生物电化学系统还原降解氯霉素   总被引:2,自引:0,他引:2  
为了探讨低温(12±2℃)条件下还原降解硝基芳香类抗生素氯霉素,采用序批式生物电化学系统(Bioelectrochemical system,BES)阴极还原的方式(外加0.5 V电压),主要研究氯霉素在BES生物阴极与非生物阴极中的不同降解速率、代谢途径和氯霉素在电化学系统中被还原为胺类产物从而脱除细菌抗性.实验数据表明,BES反应器整体的欧姆内阻随着磷酸盐缓冲液浓度的增加而减小;当葡萄糖和污泥发酵液分别存在时,生物阴极24 h的氯霉素还原效率分别为(86.3±1.69)%和(74.1±1.44)%,而相同条件下的非生物阴极24 h氯霉素还原效率仅为(57.9±1.94)%.研究结果表明,生物电化学系统还原降解氯霉素使其失去生物抗性是可行的,并且在低温地区含氯霉素废水的处理过程中,生物阴极是极具潜力的一项处理工艺.  相似文献

【目的】为明确变温条件对二点委夜蛾Athetis lepigone (Möschler) 实验种群生长发育的影响。【方法】分别设置6个变温组合(20/24℃, 20/28℃, 20/32℃, 24/28℃, 24/32℃和28/32℃),通过测定和计算各虫态发育起点温度和有效积温及生命表参数来研究变温对二点委夜蛾卵、幼虫、蛹、世代平均发育历期、存活率和繁殖力的影响。【结果】不同变温组合对二点委夜蛾各虫态的发育历期、存活率及种群繁殖力有显著影响。发育历期随着变温组合平均温度的升高而缩短,二点委夜蛾世代存活率由大到小的顺序为24/28℃>20/32℃>28/32℃>20/28℃>24/32℃>20/24℃。24/28℃时产卵量最高,单雌产卵量为390.70粒。世代发育起点温度和有效积温分别为10.37℃和663.17日·度。在不同变温组合下内禀增长率rm值随着平均温度的升高先上升后降低。在24/28℃时,二点委夜蛾rm、净增殖率R0和周限增长率λ最大,种群加倍时间 t 和世代平均周期 T 最小。【结论】24/28℃是二点委夜蛾实验种群生长发育及繁殖的最适变温组合,本研究结果为二点委夜蛾的预测预报及综合防治提供了科学依据。  相似文献

The temperature dependence of the oligomycin-sensitive ATPase (complex V) kinetic parameters has been investigated in enzyme preparations of different phospholipid composition. In submitochondrial particles, isolated complex V, and complex V reconstituted in dimirystoyl lecithin vesicles, the Arrhenius plots show discontinuities in the range 18–28°C, while no discontinuity is detected with dioleoyl lecithin recombinant. Van't Hoff plots ofK m also show breaks in the same temperature interval, with the exception of the dioleoylenzyme vesicles, whereK m is unchanged. Thermodynamic analysis of the ATPase reaction shows that DMPC-complex V has rather larger values of activation enthalpy and activation entropy below the transition temperature (24°C) than those of the other preparations, while all enzyme preparations show similar free energies of activation (14.3–18.5 kcal/mol). The results indicate that temperature and lipid composition influence to a different extent both kinetic and thermodynamic parameters of ATP hydrolysis catalyzed by the mitochondrial ATPase.  相似文献

