The callus cultures obtained from unpollinated young ovaries of willow leaf tobacco were transplanted separately on subculture medium (M-l) and differentiation medium (M-2). They were called M-1 callus and M-2 callus respectively. Only meristematic cells and parenchymas could be observed in M-1 callus. The M-2 callus, however, was able to differentiation into vascular systems, embryoids and bud primordia. The respiration of M-1 callus exhibited CN-insensitive. The relative contributions of alternative pathway and eytochrome pathway to the total respiration were 29–38% and 44–51% respectively during the culture period of 25 days. It was obvious that the bulk of the respiratory electron flux was mediated by the eytochrome pathway. The greater part of the respiration in M-2 callus was insensitive to cyanide. The relative contributions of alternative pathway and cytochrome pathway to the total respiration were 41–47% and 29–32% respectively. These results suggested that the increase in the participation of the alternative pathway in total respiration was probablely related to the differentiation of the callus culture.  相似文献

根据呼吸抑制剂试验和氧肟酸滴定法测定结果表明,甘肃黄花烟草愈伤组织呼吸中有明显的抗氰交替途径运行,平均占总呼吸的31%;但仍以细胞色素途径为主,平均占总呼吸的46%;还有23%不受 KCN 加 m-CLAM 抑制的未知剩余呼吸。改变培养基的激素成分和浓度,在不引起愈伤组织发生明显分化条件下,愈伤组织的生长和呼吸速率虽有不同,但抗氰交替途径和细胞色素途径对总呼吸的相对贡献程度和二者的变化趋势基本一致。  相似文献

In attempting to examine whether CN-resistant respiratory pathway is present in callus culture, we used tobacco callus cultures grown on different media. The M-1 medium contained tbe mineral and organic elements of MS medium and was supple,nented with 6-BA (0.5 mg/l) and 2,4-D (2 mg/l), and M-2 medium with 6-BA (2mg/l) and IAi (1 mg/l). No differentiation was observed in both of them. The respiration of M-1 callus was partly resistant to CN, and was markedly inhlbited by m-CLAM in the presence or absence of CN. Experiments of m-CLAM titration showed that the averages of relative contribution of alternative and cytochrome pathway in M-1 callus were 31% awl 46%of the total respiration respectively during the euliure period of 25 days. A same experiment was made on the M-2 callus. It was found that the pereeutages of relative contributions of the two electron transport pathways to the total respiration were approximately the same as those of the M-1 callus, although the respiratory rate was higher in M-2 callus. The above results showed that the bulk of respiratory electron flux was mediated by the eytoehrome pathway, although the alternative pathway was operative in callus of tobacco. The change of exogenous hormones added in the medium could not produee significant effects on the degree of relative contribution of two electron transport pathways under non-differentiation conditions.  相似文献

The effects of KCN (0.5mmol/L) and NaN3 (0.01 mmol/L) pretreatment on the operation of the alternative pathway in subcultured tobacco (Nicotiana rustica L. cv. Gansu yellow flower) callus were analyzed. After treatment with KCN and NaN3 for 12 h, the total respiration rate (Vt) decreased by 12% and 17%, whereas oxygen consuption by the cytochrome pathway decreased by 22% and 28% respectively. The capacity of the alternative pathway (Valt) remained constant, while the activity of the alternative pathway (ρ· Valt ) inreased slightly. This changing pattern led to a declined contribution of the cytochrome pathway to the total respiration rate and an increased activity of the alternative pathway. Treatment with KCN for 24 h brought about a slight rise of oxygen consumption by the cytochrome pathway as compared with that in callus treated for 12 h, but the oxygen consumption was still lower than that in the untreated callus. Treatment with NaN3 for 24 h resulted in a profound decrease of the cytochrome pathway operation and a continuing increase of the alternative pathway operation. These data indicated that the enhanced operation of the alternative pathway played a compensatory role to the total respiration when the cytochrome pathway was partially inhibited in tobacco callus.  相似文献

The increasement of the operation (ρValt) and the capacity of the alternative pathway (Valt) in tobacco (Nicotiana rustica L. cv. Gansu Yellow Flower) callus that treated by low temperature (4 ℃) were observed. The two electron transport pathways were changed alternatively, the cyanide-resistant respiration was the main respiratory pathway from the 2nd day to the 5th day, while the cytochrome pathway was the main pathway consistently in control callus. Cycloheximide and chloramphenicol treatment which inhibited protein synthesis resulted in dramatic and partly inhibition of Valt induced by low temperature respectively. This further proved above result that low temperature induced Valt. The same result was found when two inhibitors were used together. The analysis of heat production of callus indicated that the operation of the alternative pathway could be regulated to improve the micro-environmental temperature of tobacco callus to adapt to low temperature. The possible participation of the alternative pathway in the response to low temperature and its physiological roles were discussed.  相似文献

