Continuous exposure of Biomphalaria glabrata snails to 0.1% colchicine resulted in a significant increase, relative to non-exposed snails, in the number of arrested mitotic figures in the amebocyte-producing organ (APO) as soon as 4 h, with peak numbers after 12 h of exposure. The number of circulating hemocytes was significantly elevated at 24 h. However, by 72 h both the number of mitotic figures in the APO and the concentration of circulating hemocytes in the hemolymph had returned to control levels. Hemocytes appeared to possess normal morphology throughout this exposure, including the formation of long filopodia with supporting rodlike structures that have been reported to contain microtubules. Snail survival decreased as a function of exposure time. Significantly fewer snails, relative to controls, survived a 48-h exposure, and only 1 out of 30 snails recovered from a 72-h exposure to 0.1% colchicine. Colchicine-exposed snails displayed intoxicated behavior, even upon removal from the colchicine solution, although no histopathology was evident in the CNS of snails exposed for 72 h.  相似文献

Suspensions of miracidia and cercariae of Schistosoma mansoni were subjected to repeated freeze-thaw cycles and then injected into resistant Salvador strain Biomphalaria glabrata snails. A pronounced increase in the number of mitotic figures, relative to uninjected, sham-injected, or diluent (water)-injected controls, was observed in the amebocyte-producing organ (APO) at 3 days postinjection (PI). After centrifugation of miracidia freeze-thaw extract (FTE), the resulting supernatant (FTS) and pellet possessed equal stimulatory activity that was approximately half that seen with FTE. Ultracentrifugation of miracidia FTS resulted in a supernatant that retained full activity, indicating a soluble molecule. Heat treatment of miracidia FTE reduced but did not eliminate activity, suggesting a nonprotein active component. Concentration or dilution of FTS by a factor of 10 gave a nonlinear dose-response relationship. Susceptible NIH albino snails injected with miracidia FTE had increased mitotic activity in the APO, which was much less than that seen in Salvador snails, whereas injection of miracidia FTE into Helisoma duryi had no discernable effect. Measurement of mitotic activity as a function of time PI showed no increase in numbers of mitotic figures in the APO at 18 hr but a large increase at 24 hr PI. Mitotic activity returned to preinjection levels by 96 hr PI, although a subsequent increase occurred at 120 hr PI.  相似文献

Balb/c/nu nude mice transplanted with a moderately differentiated squamous cell carcinoma were injected intraperitoneally with different doses of aqueous skin extracts containing the epidermal G2 chalone. The mitotic counts and the mitotic rates were determined in histological sections using a stathmokinetic method with vinblastine sulphate. The mitotic duration was calculated from the mitotic rates and counts. Skin extracts containing epidermal G2 chalone increased the mitotic duration in the epidermis, and a similar trend was seen in the tumour. The higher the dose of chalone, the longer the mitotic duration tended to be. A straight line of best fit used to indicate the dose/response relationship was steeper for the epidermis than for the tumour. The study thus shows that the epidermal G2 chalone not only prevents epidermal cells from entering mitosis, it also prolongs the mitotic duration. Further, the results do not contradict the theory that tumour cells may be less sensitive to chalone than normal cells.  相似文献

The synthesis of post-translationally modified tubulins was examined during Artemia development. Tubulin, either purified to homogeneity or in cell-free extracts, was blotted to nitrocellulose and probed with a panel of antibodies. When purified tubulin was examined, tyrosinated tubulin underwent a large decrease as development progressed and this was accompanied by the appearance of detyrosinated tubulin in samples from organisms developed 24 hr. The inclusion of carboxypeptidase inhibitors had a small effect on the relative amounts of tyrosinated and detyrosinated tubulins in 24-hr preparations. The amount of alpha- and beta-tubulin in cell-free extracts of Artemia either remained relatively constant during development or increased slightly. The same result was obtained for acetylated and tyrosinated tubulin. Detyrosinated tubulin first appeared in 24-hr cell-free extracts and was only post-translationally modified tubulin to increase, relative to the total amount of tubulin, as the brine shrimp developed. As revealed by immunofluorescence staining, detyrosinated tubulin occurred in many cell types of developing nauplii and was prominently displayed in mitotic figures. Artemia, a complex metazoan animal, is thus able to grow for an extended period of time in the absence of detyrosinated tubulin. This isoform is however, synthesized in early larvae and may be required for the development of elongated cells including those which encircle the gut. Detyrosination remains as the only developmentally related change observed for brine shrimp tubulin.  相似文献

