The ultrastructure of the seminal vesicle's spermatozoa of the butterfly Euptoieta hegesia was analyzed. The apyrene spermatozoa measure about 300 microm in length and swim freely in a secretion. The anterior end consists in a cap with a cylindrical extension and a globular structure. The flagellum has a 9+9+2 axoneme, two mitochondrial derivatives with paracrystalline matrices and an external coat formed by concentric layers. The eupyrene spermatozoa measure about 550 microm in length and are grouped into bundles. The anterior end consists in an amorphous globule. Posterior to this globule, a coat with a dense material covers the spermatozoon where an acrosome and a nucleus appear. The flagellum has a 9+9+2 axoneme and two mitochondrial derivatives. External to the coat and attached to the dense material, there is a reticular appendage, which has a paracrystalline core and extends to the distal tip of the spermatozoon.  相似文献

The cholinergic, histaminergic and adrenergic features of regulation of the small muscles contractile activity in a vascular wall of a pulmonary artery in rabbits and involvement of an endothelium in these processes, were investigated. The cholinergic release phenomenon of small muscles of the rabbit pulmonary artery has a two-component character of dose dependence. The low-threshold components of Pilocarpinum relaxing effect has an endothelium-dependent nature. The important feature of histaminergic regulation of contractile activity of segments involves a direct contractile effect of histamine that is not inherent. The endothelium renders a suppressing effect on histaminergic contraction of small muscles of the rabbit pulmonary artery. A basic feature of adrenergic regulation of the pulmonary artery involves registered-beta-adrenergic contractile effects in small muscles of a vascular wall. The activation of the cAMP-dependent signal system in small muscles of a pulmonary artery is capable of rendering a contractile effect. The detected features of a regulation in the small circle can have an essential clinical-physiological value.  相似文献

The alimentary canal of Daphnia pulex consists of a tube-shaped foregut, a midgut (mesenteron) with an anterior pair of small diverticula, and a short hindgut. The foregut and hindgut are structurally similar. Each is formed by a low cuboidal epithelium 5 mum tall and lined with a chitinous intima. The midgut wall consists of a simple epithelium resting on a thick beaded basal lamina which is surrounded by a spiraling muscularis. Anteriorly the midgut cells are columnar in shape being 30 mum in height each having a basal nucleus, anteriorly concentrated mitochondria and in apical border of long thin microvilli. Posteriorly the midgut cells become progressively shorter so that in the posteriormost region of the midgut the cells are 5 mum tall and cuboidal in shape. The microvilli concomitantly become shorter and thicker. All mesenteron cells contain the usual cytoplasmic organelles. The paired digestive diverticula are simple evaginations of the midgut. The wall of each consists of a simple epithelium of cuboidal cells 25 mum in height, each with a brushed border of long thin microvilli. Enzyme secretion appears to be holocrine in mode and not confined to any one region of the mesenteron though definitely polarized anteriorly. The thin gut muscularis encircles the entire length of the midgut and caeca. Thick and thin filaments appear to be in a 6:1 ratio.  相似文献

The healing of wounds is a complex process and the contraction of the resulting scar can have a negative impact on the neighbouring skin. A finite element model of skin simulating the contraction of a scar and deformation of the surrounding skin is presented. The skin is represented by an orthotropic–viscoelastic constitutive law, which is validated against experimental data in the literature. A simplified experimental model of a contracting scar in real skin is also developed. The pattern and size of the wrinkles formed around the contracting scar in the finite element model compare favourably with those formed in the experimental model. The orthotropic nature of skin plays a significant role in the behaviour of skin around scars—the wrinkles have a preferential orientation that corresponds to a direction perpendicular to the Langer's lines in the skin. The pre-stress in skin (a property that is ignored in many skin models) is shown to be an important factor in wrinkle formation around scars. The proposed model can be used to analyse the suturing and closure of wounds of various shapes.  相似文献

