This report describes the ultrastructural development of plasmodial tapetum and pollen wall of Butomus umbellatus. The tapetum contains extensive arrays of rough endoplasmic reticulum, vesicles from which are responsible for the formation of sporopollenin-like bodies. The tapetum is also involved in the formation of other forms of sporopollenin precursors. Development of pollen wall continues after microspores are released from their callosic walls; they are then enclosed by plasmodial tapetum. The activity and products of the plasmodial tapetum show substantial correlation with pollen wall development, particularly ektexine formation. In B. umbellatus, the tapetum intrudes into the anther locule at early microspore stage. This timing of plasmodial intrusion occurs at a later stage of pollen development as compared to those in the advanced monocotyledons. We report the rough endoplasmic reticulum origin of sporopollenin-like bodies and their occurrence in the plasmodial tapeta of B. umbellatus.  相似文献

Summary Male sterility in Pennisetum americanum (L.) Leeke, inbred line IP 482, was found to be inherited as a monofactorial recessive phenotype. Homozygosity for the gene designated ms 2, produced in addition to pollen abortion, plasmodial tapetum, plasmodial pollen mother cells, delayed and asynchronous meiotic development, desynapsis and blockage of meiosis. Low temperatures during rice (Oryza sativa L.) pollen development cause pollen sterility and decreased grain yield. We show that the time of highest sensitivity to cold coincides with the time of peak tapetal activity: the transition of the tetrad to early uni-nucleate stage (young microspore, YM stage). Low temperatures at this stage of pollen development result in an accumulation of sucrose in the anthers, accompanied by decreased activity of cell wall bound acid invertase and depletion of starch in mature pollen grains. Expression analysis of two cell wall (OSINV1, 4) and one vacuolar (OSINV2) acid invertase genes showed that OSINV4 is anther-specific and down-regulated by cold treatment. OSINV4 is transiently expressed in the tapetum cell layer at the YM stage, and later from the early binucleate stage in the maturing microspores. The down-regulation of OSINV4 expression in the tapetum at YM may cause a disruption in hexose production and starch formation in the pollen grains. In a cold-tolerant cultivar, OSINV4 expression was not reduced by cold; sucrose did not accumulate in the anthers and starch formation in the pollen grains was not affected.  相似文献

The developmental stages of the pollen wall and tapetum, together with exine morphology were studied in a number of Crocus species, by light and scanning electron microscopy. Gametogenesis was characterized by: 1) development of a thick intine, 2) single mitosis, and 3) terminal amylolysis. The tapetum was of the secretory type. In C. cartwrightianus cv. albus, abnormal sporogenesis and gametogenesis produced vacuolate pollen grains with a reduced-or no intine layer, and rich with starch granules; the tapetum was either of the parietal-or amoeboid type. The exine was echinate and the pollen grains had different types of aperture: furrows, colpi or pores. The ornamentation varied from microreticulate to irregularly perforate. The exine framework was overlaid by a pellicle resistant to chloroform-carbon disulphide, on which a layer of pollenkitt was deposited. The results are discussed from both cytological and evolutionary viewpoints.  相似文献

Pollen grain morphology, sculpturing, and wall ultrastructure are investigated in two species ofCoris (Primulaceae),C. monspeliensis L. andC. hispanica Lange. The study includes both acetolysed and unacetolysed pollen. No evidence of any major palynological difference is recorded between these two species, apart from a somewhat larger pollen inC. monspeliensis. However,Coris can be distinguished from the remaining members of thePrimulaceae by the conjunction of relatively large pollen grains, prominent margo, and particular tectal pattern causing a reticulate surface with minute luminal perforations decreasing towards the colpi. From both these distinctive features, and others typically primulaceous, some evolutionary considerations are inferred. Finally, the higher proportion of irregular grains inC. hispanica is interpreted in light of environmental stress.  相似文献

The Arabidopsis anther has a bilateral symmetry with four lobes, each consisting of four distinct layers of somatic cells from the outer to inner side: epidermis, endothecium, middle layer and tapetum. The tapetum is a layer of cells comprising the inner surface of the pollen wall. It plays an important role in anther development by providing enzymes, materials and nutrients required for pollen maturation. Genes and molecular mechanisms underlying tapetum formation and pollen wall biosynthesis have been studied in Arabidopsis. However, tapetum degeneration and anther dehiscence have not been well characterized at the molecular level. Here, we report that an Arabidopsis gene, designated reduced male fertility (RMF), regulates degeneration of tapetum and middle layer during anther development. The Arabidopsis dominant mutant rmf-1D overexpressing the RMF gene exhibited pleiotropic phenotypes, including dwarfed growth with small, dark-green leaves and low male fertility. Tapetum development and subsequent degeneration were impaired in the mutant. Accordingly, pollen maturation was disturbed, reducing the male fertility. In contrast, tapetum degeneration was somewhat accelerated in the RMF RNAi plants. The RMF gene was expressed predominantly in the anther, particularly in the pollen grains. Notably, the RMF protein contains an F-box motif and is localized to the nucleus. It physically interacts with the Arabidopsis-Skp1-like1 protein via the F-box motif. These observations indicate that the RMF gene encodes an F-box protein functioning in tapetum degeneration during anther development.  相似文献