Abstract
Data concerning changes in the rate of cell proliferation of stratified epithelia with increasing age are conflicting. In the present study young (3-month-old) and old (22-month-old) C57B1/6NNia male mice were injected intraperitoneally with 2, 3, 4 or 8 mg vinblastine sulfate/kg body weight and killed after 1.5, 3, 4.5 or 6 h. The number of arrested metaphase figures per 1000 basal cells was counted in histological sections. Data were analysed using a multivariate analysis of variance. There was a significant difference between the accumulation of mitotic figures in footpad epidermis and palate epithelium and both tissues contained an increased number of mitotic figures with increasing periods of accumulation at all dose levels. In the footpad epidermis neither the age of the animal nor the dose of vinblastine had a significant effect on the number of mitotic figures. In contrast, for palate epithelium the accumulation of mitotic figures was significantly less in the old mice compared with the young mice and at a dose of vinblastine of 2mg/kg compared with the higher doses. There was a statistically signifycant interaction between the dose of vinblastine and its period of action. It was concluded that the different tissues manifest a differential sensitivity to vinblastine and that only palate epithelium showed a significant reduction in proliferative activity with age.  相似文献

Snail hosts of different ages constitute different resource environments for larval trematodes because each individual has a particular energy budget and energy allocation pattern at the time of infection. Effects of monomiracidial trematode infections on shell growth, body dry weight and reproductive effort of the snail host were compared between controls and infected juvenile and adult snails during the prepatent and patent periods. The results demonstrate phenotypic plasticity in the host response. There is an age/size-specific effect characterized by limitation of growth rate when snails are infected as juveniles and reduction of reproductive effort when snails are infected as adults, and a time-specific effect with early enhancement of growth rate and reproductive effort for infected juvenile and adult snails respectively during prepatency, before reduction and cessation during patency. The parasite is considered as a generalist in its energy exploitation strategy of the snail host, but taking into account the differential host responses observed relative to the host age, the possibility of a host preference for juvenile or adult snails is discussed. Responses of the hematopoietic organ (HO) in Biomphalaria glabrata snails to extracts and excretory-secretory (E-S) products of Echinostoma paraensei larvae were studied to understand the HO-activating mechanism. M-line B. glabrata snails were injected with materials from E. paraensei larvae, and the size of the HO was ascertained in histological sections. The size of HO in snails injected with extracts and E-S products from sporocysts and rediae was significantly larger than that in snails injected with culture medium. E-S products of sporocysts were fractionated using ultrafiltration membranes, polyacrylamide gel electrophoresis, and electrophoretic elution. Examination of fractionated E-S products of sporocysts revealed that specific components of E-S products were responsible for HO-stimulating activity.  相似文献

The extracts from ocular tentacles of Helix aspersa hare a heterochronic inhibitive action on the development of albumen gland. In view of elucidating the origin of this heterochrony, we make an electrophoretic study of the different extracts from juveniles, adults in activity, in natural sleep and awakning. This study has allowed us to spot 19 proteic fractions common to the four extracts of the tentacles under study, but these fractions show only differences in concentration. Particularly, the proteinic fractions 7 and 8 are more concentrated in the extract of juveniles snails tentacles than in the extract of adults snails. This difference in proteic concentration could explain the heterochronic inhibitive action of ocular tentacles extracts on the development of albumen gland of this snail.  相似文献

