Effects of lipid infusion into postpartum (PP) beef heifers on plasma concentrations of linoleic acid and prostaglandin (PG) F(2alpha) metabolite (PGFM), days to first estrus, and subsequent pregnancy rate were examined. Treatments (n = 5 per group) of 1 L intralipid (20% soybean oil; IL), 1 L 50% dextrose (DEXT; isocaloric to IL), 0.5 L intralipid (0.5 IL), and 1 L physiological saline (SAL) were infused i.v. over 4 h on each of Days 7 through 11 PP. Capacity of the uterus to produce PG was evaluated after i.v. injection of 150 IU of oxytocin (OT) to IL- and DEXT-treated heifers Day 12 PP. Change in plasma concentrations of PGFM from 0 to 4 h was greater for IL-treated heifers than for heifers given other treatments on Day 7 (P = 0.04) and on Day 11 (P = 0.01), but not on Day 9 (P>0.10). Plasma linoleic acid on Day 11 and OT-induced release of PGFM on Day 12 were greater in IL-treated heifers compared with DEXT-treated heifers (P<0.06 and P = 0.01, respectively). There were no significant differences among treatments for mean days to first estrus or pregnancy rate. Infusion of lipid increased systemic concentrations of linoleic acid and increased the capacity of PP heifers to produce uterine PGF(2alpha) as indicated by plasma PGFM concentration after OT injection.  相似文献

Plasma prolactin and F-prostaglandins (PGF) were measured anesthetized male Sprague-Dawley rats before and at 15, 30, 45 and 60 minutes following i.v. injection of either PGF2alpha (4 mg/kg), chlorpromazine, 1 mg/kg or chlorpormazine (1 mg/kg) after pretreatment with i.p. indomethacin (2 mg/kg). Following PGF2alpha administration, plasma prolactin levels increased significantly only at 15 and 30 minutes in spite of extremely high PGF levels throughout 60 minutes. Besides the expected rise in plasma prolactin, chlorpromazine caused a transient but statistically significant increase in PGF. Indomethacin blocked the chlorpormazine-induced PGF rise but not prolactin increase. Animals stressed with ether anesthesia showed elevation of plasma prolactin, which was not blocked by indomethacin although PGF concentration fell. Theese results indicate that PGF2alpha can stimulate prolactin release. This effect does not appear to be physiologic since very high PGF levels are required. Furthermore, blockade of prostaglandin synthesis by indomethacin does not prevent the release of prolactin in response to chlorpormazine or stress. Our findings do not support a possible role of PGFs as intermediaries in prolactin release. However, it is possible that PGFs may work through other mechanisms not investigated in our study.  相似文献

Peripheral plasma levels of the main blood plasma metabolite of PGF(2alpha) (15-keto-13,14-dihydro-PGF(2alpha)) and progesterone were investigated during the immediate, post-partum period in 59 normally calving cows. Uterine involution was monitored by weekly rectal palpations. The levels of the prostaglandin metabolite were high at parturition and remained thereafter elevated for periods varying up to 7-23 days. Uterine involution was completed during periods ranging from 16-53 days. According to the clinical findings, the animals were divided into three groups. Group A comprises 46 animals which had an uncomplicated, puerperal period. A significant (p<0.001) correlation between the duration of elevated prostaglandin levels and the time for completed uterine involution (Y=29.6 - 1.3 (X - 13.5)) was found for these animals. Group B animals (n=8) had periods of varying length with uterine discharge during the first 30 days post-partum. When compared to group A animals, the animals in group B had comparatively longer periods of prostaglandin release and also longer periods for completion of uterine involution. Group C animals (n=5) at times had palpable, thin-walled, cystlike structures in the ovaries during the first 30 days post-partum. In this group of animals, the periods of high prostaglandin levels, as well as for the completion of uterine involution, were similar to those for the animals in group A. Progesterone levels remained low during the immediate post-partum period and in no case were elevated levels found until the prostaglandin release had ceased.  相似文献

