A study was made of the age dynamics of polyploidization and dynamics of DNA synthesis in neuron cell nuclei during the postnatal growth of the gastropod pulmonate snail Succinea lauta. According to cytophotometrical results, the degree of polyploidization in neuron nuclei increases from young to adult individuals, varying from 2c to 16,384c. In the visceral complex, the maximum and medium ploidy values of the neuron nuclei are higher by almost 4-8 times than those in cerebral and pedal ganglia. The medium level of ploidy in adult snails increases by 5.7 times in the visceral complex of ganglia and by 4.1-4.2 times in the pedal and cerebral ganglia. According to 3H-thymidine autoradiography, DNA synthesis in neuron nuclei occurs during the whole life of the snail. In young individuals the neurons have the highest activity of DNA synthesis--the index of labeled nuclei of neurons making in total 50.2%. In older age, a steady decrease in the index of labeled nuclei is observed--in total to 35.8% and 7.0% in small and large adult snails, respectively. The state of summer hibernation completely stops DNA syntheses in neurons, but emergency from hibernation is accompanied by restoration of DNA syntheses.  相似文献

In experiments on the isolated nervous system, responses of the same command neurones of avoidance behaviour to electrical stimulation of the intestinal nerve were compared in adult snails (Helix pomatia) and in snails 1-20 days old. Dynamics of spike reactions of the command neurones to rhythmic nerve stimulation differed in adult and newborn snails. In the neurones of young snails, no sensitization, common in the adult animals, was observed. Study of excitatory input in the command neurones during rhythmic nerve stimulation suggests independence of habituation of synaptic input amplitude and spike response sensitization, because dynamics of habituation coincided in the young and adult snails while spike response underwent sensitization only in adult animals.  相似文献

Nitric oxide (NO) is a gas produced through the action of nitric oxide synthase that acts as a neurotransmitter in the central nervous system (CNS) of adult gastropod mollusks. There are no known reports of the presence of NOS-containing neurons and glial cells in young and adult Megalobulimus abbreviatus. Therefore, NADPH-d histochemistry was employed to map the nitrergic distribution in the CNS of young and adult snails in an attempt to identify any transient enzymatic activity in the developing CNS. Reaction was observed in neurons and fibers in all CNS ganglia of both age groups, but in the pedal and cerebral ganglia, positive neurons were more intense than in other ganglia, forming clusters symmetrically located in both paired ganglia. However, neuronal NADPH-d activity in the mesocerebrum and pleural ganglia decreased from young to adult animals. In both age groups, positive glial cells were located beneath the ganglionic capsule, forming a network and surrounding the neuronal somata. The trophospongium of large and giant neurons was only visualized in young animals. Our results indicate the presence of a nitrergic signaling system in young and adult M. abbreviatus, and the probable involvement of glial cells in NO production.  相似文献

Factors that reduce the reproductive output of self-fertilizing hermaphrodites are receiving increasing attention. The combined effects of reduced fecundity of selfing parents and inbreeding depression of the progeny have been referred to as self-fertilization depression. In isolated freshwater snails the reproductive output of selfing individuals also decreases due to the lack of social facilitation (absence of a conspecific). We examined the effect of social facilitation on lifetime reproductive success (number of young produced and longevity) over two generations in the simultaneously hermaphroditic land snail Balea perversa . In a parallel study we showed that B. perversa kept singly and in pairs reproduced exclusively by self-fertilization. In the parent generation, snails kept singly produced less offspring than snails kept in pairs. The difference in lifetime number of young was mainly due to differences in adult life span. Snails of the two groups did not differ in reproductive rate (number of young produced per 100 d of reproductive life) and hatchling size. In the offspring generation, snails kept singly did not differ from individuals kept in pairs in the lifetime number of young and hatchling survival. As in the parent generation, snails kept singly reproduced during a shorter period than snails kept in pairs. However, the shorter reproductive life span of snails kept singly was compensated for by a slightly (but not significantly) higher reproductive rate which resulted in a similar number of offspring produced for both groups. In both generations, snails of the two groups did not differ in size at first reproduction, adult growth rate and size at death. These findings suggest that social facilitation may affect longevity in selfing B. perversa .  相似文献

