This study was undertaken to clarify the suppression phenomenon of sex pheromone production after mating and its relationship to the physiological mechanism in adult females of Helicoverpa assulta, and determine the mating factor from males causing depletion of sex pheromonc production. Sex pheromone production of H. assulta females was mostly terminated in 3 hours after mating. Mated females maintained with a low titer of sex pheromone until 3 days when it started to increase again, which showed a characteristic of species mating more than once. The mated female again produced pheromone upon injection of pheromone biosynthesis activating neuropeptide (PBAN) or extracts of brain-suboesophageal ganglion complexes (Br-Sg) of mated female, which were shown similar pheromonotropic activities as compared with virgin females. These results indicated that the mating did not inhibit the receptivity of pheromone gland itself and PBAN biosynthesis in suboesophageal ganglion of the mated females. And it seems to support that the depletion of sex pheromone production is responsible for blocking of PBAN release from head. To investigate the mating factor from adult males, when extracts of reproductive organs of male were injected into hemocoel of virgin females evoking depletion of sex pheromone production as shown in mated female. The results suggest that a chemical substance(s) from the male reproductive organs could be responsible for the loss of sex pheromone biosynthesis in H. assulta.

Male volatile mating pheromone is released by male houseflies placed in close proximity to females or in contact with female hydrocarbon extract. The pheromone induces the aggregation of virgin females in the vicinity of the male. Preparations of macerated testicular and ejaculatory duct tissue exert a similar effect, and these organs are, apparently, the source of the pheromone. Such preparations are not attractive to males or mated females. The pheromone also appears to induce female receptivity, since females that are exposed to emanations from male system preparations show a quicker rate of pairing with males. The pheromone effect results from the testes and ejaculatory ducts acting together, as preparations of each of these alone are inactive. Pheromone activity is age dependent and the greatest female reaction is observed when tissues of 6–10-day old males are used.

The present study investigates the effects of age and mating status on the circadian variations of gland sex pheromone titre in female Spodoptera litura Fabricius. Similar to other nocturnal moths, S. litura females exhibit circadian variations of gland sex pheromone contents, with higher levels during scotophase and lower levels during photophase. The sex pheromone titre in the glands peaks during the first scotophase after eclosion and sharply declines afterwards. Higher pheromone contents during scotophase may facilitate female reproductive activities, and the negative relationship between pheromone titre and female calling is likely the result of pheromone release during female calling. Interestingly, the present study demonstrates that mated S. litura females have significantly higher sex pheromone titre in their pheromone glands (PGs) than virgin females. This finding contrasts with all previous studies of other insect species, in which mating generally reduces the sex pheromone titre in female PGs. In S. litura, mating and male accessory gland fluids can suppress female calling behaviours and re‐matings. These results suggest that the suppression of female calling behaviours by mating and male accessory gland fluids may significantly reduce the release of sex pheromones and thus result in higher sex pheromone titre in the PGs of mated females.

ABSTRACT. The responses of Dasineura brassicae Winn. (Diptera, Cecidomyiidae) to odours from both male and female midges and from their host ( Brassica napus ) and non-host ( Vicia faba ) plants were studied using a Y-tube olfactometer. Female but not male D. brassicae are attracted to crushed B. napus leaves but not to those of V. faba. Males are attracted to live virgin females, to crushed virgin and mated females and to hexane washes of females, suggesting that the female produces a sex pheromone that attracts males. Males were also attracted to hexane washes of female ovipositors, but not to washes of the bodies of females without ovipositors, suggesting that the ovipositor is the site of pheromone release or production. Males were not attracted to live mated females or to other live or crushed males and mated females were not attracted to other mated females.

A secondary sexual character may act as an honest signal of the quality of the individual if the trait bears a cost and if its expression is phenotypically condition dependent. The cost of increasing the trait should be tolerable for individuals in good condition but not for those in a poor condition. The trait thus provides an honest signal of quality that enables the receiver to choose higher quality mates. Evidence for sex pheromones, which play a major role in shaping sexual evolution, inflicting a signaling cost is scarce. Here, we demonstrate that the amount of the major component of the pheromone in glands of Lobesia botrana (Lepidoptera) females at signaling time was significantly greater in large than in small females, that male moths preferred larger females as mates when responding to volatile signals, and small virgin females, but not large ones, exposed to conspecific pheromone, produced, when mated, significantly fewer eggs than nonexposed females. The latter indicates a condition-dependent cost of signaling. These results are in accordance with the predictions of condition-dependent honest signals. We therefore suggest that female signaling for males using sex pheromones bears a cost and thus calling may serve as honest advertisement for female quality.

