The microbially mediated reductive dehalogenation of aromatic compounds is potentially important in removal of chlorinated aromatic compounds from the environment. Thermodynamic data are presented which show that the reductive dechlorination of 3-chlorobenzoate to benzoate is exergonic, which led to the hypothesis that reductive elimination of chlorine from 3-chlorobenzoate yields biologically useful energy. In the present paper this hypothesis is tested. Experimental data were obtained with a defined 3-chlorobenzoate degrading methanogenic consortium. These data showed that (i) the molar growth yield of a defined 3-chlorobenzoate degrading consortium increased from 4.9 g protein per mol benzoate metabolized to 6.8 g protein per mol 3-chlorobenzoate when 3-chlorobenzoate replaced benzoate as energy source, and that (ii) the ATP level in starved consortium cells was twice as high when the cells were fed 3-chlorobenzoate than when fed benzoate. These observations show that the electrochemical potential between the redox partners of the H+/H2 (electron-donating) and 3-chlorobenzoate/benzoate (electron-accepting) couples is a potential source of energy and are consistent with the hypothesis that reductive dechlorination of aromatic compounds is coupled to a novel type of microbial chemotrophy.  相似文献

The potential of using nitrate as a terminal electron acceptor to stimulate anaerobic degradation of mixtures of monochlorophenols (MCPs) or dichlorophenols (DCPs) was evaluated. Contaminated and non-contaminated soils were added to water saturated anaerobic microcosms supplemented with 1 mM or 5 mM nitrate. Denitrification and dechlorination activity were present in three diverse soil types and were maintained upon refeeding both nitrate and the appropriate chlorophenol. However, dechlorination activity could only be serially transferred in enrichments with an added electron donor such as acetate. Dehalogenation activity in enrichments from four of the primary microcosms showed at least five different dechlorination reactions, each mediated by different microbial communities. Three of these are distinct ortho-dechlorinating paths; two are meta-dechlorinating and one is the para-dechlorination of 3,4-DCP. Simultaneous dechlorination and denitrification was observed and both activities could be maintained in microcosms but only in the presence of low nitrate concentrations. Dechlorination and denitrification were mediated by two separate microbial communities; one that dechlorinates without use of nitrate and one that denitrifies while oxidizing the dechlorinated aromatic ring. There was no evidence that dechlorination is mediated by the denitrifying community, however the maintenance of a denitrification potential using low (< 1 mM) nitrate concentrations may be useful for completing the food chain by stimulating the mineralization of phenol and benzoate.  相似文献

Chloroform (CF) is an important priority pollutant contaminating groundwater. Reductive dechlorination by anaerobic microorganisms is a promising strategy towards the remediation of CF. The objective of this study was to evaluate the use of redox active vitamins as electron shuttles to enhance the anaerobic biodegradation of CF in an unadapted methanogenic consortium not previously exposed to chlorinated compounds. Only negligible degradation of CF was observed in control cultures lacking redox active vitamins. The addition of riboflavin (RF), cyanocobalamin (CNB12), and hydroxycobalamin (HOB12) enabled biodegradation of CF. The reactions were predominantly catalyzed biologically as evidenced by the lack of any CF conversion in heat-killed controls amended with the cobalamins or minor conversion with RF. In live cultures, significant increases in the rate of CF conversion was observed at substoichiometric molar ratios as low as 0.1 to 0.01 vitamin:CF for RF and CNB12, respectively. At the highest molar vitamin:CF ratios tested of 0.2, the first-order rate constant of CF degradation was 5.3- and 91-fold higher in RF and CNB12 amended cultures, respectively, compared to the unamended control culture. The distribution of biotransformation products was highly impacted by the type of redox active vitamin utilized. Cultures supplemented with RF provided high yields of dichloromethane (DCM). On the other hand, cobalamins promoted the near complete mineralization of organochlorine in CF to inorganic chloride and lowered the yield of DCM. In cultures where no or little CF bioconversion occurred, prolonged exposure to CF resulted in cell lysis, as evidenced by the release of intracellular chloride. The results taken as a whole suggest that the anaerobic bioremediation of CF-contaminated sites can greatly be improved with strategies aimed at increasing the concentration of redox active vitamins.  相似文献