Theory and performance of an infrared heater for ecosystem warming   总被引:5,自引:0,他引:5  
In order to study the likely effects of global warming on future ecosystems, a method for applying a heating treatment to open-field plant canopies (i.e. a temperature free-air controlled enhancement (T-FACE) system) is needed which will warm vegetation as expected by the future climate. One method which shows promise is infrared heating, but a theory of operation is needed for predicting the performance of infrared heaters. Therefore, a theoretical equation was derived to predict the thermal radiation power required to warm a plant canopy per degree rise in temperature per unit of heated land area. Another equation was derived to predict the thermal radiation efficiency of an incoloy rod infrared heater as a function of wind speed. An actual infrared heater system was also assembled which utilized two infrared thermometers to measure the temperature of a heated plot and that of an adjacent reference plot and which used proportional–integrative–derivative control of the heater to maintain a constant temperature difference between the two plots. Provided that it was not operated too high above the canopy, the heater system was able to maintain a constant set-point difference very well. Furthermore, there was good agreement between the measured and theoretical unit thermal radiation power requirements when tested on a Sudan grass (Sorghum vulgare) canopy. One problem that has been identified for infrared heating of experimental plots is that the vapor pressure gradients (VPGs) from inside the leaves to the air outside would not be the same as would be expected if the warming were performed by heating the air everywhere (i.e. by global warming). Therefore, a theoretical equation was derived to compute how much water an infrared-warmed plant would lose in normal air compared with what it would have lost in air which had been warmed at constant relative humidity, as is predicted with global warming. On an hourly or daily basis, it proposed that this amount of water could be added back to plants using a drip irrigation system as a first-order correction to this VPG problem.  相似文献

The content and composition of pigments were examined in the third leaf of Zea mays L. plants grown under controlled environment at near-optimal temperature (24°C) or sub-optimal temperature (14°C) at a light intensity of either 200 or 600 μmol m?2 s?1. Compared to leaves grown at 24°C, leaves grown at 14°C showed a large reduction in the chlorophyll (Chl) content, a marked decrease in the Chl a/b ratio, and a large increase in the ratio of total carotenoids/Chl a+b. Leaves grown at 14°C showed a much lower content of β-carotene than leaves grown at 24°C, while the content of the carotenoids of the xanthophyll cycle (violaxanthin [V] + antheraxanthin [A] + zeaxanthin [Z]) was markedly higher in the former leaves as compared to the latter leaves; neoxanthin and lutein were affected by the growth temperature to a much lesser extent. The xanthophylls/β-carotene ratio was about three times higher in leaves grown at 14°C as compared to leaves grown at 24°C. On a chlorophyll basis, the two types of leaves hardly differed in their level of β-carotene, while the levels of the xanthophylls (including lutein and neoxanthin) were higher in 14°C-grown leaves as compared to 24°C-grown leaves. In leaves grown at 14°C, 40 and 56% of the V+A+Z pool was in the form of zeaxanthin at low light intensity and high light intensity, respectively. Only trace amounts of zeaxanthin, if any, were present in leaves grown at 24°C. The changes in the pigment composition induced by growth at sub-optimal temperature were more pronounced at a light intensity of 600 as compared to 200 μmol m?2 s?1. In the given range, the light intensity slightly affected the composition of pigments in leaves grown at 24°C. The physiological significance of the modifications to the pigment composition induced by growth at sub-optimal temperature is discussed.  相似文献

These experiments were undertaken to measure heat production of fetal lambs in utero by using differential calorimetry. We used the principle that fetal heat production, H(fetus), can be calculated from measurements of base-line temperature difference between mother and fetus, delta T(fetus), heat introduced from an external source, H(heater), and the increase in body temperature, delta T(heater), that results, i.e., H(fetus) = H(heater) X delta T(fetus)/delta T(heater). We placed microheaters (1.8 mm diam) in the inferior vena cavae of eight near-term lambs and placed thermistors and catheters into maternal and fetal vessels and amniotic fluid. Five days later, fetal arterial temperature averaged 0.54 +/- 0.02 degrees C (SE) higher than maternal arterial temperature. When the heater was turned on to dissipate 29-103 cal/min, fetal temperature increased to approach 0.1-0.5 degrees C higher than control; the final temperature was estimated using the rate of increase during the first 20 min. Fetal heat production averaged 47.1 +/- 4.1 cal X min-1 X kg-1 during the warming phase in these lambs, which weighed 3.26 +/- 0.36 kg. This value would be 3-4% less if corrected for the increase in metabolic rate caused by heating, assuming a Q10 of 2.5. Fetal heating did not alter fetal heart rate, blood pressure, or blood gas values significantly, nor was hemolysis visible in plasma samples. When heat production was calculated from the decrease in fetal temperature after the heater was turned off, an average value of 41.2 +/- 2.5 cal X min-1 X kg-1 was found. Because this value is comparable to the heating phase, fetal metabolic rate and the insulating properties of the fetal shell are not likely to have been changed by the heating.  相似文献

