Summary The corynebacteria seem to be the most suitable microorganisms for cloning genes involved in the production of amino acids, nucleotides, and other products of industrial interest. A plasmid, pCCl, from Corynebacterium callunae has been found with a size of 4.3 kb. A physical map obtained with restriction endonucleases is presented. pCCl has single restriction sites for Kpn I, Sma I, Bal I, and Hind III. Copy number of this plasmid has been estimated to be about 30. A number of hybrid plasmids have been constructed between pCCl and pBR329 from Escherichia coli and transformed into corynebacteria. The thiostrepton resistance gene (tsr) from Streptomyces azureus has been inserted into them.  相似文献

The genome ofAzotobacler vinelandii has been taggedin vivo with transposons. The cells have then been allowed to divide and the pattern of segregation of the genomes has been studied. The results suggest the presence of multiple (possibly identical) copies of the genome inA. vinelandii. Only a fraction of the total number of genomes seem to have been tagged with transposon and an equilibrium between alleles of the same gene with and without the transposon was evident during random segregation.  相似文献

The Xist sequence has several characteristics that make it a potential candidate for the X-inactivation center. To investigate the role of Xist and adjacent sequences lying within the X-inactivation center candidate region, a 460-kb region surrounding the murine Xist sequence has been arrayed in lambda contigs with a combination of IRS-PCR-based hybridization and YAC fragmentation. The orientation of the Xist sequence in relation to the telomere and centromere of the X Chromosome (Chr) has been established with this contig and shown to be inverted compared to that in human.  相似文献

The body wall muscle fibers of Rinchelmis limosella and Enchytraeus albidus have been hystochemically characterized. The results have been compared with similar data on Lumbricus terrestris and Hirudo medicinalis. The various enzymatic activities have been correlated with the ultrastructural data and a possible functional meaning of the different muscle layers has been proposed.  相似文献

The discovery of a new maize (Zea mays L.) transposon system, Mutator, and the cloning of the 1.4 kilobase transposon, Mul, have made feasible the isolation of nuclear photosynthetic genes which are recognized only by their mutant phenotype. Mutant maize plants which express a high chlorophyll fluorescent (hcf) phenotype due to a defect in the electron transport or photophosphorylation apparatus have been isolated following mutagenesis with an active Mutator stock. The affected genes and their products in these mutants are inaccessible to classical methods of analysis. However, mutagenesis with the Mutator transposon makes it possible to isolate these genes.Although the PSII-deficient mutant hcf3 has been thoroughly studied by classical photo-biological methods, the nature of the lesion which results in the observed phenotype has not been established. A Mutator-induced allele of hcf3 has been isolated. A fragment of genomic DNA has been identified which is homologous to Mul and co-segregates with the mutant phenotype. This fragment is expected to contain a portion of the hcf3 locus which will be used to clone the normal gene. Direct study of the gene can provide insight into the nature and function of its polypeptide product.This approach can be used to study any photosynthetic gene which has been interrupted by a transposon. The isolation of more than 100 different chemically-induced hcf mutants, most of which can not be fully characterized using classical means, indicates the wealth of information which can be obtained using a transposon tagging technique.  相似文献

An Agrobacterium rhizogenes-mediated transformation system for Aesculus hippocastanum L. has been developed. Wounded androgenic embryos of A. hippocastanum were inoculated with bacteria containing the pRiA4 plasmid, with the uid A sequence as a reporter gene. The hairy roots emerging from the wounded sites of androgenic embryos were isolated and maintained in Murashige and Skoog's (MS) liquid hormone-free medium. Five hairy root lines have been maintained in vitro for 4 years with unchanged growth rate and might be a suitable source for secondary metabolite production. The transformation events have been confirmed by a polymerase chain reaction specific to the rol A, B, C and D genes. The absence of residual contaminating bacteria has been shown by a polymerase chain reaction specific to the vir D1 sequence.  相似文献

