Effects of corrugation of the dragonfly wing on gliding performance   总被引:2,自引:0,他引:2  
We investigate the aerodynamic performance of the dragonfly wing, which has cross-sectional corrugation, via a static 2-dimensional unsteady simulation. Computational conditions are Re=150, 1400, and 10,000 with angles of attack ranging from 0° to 40°. From the computational results, lift coefficients are increased by the wing corrugation at all Reynolds number. However, the corrugation has little influence on the drag coefficients. The flows such as vortex in the valley of corrugation and near the edge of the corrugation are locally different from those of an elliptic wing. However, such local flows have little influence on the time averaged wing performance. From the numerical experiment presented in this study, it is determined that suction side corrugations of the wing have very little influence on increase of the lift coefficient at a positive angle of attack.  相似文献

High-resolution Particle-Image Velocimetry (PIV) and time-resolved force measurements were performed to analyze the impact of the comb-like structure on the leading edge of barn owl wings on the flow field and overall aerodynamic performance. The Reynolds number was varied in the range of 40,000 to 120,000 and the range of angle of attack was 0° to 6° for the PIV and -15° to +20° for the force measurements to cover the full flight envelope of the owl. As a reference, a wind-tunnel model which possessed a geometry based on the shape of a typical barn owl wing without any owl-specific adaptations was built, and measurements were performed in the aforementioned Reynolds number and angle of attack: range. This clean wing model shows a separation bubble in the distal part of the wing at higher angles of attack. Two types of comb-like structures, i.e., artificial serrations, were manufactured to model the owl's leading edge with respect to its length, thickness, and material properties. The artificial structures were able to reduce the size of the separation region and additionally cause a more uniform size of the vortical structures shed by the separation bubble within the Reynolds number range investigated, resulting in stable gliding flight independent of the flight velocity. However, due to increased drag coefficients in conjunction with similar lift coefficients, the overall aerodynamic performance, i.e., lift-to-drag ratio is reduced for the serrated models. Nevertheless, especially at lower Reynolds numbers the stabilizing effect of the uniform vortex size outperforms the lower aerodynamic performance.  相似文献

Quarantine treatment enables export of avocado fruit (Persea americana) to parts of the world that enforce quarantine against fruit fly. The recommended cold-based quarantine treatment (storage at 1.1°C for 14 days) was studied with two commercial avocado cultivars ‘Hass’ and ‘Ettinger’ for 2 years. Chilling injuries (CIs) are prevalent in the avocado fruit after cold-quarantine treatment. Hence, we examined the effect of integrating several treatments: modified atmosphere (MA; fruit covered with perforated polyethylene bags), methyl jasmonate (MJ; fruit dipped in 2.5 μM MJ for Hass or 10 μM MJ for Ettinger for 30 s), 1-methylcyclopropene (1-MCP; fruit treated with 300 ppb 1-MCP for 18 h) and low-temperature conditioning (LTC; a gradual decrease in temperature over 3 days) on CI reduction during cold quarantine. Avocado fruit stored at 1°C suffered from severe CI, lipid peroxidation, and increased expression of chilling-responsive genes of fruit peel. The combined therapeutic treatments alleviated CI in cold-quarantined fruit to the level in fruit stored at commercial temperature (5°C). A successful therapeutic treatment was developed to protect ‘Hass’ and ‘Ettinger’ avocado fruit during cold quarantine against fruit fly, while maintaining fruit quality. Subsequently, treated fruit stored at 1°C had a longer shelf life and less decay than the fruit stored at 5°C. This therapeutic treatment could potentially enable the export of avocado fruit to all quarantine-enforcing countries. Similar methods might be applicable to other types of fruit that require cold quarantine.  相似文献

