Astacus leptodactylus is a decapod crustacean fully adapted to freshwater where it spends its entire life cycle after hatching under huge osmoconcentration differences between the hemolymph and surrounding freshwater. We investigated the expression of mRNA encoding one ion transport-related protein, Na+/K+-ATPase α-subunit, and one putative housekeeping gene, β-actin, during crayfish ontogenesis using quantitative real-time PCR. A 216-amino acid part of the open reading frame region of the cDNA coding for the Na+/K+-ATPase α-subunit was sequenced from total embryo, juvenile and adult gill tissues. The predicted amino acid sequence showed a high percentage similarity to those of other invertebrates (up to 95%) and vertebrates (up to 69%). β-actin expression exhibited modest changes through embryonic development and early post-embryonic stage. The Na+/K+-ATPase α-subunit gene was expressed in all studied stages from metanauplius to juvenile. Two peaks of expression were observed: one in young embryos at 25% of embryonic development (EI = 100 μm), and one in embryos just before hatching (at EI = 420 μm), continuing in the freshly hatched juveniles. The Na+/K+-ATPase expression profile during embryonic development is time-correlated with the occurrence of other features, including ontogenesis of excretory antennal glands and differentiation of gill ionocytes linked to hyperosmoregulation processes and therefore involved in freshwater adaptation.  相似文献

Evidences indicate the relationship between neurotensinergic and dopaminergic systems. Neurotensin inhibits synaptosomal membrane Na+, K+-ATPase activity, an effect blocked by SR 48692, antagonist for high affinity neurotensin receptor (NTS1) type. Assays of high affinity [3H]-ouabain binding (to analyze K+ site of Na+, K+-ATPase) show that in vitro addition of neurotensin decreases binding. Herein potential interaction between NTS1 receptor, dopaminergic D2 receptor and Na+, K+-ATPase was studied. To test the involvement of dopaminergic D2 receptors in [3H]-ouabain binding inhibition by neurotensin, Wistar rats were administered i.p.with antipsychotic drugs haloperidol (2 mg/kg) and clozapine (3, 10 and 30 mg/kg). Animals were sacrificed 18 h later, cerebral cortices harvested, membrane fractions prepared and high affinity [3H]-ouabain binding assayed in the absence or presence of neurotensin at a 10 micromolar concentration. No differences versus controls for basal binding or for binding inhibition by neurotensin were recorded, except after 10 mg/kg clozapine. Rats were administered with neurotensin (3, 10 y 30 μg, i.c.v.) and 60 min later, animals were sacrificed, cerebral cortices harvested and processed to obtain membrane fractions for high affinity [3H]-ouabain binding assays. Results showed a slight but statistically significant decrease in binding with the 30 μg neurotensin dose. To analyze the interaction between dopaminergic D2 and NTS1 receptors, [3H]-neurotensin binding to cortical membranes from rats injected with haloperidol (2 mg/kg, i.p.) or clozapine (10 mg/kg) was assayed. Saturation curves and Scatchard transformation showed that the only statistically significant change occurred in Bmax after haloperidol administration. Hill number was close to the unit in all cases. Results indicated that typical and atypical antipsychotic drugs differentially modulate the interaction between neurotensin and Na+, K+-ATPase. At the same time, support the notion of an interaction among dopaminergic and neurotensinergic systems and Na+, K+-ATPase at central synapses.  相似文献

This study assessed the endocrine and ionoregulatory responses by tilapia (Oreochromis mossambicus) to disturbances of hydromineral balance during confinement and handling. In fresh water (FW), confinement and handling for 0.5, 1, 2 and 6 h produced elevations in plasma cortisol and glucose; a reduction in plasma osmolality was observed at 6 h. Elevations in plasma prolactins (PRL177 and PRL188) accompanied this fall in osmolality while no effect upon growth hormone (GH) was evident; an increase in insulin-like growth-factor I (IGF-I) occurred at 0.5 h. In seawater (SW), confinement and handling increased plasma osmolality and glucose between 0.5 and 6 h; no effect on plasma cortisol was seen due to variable control levels. Concurrently, both PRLs were reduced in stressed fish with only transient changes in the GH/IGF-I axis. Next, the branchial expression of Na+/K+/2Cl? cotransporter (NKCC) and Na+/Cl? cotransporter (NCC) was characterized following confinement and handling for 6 h. In SW, NKCC mRNA levels increased in stressed fish concurrently with elevated plasma osmolality and diminished gill Na+, K+-ATPase activity; NCC was unchanged in stressed fish irrespective of salinity. Taken together, PRL and NKCC participate in restoring osmotic balance during acute stress while the GH/IGF-I axis displays only modest responses.  相似文献

