Little is known about the presence/absence and prevalence of Rickettsia spp, Bartonella spp. and Yersinia pestis in domestic and urban flea populations in tropical and subtropical African countries.

Methodology/Principal findings

Fleas collected in Benin, the United Republic of Tanzania and the Democratic Republic of the Congo were investigated for the presence and identity of Rickettsia spp., Bartonella spp. and Yersinia pestis using two qPCR systems or qPCR and standard PCR. In Xenopsylla cheopis fleas collected from Cotonou (Benin), Rickettsia typhi was detected in 1% (2/199), and an uncultured Bartonella sp. was detected in 34.7% (69/199). In the Lushoto district (United Republic of Tanzania), R. typhi DNA was detected in 10% (2/20) of Xenopsylla brasiliensis, and Rickettsia felis was detected in 65% (13/20) of Ctenocephalides felis strongylus, 71.4% (5/7) of Ctenocephalides canis and 25% (5/20) of Ctenophthalmus calceatus calceatus. In the Democratic Republic of the Congo, R. felis was detected in 56.5% (13/23) of Ct. f. felis from Kinshasa, in 26.3% (10/38) of Ct. f. felis and 9% (1/11) of Leptopsylla aethiopica aethiopica from Ituri district and in 19.2% (5/26) of Ct. f. strongylus and 4.7% (1/21) of Echidnophaga gallinacea. Bartonella sp. was also detected in 36.3% (4/11) of L. a. aethiopica. Finally, in Ituri, Y. pestis DNA was detected in 3.8% (1/26) of Ct. f. strongylus and 10% (3/30) of Pulex irritans from the villages of Wanyale and Zaa.

Conclusion

Most flea-borne infections are neglected diseases which should be monitored systematically in domestic rural and urban human populations to assess their epidemiological and clinical relevance. Finally, the presence of Y. pestis DNA in fleas captured in households was unexpected and raises a series of questions regarding the role of free fleas in the transmission of plague in rural Africa, especially in remote areas where the flea density in houses is high.  相似文献

Rickettsia felis is a rickettsial pathogen primarily associated with the cat flea, Ctenocephalides felis. Although laboratory studies have confirmed that R. felis is maintained by transstadial and transovarial transmission in C. felis, distinct mechanisms of horizontal transmission of R. felis among cat fleas are undefined. Based on the inefficient vertical transmission of R. felis by cat fleas and the detection of R. felis in a variety of haematophagous arthropods, we hypothesize that R. felis is horizontally transmitted between cat fleas. Towards testing this hypothesis, flea transmission of R. felis via a bloodmeal was assessed weekly for 4 weeks. Rhodamine B was used to distinguish uninfected recipient and R. felis-infected donor fleas in a rickettsial horizontal transmission bioassay, and quantitative real-time PCR assay was used to measure transmission frequency; immunofluorescence assay also confirmed transmission. Female fleas acquired R. felis infection more readily than male fleas after feeding on a R. felis-infected bloodmeal for 24 h (69.3% and 43.3%, respectively) and both Rickettsia-uninfected recipient male and female fleas became infected with R. felis after cofeeding with R. felis-infected donor fleas (3.3-40.0%). Distinct bioassays were developed to further determine that R. felis was transmitted from R. felis-infected to uninfected fleas during cofeeding and copulation. Vertical transmission of R. felis by infected fleas was not demonstrated in this study. The demonstration of horizontal transmission of R. felis between cat fleas has broad implications for the ecology of R. felis rickettsiosis.  相似文献

Los Angeles and Orange Counties are known endemic areas for murine typhus in California; however, no recent reports of flea‐borne rickettsioses are known from adjacent San Bernardino County. Sixty‐five opossums (Didelphis virginiana) were trapped in the suburban residential and industrial zones of the southwestern part of San Bernardino County in 2007. Sixty out of 65 opossums were infested with fleas, primarily cat fleas, Ctenocephalides felis (Bouché, 1835). The flea minimum infection rate with Rickettsia felis was 13.3% in pooled samples and the prevalence was 23.7% in single fleas, with two gltA genotypes detected. In spite of historic records of murine typhus in this area, no evidence for circulation of R. typhi in fleas was found during the present study. Factors contributing to the absence of R. typhi in these cat fleas in contrast to its presence in cat fleas from Orange and Los Angeles Counties are unknown and need to be investigated further in San Bernardino County.  相似文献

