The process of the disease due to herpes simplex virus types 1 and 2 (HSV-1 and 2) was studied on white uninbred mice weighing 10 to 12 g. The animals were infected intracerebrally or intraperitoneally. Intraperitoneal contamination of the animals with MS strain of HSV-2 was used for the experimental model of the herpes simplex infection. The prophylactic antiherpes action of ultralow doses of the human gamma-interferon antibodies (ULD of anti-IFN-gamma) at a course of its intragastral administration was evaluated. The preparation was shown to have a significant (p < 0.05) protective effect in a dose of 10 LD50, evident from a 10-fold decrease of the HSV-2 accumulation in the brain, a lower percentage of the animal deaths and an increase of the average lifespan of the animals by 3.3 days. The study of the therapeutic action of ULD of anti-IFN-gamma at a course of its intragastral administration showed that the preparation had no significant positive effect on the disease process in the animals infected with HSV-2 in a dose of 10 LD50. However, a positive effect associated with delayed virus replication in the brain was observed in the study on the therapeutic effect of ULD of anti-IFN-gamma after its intragastral administration to the mice infected with a sublethal dose of the virus.  相似文献

Influence of dsRNA isolated from killer yeast of S. cerevisiae on humoral and cellular immune responses in mice CBA/CaY and C57Bl/6Y with opposing reaction to the antigen was studied. It was shown that after administration of the yeast dsRNA preparation to the animals simultaneously with the antigen there was an increase in the number of the antibody forming cells in the spleen and the titer of hemolytic antibodies in blood serum of the animals with high and low reactions to the antigen. After sensitization with different doses of sheep red blood cells (10(7) and 10(8)) the preparation had immunomodulating action on development of DTH in mice CBA/CaY. The effect of the dsRNA preparation on the immunity system depended on the preparation dose, antigen loading and animal genotype and was the most marked in mice CBA/CaY with interferon levels in blood serum 2-3 times higher than those in mice C57Bl/6Y.  相似文献

The effect of the putative endogenous ligand for alpha(2)-adrenoceptors and imidazoline receptors agmatine was studied in sympathetic neurotransmission in the rat epididymal vas deferens. Tissues were obtained from N(varpi)-nitro-l-arginine methyl ester (l-NAME)-treated or normal animals and were contracted by electrical stimulation or by exogenous adenosine 5'-triphosphate (ATP). In the electrically stimulated epididymal end, agmatine produced an inhibitory effect on twitch contraction that was partially reversed in l-NAME-treated animals, whereas the inhibition produced by clonidine was not affected by l-NAME treatment. The nitric oxide (NO)-donor S-nitroso-N-acetyl-penicillamine (SNAP) also inhibited twitch contraction. Neither agmatine nor SNAP interfered with the responses induced by exogenous ATP in the epididymal end. Removal of the epithelium of the preparation did not modify the agmatine response. We conclude that a nitrergic pathway activated by agmatine plays a role in its inhibitory effect in rat vas deferens, but it remains to be investigated whether it results from a direct action on the enzyme NO-synthase or a receptor-mediated mechanism.  相似文献

Biological activity of a new natural interferon inductor, the replicative RNA form of phage f2 (RFf2) was studied. A possibility of using RFf2 for production of highly active interferon under conditions of superinduction providing an increase in the interferon yield by to 256--512 times as compared to the control samples was shown. The protective interferonogenic and antiviral effect of RFf2 in mice infected with Semliki forest virus (SFV) and tickborne encephalitis virus (TBEV) was studied on administration of the inductor by various routes. It was found that intraperitoneal administration of RFf2 in a dose of 10 gamma per a mouse protected the infected animals from death. It was accompanied by production of up to 1280 units/ml of interferon in the blood serum of the animals. Maximum protection of the animals from death under conditions of the experiment (80 per cent on infection with SFV and 65 per cent on infection with TBEV) was observed when the preparation was administered twice: 4 hours after the infection. Combined use of RFf2 with chemotherapeutics (rimantadine) increased the protective effect both in the tissue culture and in vivo.  相似文献