The Malpighian (renal) tubules play important roles in ionic and osmotic homeostasis in insects. In Lepidoptera, the Malpighian tubules are structurally regionalized and the concentration of Na+ and K+ in the secreted fluid varies depending on the segment of tubule analyzed. In this work, we have characterized fluid and ion (Na+, K+, H+) transport by tubules of the larval stage of the cabbage looper Trichoplusia ni; we have also evaluated the effects of fluid secretion inhibitors and stimulants on fluid and ion transport. Ramsay assays showed that fluid was secreted by the iliac plexus but not by the yellow and white regions of the tubule. K+ and Na+ were secreted by the distal iliac plexus (DIP) and K+ was reabsorbed in downstream regions. The fluid secretion rate decreased > 50% after 25 μM bafilomycin A1, 500 μM amiloride or 50 μM bumetanide was added to the bath. The concentration of K+ in the secreted fluid did not change, whereas the concentration of Na+ in the secreted fluid decreased significantly when tubules were exposed to bafilomycin A1 or amiloride. Addition of 500 μM cAMP or 1 μM 5-HT to the bath stimulated fluid secretion and resulted in a decrease in K+ concentration in the secreted fluid. An increase in Na+ concentration in the secreted fluid was observed only in cAMP-stimulated tubules. Secreted fluid pH and the transepithelial electrical potential (TEP) did not change when tubules were stimulated. Taken together, our results show that the secretion of fluid is carried out by the upper regions (DIP) in T. ni Malpighian tubules. Upper regions of the tubules secrete K+, whereas lower regions reabsorb it. Stimulation of fluid secretion is correlated with a decrease in the K+/Na+ ratio.  相似文献

The effects of chronic exposure to total ammonia nitrogen (TAN) concentrations on the brain monoamines and ATPases of Nile tilapia, Oreochromis niloticus fingerlings, were studied. The period of exposure was 70 consecutive days, and the initial weight of the fingerlings was 18 ± 2.1 g. In addition to the control, three treatment groups exposed to 2.5 (low), 5 (medium), and 10 (high) mg TAN L?1 concentrations were tested. The unionized ammonia nitrogen (NH3) levels calculated in mg L?1 were 0.059, 0.185, and 0.575 in aquaria at 26 °C. The brain monoamines were serotonin (5-HT), dopamine (DA), and norepinephrine (NE), as well as their derivatives, 5-hydroxyindoleacetic acid (5-HIAA) and dihydroxyphenylacetic acid (DOPAC). Compared with the controls, the levels of brain monoamines and Na+/K+- and Ca2+-ATPase activities were not significantly altered in fish exposed to low TAN concentration. However, there was a significant decrease in 5-HT, DA, and NE levels, and a significant increase in both serotonergic (5-HIAA/5-HT) and dopaminergic (DOPAC/DA) activities of fish exposed to medium TAN and high TAN concentrations. The activities of brain Na+/K+- and Ca2+-ATPases of fish exposed to medium TAN and high TAN concentrations significantly increased, while Mg2+-ATPase did not significantly change compared with that of the controls. The quantity of the detected alterations increased in fish exposed to high TAN concentration.  相似文献

Three bis(choloyl) conjugates bearing a rigid p-phenylenediamine/p-bis(aminomethyl)benzene linker and amino/acetamido groups were synthesized, and fully characterized on the basis of 1H NMR, ESI-MS and HRMS. Their ionophoric activities were investigated by means of pH discharge assay. The results indicate that these conjugates exhibit potent ionophoric activities across egg-yolk l-α-phosphatidylcholine (EYPC)-based liposomal membranes, via a cation/proton antiport mechanism. They show moderate ion selectivity among alkali metal ions. Of the three conjugates, the ones having amino groups transport alkali metal ions in the order of Na+ > Li+ > K+  Rb+  Cs+, whereas the one having acetamido groups functions in the order of Li+ > Na+ > K+  Rb+  Cs+.  相似文献

