Research into infertility in the dromedary bull, as reported during the last two decades, is reviewed with emphasis on causes and effects. Reproductive activity of such animals is naturally limited by a breeding season, though with enough encouragement some may mate with oestrous females out of season but a full fertilization potential can in no way be expected. It is essential that any female presented to a bull is capable of reproducing. The presentation of a subfertile or infertile female due to infection or physiological abnormality will adversely affect the female's ability to conceive and, therefore, the apparent fertility rates of the bull she was put to. The average number of successful services a bull could be expected to perform is two per day. Dromedary bulls with large testes have larger sperm outputs and can cope with more than two females per day providing that they are given adequate periods of rest, 1-2 days every 10 days or so, in conjunction with appropriate nutrition throughout the season. Anabolic steroids or testosterone therapies, which are sometimes used in an attempt to improve male characteristics and bull libido, are not recommended for dromedary bulls in breeding work. Such steroids result in a decrease in testicular size and weight with fewer sperm per gram of testicular tissue being found and the sperm produced also have lower motility rates. Pain associated with the act of mating a she-camel, due to injuries or inflammation in the scrotum, testes, prepuce and sheath, can cause a permanent reduction in bull libido. Camel bulls achieving pregnancy rates more than 60% have had consistently higher spermatozoal concentrations and kinematic variables derived by the computerized cell motion analyzer (CMA) system. As far as physical capabilities are concerned, 3-year-old dromedary bulls, which have reached puberty, have been shown to be perfectly capable of fertilizing a female, but they have a limited sperm production to perform consistently throughout the season in a large herd. By 4.5-5.0 years of age, they are capable of producing adequate numbers of sperm to mate as many as females as an adult bull but fertilizing capacity is not attained until 6 years of age on average. Hyperoestrogenaemia, associated with autoimmune thyroiditis and trypanosomiasis, suppresses the secretion of testicular testosterone and augments the release of testicular histamine, which appears mandatory for quantitative reduction/loss of advanced spermatogenic cells in infertile dromedary bulls.  相似文献

Experiments were conducted to evaluate two extenders (egg-yolk Tris and egg-yolk lactose), varying concentrations of two cryopreservatives (glycerol and dimethyl sulfoxide), and rates for cooling to 5 degrees C, cooling from 5 to -100 degrees C, and warming for canine spermatozoa packaged in 0.5-ml French straws. At optimal concentrations of glycerol, egg-yolk Tris extender was superior to egg-yolk lactose in preserving spermatozoal motility. Addition of dimethyl sulfoxide, alone or in combination with glycerol in either extender, was not beneficial to spermatozoal survival after thawing. Canine spermatozoa withstood a range of cooling and equilibration times with no detrimental effect on spermatozoal motility prior to freezing. However, there were differences in spermatozoal motility immediately after thawing; these differences were variable, resulting in a cooling time by equilibration time interaction. Spermatozoal motility after thawing was best preserved by freezing in egg-yolk Tris extender containing 2-4% glycerol, using a moderate rate of cooling from 5 to -100 degrees C (-5 degrees C/min from 5 to -15 degrees C, then -20 degrees C/min from -15 to -100 degrees C). Three of 12 bitches inseminated intravaginally with semen frozen using this protocol became pregnant.  相似文献

A total of 92 range beef bulls (Hereford = 60; Angus = 32) were given a breeding soundness examination (BSE) and two assessments for sex drive prior to their use in 23 breeding trials employing estrous synchronized females. Bulls were in three age groups: yearlings (n=29), two year olds (n=36), and three year olds and older (n=27). All yearling bulls were virgins, but the majority of the older bulls had previous mating experience. Angus bulls were superior (P<0.01) to Herefords in spermatozoal morphology and BSE score. Scrotal circumference increased with age beyond two years in Angus bulls but not in Herefords. Spermatozoal abnormalities generally decreased with age. BSE scores did not differ significantly among age groups. Apart from number of mounts, measures of sex drive did not differ with age or breed of bulls. This represents qualified justification for the current practice of using the same sex-drive assessment procedures for Bos taurus bulls of various ages and breeds.  相似文献

