1. Consistency of differences in standard metabolic rate (SMR) between individual juvenile salmonids and the apparently limited ability of individuals to regulate their SMR has led many researchers to conclude that differences in individual SMR are fixed (i.e. genetic). 2. To test for the effects of food ration on individual performance and metabolism, SMR was estimated by measuring oxygen consumption using flow-through respirometry on individually separated young of the year coho salmon (Oncorhynchus kisutch) placed on varying food rations over a period of 44 days. 3. Results demonstrate that the quantity of food consumed directly affects SMR of juvenile coho salmon, independent of specific dynamic action (SDA, an elevation in metabolic rate from the increased energy demands associated with digestion immediately following a meal) and indicates that higher food consumption is a cause of elevated SMR rather than a consequence of it. Juvenile coho salmon therefore demonstrated an ability to regulate their SMR according to food availability and ultimately food consumption. 4. This study indicates that food consumption may play a pivotal role in understanding individual variation in SMR independent of inherent genetic differences. We suggest that studies involving SMR need to be cautious about the effects of intra-individual differences in food consumption in communal tanks or in different microhabitats in the wild as disproportionate food consumption may contribute more to variation in SMR than intrinsic (genetic) factors. 5. In general, our results suggest that evolutionary changes in SMR are likely a response to selection on food consumption and growth, rather than SMR itself.  相似文献

Measuring standard metabolic rate (SMR) and specific dynamic action (SDA) has yielded insight into patterns of energy expenditure in snakes, but less emphasis has been placed on identifying metabolic variation and associated energy cost of circadian rhythms. To estimate SMR, SDA, and identify metabolic variation associated with circadian cycles in nocturnally active African house snakes (Lamprophis fuliginosus), we measured oxygen consumption rates (VO2) at frequent intervals before and during digestion of meals equaling 10%, 20% and 30% of their body mass. Circadian rhythms in metabolism were perceptible in the VO2 data during fasting and after the initial stages of digestion. We estimated SMR of L. fuliginosus (mean mass=16.7+/-0.3 g) to be 0.68+/-0.02 (+/-SEM) mL O2/h at 25 degrees C. Twenty-four hours after eating, VO2 peaked at 3.2-5.3 times SMR. During digestion of meals equaling 10-30% of their body mass, the volume of oxygen consumed ranged from 109 to 119 mL O2 for SMR, whereas extra oxygen consumed for digestion and assimilation ranged from 68 to 256 mL O2 (equivalent to 14.5-17.0% of ingested energy). The oxygen consumed due to the rise in metabolism during the active phase of the daily cycle ranged from 55 to 66 mL O2 during digestion. Peak VO2, digestive scope, and SDA increased with increasing meal size. Comparisons of our estimates to estimates derived from methods used in previous investigations resulted in wide variance of metabolic variables (up to 39%), likely due to the influence of circadian rhythms and activity on the selection of baseline metabolism. We suggest frequent VO2 measurements over multiple days, coupled with mathematical methods that reduce the influence of undesired sources of VO2 variation (e.g., activity, circadian cycles) are needed to reliably assess SMR and SDA in animals exhibiting strong circadian cycles.  相似文献

Estimating energy costs by respirometry is fundamental to many studies of the ecology, behavior and evolution of reptiles. However, traditional respirometry procedures seldom incorporate objective techniques for removal of outliers from estimates of metabolic parameters. We demonstrate how computer-automated respirometry equipment, which records many respiratory measurements over short intervals, can be coupled with mathematical procedures to produce robust estimates of pre- and post-prandial metabolism in banded water snakes (Nerodia fasciata fasciata). Standard metabolic rate of N. f. fasciata was estimated to be 1.21 ml O2/h (mass = 30.21 +/- 0.74 g) at 25 degrees C. After ingestion of a fish equaling 20% of their body mass, snakes exhibited a fivefold increase in metabolic rate with peak O2 consumption rate (VO2) reaching 6.5 ml O2/h. Total cost of digestion was 5.44 kJ, equivalent to approximately 21% of the energy in the meal. Repeated measurements of metabolism in the same individuals revealed that our methods yielded similar results, even when individuals exhibited different patterns of VO2 variation between respiratory trials. Our results underscore the importance of obtaining many VO2 measurements, coupled with objective removal of outlier values from estimates of metabolic rate, especially when metabolic values are to be interpreted in a comparative context.  相似文献

