A qualitative and quantitative analysis of free amino acids and related compounds in the haemolymph of Rhynchosciara americana was carried out for different periods of the fourth larval instar. Threonine, serine, proline, and glutamic acid make up 50 per cent of the total free amino acids in R. americana haemolymph just before the larvae start spinning the communal cocoon; after this the titre of most of the amino acids declines continuously. There are few peptides but these are present in high titres; they consist of two to three amino acid residues, of which the most important are histidine and aspartic acid. The fall in the haemolymph amino acid and peptide titres is insufficient to account for the silk protein which accumulates on the communal cocoon during the same period. The results are consistent with a silk protein origin from haemolymph proteins and haemolymph free amino acids. The origin and metabolic rôle of some haemolymph ninhydrin-positive compounds are discussed. 相似文献

The amount of protein, carbohydrate, lipids, and free amino acids were examined in the spinning stage in the fat body, haemolymph, skeletal muscle, and gut of Rhynchosciara americana. Protein and lipids increase in the fat body soon after the animal stopped feeding, probably at the expense of the digestion of the gut contents and of the reserves of the gut wall. Afterwards there is a fall in protein and lipids in the fat body. Haemolymph protein rises a little at the beginning of spinning and then decreases steadily during cocoon production. Carbohydrate and free amino acids decrease from the beginning of spinning in all tissues studied. Quantitatively, the most important decrease of carbohydrate during spinning occurs in the fat body whereas that of free amino acids occurs in the haemolymph. Lipid increases during spinning in the skeletal muscle, probably due to enlargement of the lateral fat body which occurs as a contaminant in the skeletal muscle preparation. The Malpighian tubules contain a large amount of calcium carbonate, which is eliminated during spinning. A correlation of our chemical data with histochemical data recently published is presented and the physiological implications of our findings are discussed in comparison to other insects. 相似文献

The salivary secretion of Rhynchosciara americana was chemically analysed. The secretion shows a yellow colour, with a pH of 7·5 and protein as its major component (94·5 per cent of the secretion dry weight). Carbohydrates are minor components of the secretion which amount to 3·4 per cent of the secretion dry weight, of which 2·3 per cent are neutral carbohydrates and 1·1 per cent are galactosamine. The major amino acids present in the secretion proteins are aspartic acid, glycine, serine, and glutamic acid. The salivary secretion proteins can be separated into eleven protein fractions by urea-acrylamide gel electrophoresis from which nine fractions are PAS positive. The salivary pigment moves together with the protein fraction No. 8, which is quantitatively the most important one, and has spectral characteristics identical to a haemolymph pigment. The higher rate of gland protein labelling by 14C-phenylalanine determined in vivo and in vitro occurs around the middle of the spinning stage at the same time as the appearance of the large chromosomal puffs. The rôle of the salivary secretion in cocoon production is discussed. 相似文献

An acrylamide gel electrophoretical analysis of the haemolymph proteins of R. americana was carried out at different stages of development. In mature larvae there are about 14 haemolymph protein fractions from which one stains heavily and two others faintly for lipoprotein, while three fractions stain for glycoprotein. The haemolymph protein fraction with Rm 0·25 decreases remarkably in mass during spinning, while the others decrease to a lesser extent. The protein fractions could be used in cocoon spinning since previous work suggests that haemolymph proteins are a major pool of cocoon protein precursors. The finding of a protein fraction in the salivary glands with an electrophoretical mobility similar to that of the haemolymph fraction with Rm 0·25 reinforces our hypothesis. 相似文献

Among many species of insects, females gain fitness benefits by producing numerous offspring. Yet actions related to producing numerous offspring such as mating with multiple males, producing oocytes and placing offspring in sub-optimal environments incur costs. Females can decrease the magnitude of these costs by retaining gametes when suitable oviposition sites are absent. We used the pomace fly, Drosophila melanogaster, to explore how the availability of fresh feeding/oviposition medium influenced female fitness via changes in offspring survivorship and the modulation of gamete release. Availability of fresh medium affected the absolute number and temporal production of offspring. This outcome was attributable to both decreased larval survival under crowded conditions and to female modulation of gamete release. Direct examination of the number of sperm retained among the different female storage organs revealed that females ‘hold on’ to sperm, retaining more sperm in storage, disproportionately within the spermathecae, when exposed infrequently to fresh medium. Despite this retention, females with lower rates of storage depletion exhibited decreased sperm use efficiency shortly after mating. This study provides direct evidence that females influence the rate of sperm depletion from specific storage sites in a way that can affect both female and male fitness. The possible adaptive significance of selective gamete utilization by female Drosophila includes lowering costs associated with frequent remating and larval overcrowding when oviposition sites are limiting, as well as potentially influencing paternity when females store sperm from multiple males. 相似文献