The effects of four vaginal lubricants on progressive spermatozoal motility were evaluated. Neat semen was exposed to 0, 5, or 10% (w/v) of H-R, sterile K-Y, nonsterile K-Y or Maxilube lubricating jellies for 10 min at 37 degrees C and then extended to 10x10(6) spermatozoa/ml. Spermatozoal motility was evaluated after 0, 1, 2, 4 and 6 or 8 h of incubation at 37 degrees C. For bovine spermatozoa, sterile K-Y jelly at 10% suppressed motility (P<0.05), but nonsterile K-Y, H-R and Maxilube jellies had no effect. Maxilube was toxic (P<0.01) to canine spermatozoa and is not recommended for use during collection or insemination of canine semen. Exposure of equine semen to 10% H-R jelly had no effect on spermatozoal motility, whereas 10% sterile K-Y, nonsterile K-Y or Maxilube jellies suppressed (P<0.05) motility. For all three species, the new, nonsterile K-Y jelly was no more deleterious to spermatozoal motility than the old, sterile K-Y jelly, and H-R jelly also was satisfactory. Fertility tests are required to determine the effect of these products on fertility.  相似文献

Spermatozoa from different bucks were stained with different fluorochromes, mixed, and inseminated heterospermically. By altering the interval between insemination and luteinizing hormone injection, spermatozoa were allowed to reside in the female tract approximately 5, 10, or 15 h prior to ovulation. The number of functional spermatozoa, from each male of a pair used, that was transported to the site of fertilization was estimated by counting total number of differently stained spermatozoa that surrounded or fertilized each oocyte. Spermatozoa from split ejaculates within a male competed against each other equally, indicating that the staining procedure did not affect fertilization or functional spermatozoal transport rates. Three pairs of males with high initial semen quality (greater than 80% motility) differed in fertility primarily due to functional spermatozoal transport. Spermatozoal survival in the female tract and capacitation time played a role in differences in male fertility when heterospermic insemination occurred at variable times relative to ovulation. Differences in fertilization not accounted for by spermatozoal transport ratio raised the possibility that rate of egg penetration due to acrosomal enzyme differences may be important in determining male fertility. Therefore, total acrosin, hyaluronidase, and arylsulfatase activity in spermatozoa from specific bucks used in fertilization experiments were determined. Although there were trends favoring high fertility when enzyme content was higher, the difference was significant only for arylsulfatase in one buck.  相似文献

Post-thaw motility, velocity and acrosome integrity of frozen semen were determined in 18 bulls with varying fertility (average non-return rates: 71.3 (± 2.8) - range: 65.2-75.7). Five semen straws were investigated from each bull. The average values for sperm motility (percentage motile spermatozoa), sperm velocity (graded from 0-3) and acrosome integrity (proportion of spermatozoa with intact acrosome) were 67.5%, 2.5 and 79.3%, respectively. Significant correlations were found between sperm motility and velocity, but not between sperm motility and acrosome integrity. Both sperm motility and velocity were significantly related to bull fertility. It was concluded that of the post-thaw semen characteristics investigated in this study these 2 parameters provided a reliable basis for prediction of bull fertility.  相似文献

Two experiments were designed to evaluate cooling rates and storage temperatures for stallion spermatozoa extended in caprogen (CAP), Cornell University extender (CUE), heated skimmilk (SM) and a nonfat-dried milk solids glucose extender (NFDMS-glucose). In Experiment 1, each extender was evaluated in a separate but similar 4 × 4 × 6 factorial trial using two ejaculates from each of six stallions. Aliquots of 66 × 106 spermatozoa were transferred to each of 16 coded tubes and extended to 6 ml with SM, CAP, CUE or NFDMS-glucose. Four tubes of extended semen were either plunged into 5C water or cooled at a rate of ?1.0, ?0.5, or ?0.2C/min. Within each treatment, one tube of extended semen was maintained at 20C, 15C, 10C or 5C. Progressive spermatozoal motility was estimated immediately after dilution (0 h) and at 4, 8, 12, 24 and 36 h. Regardless of extender, all three slower cooling rates were superior (P<0.05) to plunging to 5C; storage temperatures of 20C and 15C were superior (P<0.05) for maintaining spermatozoal motility. Experiment 2 was designed so that all extenders could be evaluated simultaneously. Since CUE resulted in an immediate depression of spermatozoal motility, it was not evaluated further. Semen was collected from 12 stallions and each ejaculate was extended in SM, CAP and NFDMS-glucose. Semen was cooled at ?1.0C/min and maintained at either 20C or 15C. Spermatozoal motility was assessed as in Experiment 1. Overall, the CAP and NFDMS-glucose extenders were superior (P<0.05) to SM for maintenance of spermatozoal motility. Storage at 20C or 15C resulted in similar (P>0.05) spermatozoal motility. Two fertility trials compared the use of SM and NFDMS-glucose extenders. Embryo recovery 6 days post-ovulation (Experiment 3) and pregnancy rate 50 days postestrus (Experiment 4) was similar (P>0.05) for mares inseminated with spermatozoa extended in SM or NFDMS-glucose.  相似文献