General morphology of chromatin, the number of chromosomes and chromocenters in normal condition and at the increase of bivalent cation (Ca2+, Mg2+) concentration were studied with the purpose to reveal mechanisms of polyploidization of neuron nuclei in the snail Succinea lauta (Gastropoda, Pulmonata). The morphology of nuclei was studied on squashed preparations. Normal diploid mitoses are described in the cerebral ganglia. A possibility is supposed that part of neurons or neuroblasts in the central nervous system (CNS) of succineid snail may divide mitotically. It has been shown that the basic mechanism of neuron postnatal growth is endomitotic polyploidization of nuclei. The transition from ordinary mitosis to polyploid cycles occurs via restitutional (polyploidizing) mitosis (4c2n-->4c4n). The next endocycles are carried out by means of classic endomitosis up to reaching the highest ploidy levels--4096n--16,384n. The study of general morphology of chromatin and chromocenters at normal condition and at artificial compactization enabled us to exclude any probability of polyteny in the CNS of lauta.  相似文献

Heterogenous properties of dentate granule neurons in the adult rat   总被引:18,自引:0,他引:18  
Postnatal neurogenesis contributes substantially to the neuronal population of the adult dentate gyrus. We report here that the neurons located in the deep aspects of the granule cell layer, near the proliferative zone, have different properties from those located in the superficial layers. The former group of neurons, tentatively designated as young, can be readily identified in a standard hippocampal slice preparation by morphological, immunohistochemical, and electrophysiological criteria. Electrophysiological recordings and imaging with Lucifer yellow from these neurons in the standard hippocampal slice preparation showed one or two main dendrites and conically shaped branches possessing varicose protrusions. These features are in agreement with the appearance of the same population of young neurons immunopositive for TOAD-64, a marker for immature neurons. In disinhibited slices, these putative young neurons are distinguished from the mature neurons, located in the superficial granule cell layer, by showing paired pulse facilitation and having a lower threshold for induction of long-term potentiation. The putative young neurons are completely unaffected by GABA(A) inhibition and always display robust long-term potentiation. In contrast, the mature neurons never produce long-term potentiation when the GABA(A) inhibition is intact. We propose that the heterogeneity of the functional properties of the granule neurons is related to the ongoing neurogenesis in the adult animals.  相似文献

The morphology of the serotonergic neurons modulating withdrawal behavior in the CNS of terrestrial snail was studied. It was shown that only one Pd4 neuron projects to the pleural and parietal ganglia. Its intracellular stimulation caused the same effects as the stimulation of the whole group of modulatory neurons. In juvenile snails, the number of serotonergic neurons and their relative sizes are smaller than in adult animals, which can be the reason for the observed age-related differences in the withdrawal behavior.  相似文献

In zebra finches, only males sing, and the neural regions controlling song exhibit prominent, hormone-induced sex differences in neuron number. In order to understand how sexual differentiation regulates neuron number within one song nucleus, the lateral magnocellular nucleus of the anterior neostriatum (IMAN), we studied the development of sex differences among IMAN neurons that project to the robust nucleus of the archistriatum (RA). The IMAN is implicated in song learning, and previous ontogenetic studies have indicated that males lose over 50% of their IMAN neurons during the juvenile song learning period. Based on developmental changes in both the extent of androgen accumulation within the IMAN and its appearance in Nissl-stained tissue, it had been hypothesized that IMAN neuron loss was even greater in young females, resulting in sex differences in neuron number. However, this hypothesis has not been tested directly because the Nissl-stained boundaries of the IMAN sometimes are ambiguous in young animals, and are not evident at all in adult females. To circumvent these problems, we employed the retrograde tracer fast blue to study the development of IMAN neurons defined on the basis of their projections to the RA. We find that the number of these IMAN-RA projection neurons is much greater in adult males than in females, and that this sex difference develops during the juvenile period of sexual differentiation and song learning because a significant number of these neurons are lost in females but not in males. With respect to sexual differentiation, we conclude that masculinization (which is stimulated by the hormone estradiol) promotes the retention of IMAN-RA projection neurons. In addition, our results indicate that any loss of IMAN neurons that may occur in young males does not include cells projecting to the RA.  相似文献

A group of 20 young and another of 20 adult Lymnaea truncatula were abundantly supplied with food and kept continuously under cold conditions (5 degrees C) in the laboratory for 3 months and the effects of low temperature on their behavior, growth and reproduction were studied. The results indicate that at low temperature the activity of L. truncatula was markedly reduced but complete hibernation did not occur. The snails seem to be unaffected by the low temperature itself since none (both young and adult) died during the 3 months that they were kept at 5 degrees C. Reduced feeding, even in the presence of abundant food, during the cold conditions caused an almost total inhibiton of growth. Of even more significance was the suppression of reproduction which was connected with the metabolic rate of adult snails kept at low temperature. Young snails seem to profit by exposure to low temperature. On the return to normal laboratory temperature (16-22 degrees C) the young snails became very active, fed voraciously, grew rapidly, tended to live longer and produced more offspring than the controls. Low temperature, however, appears to have an adverse after-effect on the growth and reproduction of mature snails. Relatively, fewer eggs were deposited in this case. The results indicate that under natural field conditions in England, where temperature fluctuations during the usually mild winter months are common, the greater burden of increasing the population in overwintered snails must rest on the younger members of the community.  相似文献