The effects of changes in growth temperature on photosynthesis and carotenoid composition were examined in Zea mays L. leaves of different age and different developmental history. The plants were first grown at sub-optimal temperature (14°C) until the full development of the third leaf. At that time, the mature third leaf and the immature fourth leaf had a low chlorophyll (Chl) content, a low Chl a/b ratio, a high carotenoid/Chl a+b ratio, a high xanthophyll/β-carotene ratio, and about 80% of the xanthophyll cycle pool (violaxanthin [V] + antheraxanthin [A] + zeaxanthin [Z]) was in the form of zeaxanthin and antheraxanthin. When the temperature was increased from 14°C to 24°C for three days, increased Chl synthesis, accompanied by an increase in the Chl a/b ratio, took place. The ratios of lutein, neoxanthin, and V+A+Z to Chl a+b decreased markedly, whereas no significant changes appeared in the β-carotene/Chl a+b ratio. Furthermore, there was a sharp decrease in the xanthophyll/β-carotene ratio and most of zeaxanthin was converted to violaxanthin in the xanthophyll cycle. The third leaf and the tip segment of the fourth leaf, both expanded at 14°C, showed little difference in their pigment contents. However, the rate of CO2 assimilation of the tip segment of the fourth leaf was nearly twice that of the third leaf on the third day at 24°C, while the photosynthetic activity was similar in both leaves before the transfer to 24°C. During the warm period at 24°C, new leaf tissue (basal segment of the fourth leaf and part of a fifth leaf) was formed. On the third day at 24°C, the pigment content of 24°C-grown leaf tissue did not differ much from that of 14°C-grown leaf tissue with the exception that the total carotenoid content was lower in the former as compared to the latter, mainly because of a lower V+A+Z content. The rate of CO2 assimilation of 24°C-grown leaf tissue was comparable to that of the tip segment of the fourth leaf. Regardless of which leaf tissue is considered, reducing the temperature from 24°C to 14°C for 5 days slightly affected the pigment content, but violaxanthin was largely converted to zeaxanthin and antheraxanthin in the xanthophyll cycle. The results indicate that compared to old leaf tissue of mature leaves, physiologically younger leaf tissue of immature leaves is much more able to recover from depressions in the photosynthetic activity induced by growth at sub-optimal temperature when the plants experience optimal growth temperatures, but that factors other than the pigment content must determine this capability.  相似文献