Eighty Holstein dairy cows were treated with 25 mg of prostaglandin F2alpha (PGF2alpha) on Days 14 to 16 post calving. Eighty-four herd mates served as saline controls in the double blind study. The reproductive parameters used to measure fertility were mean days to first service on all cows and mean days open, first service conception rates and services per pregnancy on cows that became pregnant. Mean days to first service was similar in both groups (71.8 +/- 27, treated; 68.5 +/- 28.6, controls; P = 0.5352). The mean days open was 98.6 +/- 52.0 d for treated cows compared to 118.8 +/- 71.2 d for controls (P = 0.0680). First service pregnancy rates (treated, 41.3%; control, 35.7%) were not significantly different (P = 0.0630); however, the mean services per pregnancy (treated, 1.64; control, 2.33) were significantly (P = 0.0021) improved in the treated group. Ten cows in the treated group and twelve cows in the control group were diagnosed as having retained fetal membranes and/or metritis. For treated and control cows mean days to first service were 82.2 +/- 34.8 d and 82.8 +/- 58.7 d (P = 0.5652). Days open were 97.0 +/- 32.5 d and 133.4 +/-58.4 d (P = 0.3636); services per conception were 1.83 and 2.50 (P = 0.3178), respectively. In all treated cows reproductive parameters were similar whether cows had high serum progesterone ( > 1 ng/ml) or low serum progesterone ( < 1 ng/ml) on the treatment day. This study suggests that early postpartum treatment of lactating dairy cows with prostaglandin produces an improvement in fertility.  相似文献

The objective of the present study was to determine whether treatment of postpartum multiparous and primiparous anestrous beef cows with an intravaginal progesterone-releasing insert (CIDR) and PGF(2alpha), with and without the addition of GnRH or estradiol cypionate (ECP) at the time of CIDR insertion, is effective in stimulating onset of estrous cycles. Postpartum lactating Angus primiparous (n=47, 2 years of age, 495+/-6 kg) and multiparous (n=76, >or=3 years of age, 553+/-9 kg) cows were assigned by calving date to four blocks spaced 21-day apart. Cows were assigned sequentially by calving date to four treatment groups: (1) PGF(2alpha) (n=30), (2) CIDR-PGF(2alpha) (n=30), (3) GnRH-CIDR-PGF(2alpha) (n=33), and (4) ECP-CIDR-PGF(2alpha) (n=27). Intravaginal CIDR inserts were in place from days -7 to 0. A single 100 microg injection of GnRH or 2 mg ECP were administered on day -7, and 25mg PGF(2alpha) was administered on day 0. Day 0 averaged 38+/-1 day postpartum. Blood samples were collected on days -19, -9, 0, 5, 9, 12, 16, 19, 23, 26, and 30 for determination of plasma progesterone concentrations. Pre-treatment luteal activity (progesterone>or=1 ng/ml) was detected in 19% of primiparous and 8% of multiparous cows. Progesterone concentrations on day 0 were greater (P<0.001) in primiparous (3.2+/-0.3 ng/ml) than multiparous (2.0+/-0.2 ng/ml) cows. Following CIDR withdrawal, progesterone concentrations from days 5 to 30 were used to categorize response profiles as either: (1) treatment-induced onset of estrous cycles, (2) continued anestrus, or (3) spontaneous ovulation and subsequent formation of a CL. Incidence of treatment-induced onset of estrous cycles, which was defined as progesterone concentrations >or=1 ng/ml in three or more consecutive samples from days 9 to 19, was influenced by treatment and parity. Percentages of cows initiating estrous cycles were greater (P<0.001) in the three CIDR-treated groups than in the PGF(2alpha) group (55 and 8%, respectively). Percentages of cows initiating estrous cycles in the CIDR-PGF(2alpha), GnRH-CIDR-PGF(2alpha), and ECP-CIDR-PGF(2alpha) groups were 55, 58, and 52%, respectively. Incidence of treatment-induced estrous cycles in the three CIDR-treated groups of cows was greater (P=0.008) in primiparous (76%) than multiparous (43%) cows. Treatment of postpartum anestrous primiparous and multiparous beef cows with CIDR-PGF(2alpha) approximately 40-day postpartum provides an approach to increase the percentage of cows that have reinitiated estrous cycles by the start of the breeding season.  相似文献

Prostaglandin (PG) E(2) promotes osteoclastic cell differentiation, but the physiological function of PGF(2alpha) remains unclear. We examined the physiological effects of PGF(2alpha) on osteoclast differentiation using a murine cell line, RAW264, and the column-purified murine bone marrow cells, both of which are differentiable into osteoclast-like multi-nuclear cells. Although PGF(2alpha) did not affect the number of differentiated osteoclasts, PGF(2alpha) reduced the bone resorption activity of osteoclasts developed from both cell types in a pit formation assay. Thus, PGF(2alpha) inhibits bone resorption without affecting the number of osteoclasts, providing a novel molecular mechanism underlying bone metabolism.  相似文献

