A new teleomorphic genus Ascobotryozyma, with a single species, A. americana, is proposed. Its anamorph is a Botryozyma that differs from the type species, B. nematodophila, on distributional, physiological, and molecular criteria; it is described as Botryozyma americana, anam. sp. nov. Ascobotryozyma is characterized by globose asci bearing four lunate ascospores. Fusion of thallus cells precedes ascus formation. Ascobotryozyma americana was isolated from the surface of nematodes (Panagrellus dubius) associated with galleries of the poplar borer (Saperda calcarata) in trembling aspen (Populus tremuloides) in eastern Washington, USA. The teleomorph has not been produced in pure culture.  相似文献

Experiments were conducted in the greenhouse to assess root galling and egg production of three root-knot nematode species, Meloidogyne arenaria, M. incognita, and M. javanica, on several weeds common to Florida agricultural land. Weeds evaluated were Amaranthus retroflexus (redroot pigweed), Cyperus esculentus (yellow nutsedge), Eleusine indica (goosegrass), Portulaca oleracea (common purslane), and Solanum americanum (American black nightshade). Additionally, although it is recommended as a cover crop in southern regions of the U.S., Aeschynomene americana (American jointvetch) was evaluated as a weed following the detection of root galling in a heavy volunteer infestation of an experimental field in southeastern Florida. Weeds were propagated from seed and inoculated with 1000 nematode eggs when plants reached the two true-leaf stage. Tomato (Solanum lycopersicum ‘Rutgers’) was included as a positive control. Aeschynomene americana and P. oleracea roots supported the highest number of juveniles (J2) and had the highest number of eggs/g of root for all three species of Meloidogyne tested. However, though P. oleracea supported very high root levels of the three nematode species tested, its fleshy roots did not exhibit severe gall symptoms. Low levels of apparent galling, combined with high egg production, increase the potential for P. oleracea to support populations of these three species of root-knot nematodes to a degree that may not be appropriately recognized. This research quantifies the impact of P. oleracea as a host for M. arenaria, M. incognita, and M. javanica compared to several other important weeds commonly found in Florida agricultural production, and the potential for A. americana to serve as an important weed host of the three species of root-knot nematode tested in southern regions of Florida.  相似文献

Larvae removed at one-day intervals from laboratory infected intermediate hosts provided material for a comparative study of presomal development in Prosthorhynchus formosus (Van Cleave, '18) Travassos, '26, Prosthenorchis elegans (Diesing, 1851) Travassos, '15, and Moniliformis dubius Meyer, '33. Acanthellae begin development soon after entering intermediate hosts' hemocoels, and by the 18th day all three species possess three nuclear masses representing primordia of the proboscis, proboscis receptacle and ganglion, and trunk musculature and genitalia. Presomal development of P. formosus and P. elegans results in structures concurring with morphology of other adult palaeacanthocephalans and archiacanthocephalans. Development of M. dubius, however, differs from that of other archiacanthocephalans in that the muscular receptacle wall lines the entire surface of the nonmuscular sheath, failing to form a ventral cleft characteristic of other archiacanthocephalans. Unlike receptacle protrusor muscles of other archiacanthocephalan species, those of M. dubius spiral around the receptacle as they extend posteriad to attach individually to a pouchlike, muscular thickening at the receptacle's base. These protrusor muscles are distinct from the receptacle wall, as attested by their development alongside neck retractor muscles, not from the receptacle primordium, and the manner in which they are left to trail behind the receptacle when it is drawn anteriorly into the proboscis during larval development. The proboscis receptacle of M. dubius should not be thought of as being double-walled, as envisioned by previous workers.  相似文献

Nematospiroides dubius, unlike the closely related nematode Nippostrongylus brasiliensis, lacks a sterol requirement for the completion of its development to the L3 stage in an aqueous suspension of formalin-killed Escherichia coli. These larvae were justascapable of completing their development to adults in mice as those grown on charcoal-feces cultures. In addition, sterols had a toxic effect on the larvae of N. dubius which was removed by substituting a Krebs-Ringer solution for distilled water.In order to interpret the absence of a sterol requirement for the development of N. dubius larvae, an analysis of the egg lipids for both species was made. N. brasiliensis had about twice the sterol concentration of N. dubius eggs. A comparison of the development for both species showed, however, that N. brasiliensis grows more than twice as much as N. dubius to reach the L3 larval stage. Therefore, the amount of endogenous sterol is insufficient for N. brasiliensis to develop from the egg to the L3 stage.N. dubius and N. brasiliensis also differ in their porphyrin requirements. Unlike N. brasiliensis, coproporphyrin did not increase the size of N. dubius larvae so that they were comparable to those grown on charcoal-feces cultures. Coproporphyrin did result in larvae that were more robust in appearance than those grown without a supplement.  相似文献

