Schoenus ferrugineus andS. nigricans have restricted distributions in Sweden and are almost exclusively confined to calcareous fen habitats. AtS. nigricans sites,S. ferrugineus is usually also present, and hybrids are frequently found. In this report, I used allozymes to estimate the amount of gene flow between the two species, and to compare the partitioning of genetic diversity in each of them. Thirteen loci were analysed at eight different enzyme systems. Seven loci were variable between or within the species. The two species had completely different alleles at two of the seven variable loci, whereas there was overlap at five loci. In all, 22 different alleles were found. Six of these alleles were confined toS. nigricans, and five alleles were confined toS. ferrugineus. Nei's genetic identity was 0.55.—InS. ferrugineus, three loci (23%) were polymorphic, and the average number of alleles per polymorphic locus was 2.0 (each polymorphic locus had two alleles). InS. nigricans, three loci (23%) were polymorphic, and the average number of alleles per polymorphic locus was 2.3.—The proportion of genetic diversity due to variation among sites (G ST) was fairly similar in the two species, mean over loci = 0.12 inS. ferrugineus and 0.15 inS. nigricans. However, the proportion of genetic diversity due to variation among individuals within sites (G IS) differed markedly between the two species, mean over loci = 0.54 inS. ferrugineus and 0.17 inS. nigricans. Accordingly, there was a much higher individual heterozygosity inS. nigricans than inS. ferrugineus. — Most hybrids were interpreted as F1 hybrids. However, a small proportion, 0.5–1.6 %, were Fn hybrids or back-crosses.—On the Swedish mainland, all former occurrences ofS. nigricans are extinct, but viable hybrids are still present at a few sites in southernmost Sweden.  相似文献

Larval feeding was assayed in a generalist caterpillar (Heliothis virescens (F.)), a specialist caterpillar (Heliothis subflexa (Gn.)), their F1 hybrids and a backcross withH. s. to gain a preliminary understanding of the genetic basis of host use inH. v. Plants used in these experiments were tobacco (Nicotiana tabaccum), cotton (Gossypium hirsutum), soybean (Glycine max) (hosts ofH. v.) and ground cherry (Physalis pubescens) (host ofH.s.). A feeding study ofH.v., H.s. and their reciprocal hybrids showed that, after the first eight days of feeding,H.v. had its highest survival and weight gain on soybean and had lowest survival and weight gain on its non-host,Physalis. H.s. had its highest survival and weight gain on its host plant,Physalis, and performed very poorly on the three non-host plants. The two F1 hybrids were SV0 (offspring ofH.s. female andH.v. male) and VS0 (offspring ofH.v. female andH.s. male). The hybrids did not differ from each other, indicating no sex linkage or maternal effects, except that VS0 had greater weight gain on tobacco than did SV0. The hybrids, unlike their parents, survived well on all four host plants and their weight gain was intermediate on all four host plants. In a separate experiment the VS0 hybrid was mated to the specialist to produce the VS1 backcross. In contrast to the F1, the backcross had significantly lower survival than the generalist on soybean, cotton, tobacco and weight gain was lower on soybean, cotton and tobacco but higher onPhysalis. Survival and weight gain on cotton and tobacco were inherited as partially dominant traits; onPhysalis there was overdominance for survival and complete dominance for weight gain; on soybean both survival and weight gain were additive. Survival on cotton,Physalis, soybean and tobacco and weight gain onPhysalis could not be completely explained by a model that included only dominance and additive effects. These traits may be influenced by epistatic and/or environmental effects.  相似文献

Artificial second generation and backcross hybrids involvingEchinocereus dasyacanthus (Texas rainbow cactus),E. coccineus (a species of claret-cup cactus), andE. ×lloydii (Lloyd's hedgehog cactus) were examined for morphological and reproductive characteristics. The hybrids grew from seed to flower in about four years, and they exhibited high fertility as measured by pollen stainability, crossability, and fruit-set. A rare breeding system inCactaceae, dioecy, was further examined inE. coccineus, using field and controlled pollination tests. Functional dioecy was documented forE. coccineus in Trans-Pecos Texas. All results suggested thatE. ×lloydii in eastern Pecos County, Texas, is a complex and dynamic population probably comprising first and later generation hybrids, including backcross hybrids. The experimental hybridizations also tended to support the occurrence of introgression into both putative parental species.  相似文献

