The formation of large aggregates by ionic interactions between acidic glucosaminoglycans and cationic secretory proteins has been proposed as one of the critical steps in the concentration process in the condensing vacuoles of secretory cells. In this paper, this hypothesis was tested by studies on the interactions between bovine chymotrypsinogen A and chondroitin sulfate as a simplified model. Small amounts of chondroitin sulfate were found able to induce chymotrypsinogen precipitation. Like zymogen granules, the resulting aggregates were moderately sensitive to ionic strength and insensitive to osmolality. Moreover, their pH dependence was similar to that of isolated zymogen granules. When sulfated glucosaminoglycans isolated from the zymogen granules of the guinea pig pancreas were used instead of chondroitin sulfate, the same kind of interactions with chymotrypsinogen were obtained. Our data support the hypothesis that the strong ionic interactions between those sulfated glucosaminoglycans and cationic proteins could be responsible for the concentration process.  相似文献

研究了黄土高原马栏林区主要植被类型中的辽东栎幼苗的数量特征,更新层幼苗、幼树的实生和萌生特性及其辽东栎在垂直结构上的数量分布。结果表明:(1)辽东栎幼苗在黄土高原马栏林区分布广泛且数量充足。不同的植被类型中辽东栎种群表现出不同的大小级结构,在油松—辽东栎混交林、油松林和辽东栎林中辽东栎种群的幼苗、幼树和成树均占一定的比例,而在油松—白桦混交林、白桦林、山杨林和山杨—白桦混交林中辽东栎种群则以幼苗和幼树为主。表明辽东栎种群在该地区植被的发展过程中将产生重要的作用。(2)辽东栎在这一地区是由实生和萌生的个体混合组成的。在各种植被类型中实生植株的密度都高于萌生植株,辽东栎种群的更新在该地区可能主要是通过实生植株来完成的,即辽东栎实生植株在更新过程中起重要的作用。但萌生植株作为辽东栎顺利通过瓶颈的一种手段,作为辽东栎种群繁衍和稳定的一种途径,在辽东栎种群的更新中也起着一定的作用。  相似文献

In this work, we analysed histochemical, biochemical and functional modifications of the glomerular basement membrane (GBM), occurring for aging, in the Rat. The results suggest an increase of collagenous components and a decrease of sulfated glucosaminoglycans as a function of age. In other respects, fixed anionic sites of the GBM, disclosed by colloidal iron, are almost exclusively restricted to the laminae rarae in one month-old rats, whereas the marker appears randomly scattered among the lamina densa in 12 month-old animals. These changes could be the cause of increased permeability of the GBM during aging.  相似文献

Changes in glycosaminoglycans (GAGs) were investigated in relation to cell density, growth and transformation of human fibroblasts. Relative amounts (percentages of the total GAGs) of heparan sulfate (HS) increased and those of hyaluronic acid (HA) decreased in growth-reduced (serum-starved, exogenous HS-treated and dense) cultures of normal (WI-38) cells. In contrast, transformed (WI-38 CT-1) cells exerted such GAG changes only in serum-starved cultures, but not in HS-treated or dense cultures. These results indicate that the changes in glucosaminoglycans (G1cAGs) (HS and HA) is coupled exclusively with cell growth.  相似文献

The incidence of coiled-sprout was determined in Scotch and local-grown Arran Pilot and Duke of York seed tubers which had been stored at 10°, 4° and 15° C. and in a farm store with no temperature control. All four tuber types were planted in the field and, in addition, the two types of Duke of York were planted in Perlite at 7°, 10° and 15° C. In the field, and when maintained at 7° and 10° C, the percentage of sprouts coiling and the intensity of coiling was greater in tubers stored at 10° and 15° C. than at 4° C. There was no coiling when the Duke of York tubers were planted at 15° C. In a further experiment tubers were stored at 20° C. in the light and dark and samples were planted monthly for 3 months at temperatures of 7°, 10° and 15° C. During the following 3-month period only light-stored were planted because of the excessive amount of tip-death in the tubers stored in the dark. There was very little coiling in the dark-stored tubers. In the first two plantings of the light-stored tubers there was virtually no coiling of those planted at 15° C. There was some, however, at 7° and 10° C. In subsequent plantings there was more coiling and no effect of planting temperature. Attempts to isolate Verticiculum nubilum from sprouts were successful in only a small percentage of attempts and it was not possible to demonstrate any difference between its distribution on coiled and normal sprouts. It was not possible to induce coiling by infection of sprouts with spores of V. nubilum. Over a wide range of sprout sizes the amount of coiling was a function of sprout size at planting. However, the parts which coiled were those in the apical bud of the sprout at the time of planting and hence contributed only a small amount to the total sprout size. It is likely, therefore, that the correlation between coiling and sprout size reflects the changing metabolism of the elongating regions of sprouts with their increase in length, these regions developing in such a way as to produce a greater tendency to coiling. The internal reactions concerned in these changes, however, are not known.  相似文献