Nollen P. M. 1984. Mating behavior of Philophthalmus megalurus and P. gralli in concurrent infections of chicks. International Journal for Parasitology14: 71–74. Mating patterns were determined in concurrent infections of the eyeflukes of birds, Philophthalmus megalurus and P. gralli, by detection of 3H-tyrosine-labeled sperm on autoradiograms. When labeled P. megalurus adults were transplanted singly with unlabeled P. gralli adults, interspecies mating took place. When both unlabeled P. gralli and P. megalurus were transplanted with single labeled P. megalurus, sperm were transferred to both species. However, the insemination rate of P. gralli was much lower than when unlabeled P. meglaurus adults were absent. Labeled P. gralli transplanted singly with unlabeled P. megalurus or unlabeled adults of both species did not show interspecies mating, but carried on intraspecies mating at a normal rate. A low rate of self-insemination was observed in labeled worms when the opposite species was present, but not when the same species was present. Evidence for interspecies fertilization and hybridization between P. megalurus and P. gralli was not found.  相似文献

 The taxonomic status of two nominal species of Polynemus, viz. P. dubius Bleeker, 1853 and P. longipectoralis Weber and de Beaufort, 1922, is revised. Although regarded as separate taxa up to the present time, examination of the holotype of P. longipectoralis revealed its close similarity to the type series of P. dubius, in the synonymy of which the former is now included. Polynemus dubius is redescribed as a valid species and a lectotype of the species is designated. In addition, a new species, P. aquilonaris, previously identified as P. dubius or P. longipectoralis, is described from Indochina on the basis of 28 specimens. Polynemus aquilonaris differs from P. dubius in having higher counts of pored lateral-line scales [80–86 (mode 81) vs. 69–79 (78) in the latter] and scale rows below the lateral line [14–17 (mode 14, rarely 13 or 17) vs. 13 (rarely 12)], and lower counts of gill rakers [25–29 (mode 27) vs. 29–33 (30), respectively]. The former is known from Indochina (Chao Phraya and Mekong River systems including Lake Tonle Sap), whereas the latter is currently known from the Malay Peninsula, Sumatra, and Kalimantan. Proteases, lipase, and chitinase killed Tylenchorhynchus dubius in vitro and in soil. Tylenchorhynchus dubius was more susceptible to the enzymes than Pratylenchus penetrans. Papain was the most effective protease, and other enzymes were less effective. Heating enzymes to 80 C for 10 min greatly reduced nematicidal effectiveness. Scanning electron micrographs showed that papain and chitinase produced structural changes in the cuticle of T. dubius. Lipase removed a thin outer layer. Papain removed material filling the striata, or furrow, between the horizontal bands. When added to soil, chitinase, lipase, collagenase, and proteases (papain and bromelain) decreased motility of T. dubius populations up to 75%. Bromelain was the most active in soil against T. dubius, and collagenase was the most active in soil against P. penetrans.  相似文献

This study demonstrates that homoeologous genes in two diploid plant species that specify different amounts of an enzyme maintain the same relative level of expression in an allotetraploid derivative. The three predominant alcohol dehydrogenase (ADH) isozymes (DD, DP, PP) in seeds of the recently evolved allotetraploid plant Tragopogon miscellus (Compositae) are dimers specified by Adh3-D and Adh3-P genes derived from its diploid progenitors T. dubius and T. pratensis. Seeds of T. pratensis contain twice as much ADH activity as those of T. dubius, while T. miscellus is intermediate. The three isozymes were similar in a number of catalytic properties; the densitometric ratio of the isozymes purified from T. miscellus was 1 DD:4DP:4PP for both ADH activity and protein; and dissociation-reassociation of the DP enzyme gave a 1:2:1 ratio of the three isozymes. Therefore, the enzymes were similar in specific activity, but twice as many P as D subunits were present in active enzymes in T. miscellus, precisely the difference in activity between the parents. In T. miscellus, the specific activity of ADH and its activity per mg tissue are intermediate to those of the diploids, because relative expression of the Adh gene in each genome is not influenced by the presence of the other genome.  相似文献