The effect of methyl mercuric chloride (MeHg) on short-circuit current (ISC) was studied in the isolated perfused epipodite preparation from the branchial chamber of European lobster (Homarus gammarus) acclimated to dilute seawater. When applied at the apical surface, 0.2, 1.0 and 3.0 μM MeHg depressed ISC by a 26%, 81% and 98%, respectively. The half-maximal inhibitory concentration (IC50) of apically applied MeHg was 0.6 μM. Basolaterally added MeHg (3.0 μM) had no effect on ISC, whereas addition of the specific Na+,K+-ATPase inhibitor ouabain (1.5 mM) reduced ISC by ~ 90%. Ouabain effects were reversible, and ISC fully recovered upon removal of ouabain. The MeHg-induced block of ISC was partially reversed by the reducing agent, 1,4-dithiothreitol, suggesting that the formation of S–Hg–S bridges is important in the inhibitory mechanism. A significant reduction of ISC and conductance occurred when low Na+ and Cl? salines were substituted. Furthermore, in the low Na+ saline, JClA  B fluxes were reduced by about 50%. In the highly conductive epipodite epithelium, coupling of Na+ and Cl? fluxes was suggested. The effects of MeHg on ISC in the lobster epipodite are attributed to inhibition of an apical Cl? influx.  相似文献

Saline environments of terrestrial halophytes are often prone to waterlogging, yet the effects on halophytes of combined salinity and waterlogging have rarely been studied. Either salinity or hypoxia (low O2) alone can interfere with K+ homeostasis, therefore the combination of salinity or hypoxia is expected to impact significantly on K+ retention in roots. We studied mechanisms of tolerance to the interaction of salinity with hypoxia in Puccinellia ciliata and Thinopyrum ponticum, halophytic grasses that differ in waterlogging tolerance. Plants were exposed to aerated and stagnant saline (250 mM NaCl) treatments with low (0.25 mM) and high (4 mM) K+ levels; growth, net ion fluxes and tissue ion concentrations were determined. P. ciliata was more tolerant than T. ponticum to stagnant-saline treatment, producing twice the biomass of adventitious roots, which accumulated high levels of Na+, and had lower shoot Na+. After 24 h of saline hypoxic treatment, MIFE measurements revealed a net uptake of K+ (∼40 nmol m−2 s−1) for P. ciliata, but a net loss of K+ (∼20 nmol m−2 s−1) for the more waterlogging sensitive T. ponticum. NaCl alone induced K+ efflux from roots of both species, with channel blocker tests implicating GORK-like channels. P. ciliata had constitutively a more negative root cell membrane potential than T. ponticum (−150 versus −115 mV). Tolerance to salinity and hypoxia in P. ciliata is related to increased production of adventitious roots, regulation of shoot K+/Na+, and a superior ability to maintain negative membrane potential in root cells, resulting in greater retention of K+.  相似文献

Teleost fish often live in an environment in which osmoregulatory mechanisms are critical for survival and largely unknown in larval fish. The effects of a single important marine ion (K+) on survival and ion regulation of larval Gulf killifish, an estuarine, euryhaline teleost, were determined. A four-week study was completed in four separate recirculating systems with newly hatched larvae. Salinity in all four systems was maintained between 9.5 and 10‰. Two systems were maintained using crystal salt (99.6% NaCl) with K+ supplementation (1.31 ± 0.04 mmol/L and 2.06 ± 0.04 mmol/L K+; mean ± SEM), one was maintained with crystal salt and no K+ supplementation (0.33 ± 0.05 mmol/L K+), the fourth system was maintained using a standard marine mix salt (2.96 ± 0.04 mmol/L K+), the salt mix also included standard ranges of other ions such as calcium and magnesium. Larvae were sampled throughout the experiment for dry mass, Na+/K+-ATPase (NKA) activity, whole body ion composition, relative gene expression (NKA, Na+/K+/2Cl? cotransporter (NKCC) and cystic fibrosis transmembrane conductance regulator (CFTR)), and immunocytochemistry staining for NKA, NKCC, and CFTR. Larvae stocked into water with no K+ supplementation resulted in 100% mortality within 24 h. Mortality and dry mass were significantly influenced by K+ concentration (P  0.05). No differences were observed among treatment groups for NKA activity. At 1 dph NKA mRNA expression was higher in the 0.3 mmol [K+] group than in other treatment groups and at 7 dph differences in intestinal NKA and CFTR staining were observed. These data indicate that the rearing of larval Gulf killifish may be possible in ion deficient water utilizing specific ion supplementation.  相似文献

