone formation. Here we review current understanding of the role of STC1 and its possible molecular mechanisms in the skeleton and elsewhere.  相似文献

Summary The ultrastructure of the pinealocytes of the Malaysian rat (Rattus sabanus), a mammal inhabiting a zone near the equator where the annual variations of daylength are inconspicuous, was examined and compared with that of pinealocytes of other mammals.On the basis of the presence of granular vesicles, only one population of pinealocytes was found. A large number of granular vesicles and vesicle-crowned rodlets is characteristic of the pinealocytes of this equatorial species. Vesicle-crowned rodlets are especially numerous in the endings of the pinealocyte processes and; they most often found in direct topographical connection with the perivascular spaces.The physiological significance of the presence of such large amounts of vesicle-crowned rodlets and of the secretory process characterized by the formation of granular vesicles is discussed.
Résumé Les pinéalocytes d'un Rat de Malaisie (Rattus sabanus), un animal vivant près de l'équateur donc dans une zone où les variations annuelles de la longueur des jours sont minimes, examinés au microscope électronique ont été comparés à ceux d'autres Mammifères. Une seule population de pinéalocytes a été observée. La présence d'un grand nombre de vésicules granulaires et de rubans circonscrits par des vésicules est la caractéristique premi`ere des pinéalocytes de cette espèce. Les rubans circonscrits par des vésicules ont été plus spécialement observés dans les terminaisons des prolongements des pinéalocytes où ils sont nombreux et la plus souvent en contact directe avec l'espace périvasculaire.L'importance physiologique de ces rubans circonscrits par des vésicules et des vésicules granulaires est discuté.

An interleukin-2 (IL-2) dependent cell line which could be grown continuously with crude concanavalin A-stimulated rat or mouse spleen cell culture supernatant could not survive for over 48 hr in the culture medium supplemented with partially purified or recombinant IL-2. The cell growth was restored by adding another factor obtained from the same crude culture supernatant. This potentiating activity which was physicochemically inseparable from serum albumin was also obtained from the culture medium containing 2-mercaptoethanol (2-ME) and incubated at 37 C for 24 hr without the spleen cells. By further experiments, it was demonstrated that 2-ME itself had the potentiating activity on the IL-2-dependent proliferation of this cell line and cysteine mediated the activity of 2-ME. The cells could not enter S-phase in the absence of 2-ME even in the presence of IL-2.  相似文献

Summary In the pineal gland of the pipistrelle bat two different populations of pinealocytes and glial cells were observed electron microscopically. The pinealocytes of populations I and II differ in their content of metabolically active cell organelles. In the pinealocytes of population I, granular vesicles originating from the Golgi apparatus were found in the perikaryon and especially in the endings of the pinealocyte processes. Granular vesicles appeared to be more numerous in hibernating nulliparous females. The pinealocytes of population II are characterized by the presence of small cytoplasmic vacuoles, probably originating from cisternae of the granular endoplasmic reticulum and containing flocculent material of moderate electron density. Longitudinal (T1) and transverse (T2) nuclear magnetic resonance relaxation times were measured in vitro at 37, 30, 25, 15, and 5 degrees C on serum, brain, liver, kidney, and heart samples from a hibernator, the European hamster, active in summer (SA), active in winter, or in the hibernating state in winter; from a less efficient hibernator, the golden hamster; and from a homeotherm, the rat. T1 and T2 relaxation times varied between species and in the European hamster between the active and hibernating subjects. Despite the major relaxation time differences between the organs, NMR relaxation time measurements showed a general trend to an increase in the viscosity of water for the European hamster in the active state. Although these modifications were not directly related to the process of hibernation itself, the relaxation times observed in the hibernating animals were closer to those seen in the rat. This evidenced that changes of physical properties of water reflect a better adaptation to low temperatures of the hamster, as compared to the nonhibernator, given that the low water viscosity of SA hamster allows the decrease of the viscosity with temperature during the hibernating state. These in vitro studies permit the study the viscosity which is an important physicochemical parameter involved in NMR longitudinal relaxation time of water proton. More detailed studies of other physiological parameters must be undertaken by further in vivo measurements.  相似文献

Plasma atrial natriuretic factor (ANF) was measured in 16 marmots at various times of the year. Nonhibernating males (n = 6) had an average plasma concentration of 56 +/- 8 pg/ml; nonhibernating females (n = 6) had an average plasma concentration of 61 +/- 4 pg/ml. During hibernation an additional group of females (n = 4) showed an average of 25 +/- 5 pg/ml. Plasma ANF of both groups of nonhibernating marmots was significantly higher (P less than 0.01) than that the hibernating group, but there was no difference between nonhibernating males and females.  相似文献

