Carbachol chronically and peripherally administered, depending on the circadian time, caused a phase shift in the free-running locomotor activity rhythm, resynchronized the rhythm, or had a masking effect of this rhythm in mice. The locomotor activity rhythm of the sandhopper Talitrus saltator (Amphipoda Talitridae) was investigated in Madfoun beach. The rhythm was monitored in individual and group animals during 21 days at constant temperature. During the first two weeks, animals were kept under light–dark cycle (d1–d7) and constant darkness (d8–d14). Light pulses of 2 h were applied, at the end of the experimental night, during the first 3 days of the third week (d15–d18). Then, constant darkness was restored for the rest of the experiment (d19–d21). Results revealed that Talitrus saltator individuals as well as groups concentrated their activities in experimental night under light–dark cycle and in the presence of pulses; whereas, a clear drift of activity is observed under constant darkness. Periodogram analysis showed the presence of circadian component whatever the recording type. An ultradian period was revealed in individual recording whatever the regimen, while for groups, this component appeared only under nLD cycle. Furthermore, the locomotor rhythm is more defined under nLD cycle for individuals and in constant darkness for groups. 相似文献

Effects of single intranasal administration of 0.2 ng insulin at different moments of the projected daily cycle (ZT = 1, ZT = 7, ZT = 13 and ZT = 19) on the circadian rhythms of voluntary locomotor activity (wheel-running) were studied in Wistar male rats. Insulin administered at ZT-7 or ZT-13 induced a statistically significant phase advance by 4.4 and 5.5 hours, respectively. The administration of insulin at ZT-13 additionally induced a reduction of the period of the circadian rhythm of voluntary locomotor activity. Intranasal administration of insulin at other moments of the projected daily cycle (ZT = 1 or ZT = 19) did not induce any statistically significant change in phase or period duration of the circadian rhythms. Insulin did not cause changes in total daily activity irrespective of administration time. The results of the study suggest the role of endogenous insulin as entrainment factor for circadian oscillator in absence of the main physiological zeitgeber--cyclic afferent input from retina photoreceptors. 相似文献

An emerging body of evidence involves the hypothalamic neuropeptide melanin-concentrating hormone (MCH) in the regulation of emotional states. We have reported a pro-depressive effect induced by MCH after its microinjection into the dorsal raphe nucleus (DR) evaluated in the forced swimming test (FST) in rats. Here we extended this study to the median raphe nucleus (MnR). Firstly, the presence of MCH-containing fibers in the rat MnR was analyzed by means of immunohistochemistry. Secondly, the behavioral effect induced by the microinjection of MCH into the MnR was assessed using the FST. Morphological results showed a large density of MCHergic fibers within the MnR. Behavioral results indicated that 100 ng of MCH (but not 50 ng) significantly increased the immobility time and decreased the swimming time, demonstrating a depressive-like effect. In contrast, climbing behavior was not significantly affected. Present findings revealed that the MnR neurons participate in the MCHergic control of affective-related behavioral responses. However, the behavioral patterns induced by MCH in the MnR and DR were different. This could be explained by anatomical and physiological differences between both nuclei. 相似文献

Extracellular recordings were made of spontaneously active neurons in the dorsal raphe nucleus (DRN) of neonatal rats. The firing pattern and rate of these neurons were similar to those characterized for 5-HT-containing cells in the DRN of adult rats. Neonatal DRN cells were also inhibiteby small systematic doses of LSD, as previously described for 5-HT-containing DRN neurons in adult rats. These results indicate that DRN neurons in neonatal rats are physiologically active and display many characteristics similar to mature 5-HT-containing DRN neurons. 相似文献

