Summary An electronic simulated target apparatus was used in a two-experiment study to compare the target detection performance of an echolocating bottlenose dolphin with an optimal receiver. Random Gaussian noise with a relatively flat spectrum from 20 to 160 kHz was used as a masking source. Experiment I was conducted to establish a technique for estimating the echo energy-to-noise ratio,E e /N, at the dolphin's threshold of detection. Dolphins typically vary the amplitude of their emitted signal over a large range making it difficult to estimateE e /N. In the first part of experiment I, the simulated echo was a double click, the pulses separated by 200 s, with each pulse being a replica of the dolphin's transmitted signal. A staircase psychophysical procedure was used to obtain the detection threshold, and the echo energy-to-noise ratio based on the highest amplitude click emitted per trial, (E e /N)max, was determined at each reversal point. The second echo type consisted of one of the animal's echolocation clicks, previously measured, digitized and stored in an erasable programmable read-only memory (EPROM). The electronic target simulator was modified so that every time the dolphin emitted an echolocation signal, the EPROM was triggered to produce two pulses separated by 200 s. On any trial, the EPROM signal was played back at a fixed amplitude, regardless of the amplitude of the dolphin's emitted signal. TheE e /N obtained with the EPROM signal at threshold was found to be 2.9 dB lower than (E e /N)max obtained with the normal phantom target. Therefore an estimate ofE e /N can be obtained by subtracting 2.9 dB from (E e /N)max.Experiment II was conducted to obtain isosensitivity data that could be plotted in an ROC (receiver operating characteristic) format. The response bias of the dolphin was manipulated by varying the food reinforcement payoff matrix. In terms of the ratio of correct detections to correct rejections, the payoff matrix was varied over four values: 11, 41, 81, and 14. A modified method of constants procedure was used to obtain the dolphin target detection performance data. Each session consisted of two 20-trial blocks in which a strong echo was used in the first block and a weak echo in the second block. The energy-to-noise ratio required by an optimal detector to approximate the dolphin's performance was obtained by determining the appropriate detection sensitivity,d, that best fitted the dolphin's data plotted in an ROC format. Molecular and biochemical techniques have been used to explore the reasons behind low E chain expression in the E + E I-region recombinant strain, A.TFR5. A.TFR5 (A f E k, ap5), a recombinant between A.CA (A f E f) and A.TL (A k E k), carries the E k subregion. Previous results have shown that it expresses the E chain, but at reduced levels relative to E + E + strains. No E chains were detected, which is consistent with the A.TFR5E gene being derived from the A.CA parent, which carries the null E f allele. In this paper, the defect in E-chain expression is explored. Restriction fragment length polymorphism analysis has localized the recombination event in A.TFR5 approximately 30 kb upstream of E, in the region of the large intervening sequence of E. Northern blot analysis of total RNA from A.TFR5 shows normal amounts of the E message, but no E message. Two-dimensional gel analysis of 15 min pulse-labeled A.TFR5, A.CA, and A.TL E immunoprecipitates shows decreased levels of the intracellular E chain in A.TFR5 relative to A.TL. However, analysis of total cell extracts shows normal levels of this protein. A glycoprotein fraction isolated from total cell extracts of 5 h labeled cells contains normal amounts of intracellular E, but decreased amounts of the mature cell-surface protein. These data suggest that in the absence of E, the E chain (1) takes on an altered conformation that is not as efficiently recognized by alloantibodies, and (2) is found in normal levels as the partially glycosylated intracellular precursor, but is not processed and/or transported efficiently to the cell surface.  相似文献

