Bot. 38: 1937–1948. Associations between photosynthesis, the activation state ofRuBP carboxylase and the contents of photosynthetic intermediateswere compared in soybean and wheat leaves before and after exposureto photoinhibitory treatments in the presence of atmosphericO2. Exposing attached leaves to a supra-saturating irradiance(3 800 µmol quanta m– 2 s–1) for 2 h in CO2-freeair decreased carboxylation efficiency and the light-saturatedphotosynthetic rate in air by approximately 50%. Exposure tothe photoinhibitory treatment for periods in excess of 2 h didnot cause a further decrease of photosynthesis in soybean. Althoughphotosynthesis was reduced, the initial and total (fully-activated)activities of ribulose 1,5-bisphosphate carboxylase (RuBPCase)in leaf extracts were unaltered in each species by the photoinhibitorytreatment. This was true for leaves sampled under both air andat a rate-limiting intercellular CO2 partial pressure (Ci) of75 µPa Pa–1. The contents of ribulose l,5-bisphosphate(RuBP) and 3-phosphoglyceric acid (3-PGA) were reduced by thephotoinhibitory treatment in soybean leaves sampled in air andat a rate-limiting Ci, although the RuBP/3-PGA ratio was unaffected.The relative reduction of RuBP content in soybean leaves atrate-limiting C1 was similar to the corresponding reductionof carboxylation efficiency. For wheat,the relative reductionof RuBP content at rate-limiting Ci (–19%) caused by thephotoinhibitory treatment was considerably less than the correspondingdecrease of carboxylation efficiency (–49%).The RuBP/3-PGAratio of wheat was also increased significantly by the photoinhibitorytreatment The significance of these observations to the regulationof CO2-limited photosynthesis in leaves experiencing photoinhibitionunder atmospheric oxygen is discussed. Consideration is alsogiven to the previous contention that contemporary measurementsof initial activity in crude extracts may provide a spuriousindication of the amount of the enzyme-CO2-Mg2 + form of RuBPcarboxylase present in the leaf. Effects of growth temperature and irradiance on nitrogen partitioning among photosynthetic components were studied. Plantago asiatica was grown under different temperature and light conditions. Growth conditions were regulated such that the Chl a/b ratio in leaves grown at a low temperature with a low irradiance was similar to that in leaves grown at a high temperature with a high irradiance, suggesting that the balance between acquisition and utilization of light energy in the photosynthetic apparatus was similar between the two growth conditions. When plotted against the leaf nitrogen content, the RuBP (ribulose-1,5-bisphosphate) carboxylase content did not significantly differ depending on growth conditions. Both high irradiance and low temperature decreased nitrogen partitioning to Chl-protein complexes. Low temperature increased nitrogen allocation to stroma FBPase (fructose-1,6-phosphatase) irrespective of growth irradiance. Gas exchange measurement indicated that the ratio of the electron transport (J(max)) to the maximum carboxylation rate (V(cmax)) was not affected by growth irradiance but by growth temperature. It is concluded that nitrogen partitioning between acquisition and utilization of light energy responds to both growth temperature and irradiance, while nitrogen partitioning between carboxylation and regeneration of RuBP responds only to growth temperature.  相似文献

Abstract Associations between photosynthetic responses to CO2 at rate-saturating light and photosynthetic enzyme activities were compared for leaves of maize grown under constant air temperatures of 19, 25 and 31°C. Key photosynthetic enzymes analysed were ribulose bisphosphatc (RuBP) carboxylase, phosphoenolpyruvate (PEP) carboxylase, NADP-malic enzyme and pyruvate, Pi dikinasc. Rates of CO2-saturated photosynthesis were similar in leaves developed at 19°C and 25°C but were decreased significantly by growth at 31°C. In contrast, carboxylation efficiency differed significantly between all three temperature regimes. Carboxylation efficiency was greatest in leaves developed at 19°C and decreased with increasing temperature during growth. The changes of carboxylation efficiency were highly correlated with changes in the activity of pyruvate, Pi dikinase (r= 0.95), but not with other photosynthetic enzyme activities. The activities of these latter enzymes, including that of RuBP carboxylase, were relatively insensitive to temperature during growth. The sensitivity of quantum yield to O2 concentration was lower in leaves grown at 19°C than in leaves grown at 31°C. These observations support the novel hypothesis that variation in the capacity for CO2 delivery to the bundle sheath by the C4 cycle, relative to the capacity for net assimilation by the C2 cycle, can be a principal determinant of C4 photosynthetic responses to CO2.  相似文献