An excitation-induced hypertrehalosemic (EXIT) response has been demostrated in the American cockroach, Periplaneta americana. The response occurs in normal insects and in insects that were ligated at the neck 24 hr prior to experimentation. Several known and putative insect neurotransmitters were tested for their ability to elicit rapidly a hypertrehalosemic state in the cockroach. Octopamine was the most effective hypertrehalosemic agent, and produced a 100% increase in haemolymph trehalose levels within 15 min of being injected into the haemocoel. Less pronounced hypertrehalosemic responses were obtained with dopamine and tyramine, but not with synephrine or 5-hydroxytryptamine. Hypertrehalosemia was observed also with normetanephrine, but not with isoproterenol. A slight inhibition of the EXIT response occurred with the β-blocking agents, propranolol and dichloroisoproterenol, but the monoamine oxidase inhibitors, iproniazid phosphate and nialamide, were without effect. It is suggested that the EXIT-response may be one of several octopamine-mediated responses to excitation in the insect. 相似文献

Two new saponins, agavasaponin E and agavasaponin H have been isolated from the methanolic extract of Agave americana leaves and their structures elucidated. Plant microRNA (miRNA) is a crucial regulator of gene expression. It has been reported that more than one miRNA/miRNA* duplex could be produced from a microRNA precursor (pre-miRNA). In this study, we performed a comprehensive search for the novel miRNA candidates on the pre-miRNAs of Arabidopsis. AGO1 enrichment, co-existence of the miRNA*-like coordinates, and unique genome-wide match sites were taken into consideration for candidate screening. As a result, 43 miRNA-like candidates derived from 25 pre-miRNAs were identified. Among these candidates, 31 strong candidates from 22 pre-miRNAs passed all the filtering steps. Interestingly, some of these miRNA-like candidates showed organ-specific expression patterns. After target prediction and degradome sequencing data-based validation, five miRNA candidate–target pairs (ath-miR863-5p.2–AT1G76550.1, ath-miR822.2–AT5G03552.1, ath-miR822.3–AT5G02350.1, sRNA4–AT1G66290.1 and sRNA6–AT1G66310.1) were identified, providing a basis for in-depth functional analysis of these miRNA candidates. Induction of egg diapause in the silkworm, Bombyx mori by some cephalo-thoracic organs of the cockroach, Periplaneta americana was examined. All tissues tested such as brain, corpora cardiaca, corpora allata, suboesophageal and thoracic ganglia and nerve cords between thoracic ganglia were able to produce diapause eggs in non-diapause egg producers both by transplantation and injection of their crude homogenates. The homogenate of thoracic ganglia was effective even in pharate adults with the suboesophageal ganglion removed or in isolated abdomens of pharate adults.From these results, it was surmised that some endocrine organs, as well as the central nervous system in the cephalo-thorax of Periplaneta americana, contained the active principle responsible for egg diapause in Bombyx mori. 相似文献

The identity of chrysanthemaxanthin as a major pigment in avocado pulp has been confirmed by MS and the identity of neoxanthin similarly established. A carbonyl pigment was identified as 3-hydroxy-sintaxanthin. Two new UV fluorescent apocarotenoids were isolated. On the basis of spectrum, behaviour in acid and MS, one of these is assigned the structure 5,8-epoxy-5,8-dihydro- 10′-apo-β-caroten-3, 10′-diol. The other has an acid labile pentaene chromophore, and structure (1) has been tentatively assigned on similar evidence. The sex attractant of the cockroach, Periplaneta americana, has been shown in earlier work to be largely inactivated by dissected midgut from males and from mated females. It is only slightly inactivated by the midgut from virgin females. In this paper, the sex pheromone inactivating system is further studied and shown to be active in late instar larvae of both sexes. In the male this pheromone inactivation is inhibited by piperonyl butoxide, a microsomal oxidase inhibitor. This compound appears to act on the midgut tissue directly. In the mated female, piperonyl butoxide has little effect. When the pheromone inactivating capacity is partitioned into soluble and tissue components, it appears that the soluble component is most active in the male, whereas the tissue component is most active in the female. Evidence from heat inactivation, trichloracetic acid precipitation, and the use of soy bean trypsin inhibitor, as well as the time course of the reaction, suggest that the factor or factors inactivating pheromone are proteins, probably enzymes. Evidence that at least part of the pheromone inactivating capacity is due to microsomal oxidases is considered. It is also observed that both pheromone and piperonyl butoxide absorb to membranes. 相似文献

The cockroach, Periplaneta americana represents a basal insect lineage that undergoes the ancestral hemimetabolous mode of development. Here, we examine the embryonic and post-embryonic functions of the hox gene Scr in Periplaneta as a way of better understanding the roles of this gene in the evolution of insect body plans. During embryogenesis, Scr function is strictly limited to the head with no role in the prothorax. This indicates that the ancestral embryonic function of Scr was likely restricted to the head, and that the posterior expansion of expression in the T1 legs may have preceded any apparent gain of function during evolution. In addition, Scr plays a pivotal role in the formation of the dorsal ridge, a structure that separates the head and thorax in all insects. This is evidenced by the presence of a supernumerary segment that occurs between the labial and T1 segments of RNAiScr first nymphs and is attributed to an alteration in engrailed (en) expression. The fact that similar Scr phenotypes are observed in Tribolium but not in Drosophila or Oncopeltus reveals the presence of lineage-specific variation in the genetic architecture that controls the formation of the dorsal ridge. In direct contrast to the embryonic roles, Scr has no function in the head region during post-embryogenesis in Periplaneta, and instead, strictly acts to provide identity to the T1 segment. Furthermore, the strongest Periplaneta RNAiScr phenotypes develop ectopic wing-like tissue that originates from the posterior region of the prothoracic segment. This finding provides a novel insight into the current debate on the morphological origin of insect wings. 相似文献