The American cockroach, Periplaneta americana, excretes feces containing aggregation pheromones to attract conspecific individuals. It is thought that these pheromones play an important role in establishing and maintaining colonies. However, despite previous extensive efforts, the aggregation pheromones of P. americana have not been isolated. It is also unclear whether the aggregation pheromones of P. americana are truly biosynthesized by the insect, as most previous experiments extracted chemicals from feces. Here, we investigate the tissue localization of aggregation agents in P. americana. To reduce the effects of diet-derived odorants, we developed a new diet for P. americana consisting of only agar and sugars, and tested the attractiveness of fecal extracts from animals reared on this new diet. Our results show that the aggregation agents of fecal extracts are insect-derived, and the extracts from the midgut evoke stronger behavioral responses than those from other parts of the alimentary tract. This suggests that the midgut may be the production site or the storage organ of the aggregation agents. Thus, our data will facilitate the future identification of the aggregation pheromones of P. americana.  相似文献

The Pantanal is the largest wetland in the world with extremely high plant and animal diversity, but large areas have been invaded by Vochysia divergens Pohl (Vochysiaceae), a tree that is native to the Amazon Basin, and Curatella americana L. (Dilleniaceae), a tree that is native to the Brazilian savanna (cerrado). V. divergens is reportedly floodadapted, thus its ability to invade the Pantanal may not be surprising, but the invasion of C. americana is counterintuitive, because this species is adapted to the well-drained soils of the cerrado. Thus, we were interested in comparing the photosynthetic capacity, in terms of CO2 conductance, carboxylation, and electron transport of these species over a seasonal flooding cycle. Given that V. divergens is reportedly flood-adapted, we predicted that this species would have a higher photosynthetic capacity than C. americana, especially under flooding. To test this hypothesis we measured the photosynthetic CO2 response (P N/C c) of V. divergens and C. americana within 1 year to determine, if photosynthetic capacity varied systematically over time and between species. Contrary to our hypothesis, V. divergens did not always have a higher photosynthetic capacity than C. americana. Rather, species differences were influenced by temporal variations in flooding and the leaf age. Leaf CO2 assimilation and photosynthetic capacity of both species were lower during the flood period, but the differences were not statistically significant. The physiological performance of both species was strongly related to leaf N and P concentrations, but P limitation appeared to be more important than N limitation for these species and ecosystem. Photosynthetic capacity was higher and more stable for V. divergens, but such an advantage did not result in a statistically significant increase in P N. Our results suggest that both species are tolerant to flooding even though they are adapted to very different hydrological conditions. Such physiological plasticity, especially for C. americana, might be a key feature for the ability to survive and persist in the seasonally flooded Pantanal.  相似文献

The Upper Pleistocene/Lower Holocene fossil-bearing sites of the Serra da Capivara National Park Region have yielded three cervid species: Mazama gouazoubira, M. americana and Blastocerus dichotomus, all currently living in South America, the two first in the region. A grand total of more than one hundred remains demonstrates the presence of Mazama gouazoubira in seven sites, mainly the Toca das Moendas, the Toca do Serrote do Artur, the Toca da Cima dos Pilão. This small species shows, since the Upper Pleistocene, a conspicuous tendency to reduce the average dimensions of its teeth and long bones. From the taller M. americana, only a dozen of remains were found in four sites, mainly the Sitio do Meio. In all of these it is sympatric with M. gouazoubira. It differs from this last one by its cheek teeth and its limb bones size and proportions. The oldest site where the species is known is Tarija (Bolivia, Middle Pleistocene) and it does not show any significant changes in size and proportions between recent and fossil samples. Sixteen remains of the large B. dichotomus were found in five sites, mainly the Toca das Moendas and the Toca da Barra do Antonião. The species is a rare fossil, but is frequently figured in the rock art painting of the region, where it is presently unknown.  相似文献