This study was undertaken to investigate the relationship between the cationic polymer (poly-L-lysine) concentration and microcapsule membrane thickness, maintenance of spermatozoal motility in vitro, and pregnancy rate in 335 oestrous synchronized Friesian heifers. Semen was extended in CAPROGEN containing 5% egg yolk and a final encapsulated spermatozoal concentration of 20 × 106 spermatozoa ml−1. Four concentrations of poly-L-lysine were studied (0.025, 0.05, 0.075, and 0. 1%, w/v). Microcapsule membrane thickness resulting from these concentrations was 3.22 ± 0.54 μm (mean ± SD), 5.30 ± 0.31 μm, 7.12 ± 0.41 μm, and 7.44 ± 0.85 μm, respectively (P < 0.05). Spermatozoal viability, as assessed by motility estimates at 24 h intervals during 120 h of incubation at 37°C, was not influenced by polymer concentration or different than unencapsulated controls. For fertility evaluation approximately 65 Friesian heifers were inseminated with spermatozoa either unencapsulated or encapsulated with one of the four polymer concentrations. Oestrous synchronization was accomplished with the combination of a progesterone-impregnated CIDR-B® device containing a 10 mg oestradiol benzoate capsule inserted for 10 days with administration of 12.5 mg of prostaglandin F on Day 6 of CIDR-B® insertion. Heifers were inseminated in the uterine corpus at 24 h after CIDR-B® removal which constituted the pro-oestrous stage of the cycle for 95.5% of the heifers. Inseminate dose rate was 5 × 106 spermatozoa in 0.25 ml. Pregnancy rates were similar for heifers inseminated with encapsulated and unencapsulated spermatozoa (49.4 vs. 48.6%).From these studies we conclude that poly-L-lysine concentration does influence the microcapsule membrane thickness without affecting maintenance of spermatozoal motility in vitro or fertility of oestrous synchronised Friesian heifers.  相似文献

This study determined the relationship between two measures of field fertility of 11 high-use Australian artificial insemination (AI) dairy bulls and thirty standard laboratory assessments of spermatozoal post-thaw viability.The two measures of field fertility used, conception rates (cCR) and non-return rates (cNRR), were both corrected for all major non-bull variables. Sperm viability assessments were conducted on semen collected within the same season as that used to derive the field fertility estimates. These assessments measured sperm concentration, motility, morphology and membrane integrity at thawing, after 2h incubation and after the swim-up sperm selection procedure. Derivations of these measures and in vitro embryo fertilizing and developmental capacity were also determined. The Genstat Statistical Package [Genstat 5 Release 4.2 Reference Manual, VSN International, Oxford, 2000] was used to conduct an analysis of variance on the viability parameters across semen straws and bulls, and to calculate the strength of correlation between each semen parameter, cNRR and cCR in a correlation matrix. Step forward multiple regression identified the combination of semen parameters that were most highly correlated with cCR and with cNRR.The sperm parameters identified as being most predictive of cCR were the percentage of morphologically normal sperm immediately post-thaw (zeroNorm), the number of morphologically normal sperm after the swim-up procedure (nSuNorm), and the rate of zygote cleavage in vitro (Clv); the predictive equation formed by these parameters accounted for 70% of variance. The predictive equation produced for cNRR contained the variables zeroNorm, the proportion of membrane intact sperm after 2h incubation at 37 degrees C (twoMem) and Clv and accounted for 76.5% of the variation. ZeroNorm was found to be consistent across straws and semen batches within-bull and the sperm parameter with the strongest individual predictive capacity for both cCR (P=0.1) and cNRR (P=0.001). Post-thaw sperm parameters can be used to predict field fertility of Australian dairy sires; the calculated predictive equations are particularly useful for identifying and monitoring bulls of very high and very low potential fertility within a group.  相似文献