In comparison with its wild type (WT), the transgenic (TG) rice with silenced OsBP-73 gene had significantly lower plant height, grain number per panicle, and leaf net photosynthetic rate (P N). Also, the TG rice showed significantly lower chlorophyll (Chl), ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO), RuBPCO activase, and RuBP contents, photosystem 2 (PS2) photochemical efficiency (Fv/Fm and ΔF/Fm′), apparent quantum yield of carbon assimilation (Φc), carboxylation efficiency (CE), photosynthetic electron transport and photophosphorylation rates as well as sucrose phosphate synthase activity, but higher intercellular CO2 concentration, sucrose, fructose, and glycerate 3-phosphate contents, and non-photochemical quenching of Chl fluorescence (NPQ). Thus the decreased P N in the TG rice leaves is related to both RuBP carboxylation and RuBP regeneration limitations, and the latter is a predominant limitation to photosynthesis.  相似文献

Net photosynthetic rate (P N) measured at the same CO2 concentration, the maximum in vivo carboxylation rate, and contents of ribulose-1,5-bisphosphate (RuBP) carboxylase/oxygenase (RuBPCO) and RuBPCO activase were significantly decreased, but the maximum in vivo electron transport rate and RuBP content had no significant change in CO2-enriched [EC, about 200 μmol mol−1 above the ambient CO2 concentration (AC)] wheat leaves compared with those in AC grown wheat leaves. Hence photosynthetic acclimation in wheat leaves to EC is largely due to RuBP carboxylation limitation.  相似文献

Summary The photosynthetic capacity of leaves is related to the nitrogen content primarily bacause the proteins of the Calvin cycle and thylakoids represent the majority of leaf nitrogen. To a first approximation, thylakoid nitrogen is proportional to the chlorophyll content (50 mol thylakoid N mol-1 Chl). Within species there are strong linear relationships between nitrogen and both RuBP carboxylase and chlorophyll. With increasing nitrogen per unit leaf area, the proportion of total leaf nitrogen in the thylakoids remains the same while the proportion in soluble protein increases. In many species, growth under lower irradiance greatly increases the partitioning of nitrogen into chlorophyll and the thylakoids, while the electron transport capacity per unit of chlorophyll declines. If growth irradiance influences the relationship between photosynthetic capacity and nitrogen content, predicting nitrogen distribution between leaves in a canopy becomes more complicated. When both photosynthetic capacity and leaf nitrogen content are expressed on the basis of leaf area, considerable variation in the photosynthetic capacity for a given leaf nitrogen content is found between species. The variation reflects different strategies of nitrogen partitioning, the electron transport capacity per unit of chlorophyll and the specific activity of RuBP carboxylase. Survival in certain environments clearly does not require maximising photosynthetic capacity for a given leaf nitrogen content. Species that flourish in the shade partition relatively more nitrogen into the thylakoids, although this is associated with lower photosynthetic capacity per unit of nitrogen.  相似文献