Conventional gas-exchange instruments are confined to the measurement of O(2) consumption (VO(2)) and CO(2) production (VCO(2)) and are subject to a variety of errors. This handicaps the performance of these devices at inspired O(2) fraction (FI(O(2))) > 0.40 and limits their applicability to indirect calorimetry only. We describe a device based on the automation of the Douglas bag technique that is capable of making continuous gas-exchange measurements of multiple species over a broad range of experimental conditions. This system is validated by using a quantitative methanol-burning lung model modified to provide reproducible (13)CO(2) production. The average error for VO(2) and VCO(2) over the FI(O(2)) range of 0.21-0.8. is 2.4 and 0.8%, respectively. The instrument is capable of determining the differential atom% volume of known references of (13)CO(2) to within 3.4%. This device reduces the sources of error that thwart other instruments at FI(O(2)) > 0. 40 and demonstrates the capacity to explore other expressions of metabolic activity in exhaled gases related to the excretion of (13)CO(2).  相似文献

Carp (Cyprinus carpio) and tilapia (Oreochromis mossambicus) were kept in a flow-through respirometer for a week, while continuous oxygen consumption measurements were carried out, at a constant water O2 concentration.2. Our measurements showed that tilapia decreased its standard metabolic rate (SMR), its average oxygen consumption, and its maximum metabolic rate in acid water (pH 4.0), whereas carp did not.3. Changes in the cellular metabolism assessed by the variability of oxygen consumption (VO(2) ) and carbon dioxide production (VCO(2) ) as well as the association of serum glucose and insulin to energy spectral density (ESD) of VO(2) and VCO(2) were evaluated. Ten nonglucose intolerant and 10 glucose intolerant subjects, aged 21-70 years, were included. Glucose and insulin concentrations and VO(2) and VCO(2) records were collected every 10 min during 3 h. ESD of VO(2) and VCO(2) was estimated and associated with glucose and insulin concentrations. Statistical significance in glucose levels, insulin, and ESD of VO(2) and VCO(2) among nonglucose intolerant subjects and glucose and insulin among glucose intolerance subjects at postload glucose (PLG) state compared with basal state was found. Moreover, glucose was significantly higher in glucose intolerance subjects than nonglucose intolerant subjects for basal and PLG states. These results show an increment in ESD of VO(2) and VCO(2) at PLG state among nonglucose intolerant subjects and suggest that their measurement may be a key indicator of the variability of cellular metabolic activity and contribute to confirm disturbances in glucose metabolism.  相似文献

Compared with morphological and life history traits, quantitative genetic variation of metabolic and related traits in animals has been poorly studied. We used flow-through VCO(2) respirometry and simultaneous activity measurement on nymphs of the sand cricket (Gryllus firmus) from inbred lines to estimate broad-sense heritability of four metabolic variables. In addition, we measured a number of linear dimensions in the adults from the same inbred lines. There were significant multivariate effects of inbred lines for all traits and broad-sense heritability for physiological traits was 4.5%, 5.2%, 10.3% and 8.5% for average, resting, minimum and maximum CO(2) production in nymphs, respectively. Though the MANOVA indicated significant genetic variation among inbred lines in adult morphology, the broad-sense heritabilities were relatively low ranging from 0-18%. Our results indicate that the heritabilities of metabolic measures are large enough to potentially respond to selection.  相似文献

Abstract. This study employed flow-through respirometry to measure the oxygen consumption rates (VO2) of inactive male house crickets, Acheta domesticus L. (Orthoptera: Gryllidae), and to quantify the effects of body mass and adult age on standard metabolic rate (SMR). The Vo2 increased with male body mass at a rate similar to that found in other studies of insect aerobic metabolism. The data reported in this study are combined with published data from other species of Orthoptera to generate a consensus allometric relationship between SMR and body mass for the Order. In general, the Orthoptera expend 2–3 times the energy per unit of body mass when inactive as compared to other arthropods, such as tenebrionid beetles, ants and spiders. Possible explanations for this substantial difference are discussed. By contrast to body mass, mass-specific Vo2 decreased with increasing male age. This age effect has previously been reported for mammals but is not well established for insects, and its implications for the preference of cricket females for older mates is discussed. As energy expended for metabolic maintenance comprises 78% of a male cricket's daily energy budget, changes in SMR may have a substantial effect on the energy available for reproduction.  相似文献