The persistence of covalently closed circular (ccc) DNA of Hepatitis B virus (HBV) in liver cells is believed to be the major reason for relapse after completion of HBV antiviral therapy. Up to now, there is no sensitive method to quantify cccDNA in infected liver cells. We designed a set of primers to specifically amplify DNA fragments from HBV cccDNA but not from viral genomic DNA. A good linear range was obtained when 100-10(7) copies of HBV cccDNA were used as template in the quantitative real-time PCR. Not only is this method rapid, economical, highly sensitive, it can be used to monitor HBV cccDNA in infected human liver biopsies and to guide patients undergoing long-term anti-HBV therapy.  相似文献

Seventy six apparently anoestrous buffaloes, with a palpable corpus luteum at day 43 to 547 post partum, were treated intramuscularly with Prostaglandin, using either 25 mg single dose (Group I), 25 mg double dose 11 days apart (Group II), or 50 mg single dose (Group III). Animals exhibiting oestrus, without any treatment, during the period of experimentation served as control (Group IV). The overall conception rate for the treated animals was 28.8, 51,5, and 69.7 percents after one, two, and three inseminations, respectively. The conception rate, at induced as well as subsequent oestrus, was comparable to control animals. The intergroup differences among treated animals were not significant. The conception rate was greater when the induced heat was more intensely expressed. The subsequent mean oestrus cycle length was similar to controls when all of the treated animals were considered together.  相似文献

A study was performed to examine the release patterns of prolactin and LH of young beef cows with one (single calf) or two calves (double calf) throughout the postpartum interval. The effect on prolactin release of intramuscular and intra-carotid administration of lergotrile and intra-carotid administration of L-dopa was also examined. In approximately 50% and 65% of the cases, no prolactin release could be detected after the beginning of or during the suckling stimulus in cows with one or two calves respectively. LH plasma concentrations remained constant throughout the experiment in all animals. The chosen intramuscular lergotrile treatment lowered plasma prolactin concentrations to baseline levels but had no effect on the length of the postpartum interval. No effect on prolactin release was observed by the given intra-carotid treatments of both lergotrile and L-dopa. First postpartum estrus was observed on days 67 and 88 in the single and double calf cows respectively. The number of suckling periods did not change during the postpartum period but their duration decreased during the same period. These results demonstrate that in at least half of the cases the suckling stimulus does not cause a release of prolactin from the pituitary in the young beef cow. Also, the inhibitory effect of suckling on the resumption of ovarian cyclic function postpartum appears to be of a quantitative nature and mediated by a factor other than prolactin.  相似文献

Two trials were conducted to evaluate treatments combining progesterone pretreatment and prostaglandin F(2alpha) (PGF(2alpha)) on estrus response, pregnancy and calving rate in heifers. Treatments in Trial 1 were 1) control (T(1); n=59), 2) 25 mg PGF(2alpha) on Day 0 (T(2); n=58), 3) 150 mg progesterone (P(4), i.m.) in corn oil on Day -24 plus PGF(2alpha) (T(3); n=61), and 4) 150 mg P(4) on Day -5 plus PGF(2alpha) (T(4); n=59). Trial 2 had T(2) and T(4) only. Heifers were artificially inseminated 8 to 16 h after detection of estrus for 10 and 5 d in Trials 1 and 2, respectively. In Trial 1 more heifers in T(3) and T(4) showed estrus by 72 h compared to T(1) and T(2). In T(3), percentages were greater at 84 and 96 h than in T(1) and T(2). There were no differences between T(3) and T(4) or T(1) and T(2) over time. Cumulative distributions of responses showed that more heifers in T(3) and T(4) were in estrus by 84 h after PGF(2alpha) than after other treatments, while T(3) showed the greatest total number of heifers in estrus by 84 h; this difference persisted for 180 h. In Trial 2, percentages of heifers observed in estrus for T(1) and T(4) were not different. Average interval from PGF(2alpha) to estrus was shorter in Trial 1 for T(3) heifers compared to other treatments. No difference was observed in interval to estrus for T(2) and T(4) in Trial 2; this interval averaged 58 h. Artificial insemination pregnancy rates were not different among treatments in either trial and averaged 67.4%. In Trial 1, a greater proportion of heifers in T(2), T(3) and T(4) calved by 35 days into the calving season compared to T(1), but in Trial 2 calving rates for T(2) and T(4) were not different. Progesterone pretreatment combined with PGF(2alpha) appeared to enhance estrus synchronization without influencing either pregnancy or calving rates.  相似文献