Amebocyte-producing organs (APOs) of Biomphalaria glabrata were maintained in nonnutritive saline with, or without, extracts of miracidia and adults of Schistosoma mansoni, and examined histologically. The hematopoietic cells remained viable and showed measurable mitotic activity for up to 6 days, with little evidence of tissue death. APOs accumulated fluid and became swollen by as soon as 24 hr, but no cell exomigration was observed. Parasite extracts elicited an increase in the number of dividing cells in the APO, suggesting that the extract may directly stimulate a response from the hematopoietic cells by providing either nutrients or mitogenic growth factors.  相似文献

The effects of aestivation or starvation on the neutral lipid and phospholipid content of Biomphalaria glabrata patently infected with Schistosoma mansoni were determined by high-performance thin-layer chromatography-densitometry. Infected-aestivated snails were maintained in a moist chamber at 24 +/- 1 C and a relative humidity of 98 +/- 1%. Infected-starved snails were maintained in artificial spring water (ASW) at 23 +/- 1 C without exogenous food. Infected snails (the controls) were maintained in ASW at 23 +/- 1 C and fed lettuce ad libitum. The 3 groups were maintained in the laboratory for 7 days, and then the lipids from the digestive gland-gonad complex (DGG) were extracted and analyzed by class. Infected-aestivated snails exhibited greater mortality rate and weight loss after 7 days than did the infected-starved snails. The steryl ester concentration in the infected-starved snails was significantly increased (P = 0.010) compared with the controls but not compared with infected-aestivated snails; the concentration of phosphatidylcholine in infected-aestivated snails was significantly decreased (P = 0.007) compared with the controls but not when compared with the infected-starved snails. Aestivation or starvation had a significant effect on the concentration of certain lipid classes in the DGG of B. glabrata infected with S. mansoni.  相似文献

The functional significance of AMP deaminase and adenosine deaminase has been studied in hepatopancreas of active, aestivated and ganglionic extracts-administered snails. The activity levels of both enzymes decreased in aestivating snails. Active snails injected with ganglionic extracts of aestivated snails also showed decreased activity. Contrastingly, the hepatopancreas of aestivated snails when treated with ganglionic extracts of active snails showed increased specific activities of both enzymes. The decrease or increase in the specific activities varied with different ganglionic extracts and the significance of the same is discussed.  相似文献

Root tips (1.0 cm) were excised, transferred to culture medium,sampled at intervals, embedded in paraffin and sectioned. Themitotic index in each primary meristem in the 2.0 mm tips wasreduced immediately after excision. After 8–12 h mitosisresumed. The position of mitotic figures in the protoderm, groundmeristem, procambium, and root cap was determined. The greatestnumber of mitotic figures occurred in the procambium, and uponexcision the smallest percent reduction also occurred in thisregion. The ground meristem showed the greatest reduction inmitotic figures. After excision, the relative contribution tothe total number of mitotic figures increased in the procambiumtissue and decreased in the ground meristem. Cell division inall primary meristems of the root tip is reduced after tip excision,but the ground meristem (cortical region) is the primary targettissue.  相似文献

Global warming may affect snail–periphyton–macrophyte relationships in lakes with implications also for water clarity. We conducted a 40-day aquaria experiment to elucidate the response of submerged macrophytes and periphyton on real and artificial plants to elevated temperatures (3°C) under eutrophic conditions, with and without snails present. With snails, the biomass and length of Vallisneria spinulosa leaves increased more at the high temperature, and at both temperatures growth was higher than in absence of snails. The biomass of periphyton on V. spinulosa as well as on artificial plants was higher at the highest temperature in the absence but not in the presence of snails. The biomass of Potamogeton crispus (in a decaying state) declined in all treatments and was not affected by temperature or snails. While total snail biomass did not differ between temperatures, lower abundance of adults (size >1 cm) was observed at the high temperatures. We conclude that the effect of elevated temperature on the snail–periphyton–macrophyte relationship in summer differs among macrophyte species in active growth or senescent species in subtropical lakes and that snails, when abundant, improve the chances of maintaining actively growing macrophytes under eutrophic conditions, and more so in a warmer future with potentially denser growth of periphyton.  相似文献