The effect of lipid preparations Nos. 59 and 60 isolated from Protozoa on experimental leishmaniosis of golden hamsters was studied. Preparations Nos. 59 and 60 are water-soluble derivatives of fatty acids isolated from Trypanosoma lewisi and Astasia longa respectively. It was found that both preparations administered to the animals simultaneously with the causative agent or after formation of the leishmaniosis infiltrates inhibited the development of the disease clinical signs. Such an effect of the preparations was observed only during the period of the animals exposure to them. After discontinuation of the treatment the development of the clinical process proceeded the same as in the control animals. The lipid preparation No. 60 differing from preparation No. 59 by high content of C20-poly-unsaturated fatty acids was more active and induced a significant decrease in the size of the leishmaniosis infiltrates formed. Preparation No. 60 was more effective in the treatment of the animals infected with L. tropica major than in the animals infected with L. tropica minor. Preparation No. 59 had no significant effect on multiplication and morphology of the parazites.  相似文献

Long-term rectal administration of enzyme mixture containing papain, trypsin and chymotrypsin in the same ratio as the preparation Wobe-Mugos E (Mucos Pharma, Germany) was evaluated for their antitumor effects in C57B16 inbred mice inoculated with B16 melanoma cells. 30% of animals in the test group (3 pcs) have been cured of cancer. In the rest of animals (70%) the survival time was prolonged by 58.3% compared to the control group (from average survival time of 24 days in control group to 38 days in the test group). Based on histological and immunohistochemical evaluation a faster process of metastasizing was found in control group than in the group treated with the polyenzyme preparation. In the case of melanoma B16 an antimetastatic effect of the preparation was thus proved. © 1997 Elsevier Science Inc.  相似文献

Toxicity-pathogenicity test of viroden, a new preparation, and its acting agent--a mosquito densonucleosis virus (MDV) has been carried out on warm-blooded animals. It is shown that the preparation is not toxic for laboratory animals (white common mice, rats, guinea pigs, rabbits), chicken embryos and cell cultures of warm-blooded animals. The MDV is not adapted to a warm-blooded organism with different ways of introduction and in passages. Using electron and luminescent microscopy, serological reactions, specific test systems and a biological test for sensitive insects no explicit or latent infection was found in animals, chicken embryos and cell cultures of vertebrates with primary infection and in passages. Sensibilized animals shown an immunological rearrangement of the organism proceeding by the retarded hypersensitivity type.  相似文献

Reactive oxygen species play an important role in cancer and metastasis. Kalpaamruthaa is a modified Siddha preparation, which has been formulated in our laboratory. The preparation is an amalgamation of Semecarpus anacardium (SA), Emblica officinalis (EO) and honey, which gives an extra protectiveness to mammary carcinoma bearing animals (Sprague-Dawley stains were used for this study). The aim of our research is to determine the therapeutic efficiency of the drug with respect to lipid peroxidation and antioxidant status. The levels of lipid peroxides and antioxidant levels were measured in blood, and vital organs (liver, kidney and breast tissue) of control and experimental animals. In cancer condition, the LPO was increased and antioxidant levels were decreased. On drug (SA and KA) administration, decreased LPO and increased antioxidant levels were seen in control and experimental animals. This may be due to additive property of the drugs (SA, Emblica and honey), which possesses anticancer effect. The present study shows the good therapeutic efficacy of Kalpaamruthaa against mammary carcinoma.  相似文献

Terrilitin is studied for its effect on proteolytic activity of blood and formation of immunostimulating factors by spleen cells. The preparation is shown to induce isolation of the immunostimulating factor (molecular mass 10-15 kDalton) from the spleen cells. The preparation is destroyed by trypsin and RNAase and is stable to the action of lysozyme. Spleen cell factor of the animals with administered terrilitin increases general antiproteolytic activity of the blood serum and concentration of alpha 2-macroglobulins. At the same time, it decreases the general proteolytic activity and callicrein activity of blood serum for syngenic animals.  相似文献

