A study was made of the functional significance of GABA-ergic structures of the substantia nigra (SN) and the caudate nucleus (CN) and their role in food-procuring behaviour of cats. Analysis was made of behavioral and EEG-effects of local GABA and the GABA antagonist, picrotoxin, microinjections into the studied brain structures. Stimulation of the GABA-ergic structures of the SN produced a sedative effect and depression of the cat food-procuring behaviour. Effects of stimulation of the CN GABA-ergic structures were to a great degree reverse. The conclusion has been made that GABA-ergic structures of the SN and the CN play different roles in controlling the CN inhibitory influence upon food-procuring behaviour.  相似文献

Using halothane-anaesthetized cats implanted with push-pull cannulae, we investigated the effects of GABA application into the VM/VL on the release of [3H] DA continuously synthetized from [3H] tyrosine in the ipsilateral CN and SN and on single unit activity of nigral DA cells. GABA was applied (30 min) at a concentration of 10(-3) or 10(-5) M since the higher concentration reduces the local multi-unit activity in the VM/VL while the opposite response is observed with the lower one. The application of GABA into the VM/VL at a concentration of 10(-3) M resulted in an increase in nigral [3H] DA release, an inhibition of DA cell firing and a decrease in [3H] DA release in the CN. The latter effect is likely due to the inhibition of DA neuron activity which is triggered through DA autoreceptors by DA released from dendrites. In contrast, when applied at a concentration of 10(-5) M into the VM/VL, GABA stimulated [3H] DA release in the CN despite its inhibitory effect on single unit activity of DA cells. Furthermore, the nigral release of [3H] DA was no longer affected. These results indicated that DA release from nerve terminals was not dependent on nerve activity and they favour the existence of a potent facilitatory presynaptic regulation of DA release. The intervention of a presynaptic mechanism was further established by examining the effect of GABA (10(-5) M) application into the VM/VL on [3H] DA release in the CN shortly after a complete ipsilateral hemisection of the brain made at the meso-diencephalic level.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Microiontophoretic studies using cats anesthetized with alpha-chloralose were performed to determine whether or not dopamine D-1 and D-2 receptors co-exist in the same caudate nucleus (CN) neurons that receive inputs from the substantia nigra (SN), and in which spikes elicited by SN stimulation were blocked by domperidone, a selective D-2 antagonist. Iontophoretic application of dopamine produced a dose-dependent inhibition of spontaneous firing in 2 of 4 spontaneously active CN neurons and an increase in firing in the remaining 2 neurons. However, dopamine inhibited the glutamate-induced firing in 31 of 32 CN neurons that were not spontaneously active. Similar inhibition with iontophoretically applied SKF 38393, a selective D-1 agonist, was observed in 33 of 34 spontaneously inactive neurons tested. When the effects of dopamine, SKF 38393 and bromocriptine (D-2 agonist) were examined on the same CN neurons, the inhibitory effects of both dopamine and SKF 38393 were seen in 14 of 15 neurons, and both an inhibition by SKF 38393 and an excitation by bromocriptine were observed in 15 of 17 neurons. The inhibitory effects of dopamine and SKF 38393 were antagonized by haloperidol and SCH 23390 (D-1 antagonist) without being affected by domperidone. Furthermore, the dopamine-induced inhibition was converted to an excitation during simultaneous application of SCH 23390 in 6 of 10 CN neurons, and this excitation was antagonized by domperidone. These results strongly suggest that the inhibitory D-1 and excitatory D-2 receptors co-exist on the same CN neurons receiving inputs from the SN.  相似文献

The feeding behaviour of the land snails Achatina fulica was used for investigation of the effects of different doses of ethanol. The time from the beginning of food (carrot) exposure till the removal of the carrot from the needle was measured. Regression analysis revealed the opportunity of approximation with exponential mathematical model of feeding behaviour time progressive decrease during food exposure. 1% ethanol solution injected into cephalopedal sinus inhibited the feeding behaviour to a small extent whereas 5% and 10% ethanol led to the facilitation of this behaviour. The increase of ethanol concentration from 20% to 40% in injected solution enhanced the inhibitory effect. The feeding behaviour is proposed to be employed as a model for investigation of ethanol neuronal effects.  相似文献