The efficacy of clarithromycin retarded release dosage form in the treatment of patients with community-acquired pneumonia was estimated in a open prospective comparative study. The drug was administered in a dose of 500 mg once a day (29 patients) and in a dose of 500 mg twice a day (28 patients). The clinical efficacy in the compared regimens amounted to 90.6 and 93.3% of the patients respectively and the bacteriological efficacy was 93.3 and 84.6% respectively. The adverse reactions were observed in 15.6 and 30.0% of the cases respectively.  相似文献

The Gregarinia, all parasitic in invertebrates have appeared in marine hosts (Polychaeta). The most primitive are the Archigregarinida (Selenidium) which are intestinal parasites. The Eugreganida (intestinal or coelomic) are found in various marine invertebrates as well as in terrestrial or fresh water arthropods. The Neogregarinida which parasitize only insects have a very reduced number of spores compensated by a schizogony (merogony). The Gregarinia constitute a perfect example of a concomitant evolution between a group of parasites and their hosts.  相似文献

A model of intima thickening proposed by Friedman and his coworkers (1,2) is incorporated in our computer code to simulate the growth of intima under shear. The computer code is based on a finite volume method in a boundary-fitted coordinate system. It is found that the model yields an evenly-distributed thickening over a straight, smooth vessel wall. However, in a stenosed or a curved artery, thicker intima can be formed in preferential regions due to unevenly-distributed wall shear stresses. The results clearly demonstrate the correlations among the geometry, wall shear rate and the plaque localization in arteries. The model is applied to a straight artery with a stenosis or sinus, a smooth curved artery and a stenosed curved artery. The effects of stenosis/sinus and lumen curvature on the flows and the intimal thickening are studied. The simulation provides a numerical visualization of the intimal thickening in a dynamic way.  相似文献

The incorporation of caproic acid in the sn-1 position of phosphatidylcholine (PC) catalyzed by lipase from Rhizopus oryzae was investigated in a water activity-controlled organic medium. The reaction was carried out either as esterification or transesterification. A comparison between these two reaction modes was made with regard to product yield, product purity, reaction time, and byproduct formation as a consequence of acyl migration. The yield in the esterification and transesterification reaction was the same under identical conditions. The highest yield (78%) was obtained at a water activity (a(w)) of 0.11 and a caproic acid concentration of 0.8 M. The reaction time was shorter in the esterification reaction than in the transesterification reaction. The difference in reaction time was especially pronounced at low water activities and high fatty acid concentrations. The loss in yield due to acyl migration and consequent enzymatic side reactions was around 16% under a wide range of conditions. The incorporation of a fatty acid in the sn-1 position of PC proved to be thermodynamically much more favorable than the incorporation of a fatty acid in the sn-2 position.  相似文献

The resistance of a Saccharomyces cerevisiae population against gamma-irradiation (400 krd), heating (60 degrees C), repeated freezing--thawing and rehydration of dehydrated cells was shown to depend on the density of a cell suspension. The percentage of damaged cells (stained with primulin) was found to increase in a suspension with decreasing cell concentrations. The effect was also exerted in an oxygen-free medium. The percentage of undamaged cells increased in populations with a lower cell concentration when a more viscous medium (0.2% agar) was used. The intracellular pH dropped down in a denser intact yeast suspension and rose with an increase in the concentration of dehydrated-rehydrated cells.  相似文献

This study aimed to predict the distribution of bone trabeculae, as a density change per unit time, around a dental implant based on applying a selected mathematical remodelling model. The apparent bone density change as a function of the mechanical stimulus was the base of the applied remodelling model that describes disuse and overload bone resorption. The simulation was tested in a finite element model of a screw-shaped dental implant in an idealised bone segment. The sensitivity of the simulation to different mechanical parameters was investigated; these included element edge length, boundary conditions, as well as direction and magnitude of the implant loads. The alteration in the mechanical parameters had a significant influence on density distribution and model stability, in particular at the cortical bone region. The remodelling model could succeed to achieve trabeculae-like structure around osseointegrated dental implants. The validation of this model to a real clinical case is required.  相似文献