Forty Santa Gertrudis bulls were used to examine relationships among scrotal circumference, seminal quality, libido, and fertility [assessed as the percent pregnant of estrous females (PE rate) and the percent pregnant of females mated (PM rate)]. These bulls were selected from 220 two year old bulls to represent variations in scrotal circumference and seminal quality. Each of the 40 bulls were exposed to 100 cyclic Santa Gertrudis heifers for a 4-day (96 hr) breeding period. The number of estrous females available to each bull varied from 12 to 27. A breeding soundness examination (BSE) was conducted on each bull approximately 45 days prior to the 4-day breeding period and immediately after the 4-day breeding period. The three components of the BSE scroe (scrotal circumference, spermatozoal abnormalities and spermatozoal motility) were not significantly correlated with PE rate or PM rate at either evaluation. There was no significant correlation between PM rate and scrotal circumference; however, each of the 4 bulls having a scrotal circumference less than 30 cm had a PM rate below 31%. Relationships between seminal quality and PM rate were unclear and differed between the two evaluations. There was a trend for bulls having poor seminal quality at the first evaluation to improve by the second evaluation. Consequently, fluctuations in seminal quality between evaluations is one possible explanation for low correlations between seminal parameters and PM rate. Libido (number mated/number in estrus x 100) was positively correlated (r = 0.44) with PE rate. Using a stepwise regression procedure, the independent variables accounting for the most variation in PE rate (dependent variable) included libido, secondary spermatozoal abnormalities, and BSE score (r(2) = 0.44). Results of this study indicate that current methods of fertility evaluation did not accurately predict the fertility of individual Santa Gertrudis bulls as measured by PE rate and PM rate during a 4 day breeding period.  相似文献

This study is a retrospective analysis comparing data on yearling bull breeding soundness evaluation (BSE) subjected to 3 different classification systems: the Society for Theriogenology (SFT) 1983 and 1993 systems, and the Western Canadian Association of Bovine Practitioners (WC) 1993 system. Data were collected at 5 performance bull-test stations located in South Carolina and Tennessee from 1986 through 1996. Yearling bulls (n=2898) that were 10 to 20 mo of age were used in the analysis. To simplify analysis, bulls were determined to be either satisfactory or unsatisfactory potential breeders (including those classified as questionable, deferred or unsatisfactory). Data were analyzed 1) within location where a location was treated as an individual experiment, 2) with breeds and locations collapsed, and 3) within age-group where each age-group was treated as an individual experiment. An ANOVA was performed using GLM procedures of SAS, and this model was used to generate least square means for the proportion of bulls classified as satisfactory and the 5 possible unsatisfactory outcomes due to inadequacies in scrotal circumference, spermatozoal morphology, spermatozoal motility, a combination of inadequate spermatozoal morphology and motility, or physical abnormalities. Inadequate scrotal circumference or physical abnormality did not affect differences for BSE outcomes among systems. Using the SFT93 system, bulls failed at a higher rate due to inadequate spermatozoal morphology (P < 0.05) than when subjected to the SFT83 system. In the WC93 system, a higher percentage of bulls failed due to inadequate spermatozoal motility and to a combination of inadequate spermatozoal morphology and motility than in the other 2 systems (P < 0.05). The proportion of bulls in this data failing under the WC93 system appears to be the result of overestimation.  相似文献

Better procedures for freezing and thawing equine sperm are needed since variable fertility is obtained when cryopreserved sperm are used. To evaluate current methods of freezing equine sperm, we examined spermatozoal quality by means of two new techniques. These measured the integrity of plasma-acrosomal membranes by immunofluorescent analyses of binding of an antibody specific to the acrosome and evaluated eight parameters of spermatozoal motion using a fully automated computerized system. Five ejaculates from each of eight stallions were processed for freezing in egg yolk-lactose extender with 4% glycerol. Spermatozoal quality was assessed at four different points: at less than 15 min after collecting and before processing (Step 1); after centrifugation and just before freezing (Step 2); immediately after thawing less than 3 h after freezing (Step 3); and immediately after thawing 10 to 20 d after freezing (Step 4). Acrosome-specific monoclonal antibody detected differences (P <0.05) among steps and ejaculates within stallions. All parameters of spermatozoal motion, including the percentage of motile sperm, percentage of progressively motile sperm, curvilinear velocity, straight line velocity, linearity, amplitude of lateral head displacement, and radius of the average path for circularly swimming sperm, differed (P <0.05) among steps, and most of these parameters differed among ejaculates within a stallion and among stallions. For Steps 2 and 3, 62 and 37% of the sperm were motile, and 56 and 23% of all motile sperm had a curvilinear velocity of >100 mum/sec. Most damage to sperm occurred as a result of freezing-thawing, whereas centrifugation of sperm caused only minor damage.  相似文献