Previous studies have yielded equivocal results concerning the 24-hour rhythmicity of mitotic activity in the rat pineal. The aim of the present study was to re-investigate this problem by carrying out three separate 24-hour experiments on alternate days. The results obtained confirm previous findings showing that in the pineal gland of adults mitotic activity is low. On average 22.3 mitotic figures of pinealocytes are seen per pineal gland, corresponding to a mitotic index of 0.2-0.6/1,000 pinealocytes. Mitotic activity is distinctly higher at daytime than at night. The timing of the peaks and troughs differs slightly from experiment to experiment. The majority of observations now indicate that in the rat pineal gland mitotic activity is higher at day time than at night.  相似文献

We have shown that inhibition of mTOR in granulosa cells and ovarian follicles results in compromised granulosa proliferation and reduced follicle growth. Further analysis here using spontaneously immortalized rat granulosa cells has revealed that mTOR pathway activity is enhanced during M-phase of the cell cycle. mTOR specific phosphorylation of p70S6 kinase and 4E-BP, and expression of Raptor are all enhanced during M-phase. The predominant effect of mTOR inhibition by the specific inhibitor Rapamycin (RAP) was a dose-responsive arrest in the G1 cell cycle stage. The fraction of granulosa cells that continued to divide in the presence of RAP exhibited a dose-dependent increase in aberrant mitotic figures known as anaphase bridges. Strikingly, estradiol consistently decreased the incidence of aberrant mitotic figures. In mice treated with RAP, the mitotic index was reduced compared to controls, and a similar increase in aberrant mitotic events was noted. RAP injected during a superovulation regime resulted in a dose-dependent reduction in the numbers of eggs ovulated. Implications for the real-time regulation of follicle growth and dominance, including the consequences of increased numbers of aneuploid granulosa cells, are discussed.  相似文献

Lymphocyte transformation, utilizing the mitotic index parameter has been assessed in newborns' cord blood, adults and women who had been on oral contraceptives and their babies. The results show that the newborns' cord blood response is significantly higher to PHA when compared with the respective adults' response. Levels of lysozyme activity were determined in the hemolymph, digestive gland, and headfoot extracts of M-line stock of snails, Biomphalaria glabrata, during infection with the PR-1 and Lc-1 strains of the trematode, Schistosoma mansoni. At 3 hr postexposure there was a 10-fold increase in the levels of enzyme activity in the hemolymph of snails infected with the Lc-1 strain to which the snail is resistant. This increase was considerably higher when compared to the threefold increase in the PR-1-infected snails. The infection also induced a gradual depletion of lysozyme activity in the headfoot muscles of the two groups of infected snails. There were no changes in the levels of enzyme activity in the digestive gland extracts of the control and the two groups of infected snails. Similar changes in the levels of enzyme activity in the hemolymph and headfoot extracts of infected snails suggest a nonspecific response to a parasite infection and do not indicate that lysozyme is primarily responsible for the destruction of schistosome parasite in a resistant snail host.  相似文献

The distribution of mobile species such as grazing snails may be influenced by migration patterns, which often are excluded from ecological studies. To highlight this point, the migration patterns of Littoraria scabra on mangrove trees were investigated during incoming and outgoing tides at Nanaru-i-ra, Fiji Islands. Marked snails were used to track the position of snails, relative to the ground, during advancing and receding tides. Snails were found to move quickly upward during incoming tides, potentially to avoid immersion. During the outgoing tide, snails migrated downward, but at a slower pace than upward movement, presumably as a consequence of active feeding on the newly replenished micro-organisms on the mangrove structures (i.e., stems and roots). Aggregation behavior during migration was also recorded by measuring the distance of marked snails to two nearest neighbors. Affinity to neighboring snails was found to be maintained throughout tidal cycles, although greater affinity was observed during incoming tides compared to outgoing tides. These migratory and aggregation behaviors may be a result of increased feeding and reproductive efficiency.  相似文献