We previously identified a 0.7 Kb cDNA fragment of Zm401, a novel pollen-specific gene in maize (Zea mays). However, little information is known about the function of Zm401 in pollen development. The full-length of Zm401 cDNA was amplified by 5′ RACE and 3′ RACE and both sequence analysis and in vitro translation of Zm401 showed that it belonged to an mRNA-like non-coding gene. To analyze its possible biological roles in pollen development, the Zm401 cDNA was overexpressed in transgenic maize under the control of a pollen specific promoter Zm13 or a CaMV 35S promoter. RT-PCR and RNA gel blot analysis indicated that the expression level of Zm401 in leaves and anthers of transgenic plants was much higher than that of non-transformants. Compared with the non-transformed maize, transgenic maize showed distinct phenotypes, such as abnormal tassels and degenerate anthers. The histological observation showed that the development of pollen grains and anthers in transgenic plants were abnormal. These abnormalities include delayed degradation of tapetum, asynchronous fusion of pollen sacs, and aborted pollen grain development. Furthermore, the pollen viability in six transgenic plants ranged from 1.24% to 6.63%. The reduced pollen viability cosegregated with the transgene in a selfed progeny. These results suggest that Zm401 is involved in the regulation of pollen development. This article demonstrated Zm401, as a non-coding RNA, plays an essential role in pollen development. Chinese narcissus (Narcissus tazetta var. chinensis Roem) blooms but has no seeds. Embryological studies on the species were conducted to discover the causes of its sterility. Its anther wall is composed of four layers of cells, and its tapetum is of the secretory type. The cytokinesis of microspore mother cells is of the successive type, and the tetrad is tetrahedral. During meiosis of microspore mother cells, some chromosomes lagged, and several micronuclei were found in tetrads. Only 27.7% of the pollen grains contained full cytoplasm, and 1.3% of them germinated in culture medium. No pollen grain, however, could germinate on the stigma. The ovary is trilocular with axile placenta, and the ovules are bitegmic, tenuinucellate, and anatropous. Its embryo sac is of the polygonum type. Most embryo sacs degenerated, and only about 4.5% of the ovules contained a normal embryo sac with an egg cell, two synergids, three antipodal, and a central cell containing two polar nuclei. One reason for the sterility of Chinese narcissus is the abnormality of microsporogenesis and megasporogenesis, in which only a few functional pollen grains and embryo sacs are produced. In the anthers of flowering plants, gymnosperms, and seed ferns, tiny (±1?μm) granules might occur on the radial and innermost tangential wall of secretory tapetum cells. These sporopollenin granules develop simultaneously with the pollen exine and are called orbicules or Ubisch bodies. The present paper focuses on two quite different topics associated with orbicules.

The morphological and ultrastructural diversity of orbicules in the order Gentianales is summarized, and it is demonstrated that orbicules are a plesiomorphic feature in the order. Furthermore, orbicule characters seemed to be correlated with evolutionary trends in pollen dispersal unit and tapetum type features.

In the second part, we report on our investigation of Corylus avellana L. (Hazel) pollen, using immunogold electron microscopy to gain an insight into the possible role that orbicules may play as a vector of pollen allergens. During the pollen season orbicules are dispersed into the atmosphere along with Hazel pollen grains. The localisation of homologues of the new birch pollen allergen Bet v 7 was studied at the subcellular level in Hazel anthers. The results of this study indicate that orbicules and pollen of Hazel might act as very effective vectors for homologues of Bet v 7 and that debris of Hazel anthers represent vectors of allergens after the pollen season.  相似文献

In flowering plants, the tapetum, the innermost layer of the anther, provides both nutrient and lipid components to developing microspores, pollen grains, and the pollen coat. Though the programmed cell death of the tapetum is one of the most critical and sensitive steps for fertility and is affected by various environmental stresses, its regulatory mechanisms remain mostly unknown. Here we show that autophagy is required for the metabolic regulation and nutrient supply in anthers and that autophagic degradation within tapetum cells is essential for postmeiotic anther development in rice. Autophagosome-like structures and several vacuole-enclosed lipid bodies were observed in postmeiotic tapetum cells specifically at the uninucleate stage during pollen development, which were completely abolished in a retrotransposon-insertional OsATG7 (autophagy-related 7)-knockout mutant defective in autophagy, suggesting that autophagy is induced in tapetum cells. Surprisingly, the mutant showed complete sporophytic male sterility, failed to accumulate lipidic and starch components in pollen grains at the flowering stage, showed reduced pollen germination activity, and had limited anther dehiscence. Lipidomic analyses suggested impairment of editing of phosphatidylcholines and lipid desaturation in the mutant during pollen maturation. These results indicate a critical involvement of autophagy in a reproductive developmental process of rice, and shed light on the novel autophagy-mediated regulation of lipid metabolism in eukaryotic cells.  相似文献