Gas exchange and contents of photosynthetic intermediates of leaves of Arbutus unedo L. were determined with the aim of recognizing the mechanisms of inhibition that were responsible for the midday depression of photosynthesis following exposure to dry air, and the decline in photosynthetic capacity following application of abscisic acid (ABA). Rapidly killed (<0.1 s) leaf samples were taken when gas analysis showed reduced CO2 assimilation. Determination of the contents of 3-phosphoglyceric acid (PGA), ribulose 1,5-bisphosphate (RuBP), triose phosphates, fructose 1,6-bisphosphate and hexose phosphates in the samples showed that significant variation occurred only in the level of PGA. As a result, the ratio PGA/RuBP decreased with increasing inhibition of photosynthesis, particularly when application of ABA had been the cause. A comparison of metabolite patterns did not bring out qualitative differences that would have indicated that effects of ABA and of dry air had been caused by separate mechanisms. Sunflower (Helianthus annuus L. cv Asmer) and maize (Zea mays L. cv Eta) plants were grown under controlled environmental conditions with a nutrient solution containing 0, 0.5, or 10 millimolar inorganic phosphate. Phosphate-deficient leaves had lower photosynthetic rates at ambient and saturating CO2 and much smaller carboxylation efficiencies than those of plants grown with ample phosphate. In addition, phosphate-deficient leaves contained smaller quantities of total soluble proteins and ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) per unit area, although the relative proportions of these components remained unchanged. The specific activity of Rubisco (estimated in the crude extracts of leaves) was significantly reduced by phosphate deficiency in sunflower but not in maize. Thus, there was a strong dependence of carboxylation efficiency and CO2-saturated photosynthetic rate on Rubisco activity only in sunflower. Phosphate deficiency decreased the 3-phosphoglycerate and ribulose-1,5-bisphosphate (RuBP) contents of the leaf in both species. The ratio of 3-phosphoglycerate to RuBP decreased in sunflower but increased in maize with phosphate deficiency. The calculated concentrations of RuBP and RuBP-binding sites in the chloroplast stroma decreased markedly with phosphate deficiency. The ratio of the stromal concentration of RuBP to that of RuBP-binding sites decreased in sunflower but was not affected in maize with phosphate deficiency. We suggest that a decrease in this ratio made the RuBP-binding sites more vulnerable to blockage or inactivation by tight-binding metabolites/inhibitors, causing a decrease in the initial specific activity of Rubisco in the crude extract from phosphate-deficient sunflower leaves. However, the decrease in Rubisco specific activity was much less than the decrease in the RuBP content in the leaf and its concentration in the stroma. A large ratio of RuBP to RuBP-binding sites may have maintained the Rubisco-specific activity in phosphate-deficient maize leaves. We conclude that the effect of phosphate deficiency is more on RuBP regeneration than on Rubisco activity in both sunflower and maize.  相似文献

苜蓿RuBP羧化酶的初活性和活化作用在不饱和光强下与光合速率一样随光强增加而增加。缺硫培养苜蓿叶片的光合速率和RuBP羧化酶的含量、初活性及总活性均比对照有不同程度的降低,其中酶的初活性与光合速率两者减少的趋势比较接近,说明RuBP羧化酶的初活性可能在光合CO_2固定作用中具有决定作用。然而,缺硫植株中酶的活化作用比对照明显增高。酶的活化作用与叶片中的叶绿素,6-PG,NADPH及ATP相对酶含量的比值成正比,与体内的酶量成反比。  相似文献

Abstract. A mechanistic model of photosynthesis is developed based on the characteristics of ribulose 1,5-bisphosphate (RuBP) carboxylase and the assimilation of CO2 as an ordered reaction with RuBP binding before CO2. An equation is derived which considers the effects of light (for regeneration of RuBP) and CO2. Taking values for the maximum turnover of RuBP carboxylase at substrate saturation, the maximum carboxylation efficiency (maximum increase in rate per increase in CO2 concentration) and the minimum quantum requirement for the C3 pathway, photosynthesis in the absence of photorespiration is simulated. In the model, at varying concentrations of CO2, the efficiency of light utilization approaches a maximum value as photon flux density decreases. Similarly, with a given maximum carboxyation capacity, at varying photon flux densities the carboxylation efficiency approaches a constant maximum value (equal to V max/ K m CO2) as CO2 is decreased. However, a decrease in the state of activation of RuBP carboxylase under low light results in a lower carboxylation efficiency. Limits on the rate of photosynthesis, as the maximum capacity for regeneration of RuBP is reduced relative to carboxylation potential, or as the maximum capacity of the carboxylase varies, are considered.  相似文献