Developmental changes that occur in the prefrontal cortex during adolescence alter behavior. These behavioral alterations likely stem from changes in prefrontal cortex neuronal activity, which may depend on the properties and expression of ion channels. Nav1.9 sodium channels conduct a Na+ current that is TTX resistant with a low threshold and noninactivating over time. The purpose of this study was to assess the presence of Nav1.9 channels in medial prefrontal cortex (mPFC) layer II and V pyramidal neurons in young (20‐day old), late adolescent (60‐day old), and adult (6‐ to 7‐month old) rats. First, we demonstrated that layer II and V mPFC pyramidal neurons in slices obtained from young rats exhibited a TTX‐resistant, low‐threshold, noninactivating, and voltage‐dependent Na+ current. The mRNA expression of the SCN11a gene (which encodes the Nav1.9 channel) in mPFC tissue was significantly higher in young rats than in late adolescent and adult rats. Nav1.9 protein was immunofluorescently labeled in mPFC cells in slices and analyzed via confocal microscopy. Nav1.9 immunolabeling was present in layer II and V mPFC pyramidal neurons and was more prominent in the neurons of young rats than in the neurons of late adolescent and adult rats. We conclude that Nav1.9 channels are expressed in layer II and V mPFC pyramidal neurons and that Nav1.9 protein expression in the mPFC pyramidal neurons of late adolescent and adult rats is lower than that in the neurons of young rats. © 2017 Wiley Periodicals, Inc. The anterior forebrain (AF) pathway of songbirds has an essential but poorly understood function during song learning, a process requiring auditory experience. Consistent with a role in processing auditory information, two nuclei of the AF, the lateral magnocellular nucleus of the anterior neostriatum (lMAN) and Area X (X), contain some of the most complex auditory neurons known. In adult zebra finches, these neurons are strongly selective for both spectral and temporal properties of song: They respond more robustly to the bird's own song (BOS) than to songs of conspecific individuals, and they respond less well to BOS if it is played in reverse. lMAN and X neurons of young finches early in the process of song learning (30–45 days of age) are also song responsive, but lack the song and order selectivity present in adult birds. By an intermediate stage of learning (60 days), when birds have experience of both tutor song and their own developing (plastic) song, AF neurons have significant song and order selectivity for both tutor song and BOS (in this case, plastic song). The degree of BOS selectivity is still less than that found in adults, however. In addition, neurons at 60 days are heterogenous in their preference for BOS versus tutor song: Most prefer BOS, some prefer tutor song, and others respond equally to both songs. The selectivity of adult AF auditory neurons therefore arises rapidly during development from neurons that are initially unselective. These neurons are one of the clearest examples of experience-dependent acquisition of complex stimulus selectivity. Moreover, the neural selectivity for both BOS and tutor song at 60 days raises the possibility that experience of both songs during learning contributes to the properties of individual AF neurons. © 1997 John Wiley & Sons, Inc. A chronic intoxication with fenthion was induced in adult and young snails. The fecundity was only reduced by the concentratration of 4 ppm in adults. In the snails intoxicated by fenthion from hatching the shell growth and the fecundity were both reduced from a concentration of 2 ppm. Above 2.15 ppm the rearing of young snails was not possible.

为探讨青年猫和老年猫小脑皮质GABA能神经元及其表达的年龄相关性变化,利用Nissl染色显示小脑皮质结构及神经元,免疫组织化学ABC法标记GABA免疫阳性神经元。光镜下观察,采集图像,并利用图像分析软件对分子层、蒲肯野细胞层和颗粒层神经元及GABA免疫阳性神经元及其灰度值进行分析统计。结果显示,GABA免疫阳性神经元、阳性纤维及终末在青年猫和老年猫小脑皮质各层均有分布。与青年猫相比,老年猫分子层、蒲肯野细胞层神经元和GABA免疫阳性神经元密度及其GABA免疫阳性反应强度均显著下降(P<0.01),颗粒层神经元密度和GABA免疫阳性强度也显著下降(P<0.01),但其GABA免疫阳性神经元密度无显著变化(P>0.05);蒲肯野细胞的胞体萎缩,阳性树突分枝减少。因此认为,衰老过程中猫小脑皮质GABA能神经元的丢失和GABA表达的下降,可能是老年个体运动协调、精确调速和运动学习等能力下降的重要原因之一。  相似文献