A new protein crosslinking agent, 2,3-dibromopropionyl-N-hydroxysuccinimide ester, has been synthesized and characterized. The potential use of this compound as a temperature-controllable heterobifunctional crosslinking agent has been investigated using model systems and its reactivity compared with that of chlorambucil-N-hydroxysuccinimide ester. The coupling of14C-labeled phenylethylamine to lysozyme has been used to illustrate the feasibility of the use of this crosslinking agent for the synthesis of immunotoxins.  相似文献

The distribution of alleles at the Lr34 locus associated with leaf rust resistance has been studied in soft winter wheat (Triticum aestivum L.) cultivars developed in Ukraine. To determine the allelic state of the Lr34 locus, codominant molecular marker cssfr5 has been used. Cultivars with the revealed Lr34(+) and Lr34(−) alleles have been identified as potentially resistant or susceptible, respectively. A collection of 81 cultivars from the main breeding centers of Ukraine has been examined; the Lr34(+) allele has been revealed in 44% of the tested cultivars. The obtained results have been compared with general data on the leaf rust resistance of wheat cultivars from different countries.  相似文献

The pattern of late labeling spots in the X chromosome ofDrosophila melanogaster has been studied by H3-thymidine autoradiography. The pattern has been found to be identical with that of the “weak spots”, or places of “ectopic pairing”. The late replicating spot in region 3C has been found to lie close to the right of the locus ofwhite. A triplication and a deficiency involving the right half of thewhite region and exhibiting changes in their interaction with the mutantzeste have been found to be associated with changes in the frequency and intensity of labeling of the late material in 3 C. Twoz + revertants derived from the triplication by X irradiation again show concomitant changes in labeling behavior at 3C.  相似文献

Summary A gene coding for an endoglucanase from the anaerobic cellulolytic bacteriumClostridium cellulolyticum has been cloned by direct selection inEscherichia coli, using the carboxymethyl cellulose-Congo Red assay. The cloned gene has been subcloned in the two possible orientations in pUC plasmids. One of the two resulting constructs exhibited a higher level of expression, which was associated with a high level of plasmid instability. The enzyme synthesized inE. coli from the cloned gene has been characterized by two procedures, maxicells and gel filtration chromatography, as a polypeptide of approximately 40 kilodaltons.  相似文献

The Euplotes cytoskeleton obtained by treatment with Triton X-100 in a microtubule-stabilizing buffer has been studied by electron microscopy and SDS-polyacrylamide electrophoresis. An antiserum has been prepared using the tubulin band from preparative gels of Euplotes cytoskeletons as an antigen. This antiserum reacts with the tubulin from different ciliated protozoa but fails to recognize the vertebrate tubulin by immunoblotting. Immunoblotting studies have demonstrated a slower electrophoretic mobility for the α-tubulin of Euplotes and Oxytricha than that of Paramecium and Tetrahymena. A cytoplasmic microtubular network in Euplotes has been revealed by indirect immunoftuorescence using both an anti-α-tubulin monoclonal antibody directed against chick brain tubulin and an antiserum raised against the tubulin of Euplotes.  相似文献

The fine structure of the alga Glaucocystis nostochinearum Itzigs. has been studied by thinsectioning and freeze-etching. Many of the features already established by sectioning have been verified but different conclusions are drawn, particularly with regard to the photosynthetic organs. The action of glycerol solutions on the morphology of the plasmalemma as revealed by freeze-etching is shown to be of general importance for the freeze-etching of plant cells and of particular importance in elucidating the true form of the plasmalemma in Glaucocystis. The literature regarding the taxonomic position of Glaucocystis has been reviewed, and, along with a completely new demonstration of plasmalemma structure, an attempt has been made to reclassify this organism.  相似文献

The complete nucleotide sequence of Citrus limon 26S rDNA has been determined. The sequence has been aligned with large ribosomal RNA (L-rRNA) sequences of Escherichia coli, Saccharomyces cerevisiae and Oryza sativa. Nine extensive expansion segments in dicot 26S rRNA relative to E. coli 23S rRNA have been identified and compared with analogous segments of monocot, yeast, amphibian and human L-rRNAs. A secondary structure model for lemon 26S rRNA has been derived based on the refined model of E. coli 23S rRNA. It has been compared with other eukaryotic L-rRNAs models in terms of location of functionally important regions. Origin and evolution of L-rRNA expansion segments are discussed.  相似文献

