We report on the isolation and characterization of eight microsatellite markers from enriched libraries for Arenaria nevadensis, one of the most critically endangered plant species in the Iberian Peninsula. These are the first microsatellite loci reported
for Arenaria species. The number of alleles ranged from two to eight, and the expected heterozygosity from 0.067 to 0.873. These markers
will be useful for characterizing the genetic diversity in A. nevadensis and understanding its population structure, and will provide important genetic data for the conservation and recovery of

Ostryopsis (Betulaceae) is a samll genus endemic to China with only two species. Both of them play an important role in restoring the
local ecosystems. The distribution of genetic diversity between and within populations in each species are important to further
utilize the wild genetic resources and explore the interspecific divergence. In this study, we developed 10 microsatellite
loci from O. davidiana by the combining biotin capture method for the first time. A total of 27 microsatellite sequences were recovered through
screening the library and 10 of them are polymorphic. The number of alleles per locus in 18 sampled individuals ranged from
3 to 6, expected heterozygosity and observed heterozygosity ranged from 0.2958 to 0.4767 and from 0.1591 to 0.2997, respectively.
In addition, all markers have been crossly checked in the other congeneric species. These microsatellite markers would together
provide a useful tool for investigating the genetic diversity and structure of both species and speciation mechanism between

Rhubarb is an important Traditional Chinese Medicine. However, the wild resource has been declining. In order to design appropriate
conservation methods for the official species across their natural distributions, it is important to characterize their genetic
diversity. Here, we describe the development of 10 new microsatellite loci for AC/TG/CCA in Rheum tanguticum. The microsatellites were enriched using the combined biotin capture method. The polymorphism of each locus was further assessed
in 12 individuals from four geographically distinct populations of this species. The number of alleles ranged from three to
seven and the expected heterozygosity ranged from 0.53 to 0.73. All markers have been checked in the other three species in
the genus and two of them together comprise the official medicinal rhubarb resource with R. tanguticum. These microsatellite markers could provide a useful tool for genetic and conservation studies of the rhubarb species. 相似文献

Juniperus przewalskii (Cupressaceae) is a dominant tree species endemic to the northeast Qinghai-Tibetan Plateau. This species plays an important
role in maintaining the arid ecosystem in this region. However, natural distributions of this species have been declined.
In order to develop effective conservation methods, it is important to know the distribution of the genetic diversity within
and among populations. In this study, we developed nine new microsatellite loci for this species. We used the combining biotin
capture method to enrich AG/CT/AC/GGT microsatellites. The polymorphisms of each locus were further assessed in 12 individuals
from four geographically distant populations. The number of alleles per locus varied from three to six and expected heterozygosity
ranged from 0.58 to 0.70. These loci together provide a useful tool to investigate the genetic diversity of this species.

This paper reports the development of microsatellite primers for Nelumbo nucifera Gaerten. By screening genomic libraries enriched with 10 kinds of probes, Seventeen polymorphic loci were isolated and primers
were designed. Polymorphism of these 17 loci was assessed in 24 individuals. All the 17 loci are polymorphic and the number
of alleles ranged from two to seven. Observed heterozygosity and expected heterozygosity ranged from 0.0000 to 0.9176 and

This paper reports the studies of megasporogenesis and microsporogenesis, development of female and male gametophytes, fertilization, and development of embryo and endosperm, The anther wall consists of four layers, i.e. epidermis, endothecium, middle layer and tapetum. Part of the tapetum cells originates from the primary parietal cells, and the other part comes from the basic tissue of the anther partition. Tapeta? cells are uninucleate or binucleate, and belong to the secretory type. Microsporocyte originates directly from the primary sporogenous cell, Cytokinesis is of the simultaneous type. Arrangement of microspores in tetrad is isobilateral. Mature pollen grain is of the 2-celled type. The ovary is tricarpellum, trilocular with many ovules. The ovule is mono-integinous, tenui-nucellar and anatropous. The embryo sac originates from the single-archesporial cell. The one chalazal megaspore in linear tetrad is the functional megaspore. The development of embryo sac is of the Polygonum type. Before fertilization, two polar nuclei fuse in to a secondary nucleus and the antipodal cells degenerate. Fertilization is porogamy, fusion of one sperm with secondary nucleus is faster than that of one sperm with egg nucleus. The development of endosperm is of the cellular type. The first three divisions of endosperm ceils are regular. Two endosperm cells near the ends of chalaza and the micropyle develop into haustorium without division. The haustoria gradually degenerate at the late stage of globular embryo. The mature seeds contain abundant endosperm. The development of embryo is of the Solanad type. The suspensor consists of 12–20 cells. The optimum development of the suspensor is at the early stage of the globular embryo. It begins to degenerate after late globular stage. A total of twelve polymorphic microsatellite loci were developed from polyploid endangered species, Omphalogramma vincaeflora (Primulaceae). These loci were screened for variability among 45 individuals from three populations in China. The primers
amplified loci with allele number ranging from 3 to 9, with an average of 4.25 per locus. Polymorphism information content
ranged from 0.23 to 0.86. Nei’s genetic diversity ranged from 0.34 to 0.86. These primers provide an opportunity to use polymorphic