Temperature affects both the timing and outcome of animal development, but the detailed effects of temperature on the progress of early development have been poorly characterized. To determine the impact of temperature on the order and timing of events during Drosophila melanogaster embryogenesis, we used time-lapse imaging to track the progress of embryos from shortly after egg laying through hatching at seven precisely maintained temperatures between 17.5°C and 32.5°C. We employed a combination of automated and manual annotation to determine when 36 milestones occurred in each embryo. D. melanogaster embryogenesis takes 33 hours at 17.5°C, and accelerates with increasing temperature to a low of 16 hours at 27.5°C, above which embryogenesis slows slightly. Remarkably, while the total time of embryogenesis varies over two fold, the relative timing of events from cellularization through hatching is constant across temperatures. To further explore the relationship between temperature and embryogenesis, we expanded our analysis to cover ten additional Drosophila species of varying climatic origins. Six of these species, like D. melanogaster, are of tropical origin, and embryogenesis time at different temperatures was similar for them all. D. mojavensis, a sub-tropical fly, develops slower than the tropical species at lower temperatures, while D. virilis, a temperate fly, exhibits slower development at all temperatures. The alpine sister species D. persimilis and D. pseudoobscura develop as rapidly as tropical flies at cooler temperatures, but exhibit diminished acceleration above 22.5°C and have drastically slowed development by 30°C. Despite ranging from 13 hours for D. erecta at 30°C to 46 hours for D. virilis at 17.5°C, the relative timing of events from cellularization through hatching is constant across all species and temperatures examined here, suggesting the existence of a previously unrecognized timer controlling the progress of embryogenesis that has been tuned by natural selection as each species diverges.  相似文献

Aerodynamic study of a simplified Dragonfly airfoil in gliding flight at Reynolds numbers below 10,000 is motivated by both pure scientific interest and technological applications. At these Reynolds numbers, the natural insect flight could provide inspiration for technology development of Micro UAV’s and more. Insect wings are typically characterized by corrugated airfoils. The present study follows a fundamental flow physics study (Levy and Seifert, 2009), that revealed the importance of flow separation from the first corrugation, the roll-up of the separated shear layer to discrete vortices and their role in promoting flow reattachment to the aft arc, as the leading mechanism enabling high-lift, low drag performance of the Dragonfly gliding flight. This paper describes the effect of systematic airfoil geometry variations on the aerodynamic properties of a simplified Dragonfly airfoil at Reynolds number of 6000.The parameter study includes a detailed analysis of small variations of the nominal geometry, such as corrugation placement or height, rear arc and trailing edge shape.Numerical simulations using the 2D laminar Navier-Stokes equations revealed that the flow accelerating over the first corrugation slope is followed by an unsteady pressure recovery, combined with vortex shedding. The latter allows the reattachment of the flow over the rear arc. Also, the drag values are directly linked to the vortices’ magnitude. This parametric study shows that geometric variations which reduce the vortices’ amplitude, as reduction of the rear cavity depth or the reduction of the rear arc and trailing edge curvature, will reduce the drag values. Other changes will extend the flow reattachment over the rear arc for a larger mean lift coefficients range; such as the negative deflection of the forward flat plate. These changes consequently reduce the drag values at higher mean lift coefficients.The detailed geometry study enabled the definition of a corrugated airfoil geometry with enhanced aerodynamic properties, such as range and endurance factors, as compared to the nominal airfoil studied in the literature.  相似文献

Summary The mode is demonstrated in which the three regions of the forewings of large grasshoppers, i.e. the costal, radial and anal parts (Fig. 2) are folded against each other during up- and downstroke (Fig. 1; measurements made by W. Zarnack). The aerodynamic effects of this wing folding are determined from the measurements of lift carried out on wing models in parallel air stream (Fig. 3) and on rotating wing models (Fig. 4). Accordingly wing bending during downstroke generates distinctly higher lift at small and medium angles of attack than a flat wing having the same dimensions and moving at the same speed. It generates less lift at high angles of attack. Wing bending during upstroke generates higher lift only at>15°. Lift remains greater than that of a flat wing up to the highest angles of attack. These conclusions are supported by measurements of downstroke wind velocity (Fig. 5). Therefore it is possible to change the lift of right and left wings in order to generate moments for flight control around all three axes of the animal's system of co-ordinates without changing the rough kinematics of the beating wings.

Flight in flies results from a feedback cascade in which the animal converts mechanical power produced by the flight musculature into aerodynamic forces. A major goal of flight research is to understand the functional significance of the various components in this cascade ranging from the generation of the neural code, the control of muscle mechanical power output, wing kinematics and unsteady aerodynamic mechanisms. Here, I attempted to draw a broad outline on fluid dynamic mechanisms found in flapping insect wings such as leading edge vorticity, rotational circulation and wake capture momentum transfer, as well as on the constraints of flight force control by the neuromuscular system of the fruit fly Drosophila. This system-level perspective on muscle control and aerodynamic mechanisms is thought to be a fundamental bridge in any attempt to link the function and performance of the various flight components with their particular role for wing motion and aerodynamic control in the behaving animal. Eventually, this research might facilitate the development of man-made biomimetic autonomous micro air vehicles using flapping wing motion for propulsion that are currently under construction by engineers.  相似文献

Introduction

The EOS stereoradiography system has shown to provide reliable varus/valgus (VV) measurements of the lower limb in 2D (VV2D) and 3D (VV3D) after total knee arthroplasty (TKA). Validity of these measurements has not been investigated yet, therefore the purpose of this study was to determine validity of EOS VV2D and VV3D.