The effects were determined of dietary fish oil on the polyunsaturated fatty acid desaturation in rats fed on fish oil-containing diets (FS group) and on non-fish oil diets (CN group) during the fetal to postnatal periods. Although the desaturase activity in the liver microsomes of the FS group was higher than that of the CN group before birth, this was not altered by dietary fish oil after birth. However, a lower 20:4n-6 concentration before and after birth, and lower linoleic acid desaturation index ((dihomo-γ-linolenic acid + arachidonic acid)/linoleic acid)) at 10 wk of age in the FS group than in the CN group were observed in the liver microsomal phospholipids. The Δ6-desaturase activity in the brain microsomes of the FS group was lower than that of the CN group. These findings suggest that an intake of dietary fish oil by dams and postnatal rats affected the arachidonic acid concentration due to the decreased desaturase activity in the rats’ microsomes.  相似文献

We examined the effects of fermented bean pastes derived from bean vinegar by-products on serum cholesterol in rats. The rats were fed boiled paste from adzuki (A), kintoki (K), or tebou (T), or fermented paste from adzuki (AP), kintoki (KP), or tebou (TP) for 4 weeks. The serum non-high-density lipoprotein (HDL) cholesterol levels in all the experimental groups, except for A group, were significantly lower than in the control (CN) group. Likewise, the serum triglyceride levels in K and all the fermented bean groups were significantly lower than in the CN group. The levels of hepatic 3-hydroxy-3-methylglutaryl-CoA (HMG-CoA) reductase mRNA in all the experimental groups except for A were significantly lower than in the CN group. These findings indicate that fermented bean pastes also suppress cholesterol synthesis, resulting in a reduced serum cholesterol concentration. These effects might be related not only to the resistant starch but also to the protein or peptide in the fermented bean paste.  相似文献

The thermal and rheological properties of spray-dried, ethanol-precipitated, purified, and deacetylated spruce galactoglucomannans (GGM) were investigated by rheological measurements and differential scanning calorimetry. The shear rate dependence of viscosity and the effects of the drying method, temperature, ionic strength, and deacetylation on rheological properties were studied. GGM solutions exhibited a shear thinning behaviour. GGM solutions did not obey the Cox–Merz rule. The storage modulus of GGM solutions increased with an increase in concentration; gradually until a concentration of 5%, but rapidly at higher concentrations. Ethanol-precipitated GGM solutions showed a more elastic behaviour than spray-dried GGM solutions. Deacetylation caused an increase in apparent viscosity and more significantly in storage modulus. The storage modulus increased slightly with a decrease in temperature. A small amount addition of NaCl slightly changed the oscillatory behaviour. The effects of above factors were discussed in terms of molecular interactions. The rheological measurements of GGM solutions provide the basis of functionalities of GGM solutions.  相似文献

The purpose of this investigation is to test the feasibility of a nitroxide regeneration system involving liposomes as an approach toward solving the "reduction problem" when nitroxides are used as contrast enhancing agents in MRI applications. It is shown that the inclusion of an entrapped oxidant (K3Fe(CN)6) in the aqueous compartment of nitroxide-doped liposomes causes a 4-5-fold increase in the duration of the nitroxide ESR signal in the presence of the external reductant sodium ascorbate. Confirmation was obtained by monitoring the concentration of the internalized Fe(CN)6(3-) ion versus time by visible spectroscopy at 410 nm. Trans bilayer (flip-flop) motion of the long chain nitroxide ester is the likely mechanism of this nitroxide regeneration system.  相似文献

盆栽条件下,采用单接种Suillus bovines(以下简称SB)和混合接种(Suillus luteus、Suillus grevillea、Tricholoma fulvum、Boletinus grisellus、Suillus bovinus、Leucocortinarius bulbiger、Rhizopogon luteolus、Pisolithus tinctorius 8个菌种的等量混合菌剂接种,以下简称HJ)方法对樟子松(Pinus sylvestris var.mongolica)2个月幼苗进行接种处理,幼苗生长6个月形成菌根后对樟子松幼苗进行模拟氮沉降实验,5个月后通过测定幼苗生长指标、根系形态结构和菌根侵染率,旨在探讨模拟氮沉降对樟子松不同接种处理幼苗生长的影响。结果表明:①随着氮浓度的增加,单接种乳牛肝(SB)和混合接种(HJ)菌根侵染率均呈下降趋势,且氮沉降对SB处理的菌根侵染率影响更显著。②接种显著促进幼苗地上和地下生物量积累,且混合接菌效果更好;同一接菌处理中,氮浓度增加对地上生物量的积累有抑制作用;接种和氮浓度对地径和地上生物量有显著交互作用,但对苗高和地下生物量无交互作用。③HJ处理中不施氮(CKN)和1倍氮浓度(CN)较不接菌(CK)和SB处理显著促进幼苗根系的平均直径、总根长和总表面积;氮浓度增加显著促进CK和SB处理的幼苗根系平均直径、总根长和总表面积,但是对HJ处理的根系各形态结构指标均产生抑制作用。接种和氮浓度以及两者的交互作用均极显著影响幼苗根系形态结构。  相似文献