Seven polymorphic nuclear microsatellite markers for Taxus baccata L. (English yew) were developed using an enriched-library method. An additional polymorphic SSR was obtained by testing eight
primer pairs from the congeneric species Taxus sumatrana. Mendelian inheritance for the seven Taxus baccata SSRs was proved by genotyping 17 individuals and 124 megagametophytes (conifer seed haploid tissue). A total of 96 individuals
from 5 different populations (10–26 samples per population) were used to estimate genetic diversity parameters. High levels
of genetic diversity, with values ranging from 0.533 to 0.929 (6–28 alleles per SSR) were found. No linkage disequilibrium
between pairs of loci was detected. All loci but one showed significant departures from Hardy–Weinberg equilibrium. Excess
of homozygosity was probably due to high inbreeding in English yew populations, an outcome of low effective population size
and/or fragmented distribution. Highly polymorphic SSRs will be used to conduct population genetic studies at different geographical
scales and to monitor gene flow. 相似文献

The water-soluble polysaccharide (CPP), with a molecular mass of 1.1x10(4) Da, was obtained from the roots of Codonopsis pilosula. Structure feature investigation by a combination of chemical and instrumental analysis revealed that CPP had a backbone consisting of (1-->3)-linked-beta-D-galactopyranosyl, (1-->2, 3)-linked-beta-D-galactopyranosyl and (1-->3)-linked-alpha-D-rhamnopyranosyl residues, which were branched with two glycosyl residues composed of alpha-L-arabinose-(1-->5)-alpha-L-arabinose(1-->linked residues at the O-2 position of galactosyl along the main chain in the ratio of 1:1:2:1:1. Preliminary immunological tests in vitro showed CPP could stimulate concanavalin A (ConA)- or lipopolysaccharide (LPS)-induced lymphocyte proliferation in a dose-dependent manner. 相似文献

Microsatellite-containing regions were isolated and characterized in Przewalskia tangutica Maxim (Solanaceae), an endemic and endangered species to the Qinghai-Tibetan Plateau of China. An enrichment protocol yielded
200 positive clones. We designed primers to amplify 29 unique microsatellites, 12 of which amplified cleanly and were polymorphic.
A survey of 17 individuals showed that these loci are highly variable with the number of alleles ranging from 3 to 12, and
expected heterozygosity ranged from 0.2929 to 0.4947. Those markers will be useful for studies of population structure and

In the present study, 10 polymorphic microsatellite DNA loci from Atlantic halibut (Hippoglossus hippoglossus) were isolated and characterized. The number of alleles for these loci ranged from 2 to 4 in tested 24 individuals. Observed
and expected heterozygosities per locus varied from 0.21 to 0.70 and from 0.31 to 0.65, respectively. Most of these 10 microsatellite
loci were successfully amplified and showed polymorphic in five related species. These loci will be useful for the assessment
of genetic diversity and population structure of Atlantic halibut.

The intermediate horseshoe bat (Rhinolophus affinis) is a widespread species in Southeast Asia. We developed 19 novel microsatellite loci from an enriched genomic library of
the bat, and tested their polymorphism using a single population from Guangdong province, southern China. The number of alleles
ranged from 3 to 15 per locus with the expected and observed heterozygosity ranging from 0.397 to 0.920 and 0.280–0.926, respectively.
Three markers significantly deviated from the Hardy-Weinberg expectations after Bonferroni correction and no linkage disequilibrium
was detected in any of loci. These microsatellite loci will be useful in studying the phylogeography of this species. 相似文献