The recent model showed that seawater (SW) mitochondrion-rich (MR) cells with hole-type apical openings secrete Cl? through the transporters including the Na+, K+-ATPase (NKA), Na+, K+, 2Cl? cotransporter (NKCC), and cystic fibrosis transmembrane conductance regulator (CFTR). The present study focused on the dynamic elimination of the Cl? secretory capacity and illustrated different phases (i.e., acute and regulatory phases) of branchial MR cells in response to hypoosmotic challenge. Time-course remodeling of the cell surfaces and the altered expressions of typical ion transporters were observed in the branchial MR cells of SW-acclimated brackish medaka (Oryzias dancena) when exposed to fresh water (FW). On the 1st day post-transfer, rapid changes were shown in the acute phase: the flat-type MR cells with large apical surfaces replaced the hole-type cells, the gene expression of both Odnkcc1a and Odcftr decreased, and the apical immunostaining signals of CFTR protein disappeared. The basolateral immunostaining signals of NKCC1a protein decreased throughout the regulatory phase (> 1 day post-transfer). During this period, the size and number of NKA-immunoreactive MR cells were significantly reduced and elevated, respectively. Branchial NKA expression and activity were maintained at constant levels in both phases. The results revealed that when SW-acclimated brackish medaka were transferred to hypoosmotic FW for 24 h, the Cl? secretory capacity of MR cells was eliminated, whereas NKCC1a protein was retained to maintain the hypoosmoregulatory endurance of the gills. The time-course acute and regulatory phases of gill MR cells showed different strategies of the euryhaline medaka when subjected to hypoosmotic environments.  相似文献

In teleosts, prolactin (PRL) and growth hormone (GH) act at key osmoregulatory tissues to regulate hydromineral balance. This study was aimed at characterizing patterns of expression for genes encoding receptors for the GH/PRL-family of hormones in the gill and kidney of Mozambique tilapia (Oreochromis mossambicus) during freshwater (FW)-acclimation. Transfer of seawater (SW)-acclimated tilapia to FW elicited rapid and sustained increases in plasma levels and pituitary gene expression of PRL177 and PRL188; plasma hormone and pituitary mRNA levels of GH were unchanged. In the gill, PRL receptor 1 (PRLR1) mRNA increased markedly after transfer to FW by 6 h, while increases in GH receptor (GHR) mRNA were observed 48 h and 14 d after the transfer. By contrast, neither PRLR2 nor the somatolactin receptor (SLR) was responsive to FW transfer. Paralleling these endocrine responses were marked increases in branchial gene expression of a Na+/Cl? cotransporter and a Na+/H+ exchanger, indicators of FW-type mitochondrion-rich cells (MRCs), at 24 and 48 h after FW transfer, respectively. Expression of Na+/K+/2Cl? cotransporter, an indicator of SW-type MRCs, was sharply down-regulated by 6 h after transfer to FW. In kidney, PRLR1, PRLR2 and SLR mRNA levels were unchanged, while GHR mRNA was up-regulated from 6 h after FW transfer to all points thereafter. Collectively, these results suggest that the modulation of the gene expression for PRL and GH receptors in osmoregulatory tissues represents an important aspect of FW-acclimation of tilapia.  相似文献

KEA genes encode putative K+ efflux antiporters that are predominantly found in algae and plants but are rare in metazoa; however, nothing is known about their functions in eukaryotic cells. Plant KEA proteins show homology to bacterial K+ efflux (Kef) transporters, though two members in the Arabidopsis thaliana family, AtKEA1 and AtKEA2, have acquired an extra hydrophilic domain of over 500 residues at the amino terminus. We show that AtKEA2 is highly expressed in leaves, stems and flowers, but not in roots, and that an N-terminal peptide of the protein is targeted to chloroplasts in Arabidopsis cotyledons. The full-length AtKEA2 protein was inactive when expressed in yeast; however, a truncated AtKEA2 protein (AtsKEA2) lacking the N-terminal domain complemented disruption of the Na+(K+)/H+ antiporter Nhx1p to confer hygromycin resistance and tolerance to Na+ or K+ stress. To test transport activity, purified truncated AtKEA2 was reconstituted in proteoliposomes containing the fluorescent probe pyranine. Monovalent cations reduced an imposed pH gradient (acid inside) indicating AtsKEA2 mediated cation/H+ exchange with preference for K+ = Cs+ > Li+ > Na+. When a conserved Asp721 in transmembrane helix 6 that aligns to the cation binding Asp164 of Escherichia coli NhaA was replaced with Ala, AtsKEA2 was completely inactivated. Mutation of a Glu835 between transmembrane helix 8 and 9 in AtsKEA2 also resulted in loss of activity suggesting this region has a regulatory role. Thus, AtKEA2 represents the founding member of a novel group of eukaryote K+/H+ antiporters that modulate monovalent cation and pH homeostasis in plant chloroplasts or plastids.  相似文献