Dobson C., Brindley P. J. and Sitepu P. 1982. Influence of serum donor and recipient mouse genotype on the passive transfer of protective immunity with serum against Nematospiroides dubius. International Journal for Parasitology12: 567–572. Different strains of serum donor mice showed variations in innate immunity to primary infections with Nematospiroides dubius. Different levels of anti-N, dubius antibodies were detected in sera from these mouse strains; there was no correlation between antibody titre and numbers of worms recovered. Serum from donor wild and six laboratory strains of mice protected female Quackenbush (Q) recipients against N. dubius infections; donor mouse strain influenced the degree of protection conferred and donor serum antibody titre related to the degree of stunting of worm growth in recipient mice. Five laboratory strains of mice developed different levels of protective immunity following multiple experimental infections with N. dubius. Antibody titres in these mice were strongly correlated with the percentage protection observed after 1–4 infections: Q and CBA mice produced more anti-N. dubius antibody and were better protected than DBA/2, BALB/c and C3H mice. However BALB/c, C3H and CBA mice attained similar anti-N. dubius antibody titres after a single infection with N. dubius but serum from BALB/c gave better protection when transferred to female Q recipients than that from the other two strains. This suggested qualitative differences in the protective antibodies in sera between mouse strains. Five mouse strains were passively immunized with a uniform dose of serum from female Q donors: DBA/2 female recipients showed the least, BALB/c and C3H females were intermediate, and Q and CBA female mice attained the greatest level of passive protection against N. dubius. A close positive correlation existed between the degree of actively acquired and the level of passively acquired protection between the five strains of mice.  相似文献

The infectivity of Nematospiroides dubius was increased for Quackenbush (Q) mice by ten serial passages through this host. At the same time C(In3)H mice became more refractory to infection with successive Q generations of the parasite. Both strains of mice rejected the most highly selected parasites more readily than parasites from the earlier generations. These responses were shown to be immunological in nature by infections in hypothymic Balb/c CBA nu/nu and nu/+ mice and to be dose dependent. The selection of N. dubius for increased infectivity in Q mice enhanced its immunogenicity in this and other mouse strains possibly because of increased genetic homogenicity in the selected populations. N. dubius selected for Q mice showed a degree of immunologieal adaptation to Q but not to C3H mice.  相似文献

The isolation of a myotropic substance, proctolin, associated with the innervation of the viscera of the cockroach, Periplaneta americana, is reported. Proctolin, previously described as a neurotransmitter or neurohormone, is a small basic peptide with an estimated molecular weight of 500 to 700. The proctodeal (hindgut) muscles of P. americana contract at threshold levels of about 0·5 ng/ml, equivalent to about 10−9 M proctolin. Approximately 180 μg proctolin were isolated from 125 kg of whole cockroaches. The isolated peptide was homogeneous on paper and thin-layer chromatography and on high-voltage paper electrophoresis at pH 3·5 and 6·4.  相似文献

Jamesia (Hydrangeaceae) is endemic to western North America from the southern Rocky Mountains in southeastern Wyoming and Colorado west across the Great Basin to the southern Sierra Nevada and south through New Mexico and southeastern Arizona to Chihuahua and Nuevo León. Its distinctiveness and Oligocene fossil record suggest that it is an old genus. The genus comprises five geographically distinct taxa that can be grouped into two species. One of these,Jamesia americana, is further divided into four varieties, the second of which is new, and the fourth of which assumes a name older than that in current use:J. americana var.americana, var.zionis, var.macrocalyx, and var.rosea. The plant-parasitic nematodes Criconemoides lobatum, Hoplolaimus tylenchiformis, and Tylenchorhynchus dubius were present in the top 7.5 cm of sod consisting of numerous stolons and fibrous roots. Phenamiphos and 1,2-dibromo-3-chloropropane (DBCP) controlled all three species, whereas ethoprop and oxamyl controlled H. tylenchiformis and T. dubius but not C. lobatum. Benomyl and carbofuran controlled H. tylenchiformis but had poor control of C. lobatum and T. dubius. The effectiveness of carbofuran varied with the type of formulation, being most effective as a quick-release formulation. C. lobatum was the most difficult to control with chemicals. No chemical treatment improved the growth of ''Astoria colonial'' bentgrass (A. tenuis Sibth.) or Kentucky bluegrass (Poa pratensis L.) under the moist condtions prevalent in these tests.  相似文献