Blood vessels are essential conduits of nutrients and oxygen throughout the body. The formation of these vessels involves angiogenic sprouting, a complex process entailing highly integrated cell behaviors and signaling pathways. In this review, we discuss how endothelial cells initiate a vessel sprout through interactions with their environment and with one another, particularly through lateral inhibition. We review the composition of the local environment, which contains an initial set of guidance cues to facilitate the proper outward migration of the sprout as it emerges from a parent vessel. The long-range guidance and sprout stability cues provided by soluble molecules, extracellular matrix components, and interactions with other cell types are also discussed. We also examine emerging evidence for mechanisms that govern sprout fusion with its target and lumen formation.  相似文献

景宁玉兰是萌生能力很强的灌木树种,为了解不同生境下景宁玉兰萌枝形态及其生殖特征,选择了灌丛(SH)、黄山松林(PTF)、落叶阔叶林(DBF)、杉木林(CLF)和林缘(FE)等5种坡向一致的生境类型,通过典型样地调查,比较分析了5种不同生境下景宁玉兰萌枝数量、大小、高度、枝系、开花及结实等性状。结果表明:(1)景宁玉兰在黄山松林下的分布密度最高,但5种生境下景宁玉兰每丛的萌枝数量没有差异,每丛最大萌枝基径对每丛萌枝数有一定影响,但最大萌枝高度与每丛萌枝数没有关系。在所有调查的景宁玉兰居群中,大于2根以上萌枝的灌丛达82.5%,说明萌生更新在种群繁衍中发挥着重要作用。(2)在落叶阔叶林下的萌枝基径显著小于其他生境(P0.05),黄山松林下一级枝粗度和长度显著小于其他生境(P0.05);灌丛中的总分枝率和逐步分枝率最高,灌丛和林缘生境的逐步分枝率(SBR2:3)高于其他生境。(3)生殖萌枝基径(r=0.320,P0.05)和高度(r=0.349,P0.05)与花量均呈显著正相关关系。杉木林下景宁玉兰生殖萌枝的花量显著高于黄山松林、落叶阔叶林和林缘生境(P0.05)。虽然黄山松林下景宁玉兰萌枝开花率及开花萌枝比例最低,但其果实大小、单果种子粒数及每丛结实率却较高(P0.05),(4)景宁玉兰对环境变化极为敏感,生境类型和海拔对萌枝形态和生殖性状均有一定影响。研究表明,景宁玉兰萌生特征主要受其内在生物学特性所控制,而萌枝形态及生殖特征则与其所处环境条件更为密切。  相似文献

针对大兴安岭地区塔河林业局不同林型下,不同火烧强度的火烧迹地的森林植被更新及恢复情况进行了调查研究。结果表明:(1)针叶林过火迹地上,落叶松幼苗较少,阔叶树萌条更新强度同火烧程度成正比;火烧前生长有白桦、赤杨的林地上,火烧后阔叶树成为主导树种,林相完全发生变化。(2)重度火烧下有利于天然更新,促使白桦、山杨萌生。中度火烧最初形成以杨桦为优势树种的阔叶林,后逐渐演变为针阔混交林。轻度火烧有利于针叶林的更新。(3)火烧后不同林型下灌木草本植被种类变化不同。从盖度方面来看,杜鹃落叶松林型中林下灌木草本更新最为良好。  相似文献