Summary Big brown bats (Eptesicus fuscus) were tested for their ability to detect an electronically simulated target, and to discriminate differences in range to two simulated targets, when receiving either a model of their own sonar emissions or the model reversed in time as the echo. The theory of matched detection predicts a large decrease in performance if bats use matched filtering, unless they are somehow able to adjust their filter to match the novel, time-reversed signal. The detection thresholds we obtained were much lower than Møhl's (1986), but like him we found no difference in threshold for reversed models (Table 2). This suggests either that bats do not use matched filtering for target detection, or, possibly, that they are able to adapt their filter to a highly unnatural signal in some way as yet unknown.Unlike detection, range discrimination was much poorer with reversed echoes (Table 3). Threshold increased from about 1 cm range difference with normal model echoes to 18 cm or more with reversed model echoes. This suggests that range determination, which is based on measuring the time of arrival of echoes, does involve matched filtering. Whether such filtering is ideal (i.e., equivalent to cross-correlation detection) cannot be decided by our results, but there are some indications that the match between an echo and the presumed internal template (the match of matched filtering) must be fairly precise. Also, since performance with phantom targets generated using model echoes was as good as has been found with real targets, the internal template is probably fixed (or only slowly modifiable) rather than re-programmed with each sonar emission. Finally, because synchronization of emission and model echo was not perfect, the apparent distance to targets probably varied by 2 to 4 cm from emission to emission, although both targets would appear to move together thus keeping the range difference constant. Summary Mössbauer spectroscopy was used to investigate the cellular acquisition of iron byPseudomonas aeruginosa which had been incubated with ferripyoverdine for 20, 40, 60, 120 or 360 min. Studies revealed that no ferripyoverdine accumulated in the cells at any of these times and that the amounts and kinds of iron complexes produced by cellular metabolism vary with time. At 20 and 40 min a ferric species, with isomer shift =0.38–0.42 mm/s and quadrupole splitting E Q=0.94–0.92 mm/s, was the major iron metabolite comprising approximately 80% of the iron. At later times at least three other ferric species appeared with =0.54 0.72, E Q = 0.84 1.07 mm/s. Ferrous species, =1.43 1.77 mm/s and E Q = 2.69 1.82 mm/s, were also seen at times as early as 20 min and comprised as much as 17% of the total iron at 20 and 40 min. The parameters of all these species identify them as being six-coordinated high-spin complexes. In addition a low-spin species, =0.19 mm/s E Q=0.67 0.91 mm/s, never before reported in cells, appeared at 60, 120, and 360 min as one of the major iron metabolites (50% or more). Crude extracts or supernatants of broken cells of Clostridium formicoaceticum reduce unbranched, branched, saturated and unsaturated carboxylates at the expense of carbon monoxide to the corresponding alcohols. The presence of viologens with redox potentials varying from E 0=-295 to-650 mV decreased the rate of propionate reduction. The more the propionate reduction was diminished the more formate was formed from carbon monoxide. The lowest propionate reduction and highest formate formation was observed with methylviologen. The carbon-carbon double bond of E-2-methyl-butenoate was only hydrogenated when a viologen was present. Formate as electron donor led only in the presence of viologens to the formation of propanol from propionate. The reduction of propionate at the expense of a reduced viologen can be followed in cuvettes. With respect to propionate Michaelis Menten behavior was observed. Experiments are described which lead to the assumption that the carboxylates are reduced in a non-activated form. Synopsis The standard and routine oxygen consumptions of Astyanax fasciatus from one surface population (Rio Teapao) and three cave populations (Chica, Micos and Pachon caves: sAnoptichthys jordani, the Micosfish and Anoptichthys antrobius) were determined individually over 24 hours by the use of a flow-through respirometer and polarographic oxygen electrodes. The phylogenetically oldest Pachon fish had a significantly lower standard metabolic rate (0.230 ± 0.036 mg O2 g-1 h-1) than the epigean Teapao