Background

Since its first detection, characterization of R. felis has been a matter of debate, mostly due to the contamination of an initial R. felis culture by R. typhi. However, the first stable culture of R. felis allowed its precise phenotypic and genotypic characterization, and demonstrated that this species belonged to the spotted fever group rickettsiae. Later, its genome sequence revealed the presence of two forms of the same plasmid, physically confirmed by biological data. In a recent article, Gillespie et al. (PLoS One. 2007;2(3):e266.) used a bioinformatic approach to refute the presence of the second plasmid form, and proposed the creation of a specific phylogenetic group for R. felis.

Methodology/Principal Findings

In the present report, we, and five independent international laboratories confirmed unambiguously by PCR the presence of two plasmid forms in R. felis strain URRWXCal2 T, but observed that the plasmid content of this species, from none to 2 plasmid forms, may depend on the culture passage history of the studied strain. We also demonstrated that R. felis does not cultivate in Vero cells at 37°C but generates plaques at 30°C. Finally, using a phylogenetic study based on 667 concatenated core genes, we demonstrated the position of R. felis within the spotted fever group.

Significance

We demonstrated that R. felis, which unambiguously belongs to the spotted fever group rickettsiae, may contain up to two plasmid forms but this plasmid content is unstable.  相似文献

A total of 20 plants belonging to different genera (Acalypha, Ageratum, Ambrosia, Bidens, Blechum, Caesalpinia, Calea, Carlowrightia, Croton, Eugenia (2), Furcraea, Stenandrium, Tephrosia, Trichilia (2), Randia (3), and Vitex) were selected from native flora of the Yucatan peninsula. All plants selected were collected and separated in to leaves, stems and roots. These were then extracted with ethanol and their crude extracts (54) were evaluated against Alternaria tagetica, Colletotrichum gloeosporioides, Fusarium oxysporum and Rhizopus sp. using the filter paper disc diffusion assay. Results lead to the selection of 33 crude extracts active against at least one of the target strains, which were assessed to determine their ability to inhibit the mycelial growth of the pathogenic fungi in a second antifungal assay. The results of this assay indicated that extracts from the roots of Croton chichenensis were the most promising, with a wide activity spectrum against all pathogens tested in both assays with inhibition percentages of greater than 60%. Furthermore, extracts from leaves of Ambrosia hispida, Trichilia minutiflora, and roots of Acalypha gaumeri were able to cause growth inhibition against two or three pathogen strains (≥50%). Studies of these active extracts should be continued at different levels. In general, results revealed a good bioactive potential of the flora from the Yucatan peninsula to produce metabolites with potential applications as botanical pesticides in the near future.  相似文献

Overexploitation of Neotropical ecosystems for wild meat constitutes a threat to biodiversity and might seriously affect local people who depend on wild animals as a primary food resource. To assess this problem at the local level, social use of wild animals was evaluated in a Mayan community of the Yucatan Peninsula. Over 8 months, peasant-hunters carried out a total of 175 hunting trips, gathering 253 animals of 12 species (mammals, birds and reptiles). Most preys (95%) were taken in 4 months (January–April) during the dry season and the main target species were white-tailed deer and iguana (jointly representing 59% of preys). During an 11-week period, total per capita wild meat consumption (182 kg; n = 14 families) was three times greater in nuclear than in extended families. The observed wild fauna use is a reflection of traditional knowledge and represents an important subsistence strategy for the local people.  相似文献