The marine planktonic diatom Coenobiodiscus muriformis has been reinvestigated using light microscopy and scanning and transmission electron microscopy; as a result it is proposed, to transfer the species to the genus Planktoniella. The structure and formation of colonies are discussed and compared with data from an earlier study. Further details of valve and girdle baud structure have been ascertained and the exact form of the central and peripheral tubes has been described. The relationship of the present taxon to Thalassiosira and Coscinodiscus is discussed.  相似文献

The growth of a mixed methane-utilizing culture in a continuous flow fermenter has been studied under both methane and oxygen limitation. Small additions of methanol have been shown to inhibit the methane-utilizing moiety in the culture and it has been shown that the Hyphomicrobium sp. in the mixed culture removes any inhibitory methanol. The interaction between the methane-utilizing Pseudomonas sp., and the Hyphomicrobium sp. has been explained and a model of the continuous mixed culture under oxygen limitation has been formulated. Qualitative predictions of transient phenomena by the model have been verified experimentally.  相似文献

The closed circular DNA of pea chloroplast has been digested with restriction endonucleases SalI, SmaI, BamHI, XbaI, XhoI, HindIII, and EcoRI. A physical restriction map of pea ctDNA has been constructed by mapping the SalI and SmaI sites. The pea ctDNA has been found to contain one set of ribosomal RNA genes by Southern hybridization of restriction endonuclease digest, R-loop studies, and DNA-DNA heteroduplex mapping. The 23 S and 16 S RNA genes are confined to a DNA region of 3.0 and 1.5 kbp, respectively. The two rRNA chains are separated by a spacer region of 2.2 kbp.  相似文献

Summary The red cell glutamic-pyruvic transaminase phenotype has been determined in 294 individuals from the region of the Po delta (Ferrara, Northern Italy). No correlation with past malarial morbidity has been detected. The gene frequencies found in this survey are similar to those reported for other Caucasian populations. Summary The gene rpoA, encoding a protein homologous to the alpha subunit of RNA polymerase from Escherichia coli has been located in pea chloroplast DNA downstream of the petD gene for subunit IV of the cytochrome b-f complex. Nucleotide sequence analysis has revealed that rpoA encodes a polypeptide of 334 amino acid residues with a molecular weight of 38916. Northern blot analysis has shown that rpoA is co-transcribed with the gene for ribosomal protein S11. A lacZ-rpoA gene-fusion has been constructed and expressed in E. coli. Antibodies raised against the fusion protein have been employed to demonstrate the synthesis of the rpoA gene product in isolated pea chloroplasts. Western blot analysis using these antibodies and antibodies against the RNA polymerase core enzyme from the cyanobacterium, Anabaena 7120, has revealed the presence of the gene product in a crude RNA polymerase preparation from pea chloroplasts.  相似文献

The minimal mono-heme ferricytochrome c from Bacillus pasteurii, containing 71 amino acids, has been further investigated through mutagenesis of different positions in the loop containing the iron ligand Met71. These mutations have been designed to sample different aspects of the loop structure, in order to obtain insights into the determinants of the stability of the iron(III) environment. In particular, positions 68, 72 and 75 have been essayed. Gln68 has been mutated to Lys to provide a suitable alternate ligand that can displace Met71 under denaturing conditions. Pro72 has been mutated to Gly and Ala to modify the range of allowed backbone conformations. Ile75, which is in van der Waals contact with Met71 and partly shields a long-lived water molecule in a protein cavity, has been substituted by Val and Ala to affect the network of inter-residue interactions around the metal site. The different contributions of the above amino acids to protein parameters such as structure, redox potential and the overall stability against unfolding with guanidinium hydrochloride are analyzed. While the structure remains essentially the same, the stability decreases with mutations.