fish, the hybrid Chica fish and the phylogenetically younger Micos fish (0.314 ± 0.081 mg O2g--1h-1, 0.284 ± 0.048 mg O2g-1h-1, 0.277 ± 0.063 mg O2g-1h-1). No significant differences could be determined among the latter three populations. A significant difference in routine metabolic rate existed only between the Pachon fish (0.309 ± 0.0.56 mg O2g-1h-1) and the Teapao fish (0.415 ± 0.071 mg O2g-1h-1). The Chica fish (0.356 ± 0.084 mg O2g-1h-1) and the Micos fish (0.355 ± 0.080 mg O2 g-1h-1) could not be separated from either the Teapao or the Pachon fish, but a decreasing trend from the surface population through the Chica and the Micos to the Pachon population was obvious. During a starvation period of 29 days the metabolic rate of epigean Teapao and hypogean Pachon fish decreased significantly by 32.5% and 34.8% (standard oxygen consumption rate) and 27.5% and 28.2% (routine oxygen consumption rate), respectively. Body mass loss during the starvation period was 16.3% for the Teapao fish and 9.5% for the Pachon fish.  相似文献

We determined for Vicia faba L the influence of nitrogen uptake and accumulation on the values of photon saturated net photosynthetic rate (P Nmax), quantum yield efficiency (), intercellular CO2 concentration (C i), and carboxylation efficiency (C e). As leaf nitrogen content (NL) increased, the converged onto a maximum asymptotic value of 0.0664±0.0049 mol(CO2) mol(quantum)–1. Also, as NL increased the C i value fell to an asymptotic minimum of 115.80±1.59 mol mol–1, and C e converged onto a maximum asymptotic value of 1.645±0.054 mol(CO2) m–2 s–1 Pa–1 and declined to zero at a NL-intercept equal to 0.596±0.096 g(N) m–2. fell to zero for an NL-intercept of 0.660±0.052 g(N) m–2. As NL increased, the value of P Nmax converged onto a maximum asymptotic value of 33.400±2.563 mol(CO2) m–2 s–1. P N fell to zero for an NL-intercept of 0.710±0.035 g(N) m–2. Under variable daily meteorological conditions the values for NL, specific leaf area (L), root mass fraction (Rf), P Nmax, and remained constant for a given N supply. Plastic and elastic in-vitro extensibilities (E pland E el ) of cell walls from growing maize (Zea mays L.) coleoptile segments were measured by stretching frozen-thawed tissue, pre-extended to its in-vivo length, at constant force (creep test) in a custom-buildt extensiometer, equipped with a linear-displacement transducer. The indole-3-acetic acid (IAA)-induced change of E pl (E pl ) is strictly correlated with the growth rate for a period of 3–4 h. Subsequently, E plremains constant while the growth rate is slowing down. Since this discrepancy can be accounted for by a growth-dependent reduction of osmotic pressure, it is concluded that E plrepresents quantitatively the relative increase of in-vivo extensibility (cell wall loosening) involved in IAA-mediated cell growth over a much longer time. On the other side it is argued that the growth rate may not be strictly correlated with wall extensibility during long-term growth. Abscisic acid (ABA) inhibits segment growth induced by auxin, fusicoccin, or exogenous acid, and this effect can be quantitatively attributed to an ABA-mediated reduction of cell wall extensibility as determined by the E plmeasurement. Both, IAA and ABA have no effect on total protein synthesis, RNA synthesis, and amount of osmotic solutes. Fusicoccin-induced proton excretion is only slightly inhibited by ABA. In contrast to ABA, growth inhibition by cycloheximide (CHI) is always much larger than the concomitant reduction of E pl , indicating that a further growth parameter is also involved in the inhibition of cell growth by CHI. E el is not affected by either IAA, ABA, or CHI. It is concluded that E pl as determined by the applied method, represents a relative measure of the actual in-vivo extensibility of the growing cell wall at the very moment when the tissue is killed, rather than an average extensibility accumulated over some immediate-past period of time as suggested by Cleland (1984, Planta 160, 514–520). Laboratory tests were conducted to determine the effects of five pyrethroid insecticides—permethrin (FMC 33297) [3-phenoxybenzyl (±)-3-(2,2-dichlorovinyl)-2,2-dimethylcyclopropanecarboxylate]; FMC 45498 [(S)--cyano-3-phenoxybenzyl-(R)-cis-2-(2,2-dibromovinyl)-3,3-dimethylcyclopropanecarboxylate]; Shell WL 