Bioluminescence induced by multifarious stimuli has long been observed and is remains under investigation because of its great complexity. In particular, the exact mechanism underlying bioluminescence is not yet fully understood. This work presents a new experimental method for studying Noctiluca sp. bioluminescence under temperature change stimulation. It is a study of Noctiluca sp. bioluminescence using controlled temperature changes in a tank. A characteristic of this experiment is the large volume of water used (1 m3 in a tank of 2 × 1 × 1 m). Temperature changes were controlled by two methods. In the first, a flask filled with hot water was introduced into the tank and in the second, a water heater was used in the tank. Temperature changes were recorded using sensors. Noctiluca sp. bioluminescence was recorded using a Canon 5D Mark II and this allowed the characteristics of Noctiluca sp. bioluminescence under temperature change stimulation to be monitored. There are large inter- and intraspecific differences in the temperature dependence of photosynthesis, but the physiological cause of the variation is poorly understood. Here, the temperature dependence of photosynthesis was examined in three ecotypes of Plantago asiatica transplanted from different latitudes, where the mean annual temperature varies between 7.5 and 16.8 degrees C. Plants were raised at 15 or 30 degrees C, and the CO(2) response of photosynthetic rates was determined at various temperatures. When plants were grown at 30 degrees C, no difference was found in the temperature dependence of photosynthesis among ecotypes. When plants were grown at 15 degrees C, ecotypes from a higher latitude maintained a relatively higher photosynthetic rate at low measurement temperatures. This difference was caused by a difference in the balance between the capacities of two processes, ribulose-1,5-bisphosphate regeneration (J(max)) and carboxylation (V(cmax)), which altered the limiting step of photosynthesis at low temperatures. The organization of photosynthetic proteins also varied among ecotypes. The ecotype from the highest latitude increased the J(max) : V(cmax) ratio with decreasing growth temperature, while that from the lowest latitude did not. It is concluded that nitrogen partitioning in the photosynthetic apparatus and its response to growth temperature were different among ecotypes, which caused an intraspecific variation in temperature dependence of photosynthesis.  相似文献

PurposeTo develop an MRI-compatible resistive heater, using high frequency alternating current (AC), for temperature maintenance of anaesthetised animals.ResultsImages and spectra acquired in the presence and absence of 50–100 kHz AC through the wire heater were indistinguishable, whereas DC power created field shifts and lineshape distortions. Temperature lost during induction of anaesthesia was recovered within approximately 20 minutes and a stable temperature was reached as the mouse’s temperature approached the set target.ConclusionThe AC-powered wire heater maintains adequate heat input to the animal to maintain body temperature, and does not compromise image quality.  相似文献

The emergence of Austrobilharzia terrigalensis cercariae from the snail Velacumantus australis decreased with decreasing salinity and temperature. This was most marked between 23 and 25 and between 24 and 19°C. Very few cercariae were shed below 23 regardless of temperature. The decrease in cercarial emergence was associated with the decreasing metabolic activity of the intermediate host. The emergence pattern of A. terrigalensis cercariae from V. australis was diurnal. There was a major peak between 08h00 and 10h00 and a minor second one between 16h00 and 18h00. No cercariae were shed between 22h00 and 04h00. Cercariae survived in sea water (35) for up to 48h, with a half-life of 30h. There was evidence that the prevalence of A. terrigalensis infection in the intermediate host population in the Swan estuary was higher during the summer than during the winter. The production of cercariae in the sporocyst also appeared to be a seasonal phenomenon. Mature cercariae were rare in sporocysts examined during the winter months (May–October) and absent altogether from those examined in September.  相似文献

To assess risk factors associated with the contamination of the domestic environment by legionellae, 211 houses in the Quebec City area were randomly selected and water samples were collected from the hot water tank, the shower heads, and the most frequently used faucet. After centrifugation, concentrated samples were seeded in triplicate on BCYE and GPV media. Data on the characteristics of the hot water system and plumbing in the house and on the personal habits of the occupants were collected for each house. Among these 211 houses, hot water was provided by either an oil or gas heater in 33 and by an electric heater in 178. Legionellae were isolated from none of the samples from houses with oil or gas heaters and from 39% (69 of 178) of those with electric water heaters (P less than 0.0001). This association remained highly significant after control for water temperature and other variables in a stratified analysis. In the 178 houses with an electric heater, 12% of the faucets, 15% of the shower heads, and 37% of the water heaters were contaminated. Legionella pneumophila serogroups 2 and 4 were the most frequently isolated strains. Logistic regression showed that factors associated with electric water heater contamination were (i) location of the house in older districts of the city (P less than 0.0001), (ii) old age of the water heater (P = 0.003), and (iii) low water temperature (P = 0.05). We conducted microcosm experiments with two contrasting freshwater ciliates on functional traits (FTs) related to their growth rate (numerical response, NR) and ingestion rate (functional response, FR) over a range of ecologically relevant temperatures. Histiobalantium bodamicum and Vorticella natans are common planktonic ciliates but their abundance, swimming behavior, and temperature tolerance are different. In contrast to most sessile peritrich species, the motile V. natans is not strictly bacterivorous but also voraciously feeds upon small algae. We observed three main alterations in the shape of NR of both species with temperature, that is, change in the maximum growth rate, in the initial slope and in the threshold food level needed to sustain the population. Similarly, maximum ingestion rate, gross growth efficiency (GGE), and cell size varied with temperature and species. These findings caution against generalizing ciliate performance in relation to the ongoing global warming. Our results suggest that V. natans is the superior competitor to H. bodamicum in terms of temperature tolerance and bottom-up control. However, the abundance of V. natans is usually low compared to H. bodamicum and other common freshwater ciliates, suggesting that V. natans is more strongly top-down controlled via predation than H. bodamicum. The taxonomic position of V. natans has been debated. Therefore, to confirm species and genus affiliation of our study objects, we sequenced their small subunit ribosomal RNA (SSU rDNA) gene.  相似文献