Allergen extracts from dust mites and cockroaches commonly found in Korean homes were used to evaluate their enzymatic activity as they are believed to influence allergenicity. Allergen extracts were prepared from 3 dust mite species (Dermatophagoides farinae, D. pteronyssinus, and Tyrophagus putrescentiae) and 3 cockroach species (Blattella germanica, Periplaneta americana, and P. fuliginosa) maintained in the Korea National Arthropods of Medical Importance Resource Bank. Proteins were extracted in PBS after homogenization using liquid nitrogen. The activities of various enzymes were investigated using the API Zym system. No significant difference in phosphatase, lipase, or glycosidase activity was observed among the 6 allergen extracts, but much difference was observed in protease activity. Protease activity was assessed in more detail by gelatin zymography and the EnzChek assay. Extract from T. putrescentiae showed the highest protease activity, followed by those of the cockroach extracts. Extracts from D. farinae and D. pteronyssinus showed only weak protease activity. Gelatinolytic activity was detected mainly in a 30-kDa protein in D. farinae, a 28-kDa protein in D. pteronyssinus, a > 26-kDa protein in T. putrescentiae, a > 20-kDa protein in B. germanica, and a > 23-kDa protein in P. americana and P. fuliginosa. The information on various enzymatic activities obtained in this study may be useful for future studies. In particular, the strong protease activity found in cockroach extracts could contribute to sensitization to cockroach allergens, which is known to be associated with the development of asthma.  相似文献

DNA preparations obtained from several tissues of Rhynchosciara americana and two related species, R. milleri and R. papaveroi, were hybridized to R. americana rRNA. The percentage of hybridization was found to be higher in tissues with low polyteny than in tissues with high polyteny, suggesting a relationship between the amount of rDNA and the tissue polyteny. This could be explained by under-replication of ribosomal cistrons in polytene cells, such as those from the salivary gland. Only slight tissue-dependent changes in the percentages of hybridization can be observed in heterologous hybridization using Xenopus laevis rRNA. The possibility that these experiments could not detect differences in the amount of ribosomal cistrons among tissues is discussed. The female:male ratio for the percentages of hybridization in the salivary gland of R. americana agrees with the results obtained by in situ hybridization experiments (16, 17) which have shown that the rRNA cistrons are distributed among chromosomes other than chromosome X.  相似文献

There have been recent disagreements as to how many taxa of titi monkeys, genus Callicebus, occur in the region between the Purus and Madeira rivers in western Brazilian Amazonia. Three parapatric taxa were proposed for the area: Callicebus caligatus, Callicebus stephennashi, and Callicebus dubius, but the latter has recently been considered a synonym of C. caligatus, even though both form monophyletic groups and are morphologically distinct. We analyzed the geographic variation in the pelage of Callicebus occurring between the Madeira and Purus rivers and concluded that the phenotypes attributed to C. caligatus and C. dubius are not individual morphs, but rather well-marked and geographically restricted varieties. For this reason, we classify Callicebus caligatus as a polytypic species with two subspecies: Callicebus caligatus caligatus and Callicebus caligatus dubius. This classification is corroborated by molecular evidence as well. The morphological and distributional data indicate that Callicebus stephennashi is a hybrid form of C. c. caligatus and C. c. dubius, due to the presence of intermediate characters. Therefore, until more precise locality records are provided and further evidence is presented, we consider Callicebus stephennashi to be a homonym of the two parental forms.  相似文献

Pigmentation is one of the most variable traits within and between Drosophila species. Much of this diversity appears to be adaptive, with environmental factors often invoked as selective forces. Here, we describe the geographic structure of pigmentation in Drosophila americana and evaluate the hypothesis that it is a locally adapted trait. Body pigmentation was quantified using digital images and spectrometry in up to 10 flies from each of 93 isofemale lines collected from 17 locations across the United States and found to correlate most strongly with longitude. Sequence variation at putatively neutral loci showed no evidence of population structure and was inconsistent with an isolation-by-distance model, suggesting that the pigmentation cline exists despite extensive gene flow throughout the species range, and is most likely the product of natural selection. In all other Drosophila species examined to date, dark pigmentation is associated with arid habitats; however, in D. americana, the darkest flies were collected from the most humid regions. To investigate this relationship further, we examined desiccation resistance attributable to an allele that darkens pigmentation in D. americana. We found no significant effect of pigmentation on desiccation resistance in this experiment, suggesting that pigmentation and desiccation resistance are not unequivocally linked in all Drosophila species.  相似文献