41706 [(±)--cyano-3-phenoxybenzyl 2,2,3,3-tetramethylcyclopropane-carboxylate]; Shell WL 43467 [(±)--cyano-3-phenoxy benzyl (±)-cis,trans-2-(2,2-dichlorovinyl)-3,3-dimethylcyclopropanecarboxylate]; and Shell WL 43775 [(±)--cyano-3-phenoxybenzyl (±)-2-(4-chlorophenyl)-3-methylbutyrate]—at 0.5 and 5g/g on microbial populations and activities in a sandy loam. The insecticides had antimicrobial activity in early stages of incubation. The populations recovered after 2 to 4 weeks and stimulatory effects on populations were also observed in later stages. No inhibition of acetylene (C2H2) reduction was evident with any of the insecticides. However, WL 43467 at both concentrations and permethrin and WL 41706 at 5 g/g increased nitrification after 4 weeks. Soil microbial respiration, as indicated by oxygen consumption, increased with increasing concentration of insecticides, suggesting the possibility of microbial degradation of the insecticides. Dehydrogenase activity showed that none of the insecticides inhibited formazan (2,3,5-triphenyltetrazolium formazan) formation, whereas urease activity was stimulated in most cases. The studies indicated that some of the pyrethroid insecticides may exert transient effects on populations and activities of the microflora in a sandy loam, but these were short-lived and minor in nature.  相似文献

Summary Determinations of current-voltage relationships are widely employed in the characterization of epithelial sodium transport. In order to determine the protocol dependence of transport parameters in the toad urinary bladder, studies were carried out in the presence and absence of amiloride, an inhibitor of active sodium transport. With symmetric positive and negative perturbations of the transepithelial electrical potential difference (0±100 mV) for 30 sec, the amiloride-sensitive current-voltage (i a -) relationship was near linear over the range –75+100 mV, indicating constancy of the conductance a and the apparent electromotive force E Na, lumped parameters of the standard electrical equivalent circuit model of the active transport system. With a reverse protocol (±1000 mV) or 15 min perturbations thei a - relationships were highly nonlinear. Nonlinearity reflected voltage dependence of parameters: perturbations that increased active transport decreased E Na and increased a, as evaluated from 10 sec perturbations of ; slowing of active transport produced the converse changes. These effects are usefully analyzed in both quasi-steady states and true steady states by means of a detailed equivalent circuit incorporating the significant ionic currents across each plasma membrane. Water relation parameters of leaf cells of the aquatic plant Elodea densa have been measured using the pressure probe. For cells in both the upper and lower epidermis it was found that the elastic modulus () and the hydraulic conductivity (Lp) were dependent on cell turgor (P). Lp was (7.8±5.5)·10-7 cm s-1 bar-1 (mean±SD; n=22 cells) for P>4 bar in cells of the upper epidermis and was increasing by a factor of up to three for P0 bar. No polarity of water movement or concentration dependence of Lp was observed. For cells of the lower epidermis the Lp-values were similar and the hydraulic conductivity also showed a similar dependence on turgor. No wall ingrowth or wall labyrinths (as in transfer cells) could be found in the cells of the lower epidermis. The elastic modulus () of cells of the upper epidermis could be measured over the whole pressure range (P=0–7 bar) by changing the osmotic pressure of the medium. increased linearly with increasing turgor and ranged between 10 and 150 bar. For cells of the lower epidermis the dependence of on P was similar, although the pressure dependence could not be measured on single cells. The Lp-values are compared with literature data obtained for Elodea by a nuclear magnetic resonance (NMR)-technique. Summary In separated outer medullary collecting duct (MCD) cells, the time course of binding of the fluorescent stilbene anion exchange inhibitor, DBDS (4,4-dibenzamido-2,2-stilbene disulfonate), to the MCD cell analog of band 3, the red blood cell (rbc) anion exchange protein, can be measured by the stopped-flow method and the reaction time constant, DBDS, can be used to report on the conformational state of the band 3 analog. In order