The resistance of Mycobacterium tuberculosis to isoniazid is commonly linked to inactivation of a catalase-peroxidase, KatG, that converts isoniazid to its biologically active form. Loss of KatG is associated with elevated expression of the alkylhydroperoxidases AhpC and AhpD. AhpD has no sequence identity with AhpC or other proteins but has alkylhydroperoxidase activity and possibly additional physiological activities. The alkylhydroperoxidase activity, in the absence of KatG, provides an important antioxidant defense. We have determined the M. tuberculosis AhpD structure to a resolution of 1.9 A. The protein is a trimer in a symmetrical cloverleaf arrangement. Each subunit exhibits a new all-helical protein fold in which the two catalytic sulfhydryl groups, Cys-130 and Cys-133, are located near a central cavity in the trimer. The structure supports a mechanism for the alkylhydroperoxidase activity in which Cys-133 is deprotonated by a distant glutamic acid via the relay action of His-137 and a water molecule. The cysteine then reacts with the peroxide to give a sulfenic acid that subsequently forms a disulfide bond with Cys-130. The crystal structure of AhpD identifies a new protein fold relevant to members of this protein family in other organisms. The structural details constitute a potential platform for the design of inhibitors of potential utility as antitubercular agents and suggest that AhpD may have disulfide exchange properties of importance in other areas of M. tuberculosis biology.  相似文献

Summary The application of iron to some glasshouse crops in soilless culture was studied in the course of an investigation. The growing systems consisted of nutrient film technique and rockwool. The test crops were tomatoes, cucumbers and sweet peppers.Various iron chelates were compared in a range of concentrations in a nutrient film system as well as in a rockwool system. The results showed that higher iron levels in the nutrient solution are required in the nutrient film technique than in the rockwool system. The level of the iron content in the nutrient solution had a significant effect on the structure of the root system. Crop analyses showed that on the whole no clear relationship could be found between the iron content of the plant tissues and iron chlorosis in the crop. Only in a few instances could a relationship be established and them only by averaging the results of a series of samplings.  相似文献

COMPOSITION OF CEREBRAL LIPIDS IN MURINE LEUCODYSTROPHY: THE QUAKING MUTANT   总被引:3,自引:3,他引:0  
The composition of sphingolipids and phospholipids of mouse brain during myelination was determined in the Quaking mutant, which manifests a genetic disorder of myelin formation, and in littermate controls. The biochemical changes during myelination in the brains of the controls corresponded quantitatively with previous findings in a different strain of mice. The Quaking mutant exhibited concentrations of sphingolipids and phospholipids in brain which were comparable to those of controls in the early stage of myelination but the tissue content failed to increase with maturation. The greatest differences occurred in the cerebrosides which at 65 days of postnatal age were only 10 per cent of control levels. During development the pattern of cerebral levels of sphingomyelin, plasmalogen and total phospholipid in the mutants tended to resemble that of the cerebrosides. The defect in the Quaking mutant is compatible with a failure in maturation of myelin. These findings have been compared with those in the Jimpy mutant, a different genetic disorder of myelin in the mouse previously studied in a similar fashion. The Jimpy mutant is characterized by a quantitatively more pronounced deficiency of myelin lipids and a decline in cerebrosides during brain development.  相似文献

The results of a bioindication study based on the diversity of epiphytic lichens in a semi-arid area of South Italy are presented. The area features the presence of an industrial zone located in a rural context. The survey was performed in 20 sampling sites. The lichen diversity values (LDV) were determined by the frequencies of all lichen species within a sampling grid placed on tree trunks. Lichens demonstrated a condition of environmental alteration extending to the industrial zone, and tending towards a recovery in the rural surroundings. The survey also showed evidence of eutrophication all over the study area.  相似文献