The ensuing hydrogen gradients dissipate part of the potential energy that would otherwise be available to the hydrogen-consuming organisms. A metagenomic approach was taken to investigate the genetic basis for the ability of an anaerobic consortium to grow on either 4-chlorobenzoate or 4-bromobenzoate under denitrifying conditions. Degenerate PCR primers were designed for the family of 4-chlorobenzoyl-CoA dehalogenase genes. The primers were utilized to screen a metagenome library and two overlapping clones were identified which yield a PCR product. The complete sequence of one metagenome clone was determined and genes encoding 4-chlorobenzoyl-CoA ligase (FcbA) and 4-chlorobenzoyl-CoA dehalogenase (FcbB) were identified. Analysis of the ORFs present in the nucleotide sequence suggests that the metagenome clone originated from an uncultured denitrifying microorganism belonging to the Betaproteobacteria. Interestingly, unlike similar gene clusters reported in aerobes, a gene encoding 4-hydroxybenzoyl-CoA thioesterase was not present in the gene cluster. This suggests that 4-hydroxybenzoyl-CoA is further degraded via the anaerobic reduction pathway in the corresponding microorganism instead of through thioester hydrolysis to yield 4-hydroxybenzoate.  相似文献

An anaerobic enrichment culture with glucose as the sole source of carbon and energy plus trichloroethene (TCE) as a potential electron acceptor was inoculated with material from a full size anaerobic charcoal reactor that biologically eliminated dichloromethane from contaminated groundwater (Stromeyer et al. 1991). In subcultures of this enrichment complete sequential transformation of 10 µM TCE viacis-dichloroethene and chloroethene to ethene was reproducibly observed. Maintenance of this activity on subcultivation required the presence of TCE in the medium. The enrichment culture was used to inoculate an anaerobic fixed-bed reactor containing sintered glass Raschig elements as support material. The reactor had a total volume of 1780 ml and was operated at 20 °C in an up-flow mode with a flow rate of 50 ml/h. It was fed continuously with 2 mM glucose and 55 µM TCE. Glucose was converted to acetate as the major product and to a minor amount of methane; TCE was quantitatively dehalogenated to ethene. When, in addition to TCE, tetrachloroethene or 1,2-dichloroethane were added to the system, these compounds were also dehalogenated to ethene. In contrast, 1,1,1-trichloroethane was not dehalogenated, but at 40 µM severely inhibited acetogenesis and methanogenesis. When the concentration of TCE in the feed was raised to 220 µM, chloroethene transiently accumulated, but after an adaptation period ethene was again the only volatile product detected in the effluent. The volumetric degradation rate at this stage amounted to 6.2 µmol/l/h. Scavenging can have important consequences for food web dynamics, for example, it may support additional consumer species and affect predation on live prey. Still, few food web models include scavenging. We develop a dynamic model that includes two facultative scavenger species, which we refer to as the predator or scavenger species according to their natural scavenging propensity, as well as live prey, and a carrion pool to show ramifications of scavenging for predation in simple food webs. Our modeling suggests that the presence of scavengers can both increase and decrease predator kill rates and overall predation in model food webs and the impact varies (in magnitude and direction) with context. In particular, we explore the impact of the amount of dynamics (exploitative competition) allowed in the predator, scavenger, and prey populations as well as the direction and magnitude of interference competition between predators and scavengers. One fundamental prediction is that scavengers most likely increase predator kill rates, especially if there are exploitative feedback effects on the prey or carrion resources like is normally observed in natural systems. Scavengers only have minimal effects on predator kill rate when predator, scavenger, and prey abundances are kept constant by management. In such controlled systems, interference competition can greatly affect the interactions in contrast to more natural systems, with an increase in interference competition leading to a decrease in predator kill rate. Our study adds to studies that show that the presence of predators affects scavenger behavior, vital rates, and food web structure, by showing that scavengers impact predator kill rates through multiple mechanisms, and therefore indicating that scavenging and predation patterns are tightly intertwined. We provide a road map to the different theoretical outcomes and their support from different empirical studies on vertebrate guilds to provide guidance in wildlife management.  相似文献