to validate the method we have now shown that the ID50,DBDS,MCD (0.5±0.1 m) for the H2-DIDS (4,4-diisothiocyano-2,2-dihydrostilbene disulfonate) inhibition of DBDS is in agreement with the ID50,Cl ,MCD (0.94±0.07 m) for H2-DIDS inhibition of MCD cell Cl flux, thus relating DBDS directly to anion exchange. The specific cardiac glycoside cation transport inhibitor, ouabain, not only modulates DBDS binding kinetics, but also increases the time constant for Cl exchange by a factor of two, from Cl=0.30±0.02 sec to 0.56±0.06 sec (30mm NaHCO3). The ID50,DBDS,MCD for the ouabain effect on DBDS binding kinetics is 0.003±0.001 m, so that binding is about an order of magnitude tighter than that for inhibition of rbc K+ flux (K I,K +,rbc=0.017 m). These experiments indicate that the Na+,K-ATPase, required to maintain cation gradients across the MCD cell membrane, is close enough to the band 3 analog that conformational information can be exchanged. Cytochalasin E (CE), which binds to the spectrin/actin complex in rbc and other cells, modulates DBDS binding kinetics with a physiological ID50,DBDS,MCD (0.076±0.005 m); 2 m CE also more than doubles the Cl exchange time constant from 0.20±0.04 sec to 0.50±0.08 sec (30mm NaHCO3). These experiments indicate that conformational information can also be exchanged between the MCD cell band 3 analog and the MCD cell cytoskeleton.  相似文献

A dual-wavelength fluorimeter was constructed, which used two light emitting diodes (LEDs) to excite the fluorescence dye RH 421 alternately with two different wavelengths. The ratio of the emissions at the two excitation wavelengths provided a drift-insensitive signal, which allowed detection of very small changes of the fluorescence intensity. Those small changes were induced by ion binding and release in conformation E1 of the Na,K-ATPase. Titration experiments were performed to determine equilibrium dissociation constants (± standard deviation) for each step in the complete binding and release sequence: 0.12 ± 0.01 mM (E2(K2) KE1), 0.08 ± 0.01 mM (KE1 E1), 3.0 ± 0.2 mM (NaE1 E1), 5.2 ± 0.4 mM (Na2E1 NaE1) and 6.5 ± 0.4 mM (Na3E1 Na2E1) at pH 7.2 and T=16°C. These numbers show that the affinities of the binding sites exposed to the cytoplasm, are higher for K+ than for Na+ ions, similar to what was found on the extracellular side. The physiological requirement for extrusion of Na+ from the cytoplasm, and for import of K+ from the extracellular medium seems to be facilitated not by favorable binding affinities in state E1 but by the two ATP-driven reaction steps of the cycle, E2(K2) + ATP K2E1 · ATP and Na3E1 · ATP (Na3) El-P, which border the ion exchange reactions at the binding sites in conformation E1. Summary Using manometric and gas analytical methods oxygen consumption , carbon dioxide production , respiratory quotientRQ, (Fig. 1A-C) and thorax surface temperature difference T ts (Fig. 3) were determined in single bees. The animals were either sitting in respiratory chambers or were suspended by the scutum, in which case they were resting, walking (turning a small polystyrene ball) or flying in a closed miniature wind tunnel.During resting (sitting in Warburg vessels) at an ambient temperatureT a=10°C,RQ was 1.01±0.2 (n=905) with variations due to method (Fig. 1D, E).RQ values during walking were determined in single cases. In no case were they significantly different from 1.00. After the first 10 min of flight meanRQ was 1.00±0.04. It was significantly smaller than 1.00 (RQ=0.97) only during the last 5% of long-time flights (mean flight duration 58.8±28.8 min). With the exception of near-exhaustion conditions no signs of fuels other than carbohydrates were found.Metabolic rateP m was 19.71±21.38 mW g–1 during resting at 20°CT a30°C indicating that many resting bees actively thermoregulate at higherT a. After excluding bees which were actively thermoregulating, by an approximationP m was 5.65±2.44 mW g–1 at 20°CT a30°C. Summary Confluent monolayers of the established opossum kidney cell line were exposed to NH4Cl pulses (20 mmol/liter) during continuous intracellular measurements of pH, membrane potential (PD m ) and membrane resistance (R m) in bicarbonate-free Ringer. The removal of extracellular NH4Cl leads to an intracellular acidification from a control value of 7.33±0.08 to 6.47±0.03 (n=7). This