对人类和动物的心理学研究证实,老年个体的视觉对比敏感度相对青年个体显著下降。为揭示其可能的神经机制,采用在体细胞外单细胞记录技术研究青、老年猫(Felis catus)初级视皮层(primary visual cortex,V1)细胞对不同视觉刺激对比度的调谐反应。结果显示,老年猫V1细胞对视觉刺激反应的平均对比敏感度比青年猫显著下降,这与灵长类报道的研究结果相一致,表明衰老影响视皮层细胞对视觉刺激反应的对比敏感度是灵长类和非灵长类哺乳动物中普遍存在的现象,并可能是介导老年性视觉对比敏感度下降的神经基础。另外,与青年猫相比,老年猫初级视皮层细胞对视觉刺激的反应性显著增强,信噪比下降,感受野显著增大,表明衰老导致的初级视皮层细胞对视觉刺激反应的对比敏感度下降伴随着皮层内抑制性作用减弱。  相似文献

Of the approximately 34 identified Biomphalaria species,Biomphalaria alexandrina represents the intermediate host of Schistosoma mansoni in Egypt. Using parasitological and SOD1 enzyme assay, this study aimed to elucidate the impact of the age of B. alexandrina snails on their genetic variability and internal defence against S. mansoni infection. Susceptible and resistant snails were reared individually for self-reproduction; four subgroups of their progeny were used in experiment. The young susceptible subgroup showed the highest infection rate, the shortest pre-patent period, the highest total cercarial production, the highest mortality rate and the lowest SOD1 activity. Among the young and adult susceptible subgroups, 8% and 26% were found to be resistant, indicating the inheritance of resistance alleles from parents. The adult resistant subgroup, however, contained only resistant snails and showed the highest enzyme activity. The complex interaction between snail age, genetic background and internal defence resulted in great variability in compatibility patterns, with the highest significant difference between young susceptible and adult resistant snails. The results demonstrate that resistance alleles function to a greater degree in adults, with higher SOD1 activity and provide potential implications for Biomphalaria control. The identification of the most susceptible snail age enables determination of the best timing for applying molluscicides. Moreover, adult resistant snails could be beneficial in biological snail control.  相似文献

为阐明脊髓背角神经元痛放电的年龄相关的动力学变化,本研究采用非线性预报方法,对两组不同年龄大鼠(成年青龄鼠3~4月龄,老年鼠>22月龄)组织损伤诱发的脊髓背角神经元痛放电峰峰间期序列进行了确定性行为的定量分析.结果显示,皮下注入蜜蜂毒,在两组大鼠均诱发脊髓背角广动力域神经元长时程放电,而老龄大鼠的痛放电峰峰间期序列表现出更高的可确定性.本研究表明,单个神经元的痛放电动力学在整个生命过程中并不是恒定不变的,伤害性神经元活动的年龄相关动力学变化可能是老年人群中多样化痛反应的内在机制之一.  相似文献

Electric reactions of the identified neurons in adult (10-12 months) and old (22-24 months) molluscs Lymnaea stagnalis to endogenous peptide isolated from the water soluble fraction of the Helix Pomatia brain homogenate as well as to vasopressin are studied. The endogenous peptide causes a significant activation of the burst both in adult and old neurons of the molluscs. No significant age-related differences in the pronouncement of the above shifts of the molluscan neurons are found. There is an increased sensitivity to the identified peptide, vasopressin, in the old individuals. A non-uniform change in the sensitivity of mollusk neurons to various neuropeptides may affect the synaptic conduction in old age.  相似文献

Summary

To evaluate the influence of repeated mating on female fecundity in the simultaneously hermaphroditic, self-incompatible land snail Arianta arbus-torum, the number and size of clutches, egg size and hatching success of individuals from 3 populations were determined under conditions of isolation and grouping during one breeding season in a field cage experiment. Only adult snails which had mated and oviposited in the preceding year were used.

Sperm storage enabled isolated individuals to continue with the production of fertilized eggs. Snails kept singly or in groups differed neither in number of clutches laid nor in egg size. But isolated snails laid smaller clutches than did grouped snails. As a result isolated snails produced fewer eggs per breeding season. Hatching success varied greatly between populations and rearing conditions. In general, isolated snails had fewer hatchlings than grouped snails, indicating that individuals prevented from remating suffered a reduced fitness.  相似文献