In situ hybridization of biotin-labeled mouse major satellite DNA clone pMR196 was applied toMus domesticus andMus spretus chromosomes (Chr). The same karyotypes were counterstained with distamycin A-DAPI to identify AT-rich heterochromatin. Chromosomes from the laboratory mouse, C57BL/6Ros (BL/6;M. domesticus) were uniformly labeled at the centromere except for the Y, while chromosomes from the divergentMus speciesM. spretus showed little or no hybridization. Differences betweenMus species in copy number of the major satellite DNA sequence were used to identify chromosomes ofM. domesticus andM. spretus in their F1 hybrids and to discriminatedomesticus andspretus centromeres in backross progeny. The distribution pattern of heterochromatic regions demonstrated by distamycin A-DAPI counterstaining was comparable with that of in situ hybridization with pMR196, suggesting that A-T rich heterochromatin inM. domesticus is mainly constructed by the pMR196-related sequence. The in situ technique was used to examine segregation ofdomesticus centromeres in backcross progeny obtained by mating F1 hybrid females withM. domesticus orM. spretus males. The segregation of centromeres did not deviate from the expected among the backcross progeny from C57BL/6Ros males, whereas chromosomes withM. domesticus centromeres were prone to appear in the progeny from backcross matings byM. spretus males.  相似文献

By means of thin layer chromatography in conjunction with mass spectrometry the pyrrolizidine alkaloid patterns derived fromSymphytum asperum, several cytotypes ofS. officinale agg. and the artificial hybrids of the former taxa, were compared. The obtained patterns were not essentially affected by variation in cytotype, harvesting times and -location of plants. Lycopsamine, acetyl-lycopsamine and symphytine or their isomers were generally found in theS. officinale cytotypes, echimidine and symphytine inS. asperum. The interspecific hybrids contained all alkaloids mentioned. The definite lack of echimidine in the 2 n=40 cytotype proves that it is conspecific withS. officinale and does not belong to a hybrid-swarmS. asperum × S. officinale with 2 n=48. The breakdown of species integrity was studied in three sympatricSatyrium (Orchidaceae) species at a site in the Western Cape province of South Africa. Hybrids with intermediate morphology were identified using multivariate analysis.Satyrium erectum ×S. bicorne andS. erectum ×S. coriifolium hybrids were common, while only oneS. bicorne ×S. coriifolium hybrid was found (a new record). Reciprocal artificial crossing experiments confirmed that genetic compatibility exists between all three species. However, the species have different pollinators: moths inS. bicorne, bees inS. erectum and sunbirds inS. coriifolium. Pollinators, rather than genetic factors, therefore, appear to be responsible for reproductive isolation between these species. Breakdown in pollinator specificity results in interspecific pollen transfer and the formation of hybrids. Overlap in floral signals, including nocturnal scent in bothS. erectum andS. bicorne, and bright coloration in bothS. erectum andS. coriifolium, facilitates the formation of hybrids between these species pairs. The brown, scented flowers ofS. bicorne, on the other hand, share no obvious floral signals with the bright orange, unscented flowers ofS. coriifolium, accounting for the rarity of hybrids between this species pair. Differences in floral morphology also result in mechanical isolation between these two species. Since the three species are sympatric in a very small part of their geographic ranges, it is unlikely that avoidance of hybridization has been an important selective factor in the evolution of specialized pollination systems.  相似文献

Stylar proteins involved in the self-incompatible (SI) response ofLycopersicon hirsutum have been identified and mapped to the locus that controls SI (S locus).L. esculentum, a self-compatible (SC) species of cultivated tomato, does not display these proteins. Hybrids between SCL. esculentum and SIL. hirsutum are self-sterile despite these individuals bearing pollen containing theS allele ofL. esculentum. In progeny derived from backcrossing the hybrids toL. esculentum, there was a strong correlation between the presence of theS allele fromL. hirsutum and self-infertility. However, this relationship was uncoupled in a number of backcross (BC) progeny. The SI response appeared to be nonexistent in two self-fertile BC individuals that were heterozygous for theS allele ofL. hirsutum, based on Mendelian segregation of a tightly linked DNA marker,CD15, in selfed progeny. Among these progeny self-fertile individuals that were homozygous for theL. hirsutum allele of the linked marker were also determined to be homozygous for anS-related protein ofL. hirsutum through test crosses withL. esculentum. Therefore, plants were produced that were homozygous for a functionalS allele but were self-fertile. This result and other evidence suggest that theS-related proteins are not sufficient to elicit a self-incompatible response inL. esculentum and that there is a mutation(s) inL. esculentum somewhere other than theS locus that leads to self-compatibility.  相似文献