The effect of temporary droughts on photosynthesis, total conductanceto water vapour, intercellular CO2 concentration, CO2 compensationpoint, light-response curves, photorespiration, dark respiration,chlorophyll content, and ribulose 1,5-bisphosphate (RuBP) carboxylase(EC 4.1.1.39 [EC] ) activity has been examined in nodulated alfalfaplants (Medicago saliva cv. Aragón). Plants were subjectedto moderate (S1/RS1) or severe (S2/RS2) cycles of drought (drought/recovery).Photosynthetic light-response curves showed decreased light-saturatedphotosynthetic capacity and decreased apparent quantum yield.Upon rewatering, photosynthesis did not recover whereas conductancedid in moderately stressed plants. Calculated electron transportrate also declined in drought-stressed plants, but upon rewatering,moderately stressed plants exhibited a total recovery. Comparison of photosynthetic intercellular CO2 response curvesin well-watered and stressed leaves led to the assertion thateffects in chloroplast metabolism contribute significantly tophotosynthetic inhibition. Although the validity of this entireline of research has been questioned by some recent studiesbecause the occurrence of heterogeneous stomatal closure wouldaffect these curves, in our case, the effect of water stresswas investigated in experimental systems where stomata had beenremoved. Measurements of in vitro RuBP carboxylase activityand protein content showed a strong decline during drought treatmentsand upon rewatering no recovery was observed. Therefore, ourresults suggest the major implication of non-stomatal factorsin the decline of photosynthesis in alfalfa plants under cyclicdrought conditions. Drought- and ABA-induced changes in photosynthesis of barley plants   总被引:1,自引:0,他引:1  
The changes caused by drought stress and abscisic acid (ABA) on photosynthesis of barley plants (Hordeum vulgare. L. cv. Alfa) have been studied. Drought stress was induced by allowing the leaves to lose 12% of their fresh weight. Cycloheximide (CHI), an inhibitor of stress-induced ABA accumulation, was used to distinguish alterations in photosynthetic reactions that are induced after drought stress in response to elevated ABA levels from those that are caused directly by altered water relations. Four hoars after imposition of drought stress or 2 h after application of ABA, Ihe bulk of the leaf's ABA content measured by enzyme-amplified ELISA, increased 14- and 16-fold, respectively. CHI fully blocked the stress-induced ABA accumulation. Gas exchange measurements and analysis of enzyme activities were used to study the reactions of photosynthesis to drought stress and ABA. Leaf dehydration or ABA treatment led to a noticeable decrease in both the initial slope of the curves representing net photosynthetic rate versus intercellular CO2 concentration and the maximal rate of photosynthesis; dehydration of CHI-treated plants showed much slower inhibition of the latter. The calculated values of the intercellular CO2 concentration, CO2 compensation point and maximal carboxylating efficiency of ribulose 1,5-bisphosphate (RuBP) carboxylase support the suggestion that biochemical factors are involved in the response of photosynthesis to ABA and drought stress. RuBP carboxylase activity was almost unaffected in ABA- and CHI-treated, non-stressed plants. A drop in enzyme activity was observed after leaf dehydration of the control and ABA-treated plants. When barley plants were supplied with ABA, the activity of carbonic anhydrase (CA, EC 4.2.2.1) increased more than 2-fold. Subsequent dehydration caused an over 1.5-fold increase in CA activity of the control plants and a more than 2.5-fold increase in ABA-treated plants. Dehydration of CHI-treated plants caused no change in enzyme activity. It is suggested that increased activity of CA is a photosynthetic response to elevated ABA concentration.  相似文献

Application of a 17-millimolar solution of glyphosate (GLP) to sugarbeet (Beta vulgaris L.) leaves resulted in an immediate and rapid decline in the level of ribulose bisphosphate (RuBP). Phosphoglyceric acid level began to decrease about 2 hours following the decline in RuBP level. Photosynthesis rate declined linearly with RuBP level, but only when the RuBP level had decreased to about twice the RuBP carboxylase active site concentration. This occurred about 4 hours following GLP-application. At this time starch synthesis also declined abruptly. The activation state of RuBP carboxylase did not change for 8 hours following GLP application and then decreased slightly from 70 to 50% when the RuBP level fell below the RuBP carboxylase active-site concentration. Triose-phosphate, hexose-phosphate, and adenylate energy charge did not change for 8 hours following GLP-application. These data indicate that GLP induced a depletion of carbon or phosphate or both from the photosynthetic carbon reduction cycle, reducing the rate of regeneration of RuBP, photosynthesis, and starch synthesis, while having little effect upon the rate of sucrose synthesis and transport.  相似文献

Previous reports indicate that ribulose 1,5-bisphosphate (RuBP) binds very tightly to inactive ribulose bisphosphate carboxylase (rubisco) in vitro. Therefore, we decided to investigate whether there was evidence for tight binding of RuBP associated with deactivation of rubisco in vivo. We modified a technique for rapidly separating `free' metabolites from those bound to high molecular compounds. Arabidopsis thaliana plants were illuminated at various irradiances before freezing the leaves in liquid N2 and assaying rubisco activity and RuBP. The percentage activation of rubisco varied from 37% at low irradiance (45 micromoles quanta per square meter per second) to 100% at high irradiance (800 micromoles quanta per square meter per second). The total amount of RuBP did not vary much with irradiance, but bound RuBP changed from 36% of the total at low irradiance to none at high irradiance. Bound RuBP was significantly correlated with the estimated number of inactive rubisco sites, with a ratio of about 1:1. After a step increase in irradiance, rubisco activation increased and total RuBP increased transiently, but steady levels of both occurred by 10 minutes. The amount of bound RuBP decreased with a similar time course to the estimated decrease in inactive rubisco sites. After a step decrease in irradiance, rubisco deactivated slowly for at least 25 minutes. Bound RuBP increased gradually but did so more slowly than the estimated increase in inactive rubisco sites.  相似文献