食物网是持久性有机污染物(POPs)在水生生态系统中传递的重要途径,了解其传递行为与机制是POPs生态暴露风险评价的科学基础。从4个方面展开了讨论和分析:(1)食物网主要特征(营养级和食物链长度)与POPs环境行为的关系;(2)POPs在底栖及底栖-浮游耦合食物网中的环境行为;(3)微食物网对POPs环境行为的作用;(4)食物网的变化对POPs环境行为的影响。主要结论如下:(1)已有研究对水生生物中POPs生物放大作用存在较大争议。一般营养级越高,POPs生物富集性越强,但由于各种生态和生理性质的影响,也存在例外情况。食物链长度与POPs生物富集性呈正相关。(2)POPs通过底栖食物网将沉积物中的POPs向上传递,底栖-浮游食物网的耦合提高了高营养级消费者的暴露风险,目前就POPs在底栖食物网中的生物放大性是否大于浮游食物网存在争议。(3)微生物具有较大的比表面积,是吸附POPs的重要载体。另,沉积物中的微生物通过分解有机质,将POPs释放到水柱中。微生物降解也是环境中POPs脱离环境的重要途径。(4)在内、外压力下,食物网结构和功能发生变化,使物质和能量的传递方向和效率发生改变,并与环境理化性质的变化互相耦合,影响POPs的环境行为。当前研究的重点多集中在POPs在浮游食物网,尤其是高营养级浮游食物网中的环境行为,对POPs在底栖及底栖-浮游耦合食物网和微食物网中环境行为的研究相对缺乏。有关POPs在食物网中环境行为的研究多集中在食物网的某个部分,时间尺度较短,缺乏对POPs环境行为动态变化的研究,未来需深入开展多尺度和多角度的POPs在食物网中环境行为的动态变化研究。新型POPs的生产和使用量不断增加,但有关其在食物网中环境行为的相关分析还较为匮乏,需加强研究。  相似文献

A three-state, discrete-time Markov chain is used to model the dynamics of energy flow in a tri-trophic food web. The distribution of energy in the three trophic levels is related to the rates of flow between the trophic levels and calculated for the entire range of possible flow values. These distributions are then analysed for stability and used to test the idea that plants are resource-limited and herbivores are predation-limited. Low rates of death and decomposition, when coupled with low rates of herbivory and carnivory, tend to destabilize this food web. Food webs with higher rates of death and decomposition are relatively more stable regardless of rates of herbivory and carnivory. Plants are more prone to resource-limitation and herbivores are, in general, limited by their predators, which supports Hairston et al. (Am. Nat. 94 (1960) 421). The rate of decomposition often mediates the roles of top-down and bottom-up control of energy flow in the food web.  相似文献

Abstract The interrelationships between an obligate hydrogen-producing and two different hydrogen-scavenging populations grown as synthrophic members of a 3-chlorobenzoate degrading methanogenic consortium were studied. The hydrogen producer was a benzoate degrader (strain BZ-2), and the hydrogen consumers were a 3-chlorobenzoate dechlorinating bacterium ( Desulfomonile tiedjei ) and a hydrogenotropic methanogen ( Methanospirillum strain PM-1). When a mixture of 3-chlorobenzoate plus benzoate was added to this consortium, the rate of benzoate degradation was 50% higher, at slightly lower H2 concentrations, than when benzoate alone was added. The enhanced benzoate degradation rate was apparantly triggered by the lower H2 concentration, as the rate of benzoate degradation was shown to be a function of the H2 concentration. By offering a hydrogen sink, in addition to methanogenesis, the dechlorinating hydrogen-scavenging population stimulated the rate of benzoate degradation. The lowering of the H2 concentration was very small, which was in agreement with the observation that the rate of methanogenesis was hardly affected by this lower hydrogen concentration. Thus there was no significant competition for H2 between the two hydrogen-scavenging populations in the consortium, as they practically complemented each other's hydrogen-scavenging potential at in situ hydrogen concentrations during the degradation of 3-chlorobenzoate. The H2 concentrations at which hydrogen driven methanogenesis by Methanospirillum occurred in the consortium were well below the threshold concentration extrapolated for this methanogen after growth at high H2 concentrations.  相似文献