The subtropical scarab beetle, Dasylepida ishigakiensis, has a two-year life cycle. This study showed the time of adult emergence from the soil relative to the time of dusk and the presence of the female sex pheromone. Beetles collected on Miyako Island were transported to Tsukuba where they were immediately placed under natural day lengths in February. They exhibited two emergence peaks that corresponded to the times of dusk in Tsukuba and on the island, respectively. Males emerged precociously if a lure containing synthetic female sex pheromone was placed in their container, whereas the females’ behavior was unaffected. Previous observations that mated females dig deeper in the soil than virgin females, males or mated males were confirmed. To explore the underlying mechanism controlling the behavioral change associated with mating, liquid material derived from the male accessory glands, seminal vesicles and female bursa copulatrix was injected into beetles, but without any significant influence on burrowing behavior. No significant influence was also observed in beetles injected with anisomycin, an inhibitor of protein synthesis related to memory in other animals.

Diaphorina citri Kuwayama (Hemiptera: Psyllidae) is an important worldwide pest of citrus. It vectors three phloem-restricted bacteria in the genus Candidatus Liberibacter that cause huanglongbing (citrus greening disease). Studies were conducted to examine the behavioral responses of male and female D. citri to conspecifics of the same and opposite sex, with and without associated citrus host plants, in both open-air arena choice assays and Y-tube olfactometer assays. Virgin and mated male D. citri colonized citrus plants that were currently or had been previously colonized by virgin or mated female D. citri in greater numbers than control plants without females. However, males or females did not accumulate more on plants colonized by conspecifics of the same sex compared to uninfested plants, and females showed no preference for plants pre-infested with males compared with uninfested controls. In complementary Y-tube olfactometer assays, virgin and mated males chose arms with odor sources from mated females compared with blank controls in the absence of associated citrus host plant volatiles. In both behavioral assays, mated female D. citri appeared more attractive than virgin females. The vibrational calling behavior of male D. citri was reduced when males were challenged by the odors of conspecific mated females relative to when males were challenged by the odor of other males. Collectively, our results provide behavioral evidence for a female-produced volatile sex attractant pheromone in D. citri . Future identification and synthesis of a sex attractant pheromone will be an important contribution to current monitoring and management practices for D. citri .

Prostaglandin (PG) synthetase was present in the testes, seminal vesicles, and spermatophores of the male house cricket, Acheta domesticus. The enzyme was not detected in bursa copulatrix, spermatheca, spermathecal canal, and oviducts from virgin females, while substantial activity was measured in the same tissue from mated females. The female appears to receive the enzyme from the spermatophore. A PGE2-like material was detected by radioimmunoassay in A. domesticus testes and to a lesser extent in the remainder of the male reproductive tract. PG went undetected in virgin female reproductive tissues, while the same tissues from mated females contained an average of 589 pg of PGE2-like material per female. In in vivo studies, injected PGE1, PGE2, and to a smaller degree PGF stimulated oviposition by virgin females. Moreover, N-acetyl-p-aminophenol, a PG synthetase inhibitor, suppressed oviposition in mated females. Post-copulatory PG biosynthesis in the female reproductive tract might be partially responsible for triggering oviposition in A. domesticus. Since PG synthetase appears to be acquired from the male, it could be considered a primer pheromone.

The potential for short‐range sex pheromone communication by the egg parasitoid wasp Trissolcus brochymenae (Hymenoptera: Platygastridae) was investigated in closed arena bioassays. Males of this parasitoid showed more antennal drumming and more frequent mounting behaviour on 1‐ to 2‐d‐old virgin females compared with 8‐d‐old virgin females. Male copulation attempts were fewer with previously mated females than with virgin females. Males courted and made copulation attempts with 1‐ to 2‐d‐old female cadavers, but not with male cadavers or with female cadavers rinsed in organic solvents of different polarities. Male attraction to female cadavers was re‐established by treating cadavers with acetone extracts of females, but not with ether or hexane extracts. In experiments using female cadavers dissected into head, mesosoma, and gaster, and then reassembled using one unwashed body section and two body sections washed in acetone, males were attracted only to the reassembled cadavers with an unwashed mesosoma. These findings suggest that (1) courtship behaviour in males of T. brochymenae is triggered by a short‐range sex pheromone produced by females; (2) the age and the physiological condition of females (virgin/mated) influence pheromone release or production; (3) the female's mesosoma is the source of the sex pheromone; and (4) polar components of the sex pheromone play a major role in influencing male behaviour. Our results suggest that quasi‐gregarious egg parasitoids are selected for short‐range rather than long‐range sex pheromones.

