The ability of foliar tannins to increase plant resistance to herbivores is potentially determined by the composition of the tannins; hydrolyzable tannins are much more active as prooxidants in the guts of caterpillars than are condensed tannins. By manipulating the tannin compositions of two contrasting tree species, this work examined: (1) whether increased levels of hydrolyzable tannins increase the resistance of red oak (Quercus rubra L.), a tree with low resistance that produces mainly condensed tannins, and (2) whether increased levels of condensed tannins decrease the resistance of sugar maple (Acer saccharum Marsh.), a tree with relatively high resistance that produces high levels of hydrolyzable tannins. As expected, when Lymantria dispar L. caterpillars ingested oak leaves coated with hydrolyzable tannins, levels of hydrolyzable tannin oxidation increased in their midgut contents. However, increased tannin oxidation had no significant impact on oxidative stress in the surrounding midgut tissues. Although growth efficiencies were decreased by hydrolyzable tannins, growth rates remained unchanged, suggesting that additional hydrolyzable tannins are not sufficient to increase the resistance of oak. In larvae on condensed tannin-coated maple, no antioxidant effects were observed in the midgut, and levels of tannin oxidation remained high. Consequently, neither oxidative stress in midgut tissues nor larval performance were significantly affected by high levels of condensed tannins. Post hoc comparisons of physiological mechanisms related to tree resistance revealed that maple produced not only higher levels of oxidative stress in the midgut lumen and midgut tissues of L. dispar, but also decreased protein utilization efficiency compared with oak. Our results suggest that high levels of hydrolyzable tannins are important for producing oxidative stress, but increased tree resistance to caterpillars may require additional factors, such as those that produce nutritional stress.  相似文献

In the field of plant–herbivore interactions, research methods where plant secondary metabolites are manipulated are becoming more and more popular. Among the most commonly used is tannic acid. However, recent studies have shown that different tannic acid preparations are not comparable in their tannin structures. While tannic acids are meant to contain only gallotannins (GTs), some commercial preparations compose mainly of more simple galloylglucoses (that have, e.g. much lower protein precipitation capacity than GTs) or even of gallic acid (the hydrolysis product of GTs). Another group of tannins used in feeding trials is condensed tannins (CTs), usually in the form of quebracho tannin. Quebracho, however, contains different CT structures than, e.g. leaves of many deciduous trees. Additionally, when analysed with the common acid-butanol assay for total CTs, quebracho tannins give even 30-fold lower absorbance than the CTs of those deciduous trees. In addition to above problems it has been shown that different tannins can give different response even within the same herbivore species, and that the same tannin structure can cause different response in different herbivores. Browsing ruminants have been shown to tolerate a certain amount of tannins in their natural diet, and preference trials with captive roe deer (Capreolus capreolus) have suggested an active selection for a low dose of hydrolysable tannins. In this study, we investigated the preference patterns for tannic acid, a source of hydrolysable tannins, and quebracho, a source of condensed tannins, in a series of preference trials with captive roe deer over time, using a pelleted feed that differed only in the respective tannin content. Additionally, two groups of four hand-raised roe deer fawns were fed either a control or a 3% tannic-acid containing diet and physiological parameters were compared after 7.5 months. There were large differences in preference patterns between the individual roe deer groups; quebracho was mostly avoided, whereas tannic acid was actively included in the diet in differing, low proportions. However, one group consistently preferred the quebracho diet over both the control or the tannic acid diet. For the tannic acid, the preference pattern often revealed an initial period of high preference, followed by a stable period of a moderate preference. The fawns on the tannic acid diet had a lower pellet intake and a higher relative mass gain than the fawns on the control diet; differences in salivary tannin-binding capacity and in blood antioxidant status were below significance. These results are the first indications of potential benefits of a low-dose tannin diet, which need further confirmation. The results of the preference trials demonstrate that the time pattern of tannin intake is not constant, and pose the question about the validity of short-term preference trials in general.  相似文献