In suspension cells of NT1 tobacco (Nicotiana tabacum L. cv bright yellow), inhibition of the cytochrome pathway of respiration with antimycin A induced a large increase in the capacity of the alternative pathway over a period of approximately 12 h, as confirmed in both whole cells and isolated mitochondria. The increase in alternative pathway capacity required de novo RNA and protein synthesis and correlated closely with the increase of a 35-kD alternative oxidase protein. When the cytochrome pathway of intact cells was inhibited by antimycin A, respiration proceeded exclusively through the alternative pathway, reached rates significantly higher than before antimycin A addition, and was not stimulated by p-trifluoromethoxycarbonylcyanide (FCCP). When inhibition of the cytochrome pathway was relieved, alternative pathway capacity and the level of the 35-kD alternative oxidase protein declined. Respiration rate also declined and could once again be stimulated by FCCP. These observations show that the capacities of the mitochondrial electron transport pathways can be regulated in a coordinate fashion.  相似文献

Potato tuber ( Solanum tuberosum L. cv. Bintje) callus shows a decrease in fresh weight and an increase in dry weight upon transfer to nutrient medium supplemented with 0.3 or 0.5 M mannitol. The osmolarity of the intracellular fluid increases simultaneously. Probably mannitol is taken up from the medium till the osmolarity of the tissue is in equilibrium with that of the medium. After osmotic adaptation, on a medium with 0.5 M mannitol, growth is negligible, although the tissue retains its viability.
Respiration increases upon transfer to medium with extra mannitol, especially when expressed on a fresh weight basis. On this basis cytochrome and alternative pathway capacities do not change appreciably. The respiratory increase is exclusively caused by an increased engagement of the alternative pathway. The participation of this pathway in uninhibited respiration increases from about 10 to 90% upon transfer to medium with extra mannitol. The increase in respiration is partly correlated with the decrease in fresh weight upon transfer. Per disc, the capacities of the cytochrome and alternative pathway decline. Yet, total respiration per disc significantly increases due to the increased participation of the alternative pathway. This results in an almost equal ATP-production per disc before and after transfer. We suggest, that the alternative pathway functions as a reserve capacity in potato callus, which is switched on when ATP-production coupled to the cytochrome pathway is impaired.  相似文献

The uninhibited respiration of mitochondria, isolated from potato tuber discs (Solanum tuberosum L. cv. Bintje) incubated on a callus-inducing medium at 28°C, is higher than that of mitochondria from tissue incubated at 8°C. This respiration is composed of a CN-sensitive and a CN-resistant part. The capacity of the CN-resistant alternative oxidase pathway is larger in mitochondria from 28°C tissue than in mitochondria from 8°C tissue (35% and 8% of uninhibited respiration, respectively). The alternative pathway is operative both in mitochondria from 28°C tissue and 8°C tissue.

The observed difference in uninhibited respiration, is not only caused by lower values of respiration via the alternative pathway in mitochondria from 8°C tissue, but also by lower values of respiration via the cytochrome pathway.

A positive correlation has been demonstrated between the incubation temperature (ranging from 4-37°C) and the relative capacity of respiration via alternative pathway in the mitochondria. Induction of alternative pathway is not directly correlated with growth (in terms of increase in fresh weight) of the potato tuber discs.

Cyanide-resistant, alternative respiration in Nicotiana tabacum L. cv Xanthi-nc was analyzed in liquid suspension cultures using O2 uptake and calorimetric measurements. In young cultures (4-8 d after transfer), cyanide inhibited O2 uptake by up to 40% as compared to controls. Application of 20 μm salicylic acid (SA) to young cells increased cyanide-resistant O2 uptake within 2 h. Development of KCN resistance did not affect total O2 uptake, but was accompanied by a 60% increase in the rate of heat evolution from cells as measured by calorimetry. This stimulation of heat evolution by SA was not significantly affected by 1 mm cyanide, but was reduced by 10 mm salicylhydroxamic acid (SHAM), an inhibitor of cyanide-resistant respiration. Treatment of SA-induced or uninduced cells with a combination of cyanide and SHAM blocked most of the O2 consumption and heat evolution. Fifty percent of the applied SA was taken up within 10 min, with most of the intracellular SA metabolized in 2 h. 2,6-Dihydroxybenzoic and 4-hydroxybenzoic acids also induced cyanide-resistant respiration. These data indicate that in tobacco cell-suspension culture, SA induces the activity and the capacity of cyanide-resistant respiration without affecting the capacity of the cytochrome c respiration pathway.  相似文献