inhibits the absolute K conductance (g K+), reflected by a decrease of K+ transference number from 71±3% (n=28) to 26±6% (n=5), a 2.6±0.2-fold rise ofR m, and a depolarization by 24.2±1.5 mV (n=52). In contrast, intracellular acidification during a block ofg K+ by 3 mmol/liter BaCl2 enhances the total membrane conductance, being shown byR m decrease to 68±7% of control and cell membrane depolarization by 9.8±2.8 mV (n=17). Conversely, intracellular alkalinization under barium elevatesR m and hyperpolarizes PD m . The replacement of extracellular sodium by choline in the presence of BaCl2 significantly hyperpolarizes PD m and increasesR m, indicating the presence of a sodium conductance. This conductance is not inhibited by 10–4 mol/liter amiloride (n=7). Patch-clamp studies at the apical membrane (excised inside-out configuration) revealed two Na+-conductive channels with 18.8±1.4 pS (n=10) and 146 pS single-channel conductance. Both channels are inwardly rectifying and highly selective towards Cl. The low-conductive channel is 4.8 times more permeable for Na+ than for K+. Its open probability rises at depolarizing potentials and is dependent on the pH of the membrane inside (higher at pH 6.5 than at pH 7.8).  相似文献

Summary Four zones of enzymatic activity for glutamate oxaloacetate transaminase (GOT) were found in apple tissue. A dimeric gene, GOT-1, determining the fastest migrating zone, was identified. Six alleles were found, including a near null allelle which produced detectable heterodimeric bands but not homodimeric bands. A marked deficit or absence of certain geno-types in all backcrosses and in some crosses between unrelated varieties was attributed to the close linkage (r=0.02±0.005) of GOT-1 with the incompatibility S locus. GOT-1 was also closely linked with the isocitrate dehydrogenase locus IDH-1 (0.03±0.01). Proposed incompatibility genotypes for four cultivars, and the linked GOT-1 alleles are Cox: S 1 b/S 2 d, Idared: S 3 a/S 4 c, Fiesta: S 3 a/S 2 d and Kent: S 3 a/S 1 b.The results reported in this paper are part of a PhD Thesis by the first author  相似文献

The dorsal skin of the leech Hirudo medicinalis was used for electrophysiological measurements performed in Ussing chambers. The leech skin is a tight epithelium (transepithelial resistance = 10.5±0.5 k· cm-2) with an initial short-circuit current of 29.0±2.9 A·cm-2. Removal of Na+ from the apical bath medium reduced short-circuit current about 55%. Ouabain (50mol·l-1) added to the basolateral solution, depressed the short-circuit current completely. The Na+ current saturated at a concentration of 90 mmol Na+·l-1 in the apical solution (K M=11.2±1.8 mmol·l-1). Amiloride (100 mol·l-1) on the apical side inhibited ca. 40% of the Na+ current and indicated the presence of Na+ channels. The dependence of Na+ current on the amiloride concentration followed Michaclis-Menten kinetics (K i=2.9±0.4 mol·l-1). The amiloride analogue benzamil had a higher affinity to the Na+ channel (K i=0.7±0.2 mol·l-1). Thus, Na+ channels in leech integument are less sensitive to amiloride than channels known from vertebrate epithelia. With 20 mmol Na+·l-1 in the mucosal solution the tissue showed an optimum amiloride-inhibitable current, and the amiloride-sensitive current under this condition was 86.8±2.3% of total short-circuit current. Higher Na+ concentrations lead to a decrease in amiloride-blockade short-circuit current. Sitmulation of the tissue with cyclic adenosine monophosphate (100 mol·l-1) and isobutylmethylxanthine (1 mmol·l-1) nearly doubled short-circuit current and increased amiloride-sensitive Na+ currents by 50%. By current fluctuation analysis we estimated single Na+ channel current (2.7±0.9 pA) and Na+ channel density (3.6±0.6 channels·m-2) under control conditions. After cyclic adenosine monophosphate stimulation Na+ channel density increased to 5.4±1.1 channels·m-2, whereas single Na+ channel current showed no significant change (1.9±0.2 pA). These data present a detailed investigation of an invertebrate epithelial Na+ channel, and show the similarities and differences to vertebrate Na+ channels. Whereas the channel properties are different from the classical vertebrate Na+ channel, the regulation by cyclic adenosine monophosphate seems similar. Relationships between the permeability coefficient (PHA) and partition coefficient (K m/w) of acetic acid and the surface density of DMPC:cholesterol bilayers have been investigated. Permeability coefficients were measured in large unilamellar vesicles by NMR line broadening. Bilayer surface density, , was varied over a range of 0.5–0.9 by changing cholesterol concentration and temperature. The temperature dependence of PHA for acetic acid exhibits Arrhenius behavior with an average apparent activation energy (E a ) of 22±3 kcal/mole over a cholesterol mole fraction range of 0.00–0.40. This value is much greater than the enthalpy change for acetic acid partitioning between bulk decane and water (H° = 4.8±0.8 kcal/mole) and the calculated E a (= 8.0 kcal/mole) assuming a bulk phase permeability model which includes the enthalpy of transfer from water to decane and the temperature dependence of acetic acid's diffusion coefficient in decane. These results suggest that dehydration, previously considered to be a dominant component, is a minor factor in determining E a . Values of 1n PHA decrease linearly with the normalized phospholipid surface density with a slope of = -12.4±1.1 (r = 0.90). Correction of PHA for those temperature effects considered to be independent of lipid chain order (i.e., enthalpy of transfer from water to decane and activation energy for diffusion in bulk hydrocarbon) yielded an improved correlation ( = -11.7±0.5 (r = 0.96)). The temperature dependence of Km/w is substantially smaller than that for PHA and dependent on cholesterol composition. Values of 1n Km/w decrease linearly with the surface density with a slope of = -4.6±0.3 (r = 0.95), which is 2.7-fold smaller than the slope of the plot of 1n PHA vs. . Thus, chain ordering is a major determinant for molecular partitioning into and transport across lipid bilayers, regardless of whether it is varied by lipid composition or temperature.This work was supported by grants from Glaxo, INTERx/Merck, and University of Utah Research Committee.  相似文献

Summary Two-hexameric (2×6) hemocyanins from the brachyuran crabsCancer pagurus andCallinectes sapidus, the freshwater crayfishAstacus leptodactylus and the lobsterHomarus americanus were isolated and dissociated into native subunits.The subunits of each hemocyanin were analyzed by electrophoresis and immunology. Three immunologically distinct subunit types, which were termed, and, could be identified in each case. They were isolated preparatively, and interspecifically correlated. Subunit is subdivided into several electrophoretically distinct isoforms which are immunologically closely related (Astacus) or identical (other species). InAstacus andCancer one of these isoforms was shown to dimerize and to act as inter-hexamer bridge. It represents a fourth subunit type termed. A fifth, diffuse component, which in PAGE migrated at the position of a dimer, was identified in the crossed immunoelectrophoretic patterns as denatured hemocyanin.A common feature of the four hemocyanins is the presence of 4 copies of and 8 copies of/ within the 2×6 particles. The: ratio is 4:4 in the two Astacidea and 6:2 in the two Brachyura. exists in 2 copies inAstacus andCancer which means that a single dimer- is present in a two-hexamer. This leaves 2 monomeric copies inAstacus and 4 inCancer.Every subunit from the four species except ofAstacus - was capable to form hexamers in reassembly experiments. If subunit combinations were tested, hetero-hexamers were formed preferentially. Two-hexamers were reconstituted only in the presence of all subunit types and the native subunit stoichiometry was required to obtain twohexamers in considerable yields. Factors limiting 2×6 reassembly are discussed.Authentic 2×6 molecules ofAstacus, Homarus andCancer hemocyanin were immunolabeled with subunit-specific antibody fragments (Fab) or IgG molecules, and the resulting immuno complexes were studied in the electron microscope. A topological model of the quaternary structure of decapod 2×6 hemocyanins is derived, showing the position of each copy of the four subunit types. In this model, the inter-hexamer bridge- is surrounded by two and two subunits forming the central core of the dodecamer. Two additional and two additional subunits form the periphery together with one subunit occupying the peripheral short edges of each hexameric half structure.