Mechanisms and genetics of resistance to pyrethroid insecticides were investigated in a strain of house fly (ASPR) collected from a cattle ranch in Miyagi, Gunma Prefecture, Japan. Flies were selected in the laboratory with the pyrethroid insecticide permethrin. Both sexes were resistant to pyrethroids; however, females were 22- to 245-fold more resistant than males. Permethrin resistance could be partly suppressed by the monooxygenase inhibitor piperonyl butoxide in females, but not in males. In this strain, sex was determined by a male factor on the third autosome. The relationship of the autosomal male factor to the lower resistance levels observed in the males and the mechanisms of resistance expressed in each sex are discussed.

Reproductive behavior in Drosophila has both stereotyped and plastic components that are driven by age- and sex-specific chemical cues. Males who unsuccessfully court virgin females subsequently avoid females that are of the same age as the trainer. In contrast, males trained with mature mated females associate volatile appetitive and aversive pheromonal cues and learn to suppress courtship of all females. Here we show that the volatile aversive pheromone that leads to generalized learning with mated females is (Z)-11-octadecenyl acetate (cis-vaccenyl acetate, cVA). cVA is a major component of the male cuticular hydrocarbon profile, but it is not found on virgin females. During copulation, cVA is transferred to the female in ejaculate along with sperm and peptides that decrease her sexual receptivity. When males sense cVA (either synthetic or from mated female or male extracts) in the context of female pheromone, they develop a generalized suppression of courtship. The effects of cVA on initial courtship of virgin females can be blocked by expression of tetanus toxin in Or65a, but not Or67d neurons, demonstrating that the aversive effects of this pheromone are mediated by a specific class of olfactory neuron. These findings suggest that transfer of cVA to females during mating may be part of the male's strategy to suppress reproduction by competing males.

Mating results in a strong suppression of sex pheromone (bombykol) production in the female silkworm moth, Bombyx mori. The mechanical stimulation from the insertion of a penis, inflation of the bursa copulatrix (BC), or copulation with the sterile male whose penis was removed in order to prevent ejaculation (pr-male) induced only a partial decline in bombykol production. Artificial insemination stimulates oviposition of fertilized eggs as does normal mating. However, bombykol production did not decline in artificially inseminated females. When females were artificially inseminated before or after mating with pr-males, some females had a small amount of bombykol, similar to females mated with normal males, while other females had a large amount of bombykol similar to virgin females. The former usually laid fertilized eggs, while the latter laid only unfertilized eggs though semen filled their spermatophores and spermathecae. The mechanical stimulation caused by mating with a pr-male could be replaced by covering the abdominal tip with melted paraffin. Neither implantation of the BC obtained from mated females, nor injection of the spermatophore extract, into a female mated with a pr-male could inactivate bombykol production. Injection of hemolymph from a mated female into a virgin also failed to affect bombykol production These results indicate that a combination of both the tactile stimulation of the abdominal tip and the arrival of fertile spermatozoa in the vestibulum trigger a neural inactivation mechanism of bombykol production after mating.

An oxidase that converts primary aliphatic alcohols into aldehydes was discovered in the cuticle of the sex pheromone gland and in the papillae anales on the tip of the abdomen of Manduca sexta females. Oxidase activity was not found in the epidermal cells of the pheromone gland where fatty acid precursors of the pheromonal aldehydes are found. This oxidase requires oxygen and water to function and appears to have a rather broad substrate specificity. The activity of the oxidase is reduced by the application of piperonyl butoxide, which also interferes with the PBAN induced production of the natural pheromone aldehydes. However, endogenous alcohols cannot be found in the pheromone gland. Thus, it is not yet clear whether or not the oxidase is involved in the terminal step of biosynthesis of the pheromone aldehydes in M. sexta females. © Wiley-Liss, Inc.

The presence of a mounting sex pheromone was demonstrated on the surface of fed female Rhipicephalus appendiculatus. This pheromone, which is present on the female cuticle, allows the male to recognise the female. The pheromone was removed by cleaning the female in hexane, resulting in the loss of male mating behaviour in in vitro experiments. Male mating behaviour was resumed when extract made from fed female cuticle was replaced on cleaned females. When the extract was transferred to innanimate objects typical male mating behaviour was released. Preliminary chemical analyses indicated that the active component of the extract was contained in the sterol ester fraction of the extract.