The effects of phosphate concentration on plant growth and photosyntheticprocesses in primary leaves of young sunflower (Helianthus annuusL.) plants were examined. Plants were grown for 3 weeks on half-strengthHoagland's solution containing 0, 0.1, 0.5, 1.0, and 3.0 molm–3 orthophosphate (Pi). It was shown that optimal photosynthesisand the highest light utilization capacity were achieved at0.5 mol m–3 Pi in the growth medium, which was in goodagreement with the maximum content of organic phosphorus inthe leaves. Low phosphate in the medium inhibited plant growthrate. Phosphate deficiency appreciably decreased photosyntheticoxygen evolution by leaves, the efficiency of photosystem two(PSII) photochemistry and quantum efficiency of PSII electrontransport. High oxidation state of PSII primary electron acceptorQA, at 0.1 mol m–3 Pi, however, indicates that photosyntheticelectron transport through PSII did not limit photosynthesisin Pi-deficient leaves. The results indicate that diminishedphotosynthesis under sub- and supra-optimal Pi was caused mainlyby a reduced efficiency of ribulose 1, 5-bisphosphate (RuBP)regeneration at high light intensities. These results suggestthat, under non-limiting C02 and irradiance, photosynthesisof the first pair of leaves could be diminished by both sub-and supra-optimal phosphorus nutrition of sunflower plants. Net photosynthetic rates (Pns) in leaves were compared between rice plants grown in ambient air control and free-air CO2 enrichment (FACE, about 200 micromol mol(-1) above ambient) treatment rings. When measured at the same CO2 concentration, the Pn of FACE leaves decreased significantly, indicating that photosynthetic acclimation to high CO2 occurs. Although stomatal conductance (Gs) in FACE leaves was markedly decreased, intercellular CO2 concentrations (Ci) were almost the same in FACE and ambient leaves, indicating that the photosynthetic acclimation is not caused by the decreased Gs. Furthermore, carboxylation efficiency and maximal Pn, both light and CO2-saturated Pn, were decreased in FACE leaves, as shown by the Pn-Ci curves. In addition, the soluble protein, Rubisco (ribulose-1,5-bisphosphate caboxylase/oxygenase), and its activase contents as well as the sucrose-phosphate synthase activity decreased significantly, while some soluble sugar, inorganic phosphate, chlorophyll and light-harvesting complex II (LHC II) contents increased in FACE leaves. It appears that the photosynthetic acclimation in rice leaves is related to both ribulose-1,5-bisphosphate (RuBP) carboxylation limitation and RuBP regeneration limitation.  相似文献

The influence of long-term water deficit on photosynthesis, electron transport and carbon metabolism of sunflower leaves has been examined. Water deficit was imposed from flower bud formation up to the stage of full flowering in the field on two sunflower hybrids with different drought tolerance. CO2 assimilation and stomatal conductance of the intact leaves, determined at atmospheric CO2 and full sunlight (1500-2000 mol quanta m-2 s-1), decreased with water deficit. Maximum quantum efficiency of PSII (Fy/Fm) and relative quantum yield of PSII (II) determined under similar experimental conditions, did not change significantly in severely stressed leaves. The strong inhibition of the plateau region of the light response curve, determined at high CO2 (5%) in water-deficient sunflower leaves, indicates that photosynthesis is also limited by non-stomatal factors. The decreased slope and the plateau of the CO2 response curves show that the capacity of carboxylation and RuBP regeneration decreased in severely stressed intact leaves. Rubisco specific activity decreased in severely stressed leaves, but Rubisco content increased under prolonged drought. The increase of Rubisco content was significantly higher in leaves of the drought-tolerant sunflower hybrid indicating that a higher Rubisco content could be one factor in conferring better acclimation and higher drought tolerance.  相似文献