Summary Previous work on a snail community occurring throughout lakes in southwestern Michigan showed that predation by molluscivorous sunfish had large impacts on only the rarest snail species. Thus, competition might play a major role in population limitation because dominant members of the snail community are relatively immune to predation. The present experiments were conducted to determine the extent to which the snail community depleted the abundance of food resources (epiphytes) and the extent to which epiphyte abundances limited snail production. An experimental gradient in snail densities showed that removal of snails increased epiphyte biomass by approximately 3-fold relative to that observed at natural snail densities. Enrichment of the environment with phosphorus fertilizer increased epiphyte biomass by approximately 20-fold and provided a test of food limitation in the snail community. All snail taxa exhibited positive numerical or growth responses to enrichment. The observations that snails depleted resources and that resources limited snail production demonstrated that snails competed exploitatively for epiphytes. The response of each snail species to increased food abundance differed depending on the timing of fertilization relative to the snails' life histories. Snails hatched before the experiment began were larger in fertilized treatments, due to increased growth, but their densities were similar among treatments. On the other hand, densities of snails born during the experiment were up to 15-fold greater in fertilized treatments, due in part to increased survival of newborn snails. Comparison of the responses of snails to food addition and to predator removals (based on prior experiments) suggested that food availability limits snail production more than predators do. Additionally, the large responses by algae and snails to fertilization demonstrated that both the producers and herbivores in this simplified food chain were strongly resource limited.  相似文献

Preparations of surface stretched amembranous nuclei and mitotic figures were used for revealing the high order nuclear and chromosomal structures. The preparations were obtained by dropping amembraneous nuclei and mitotic figures suspension in methanol-glacial acetic acid mixture (3:1) on wetted superclean slides. Amembraneous nuclei and mitotic figures were isolated from intact murine and human cells (lines L1210, SK-UT-1B, PHA-stimulated lymphocytes) by means of their 1-5 min prefixational capillary pipetting with freshly prepared 0.018-0.06% Triton X-100 solution in the conditional cultural medium. Stretched amembraneous nuclei and mitotic figures had no features of induced chromatin dispersion and compaction. Stretched interphase amembraneous nuclei showed spatially separated individual structures (thin chromatin fibres, nucleoli, intranuclear bodies), polymorphous pattern of perinucleolar chromatin aggregation and episodically expressed beaded thick chromatin fibres and a chromocenter. The chromomeric pattern of the spread chromosomes of mitotic figures was quite similar but hardly identical with that of G-banding. The stretched prometaphase mitotic figures in all tested cell types always contained loose "residual" nucleoli looking like typical prophase nucleoli as concerns their shape and number per cell (mitotic figure). The majority of chromosomes of stretched mitotic figures and of prophase amembraneous nuclei were attached to the nucleolar material. All tested cell lines showed almost the same variation in number of nucleolus-attached chromosomes, per both prophase amembraneous nucleus and prometaphase mitotic figure. Some chromosomes of stretched mitotic figures were colocated with "residual" nucleoli and looked shortened and strongly condensed. Other chromosomes, locally associated with "residual" nucleoli, were straight and oriented radially to these. Mutual chromosomal arrangements in mitotic cells on smears and in stretched mitotic figures were analogous. Equatorial plates from PBS-washed SK-UT-1B cells displayed a better stretching capacity than those from untreated cells. In the former case metaphase chromosomes were seen more uniformly stretched and well identified after GTG-banding procedure. The number of interchromosomal (mainly telomere-telomeric and telomere-centromeric) connections per stretched mitotic figure (or per stretched prophase amembraneous nucleus) was minimum in late prometaphase, maximum in prophase and early prometaphase, and intermediate in metaphase. The obtained data are discussed in terms of topology and longitudinal heterogeneity of mitotic chromosomes.  相似文献