Climate change affects organisms that inhabit not only in aerial but also in aquatic environments by making water more hypoxic and acidic. In the past, we evaluated morphological and functional variations in the gills of 12 species of aquatic air-breathing fishes. The aim of the present study is to examine the degree of gill modification in the aquatic air-breathing fish, Trichogaster lalius, in response to acidic stress. This provides a link between the ecological and physiological studies. We evaluated the changes in morphology and function of the gills, labyrinth organ, and kidney when the fish were subjected to acidic water and deionized water (DW). In the first experiment, fish were sampled at 1, 2, 4, and 7 days after acidic treatment. Apparent morphological modification was observed on day 4 and recovery was noted on day 7. Protein expression and enzyme activity of vacuolar-type H+-ATPase (VHA) and the protein expression of the proliferating cell nuclear antigen (PCNA) of the 1st and 4th gill arches both increased in the 4-day and 7-day acidic groups while the enzyme activity of Na+/K+-ATPase (NKA) decreased. In the second experiment, fish were tested for changes in the 1st and 4th gill arches and kidney after exposure to DW and acidic water for 4 days. The gill structure of the fish in the DW was not different from that of the control group (fresh water). The protein expression and enzyme activity of the VHA of the 1st and 4th gill arches increased in both the DW and acidic groups for 4 days. We found a decrease in the protein expression of NKA in the kidney and in the enzyme activity of NKA in the 1st and 4th gill arches in the DW and acidic groups. From these results, we suggest that T. lalius exhibited significantly different ionic regulation and acid-base regulatory abilities in the DW and acidic groups in the 1st and 4th gill arches and kidney. The responses of the gills in T. lalius were different from those fish that show apparent morphological variations between the 1st and 4th gill arches.  相似文献

β-Casomorphin-7 (β-CM-7) is regarded as the most representative milk-derived bioactive peptide. The present work studies the efficacy of β-CM-7 against myocardial injury in streptozotocin-induced diabetic rats, focusing on the following assays: (1) the level of blood glucose and advanced glycosylation end product (AGE), the activity of lactate dehydrogenase (LDH) in serum; (2) the level of hydrogen peroxide (H2O2), the activity of Na+K+-ATPase, Ca2+Mg2+-ATPase and enzymatic antioxidants such as superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px) in myocardial tissue; (3) the protein expression of glucose transporter-4 (GLUT-4) in myocardial tissue. It showed that with the influence of β-CM-7, the levels of blood glucose of β-CM-7 treatment group decreased markedly compared with model group (P < 0.01) accompanied with their alleviated symptoms of diabetes. In the antioxidant and oxidant levels, β-CM-7 treatment group signified a remarkable increase in the activity of GSH-Px, SOD and CAT of the anti-oxidation system and meanwhile demonstrated a considerable reduction in H2O2 content (all P < 0.05) in comparison with model group. We also found both the content of AGE and the activity of LDH of β-CM-7 treated group considerably reduced while the content of GLUT-4 and the activity of Na+K+-ATPase and Ca2+Mg2+-ATPase of β-CM-7 treated group increased obviously (P < 0.05). Meanwhile the cardiac indexes were significantly lessened. Thus our assay validates that the remedy employing β-CM-7 may treat diabetic cardiomyopathy with high efficacy predominantly associated with the mechanism that β-CM-7 ameliorates myocardial energy metabolism and abates free-radical-mediated oxidative stress in blood and myocardium.  相似文献