Methods

EOS images were made of a lower limb phantom containing a knee prosthesis, while varying VV angle from 15° varus to 15° valgus and flexion angle from 0° to 20°, and changing rotation from 20° internal to 20° external rotation. Differences between the actual VV position of the lower limb phantom and its position as measured on EOS 2D and 3D images were investigated.

Results

Rotation, flexion or VV angle alone had no major impact on VV2D or VV3D. Combination of VV angle and rotation with full extension did not show major differences in VV2D measurements either. Combination of flexion and rotation with a neutral VV angle showed variation of up to 7.4° for VV2D; maximum variation for VV3D was only 1.5°. A combination of the three variables showed an even greater distortion of VV2D, while VV3D stayed relatively constant. Maximum measurement difference between preset VV angle and VV2D was 9.8°, while the difference with VV3D was only 1.9°. The largest differences between the preset VV angle and VV2D were found when installing the leg in extreme angles, for example 15° valgus, 20° flexion and 20° internal rotation.

Conclusions

After TKA, EOS VV3D were more valid than VV2D, indicating that 3D measurements compensate for malpositioning during acquisition. Caution is warranted when measuring VV angle on a conventional radiograph of a knee with a flexion contracture, varus or valgus angle and/or rotation of the knee joint during acquisition.  相似文献

In the midwestern and eastern U.S. populations of Drosophila melanogaster, the Standard gene arrangements show higher frequencies in the north than in the south. In a Missouri population, and to a lesser extent in a south Texas population, the frequencies of Standard chromosomes regularly rise during the cold season and drop during the warm season, thus paralleling the north-south frequency differences. In the Missouri population in 1976 and 1978, wild males were tested for their ability to fly to bait at different ambient temperatures. In both years, males flying in nature in the temperature range of 13° to 15° showed significantly higher frequencies of Standard chromosomes than did those flying in the 16° to 28° range. Wild males flying at 13° to 15° also have different thorax/wing proportions and significantly lower wing-loading indices than do those flying at 16° to 28°. Moreover, wild flies homozygous Standard in 2R and/or 3R have significantly lower wing-loading indices than flies carrying inversions in these arms. Thus, wild flies with high frequencies of Standard chromosomes are karyotypically northern, are selectively favored during the cold season, have a relatively low wing-load and are most capable of flying at critically low ambient temperatures.—In summary, in Missouri, presence or absence of the common cosmopolitan inversions is an important factor in low temperature adaptation, and at least part of the adaptive mechanism involves control of thorax/wing proportions and thus control of wing-loading.  相似文献

A comprehensive computational fluid-dynamics-based study of a pleated wing section based on the wing of Aeshna cyanea has been performed at ultra-low Reynolds numbers corresponding to the gliding flight of these dragonflies. In addition to the pleated wing, simulations have also been carried out for its smoothed counterpart (called the 'profiled' airfoil) and a flat plate in order to better understand the aerodynamic performance of the pleated wing. The simulations employ a sharp interface Cartesian-grid-based immersed boundary method, and a detailed critical assessment of the computed results was performed giving a high measure of confidence in the fidelity of the current simulations. The simulations demonstrate that the pleated airfoil produces comparable and at times higher lift than the profiled airfoil, with a drag comparable to that of its profiled counterpart. The higher lift and moderate drag associated with the pleated airfoil lead to an aerodynamic performance that is at least equivalent to and sometimes better than the profiled airfoil. The primary cause for the reduction in the overall drag of the pleated airfoil is the negative shear drag produced by the recirculation zones which form within the pleats. The current numerical simulations therefore clearly demonstrate that the pleated wing is an ingenious design of nature, which at times surpasses the aerodynamic performance of a more conventional smooth airfoil as well as that of a flat plate. For this reason, the pleated airfoil is an excellent candidate for a fixed wing micro-aerial vehicle design.  相似文献