Summary A survey has been made of the pineal region of the brain of 11 species of marsupials belonging to 5 families and a species from both families of monotremes.The results show that the pineal body of non-eutherian mammals, although well-defined in all species, has a very varied morphology. Three types of pineal recess occur: (i) a pineal recess in sensu stricto, (ii) an intercommissural pineal recess, and (iii) an infrapineal recess. The existence of nerve fibres which pass through the pineal body and form a spatial link between the habenular and posterior commissures, has been demonstrated in marsupials and monotremes. It is also likely that these animals as well as eutherian mammals possess a nervus conarii. Nerve cells are not a constant feature of the non-eutherian pineal body.The subcommissural organ (SCO) is present in all species. It does not exhibit the same degree of morphological variation as the pineal body. Horizontal sections available for 4 species within 3 families of marsupials show it to be composed of a median portion joined to bilateral protuberances. Large nerve cells occur within the SCO in all marsupial species; they are absent from the monotreme SCO. Tentatively, the relationship of these neurons to the SCO is considered to be merely one of association.The importance of an extended comparative study of this region in non- eutherian mammals in order to add insight into its phylogeny and function is emphasized.  相似文献

The ultrastructure of pinealocytes was studied in rats with 7,12-dimethylbenz(a)anthracene-induced mammary tumors which were subjected to experimental manipulations known as enhancers of pineal actions (anosmia, underfeeding or cold exposure). In these animals we found: (I)--more nuclei with deep nuclear invaginations; (II)--a large number of cytoplasmic organelles, including lipid droplets, myeloid bodies, synaptic ribbons and lysosomes; (III)--numerous degenerative changes. In general, we found an increase in structural features related to pineal photoneuroendocrine activity. Our results indicate that pineal-dependent inhibition of neoplastic growth induced by these experimental manipulations, previously reported, can be mediated through an increase in pineal metabolic activity.  相似文献

Synthetic analogues of the bradykinin potentiating nonapeptide BPP9alpha indicate significantly different structural requirements for potentiation of the bradykinin (BK)-induced smooth muscle contraction (GPI) and the inhibition of isolated somatic angiotensin I-converting enzyme (ACE). The results disprove the ACE inhibition as the only single mechanism and also the direct interaction of potentiating peptides with the bradykinin receptors in transfected COS-7 cells as molecular mechanism of potentiation. Our results indicate a stimulation of inositol phosphates (IPn) formation independently from the B2 receptor. Furthermore, the results with La3+ support the role of extracellular Ca2+ and its influx through corresponding channels. The missing effect of calyculin on the GPI disproves the role of phosphatases in the potentiating action. These experimental studies should not only contribute to a better understanding of the potentiating mechanisms but also incorporate a shift in the research towards the immune system, in particular towards the immunocompetent polymorphonuclear leukocytes. The chemotaxis of these cells can be potentiated most likely by exclusive inhibition of the enzymatic degradation of bradykinin. Thus the obtained results give evidence that the potentiation of the bradykinin action can occur by different mechanisms, depending on the system and on the applied potentiating factor.  相似文献

The considerable variation in shape, size, structure and properties of the enamel cap covering mammalian teeth is a topic of great evolutionary interest. No existing theories explain how such variations might be fit for the purpose of breaking food particles down. Borrowing from engineering materials science, we use principles of fracture and deformation of solids to provide a quantitative account of how mammalian enamel may be adapted to diet. Particular attention is paid to mammals that feed on 'hard objects' such as seeds and dry fruits, the outer casings of which appear to have evolved structures with properties similar to those of enamel. These foods are important in the diets of some primates, and have been heavily implicated as a key factor in the evolutionary history of the hominin clade. As a tissue with intrinsic weakness yet exceptional durability, enamel could be especially useful as a dietary indicator for extinct taxa.  相似文献

Studies were carried out of the permeability of the cuticle of Trichinella spiralis (Owen, 1835) for fluorochromes and histidine, the filling of the mid-gut and the amino-acids contents in it. The quantity of glicogene in different organs was estimated. A conclusion has been grown that a larva consumes food through the integuments of its body; intestinal Trichinella use within the first 20 hours of their development the reserves of glicogene and later consume food monomeres and oxygen from the host's mucous membrane through their integuments. The mode of feeding of Trichinella was formed during their evolution as an adaptation to parasitism on the mucous membrane of the host.  相似文献

Endozoochrous seed dispersal by herbivorous mammals has been verified repeatedly and its possible influence on the structure and function of herbaceous communities has been suggested. Quantitative studies, however, are lacking in the field of seed dispersal via the dung of herbivore guilds in little-altered environments. The present paper analyses seed dispersal via rabbit, fallow deer, red deer and cow dung in a Mediterranean dehesa (open woodland used for hunting and ranching) during the seeding season. Dung seed content was determined by the glasshouse cultivation of eight dung samples from each herbivore, collected fortnightly between February and August. The four herbivores disperse many seeds (spring averages are 6–15 seeds per gram of dry dung and maxima of 25–70) from a large number of species (totals between 52 and 78). Dispersal seems to be mainly determined by seed production of the plant comminity. This is reflected in (i) the dissemination of a high percentage of the species present in the dehesa, (ii) great seasonal variability, related to seed production, in the amount of seeds and number of species dispersed, and (iii) a high semi-quantitative similarity of seed content in the four types of herbivore dung throughout the year. There is also important quantitative variation that depends on animal traits and feeding habits. These results and the characteristics of species found in dung suggest the adaptation of plant species to the dispersal of their seeds via herbivore gut. This process may well have profound implications for vegetation dynamics and the evolution of plant traits.  相似文献