The ultrastructure and synaptic relations of neurotensinergic neurons in the rat dorsal raphe nucleus (DRN) were examined. The neurotensin-like immunoreactive (NT-LI) neurons in the DRN were fusiform or spherical. The NT-LI perikarya could only be detected in colchicine-treated animals whereas the immunoreactive axon terminals could only be found in the anirnals not treated with colchicine. Although many NT-LI dendrites received synapses from nonimmunoreactive axon terminals, the NT-LI perikarya received few synapses. NT-LI axon terminals also made synapses on nonimmunoreactive dendrites. Occasionally, synapses were found between the NT-LI axon terminals and NT-LI dendrites in the cases in which the animals were not treated with colchicine. 相似文献

Entrainment of the nocturnal, endogenous locomotor activity rhythm of Talitrus saltator (Montagu) by the natural light-dark cycle is non-parametric, the phase of the rhythm shifting only in response to changes in the time of an experimentally simulated ‘dawn’ transition. The difference in response to light at ‘dusk’ and ‘dawn’ illustrates a phase-dependent responsiveness common to endogenous rhythms. Activity begins after complete darkness, with cessation always occurring during the ‘dawn’ transition and never continuing past the onset of total experimental illumination. The point of activity cessation is taken to be the position of a synchronizing cue controlling entrainment. The ‘dawn’ cue appears to be an absolute irradiance level of approximately 1.5 × 10?4 W/m2 (1.5 lux). The implications of such a cue are discussed in relation to field behaviour. 相似文献

The effects of microinfusingl-glutamate, serotonin (5-HT), (±)-8-hydroxy-2-(di-N-propylamino) tetralin (8-OH DPAT; a 5-HT1A agonist), and muscimol (a GABAA agonist) into the dorsal raphe nucleus on the extracellular levels of 5-HT, dopamine (DA) and their metabolites in the nucleus accumbens were studied in unanesthetized, freely moving, adult male Wistar rats, using the technique of microdialysis coupled with small-bore HPLC. Administration of 0.75 gl-glutamate produced a 25–50% increase (P<0.05) in the extracellular levels of both 5-HT and DA. On the other hand, infusion of 8-OH DPAT and, to a lesser extent, 5-HT produced a significant (P<0.05) decrease in the extracellular levels of both 5-HT and DA. Muscimol (0.25 or 0.50 g) had little effect on the extracellular concentrations of 5-HT or DA following its administration. In general, the extracellular levels of the major metabolites of 5-HT and DA in the nucleus accumbens were not altered by microinfusion of any of the agents. The data indicate that (a) the 5-HT neurons projecting to the nucleus accumbens from the dorsal raphe nucleus can be activated by excitatory amino acid receptors and inhibited by stimulation of 5-HT1A autoreceptors, and (b) the dorsal raphe nucleus 5-HT neuronal system may regulate the ventral tegmental area DA projection to the nucleus accumbens.Special issue dedicated to Dr. Morris H. 采用玻璃微电极在体细胞外记录法,观察了5,7-双羟色胺(5,7-dihydroxytryptamine,5,7-DHT)损毁大鼠中缝背核(dorsalraphenucleus,DRN)后,底丘脑核(subthalamicnucleus,STN)神经元电活动的变化。结果发现,对照组和DRN损毁组大鼠STN神经元的放电频率分别是(6.93±6.55)Hz和(11.27±9.31)Hz,DRN损毁组大鼠的放电频率显著高于对照组(P<0.01)。在对照组大鼠,13%的神经元呈现规则放电,46%为不规则放电,41%为爆发式放电;而在DRN损毁组大鼠,具有规则、不规则和爆发式放电的神经元比例分别为9%、14%和77%,爆发式放电的STN神经元比例明显高于对照组(P<0.01)。结果显示,DRN损毁后大鼠STN神经元的放电频率增高,爆发式放电增多,提示在正常大鼠DRN抑制STN神经元的活动。 相似文献