The growth (fresh and dry weight increase) of potato tuber ( Solanum tuberosum L. cv. Bintje) callus discs was stimulated by incubation in air with 500 ppm 2,5-norbornadiene (NBD, a competitive inhibitor of ethylene action) and inhibited by incubation in air with 4 000 ppm NBD. Ethylene formation by the callus was stimulated by NBD. The development of the alternative pathway, measured in isolated mitochondria was inhibited by NBD in a concentration-dependent way. The alternative pathway capacity, measured in vivo, was inhibited by 4 000 ppm NBD, but not by 500 ppm. Uninhibited in vivo respiration, which consists of cytochrome path activity and alternative path activity, was stimulated by the treatment with 500 ppm NBD. The main contribution to this stimulation was made by the cytochrome pathway. In 4 000 ppm NBD-treated callus, uninhibited respiration seemed to be unaffected as a consequence of an inhibited cytochrome path activity, which was compensated by a stimulated alternative path activity. Both in 500 and 4 OIK) ppm NBD-treated callus the alternative path activity in vivo was stimulated.
The regulatory role for endogenous ethylene in potato tuber callus is discussed in relation to: 1) The induction of respiratory pathways, 2) the supply of reduction equivalents in vivo and 3) growth.  相似文献

以油桃品种“曙光”为试材,采用呼吸抑制剂法研究了短日照处理下花芽在休眠诱导期两条电子传递途径的发生和运行情况.结果表明:花芽总呼吸速率(Vt)和细胞色素电子传递途径呼吸速率(ρ'Vcyt)均呈双峰曲线变化,短日照可同步诱导两者的一次峰前移、二次峰后延,抑制ρ’Vcyt,但对Vt无显著影响.交替途径容量(Valt)和实际运行活性(ρValt)亦呈双峰曲线,两者基本同步变化,短日照可以显著诱导Valt和ρValt的前期高峰期提前,提高Valt和ρValt,对后期高峰期无明显作用.细胞色素电子传递途径呼吸速率下降和交替途径呼吸速率上升是油桃花芽休眠诱导期的重要特点.从两条电子传递途径的呼吸速率对总呼吸速率的贡献率来看,细胞色素电子传递途径仍是主要电子传递途径,交替途径起辅助与分流作用.  相似文献

During incubation of potato tuber discs ( Solanum tuberosum L. cv. Bintje) on a callus-inducing medium at 28°C, a high capacity of in vivo alternative pathway respiration develops (75% of uninhibited respiration is azide-resistant). When callus induction takes place at 8°C, only 45% of respiration is resistant to azide. In the lag phase of growth the activity of alternative pathway is low. during the exponential growth phase the activity reaches its maximal rate. This in vivo activity is of the same size at both culture temperatures. As a consequence a greater part of alternative pathway capacity is operating in uninhibited respiration during growth at low temperatures.  相似文献

Developing Nicotiana tabacum L. cv. Wisconsin-38 callus grown on modified Murashige-Skoog (MS) medium with Kao organic acids (pyruvic, citric, malic and fumaric acids) contains abnormally high levels of nornicotine and total alkaloids when compared with the leaves of the donor plant. Nornicotine/nicotine ratios observed during callus development suggest that nicotine is converted into nornicotine in the callus, with subsequent movement of alkaloids into roots formed on the callus and into the agar medium. Addition of Kao organic acids to the medium increases alkaloid levels, but cannot account for the abnormal increase in nicotine demethylation. This study thus reports two new findings: (a) that the total alkaloid content of tobacco callus can be greatly enhanced to 3.75% on a dry weight basis by exogenous organic acids, and (b) that endogenous nornicotine can accumulate in tobacco tissue cultures.  相似文献

The addition of potassium bicarbonate to the electrode cuvette immediately stimulated the rate of dark O2 uptake of photomixotrophic and heterotrophic carnation (Dianthus caryophyllus L.) callus, of Elodea canadensis (Michx) leaves, and of other plant tissues. This phenomenon occurred at pH values lower than 7.2 to 7.8, and the stimulation depended on the concentration of gaseous CO2 in the solution. These stimulatory responses lasted several minutes and then decreased, but additional bicarbonate or gaseous CO2 again stimulated respiration, suggesting a reversible effect. Carbonic anhydrase in the solution increased the stimulatory effect of potassium bicarbonate. The CO2/bicarbonate dependent stimulation of respiration did not occur in animal tissues such as rat diaphragm and isolated hepatocytes, and was inhibited by salicylhydroxamic acid in carnation callus cells and E. canadensis leaves. This suggested that the alternative oxidase was engaged during the stimulation in plant tissues. The cytochrome pathway was severely inhibited by CO2/bicarbonate either in the absence or in the presence of the uncoupler carbonylcyanide m-chlorophenyl hydrazone. The activity of cytochrome c oxidase of callus tissue homogenates was also inhibited by CO2/bicarbonate. The results suggested that high carbon dioxide levels (mainly free CO2) partially inhibited the cytochrome pathway (apparently at the oxidase level), and this block in electron transport elicited a large transient engagement of the alternative oxidase when present uninhibited.  相似文献