Of 851 specimens ofTridentiger obscurus andt. brevispinis collected from Lake Hinuma (Ibaraki Prefecture, Japan) from July 1996 to February 1998, 49 (5.8%) comprised F1 hybrids and backcross progeny of the two species. Since the mitochondrial DNA haplotypes of the F1 hybrids reflected those ofT. brevispinis, most instances of hybridization are thought to have occurred between maleT. obscurus and femaleT. brevispinis. Although both allozyme and mitochondrial DNA analyses indicated backcrossing and introgression of mitochondrial DNA, the frequency of backcross progeny was relatively low, suggesting the existence of a natural selection to backrossing.  相似文献

Intergeneric crosses were made between representatives of the genomically-defined generaElymus, Agropyron, Elytrigia, Pseudoroegneria, andThinopyrum. The genomic constitution ofElytrigia repens, the type species ofElytrigia, is shown to be SSH, a genomic combination otherwise found only inElymus. The S genome ofPseudoroegneria has almost always a dominant influence on the morphology of the taxa of which it is a component.Wang (1989) showed that the J genome inThinopyrum and the S genome have considerable homoeology, with a mean c-value of 0.35 in diploid SJ hybrids. A genetic coherence from S to SJe, Je, JeJb, and Jb can be expected, agreeing with the continuous morphologic variation pattern observed. Because of the absence of morphological discontinuities between the taxa,Pseudoroegneria (S),Elymus (SH, SY, sometimes with additional genomes),Elytrigia (SSH, SSHX), andThinopyrum (SJ, SJJ, J) are best treated as a single genus,Elymus, following the generic concept ofMelderis in Flora Europaea and Flora of Turkey. The basic genomic constituents ofElymus will then be the S and/or J genomes.Agropyron, with diploids, tetraploids, and hexaploids based on the P genome is morphologically distinct from other genera inTriticeae. In a few species ofElymus andPseudoroegneria, a P genome is an additional constituent. In these cases the P genome has a negligible morphological influence. Therefore, it seems reasonable to maintainAgropyron as a separate genus.  相似文献

This study focuses on the variability of chromosomal location and number of ribosomal DNA (rDNA) sites in some diploid and autotetraploidFestuca pratensis andLolium perenne cultivars, as well as on identification of rDNA-bearing chromosomes in their triploid and tetraploidF. pratensis ×L. perenne hybrids. The rDNA loci were mapped using fluorescence in situ hybridization (FISH) with 5S and 25S rDNA probes, and the origin of parental genomes was verified by genomic in situ hybridization (GISH) withL. perenne genomicDNAas a probe, andF. pratensis genomic DNA as a block. FISH detected variation in the number and chromosomal location of both 5S and 45S rDNA sites. InF. pratensis mostly additional signals of 5S rDNA loci occurred, as compared with standardF. pratensis karyotypes. Losses of 45S rDNA loci were more frequent inL. perenne cultivars and intergeneric hybrids. Comparison of theF. pratensis andL. perenne genomes approved a higher number of rDNA sites as well as variation in chromosomal rDNA location inL. perenne. A greater instability ofF. pratensis-genome-like andL. perenne-genome-like chromosomes in tetraploid hybrids was revealed, indicating gains and losses of rDNA loci, respectively. Our data indicate that the rDNA loci physically mapped on chromosomes 2 and 3 inF. pratensis and on chromosome 3 inL. perenne are useful markers for these chromosomes in intergenericFestuca ×Lolium hybrids.  相似文献