Consistent with the effects of HIV on cell‐mediated immunity, an increased susceptibility to intracellular microorganisms has been observed. Rickettsiae are obligate intracellular microorganisms. The aim of this study was to examine Rickettsia typhi and Rickettsia felis infections in HIV+ population. Sera of 341 HIV+ patients were evaluated by indirect immunofluorescent assay. Age, sex, residential locality, risk behavior, stage according to criteria of the Center for Disease Control and Prevention, CD4+/CD8+ T cells, Hepatitis B antigen, and Hepatitis C serology were surveyed. Seroprevalences of R. typhi and R. felis infection were 7.6% and 4.4%, respectively. No associations were found between seropositivities and the assessed variables. Findings were similar to those obtained in healthy subjects from the same region.  相似文献

Extracts from 48 marine macroalgae species (17 Chlorophyta, 8 Phaeophyta and 23 Rhodophyta) from the coasts of Yucatan and Quintana Roo (Mexico) were evaluated for antioxidant activity. The antioxidant activity was measured with the DPPH (2,2-diphenyl-1-picrylhydrasyl) method, and the phenolic content of each extract were also evaluated. All species exhibited a DPPH radical scavenging activity, and three species (Avrainvillea longicaulis, Chondria baileyana and Lobophora variegata) demonstrated great antioxidant potential with very low oxidation index EC50 (1.44 ± 0.01, 2.84 ± 0.07 and 0.32 ± 0.01 mg mL−1, respectively), significantly equivalent to EC50 of some commercial antioxidants such as α-tocopherol, ascorbic acid, BHA and BHT. Moreover, extracts of the most active species exhibited reducing activities, superoxide anion radical scavenging and inhibition of lipid peroxidation. These results suggest that some macroalgae from the Yucatan peninsula have a great antioxidant potential which could be considered for future applications in medicine, food production or cosmetic industry.  相似文献

Distribution of Herbal Remedy Knowledge in Tabi, Yucatan, Mexico. The distribution of herbal remedy knowledge among a group of people is studied for two main reasons: (1) to identify plants that are promising for pharmacological analysis, and (2) to examine the factors that lead to herbal remedy knowledge erosion as opposed to dynamism in the acquisition of knowledge. The goal of this particular study, which is aligned with the second reason, is to establish the variation in herbal remedy knowledge among the Yucatec Maya in Tabi, Yucatan, Mexico. Free listing and cultural consensus analysis revealed that knowledge about a few medicinal plants and herbal remedies was distributed widely among the Yucatec Maya in Tabi, whereas the majority of knowledge was idiosyncratic. This finding was consistent with other studies of herbal remedy knowledge distribution among indigenous groups in Latin America and Africa. Assessing patterns in the distribution of herbal remedy knowledge is an important next step in determining the degree of dynamism or erosion in knowledge acquisition and transmission in Tabi.  相似文献

Aqueous leachates and a CHCl3-MeOH (1:1) extract of roots of Stauranthus perforatus showed a significant phytotoxic effect on Amaranthus hypochondriacus and Echinochloa crus-galli. Bioassay-directed fractionation of the active organic extract led to the isolation and characterization of ten secondary metabolites, which included two pyranocoumarins [xanthyletin (1) and 3-(1',1'-dimethylallyl)-xanthyletin (2)], four furanocoumarins [chalepensin (3), ammirin (4), chalepin (5) and 2'-isopropyl-psoralene (6)], two lignans [asarinin (7) and fargesin (8)], one sesquiterpene [4,5-epoxi-beta-caryophyllene (9)], and one alkamide [pellitorine (10)]. From these compounds, 2'-isopropyl-psoralene (6) or anhydromarmesin, is reported for the first time as a natural product, whereas compounds 4-10 are now reported as being present in S. perforatus. Metabolites 1, 3-5 and 10 caused significant inhibition of radicle growth of A. hypochondriacus and E. crus-galli. Furthermore, in a greenhouse experiment the decomposition of the leaves and roots in the soil had a significant inhibitory effect on the growth of weeds. The allelopathic action of the decomposition of roots was evident up to the sixth week of the experiment. The effect of leaves was comparable to that of DPCA (dimethyl tetrachloroterephthalate), a commercial herbicide. Finally different concentrations of Stauranthus root powder were combined with maize kernels and used to feed corn weevil. The treatments resulted in high mortality of this insect.  相似文献