Alterations in VO(2) or VCO(2) are amongst the more polemical physiological adaptations ascribed to insects. Generally, metabolic rate is thought to be lowered in response to arid conditions, and elevated in species from cold environments compared to their more temperate relatives. However, most studies have rarely addressed the influence of both environmental factors in unison. To this end, standard metabolic rate and its temperature dependence were measured (at 4 degrees C intervals from 16 to 32 degrees C) in six Scarabaeus dung beetle species (three flightless, three volant) from a variety of habitats (warm, arid to cool, mesic) in southern Africa using flow-through respirometry. Mass specific VCO(2) varied from 0.0158 ml g(-1) h(-1) at 16 degrees C to 0.1839 ml g (-1) h(-1) at 32 degrees C. Until recently most studies have focussed on method development for metabolic rate assessment in adult and/or juvenile fish with less focus on measurement of oxygen consumption (ṀO2) during early life history stages, including fast-growing larval fish and even less focus on nonteleostean species. In the present study we evaluated measurement techniques for standard metabolic rate (SMR), maximum metabolic rate (MMR) and aerobic scope in an Acipenseriform, the lake sturgeon Acipenser fulvescens, throughout the first year of life. Standardized forced exercise protocols to assess MMR were conducted for 5 or 15 min before or after measurement of SMR. We used different levels of oxygen decline during the measurement period of MMR post forced exercise to understand the influence these may have on the calculation of MMR. Opercular rate and tail beat frequencies were recorded by video as measures of behaviours and compared to metabolic rate recorded over a 24 h period. Results indicate that calculated values for aerobic scope were lower in younger fish. Neither exercise sequence nor exercise duration influenced metabolic rate measurements in the younger fish, but exercise duration did affect measurement of MMR in older fish. Finally, there was no strong correlation between metabolic rate and the measured behaviours in the lake sturgeon at either age. Based on the results, we recommend that a minimum of 6 h of acclimation to the respirometry chamber should be given prior to measuring SMR, a chasing protocol to elicit MMR should ideally be performed at the end of experiment, a short chasing time should be avoided to minimize variation and assessment of MMR should balance measurement limitations of the probes along with when and for how long oxygen consumption is measured.  相似文献

Aerobic metabolism (oxygen consumption, VO2, and carbon dioxide production, VCO2) has been measured in newborn rats at 2 days of age during normoxia, 30 min of hyperoxia (100% O2) and an additional 30 min of recovery in normoxia at ambient temperatures of 35 degrees C (thermoneutrality) or 30 degrees C. In normoxia, at 30 degrees C VO2 was higher than at 35 degrees C. With hyperoxia, VO2 increased in all cases, but more so at 30 degrees C (+20%) than at 35 degrees C (+9%). Upon return to normoxia, metabolism readily returned to the prehyperoxic value. The results support the concept that the normoxic metabolic rate of the newborn can be limited by the availability of oxygen. At temperatures below thermoneutrality the higher metabolic needs aggravate the limitation in oxygen availability, and the positive effects of hyperoxia on VO2 are therefore more apparent.  相似文献

We compared the precision, bias and accuracy of two techniques that were recently proposed to estimate the contributions of cuticular and respiratory water loss to total water loss in insects. We performed measurements of VCO2 and VH2O in normoxia, hyperoxia and anoxia using flow through respirometry on single individuals of the highly variable cockroach Perisphaeria sp. to compare estimates of cuticular and respiratory water loss (CWL and RWL) obtained by the VH2O-VCO2 y-intercept method with those obtained by the hyperoxic switch method. Precision was determined by assessing the repeatability of values obtained whereas bias was assessed by comparing the methods' results to each other and to values for other species found in the literature. We found that CWL was highly repeatable by both methods (R0.88) and resulted in similar values to measures of CWL determined during the closed-phase of discontinuous gas exchange (DGE). Repeatability of RWL was much lower (R=0.40) and significant only in the case of the hyperoxic method. RWL derived from the hyperoxic method is higher (by 0.044 micromol min(-1)) than that obtained from the method traditionally used for measuring water loss during the closed-phase of DGE, suggesting that in the past RWL may have been underestimated. The very low cuticular permeability of this species (3.88 microg cm(-2) h(-1) Torr(-1)) is reasonable given the seasonally hot and dry habitat where it lives. We also tested the hygric hypothesis proposed to account for the evolution of discontinuous gas exchange cycles and found no effect of respiratory pattern on RWL, although the ratio of mean VH2O to VCO2 was higher for continuous patterns compared with discontinuous ones.  相似文献