Background
The dorsal raphe nucleus (DRN) of the mesencephalon is a complex multi-functional and multi-transmitter nucleus involved in a wide range of behavioral and physiological processes. The DRN receives a direct input from the retina. However little is known regarding the type of retinal ganglion cell (RGC) that innervates the DRN. We examined morphological characteristics and physiological properties of these DRN projecting ganglion cells.Methodology/Principal Findings
The Mongolian gerbils are highly visual rodents with a diurnal/crepuscular activity rhythm. It has been widely used as experimental animals of various studies including seasonal affective disorders and depression. Young adult gerbils were used in the present study. DRN-projecting RGCs were identified following retrograde tracer injection into the DRN, characterized physiologically by extracellular recording and morphologically after intracellular filling. The result shows that DRN-projecting RGCs exhibit morphological characteristics typical of alpha RGCs and physiological response properties of Y-cells. Melanopsin was not detected in these RGCs and they show no evidence of intrinsic photosensitivity.Conclusions/Significance
These findings suggest that RGCs with alpha-like morphology and Y-like physiology appear to perform a non-imaging forming function and thus may participate in the modulation of DRN activity which includes regulation of sleep and mood. 相似文献15.

Summary Pinealectomy of house sparrows on 3L:21D (3 h light per 24 h) resulted in a significant increase in the time between the onset of perch-hopping activity and lights on (on) as well as the time between the offset of activity and lights of (off). The daily variance in on and off was also increased following the removal of the pineal gland. On longer light cycles (i.e., 5L:19D; 7L:17D), neither on or off, nor the variance of on or off was different between sham-pinealectomized and pinealectomized sparrows. Upon returning the birds to an ultrashort light cycle, 1L:23D, off, as well as the variance in on and off were again found to be significantly larger in the pinealectomized birds when compared to sham-operated controls. These results indicate that the effects of pinealectomy on the entrained rhythm of locomotor activity are most pronounced when birds are exposed to a weak entraining agent, such as an ultrashort light: dark cycle. In view of the observation that pinealectomy can alter the phase relationship between activity onset and offset, it is suggested that the pineal gland may be involved in the coupling of the oscillators that regulate activity onset and offset. 相似文献

Dopamine (DA) neurons of the ventrolateral periaqueductal gray (vlPAG) and dorsal raphe nucleus (DRN) fire spontaneous action potentials (APs) at slow, regular patterns in vitro but a detailed account of their intrinsic membrane properties responsible for spontaneous firing is currently lacking. To resolve this, we performed a voltage-clamp electrophysiological study in brain slices to describe their major ionic currents and then constructed a computer model and used simulations to understand the mechanisms behind autorhythmicity in silico. We found that vlPAG/DRN DA neurons exhibit a number of voltage-dependent currents activating in the subthreshold range including, a hyperpolarization-activated cation current (IH), a transient, A-type, potassium current (IA), a background, ‘persistent’ (INaP) sodium current and a transient, low voltage activated (LVA) calcium current (ICaLVA). Brain slice pharmacology, in good agreement with computer simulations, showed that spontaneous firing occurred independently of IH, IA or calcium currents. In contrast, when blocking sodium currents, spontaneous firing ceased and a stable, non-oscillating membrane potential below AP threshold was attained. Using the DA neuron model we further show that calcium currents exhibit little activation (compared to sodium) during the interspike interval (ISI) repolarization while, any individual potassium current alone, whose blockade positively modulated AP firing frequency, is not required for spontaneous firing. Instead, blockade of a number of potassium currents simultaneously is necessary to eliminate autorhythmicity. Repolarization during ISI is mediated initially via the deactivation of the delayed rectifier potassium current, while a sodium background ‘persistent’ current is essentially indispensable for autorhythmicity by driving repolarization towards AP threshold. 相似文献

The ultrastructure and the synaptic relationships of the orexin-A-like immunoreactive fibers in the dorsal raphe nucleus were examined with an immunoelectron microscopic method. At the electron microscopic level, most of the immunoreactive fibers, a varicosity appearance at the light microscopic level, were found as axon terminals. The large dense-cored vesicles contained in the immunoreactive axon terminals were the most intensely immunostained organellae. These axon terminals were often found to make synapses. While the axo-dendritic synapses were usually asymmetric in appearance, the axo-somatic synapses were symmetric. Orexin-A-like immunoreactive processes with no synaptic vesicles were also found. These processes often received asymmetric synapses. With less frequency, the synapses were found between the orexin-like immunoreactive processes. The results suggest that the orexin peptides are stored in the large dense-cored vesicles; the orexin-containing fibers may have influences on the physiological activities of the dorsal raphe nucleus through direct synaptic relationships. 相似文献