The bacterial community in the activated sludge of a local wastewater treatment plant was studied in an effort to understand and exploit the metabolic versatility of microorganisms for the efficient biological treatment of food waste. Microorganisms capable of and efficient in degrading domestic food waste were screened based on their ability to produce areas of clearing on selective media containing protein, fat, cellulose and starch. Nine microbial species belonging to the genera Flavobacterium, Pseudomonas, Micrococcus, Aeromonas, Xanthomonas, Vibrio and Sphingomonas were found to degrade all components of food waste. These bacteria were added to domestic wastewater and shown to cause a 60% reduction in the biochemical oxygen demand (BOD) level of wastewater compared to a control in which no microorganisms were added. The ability of the microbial consortium to degrade domestic wastewater as evidenced by the decrease in BOD levels suggests its potential for use in the biological treatment of food waste.  相似文献

Biotransformation of 2-chlorophenol by a methanogenic sediment community resulted in the transient accumulation of phenol and benzoate. 3-Chlorobenzoate was a more persistent product of 2-chlorophenol metabolism. The anaerobic biotransformation of phenol to benzoate presumably occurred via para-carboxylation and dehydroxylation reactions, which may also explain the observed conversion of 2-chlorophenol to 3-chlorobenzoate.  相似文献

From a methanogenic fixed-bed reactor fed with hydroquinone as sole energy and carbon source, a rodshaped bacterium was isolated in pure culture which could degrade hydroquinone and gentisate (2,5-dihydroxybenzoate). In syntrophic coculture with either Desulfovibrio vulgaris or Methanospirillum hungatei, also benzoate could be degraded. Other substrates such as sugars, fatty acids, alcohols, and cyclohexane derivatives were not degraded. Sulfate, sulfite, or nitrate were not used as external electron acceptor. The isolate was a Gram-negative, non-motile, nonsporeforming strict anaerobe; the guanine-plus-cytosine content of the DNA was 53.2±1.0 mol%. In pure culture, hydroquinone was degraded to acetate and benzoate, probably via an intermediate carboxylation. In syntrophic mixed cultures, all three substrates were converted completely to acetate. Phenol was never detected as a fermentation product.  相似文献

Despite recent progress made in describing microbial transformations that occur under anaerobic conditions, our understanding of the role sulfate‐reducing bacteria may play in the remediation of environmental contaminants is still very limited. The objective of this mini‐review is to summarize what is currently known of the metabolism of chlorinated aromatic compounds in the presence of sulfate. Sulfidogenic processes are discussed with respect to the thermodynamics of haloaromatic oxidation and to their potential use in the in situ bioremediation of hazardous organic wastes. A comprehensive listing is made of anaerobic transformations that involve both halogenated and nonhalogenated monoaromatic substrates by denitrifiers, dissimilatory iron‐reducing bacteria, and methanogenic consortia. In contrast to other anaerobic processes, studies involving sulfate‐mediated metabolism of hazardous organic compounds have been neglected; however, the recent success in defining methanogenic transformations, in particular, has enhanced expectations of defining an analogous role for sulfate‐reducing microbial communities in low redox environments that have become contaminated with hazardous substances.  相似文献