The influence of 90 h of acute nutritional deprivation (ND) on the cross-sectional areas of muscle fibers and the contractile and fatigue properties of the adult rat diaphragm were determined. Isometric contractile properties and fatigue resistance of the diaphragm were measured by means of an in vitro nerve-muscle strip preparation. Contractions were evoked by using phrenic nerve stimulation (left hemidiaphragm) or direct muscle stimulation (right hemidiaphragm) in the presence of curare. Acute ND resulted in a 20% reduction in body weight. No significant decrements in diaphragm or soleus weights were noted in the ND animals compared with controls (CTL), whereas the weight of the medial gastrocnemius was reduced by 20% in the ND animals. Peak twitch and tetanic tensions (normalized for the weight of the diaphragm strip) were not reduced in ND compared with CTL animals after either nerve or muscle stimulation. The fatigue index of the diaphragm was significantly reduced in ND animals only after nerve stimulation. After the fatigue test, there was rapid recovery of the additional fatigue noted with nerve stimulation. The proportions of type I and II muscle fibers of the diaphragm were similar in the CTL and ND animals. No differences in diaphragm cross-sectional areas were noted for either type I or II muscle fibers in the CTL and ND animals. It is concluded that acute ND has no effect on diaphragm contractility or morphometry and only an inconsequential influence on diaphragm fatigue.  相似文献

To verify if the peptide preparation Cortexin can be used to correct pathological processes in the CNS during perinatal ontogenesis, a set of physiological parameters (EMG, ECG, respiration, vagosympathetic balance) were recorded and analyzed in a rat perinatal hypoxic-ischemic brain damage (PHIBD) model and control infant rats. In experimental 7-day-old animals, PHIBD was modeled under inhalation ether anesthesia by ligation of the left common carotid artery and subsequent exposure to a gas mixture of 8% oxygen and 92% nitrogen. 1 h after exposure, animals were first injected intraperitoneally with cortexin (1 mg/kg); the preparation was then injected at a daily basis for 10 days. Both control animals and those after surgery but non-treated were injected with physiological solution. After 10 and 30 days after surgery, animals exhibited a delayed body weight gain versus control and significant differences in EMG intensity and spectral structure. Cortexin treatment induced on day 10 a transient improvement in EMG spectral structure but not amplitude; by day 30, the positive effect of cortexin was no longer observed. The respiration rate both in treated and non-treated animals was higher than in control. No significant changes in the heart rate were revealed in animals with PHIBD, but non-treated animals exhibited on day 30 a tendency towards its decrease. The heart rate variability (HRV) analysis showed that 10 days after trauma both non-treated and cortexin-treated animals exhibited a statistically significant shift in the vagosympathetic balance towards a parasympathetic prevalence. On day 30, cortexin treatment yields positive effects, while in non-treated animals the vagosympathetic balance shifts towards a humoral-metabolic and sympathetic prevalence. Cortexin injection to intact rats leads to significant disturbances in the vagosympathetic balance, cardiac and, to a lesser extent, respiratory rhythms, and can cause stable disturbances in activity of the somatic and vegetative nervous systems.  相似文献

The protective activity of pyoimmunogen II, lot 9, was studied on P. aeruginosa experimental sepsis used as a model. In experiments on rats the preparation was shown to be nontoxic according to the results of the determination of acute and chronic toxicity. The preparation under test produced a high protective effect in experiments on animals infected with various P. aeruginosa strains, irrespective of their virulence and immunotype. Anti-P. aeruginosa plasma, obtained by plasmapheresis from donors immunized with pyoimmunogen II, showed a curative effect when injected into experimental animals in a dose of 0.02 ml/g body weight at early stages of the infection.  相似文献