Condensed tannin and astringency levels of several grasses known to be preferred, or not preferred, by range caterpillar larvae, Hemileuca oliviae Cockrell, were determined. Comparable levels of a commercially available condensed tannin, quebracho, were applied to filter paper discs in choice-tests. Quebracho inhibited feeding in preference tests. Quebracho also was incorporated into agar-based diets; larval development and survival were inhibited. However, in these latter tests, larvae were not provided with tannin-free diet and responded to the presence of quebracho by increasing their rate of consumption. Condensed tannin (quebracho) therefore acted as a feeding deterrent to range caterpillar larvae in situations where low-tannin hosts were available, but as an antimetabolite in no-choice environments.  相似文献

To understand the ways in which condensed tannins (CT) affect primate diet selection and nutritional status, correct measurements are essential. In the majority of studies of the CT contents of primate foods, a tannin source such as "quebracho" is used to standardize CT assays, but the CT in quebracho tannin may not be similar to those in the plants of interest. We investigated how the choice of standard to calibrate CT assays affects the estimation of CT in the diets of mountain gorillas (Gorilla beringei). We purified the CT from gorilla foods and compared the actual amounts of CT in the foods with estimates produced by using the quebracho tannin. When quebracho was used, the estimates of CT contents of gorilla foods were, on average, 3.6 times the actual content of CT so that the amounts in frequently eaten gorilla foods were substantially overestimated. The overestimation for a given plant could not be predicted reliably and the ranking of plants by tannin content differed according to the standard used. Our results demonstrate that accurate measurements of CT necessitate the use of tannins purified from the plant species of interest. A reevaluation of primatology studies using interspecific comparisons of tannin content will provide new insights into primate food selection and nutritional ecology.  相似文献

Summary The concept of protein precipitation potential has recently been introduced by Wisdom et al. (1987) as a means to combine chemical and protein precipitation assays of tannins for ecological studies. The definition of protein precipitation potential was not theoretically rigorous, and data analysis was obscure. Our attempts to repeat the tannin extraction procedure gave incomplete recovery (24% loss of quebracho) of condensed tannins, the only type considered by Wisdom et al. In contrast we found their method efficient for hydrolysable tannins (80% recovery of tannic acid) which were undetected by their chemical assay of tannins. Their protein precipitation assay was confounded by chemical interference from both types of tannins. We conclude with recommendations for this type of analysis.  相似文献

The interactions among white spruce, Picea glauca (Moench) Voss, purified acetone tannin extracts (hydrolyzable and condensed tannin), Bacillus thuringiensis subsp. kurstaki Cry1A(c) delta-endotoxin strain HD-73 (Btk), and spruce budworm, Choristoneura fumiferana (Clemens) (Lepidoptera: Tortricidae) on larval survival, growth, and development were investigated over the whole larval feeding period by using artificial diet supplemented with three concentrations of Btk toxins per milliliter of diet (0, 0.021, and 1.72 microg/ml) and three concentrations of foliar tannin extract (0, 8, and 15% dry mass basis). At high Btk concentration, tannin antagonized Btk potency against spruce budworm by lowering Btk-related larval mortality from 83 to 43%. At moderate Btk concentration tannin did not affect Btk potency. Host tree tannins antagonized not only the lethal effects of Btk toxin but also sublethal Btk-related impacts in terms of larval development, pupal weight, relative consumption rate, and growth rate. When alone in the diet, tannin negatively affected larval survival, growth, and development. Maximum potency of tannins against spruce budworm larvae (60% mortality) was reached at dietary concentrations corresponding to what is found in the plant (8% dry mass). The addition of Btk toxin in food containing tannin reduced percentage of larval mortality by one-third, indicating that Btk toxin can antagonize tannin potency against the insect. Development of Btk transgenic spruce trees should consider the antagonistic effect the toxin may have on the resistance conferred by tannins that have evolved naturally in spruce trees.  相似文献