Causes, consequences and pheromonal regulation of male contest and female choice in the staphylinid beetle, Aleochara curtula (Goeze), have been investigated in the field and in the laboratory. At the feeding and mating site (carcass) the sex ratio is male biased. Polyandry is affected by prolonged copulations, spermatophore plugs and anti-aphrodisiac pheromones transferred from the male, and by female repulse behaviour as well. Aggression of competing males leads to expulsion of inferior males from the carcass. Young, starved and multiply mated males, which need access to the food resource, produce the female sex pheromone. They release homosexual responses, but also avoid intrasexual aggression. On the other hand, females behave aggressively towards individuals bearing the female sex pheromone or repulse their copulatory attempts. Those males of insufficient physiological condition produce a lighter spermatophore and fertilize less eggs. The adaptive significance of female mimicry, male mating tactics, and female choice is discussed.

Sex pheromone titers in females of two tortricid moths, Epiphyas postvittana and Planotortrix octo, did not significantly vary between the scotophase and photophase. Pheromone production in these two species is controlled by a factor located in the head of the respective females, probably the pheromone biosynthesis-activating neuropeptide (PBAN). Unlike that reported for the related tortricid, Argyrotaenia velutinana, the bursa copulatrix in female E. postvittana and P. octo does not appear to contain a factor that stimulates pheromone production. After mating, female E. postvittana permanently shut down pheromone production. In contrast, pheromone titer in mated P. octo females is reduced to a level approximately half that of similar-age virgins. While the abdominal nervous system is involved in the inactivation of pheromone production in mated E. postvittana females and probably acts to stop release of PBAN from the corpora cardiaca, the abdominal nervous system is not involved in effecting the decreased pheromone titers of mated P. octo females. It is possible that in the latter species, a humoral factor(s) is responsible for effecting the decreased pheromone titers, possibly through affecting the release of PBAN from the corpora cardiaca. Bioassaying head extracts allowed changes in PBAN titer in female E. postvittana to be inferred. PBAN titers remain roughly constant in virgins but increase after mating. This suggests that PBAN is biosynthesized throughout the life of an adult virgin female at approximately the same rate as it is released. Furthermore, it appears that the decline in pheromone titer observed in older E. postvittana females is probably due to a decline in competency of the gland to produce pheromone rather than to a decrease in PBAN titer in older females.

Direct evidence is presented for the role of a cytochrome P-450 monooxygenase (called mixed-function oxidase, or polysubstrate mono-oxygenase, PSMO) in the metabolism of the sex pheromone (Z)-9-tricosene to its corresponding epoxide and ketone in the housefly. A secondary alcohol, most likely an intermediate in the conversion of the alkene to the ketone, was also tentatively identified. The results of in vivo and in vitro experiments showed that the PSMO inhibitors, piperonyl butoxide (PB) and carbon monoxide, markedly inhibited the formation of epoxide and ketone from (9,10-3H) (Z)-9-tricosene. An examination of the relative rates of (Z)-9-tricosene metabolism showed that males exhibited a higher rate of metabolism than females with the antennae of males showing the highest activity of any tissue/organ examined. The major product from all tissues/organs was the epoxide. Data from experiments with subcellular fractions showed that the microsomal fraction had the majority of enzyme activity, which was strongly inhibited by PB and CO and required NADPH and O2 for activity. A carbon monoxide difference spectrum with reduced cytochrome showed maximal absorbance at 450 nm and allowed quantification of the cytochrome P-450 in the microsomal fraction of 0.410-nmol cytochrome P-450 mg?1 protein. Interaction of (Z)-9-tricosene with the cytochrome P-450 resulted in a type I spectrum, indicating that the pheromone binds to a hydrophobic site adjacent to the heme moiety of the oxidized cytochrome P-450.

ABSTRACT The amount of the major component of the cuticular sex pheromone, 3, 11-dimethyl-2-nonacosanone, on individual female German cockroaches, Blattella germanica (L.), as a function of age was determined by gas-liquid-chromatographic analysis. Accumulation of phermone increased with age in both virgin and mated females. During the first gono-trophic cycle, the pheromone accumulated most rapidly when oocyte growth rates were maximal (days 5–10), and least rapidly while the female carried an ootheca (days 11–32). Pheromone accumulation was similar in virgin and mated females when the same physiological stages (determined by oocyte maturation) were considered. Inhibition of Juvenile Hormone release, through allatectomy, chemicals (precocene or fluoromevalonate), or through mechanical egg case implants, suppressed or delayed pheromone production and oocyte growth. The Juvenile Hormone analogue ZR512 induced allatectomized or head-ligated females and females with chemically or mechanically inhibited corpora allata to produce pheromone and enlarge their basal oocytes. Finally, ZR512 applied to intact females stimulated pheromone production in a dose-dependent manner.