Synaptic clustering on dendritic branches enhances plasticity, input integration and neuronal firing. However, the mechanisms guiding axons to cluster synapses at appropriate sites along dendritic branches are poorly understood. We searched for such a mechanism by investigating the structural overlap between dendritic branches and axons in a simplified model of neuronal networks - the hippocampal cell culture. Using newly developed software, we converted images of meshes of overlapping axonal and dendrites into topological maps of intersections, enabling quantitative study of overlapping neuritic geometry at the resolution of single dendritic branch-to-branch and axon-to-branch crossings. Among dendro-dendritic crossing configurations, it was revealed that the orientations through which dendritic branches cross is a regulated attribute. While crossing angle distribution among branches thinner than 1 µm appeared to be random, dendritic branches 1 µm or wider showed a preference for crossing each other at angle ranges of either 50°–70° or 80°–90°. It was then found that the dendro-dendritic crossings themselves, as well as their selective angles, both affected the path of axonal growth. Axons displayed 4 fold stronger tendency to traverse within 2 µm of dendro-dendritic intersections than at farther distances, probably to minimize wiring length. Moreover, almost 70% of the 50°–70° dendro-denritic crossings were traversed by axons from the obtuse angle’s zone, whereas only 15% traversed through the acute angle’s zone. By contrast, axons showed no orientation restriction when traversing 80°–90° crossings. When such traverse behavior was repeated by many axons, they converged in the vicinity of dendro-dendritic intersections, thereby clustering their synaptic connections. Thus, the vicinity of dendritic branch-to-branch crossings appears to be a regulated structure used by axons as a target for efficient wiring and as a preferred site for synaptic clustering. This synaptic clustering mechanism may enhance synaptic co-activity and plasticity.  相似文献

Aims

The 3D geometry of individual vascular smooth muscle cells (VSMCs), which are essential for understanding the mechanical function of blood vessels, are currently not available. This paper introduces a new 3D segmentation algorithm to determine VSMC morphology and orientation.

Methods and Results

A total of 112 VSMCs from six porcine coronary arteries were used in the analysis. A 3D semi-automatic segmentation method was developed to reconstruct individual VSMCs from cell clumps as well as to extract the 3D geometry of VSMCs. A new edge blocking model was introduced to recognize cell boundary while an edge growing was developed for optimal interpolation and edge verification. The proposed methods were designed based on Region of Interest (ROI) selected by user and interactive responses of limited key edges. Enhanced cell boundary features were used to construct the cell’s initial boundary for further edge growing. A unified framework of morphological parameters (dimensions and orientations) was proposed for the 3D volume data. Virtual phantom was designed to validate the tilt angle measurements, while other parameters extracted from 3D segmentations were compared with manual measurements to assess the accuracy of the algorithm. The length, width and thickness of VSMCs were 62.9±14.9μm, 4.6±0.6μm and 6.2±1.8μm (mean±SD). In longitudinal-circumferential plane of blood vessel, VSMCs align off the circumferential direction with two mean angles of -19.4±9.3° and 10.9±4.7°, while an out-of-plane angle (i.e., radial tilt angle) was found to be 8±7.6° with median as 5.7°.

Conclusions

A 3D segmentation algorithm was developed to reconstruct individual VSMCs of blood vessel walls based on optical image stacks. The results were validated by a virtual phantom and manual measurement. The obtained 3D geometries can be utilized in mathematical models and leads a better understanding of vascular mechanical properties and function.  相似文献

The mechanism by which sequence non-specific DNA-binding proteins enhance DNA flexibility is studied by examining complexes of double-stranded DNA with the high mobility group type B proteins HMGB2 (Box A) and HMGB1 (Box A+B) using atomic force microscopy. DNA end-to-end distances and local DNA bend angle distributions are analyzed for protein complexes deposited on a mica surface. For HMGB2 (Box A) binding we find a mean induced DNA bend angle of 78°, with a standard error of 1.3° and a SD of 23°, while HMGB1 (Box A+B) binding gives a mean bend angle of 67°, with a standard error of 1.3° and a SD of 21°. These results are consistent with analysis of the observed global persistence length changes derived from end-to-end distance measurements, and with results of DNA-stretching experiments. The moderately broad distributions of bend angles induced by both proteins are inconsistent with either a static kink model, or a purely flexible hinge model for DNA distortion by protein binding. Therefore, the mechanism by which HMGB proteins enhance the flexibility of DNA must differ from that of the Escherichia coli HU protein, which in previous studies showed a flat angle distribution consistent with a flexible hinge model.  相似文献