The incorporation and electrochemical behaviour of cytochrome c (Cyt C) at glassy carbon electrodes modified with the polyestersulfonated ionomer Eastman AQ 55 are examined. The presence of the polyelectrolytic coating allows the preconcentration of the protein within the polymer and the observation of the direct electrochemistry of Cyt C at the modified electrode without addition of promoters or mediators in the solutions. The dependence of voltammetric signals on typical parameters such as solution pH and nature or concentration of the supporting electrolyte supports the ion-exchange nature of the incorporation process. The relevant role of the permselectivity of the polymeric modifier is highlighted also by the study of electrocatalytic processes which take place at the modified electrode loaded with Cyt C. No electrocatalytic effect is observed when the electrogenerated dication (ferricenylmethyl)trimethylammonium is present as possible oxidant. On the contrary, electrocatalytic current enhancements are observed for anionic substrates such as Fe(CN)(6)(3-) (oxidant) and ascorbate (reductant). Catalytic currents increase with the substrate concentration, with higher sensitivity for Fe(CN)(6)(3-). Due to ionic repulsion, the reaction with anions occurs at the polymer-solution interface. In the case of chemically unstable substrates, such as superoxide anion, ionic repulsion slow down the approach rate of the substrate so that spontaneous decomposition can prevail over the reaction with Cyt C incorporated in the coating.  相似文献

Calcineurin (CN) is thought to play an important role in the immune system by regulating cytokine production, for example, interleukin-2 (IL-2) in T-lymphocytes. We have previously shown that physiological concentrations of Zn2+ inhibit CN activity in vitro [K. Takahashi, E. Akaishi, Y. Abe, R. Ishikawa, S. Tanaka, K. Hosaka, Y. Kubohara, Zinc inhibits calcineurin activity in vitro by competing with nickel, Biochem. Biophys. Res. Commun. 307 (2003) 64-68], in spite of the fact that Zn2+ is an essential element of the CN catalytic domain. In this study, in order to assess whether Zn2+ regulates (suppresses) CN activity in vivo and whether Zn2+ can be used as an anti-inflammatory and/or immunosuppressive drug, we examined the effects of Zn2+ on IL-2 production induced by the mitogen, concanavalin A (ConA), in Jurkat T-cells. Zn2+ at 0.2 mM suppressed ConA-induced IL-2 accumulation in the medium of an in vitro culture of Jurkat cells. Zn2+ at 0.03-0.3 mM dose-dependently suppressed ConA-induced IL-2 mRNA expression in Jurkat cells. Zn2+ also suppressed IL-2 mRNA expression induced by phorbol ester (PMA) and ionomycin. Furthermore, Zn2+ and the immunosuppressant FK506 showed an additive inhibitory effect on ConA-induced IL-2 mRNA expression. These results suggest that exogenously added Zn2+ may disturb (increase) the intracellular Zn2+ concentration and inhibit CN activity, thereby suppressing IL-2 production in Jurkat cells. The present study further indicates that Zn2+ may have therapeutic potential in the treatment of T-cell related inflammation and also that Zn2+ may be utilized as a supplemental drug with FK506.  相似文献