Thermal performance curves (TPCs) compute the effects of temperature on the performance of ectotherms and are frequently used to predict the effect of environmental conditions and currently, climate change, on organismal vulnerability and sensitivity. Using Drosophila melanogaster as an animal model, we examined how different thermal environments affected the shape of the performance curve and their parameters. We measured the climbing speed as a measure of locomotor performance in adult flies and tested the ontogenetic and transgenerational effects of thermal environment on TPC shape. Parents and offspring were reared at 28 ± 0ºC (28C), 28 ± 4ºC (28V), and 30 ± 0ºC (30C). We found that both, environmental thermal variability (28V) and high temperature (30C) experienced during early ontogeny shaped the fruit fly TPC sensitivity. Flies reared at variable thermal environments shifted the TPC to the right and increased heat tolerance. Flies held at high and constant temperature exhibited lower maximum performance than flies reared at the variable thermal environment. Furthermore, these effects were extended to the next generation. The parental thermal environment had a significative effect on TPC and its parameters. Indeed, flies reared at 28V whose parents were held at a high and constant temperature (30C) had a lower heat tolerance than F1 of flies reared at 28C or 28V. Also, offspring of flies reared at variable thermal environment (28V) reached the maximum performance at a higher temperature than offspring of flies reared at 28C or 30C. Consequently, since TPC parameters are not fixed, we suggest cautiousness when using TPCs to predict the impact of climate change on natural populations.  相似文献

We have tested the hypothesis that nitric oxide (NO) arising from inducible nitric oxide synthase (iNOS) plays a role in hypothermia during endotoxemia by regulating vasopressin (AVP) release. Wild-type (WT) and iNOS knockout mice (KO) were intraperitoneally injected with either saline or Escherichia coli lipopolysaccharide (LPS) 10.0 mg/kg in a final volume of 0.02 mL. Body temperature was measured continuously by biotelemetry during 24 h after injection. Three hours after LPS administration, we observed a significant drop in body temperature (hypothermic response) in WT mice, which remained until the seventh hour, returning then close to the basal level. In iNOS KO mice, we found a significant fall in body temperature after the fourth hour of LPS administration; however, the hypothermic response persisted until the end of the 24 h of the experiment. The pre-treatment with beta-mercapto-beta,beta-cyclopentamethylenepropionyl(1), O-Et-Tyr2, Val4, Arg8-Vasopressin, an AVP V1 receptor antagonist (10 microg/kg) administered intraperitoneally, abolished the persistent hypothermia induced by LPS in iNOS KO mice, suggesting the regulation of iNOS under the vasopressin release in this experimental model. In conclusion, our data suggest that the iNOS isoform plays a role in LPS-induced hypothermia, apparently through the regulation of AVP release.  相似文献
