An equilibrium density gradient centrifugation study involving the separation of "old" and "new" membranes has been developed to determine the manner in which protein, lipid, and chlorophyll are incorporated into growing intracytoplasmic membranes (chromatophores) of Rhodopseudomonas spheroides. Chromatophores derived from cells grown in an H(2)O-medium had a density of 1.175 to 1.180 g/cm(3) and were readily separable from chromatophores having a density of 1.220 to 1.230 isolated from cells grown in a 70% D(2)O-medium. After a shift from "D(2)O-" to "H(2)O"-based media, only hybrid chromatophores derived from a combination of "heavy" (old) and "light" (new) chromatophore material could be detected. The experimentally determined, median density values for the growing intracytoplasmic membrane system followed a theoretically determined profile which was calculated from the density of full "heavy" and full "light" material assuming random, homogeneous incorporation of new material into old membrane. The distribution of the radioactive labels for protein (leucine) and chlorophyll (delta-aminolevulinic acid) were identical and showed a reproducible displacement of the "old" material to the heavy side of the optical density at 365 nm (OD(365)) absorbance and a displacement of the "new" material to the light side of the OD(365) absorbance profile. Specific phospholipid growth showed no displacement for either the "old" or "new" material from the median absorbance profile.  相似文献

A new crystal structure of O-acetylserine sulfhydrylase (OASS) has been solved with chloride bound at an allosteric site and sulfate bound at the active site. The bound anions result in a new "inhibited" conformation, that differs from the "open" native or "closed" external aldimine conformations. The allosteric site is located at the OASS dimer interface. The new inhibited structure involves a change in the position of the "moveable domain" (residues 87-131) to a location that differs from that in the open or closed forms. Formation of the external aldimine with substrate is stabilized by interaction of the alpha-carboxyl group of the substrate with a substrate-binding loop that is part of the moveable domain. The inhibited conformation prevents the substrate-binding loop from interacting with the alpha-carboxyl group, and hinders formation of the external Schiff base and thus subsequent chemistry. Chloride may be an analog of sulfide, the physiological inhibitor. Finally, these results suggest that OASS represents a new class of PLP-dependent enzymes that is regulated by small anions.  相似文献

Since Jones et al. (2000) drew attention to a "new" type of glume wheat from Neolithic and Bronze Age sites in northern Greece, several finds of this morphologically distinct tetraploid wheat form have been made across central and southeastern Europe. Charred remains of this wheat, dating from 819–1031 cal b.c., have also been discovered in a storage pit at late Bronze Age Stillfried, eastern Austria. As both chaff and grains were found, it was not only possible to match the diagnostic features of the spikelet bases to the "new" form, but also to examine the grains, which are strikingly long, slender and flat. A dorsal ridge is absent and there is no hump above the embryo. The embryo angle is relatively low and compression lines are much more distinct. Within the Stillfried store "new" glume wheat grains were also easily separable from two-grained einkorn and spelt grains. The morphology of the grains is not inconsistent with the suggestion that the "new" type glume wheat might correspond to modern Triticum timopheevi. In Stillfried "new" glume wheat was grown as a winter crop, and it seems to have been cultivated as a maslin (mixed crop) together with T. monococcum (einkorn).  相似文献

Twenty low-molecular-weight-glutenin subunit (LMW-GS) gene sequences from the D-genome from Aegilops crassa (2n = 4x = 28), Ae. cylindrica (2n = 4x = 28), Ae. tauschii (2n = 2x = 14) and Triticum aestivum (2n = 6x = 42) were obtained using five sets of specific allele primer pairs. Only the sequences of the first primer pair were complete coding sequences (cds) of LMW-GS, and had 305, 304, 306 and 305 LMW-m amino acid residues in Ae. crassa, Ae. cylindrica, Ae. tauschii and T. aestivum, respectively. The repetitive domain and repeat motif numbers of all LMW glutenin subunits showed eight conserved cysteine residues that lead to the same functional activity in different genome. Based on DNA and predicted protein sequences, phylogenetic trees for all sets of sequences were drawn. At the DNA level, the species closest to T. aestivum for the second, third, fourth and fifth set of sequences were Ae. cylindrica, Ae. tauschii and Ae. crassa, respectively. At the protein level, the species closest to T. aestivum based on the first, second and fifth set of sequences were Ae. cylindrica, Ae. crassa and Ae. crassa, respectively. For other sets of sequences, bread wheat proved to be a distinct species. The evolution of the Permian ammonoid family Spirolegoceratidae is discussed and a new phylogenetic reconstruction is proposed based on new data. Two subfamilies are recognized within the family Spirolegoceratidae: Spirolegoceratinae Nassichuk and Sverdrupitinae subfam. nov. A new genus, Archboldiceras, is proposed to accommodate the Australian species Uraloceras lobulatum and U. whitehousei. Species of the genus Sverdrupites Nassichuk, 1970 are described based on new material from the Volga-Urals Region. The new species S. bogoslovskayae from the Gerkinskaya Formation of Novaya Zemlya previously identified by M.F. Bogoslovskaya as S. amundseni is described.  相似文献