FMRFamide and the related tetrapeptide FLRFamide are highly excitatory in molluscan non-cardiac smooth muscle. They are also exceptionally excitatory in the atrium and internally perfused ventricle of Busycon canaliculatum. These two peptides, usually thought of as classic molluscan cardio-acceleratory agents are in fact simply two members of a large and ever growing superfamily, the RFamide family, whose phylogenetic distribution has been so elegantly mapped by Walker. Members of this family, often with extended peptide chains (e.g. penta, hepta and decapeptides), stretch in their known distribution from the cnidaria to the chordates. The effects of some of the members of this superfamily (FMRFamide. FLRFamide, YMRFamide, TNRNFLRFamide, SDPFLRFamide, LMS) were examined. The neuropeptides were found to be very potent at very low concentrations (10(-9) M) in the ventricle of both Buccinium and Busycon. Other neuropeptides (HFMRdFamide, SCPb, NLERFamide and pEGRFamide) were found to be without any effect. The Ca2+ dependency of these neuropeptides was also tested. The peptides appear to induce contraction of the ventricles by release of Ca2+ from internal pools. The neuropeptides appear to stimulate contraction in these cardiac muscles through a completely different pathway to Serotonin (the main excitatory neurotransmitter for the cardiac muscle). When the peptides were applied together with Serotonin an additive effect was observed clearly indicating the release of Ca2+ through different pathways. The nature of the RFamide receptor was also tested. It appears that the RFamide neuropeptides mobilize the 2nd messenger IP3 (Inositol trisphosphate), since the IP3 blocker Neomycin Sulphate inhibited the response of the neuropeptides.  相似文献

The potential for in situ bioremediation of cyanide (CN) and nitrate (NO3) contamination within the extracted ore (residue pile) and downgradient groundwater at a CN heap leach mine in New Mexico was assessed through the following steps: (1) identification of the relative abundance of CN‐degrading microorganisms in the contaminated residue pile, (2) identification of amendments to enhance aerobic CN degradation and assimilatory NO3 reduction and determination of the optimal carbon‐to‐nitrogen concentration for degradation, (3) assessment of optimum amendment's influence on biodégradation of CN and NO3 by experiments with large‐scale columns filled with residue pile material, and (4) evaluation of the potential for other adverse environmental effects, specifically acid rock drainage, due to application of the amendment. These investigations determined that application of a reduced carbon source (sucrose) significantly increased the rate of CN degradation and NO3 immobilization without increasing the probability of acid rock drainage.  相似文献

Insulin resistance leads to memory impairment. Cinnamon (CN) improves peripheral insulin resistance but its effects in the brain are not known. Changes in behavior, insulin signaling and Alzheimer-associated mRNA expression in the brain were measured in male Wistar rats fed a high fat/high fructose (HF/HFr) diet to induce insulin resistance, with or without CN, for 12 weeks. There was a decrease in insulin sensitivity associated with the HF/HFr diet that was reversed by CN. The CN fed rats were more active in a Y maze test than rats fed the control and HF/HFr diets. The HF/HFr diet fed rats showed greater anxiety in an elevated plus maze test that was lessened by feeding CN. The HF/HFr diet also led to a down regulation of the mRNA coding for GLUT1 and GLUT3 that was reversed by CN in the hippocampus and cortex. There were increases in Insr, Irs1 and Irs2 mRNA in the hippocampus and cortex due to the HF/HFr diet that were not reversed by CN. Increased peripheral insulin sensitivity was also associated with increased glycogen synthase in both hippocampus and cortex in the control and HF/HFr diet animals fed CN. The HF/HFr diet induced increases in mRNA associated with Alzheimers including PTEN, Tau and amyloid precursor protein (App) were also alleviated by CN. In conclusion, these data suggest that the negative effects of a HF/HFr diet on behavior, brain insulin signaling and Alzheimer-associated changes were alleviated by CN suggesting that neuroprotective effects of CN are associated with improved whole body insulin sensitivity and related changes in the brain.  相似文献

The effects of carbon monoxide and cyanide on the electric potentialdifference between both cut ends of a hypocotyl segment excisedfrom a bean embryo were investigated. Carbon monoxide definitely diminished the PD. This inhibitionwas photoreversible. The inhibition ratio was quantitativelyrelated to the [CO]/[O2] ratio of the gas mixture. Prolongedexposure to a CO gas mixture induced spontaneous recovery ofthe PD, and this CO-resistant electrogenic activity was sensitiveto CN and also disappeared under anoxia. Cyanide also diminished the PD and often induced oscillationof the potential. The type of oscillation and the inhibitionratio varied with the concentration of CN. Sometimes CN pretreatmentcaused changes in the anoxia response of the potential. The time course of the spontaneous change in PD after cuttingof the hypocotylsegment also was investigated. A conclusion on relationships of the septum with both the antidiuretic and antinatriuretic systems in the regulation of the aqueous-electrolytic balance and renal function has been drawn as a result of the study of the drinking behaviour and renal reaction after the destruction of the lateral septal nucleus (LSN). This conclusion is based on the revealed increase in the urinary excretion of sodium, osmotically active substances, creatinine, titrating acids and ammonia after the LSN destruction. Moreover, an increase of the vasopressin level in the blood plasma, a decrease of aldosterone concentration as well as an activation of the drinking behaviour are noted.  相似文献