The aquatic air-breathing fish, Trichogaster microlepis, can be found in fresh water and estuaries. We further evaluated the changes in two important osmoregulatory enzymes, Na+/K+-ATPase (NKA) and vacuolar-type H+-ATPase (VHA), in the gills when fish were subjected to deionized water (DW), fresh water (FW), and salinated brackish water (salinity of 10 g/L). Fish were sampled only 4 days after experimental transfer. The mortality, plasma osmolality, and Na+ concentration were higher in 10 g/L acclimated fish, while their muscle water content decreased with elevated external salinity. The highest NKA protein abundance was found in the fish gills in 10 g/L, and NKA activity was highest in the DW and 10 g/L acclimated fish. The VHA protein levels were highest in 10 g/L, and VHA activity was highest in the DW treatment. From immunohistochemical results, we found three different cell populations: (1) NKA-immunoreactive (NKA-IR) cells, (2) both NKA-IR and HA-IR cells, and (3) HA-IR cells. NKA-IR cells in the lamellar and interlamellar regions significantly increased in DW and 10 g/L treatments. Only HA-IR cells in the lamellar region were significantly increased in DW. In the interlamellar region, there was no difference in the number of HA-IR cells among the three treated. From these results, T. microlepis exhibited osmoregulatory ability in DW and 10 g/L treatments. The cell types involved in ionic regulation were also examined with immunofluorescence staining; three ionocyte types were found which were similar to the zebrafish model.  相似文献

The potential of four essential cations (K+, Ca2+, Mg2+ and Fe2+) to alleviate salt toxicity was studied in sage (Salvia officinalis L.) plants grown in pots. Two concentrations of the following chloride salts: KCl, CaCl2, MgCl2 and FeCl3, were used together with 100 mM NaCl to study the effects of these nutrients on plant growth, leaf essential oils (EOs) and phenolic diterpenes composition. The sage plants accumulated Na+ in their leaves (includers); this has affected secondary metabolites’ biosynthesis. Treatment with 100 mM NaCl slightly decreased borneol and viridiflorol, while increased manool concentrations. Addition of KCl, CaCl2 and MgCl2 increased considerably in a dose-dependent manner the oxygen-containing monoterpenes (1.8-cineole, camphor, β-thujone and borneol) in 100 mM NaCl-treated sage. Whereas, the contents of viridiflorol decreased further with the addition of KCl in 100 mM NaCl-treated sage. Our results suggest that the changes in EOs composition were more related to K+ and Ca2+ availability than to Na+ toxicity. Furthermore, treatment with NaCl decreased by 50% carnosic acid (CA), a potent antioxidant, content in the leaves. K+ and Ca2+ promoted the accumulation of CA and its methoxylated form (MCA) in the leaves. The concentration of CA was positively correlated with leaf K+ (r = 0.56, P = 0.01) and Ca2+ (r = 0.44, P = 0.05) contents. It appears that different salt applications in combination with NaCl treatments had a profound effect on EOs and phenolic diterpene composition in sage. Therefore, ionic interactions may be carefully considered in the cultivation of this species to get the desired concentrations of these secondary metabolites in leaf extracts.  相似文献

Insects lose ion and water balance during chilling, but the mechanisms underlying this phenomenon are based on patterns of ion and water balance observed in the later stages of cold exposure (12 or more hours). Here we quantified the distribution of ions and water in the hemolymph, muscle, and gut in adult Gryllus field crickets during the first 12 h of cold exposure to test mechanistic hypotheses about why homeostasis is lost in the cold, and how chill-tolerant insects might maintain homeostasis to lower temperatures. Unlike in later chill coma, hemolymph [Na+] and Na+ content in the first few hours of chilling actually increased. Patterns of Na+ balance suggest that Na+ migrates from the tissues to the gut lumen via the hemolymph. Imbalance of [K+] progressed gradually over 12 h and could not explain chill coma onset (a finding consistent with recent studies), nor did it predict survival or injury following 48 h of chilling. Gryllus veletis avoided shifts in muscle and hemolymph ion content better than Gryllus pennsylvanicus (which is less chill-tolerant), however neither species defended water, [Na+], or [K+] balance during the first 12 h of chilling. Gryllus veletis better maintained balance of Na+ content and may therefore have greater tissue resistance to ion leak during cold exposure, which could partially explain faster chill coma recovery for that species.  相似文献