Despite the importance of metabolic rate in determining flight time of tsetse and in mediating the influence of abiotic variables on life history parameters (and hence abundance and distribution), metabolic rate measurements and their repeatability have not been widely assessed in these flies. We investigate age-related changes in standard metabolic rate (SMR) and its repeatability, using flow-through respirometry, for a variety of feeding, gender and pregnancy classes during early adult development in laboratory-reared individuals of the tsetse fly, Glossina pallidipes. Standard metabolic rate (144-635 microW) was generally within 22% of previous estimates, though lower than the values found using closed system respirometry. There was no significant difference between the genders, but metabolic rate increased consistently with age, probably owing to flight muscle development. Repeatability of metabolic rate was generally high (r=0.6-.09), but not in younger teneral adults and pregnant females (r approximately equal to 0.05-0.4). In these individuals, low repeatability values are a consequence of muscle or in utero larval development. Tsetse and other flies generally have a much higher metabolic rate, for a given size, than do other insect species investigated to date.  相似文献

On three separate occasions, five zebra finches ( Taenopygia guttata) were injected intraperitoneally with 0.2 ml 0.29 M NaH(13)CO(3)solution and placed immediately into respirometry chambers to explore the link between (13)C elimination and both O(2) consumption (VO(2)) and CO(2) production (VCO(2)). Isotope elimination was best modelled by a mono-exponential decay. The elimination rate (k(c)) of the (13)C isotope in breath was compared to VO(2) (ml O(2)/min) and VCO(2) (ml CO(2)/min) over sequential 5-min time intervals following administration of the isotope. Elimination rates measured 15-20 min after injection gave the closest relationships to VO(2) ( r(2) =0.82) and VCO(2) ( r(2)=0.63). Adding the bicarbonate pool size (N(c)) into the prediction did not improve the fit. A second group of birds ( n=11) were flown for 2 min (three times in ten birds and twice in one) between 15 min and 20 min following an injection of 0.2 ml of the same NaH(13)CO(3) solution. Breath samples, collected before and after flight, were used to calculate k(c) over the flight period, which was converted to VO(2) and VCO(2) using the equation generated in the validation experiment for the corresponding time period. The energy expenditure (watts) during flight was calculated from these values using the average RQ measured during flight of 0.79. The average flight cost measured using the bicarbonate technique was 2.24+/-0.11 W (mean+/-SE). This average flight cost did not differ significantly from predictions generated by an allometric equation formulated by Masman and Klaassen (1987 Auk 104:603-616). It was however substantially higher than the predictions based on the aerodynamic model of Pennycuick (1989 Oxford University Press), which assumes an efficiency of 0.23 for flight. The flight efficiency in these birds was 0.11 using this model. Flight cost was not related to within-individual variation [general linear model (GLM) F(1,31)=1.16, P=0.29] or across-individual variations in body mass (GLM F(1,31)=0.26, P=0.61), wingspan (regression F(1,10)=0.01, P=0.94) or wing loading (regression F(1, 31)=0.001, P=0.99) in this sample of birds.  相似文献

Water loss and VCO(2) relative to temperature and oxygen tension was investigated in a log-dwelling onychophoran (Euperipatoides rowelli) and a sympatric, un-described millipede species using flow-through respirometry. Onychophorans possess a tracheal system featuring permanently open spiracles. Total body water loss was consistently very high in E. rowelli and there was a positive correlation with increasing temperature. CO(2) output was continuous, increasing with higher temperatures and decreasing under lower oxygen concentrations. The millipede which has occludible spiracles also showed continuous gas exchange; however water loss was up to an order of magnitude lower than E. rowelli. An ability to survive under hypoxia is apparent for both species and corresponds with reports of hypoxic conditions within rotting logs. The rotting log habitat common to both taxa is characterized by high relative humidity and typically cool temperatures that approach 0 degrees C at night in winter. Consequently, dispersal through the higher temperatures and lower humidity of the exposed and dry understorey between suitable habitat may be hazardous for E. rowelli due to high desiccation susceptibility.  相似文献

The purpose of this study was to determine differences in VO2max and metabolic variables between treadmill running and treadmill skating. This study also examined VO2max responses during a continuous skating treadmill protocol and a discontinuous skating treadmill protocol. Sixteen male high school hockey players, who had a mean age of 16 +/- 1 years and were of an above-average fitness level, participated in this study. All subjects completed 4 exercise trials: a 1-hour skating treadmill familiarization trial, a treadmill running trial, and 2 randomized skating treadmill trials. Minute ventilation (VE), oxygen consumption VO2), carbon dioxide production VCO2), respiratory exchange ratio (RER), and heart rate were averaged every 15 seconds up to VO2max for each exercise test. The results showed that there was a significant difference (P < 0.05) for VO2max (mL.kg.min) and maximal VCO2 (L.min) between the running treadmill protocol and discontinuous skating treadmill protocol. There was also a significant difference for maximal RER between the discontinuous and continuous skating treadmill protocol and between the discontinuous skating treadmill protocol and running treadmill protocol. In conclusion, the running treadmill elicited a greater VO2max (mL.kg.min) than the skating treadmill did, but when it comes to specificity of ice skating, the skating treadmill may be ideal. Also, there was no significant difference between the discontinuous and continuous skating treadmill protocols. Therefore, a continuous protocol is possible on the skating treadmill without compromising correct skating position and physiologic responses. However, the continuous skating treadmill protocol should undergo validation before other scientists, coaches, and strength and conditioning professionals can apply it correctly.  相似文献