Material on the Indo-Malayan Gryllacridinae (Stenopelmatidae) of the genera Zalarnaca Gor., Dinolarnaca gen. n., Phryganogryllacris Karny, Neanias Br.-W., and Aancistroger B.-Bien. is considered. A new genus, a new subgenus, 17 new species, and 2 new subspecies are described. Some insufficiently known taxa are redescribed. The Hopfield model of neural network stores memory in its symmetric synaptic connections and can only learn to recognize sets of nearly orthogonal patterns. A new algorithm is put forth to permit the recognition of general (non-orthogonal) patterns. The algorithm specifies the construction of the new network's memory matrix T ij, which is, in general, asymmetrical and contains the Hopfield neural network (Hopfield 1982) as a special case. We find further that in addition to this new algorithm for general pattern recognition, there exists in fact a large class of T ij memory matrices which permit the recognition of non-orthogonal patterns. The general form of this class of T ij memory matrix is presented, and the projection matrix neural network (Personnaz et al. 1985) is found as a special case of this general form. This general form of memory matrix extends the library of memory matrices which allow a neural network to recognize non-orthogonal patterns. A neural network which followed this general form of memory matrix was modeled on a computer and successfully recognized a set of non-orthogonal patterns. The new network also showed a tolerance for altered and incomplete data. Through this new method, general patterns may be taught to the neural network.  相似文献

Evolutionary analysis of the picornavirus family   总被引:1,自引:0,他引:1  
An exhaustive evolutionary analysis of the picornavirus family has been carried out using the amino acid sequences of several proteins of the viruses including: the capsid proteins (1D, 1B, and 1C) situated at the 5 end of the genome and responsible for the serotype of the viruses, and the viral polymerase (3D), located at the 3 end of the genome. The evolutionary relationships found among the viruses studied support the new classification, recently suggested, in contrast to the classical one, and the existence of a new genus for the picornavirus family. In the new taxonomic organization, five genera form the picornavirus family: (1) aphthoviruses, (2) cardioviruses, (3) hepatoviruses (previously classified as enteroviruses), (4) renteroviruses (which mainly constitute a combination of the previous genera rhinovirus and enterovirus), and (5) a new genus, with a new and unique representative: the echovirus 22. Our analysis also allowed us, for the first time, to propose the most probable sequence of speciation events to have given rise to the current picornavirus family.The bootstrap procedure was used to check the reliability of the phylogenetic trees obtained. The application of the method of the statistical geometry in distance space to internal branches of the tree revealed a high degree of evolutionary noise, which makes the resolution of some internal branching points difficult. Tripaphylus musteli (van Beneden, 1851) (Copepoda, Siphonostomatoida, Sphyriidae) is redescribed from an adult female collected from the branchial chamber of a starry smooth-hound, Mustelus asterias Cloquet (Carcharhiniformes, Triakidae), captured in the English Channel off Portland, UK. The new account of T. musteli is the first based on a complete adult female and highlighted the lack of a robust distinction separating Tripaphylus Richiardi, in Anonymous, 1878 and Paeon Wilson, 1919 prompting us to relegate Paeon to a junior subjective synonym of Tripaphylus. In the light of this synonymy the eight former species of Paeon are transferred to Tripaphylus as follows: T. ferox (Wilson, 1919) new combination, T. elongatus (Wilson, 1932) new combination, T. vassierei (Delamare Deboutteville & Nuñes-Ruivo, 1954) new combination, T. lobatus (Kirtisinghe, 1964) new combination, T. asymboli (Turner, Kyne & Bennett, 2003) new combination, T. versicolor (Wilson, 1919) new combination, T. australis (Kabata, 1993) new combination, and T. triakis (Castro Romero, 2001) new combination. Comparisons between terminology used in this report and that in the literature indicate that all transformed adult females of Tripaphylus probably possess a full complement of cephalic appendages and maxillipeds. All limbs, with the exception of the maxillae share a general morphological similarity to the corresponding appendages of conspecific males. The maxilla of the transformed adult female of Tripaphylus is a small digitiform protuberance associated with a swelling in some species.  相似文献