利用常规石蜡切片法和细胞学压片法,对异型花柱植物滇丁香的长花柱型植株的小孢子发生、雄配子体发育及花粉萌发进行观察。结果表明:(1)长花柱型滇丁香具5枚花药,花药4室。(2)花药壁由1层表皮、1层花药内壁、2层中层和1~3层绒毡层组成;花药壁发育方式为基本型,绒毡层类型为腺质绒毡层。(3)小孢子母细胞减数分裂的胞质分裂为同时型,四分体排列方式为四面体型,偶有左右对称型;不同药室间小孢子母细胞减数分裂不同步。(4)成熟花粉粒为二细胞型。(5)小孢子发生和雄配子体发育过程正常,表明长花柱型滇丁香属于发育正常的两性花。(6)授粉4h后,长花柱型滇丁香的花粉在长、短两种花柱柱头上的萌发率分别达(91.8±1.6)%和(93.2±1.1)%,且两者间无显著性差异(t=1.585,df=8,p=0.152),表明长花柱型滇丁香的成熟花粉粒在长、短两种花柱柱头上的萌发均正常。  相似文献

Using the monoclonal antibodies JIM 5 and 7, pectin was immunolocalized and quantitatively assayed in three anther compartments of Lilium hybrida during pollen development. Pectin levels in both the anther wall and the loculus increased following meiosis, were maximal during the early microspore stages and declined during the remainder of pollen ontogenesis. In the microspores/pollen grains, pectin was detectable at low levels during the microspore stages but accumulated significantly during pollen maturation. During early microspore vacuolation, esterified pectin epitopes were detected both in the tapetum cytoplasm and vacuoles. In the anther loculus, the same epitopes were located simultaneously in undulations of the plasma membrane and in the locular fluid. At the end of microspore vacuolation, esterified pectin epitopes were present within the lipids of the pollenkitt, and released in the loculus at pollen mitosis. Unesterified pectin epitopes were hardly detectable in the cytoplasm of the young microspore but were as abundant in the primexine matrix as in the loculus. During pollen maturation, both unesterified and esterified pectin labelling accumulated in the cytoplasm of the vegetative cell, concurrently with starch degradation. In the mature pollen grain, unesterified pectin epitopes were located in the proximal intine whereas esterified pectin epitopes were deposited in the distal intine. These data suggest that during early microspore development, the tapetum secretes pectin, which is transferred to the primexine matrix via the locular fluid. Anther development, microsporogenesis and microgametogenesis in several species of Heliconia were investigated as part of a complementary embryological study of the Heliconiaceae. All studied Heliconia species present bithecate and tetrasporangiate anthers with fertile pollen grains; only H. rivularis, a natural hybrid, presented sterile pollen grains of variable size and no content. The anther wall has an uniseriate epidermis and endothecium, the latter with helicoidal thickenings, although some cells of the middle layers also showed thickenings; the biseriate tapetum is of amoeboid non-syncytial type, since the tapetum cells did not fuse together forming a true plasmodium. The microsporogenesis is successive leading to isobilateral tetrads. The inaperturate pollen grains had a very reduced exine consisting of a thin, more or less continuous layer with small spines upon; the pollen grain shape is variable among the species, all of them presenting heteropolar pollen, except H. angusta with isopolar ones. Most of these characteristics were shared with other studied Zingiberales, although more studies need to be done.  相似文献

Abstract

« Harmomegathy »: a still open problem not yet well-known. - Wodehouse used the term « Harmomegathy » to indicate the form and volume change in pollen grains, due to water loss or assumption. Roundish and tricolporate grains in the anther become oval and decrease in volume during the dehydration preceeding anthesis. Viceversa during rehydration on the stigma they undergo the opposite process.

Pollen water content at an thesis varies from species to species and it usually ranges between 10 and 20%; nevertheless in some families as Zingiberaceae and Araceae it is higher. Anther and pollen dehydrations are affected by environmental factors, such as humidity of air and temperature, before the anthesis and when pollen is suspended in air. Besides, to avoid the further loss of water during pollen flow, an oily layer derived from tapetum degeneration is deposited outside the pollen grains.