The erythrocyte membrane is modeled as a two-dimensional viscoelastic continuum that evolves under the application of stress. The present analysis of the erythrocyte membrane is motivated by the recent development of knowledge about its molecular structure. The constitutive equations proposed in the present analysis explain in a consistent manner the data on both the deformation and recovery phases of the micropipette experiment. The rheological equations of the present study are applied in a later section to the analysis of a plane membrane deformation that is quantitatively similar to the tank-treading motion of the erythrocytes in a shear field. The computations yield useful information on how the membrane viscosity becomes a more dominant feature in tank-treading motion. The material constants appearing in the proposed constitutive equations may be useful indications of the biochemical state of the membrane in health and disease.  相似文献

The sensillum of the ear of Feltia subgothica contains two ciliated receptor cells, the A cells. The cilium of each is enclosed within a well developed scolops consisting of a cap, a set of scolopalial rods and a collar, an unusual structure contained within the dendrite. The tip of the cilium is inserted in a channel in the scolopalial cap. The cap is linked to the tympanic membrane by a series of three structures: a cap cell, a microtubular shaft and a microfibrillar plug. The two latter structures are heavily reinforced by cytoskeletal elements and the microfibrillar plug is actually continuous with the tympanic membrane. These three structures transmit the vibration of the tympanic membrane to the scolops. The simplicity and accessibility of the ear suggests that it might be a good system in which to investigate cellular events associated with transduction of sound in these receptors.  相似文献

The Arabidopsis thalianachlorophyll a/b binding protein underexpressed (cue1) mutant, which has been isolated in a screen for chlorophyll a/b binding protein (CAB) underexpressors, exhibits a reticulate leaf phenotype combined with delayed chloroplast development and aberrant shape of the palisade parenchyma cells. The affected gene in cue1 is a phosphoenolpyruvate (PEP)/phosphate translocator (PPT) of the plastid inner envelope membrane. The proposed function of the PPT in C3-plants is the import of PEP into the stroma as one of the substrates for the shikimate pathway, from which aromatic amino acids and a variety of secondary plant products derive. The mutant phenotype could be: (i) complemented by constitutive overexpression of a heterologous PPT from cauliflower; and (ii) rescued by overexpression of a C4-type pyruvate,orthophosphate dikinase (PPDK). The latter approach indicates that PEP deficiency within plastids triggers developmental constraints in cue1. The impact of the mutation on aspects of primary and secondary metabolism was assessed in cue1 as well as in the individual transformant lines. The majority of the data obtained in this and an accompanying paper suggest that the mutant phenotype is not simply caused by a general restriction of the shikimate pathway because of a defect in a PPT.  相似文献

The possibility is discussed of determining the amplitude and phase of a static resonant error field in a tokamak by means of dynamic magnetic measurements. The method proposed assumes measuring the plasma response to a varying external helical magnetic field with a small (a few gauss) amplitude. The case is considered in which the plasma is probed by square pulses with a duration much longer than the time of the transition process. The plasma response is assumed to be linear, with a proportionality coefficient being dependent on the plasma state. The analysis is carried out in a standard cylindrical approximation. The model is based on Maxwell’s equations and Ohm’s law and is thus capable of accounting for the interaction of large-scale modes with the conducting wall of the vacuum chamber. The method can be applied to existing tokamaks.  相似文献

The quasi-elastic incoherent neutron-scattering method has been used to investigate the mobility of water molecules associated with the protein, alpha-chymotrypsin, both in aqueous solution and adjacent to a charged surfactant interface. The latter was studied by solubilizing the protein as monomer in the aqueous cores of small water-in-oil microemulsion droplets (radius 3.5 nm). The droplets were stabilised by an interfacial shell of a double-chain surfactant (Aerosol-OT). The spectra of the water in both these protein-containing system contained a component corresponding to a 7-fold reduction in mobility as compared with bulk water. The integrated intensity of this 'immobilised' spectra component shows that a maximum of approx. 450 water molecules, corresponding to half complete monolayer coverage, are associated with a single protein molecule. This value of 450 may contain a contribution from exchangeable hydrogens within the protein, but this contribution is estimated to be small. The mobility of the remainder of the water is unaffected. The solvation behaviour of the protein is similar in bulk water and in the microemulsion water droplets.  相似文献