单宁酸是一种植物次生代谢物, 为探索其用于蚊幼虫防治的可能性,在室内测定了其对淡色库蚊Culex pipiens pallens抗氰戊菊酯品系和敏感品系1~4龄幼虫的毒性,并观察了其对存活幼虫生长发育的影响。结果表明,淡色库蚊敏感品系幼虫对单宁酸的敏感性比抗氰戊菊酯品系的要高,1~4龄幼虫分别高6.4、4.9、4.7和2.0倍。4个龄期幼虫中,无论是敏感品系还是抗氰戊菊酯品系,均是1龄幼虫对单宁酸的敏感性最高,3龄幼虫最低。在1 000 mg/L单宁酸持续作用下,敏感品系和抗氰戊菊酯品系各龄幼虫的存活率,均随处理时间延长而降低。与对照相比,饲养在100 mg/L~500 mg/L单宁酸溶液中的存活幼虫发育历期延长,敏感品系和抗氰戊菊酯品系发育历期分别延长了34.5~38.3 h和59.2~93.4 h。其中,125 mg/L浓度处理的敏感品系1~4龄幼虫,其发育历期与对照的差异达到了显著水平(P<0.05);抗性品系则在250 mg/L作用下也达到了差异显著水平(P<0.05)。但100~250 mg/L单宁酸处理淡色库蚊抗氰戊菊酯品系和敏感品系1龄幼虫,对其存活幼虫的化蛹率、羽化率和成虫性比均无显著影响。表明单宁酸对淡色库蚊幼虫的影响主要是延迟其生长发育,且影响程度与蚊虫对氰戊菊酯的敏感性有关。  相似文献

Summary Previous studies have shown leaves of tulip tree, Liriodendron tulipifera L. (of the Magnoliaceae) and of Populus tremuloides Michx. (of the Salicaceae) to be antixenotic/antibiotic to many Lepidoptera, including one of the most polyphagous of all phytophagous insects, the southern armyworm, Spodoptera eridania Cramer (Noctuidae). We investigated the physiological responses to this phytochemical activity on neonate and late instar armyworm larvae in controlled environments with particular emphasis upon the leaf extracts containing condensed tannins and hydrolysable tannins. These tannin-containing extracts of tulip tree leaves and quaking aspen leaves were generally toxic to neonate larvae. For later instars, growth suppression was not due to digestibility-reduction, but instead to suppressed consumption rates and greatly increased metabolic (respiratory) costs as reflected in reduced biomass conversion efficiencies.  相似文献

Tannins in forages complex with protein and reduce the availability of nitrogen to ruminants. Ruminal bacteria that ferment protein or peptides in the presence of tannins may benefit digestion of these diets. Bacteria from the rumina of sheep and goats fed Calliandra calothyrsus (3.6% N and 6% condensed tannin) were isolated on proteinaceous agar medium overlaid with either condensed (calliandra tannin) or hydrolyzable (tannic acid) tannin. Fifteen genotypes were identified, based on 16S ribosomal DNA-restriction fragment length polymorphism analysis, and all were proteolytic and fermented peptides to ammonia. Ten of the isolates grew to high optical density (OD) on carbohydrates (glucose, cellobiose, xylose, xylan, starch, and maltose), while the other isolates did not utilize or had low growth on these substrates. In pure culture, representative isolates were unable to ferment protein that was present in calliandra or had been complexed with tannin. One isolate, Lp1284, had high protease activity (80 U), a high specific growth rate (0.28), and a high rate of ammonia production (734 nmol/min/ml/OD unit) on Casamino Acids and Trypticase Peptone. Phylogenetic analysis of the 16S ribosomal DNA sequence showed that Lp1284 was related (97. 6%) to Clostridium botulinum NCTC 7273. Purified plant protein and casein also supported growth of Lp1284 and were fermented to ammonia. This is the first report of a proteolytic, ammonia-hyperproducing bacterium from the rumen. In conclusion, a diverse group of proteolytic and peptidolytic bacteria were present in the rumen, but the isolates could not digest protein that was complexed with condensed tannin.  相似文献

Summary Feeding efficiencies of ultimate instar larvae of two polyphagous tree-feeding Lepidoptera, Malacosoma disstria (Lasiocampidae) and Orgyia leucostigma (Liparidae), were measured on artificial diets containing from 0% to 8% tannic acid. Relative growth rate (RGR) of O. leucostigma was not affected by up to 8% tannic acid, suggesting that O. leucostigma has evolved an effective counteradaptation to hydrolyzable tannins. In contrast, as little as 0.5% tannic acid caused a significant reduction in RGR of M. disstria, due both to reduced efficiency of conversion of digested food (ECD) and reduced relative consumption rate (RCR), and caused a significant increase in mortality during the pupal stage. Moreover, when reared from hatching on tannin-containing diets, no M. disstria larvae survived past the fourth instar.Although tannins are commonly referred to as digestibility-reducing substances, tannic acid did not reduce the ability of M. disstria or O. leucostigma larvae to digest either the whole diet or nitrogen contained in the diet. For M. disstria, tannic acid acts as a toxin and a feeding deterrent, but not as a digestibility-reducing substance. Growing evidence that tannins commonly act as toxins warrants a reassessment of their role in anti-herbivore chemistry.  相似文献