Preliminary studies indicate the presence of PGF2alpha specific binding sites in membrane fractions prepared from equine corpora lutea. The equilibrium binding data indicate an apparent dissociation constant of 3.2 X 10(-9)M and the concentration of binding sites of -0.1 pmoles/mg membrane protein. Competition of several natural prostaglandins for equine luteal PGF2alpha specific binding sites indicates specificity for the 9alpha-hydroxyl moiety and the 5,6-cis doublebond. Significant increases in relative binding affinities were demonstrated for PGF2alpha analogs with a phenyl ring introduced at carbons 16 or 17. Specific PGF2alpha binding was demonstrated in corpora lutea collected at known stages of the estrous cycle. There was no pattern in these values based on the stage of the cycle. While specific 3H-PGE1 binding could be demonstrated, no high affinity sites could be quantitated. 3H-PGE1 binding appeared unaffected by changes in temperature or time of incubation, whereas PGF2alpha specific binding was significantly modified by both these factors.  相似文献

To examine possible mechanisms involved in resistance of the ovine corpus luteum to the luteolytic activity of prostaglandin (PG)F(2alpha), the enzymatic activity of 15-hydroxyprostaglandin dehydrogenase (PGDH) and the quantity of mRNA encoding PGDH and cyclooxygenase (COX-2) were determined in ovine corpora lutea on Days 4 and 13 of the estrous cycle and Day 13 of pregnancy. The corpus luteum is resistant to the action of PGF(2alpha) on Days 4 of the estrous cycle and 13 of pregnancy while on Day 13 of the estrous cycle the corpus luteum is sensitive to the actions PGF(2alpha). Enzymatic activity of PGDH, measured by rate of conversion of PGF(2alpha) to PGFM, was greater in corpora lutea on Day 4 of the estrous cycle (P < 0.05) and Day 13 of pregnancy (P < 0.05) than on Day 13 of the estrous cycle. Levels of mRNA encoding PGDH were also greater in corpora lutea on Day 4 of the estrous cycle (P < 0. 01) and Day 13 of pregnancy (P < 0.01) than on Day 13 of the estrous cycle. Thus, during the early estrous cycle and early pregnancy, the corpus luteum has a greater capacity to catabolize PGF, which may play a role in the resistance of the corpus luteum to the actions of this hormone. Levels of mRNA encoding COX-2 were undetectable in corpora lutea collected on Day 13 of the estrous cycle but were 11 +/- 4 and 44 +/- 28 amol/microgram poly(A)(+) RNA in corpora lutea collected on Day 4 of the estrous cycle and Day 13 of pregnancy, respectively. These data suggest that there is a greater capacity to synthesize PGF(2alpha), early in the estrous cycle and early in pregnancy than on Day 13 of the estrous cycle. In conclusion, enzymatic activity of PGDH may play an important role in the mechanism involved in luteal resistance to the luteolytic effects of PGF(2alpha).  相似文献

It is well known that prostaglandin F(2alpha) (PGF(2alpha)) is a physiological luteolysine, and that its pulsatile release from the endometrium is a luteolytic signal in many species. There is now clear evidence that the vasoactive peptides endothelin-1 (ET-1) and angiotensin II (Ang II) interact with PGF(2alpha) in the luteolytic cascade during PGF(2alpha)-induced luteolysis in the cow. Thus, we investigated the local secretion of PGF(2alpha), ET-1, and Ang II in the corpus luteum (CL) and their real-time relationships during spontaneous luteolysis in the cow. For this purpose, an in vivo microdialysis system (MDS) implanted in the CL was utilized to observe local secretion changes within the CL microenvironment. Each CL of cyclic Holstein cows (n = 6) was surgically implanted with MDS capillary membranes (18 lines/6 cows) on Day 15 (estrus = Day 0) of the estrous cycle. The concentrations of PGF(2alpha), ET-1, Ang II, and progesterone (P) in the MDS samples were determined by enzyme immunoassays. The intraluteal PGF(2alpha) secretion slightly increased from 12 h after the onset of luteolysis (0 h) and drastically increased (by about 300%) from 24 h. Intraluteal ET-1 secretion increased from 12 h. Intraluteal Ang II secretion was elevated from 0 h and was maintained at high levels (about 180%) toward estrus. In each MDS lines (in the same microenvironment) within the regressing CL, the local releasing profiles of PGF(2alpha), ET-1, and Ang II CL positively correlated with each other (P < 0.05) at high proportions in 18 MDS lines (PGF(2alpha) vs. ET-1, 44.4%; PGF(2alpha) vs. Ang II, 55.6%; ET-1 vs. Ang II, 38.9%). In contrast, there was no clear relationship among these substances released into different MDS lines implanted in the same CL (with different microenvironments). In conclusion, we propose that the increase of PGF(2alpha), ET-1, and Ang II within the CL during luteolysis is a common phenomenon for both PGF(2alpha)-induced and spontaneous luteolysis. Moreover, this study illustrated the in vivo relationships in intraluteal release among PGF(2alpha), ET-1, and Ang II during spontaneous luteolysis in the cow. The data suggest that these vasoactive substances may interact with each other in a local positive feedback manner to activate their secretion in the regressing CL, thus accelerating and completing luteolysis.  相似文献