The effect of injections of 5,6-dihydroxytryptamine, a potent and selective neurotoxic of serotonin neurons, into amygdala and dorsal raphe mesencephalic nucleus on the plasma renin activity has been studied in male Wistar rats. Plasma renin activity was estimated on 2nd, 4th, Tth and 14th day after injections in both areas. The administration of 5,6-dihydroxytryptamine in amigdala produced a significant decrease in plasmatic renin activity between 2nd and 4th day, but the inverse effect between 7th and 14th day. Similar effects were found after injections in dorsal raphe nucleus. The contents of cerebral 5-HT were simultaneously evaluated in the entire brain when the drug was implanted in dorsal raphe, and only in amygdaloid tissue when the injection was restricted to this area. A significant decrease in serotonin content was produced 7th day in both places, while partial recuperation was found toward 14th day. The results, especially the ones related to the chemical lesion of dorsal raphe nucleus, suggest that serotoninergic brain systems are involved, as stimulators, in the control of the dynamics of renin-angiotensin system. 相似文献

Crayfish Procambarus acanthophorus is a burrower that spends long periods building deep tunnels to reach the water table during the dry season; thus, its survival entertains a close ecological relationship with the sediment. The aim of this work was to determine whether the properties of the circadian rhythm of locomotor activity could be modified by the sediment availability. Experiments were conducted in both aquaria filled with sediment or filled with water, under cycles of Bright and Dim Illumination (BI:DI, 12:12 h) or under continuous DI:DI. The rhythm of locomotor activity was entrained with the photoperiod in aquaria with sediment or water; however, statistical differences between conditions were obtained comparing the period and the level of activity under free-running. These data suggest that the substrate’s sensorial perception could be encoded as a significant ecological parameter that exerts influence in the physiological mechanisms that control the temporal order in P. acanthophorus. 相似文献

In the dorsal raphe nucleus (DR), extracellular serotonin (5-HT) regulates serotonergic transmission through 5-HT1A autoreceptors. In this work we used in vivo microdialysis to examine the effects of stressful and pharmacological challenges on DR 5-HT efflux in 5-HT1A receptor knockout (5-HT1A-/-) mice and their wild-type counterparts (5-HT1A+/+). Baseline 5-HT concentrations did not differ between both lines of mice, which is consistent with a lack of tonic control of 5-HT1A autoreceptors on DR 5-HT release. (R)-(+)-8-Hydroxy-2-(di-n-propylamino)tetralin hydrobromide (8-OH-DPAT, 0.5 mg/kg) reduced 5-HT levels to 30% of basal values in 5-HT1A+/+ mice, but not in 5-HT1A-/- mice. The selective 5-HT1B receptor agonist 1,4-dihydro-3-(1,2,3,6-tetrahydro-4-pyridinyl)-5H-pyrrolo[3,2-b]pyridin-5-one dihydrochloride (CP 93129, 300 micro m) reduced dialysate 5-HT to the same extent (30-40% of baseline) in the two genotypes, which suggests a lack of compensatory changes in 5-HT1B receptors in the DR of such mutant mice. Both a saline injection and handling for 3 min increased DR dialysate 5-HT in mutants, but not in 5-HT1A+/+ mice. Fluoxetine (5 and 20 mg/kg) elevated 5-HT in a dose-dependent manner in both genotypes. However, this effect was markedly more pronounced in the 5-HT1A-/- mice. The increased responsiveness of the extracellular 5-HT in the DR of 5-HT1A receptor knockout mice reflects a lack of the autoinhibitory control exerted by 5-HT1A autoreceptors. 相似文献