Recent cDNA cloning of the glycosyltransferases involved in the synthesis of the sulfated glycosaminoglycan sidechains of proteoglycans has provided important clues to answering long-standing questions concerning the mechanisms of both chain polymerization and the biosynthetic sorting of glucosaminoglycans (heparin/heparan sulfate) and galactosaminoglycans (chondroitin/dermatan sulfate). Recovery from hematopoietic aplasia is a predominant factor in the survival of total-body-irradiated mice within 30 days after exposure. However, other radiation-induced pathophysiological events have been shown to play a role, among which an inflammatory reaction must be considered. In the present study, we evaluated the therapeutic potential of a hematopoietic growth factor (thrombopoietin, Tpo) and pleiotropic cytokines (Il4 or Il11), used alone or in combination, on the survival of mice, hematopoietic reconstitution, inflammatory reaction and vascular changes. All treatments including Tpo induced a higher level of survival than did treatment with a placebo, with combinations being the most efficient. The increased survival could not be explained solely by an improved hematopoietic recovery. Treatments with Tpo also reduced the level of the chemokine KC in plasma and the level of expression of mRNA for inflammatory and coagulation proteins in the lungs of irradiated mice. In addition, radiation- induced vascular hyperpermeability was reduced with the use of Tpo. In summary, our results show that Tpo may improve survival by limiting vascular leakage, which in turn could limit inflammatory reactions and the ensuing tissue damage.  相似文献

The tetracycline (tet)-regulated expression system allows for the inducible overexpression of protein-coding genes, or inducible gene knockdown based on expression of short hairpin RNAs (shRNAs). The system is widely used in mice, however it requires robust expression of a tet transactivator protein (tTA or rtTA) in the cell type of interest. Here we used an in vivo tet-regulated fluorescent reporter approach to characterise inducible gene/shRNA expression across a range of hematopoietic cell types of several commonly used transgenic tet transactivator mouse strains. We find that even in strains where the tet transactivator is expressed from a nominally ubiquitous promoter, the efficiency of tet-regulated expression can be highly variable between hematopoietic lineages and between differentiation stages within a lineage. In some cases tet-regulated reporter expression differs markedly between cells within a discrete, immunophenotypically defined population, suggesting mosaic transactivator expression. A recently developed CAG-rtTA3 transgenic mouse displays intense and efficient reporter expression in most blood cell types, establishing this strain as a highly effective tool for probing hematopoietic development and disease. These findings have important implications for interpreting tet-regulated hematopoietic phenotypes in mice, and identify mouse strains that provide optimal tet-regulated expression in particular hematopoietic progenitor cell types and mature blood lineages.  相似文献

The time taken from reaping coconut seednuts to the appearanceof the sprout through the husk distinguishes late germinatingNiu kafa types from early germinating Niu vai types. Resultsfrom previously unrelated germination studies, carried out independentlyin Jamaica, Sri Lanka, Tanzania and Ivory Coast, show that thecommon tall varieties in those countries, and also in Beninand Mozambique, are Niu kafa types, whilst in peninsular Malaysiaand on the Pacific coast of Panama the common tall varietiesare Niu vai types. These findings agree with others based ondifferent aspects of the palm and its ecology. The relationship between germination and the degree of waterabsorption, particularly sea water, at the time the ripe fruitfalls from the palm may account for the genetic stability ofthe wild Niu kafa type despite island conditions which wouldappear to favour genetic drift. In this paper we have demonstrated that wheat sprout extract, which has been shown to be antimutagenic towards benzo[a]pyrene (BP), reduced formation of BP metabolites by hepatic microsomes of either benzo[a]pyrene- or phenobarbital-treated rats as analyzed in high-pressure liquid chromatography (HPLC). Comparing the time dependence of profiles and values of BP metabolites, formed in experiments in which the same dose of wheat sprout extract was added to the incubation medium, it has been observed that the later this extract was added the higher the percent of BP that was metabolized. In a bacterial test (cytochrome P450 induction assay) high inhibition of mutagenic activity of cyclophosphamide and ethidium bromide, in the presence of wheat sprout extract, reflected decreased levels of cytochromes P4502B1 and P4501A1 respectively. Decreased levels of both cytochromes P4501A1 and P4502B1 were also observed in either wheat sprout extract- or wheat sprout extract plus benzo[a]pyrene-treated rats. In all of these studies it has been observed that wheat sprout extract displays much more affinity for cytochrome P4501A1 than for the P4502B1 form. On the other hand the wheat sprout extract had higher affinity for carcinogen binding protein (4S protein) than for the aryl hydrocarbon receptor. The strong inhibition of BP mutagenicity and BP metabolism with non-chlorophyllic wheat sprout extract suggests that chlorophyll is not the main compound responsible for the antimutagenic activity of wheat sprout extract. The similar chromatographic behavior of both the main inhibitory fraction, obtained from wheat sprout extract, and two pure glycosides of apigenin--shaftoside, purified from wheat sprout extract and synthetic swertisine--suggests that antimutagenic compound(s) contained in the wheat sprout extract belong(s) to this family of flavonoids.  相似文献