Spermatozoal motility and enzyme leakage during cryopreservation of ram spermatozoa was evaluated in the presence and absence of antifreeze proteins (AFP). Loss of spermatozoal motility due to the cryopreservation process was reduced by 10% in the presence of AFP. Samples of the diluted semen at various stages of the cryopreservation process were assayed for aspartate transaminase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP), acid phosphatase (ACP) and lactate dehydrogenase (LDH). The levels of AST, ALT and ACP were low in ram spermatozoa and were not considered suitable for the objective measurement of spermatozoal damage. ALP leaked during the cooling and freezing process, whereas LDH leakage was prevalent during cooling, freezing and post-thaw incubation of spermatozoa. Changes in ALP and LDH could be used as marker enzymes in the development of semen processing protocols and of semen diluents.  相似文献

Frozen semen specimens from 22 Holstein bulls representing a wide range of field fertility levels or nonreturn rates (NRR) were used in this study. Semen specimens were thawed at 37 degrees C for a minimum of 30 sec, followed by assessment via a routine semen analysis (RSA) and other sperm functional tests. The RSA was performed by assessing sperm count, motility and morphological characteristics. Other sperm functional tests were performed by assessing the acrosomal membrane integrity, sperm penetration into the cervical mucus and the sperm membrane functional integrity. Following assessment of sperm characteristics, the fertility data of the various bulls were compared to the RSA and the functional tests results. Bulls of high and low fertility were similar in terms of sperm count and progressive motility (P > 0.05). Other characteristics measured by the RSA and functional tests were significantly higher in high fertility bulls (P < 0.05). Correlation coefficients among the various sperm characteristics and fertility of bulls were highly significant (P < 0.01). The highest correlation coefficients between sperm characteristics and fertility were obtained for motility (r = 0.53; P < 0.01), normal morphology (r = 0.59; P < 0.01) and swollen spermatozoa (r = 0.57; P < 0.01). Analysis of specific sperm swelling patterns showed that those patterns considered to reflect maximal sperm swelling were indicative of high fertility.  相似文献

Prior to use in single-sire mating trials, ninety-two beef bulls were evaluated to determine the relationships between. Breeding Soundness Examination (BSE) and sex drive assessment categories. A BSE score was given to each bull based on scrotal circumference, spermatozoal motility and morphology. Eighty bulls were classified as satisfactory potential breeders, while 12 were classified as questionable potential breeders. Each bull was exposed to two, 10 min modified libido/serving capacity tests. Sixty-nine bulls were classified as high libido and 23 were classified as medium libido. Differences in scrotal circumference and spermatozoal motility did not influence (P > 0.05) the BSE classifications. However, differences in primary, secondary and total spermatozoal abnormalities were the major factors affecting the BSE classification. Bulls classified as high libido serviced three times more (P < 0.01) during the two test periods than bulls classified as medium libido. High libido bulls were superior in all sex drive traits. However, bulls in the two libido categories did not differ (P > 0.05) in semen quality or scrotal circumference. There was no relationship (P > 0.05) between BSE and sex drive traits (r = -0.16 to 0.24).  相似文献

Preserved stallion semen often has decreased spermatozoal motility and fertility that can vary significantly between individual stallions. It is not known whether the medium used for extending equine sperm contributes to these decreases by inducing premature capacitation during storage. If spermatozoa undergo capacitation or acrosome reaction prior to insemination, this could result in a diminished capacity to penetrate the cumulus mass and fertilize the egg. We hypothesized that skim milk-based semen extenders, similar to those used in cooled storage, stabilize sperm membranes and prolong sperm motility and longevity. However, this could decrease the efficiency of sperm to undergo subsequent capacitation in vivo. This study was designed to evaluate the effects from two media on sperm function. Spermatozoal motility was analyzed, intracellular calcium was measured, and the ability of sperm to undergo acrosome reaction was compared after incubation in a skim milk extender (SME) and Tyrode's medium containing albumin, lactate, and pyruvate (TALP) at 37 degrees C. Results suggest that the SME facilitated capacitation as detected by an increase in both intracellular calcium and acrosome reactions, and a decrease in motility, as compared to TALP. Our data support a shortened functional lifespan for equine sperm in skim milk extender, which indicates that further refinements in cooled semen preservation are required to improve fertility of transported equine semen.  相似文献