The timing of PGF(2alpha) release and the timing and extent of the rise in endometrial oxytocin receptors was determined in relation to the timing of the progesterone fall during luteolysis in cycling cows. In cows undergoing luteolysis (n = 6), measurement of PGF(2alpha) metabolite in hourly plasma samples collected during daily 10 h sampling periods identified a total of 2.2+/-0.5 PGF(2alpha) release episodes per animal, each of 4.0+/-0.4 h duration. In cows in which luteolysis was not observed (n = 4) no PGF(2alpha) release episodes were identified. In a further three cows in which additional repeated uterine biopsies were collected on days 15, 17, 19, 21 and 23, endometrial oxytocin receptors were initially undetectable (<15 fmol/mg protein) but had increased to 120+/-19 fmol/mg protein prior to the initiation of PGF(2alpha) release episodes. Receptor concentrations then continued to increase reaching peak concentrations of 651+/-142 after luteolysis had been completed.  相似文献

This study was designed to test the efficacy of melengestrol acetate (MGA) in combination with prostaglandin F(2alpha) (PGF(2alpha)) in synchronizing estrus in cyclic and noncyclic heifers. One hundred thirty-one cyclic and prepubertal crossbred heifers were randomly assigned to three treatment groups: Controls (n = 43); MGA (0.5 mg/d for 7 d) and PGF(2alpha) (25 mg i.m. on Day 7; n = 44); and PGF(2alpha) (25 mg i.m. on Day 7; n = 44). Observations for estrus were made at 6-n intervals throughout the 7-d treatment period followed by a 34-d artificial insemination breeding season. A greater percentage (P < 0.05) of MGA-PGF(2alpha) noncyclic heifers showed behavioral estrus (91%) than did Control (67%) or PGF(2alpha) heifers (61%) during the 34-d artificial insemination period. There was no difference (P > 0.05) between synchronization rates of the MGA-PGF(2alpha) heifers and PGF(2alpha) heifers 7 d after PGF(2alpha) administration. The percentage of control animals in estrus during the first 25 d of the breeding season did non differ from the synchronized rates of MGA-PGF(2alpha) and PGF(2alpha) heifers (P > 0.05). Conception rates (heifers pregnant/heifers inseminated) did not differ (P > 0.05) for cyclic or prepubertal heifers among Control, MGA-PGF(2alpha) or PGF(2alpha) heifers. Though conception rates did not differ, there was a trend toward lowered conception rates in MGA-PGF(2alpha) heifers.  相似文献

Following observation of estrus, 134 Holstein heifers were given injections of Prostaglandin F(2)alpha (PGF(2)alpha) between Days 5 and 10 of their cycle (estrus = Day 0). They were then randomly assigned to either a group receiving 400 mug of estradiol benzoate (E(2)B) 40 h or maintained as controls. Heifers observed in estrus within 120 h of PGF(2)alpha administration were inseminated (approximately 12 h after initial observation of estrus). Blood samples for progesterone determination were drawn from the coccygeal vein on Days 15 and 21 after insemination. Pregnancy was confirmed by palpation per rectum between Days 5.0 and 60 post insemination. When control and treated heifers were compared it was found that a higher percentage of heifers treated with E(2)B exhibited estrus after PGF(2)alpha, but there had been no effect on subsequent progesterone concentrations or pregnancy rates.  相似文献