A 10 weeks study in rats fed a balanced diet containing "hydrosoluble" vitamin A and 3,2 p. 100 by weight of tannic acid resulted in a marked decrease of the hepatic concentration of vitamin A. The same dietary dose of chestnut tree tannins resulted in a much more marked decrease. However, there was no effect of a quebracho extract (condensed tannins) given in the same conditions.  相似文献

Strategies are sought to reduce intestinal colonisation of food-producing animals by Campylobacter jejuni, a leading bacterial cause of human foodborne illness worldwide. Presently, we tested the antimicrobial activity of hydrolysable-rich blackberry, cranberry and chestnut tannin extracts and condensed tannin-rich mimosa, quebracho and sorghum tannins (each at 100 mg/mL) against C. jejuni via disc diffusion assay in the presence of supplemental casamino acids. We found that when compared to non-tannin-treated controls, all tested tannins inhibited the growth of C. jejuni and that inhibition by the condensed tannin-rich mimosa and quebracho extracts was mitigated in nutrient-limited medium supplemented with casamino acids. When tested in broth culture, both chestnut and mimosa extracts inhibited growth of C. jejuni and this inhibition was much greater in nutrient-limited than in full-strength medium. Consistent with observations from the disc diffusion assay, the inhibitory activity of the condensed tannin-rich mimosa extracts but not the hydrolysable tannin-rich chestnut extracts was mitigated by casamino acid supplementation to the nutrient-limited medium, likely because the added amino acids saturated the binding potential of the condensed tannins. These results demonstrate the antimicrobial activity of various hydrolysable and condensed tannin-rich extracts against C. jejuni and reveal that condensed tannins may be less efficient than hydrolysable tannins in controlling C. jejuni in gut environments containing high concentrations of amino acids and soluble proteins.  相似文献

The high levels of tannins in many tree leaves are believed to cause decreased insect performance, but few controlled studies have been done. This study tested the hypothesis that higher foliar tannin levels produce higher concentrations of semiquinone radicals (from tannin oxidation) in caterpillar midguts, and that elevated levels of radicals are associated with increased oxidative stress in midgut tissues and decreased larval performance. The tannin-free leaves of hybrid poplar (Populus tremula × P. alba) were treated with hydrolyzable tannins, producing concentrations of 0%, 7.5% or 15% dry weight, and fed to Lymantria dispar caterpillars. As expected, larvae that ingested control leaves contained no measurable semiquinone radicals in the midgut, those that ingested 7.5% hydrolyzable tannin contained low levels of semiquinone radicals, and those that ingested 15% tannin contained greatly increased levels of semiquinone radicals. Ingested hydrolyzable tannins were also partially hydrolyzed in the midgut. However, increased levels of semiquinone radicals in the midgut were not associated with oxidative stress in midgut tissues. Instead, it appears that tannin consumption was associated with increased metabolic costs, as measured by the decreased efficiency of conversion of digested matter to body mass (ECD). Decreased ECD, in turn, decreased the overall efficiency of conversion of ingested matter to body mass (ECI). Contrary to our hypothesis, L. dispar larvae were able to maintain similar growth rates across all tannin treatment levels, in part, because of compensatory feeding. We conclude that hydrolyzable tannins act as “quantitative defenses” in the sense that high levels appear to be necessary to increase levels of semiquinone radicals in the midguts of caterpillars. However, these putative resistance factors are not sufficient to decrease the performance of tannin-tolerant caterpillars such as L. dispar.  相似文献