Reduction of oscillations in population size is of fundamental importance to both theoretical and applied ecology. Spatial variability in population rates among different habitat regions is known to be an important mechanism that inhibits oscillations in population size. In the current study we used an individual-based model to simulate a single population of animals whose individual members are sensitive to competition only within their vicinity (i.e., within their competition neighborhood, CN). Our model extends previous studies by exploring how local interactions reduce population oscillations in competitive systems of animals, rather than in systems of plants. Our simulations explored the effects of animal mobility and interaction range separately on population dynamics. In our model, a decrease in CN dimensions tended to reduce population oscillations at all tested animal movement speeds. Yet, movement speed affected animal distribution patterns; an increase in movement speed led to more random distributions. We also found that mean population size was affected more by CN dimensions at lower mobility levels than when it was high.  相似文献

The effects of two types of mushroom (Agaricus bisporus; white, WM; brown, BM) powders on intestinal fermentation in rats were investigated in terms of the physical characteristics of animals and by bacterial and HPLC analyses of cecal contents. Short-chain fatty acid levels were found to be significantly higher in the WM group than in the BM and the control (CN) groups; coliform bacteria levels in the BM group were significantly lower than those in the CN group, with the WM group inducing an apparent but insignificant decrease in coliforms. Anaerobe levels in the WM group were significantly higher than those in the CN group and, compared with the CN group, the BM and WM groups exhibited significantly increased feces weight and cecum weight, respectively. These results indicate that the mushroom powders, and in particular the WM powder, have beneficial effects on the intestinal environment in rats.  相似文献

The titration of sonicated vesicles of egg phosphatidylcholine with ferricyanide in the presence of Ca2+ results in the formation of aggregates. The turbidity increase caused by these aggregates cannot be reversed by EDTA treatment. In addition, no rearrangement of the bilayer structure has been found in this process, either measuring leakage of vesicle content or exchange of lipids among the bilayers themselves. The aggregation is dependent on the Ca2+ content of the vesicles, the outer Ca2+ and Fe(CN)3-(6) concentration and the order of addition of Ca2+ and ferricyanide. The results can be explained by a specific adsorption of Fe(CN)3-(6) to bilayers of sonicated vesicles, in contrast to other multivalent anions. In contrast to the stability found with sonicated vesicles, the aggregation causes a leakage of the internal solution when multilamellar liposomes are titrated with Fe(CN)3-(6).  相似文献

There is now conclusive evidence for the presence of insulin and insulin receptors in the mammalian CNS and it has been postulated that they can modulate peripheral glucose homeostasis. Since a number of central neurotransmitters are also known to influence glucose levels and it is likely that CNS insulin receptors act through neurotransmitter mediation, the present study was conducted to investigate the effect of intracerebroventricularly (icv) administered insulin on rat brain dopamine (DA), noradrenaline (NA), serotonin and acetylcholine (ACh) activity in normal and alloxan-induced hyperglycaemic animals. Insulin was administered in doses (50 and 100 microU) which induced minimal hypoglycaemia, so as to obviate the likely effects of hypoglycaemia on neurotransmitter function. DA was estimated in midbrain-diencephalon (MD) and caudate nucleus (CN), NA and serotonin in MD and pons-medulla (PM), while ACh was estimated in all the three areas, namely, MD, CN and PM. The regional brain concentrations of DA, NA and serotonin were more in the hyperglycaemic rats as compared to their euglycaemic counterparts. However, the reverse was noted in case of ACh. Insulin induced a decrease in rat brain DA and NA levels, which was more marked in the hyperglycaemic animals. Conversely, insulin induced an increase in rat brain serotonin concentration which was not significantly different in normal and hyperglycaemic rats. Insulin induced marked increase in rat brain ACh levels, which was accentuated in hyperglycaemic animals. The present study reports for the first time the likely interaction between CNS insulin receptors and brain monoamines, and ACh, in euglycaemic and hyperglycaemic states.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