A complex pattern of chloroplast pigments was found to be common to pulp, peel and leaves of avocado. This is explained by the presence of chlorophyll in the ripe fruit and its peel. Two chromoplast-specific pigments were found in ripe pulp and are tentatively identified as α-citraurin and mimulaxanthin. The identity of the latter and of neochrome, was established inter alia by the MS. The xanthophylls occur esterified to a major extent in pulp but entirely free in peel. The coexistence of chloroplast and chromoplast pigments in avocado fruit make this a suitable object for the ultrastructural studies of the origin of the chromoplasts. 相似文献

Externally applied l-phenylalanine rapidly equilibrates with the cytosolic pool(s) in hyphae of emerged cultures of Penicillium cyclopium. If not incorporated into protein it is accumulated in the so called expandable pool, which is presumably localized in the vacuolar compartment. At high concentrations of exogenous l-phenylalanine practically all of the amino acid needed for protein synthesis comes from the extracellular source, contrary to alkaloid synthesis which under all conditions recruits more than 90% of the required l-phenylalanine from intracellular sources. A new and simple method of training intact cockroaches was designed. (1) Cockroaches were maintained before and during the experiment with a high motivation to look for sugar. They were kept in a chamber with water and yeast extract ad libitum but without any sugar. (2) Cockroaches were individually trained to associate two artificial scent signals, i.e. menthol and vanilla, with sugar and salt solutions. Discriminatory learning performance was measured by testing the scent preference before and after training. Care was taken to exclude other than olfactory cues in the discriminatory paradigm. (3) Cockroaches exhibited a spontaneous initial preference for vanilla over menthol. This preference could be modified by training. The insect showed fast learning and long retention. One training trial was enough to reverse the initial preference with a significant retention after 7 days. Reversal of trained preference was accomplished by retraining. (4) The method provides an opportunity to study sensory performance and memory consolidation in cockroaches. 相似文献

The haemolymph of the cockroach, Periplaneta americana, has been shown to possess pronounced lipolytic activity. The enzyme may be considered a ‘true’ lipase because of its ability to hydrolyse long chain fatty acid glycerides. The enzyme exhibits dual optima at pH 4·9 and 7·0 and is inhibited by fluoride ion. Lipase activity in the intact cockroach is unaffected by the corpus cardiacum hypolipemic factor; however, prolonged periods of starvation result in a significant decline in enzyme activity. 相似文献

Mevalonate kinase activity was demonstrated in acetone powder extracts from Agave americana leaves, flowers and scape. ATP was the most effective phosphate donor. The enzyme had an optimum pH of 7.9 in Tris-HCl buffer. Dialysis decreased the ability to phosphorylate mevalonic acid (MVA). Partially purified mevalonate kinase reached maximum activity in the presence of 2 mM Mn2+ or 6–8 mM Mg2+. Higher concentrations of Mn2+ were inhibitory, whereas higher concentrations of Mg2+ produced only a small decrease in the activity. The amount of mevalonate-5-phosphate (MVAP) formed depended on protein concentration and incubation time. During short incubations, the MVAP formed increased as protein concentration rose, whereas during prolonged incubations (1–6 hr), there was a decrease in the MVAP formed when a certain amount of protein was exceeded. It is suggested that MVAP formed was hydrolysed by a phosphatase present in the extracts. This interfering activity was eliminated when mevalonate kinase is partially purified. The apparent Km values of the enzyme from leaves were 0.05 mM for MVA and 0. 14 mM for ATP. Similar Km values are obtained with partially purified mevalonate kinase. The enzyme was purified by ammonium sulphate precipitation, Sephadex G-100 filtration and DEAE-Sephadex A-50 fractionation. 相似文献

Topical treatment of adult female Periplaneta americana with the anti juvenile hormone fluoromevalonate delays the formation of the first post-treatment oötheca. This effect is dose-related and can be counteracted by simultaneous application of a juvenile hormone analogue (hydroprene) or by a 5-fold excess of mevalonic acid. Although treatment with fluoromevalonate delayed the production of oöthecae, such treatment did not affect either the carrying period of oötheca or their weight or viability. The technique described permits rapid in vivo evaluation of candidate anti juvenile hormones as anti-gonadotropic agents and provides further opportunity for studying the role of the endocrine system in regulating reproduction in P. americana. 相似文献