The effects of norepinephrine (NE) injection (300 microgram . kg-1 of body weight) on oxygen consumption (VO2), carbon dioxide production (VCO2) and respiratory exchange ratio (R) were investigated in female rats after 1 h of running on a treadmill (21.5 m . min-1) at 10% inclination. Six groups of animals were injected respectively at various times after the exercise (1, 3, 6, 9, 21, and 47 h), and were compared to six non-exercised groups injected at corresponding times. VO2 and VCO2 were monitored continuously during the 20 min preceding injection and for the 60 min following it. The increases in VO2 and VCO2, and the decrease in R were of similar magnitude in both exercised and non-exercised rats (about 30% and 20% for VO2 and VCO2, respectively, and -12% for R). Peak VO2 and R values attained after NE injection varied however with time of injection, specially in exercised animals 1 and 9 h after the run. Exercise significantly delayed time of response to NE for VO2 and VCO2 particularly 1 and 9 h after the running bout. It is concluded that time of day, exercise, and time elapsed after exercise are important factors to consider when studying metabolic responses to catecholamines. Furthermore, it is suggested that such experimental controls might be meaningful in human studies as well.  相似文献

Humoral factors play an important role in the control of exercise hyperpnea. The role of neuromechanical ventilatory factors, however, is still being investigated. We tested the hypothesis that the afferents of the thoracopulmonary system, and consequently of the neuromechanical ventilatory loop, have an influence on the kinetics of oxygen consumption (VO2), carbon dioxide output (VCO2), and ventilation (VE) during moderate intensity exercise. We did this by comparing the ventilatory time constants (tau) of exercise with and without an inspiratory load. Fourteen healthy, trained men (age 22.6 +/- 3.2 yr) performed a continuous incremental cycle exercise test to determine maximal oxygen uptake (VO2max = 55.2 +/- 5.8 ml x min(-1) x kg(-1)). On another day, after unloaded warm-up they performed randomized constant-load tests at 40% of their VO2max for 8 min, one with and the other without an inspiratory threshold load of 15 cmH2O. Ventilatory variables were obtained breath by breath. Phase 2 ventilatory kinetics (VO2, VCO2, and VE) could be described in all cases by a monoexponential function. The bootstrap method revealed small coefficients of variation for the model parameters, indicating an accurate determination for all parameters. Paired Student's t-tests showed that the addition of the inspiratory resistance significantly increased the tau during phase 2 of VO2 (43.1 +/- 8.6 vs. 60.9 +/- 14.1 s; P < 0.001), VCO2 (60.3 +/- 17.6 vs. 84.5 +/- 18.1 s; P < 0.001) and VE (59.4 +/- 16.1 vs. 85.9 +/- 17.1 s; P < 0.001). The average rise in tau was 41.3% for VO2, 40.1% for VCO2, and 44.6% for VE. The tau changes indicated that neuromechanical ventilatory factors play a role in the ventilatory response to moderate exercise.  相似文献

A monitor was developed to track weightlessness deconditioning aboard the National Aeronautics and Space Administration (NASA) Space Station by measuring the O2 uptake (VO2) and CO2 production (VCO2) and calculating maximum VO2 and anaerobic threshold during an exercise stress test. The system uses two flowmeters in series to achieve a completely automatic flow calibration, and it uses breath-by-breath compensation for sample line transport delay. The accuracy of the system was measured over the range of VO2 and VCO2 from 100 to 800 ml/min by means of simulation. Accuracy was 0.54% for VO2 and 2.9% for VCO2. The system was further evaluated using two laboratory methods, the first method being comparison with a breath-by-breath system. As volunteers performed a maximum effort on a cycle ergometer, the mean difference in readings between the two systems was 17 ml/min for VO2 and 8.0 ml/min for VCO2. The correlation coefficient squared was greater than 0.96 for both. The second laboratory test was to use the system for 2 mo in a Human Performance Laboratory. Readings of maximum VO2 (VO2max) and anaerobic threshold were repeatable and consistent with the individual's activity level. The accuracy and convenience of operation will make this a valuable instrument aboard the Space Station.  相似文献