The remarkable maneuverability of flying animals results from precise movements of their highly specialized wings. Bats have evolved an impressive capacity to control their flight, in large part due to their ability to modulate wing shape, area, and angle of attack through many independently controlled joints. Bat wings, however, also contain many bones and relatively large muscles, and thus the ratio of bats’ wing mass to their body mass is larger than it is for all other extant flyers. Although the inertia in bat wings would typically be associated with decreased aerial maneuverability, we show that bat maneuvers challenge this notion. We use a model-based tracking algorithm to measure the wing and body kinematics of bats performing complex aerial rotations. Using a minimal model of a bat with only six degrees of kinematic freedom, we show that bats can perform body rolls by selectively retracting one wing during the flapping cycle. We also show that this maneuver does not rely on aerodynamic forces, and furthermore that a fruit fly, with nearly massless wings, would not exhibit this effect. Similar results are shown for a pitching maneuver. Finally, we combine high-resolution kinematics of wing and body movements during landing and falling maneuvers with a 52-degree-of-freedom dynamical model of a bat to show that modulation of wing inertia plays the dominant role in reorienting the bat during landing and falling maneuvers, with minimal contribution from aerodynamic forces. Bats can, therefore, use their wings as multifunctional organs, capable of sophisticated aerodynamic and inertial dynamics not previously observed in other flying animals. This may also have implications for the control of aerial robotic vehicles.  相似文献

This study investigated the effects of wrist angle, sex, and handedness on the changes in the median nerve cross-sectional area (MNCSA) and median nerve diameters, namely longitudinal diameter (D1) and vertical diameter (D2). Ultrasound examination was conducted to examine the median nerve at the proximal carpal tunnel in both dominant and nondominant hands of men (n = 27) and women (n = 26). A total of seven wrist angles were examined: neutral; 15°, 30°, and 45° extension; and 15°, 30°, and 45° flexion. Our results indicated sexual dimorphism and bilateral asymmetry of MNCSA, D1 and D2 measurements. MNCSA was significantly reduced when the wrist angle changed from neutral to flexion or extension positions. At flexion positions, D1 was significantly smaller than that at neutral. In contrast, at extension positions, D2 was significantly smaller than that at neutral. In conclusion, this study showed that MNCSA decreased as the wrist angle changed from neutral to flexion or extension positions in both dominant and nondominant hands of both sexes, whereas deformation of the median nerve differed between wrist flexion and extension.  相似文献

Microstructures on insect wings can promote directional drop shedding, and the local orientation of these structures is expected to facilitate drop removal. However, microstructures may exhibit very different orientations at different locations on the wing. Using the march fly Penthetria heteroptera, we propose that local orientation of small hairs (microtrichia) reflects a balance of three nonexclusive strategies: (1) preventing water from becoming stuck in intervenous grooves (microtrichia point upslope), (2) shedding water off the wing as readily as possible (microtrichia point towards the nearest edge), and, (3) shedding water away from the body (microtrichia point distally). We present evidence for all three and show that local microtrichial orientation is seldom determined by any one factor. We develop a mathematical model that employs factor-specific weighting values determined via optimization. Our predictions are tested against the orientation of microtrichia randomly sampled from a P. heteroptera specimen. Using the best-fit weighting parameters, the model displays a median residual of 20°; no residual is greater than 46°. The model also reproduces qualitative aspects of microtrichial orientation, such as bifurcation midway between veins and convergence toward peaks. This strong correspondence between modelled and observed orientation supports the role of microtrichia as directional antiwetting devices and highlights the importance of considering both function and wing geometry to explain the organization of natural microstructure arrays.  相似文献

Background

Trichoderma reesei is a key cellulase source for economically saccharifying cellulosic biomass for the production of biofuels. Lignocellulose hydrolysis at temperatures above the optimum temperature of T. reesei cellulases (~50°C) could provide many significant advantages, including reduced viscosity at high-solids loadings, lower risk of microbial contamination during saccharification, greater compatibility with high-temperature biomass pretreatment, and faster rates of hydrolysis. These potential advantages motivate efforts to engineer T. reesei cellulases that can hydrolyze lignocellulose at temperatures ranging from 60–70°C.