The tandemly repeated multigene families encoding 18S and 25S rRNAs were studied at the restriction enzyme level inPopulus alba L.,Populus deltoides Bartr. exMarsh.,Populus trichocarpa Torr. & Gray and in the hybrids between the last two mentioned species. The analysis of single and double digestion with EcoRI, BamHI, XbaI, and SstI endonucleases showed the presence of single repetitive unit types of 12.25 and 11.75kb inP. alba andP. trichocarpa, respectively.P. deltoides showed two rDNA gene types having the same length (12.25Kb) but different nucleotide sequence in the IGS. Seventy populations of North American annualMicroseris, Stebbinsoseris, andUropappus species were examined for chloroplast and nuclear ribosomal DNA restriction site variability to determine the origin of the allotetraploid speciesS. heterocarpa andS. decipiens. Previously identified chloroplast DNA restriction site variants were used in concert with restriction site variation forNco I in the nuclear-encoded ribosomal DNA repeat. The presence of two, mutually exclusive restriction site gains were observed in diploid populations ofM. douglasii; these same variants were also found in populations of allotetraploidS. heterocarpa, indicating mutiple origins of this species from different maternal diploid populations ofM. douglasii. Variation in the rDNA repeat between the diploid annual species and the putative paternal genome ofU. lindleyi was found to be additive inS. heterocarpa. A similar relationship was observed for the origin ofS. decipiens; cpDNA restriction site variants found inM. bigelovii andM. douglasii were present inS. decipiens. The rDNANco I variants also were additive in this purported allotetraploid. These results confirm the reticulate evolutionary pattern inStebbinsoseris and provide another example of multiple origins of intergeneric allopolyploids.  相似文献

The enzymatic spectra ofTypha angustifolia, T. latifolia and their hybrids were examined by disc electrophoresis for three enzymatic systems: esterases, alcohol deshydrogenase, and glutamate deshydrogenase. The intermediate position of the hybrids is confirmed. The study of alcohol deshydrogenase reveales the existence of an intraspecific variability inT. angustifolia between individuals from the Massif Armoricain in France and Dellys in Algeria.  相似文献

Interspecific hybrids from the crosses betweenBrassica campestris, B. carinata, B. juncea andB. napus were obtained throughin vitro ovary and ovule culture. F1 hybrids were studied morphologically and flow cytometry was used to estimate 2C nuclear DNA content both in parentalBrassica species and their hybrids. It was found that in comparison with the A genome, the B and the C genomes ofBrassica contained 26.9 % and 43.9 % more DNA, respectively. This finding may be used to distinguish interspecific hybrids containing various genome combinations. It was concluded that flow cytometric analysis of nuclear DNA content might be useful tool inBrassica breeding.  相似文献

Cell walls of two strains ofChlorella vulgaris (from fresh water, F, and saline water, S) and ofKirchneriella lunaris contained various proportion of saccharides and proteins (highest percentage in dry matter was found inK. lunaris). They differed also in presence of individual monosaccharides (5 inC. vulgaris F, 8 inC. vulgaris S and 6 inK. lunaris) and amino acids (11 in C.vulgaris F, 6 in C.vulgaris S and 7 inK. lunaris). To assist in the domestication of the sole fish Solea senegalensis, 15 microsatellites were developed from its genome (13 di‐ and two trinucleotide). These loci, along with three from S. solea that cross‐amplified, were characterized on a sample of 20 individuals of S. senegalensis from a hatchery stock. All loci were polymorphic displaying from two to 15 alleles/locus (mean 6.5). The results obtained from the hatchery population screening indicate that they might be useful for studies of genetic structuring and/or kinship analysis in S. senegalensis.  相似文献

A total of 35 electrophoretic variants were detected in four apogamous species of theDryopteris varia group: 14 variants inD. bissetiana triploids, 7 inD. sacrosancta triploids, 6 inD. varia triploids, 3 inD. pacifica triploids, and 5 in apogamous diploids. The variants differed from each other in various combinations of a few alleles which were common in some of the variants, suggesting recurrent origin.Dryopteris insularis var.insularis (2x) and var.chichisimensis (3x), other members of the group, were electrophoretically monomorphic, suggesting single origin. Banding patterns infer that triploids ofD. bissetiana andD. sacrosancta may have originated from hybrids whose parent is the sexualD. saxifraga, whereas this species was not involved in the origin of the other apogamous triploids and the apogamous diploids. The other parent was also discussed and we proposed a hypothesis that diploid offsprings fromD. pacifica triploids hybridized withD. saxifraga giving rise to triploids ofD. bissetiana andD. sacrosancta.  相似文献