Panax ginseng C.A. Meyer, commonly known as Korean or Asian ginseng, is a perennial herb native to Korea and China. Its roots are highly
prized for several medicinal properties. The present study describes development and characterization of twenty-two polymorphic
microsatellite markers for this species. A total of 99 alleles were detected with an average of 4.5 alleles per locus across
20 accessions. Values for observed (H
O
) and expected (H
E
) heterozygosities ranged from 0.05 to 1.00 and from 0.18 to 0.73, respectively. Eleven loci deviated from Hardy–Weinberg
equilibrium (P < 0.001). Significant (P < 0.05) heterozygote deficiency was observed at 13 loci. Exact test for linkage disequilibrium showed significant values
(P < 0.05) between 12 pairs of loci. These microsatellite markers provide powerful tools for understanding population and conservation

Artemisia annua is an important medicinal plant from which Artemisinin was extracted to cure malaria effectively. We developed eight microsatellite
markers from the genome of A. annua using the FIASCO protocol. Polymorphism of each locus was assessed in 54 individuals from two Chinese populations. The average
allele number of these microsatellites was 3.1 per locus, ranging from 2 to 6. The observed (H
O) and expected (H
E) heterozygosity were from 0.019 to 0.907 and from 0.055 to 0.793, respectively. These microsatellite markers would provide
a useful tool for genetic studies of A. annua.

Cupressus chenggiana S. Y. Hu (Cupressaceae) is an endemic and endangered conifer species in southwest China. In order to study the population
genetics and design the effective conservation methods, we aimed to develop microsatellite primers for this species in the
present study. We developed eight new microsatellite loci for this species through biotin capture method. Polymorphism of
each locus was further assessed in 18 individuals from three geographically distant populations. The number of alleles per
locus ranged from 6 to 11 with an average of 8.13. The observed and expected heterozygosities ranged from 0.219 to 0.296 and
from 0.374 to 0.470, with averages of 0.254 and 0.417, respectively. We further found that three of nine microsatellite loci
developed previously for another congeneric species showed polymorphic banding patters. We performed primer-crossing tests
of these loci in the other two congeneric species which are closely related to C. chenggiana (C. gigantea and C. duclouxiana). These microsatellite markers would be effective for analyzing genetic diversity and population genetic structure of this
species and its morphological differentiation with the close relatives. 相似文献

Scutellaria baicalensis is a popular medicinal plant that is on the verge of extinction due to uncontrolled harvesting, habitat destruction and deterioration of its ecosystem. We isolated and characterised 21 microsatellite loci in this species. Ninety-four individuals from six populations were used to test the polymorphism of the microsatellite loci. The number of alleles per locus ranged from 1 to 13, with a mean of 7.2. Observed and expected heterozygosities varied from 0.000 to 1.000 and 0.000 to 0.938, respectively. Among these new microsatellite markers, only two loci showed significant deviation from Hardy–Weinberg equilibrium. No locus pairs showed significant linkage disequilibrium. The 21 primer pairs were tested in other Scutellaria species. Most of these primer pairs worked successfully, except for Scut18. These new microsatellite markers could be applied to investigate the genetic diversity and population genetic structure of S. baicalensis and its closely related species. 相似文献

Loci were screened in 12 individuals of both T. polytmus and its sister species T. scitulus, also a Jamaican endemic. Microsatellites are often highly variable and abundant in most complex genomes, therefore are widely used in population genetic
studies. In this study, twelve polymorphic microsatellite loci were isolated and characterized for the Achnatherum inebrians, a plant abundant in grasslands of Northwest China. Characterization of 24 A. inebrians individuals form four geographically distant populations (Gansu, Qinghai, Xinjiang and Inner Mongolia provinces) showed moderate
to high allelic diversity ranging from 3 to 13 alleles per locus, and the expected heterozygosity ranging from 0.41 to 0.67.
No evidence of linkage disequilibrium was found for any locus pairwise comparisons. The markers described here will be useful
for investigating the genetic diversity, genetic structure and gene flow of this species.

Candidia barbata is an endemic cyprinid fish in Taiwan and suffers habitat destruction caused by human overexploitation. In this study, a
total of 14 polymorphic microsatellite loci from C. barbata were isolated and characterized using an optimized protocol to construct a microsatellite-enriched genomic library. The analysis
of variability was performed in 30 specimens of Taiwan. The mean number of alleles across loci was 4.92 ± 1.44. The levels
of expected and observed heterozygosities varied from 0.1266 to 0.5079, and from 0.0667 to 0.9667, respectively. Frequencies
of null alleles of the 14 loci are not significantly greater than zero. No linkage disequilibrium was detected between loci