Results

A B-factor guided approach for improving thermostability was used to engineer variants of endoglucanase I (Cel7B) from T. reesei (TrEGI) that are able to hydrolyze cellulosic substrates more rapidly than the recombinant wild-type TrEGI at temperatures ranging from 50–70°C. When expressed in T. reesei, TrEGI variant G230A/D113S/D115T (G230A/D113S/D115T Tr_TrEGI) had a higher apparent melting temperature (3°C increase in Tm) and improved half-life at 60°C (t1/2 = 161 hr) than the recombinant (T. reesei host) wild-type TrEGI (t1/2 = 74 hr at 60°C, Tr_TrEGI). Furthermore, G230A/D113S/D115T Tr_TrEGI showed 2-fold improved activity compared to Tr_TrEGI at 65°C on solid cellulosic substrates, and was as efficient in hydrolyzing cellulose at 60°C as Tr_TrEGI was at 50°C. The activities and stabilities of the recombinant TrEGI enzymes followed similar trends but differed significantly in magnitude depending on the expression host (Escherichia coli cell-free, Saccharomyces cerevisiae, Neurospora crassa, or T. reesei). Compared to N.crassa-expressed TrEGI, S. cerevisiae-expressed TrEGI showed inferior activity and stability, which was attributed to the lack of cyclization of the N-terminal glutamine in Sc_TrEGI and not to differences in glycosylation. N-terminal pyroglutamate formation in TrEGI expressed in S. cerevisiae was found to be essential in elevating its activity and stability to levels similar to the T. reesei or N. crassa-expressed enzyme, highlighting the importance of this ubiquitous modification in GH7 enzymes.

Conclusion

Structure-guided evolution of T. reesei EGI was used to engineer enzymes with increased thermal stability and activity on solid cellulosic substrates. Production of TrEGI enzymes in four hosts highlighted the impact of the expression host and the role of N-terminal pyroglutamate formation on the activity and stability of TrEGI enzymes.

Bending, in addition to compression, is recognized to be a common loading pattern in long bones in animals. However, due to the technical difficulty of measuring bone deformation in humans, our current understanding of bone loading patterns in humans is very limited. In the present study, we hypothesized that bending and torsion are important loading regimes in the human tibia. In vivo tibia segment deformation in humans was assessed during walking and running utilizing a novel optical approach. Results suggest that the proximal tibia primarily bends to the posterior (bending angle: 0.15°–1.30°) and medial aspect (bending angle: 0.38°–0.90°) and that it twists externally (torsion angle: 0.67°–1.66°) in relation to the distal tibia during the stance phase of overground walking at a speed between 2.5 and 6.1 km/h. Peak posterior bending and peak torsion occurred during the first and second half of stance phase, respectively. The peak-to-peak antero-posterior (AP) bending angles increased linearly with vertical ground reaction force and speed. Similarly, peak-to-peak torsion angles increased with the vertical free moment in four of the five test subjects and with the speed in three of the test subjects. There was no correlation between peak-to-peak medio-lateral (ML) bending angles and ground reaction force or speed. On the treadmill, peak-to-peak AP bending angles increased with walking and running speed, but peak-to-peak torsion angles and peak-to-peak ML bending angles remained constant during walking. Peak-to-peak AP bending angle during treadmill running was speed-dependent and larger than that observed during walking. In contrast, peak-to-peak tibia torsion angle was smaller during treadmill running than during walking. To conclude, bending and torsion of substantial magnitude were observed in the human tibia during walking and running. A systematic distribution of peak amplitude was found during the first and second parts of the stance phase.  相似文献

Understanding how the fruit microclimate affects ascorbate (AsA) biosynthesis, oxidation and recycling is a great challenge in improving fruit nutritional quality. For this purpose, tomatoes at breaker stage were harvested and placed in controlled environment conditions at different temperatures (12, 17, 23, 27 and 31°C) and irradiance regimes (darkness or 150 µmol m-2 s-1). Fruit pericarp tissue was used to assay ascorbate, glutathione, enzymes related to oxidative stress and the AsA/glutathione cycle and follow the expression of genes coding for 5 enzymes of the AsA biosynthesis pathway (GME, VTC2, GPP, L-GalDH, GLDH). The AsA pool size in pericarp tissue was significantly higher under light at temperatures below 27°C. In addition, light promoted glutathione accumulation at low and high temperatures. At 12°C, increased AsA content was correlated with the enhanced expression of all genes of the biosynthesis pathway studied, combined with higher DHAR and MDHAR activities and increased enzymatic activities related to oxidative stress (CAT and APX). In contrast, at 31°C, MDHAR and GR activities were significantly reduced under light indicating that enzymes of the AsA/glutathione cycle may limit AsA recycling and pool size in fruit pericarp, despite enhanced expression of genes coding for AsA biosynthesis enzymes. In conclusion, this study confirms the important role of fruit microclimate in the regulation of fruit pericarp AsA content, as under oxidative conditions (12°C, light) total fruit pericarp AsA content increased up to 71%. Moreover, it reveals that light and temperature interact to regulate both AsA biosynthesis gene expression in tomato fruits and AsA oxidation and recycling.  相似文献