采用活体饵料对黄东海生态系统食物网关键种食物链的中下营养层次"浮游植物-中华哲水蚤(Calanus sinicus)-鯷鱼(Engraulis japonicus)"进行了室内模拟实验研究,以了解氨基酸在该食物链中的传递过程.通过76 d的培养实验,对采集样品做了14种氨基酸含量的定量分析.结果表明,在3个营养层次中中华哲水蚤起着十分关键的承上启下作用,它不仅将小球藻的植物性蛋白转化为动物性蛋白,大幅度地提高了赖氨酸的含量,而且将氨基酸总含量从小球藻的6.10%提高到24.16%.作为该食物网主要经济鱼类重要饵料的鯷鱼则进一步将氨基酸含量提高到27.06%.中华哲水蚤的氨基酸相对组成与小球藻有明显的相关性(r=0.606,p<0.05),其中7种必需氨基酸的相关性相对较弱,而非必需氨基酸的相关性明显较强.鯷鱼的食性由合成饵料转变为活体中华哲水蚤时,其氨基酸量也随饵料的改变而有所变化,从与合成饵料相关转为与中华哲水蚤有更紧密的相关.鯷鱼排泄粪便中的氨基酸含量主要由新陈代谢的生理过程所决定.  相似文献

In anoxic marine sediments fatty acids may be oxidized directly by sulfate reducing bacteria, or may be oxidized by pathways which result in hydrogen production. Some of these latter reactions are quite sensitive to hydrogen concentrations ... in other words if hydrogen concentrations become elevated, fatty acid oxidation will cease. Thus sulfate reducers may actually play two important roles in the metabolism of fatty acids in marine sediments. The sulfate reducers both can utilize fatty acids directly, and also can oxidize hydrogen and thus control hydrogen partial pressures in the sediments. Therefore sulfate reducers may act indirectly to facilitate fatty acid oxidation by hydrogen-producing pathways. We carried out a series of incubations of slurried salt marsh sediment under high and low hydrogen partial pressures and in the presence and absence of molybdate to investigate the relative importance of sulfate reducers and other bacteria mediating hydrogen-sensitive reactions. Our results suggest that both classes of bacteria contribute significantly to fatty acid turnover in marine sediments. Studies of low molecular weight fatty acid turnover in sediment must explicitly recognize that manipulation of sediment (including addition of molydbate to inhibit sulfate reducers) may have a large impact on hydrogen partial pressures in sediment, and may thus significantly alter the pathways and/or rates of fatty acid turnover.  相似文献

Abstract Thermophilic (55°C) protein (peptone) degradation was studied in steady state, laboratory-scale reactors. Peptone was easily hydrolysed to amino acids under methanogenic conditions, and all amino acids were completely degraded to volatile fatty acids, carbon dioxide and ammonium. Under these conditions, amino acids known to be oxidatively deaminated were degraded more slowly than the other amino acids. Inhibition of methanogenesis by 2-bromoethanesulfonic acid led to the accumulation of hydrogen in the gas phase and to the immediate inhibition of both protein hydrolysis and the degradation of amino acids that are preferentially oxidatively deaminated. These effects resulted in lower concentrations of all volatile fatty acids except for butyrate and caproate, which increased in concentration. Interspecies hydrogen transfer appeared to be necessary for the complete degradation of alanine, phenylalanine, methionine, valine, leucine and isoleucine. α-Aminobutyrate also accumulated when methanogenesis was inhibited.  相似文献

Abstract The products of anaerobic and micro-aerobic (0.8% O2) metabolism of the sapropelic ciliate Trimyema compressum strain N were studied. Under anaerobic conditions ethanol was formed in large amounts representing 44% of the total carbon excreted. Acetate, lactate, formate, CO2 and H2 were minor products, while succinate was formed in hardly detectable amounts. Under micro-aerobic conditions O2 was consumed, CO2 and formate were produced as major end products and no H2, ethanol and succinate were formed.  相似文献