AimsThis study examines the effect of chronic ouabain-treatment on renal Na+ handling in 12-week and 52-week old rats.Main methodsWistar Kyoto rats aged 5 weeks or 45 weeks were treated with ouabain or vehicle during 7 weeks. Blood pressure was measured in conscious animals throughout the study. After 7 weeks of treatment urinary electrolyte concentration, Na+,K+-ATPase activity and α1-subunit expression were determined in 12-week and 52-week old rats.Key findingsIn 12-week and 52-week old rats ouabain produced a significant increase in systolic blood pressure. Although no differences were observed in Na+ excretion in these animals, 12-week old ouabain-treated rats had lower Na+,K+-ATPase activity in proximal tubules. However, 12-week old ouabain-treated rats had decreased fractional excretion of Na+. In proximal tubules of 52-week old rats Na+,K+-ATPase activity did not differ between vehicle and ouabain-treated groups.SignificanceOur results show that in Wistar Kyoto rats renal response to ouabain treatment may be age-dependent and that the hypertensive effect of ouabain is independent of the effect on renal Na+,K+-ATPase.  相似文献

Salts inhibit the activity of sweet almond β-glucosidase. For cations (Cl salts) the effectiveness follows the series: Cu+2, Fe+2 > Zn+2 > Li+ > Ca+2 > Mg+2 > Cs+ > NH4+ > Rb+ > K+ > Na+ and for anions (Na+ salts) the series is: I > ClO4 > SCN > Br  NO3 > Cl  OAc > F  SO4 2. The activity of the enzyme, like that of most glycohydrolases, depends on a deprotonated carboxylate (nucleophile) and a protonated carboxylic acid for optimal activity. The resulting pH-profile of kcat/Km for the β-glucosidase-catalyzed hydrolysis of p-nitrophenyl glucoside is characterized by a width at half height that is strongly sensitive to the nature and concentration of the salt. Most of the inhibition is due to a shift in the enzymic pKas and not to an effect on the pH-independent second-order rate constant, (kcat/Km)lim. For example, as the NaCl concentration is increased from 0.01 M to 1.0 M the apparent pKa1 increases (from 3.7 to 4.9) and the apparent pKa2 decreases (from 7.2 to 5.9). With p-nitrophenyl glucoside, the value of the pH-independent (kcat/Km)lim (= 9 × 104 M 1 s 1) is reduced by less than 4% as the NaCl concentration is increased. There is a similar shift in the pKas when the LiCl concentration is increased to 1.0 M. The results of these salt-induced pKa shifts rule out a significant contribution of reverse protonation to the catalytic efficiency of the enzyme. At low salt concentration, the fraction of the catalytically active monoprotonated enzyme in the reverse protonated form (i.e., proton on the group with a pKa of 3.7 and dissociated from the group with a pKa of 7.2) is very small (≈ 0.03%). At higher salt concentrations, where the two pKas become closer, the fraction of the monoprotonated enzyme in the reverse protonated form increases over 300-fold. However, there is no increase in the intrinsic reactivity, (kcat/Km)lim, of the monoprotonated species. For other enzymes which may show such salt-induced pKa shifts, this provides a convenient test for the role of reverse protonation.  相似文献

The effect of long-term (30 days) exposure to PCZ (0.2, 50, and 500 μg l?1) on intestine-related biochemical markers in rainbow trout was investigated. Multiple biomarkers were measured, including digestive enzymes (proteolytic enzymes and amylase), antioxidant responses (TBARS, CP, SOD, CAT, GR and GPx) and energy metabolic parameters (RNA/DNA ratio, Na+-K+-ATPase). Exposure to 500 μg l?1 PCZ led to significantly inhibited (p < 0.01) proteolytic enzyme and amylase activity. Activities of the antioxidant enzymes SOD, CAT, and GPx gradually increased at lower PCZ concentrations (0.2 and 50 μg l?1). At the highest concentration (500 μg l?1), oxidative stress was apparent as significant higher (p < 0.05) lipid peroxidation and protein carbonyls, associated with an inhibition of antioxidant enzymes activity. Moreover, energy metabolic parameters (RNA/DNA ratio, Na+-K+-ATPase) were significantly inhibited (p < 0.01) in the intestines of fish exposed to 500 μg l?1 PCZ, compared with controls. We suggest that long-term exposure to PCZ could result in several responses in intestine-related biochemical markers, which potentially could be used as indicators for monitoring residual PCZ present in the aquatic environment.  相似文献