During dehydration and rehydration pollen grains undergo a stress due to shape and volume changes expecially in the colpus and/or pore areas. Pollen walls are structured in order to avoid their own damage, which could be followed by protoplast death and/or irregularities in grain shape. Intine, formed by pectocellulosic filaments, has elastic properties, as sporopollenin, the component of exine.

Stigma can be dry or wet, namely covered or not by stigmatic exudate giving place to different hydration patterns. The structure of intine and exine has to match that of the stigma, in fact pollen will find the best place and way to proper hydration only on stigmas of its own species. Grains structure also must be sterically conform to stigma: this is well-shown in species as Linum, Lytrum and Primula which have an heteromorphic autoincompatibility.

The right keeping of shape and structure of grains, despite the hydration and dehydration, is therefore essential to the physical-morphological recognition between pollen and stigma, which preceedes the biochemical recognition.  相似文献

Pollen size and exine structure of the following five Onosma L. species from Slovakia and Italy were studied by light and scanning electron microscopy: Onosma echioides, O. tornensis, O. visianii, O. arenaria, and O. pseudoarenaria. Among the taxa studied, O. echioides has the smallest pollen grains in average (10.5–17.5 × 8.0–14.5 μm), and O. pseudoarenaria the largest grains in average (15.3–20.5 × 12.3–16.3 μm). Pollen grains of all 5 species are small sized, 3-syncolporate, subprolate, heteropolar, with ovate equatorial outlines and circular to rounded triangular polar outlines; the tectum is microechinate. A positive correlation is found between pollen size and chromosome number. The value of pollen characters for taxonomic purposes and the position of the taxa studied within the genus Onosma are discussed.  相似文献

Locule structure and organization were studied in vivo and in vitro to determine whether the disposition of pollen within barley anthers affected the response of pollen in culture. Following release from the meiotic tetrads, juvenile barley microspores become peripherally organized around the locule, with the single pollen pore oriented towards the tapetum. Scanning electron micrographs of transverse sections from freeze fractured anthers showed that some microspores failed to locate the tapetal surface and occupied a position in the centre of the locule where they continued to develop as small, abnormal pollen grains (dimorphic pollen). Previous evidence has suggested that in some species dimorphic pollen could be the source of embryonic pollen in vitro. Cultured anthers frequently dehisced to reveal a mass of dividing pollen grains, however those anthers that remained intact retained the original locule structure and could be freeze fractured permitting examination of the developing pollen in situ. This showed that pollen embryogenesis was not restricted to dimorphic pollen, and that any grain could become Embryogenic irrespective of position.  相似文献

This is the first data from a pollen survey in Vigo, an Atlantic city in northwest Spain. The pollen calendar for Vigo is presented, as well as the pollination period for the nine most important allergenic plants. Through 1995, 30 083 pollen grains belonging to 52 taxa, were recorded using the Lanzoni VPPS 2000 volumetric spore-trap. The most relevant taxa found were: Urticaceae,Pinus, Poaceae andQuercus (75% of the total pollen),Betula, Castanea, Cupressaceae, Chenopodiaceae, Ericaceae, Myrtaceae,Olea, Plantago, Platanus andRumex (21%), and the final 4% was distributed mainly among pollen types, such as:Corylus, Alnus, Fabaceae, Compositae,Artemisia andCedrus. Of the total annual pollen count, 56% was found in March and April. Another, secondary peak was recorded in June corresponding to the flowering period of herbaceous species. The high pollen total of Urticaceae (7625 grains, 25% of the total) should be highlighted. The percentages ofOlea europaea (565 grains) should be noted as well, taking into account its geographical distribution.  相似文献

Chloroplast DNA (cpDNA) restriction site analysis was used to assess relationships among samples of Eupatorieae from eastern North America. A total of 270 cpDNA variants was recorded from 35 species using 13 restriction enzymes. Phylogenetic analysis usingGalinsoga, Flaveria, andHelianthinae as outgroups indicated that samples ofAgeratina, Hofmeisteria, Mikania, andStevia, each of which have relatively high base chromosome numbers, formed an unresolved basal polytomy. The remaining samples examined formed a well-supported clade, within which there was a split betweenBrickellia, which hasx = 9, and a number of genera withx = 10. Within thex = 10 clade, there was an unresolved trichotomy, with one group ofAgeratum, Conoclinium, Fleischmannia, andCritonia, a second ofLiatris andTrilisa, and a third ofEupatorium s.s. Within theEupatorium s.s. clade there were three further clades, withE. sect.Verticillata diverging first, and a subsequent split between species originating from North America and those from Asia. The cpDNA restriction site data provided support for a relatively narrow interpretation ofEupatorium, and indicated that a high chromosome base number is plesiomorphic for Eupatorieae.  相似文献