Several parameters of the cytoplasmic enzymatic antioxidant system of the liver and brain of the rat have been investigated under conditions of immobilization stress and of an antioxidant preparation in the diet of animals. These included superoxide dismutase (SOD) and glutathione reductase (GR) activities and nonspecific NADPH oxidation. Only changes in the activity of SOD both in the liver and brain were revealed. In the liver of animals that receive no preparation, a decrease in the activity of SOD after 30-min immobilization and its restoration after a 360-min immobilization were observed. In the brain, the activity of SOD decreased only in preconditioned animals after 30 and 360 min of exposure to stress. In addition, the activity of SOD in the brain of preconditioned animals, both stressed and unstressed, was lower than in the corresponding groups of control animals. It is probable that, under the conditions of immobilization stress, the level of reactive oxygen species (ROS) and as a consequence the activity of SOD decrease. The intake of an antioxidant preparation under these conditions seems to be not correct.  相似文献

We examined the therapeutic effect of a 1% cream preparation of NND-502, a novel topical antifungal agent, in a guinea pig tinea pedis model produced by infecting the plantar skin of guinea pigs with Trichophyton mentagrophytes. Animals developing tinea pedis were divided into two groups: an untreated control group and a treated group. In the latter group, after confirming infection had been established, the infected animals were topically treated with the NND-502 cream once daily for one week. The animals were reared in a clean environment free from exposure to exogenous dermatophytes. At one week (5 weeks post-infection), 6 weeks (10 weeks post-infection) and 16 weeks (20 weeks post-infection) after completion of the treatment, plantar skin samples were taken from a certain number of both groups of animals. The results demonstrated that all of the animals in the untreated control group and none of those in the treated group were culture-positive in this animal model of tinea pedis. The topical treatment with NND-502 achieved a mycological cure. Thus NND-502 can be considered a promising candidate as a new anti-dermatophytic agent for topical use.  相似文献

BACKGROUND: Breast cancer is one of the most common cancers in women of developed and developing countries. Lipids, lipoproteins and lipid-metabolizing enzymes have been associated with the risk of breast cancer. Kalpaamruthaa (KA) is a modified Siddha preparation, which contains Semecarpus anacardium Linn. (SA), Emblica officinalis (EO) and honey. OBJECTIVE: The present study was embarked to study the variations in lipids, lipid-metabolizing enzymes and lipoproteins in cancerous animals and the effect of KA on the lipid metabolism. MATERIALS AND METHODS: Breast cancer was induced in rats by administrating 7,12-dimethylbenz(a)anthracene orally (25 mg/kg body weight). After 90 days of induction, KA (300 mg/kg body weight) and SA (200 mg/kg body weight) were administered for 14 days, by gastric intubations. The levels of lipids and lipid-metabolizing enzymes were analysed in control and experimental animals. RESULTS AND CONCLUSION: The increased levels of total cholesterol, free cholesterol, phospholipids, triglycerides and free fatty acids and decreased levels of ester cholesterol in plasma, liver and kidney found in cancer suffering animals were reverted back to near normal levels on treatment with KA and SA. In mammary carcinoma bearing animals, the activities of total lipase, cholesterol ester synthase, and cholesterol ester hydrolase were significantly (p < 0.05) increased whereas lipoprotein lipase and lecithin cholesterol-acyl transferase were decreased. The levels of very low-density lipoprotein (VLDL) and low-density lipoprotein (LDL) were increased and the level of high-density lipoprotein (HDL) was decreased. These alterations were recouped back upon treatment with KA as well as SA when compared to cancer animals. The effects of KA were found to be more effective than SA. No significant alterations were observed in herbal preparation control animals when compared to control animals.  相似文献