The relative affinities of condensed tannins purified from sorghum, pinto bean, quebracho and wattle for six dissimilar proteins have been determined by a competitive binding assay. The results indicate that tannin/protein interactions may be specific for different tannins as well as for different proteins. Eleven fungal strains (4 Penicillium commune, 2 Aspergillus niger, 2 Aspergillus rugulosa, Aspergillus terricola, Aspergillus ornatus and Aspergillus fumigatus) were isolated, characterized morphologically and by their capacity to degrade tannins. Aspergillus niger Aa-20 was used as control strain. Several concentrations of hydrolysable tannin (tannic acid) were used as sole carbon source. All strains were able to degrade hydrolysable tannins. Aspergillus niger GH1 and PSH showed the highest tannin-degrading capacity (67 and 70%, respectively). Also, the fungal capacity to degrade condensed tannin (catechin) was tested. Aspergillus niger PSH and Penicillium commune EH2 degraded 79.33% and 76.35% of catechin. The results demonstrated the capacity of fungi to use hydrolysable and condensed tannins as carbon source.  相似文献

An anaerobic diplococcoid bacterium able to degrade hydrolyzable tannins was isolated from the ruminal fluid of a goat fed desmodium (Desmodium ovalifolium), a tropical legume which contains levels as high as 17% condensed tannins. This strain grew under anaerobic conditions in the presence of up to 30 g of tannic acid per liter and tolerated a range of phenolic monomers, including gallic, ferulic, and p-coumaric acids. The predominant fermentation product from tannic acid breakdown was pyrogallol, as detected by high-performance liquid chromatography and mass spectrometry. Tannic acid degradation was dependent on the presence of a sugar such as glucose, fructose, arabinose, sucrose, galactose, cellobiose, or soluble starch as an added carbon and energy source. The strain also demonstrated resistance to condensed tannins up to a level of 4 g/liter.  相似文献

Reactivities of several proanthocyanidins (monomers of condensed tannins) and gallate esters (representing hydrolyzable tannins) with hydroxyl radicals, azide radicals, and superoxide anions were investigated using pulse radiolysis combined with kinetic spectroscopy. We determined the scavenging rate constants and the decay kinetics of the aroxyl radicals both at the wavelength of the semiquinone absorption (275 nm) and the absorption band of the gallate ester ketyl radical (400-420 nm). For most compounds second-order decay kinetics were observed, which reflect disproportionation of the semiquinones. In the case of the oligomeric hydrolysable tannins, pentagalloyl glucose and tannic acid, the decay kinetics were more complex involving sequential first-order and second-order reactions, which could only be resolved by kinetic modeling. A correlation of the reaction rates with hydroxyl radicals (k*OH) with the number of adjacent aromatic hydroxyl groups (i.e., representing catechol and/or pyrogallol structures) was obtained for both condensed and hydrolyzable tannins. Similar correlation for the reactions with azide radicals and superoxide anions are less obvious, but exist as well. We consider proanthocyanidins superior radical scavenging agents as compared with the monomeric flavonols and flavones and propose that these substances rather than the flavonoids proper represent the antioxidative principle in red wine and green tea.  相似文献

Gut microorganisms are thought to play a role in the degradation of plant secondary metabolites in herbivores. Here, we examined the presence of tannin–degrading microorganisms in the gut of two herbivores, plateau pikas (Ochotona curzoniae) and root voles (Microtus oeconomus). Tannase activity in the feces of wild plateau pikas and root voles increased from June to August, corresponding to the increase in condensed and hydrolyzable tannin concentrations in plants during this period. Using intragastric tannic acid infusion, we found that tannase activities were significantly higher following administration of three concentrations of tannic acid than that in the control group for root voles. In contrast, for plateau pikas, animals administered tannic acid exhibited lower tannase activity in low–dose and high–dose groups than that in the control group. Tannase activities in root voles were higher than those in plateau pikas among different months and in different tannic acid infusion groups. Three tannin–degrading anaerobic strains were isolated from the cecum of plateau pikas (termed A1, A2, and A3) and root voles (termed M1, M2, and M3). 16 s rDNA sequencing assigned isolates A1 and M1 to the genus Enterobacter, A2 and M2 to the genus Shigella, and A3 and M3 to the genus Staphylococcus. These data provided the first evidence of microorganisms with tannin–degrading activity in the gut of plateau pikas and root voles. We speculated that both animals could degrade tannins in the gut, but that root voles would have a greater ability to process tannins than plateau pikas.  相似文献