Frozen-thawed spermatozoa from Friesian bulls held at stud in Ireland were used to assess the effect of ionophore on motility, acrosome reaction and heterologous in vitro fertilization. Bovine spermatozoa penetrated zone-free hamster oocytes following treatment with calcium ionophore in the absence of bovine serum albumin (BSA) and in the presence of 10 mM caffeine. Sperm velocity was stimulated in concentrations of caffeine <2.5 mM following dilution with medium containing BSA. Sperm attachment to the plasmalemma showed no association with penetration rates of zona-free hamster oocytes. Penetrated oocytes in regimens with >0.1 mM ionophore did not progress through Meiosis II. Increasing concentrations of ionophore induced the acrosome reaction more rapidly, although this was associated with reduced motility. Hyperactive motility was observed in calcium ionophore-treated spermatozoa which were capable of penetrating zona-free hamster oocytes. Sperm velocity remained unchanged. whereas the track:vector ratio, a measurement of curvilinear movement, was reduced. This work may have important implications for the assessment of bovine fertility and cytogenetic analysis of bovine sperm.  相似文献

The effect of Trypanosomiasis on concentrations of plasma steroids and semen characteristics was studied in 24 dromedary bulls. Based upon the parasitological and serological diagnosis, 18 bulls were found infected with Trypanosoma evansi (Group 2) and six were found to be free from infection and served as controls (Group 1). The infected animals exhibited signs of anaemia indicated by the decrease of packed cell volume (PCV) and haemoglobin concentration (Hb), pale mucus membranes, weight loss, lethargy, weakness and dullness. However, five animals (27.8%) of the infected group revealed elevated rectal temperatures and three animals (16.7%) revealed testicular degeneration upon palpation of their scrotal contents. Concentrations of plasma oestradiol-17beta (86.5 +/- 8.6 pg/ml versus 232.5 +/- 74.4 pg/ml) and testosterone (4.8 +/- 0.7 ng/ml versus 2.7 +/- 1.5 ng/ml) were significantly different (P < 0.05) between the control and infected bulls. Evaluation of the semen collected by electroejaculation and evaluated by a computerized cell motion analyzer revealed normal semen characteristics in the control animals compared to deteriorated ones in the infected bulls. There were highly significant (P < 0.01) decreases in sperm count (12.2 +/- 1.3/ml versus 6.5 +/- 4.9 x 10(6)/ml), motility percentage (68.2 +/- 6.7% versus 27.4 +/-15.6%), percentage of live spermatozoa (73.2 +/- 8.3% versus 35.8 +/- 8.2%) and increases in percentage of morphological abnormalities (3.3 +/- 0.6% versus 15.9 +/- 1.0%) in the infected group. An examination of the plasma hormonal profiles and semen characteristics in the infected bulls indicated that altered Sertoli cell function due to formation of immune complexes in four bulls (Group 2A), pituitary dysfunction in six bulls (Group 2B), testicular degeneration in three bulls (Group 2C) and finally trypanotolerancy in five bulls (Group 2D) are possible factors responsible for poor semen characteristics and infertility induced by T. evansi infection in dromedary bulls.  相似文献

Cryopreserved spermatozoa from 8 bulls were used to examine the interrelationships among flow cytometric spermatozoal quality assessments and classical semen quality parameters and nonreturn rate estimates of fertility. The integrity of the sperm cell membrane and the functional capacity of the mitochondria were quantified by flow cytometry after concurrent staining with carboxydimethylfluorescein diacetate (CDMFDA), propidium iodide (PI), and rhodamine 123 (R123). For each sample a total of 10,000 stained spermatozoa were simultaneously quantified for the intensity of their green and red fluorescence. Three straws from each bull were each examined initially and following incubation at 37 degrees C for 3 hours to assess the rate of senescence. The proportion of spermatozoa retaining membrane integrity and having functional mitochondria, as determined by CDMFDA and R123 staining, were compared with classical semen quality assessments (sperm motility, acrosomal status, cellular and head morphology, presence of vacuoles/craters and cytoplasmic droplets) and with fertility (nonreturn to estrus rates). For individual ejaculates nonreturn rates, the range was from 61.8 to 78.8%, whereas the cumulative rates of several ejaculates for each bull ranged from 71.3 to 83.5%. The proportion of spermatozoa with functional membranes and mitochondria were positively correlated with the percentage of spermatozoa with normal morphology (r=0.82; P=0.01) and motility after 4 hours of incubation (r=0.78; P=0.02), but not with the estimates of fertility. The actual number of spermatozoa per straw staining with CDMFDA and R123 after 4 hours of incubation at 37 degrees C was correlated with the percentage of spermatozoa with normal morphology (r=0.73; P=0.04). Multiple regression equations indicated that combinations of semen quality measurements could be useful in estimating fertilizing potential.  相似文献