Biglycan and decorin are two members of a family of small extracellular matrix proteoglycans characterized by the presence of 10 leucine-rich repeats and one or two attachment sites for glucosaminoglycans. Both have thus far been described only from tetrapod species, mainly mammals. Because the extracellular matrix has played an important part in the evolution of Metazoa, the phylogeny of its components is of considerable interest. In this study, biglycan-like (BGL) cDNA sequences have been obtained from two teleost (Oreochromis cichlid and zebrafish) and two lamprey species. The analysis of the sequences suggests that, like tetrapods, the lampreys possess two types of proteoglycans, both of which are biglycan-like; decorin-like proteoglycans could not be identified in these species. The genes specifying these two types apparently arose by duplication in the lamprey lineage after its divergence from gnathostomes. The two teleost species possess a BGL proteoglycan and a bona fide decorin. The BGL proteoglycan is highly divergent from the tetrapod biglycan and related to the BGL proteoglycans of the lamprey. Hence, although the duplication generating the ancestors of biglycan and decorin genes occurred after the divergence of agnathans but before the emergence of teleosts, only decorin acquired its characteristic properties in the bony fishes. The BGL gene presumably turned into a typical biglycan only in the tetrapod lineages. The presumed acquisitions of new functions appear to have been accompanied by changes in the evolutionary rate. Solid tumors are composed of cancerous cells and non-cancerous stroma. A better understanding of the tumor stroma could lead to new therapeutic applications. However, the exact compositions and functions of the tumor stroma are still largely unknown. Here, using a Lewis lung carcinoma implantation mouse model, we examined the hematopoietic compartments in tumor stroma and tumor-bearing mice. Different lineages of differentiated hematopoietic cells existed in tumor stroma with the percentage of myeloid cells increasing and the percentage of lymphoid and erythroid cells decreasing over time. Using bone marrow reconstitution analysis, we showed that the tumor stroma also contained functional hematopoietic stem cells. All hematopoietic cells in the tumor stroma originated from bone marrow. In the bone marrow and peripheral blood of tumor-bearing mice, myeloid populations increased and lymphoid and erythroid populations decreased and numbers of hematopoietic stem cells markedly increased with time. To investigate the function of hematopoietic cells in tumor stroma, we co-implanted various types of hematopoietic cells with cancer cells. We found that total hematopoietic cells in the tumor stroma promoted tumor development. Furthermore, the growth of the primary implanted Lewis lung carcinomas and their metastasis were significantly decreased in mice reconstituted with IGF type I receptor-deficient hematopoietic stem cells, indicating that IGF signaling in the hematopoietic tumor stroma supports tumor outgrowth. These results reveal that hematopoietic cells in the tumor stroma regulate tumor development and that tumor progression significantly alters the host hematopoietic compartment.  相似文献

Evidence for the presence of a [2Fe-2S] ferredoxin in bean sprouts   总被引:3,自引:0,他引:3  
An iron-sulfur protein with properties similar to those of ferredoxins found in the leaves of higher plants has been isolated from bean sprouts--a non-photosynthetic plant tissue. The bean sprout protein has a molecular mass of 12.5 kDa and appears to contain a single [2Fe-2S] cluster. The absorbance and circular dichroism spectra of the bean sprout protein resemble those of spinach leaf ferredoxin and the bean sprout protein can replace spinach ferredoxin as an electron donor for NADP+ reduction, nitrite reduction and thioredoxin reduction by spinach leaf enzymes. Although the reduced bean sprout protein (Em = -440 mV) is a slightly stronger reductant than spinach ferredoxin and appears to be less acidic than spinach ferredoxin, the two proteins are similar enough so that the bean sprout protein is recognized by an antibody raised against spinach ferredoxin.  相似文献