Experimental data on the effect of the natural immunopeptide tuftsin and its synthetic counterpart on hemostasis parameters in vitro and in vivo when administered intranasally to animals (white laboratory rats) are given. It has been shown that in vitro both preparations induced a fibrin-depolymerization effect on fibrin that is not stabilized by factor XIIIa. For each preparation, it was noted that, when administered to rats, simultaneous antiplatelet, anticoagulant, and fibrin-depolymerization effects were observed in the blood plasma, with Selank having a more pronounced effect.  相似文献

An experimental investigation of the ability of the chondroitinsulphate preparation produced from cattle tracheas, of rumalon and chonsuride to stimulate the regeneration of cutaneous coverings in case of their injury was carried out. A strength of the surgical cicatrix and its elongation at rupture in two weeks' injections of chondroitinsulphate increased in comparison with control. A stimulating effect of chondroitinsulphate to regeneration of flesh wound in case of local single action didn't differ essentially from the effect of chonsuride. In case of application of the preparations, an area of wound for the rats received chondroitinsulphate was already on the eighth day twice as less in comparison with the animals treated with chonsuride. Thus, a stimulating effect of chondroitinsulphate preparation to the regeneration of damaged cutaneous coverings by both parenteral and local administration was shown.  相似文献

In experiments on 406 CBA mice the effect of antimicrobial preparations, used in combination with total gnotobiotic isolation, on the survival rate of totally irradiated animals was studied. The use of antimicrobial preparations was shown to considerably enhance the survival rate of these animals. Cyprofloxacin, a new preparation of the kinolone row, exhibited a good effect ensuring selective decontamination of the intestine. The use of cyprofloxacin prior to irradiation with the subsequent isolation of the animals in gnotobiotic chambers increased the survival rate of these animals by 2.7 times. But the maximum survival rate of irradiated mice was observed in those cases when antimicrobial preparations were used both before and after irradiation in combination with the total gnotobiotic isolation of the animals.  相似文献

Used four schemes of the administration of the preparation with different time of the exposition of the animals in an aerosol chamber were tested with their subsequent intraperitoneal challenge with K. pneumoniae virulent strain K16. Irrespective of the number of immunization courses, the administration of the preparation made at intervals of 1 day, or daily, did not ensure any protective effect, but only led to an insignificant increase in their survival time in comparison with nonimmunized animals. After intervals between immunizations were increased to 3 days the protective effect of aerosol immumization was obtained (the survival rate was 65-80 % and considerably differed from that of the controls). The protective effect of aerosol immunization thus obtained was comparable with the effectiveness immunization made in a single subcutaneous injection. Aerosol immunization resulted in low antibody titers to the antigens contained in the vaccine, while after a single subcutaneous injection high antibody titers to Klebsiella and Proteus antigens were detected. The antigen-stimulated blast transformation of spleen lymphocytes in mice subjected to aerosol immunizations in 5 exposures was high. After subcutaneous immunization significant changes in such characteristics were detected on day 15. The data thus obtained were indicative of good prospects in the development Immunovac VP-4 as the medicinal form intended for use in aerosols.  相似文献

The influence of ethanol, its metabolites and some opiates on enkephalinase A activity was studied in rat experiments in vitro after acute and chronic administration of ethanol. It was demonstrated that addition of ethanol to the reaction mixture activated enkephalinase A of the midbrain and hypothalamus of intact rats, the maximal effect being attained at an ethanol concentration of 10(-3) M. Multiple washings with buffer of the ethanol-preincubated membranous fraction of these brain structures in the control rats did not lead to a significant reduction in the activating effect of ethanol on enkephalinase A. No activation was recorded upon the use of an enzymatic preparation of the brain from chronically alcoholized animals. Morphine, naltrexon, beta-carbolines, salsolinol (10(-4) M) and acetaldehyde (10(-8)-10(-2) M) did not activate the enzyme. It is suggested that enkephalinase A activation in rats given ethanol is determined by a direct action of ethanol on the enzyme.  相似文献