Suspension cells of NT1 tobacco (Nicotiana tabacum L. cv bright yellow) have been used to study the effect of growth temperature on the CN-resistant, salicylhydroxamic acid-sensitive alternative pathway of respiration. Mitochondria isolated from cells maintained at 30°C had a low capacity to oxidize succinate via the alternative pathway, whereas mitochondria isolated from cells 24 h after transfer to 18°C displayed, on average, a 5-fold increase in this capacity (from 7 to 32 nanoatoms oxygen per milligram protein per minute). This represented an increase in alternative pathway capacity from 18 to 45% of the total capacity of electron transport. This increased capacity was lost upon transfer of cells back to 30°C. A monoclonal antibody to the terminal oxidase of the alternative pathway (the alternative oxidase) from Sauromatum guttatum (T.E. Elthon, R.L. Nickels, L. McIntosh [1989] Plant Physiology 89: 1311-1317) recognized a 35-kilodalton mitochondrial protein in tobacco. There was an excellent correlation between the capacity of the alternative path in isolated tobacco mitochondria and the levels of this 35-kilodalton alternative oxidase protein. Cycloheximide could inhibit both the increased level of the 35-kilodalton alternative oxidase protein and the increased alternative pathway capacity normally seen upon transfer to 18°C. We conclude that transfer of tobacco cells to the lower temperature increases the capacity of the alternative pathway due, at least in part, to de novo synthesis of the 35-kilodalton alternative oxidase protein.  相似文献

Inhibitor titration curves and discrimination against 18O2 by mitochondrial respiration in three strains of green algae (Selenastrum minutum [Naeg.] Collins, and two strains of Chlamydomonas reinhardtii Dangeard) with differing respiratory capabilities were determined. Discrimination for cytochrome pathway respiration ranged from 19.89 to 20.43%. Discrimination for alternative pathway respiration by wild-type C. reinhardtii (measured in the presence of KCN) was 25.46%, while discrimination values for a cytochrome oxidase deficient mutant of C. reinhardtii ranged from 24.24 to 24.96%. In the absence of KCN, the alternative pathway was not engaged in wild-type C. reinhardtii, the only algal strain that possessed both cytochrome and alternative pathway capacities.  相似文献

The contribution of the alternative pathway in root respiration of Pisum sativum L. cv Rondo, Plantago lanceolata L., and Plantago major L. ssp major was determined by titration with salicylhydroxamate (SHAM) in the absence and presence of cyanide. SHAM completely inhibited the cyanide-resistant component of root respiration at 5 to 10 millimolar with an apparent Ki of 600 micromolar. In contrast, SHAM enhanced pea root respiration by 30% at most, at concentrations below 15 millimolar. An unknown oxidase appeared to be responsible for this stimulation. Its maximum activity in the presence of low SHAM concentrations (1-5 millimolar) was 40% of control respiration rate in pea roots, since 25 millimolar SHAM resulted in 10% inhibition. In plantain roots, the maximum activity was found to be 15%. This hydroxamate-activated oxidase was distinct from the cytochrome path by its resistance to antimycin. The results of titrations with cyanide and antimycin indicated that high SHAM concentrations (up to 25 millimolar) block the hydroxamate-activated oxidase, but do not affect the cytochrome path and, therefore, are a reliable tool for estimating the activity of the alternative path in vivo. A considerable fraction of root respiration was mediated by the alternative path in plantain (45%) and pea (15%), in the latter because of the saturation of the cytochrome path.  相似文献

The growth of callus tissue cultures and the infectivity of twenty fiveSolanum laciniatum Ait. plants and of sixteenNicotiana tabacum L. cv. Samsun plants were investigated. The plants were obtained from callus tissue cultures derived from stem pieces of the respective plants infected with a mycoplasma-like organism (MLO) evoking potato witches’ broom. The tissues were cultivated on synthetic nutrient medium with kinetin and IAA. Allde novo obtainedS. laciniatum plants were healthy. On the contrary twelve of the sixteen reconstituted tobacco plants showed MLO presence. Summarizing these and previous results, the authors suppose that the most important factor influencing MLO persistence in callus tissues cultivated on the applied nutrient medium may be the callus growth rate and the organogenesis set. Both these conditions are determined by the metabolism of the investigated plant species.  相似文献