Phototrophic Biofilms on Ancient Mayan Buildings in Yucatan, Mexico   总被引:6,自引:0,他引:6  
Buildings at the important archaeological sites of Uxmal and Kabah, Mexico, are being degraded by microbial biofilms. Phospholipid fatty acid (PLFA) and chlorophyll a analyses indicated that phototrophs were the major epilithic microorganisms and were more prevalent on interior walls than exterior walls. Culture and microscopical techniques showed that Xenococcus formed the major biomass on interior surfaces, but the stone-degrading genera Gloeocapsa and Synechocystis were also present in high numbers. Relatively few filamentous algae and cyanobacteria were detected. The fatty acid analysis also showed that complex biofilms colonize these buildings. Circular depressions observed by scanning electron microscopy (SEM) on stone and stucco surfaces beneath the biofilm corresponded in shape and size to coccoid cyanobacteria. SEM images also demonstrated the presence of calcareous deposits on some coccoid cells in the biofilm. Phototrophic biofilms may contribute to biodegradation by (1) providing nutrients that support growth of acid-producing fungi and bacteria and (2) active “boring” behavior, the solubilized calcium being reprecipitated as calcium carbonate. In cell culture, Rickettsia felis grows only at low temperatures (< 31 °C). Therefore, its ability to enter, survive and grow in cell lines has primarily been tested in cells derived from amphibians and arthropods, which naturally grow at low temperatures, and only infrequently in mammalian cells. We subcultured R. felis in mammalian cells for more than 10 passages using media supplemented with tryptose phosphate broth (TPB) and found that TPB is critical for optimal growth of R. felis in mammalian cells.  相似文献

In the present study, we utilized an Ixodes scapularis-derived tick cell line, ISE6, to achieve isolation and propagation of R. felis. A cytopathic effect of increased vacuolization was commonly observed in R. felis-infected cells, while lysis of host cells was not evident despite large numbers of rickettsiae. Electron microscopy identified rickettsia-like organisms in ISE6 cells, and sequence analyses of portions of the citrate synthase (gltA), 16S rRNA, Rickettsia genus-specific 17-kDa antigen, and spotted fever group-specific outer membrane protein A (ompA) genes and, notably, R. felis conjugative plasmids indicate that this cultivatable strain (LSU) was R. felis. Establishment of R. felis (LSU) in a tick-derived cell line provides an alternative and promising system for the expansion of studies investigating the interactions between R. felis and arthropod hosts.  相似文献

Rickettsia felis is an emerging etiological agent of rickettsioses worldwide. The cosmopolitan cat flea (Ctenocephalides felis) is the primary vector of R. felis, but R. felis has also been reported in other species of hematophagous arthropods including ticks and mosquitoes. Canines can serve as a bacteremic host to infect fleas under laboratory conditions, yet isolation of R. felis from the blood of a vertebrate host in nature has not been realized. Cofeeding transmission is an efficient mechanism for transmitting rickettsiae between infected and uninfected fleas; however, the mechanism of transmission among different orders and classes of arthropods is not known. The potential for R. felis transmission between infected fleas and tick (Dermacentor variabilis) and mosquito (Anopheles quadrimaculatus) hosts was examined via cofeeding bioassays. Donor cat fleas infected with R. felis transmitted the agent to naïve D. variabilis nymphs via cofeeding on a rat host. Subsequent transstadial transmission of R. felis from the engorged nymphs to the adult ticks was observed with reduced prevalence in adult ticks. Using an artificial host system, An. quadrimaculatus exposed to a R. felis-infected blood meal acquired rickettsiae and maintained infection over 12 days post-exposure (dpe). Similar to ticks, mosquitoes were able to acquire R. felis while cofeeding with infected cat fleas on rats infection persisting in the mosquito for up to 3 dpe. The results indicate R. felis-infected cat fleas can transmit rickettsiae to both ticks and mosquitoes via cofeeding on a vertebrate host, thus providing a potential avenue for the diversity of R. felis-infected arthropods in nature.  相似文献