Only one isolate each of the class "Spartobacteria" (subdivision 2 of the phylum Verrucomicrobia) and of subdivision 3 of Verrucomicrobia have previously been reported to grow in laboratory culture. Using media that had been used successfully in other studies to isolate members of diverse groups of soil bacteria, we generated a collection of over 1,200 isolates from soil from a pasture. An oligonucleotide probe that targets the 16S rRNA genes of verrucomicrobia was used to screen this collection, and 14 new verrucomicrobia were identified. Nine of these belonged to the class "Spartobacteria" and were related to "Chthoniobacter flavus." Five further isolates were members of subdivision 3 and were related to the only known isolate of this subdivision. The differences in the 16S rRNA gene sequences of the new isolates and previously described isolates, of up to 10%, indicated that the new isolates represent new species and genera. All but two of the verrucomicrobial isolates were from colonies that first became visible one or more months after inoculation of plates with soil, but subcultures grew more rapidly. Analysis of PCR-amplified 16S rRNA genes in the pasture soil showed that members of the class "Spartobacteria" were more numerous than members of subdivision 3. Isolates of subdivision 3 were only found on plates receiving an inoculum that yielded a mean of 29 colonies per plate, while members of the class "Spartobacteria" were only found on plates receiving a more dilute inoculum that resulted in a mean of five colonies per plate. This suggested that colony development by members of the class "Spartobacteria" was inhibited by other culturable bacteria.  相似文献

A set of hypotheses are developed for the origin of living sharksand rays and the interrelationships of their major groups, usingsome methods of cladistic analysis to relate groups with sharedderived characters. Comparative studies on living sharks andrays combined with new data on fossil sharks suggests that theliving groups ultimately stem from a common ancestral groupof "neoselachian" sharks with many modern characters. The sociobiological fashion has for a long time discouraged the development of theories of the immediate causation of behaviour. It is only recently that new approaches are being developed to improve on the classical ethological theory originated by Lorenz and Tinbergen. One new departure is behavioural field theory (BFT) which brings back the concept of energy in behaviour, without being susceptible to the drawbacks of the energy or fuel concept of classical theory. In this paper the new theory is explained in some detail and it is shown to be illuminating, predictive and testable in examples of play-like and territorial behaviour.  相似文献

The authors show a way of reconstruction of scalp defects with excellent results using a tissue expander. This method creates "new" scalp tissue for coverage of defects with normal hair-bearing skin having a dense and even growth of hair. The hairs grow in the correct direction. The authors also show that almost all of the "new" hair-bearing scalp gained by the tissue expander is a result of stretching the scalp over the expander and its close surroundings and that only a very minute portion is gained by migration of the scalp from farther away.  相似文献

It should be noted that existing notions concerning the functioning of the oculomotor system (OMS) in human beings depend to a considerable degree on the methods of investigation chosen. Summary: The launch of a new brand and website for DMM.Regular readers of Disease Models & Mechanisms (DMM) during 2015 will have noticed some changes to the journal. This is part of the gradual implementation of a new Company brand and migration to a new, better, web platform. This culminated in October with the launch of a new website for The Company of Biologists (www.biologists.com), and a brand new look and feel for the DMM website (dmm.biologists.org) and the articles it publishes (see Fig. 1). The new DMM website (and those of its sister journals Development, Journal of Cell Science, Journal of Experimental Biology and Biology Open) is the result of a mammoth project to ensure that users have an enhanced experience when visiting our pages. 《遗传学》是生命科学相关专业本科阶段最重要的课程之一。近年来,随着生命科学领域研究的不断深入,新知识与新技术也在不断更新。但遗传学的教学模式目前仍以理论讲授为主,这使得抽象的原理难以被学生理解接受,直接影响了教学效果。因此探索新的教学模式尤为必要。2010年以来我校在生物技术专业《微生物遗传学》教学中开展了新教学模式——文献精读,文章从文献精读的前期课程基础,如何选择专业文献,怎样组织教学过程,开展文献精读对学生和教师的意义等方面全面分析了实施情况和应用价值,指出该教学模式体现了“前沿”和“经典”的结合,使书本的知识在实践中具体化,既提高学生的学习效果,激发学习兴趣,又开拓了学生的思路,锻炼其能力。这种教学模式为《遗传学》教学授课不断探索新的模式、在“精准医疗”时代下如何培养兼具临床与科研能力的医疗人才提供新思路。  相似文献