InSedum rupestre L. a polyploid series (x = 16) occurs in which aneuploid chromosome numbers and odd levels of ploidy prevail. The most common and widely distributed cytotype,S. rupestre subsp.rupestre, is 2n = 112. Plants resemblingS. rupestre subsp.rupestre can be obtained by hybridizing the tetraploid cytotypes ofS. forsterianum Sm. (2n = 48) andS. rupestre subsp.erectum 't Hart (2n = 64). Comparison of these artificial hybrids with their parents and a large number of plants ofS. rupestre subsp.rupestre (2n = 112) from nature showed thatS. rupestre subsp.rupestre and the artificial hybrids are morphologically indistinguishable, and intermediate betweenS. forsterianum andS. rupestre subsp.erectum. MorphologicallyS. rupestre subsp.rupestre is closer to subsp.erectum than toS. forsterianum. Chloroplast DNA restriction patterns ofS. rupestre subsp.rupestre, however, resembleS. forsterianum more closely. The combined results of the hybridization experiments, the analysis of the cpDNA restriction patterns, and the morphological variation indicate the allopolyploid origin ofS. rupestre subsp.rupestre. Morphological, geographical and ecological evidence suggests thatEncelia virginensis is a true-breeding diploid species derived from hybrids ofE. actoni andE. frutescens. To test this hypothesis, we examined the chloroplast and nuclear DNA of severalEncelia species. PCR amplification targeted three separate regions of chloroplast DNA:trnK-2621/trnK-11,rbcL/ORF106, andpsbA3/TrnI-51, which amplify 2600bp, 3300bp and 3200bp fragments respectively. Restriction fragment analysis of chloroplast DNA revealed no variation that could be used to discriminate between the parent species. A RAPD analysis using 109 dekamer primers was used to analyze the nuclear genome.Encelia actoni andE. frutescens were distinguished by several high-frequency RAPD markers. In populations ofE. virginensis, these markers were detected in varying proportions, and no unique markers were found. Evidence from the nuclear genome supports the hypothesis thatE. virginensis is of hybrid origin. ThatE. virginensis may have arisen by normal divergent speciation followed by later introgression remains a possibility, however, and is not formally ruled out here. Diploid hybrid speciation inEncelia differs from other documented cases in that there are no discernible chromosome differences between the species, and all interspecific hybrids are fully fertile. In addition, apparent ecological selection against backcross progeny provides an external barrier to reproduction between F1 progeny and the parental species. These characteristics suggest that hybrid speciation inEncelia may represent an alternative model for homoploid hybrid speciation involving external reproductive barriers. In particular, this may be the case for other proposed diploid hybrid taxa that also exhibit little chromosomal differentiation and have fertile F1s.  相似文献

Ischnura species (Odonata) are among the most common damselflies in the world, which often exhibit female color polymorphisms. One morph, called androchrome, is similar to males in its color pattern, whereas the other morphs, generally referred to as gynochromes, exhibit female‐specific colors. In several Ischnura species, the female polymorphism is heritable, although molecular and genetic mechanisms remain largely unknown. The dominant‐recessive patterns of the female color morphs may differ between species. For example, androchromic females are dominant to gynochromic females in Ischnura elegans, whereas androchromic females are recessive in Ischnura senegalensis. Here we report a case of interspecific hybridization between a gynochrome female of I. elegans and a male of I. senegalensis in the laboratory. We obtained 61 hybrid adult offspring, of which all 31 females were of gynochrome morph. DNA analyses of the hybrids confirmed that nuclear DNA sequences were derived from both parent species, whereas mitochondrial DNA sequences were maternally inherited. In the hybrids, the postocular spots of female heads, the shape of male appendages, and the color of female's cerci resembled those of I. elegans, whereas the size of abdominal blue spots was similar to that of I. senegalensis. The shape of prothorax and basal abdominal markings were intermediate in females. The larval developmental traits and the morphological changes in the final larval instar of the hybrids were similar to those of I. senegalensis. To our knowledge, this is the first report of hybrids between two damselfly species with different dominant‐recessive patterns of female color morphs.  相似文献
