The evolution of virulence in UK oat powdery mildew (Blumeria graminis f.sp. avenae) populations is presented along with comparative information on the deployment of resistant cultivars. Virulence frequencies have followed classical gene‐for‐gene principles, and there are no effective resistance genes currently deployed in cultivars grown in the UK. The incidence of powdery mildew in continental Europe and pathogen variation is reviewed as well as other strategies for the control of this disease. New resistant sources have been identified and are being used in breeding programmes throughout Europe.  相似文献

The powdery mildew resistance from Avena macrostachya was successfully introgressed into hexaploid oat (A. sativa). Genetic analysis of F1, F2, F3 and BC1 populations from two powdery-mildew resistant introgression lines revealed that the resistance is controlled by a dominant gene, tentatively designated Eg-5. Molecular marker analysis was conducted using bulked-segregant analysis in two segregating F3 populations. One codominant simple sequence repeats (SSR) marker AM102 and four AFLP-derived PCR-based markers were successfully developed. The SSR marker AM102 and the STS marker ASE41M56 were linked to the gene Eg-5, with genetic distances of 2 and 0.4 cM, respectively, in both mapping populations. Three STS markers (ASE45M56, ASE41M61, ASE36M55) co-segregated with Eg-5 in one population while two (ASE45M56, ASE36M55) of them linked to Eg-5 with a genetic distance of 1 cM in another population. The gene was further mapped to be in a region corresponding to linkage group 22_44+18 in the Kanota × Ogle (KO) hexaploid oat map by comparative mapping. To our knowledge, this is the first report of mapping powdery-mildew resistance in hexaploid oat. The new resistance source of A. macrostachya, together with the tightly linked markers identified here, could be beneficial in oat breeding programmes.  相似文献

Treatment of seed with the systemic fungicides triadimenol plus fuberidazole as Baytan significantly decreased powdery mildew in field experiments involving eight oat cultivars differing widely in their resistance to the disease. This was so up to growth stage G.S. 61 (Zadoks) even in a year of high mildew incidence, although the effect diminished after flowering. Significant (P≥ 0.001) interactions between fungicide treatments and cultivars occurred in each of three years at nearly all assessment times. In years of high mildew incidence (1983 and 1984), susceptible cultivars developed similar levels of mildew in seed-treated and untreated plots by about flowering time, although cultivars with adult plant resistance (APR) had significantly less mildew when seed-treated than when left untreated. Later, as the APR was fully expressed and the fungicide effectiveness was declining, similar mildew levels were recorded on treated and untreated plots. Untreated APR cultivars generally had less mildew than treated susceptible cultivars and in a year of late and light mildew, APR alone provided good protection. Seed treatments combined with foliar sprays of tridemorph as Calixin almost completely controlled mildew except on very susceptible cultivars late in the season in high mildew years. Over all cultivars, seed treatment gave yield advantages of 5.3% (1983) and 6.6% (1984) but in 1982, a year of low mildew, the response was small. The possible influence of integrated host resistance and fungicides in stabilising the pathogen population at economically unimportant levels and the environmental benefit of using host resistance to minimise fungicide usage is discussed.  相似文献

Blumeria graminis (Bgt) is a pathogenic fungus that affects severally wheat plants provoking high losses in wheat production. Biochemical parameters like enzyme activity of catalase, superoxide dismutase, or peroxidases can be used to detect changes of metabolism in response to pathogen infection. We evaluated different biochemical and biometrics parameters to assess the effect of silicon, a widely recognized beneficial nutrient, in wheat infected naturally with Bgt. Integral study and interpretation of results obtained by multivariate analysis is the challenge of present work. Wheat plants growing in hydroponic solution were fertilized with increasing concentration of silicon. Responses of wheat plants to silicon treatments were assessed through the analysis of lipid peroxidation and antioxidant enzymes activity (catalase, ascorbate peroxidase and superoxide dismutase). Furthermore, biometric measures such as Pathogen Index, dry weight of shoot and roots, tiller height, spike length, spike mass, grain number and grain mass production were assessed. Pathogen Index decreased while mass and number of grain, dry weight of shoots were significantly increased, proportionally to silicon concentration increasing in culture media; biochemical parameters were also influenced by the concentration of silicon. Multivariate analysis indicated correspondence between increase of silicon treatments and decrease of antioxidant activities of APX, CAT and SOD. Relationships between the applied doses of silicon and decreasing Bgt expansion in foliar surfaces were also observed. Chemometrics proved to be an optimal tool for integrating data about metabolic status and demonstrate, in present case, that increasing concentration of silicon and the reduction of antioxidant enzyme activity are closely related to decreasing of powdery mildew disease.  相似文献

Chitosan (CHT), a deacetylated chitin derivative, and benzo-(1,2,3)-thiadiazole-7-carbothioic acid S-methyl ester (BTH), a non toxic synthetic functional analogue of salicylic acid, were applied as foliar spray to barley plants (Hordeum vulgare L.), to compare their effectiveness in inducing resistance against Blumeria graminis f. sp. hordei and to investigate the underlying defence response. After an induction phase of 3 days (IP, time elapsed between treatment and fungal inoculation) both compounds reduced significantly the infection on the primary leaf, namely of 55.5% for CHT and of 68.9% for BTH, showing the induction of a good level of local resistance (LAR). A 5-day IP further reduced the infected areas in BTH treated plants (−77.2%) but not in CHT treated ones (−47.1%). Furthermore, both CHT and BTH also induced SAR, being the infection in the second non treated leaves reduced of 57% and 76.2%, respectively, as evaluated at 10-day IP. Both BTH and CHT induced oxidative burst and phenolic compound deposition in treated leaves, creating an hostile environment that slowed down the fungal spreading by impairing haustorium development. However, the greater efficacy of BTH was possibly due to: i) a greater reinforcement of papilla; ii) a higher level and the more homogeneous diffusion of H2O2 in the treated leaf tissues and iii) an induced hypersensitive-like response in many penetrated cells.  相似文献

The trifluoroethanol (TFE)-induced structural changes of two proteins widely used in folding experiments, bovine alpha-lactalbumin, and bovine pancreatic ribonuclease A, have been investigated. The experiments were performed using circular dichroism spectroscopy in the far- and near-UV region to monitor changes in the secondary and tertiary structures, respectively, and dynamic light scattering to measure the hydrodynamic dimensions and the intermolecular interactions of the proteins in different conformational states. Both proteins behave rather differently under the influence of TFE: alpha-lactalbumin exhibits a molten globule state at low TFE concentrations before it reaches the so-called TFE state, whereas ribonuclease A is directly transformed into the TFE state at TFE concentrations above 40% (v/v). The properties of the TFE-induced states are compared with those of equilibrium and kinetic intermediate states known from previous work to rationalize the use of TFE in yielding information about the folding of proteins. Additionally, we report on the properties of TFE/water and TFE/buffer mixtures derived from dynamic light scattering investigations under conditions used in our experiments.  相似文献

Comparisons of programmes, which differed in the dates on which high-volume sprays of dinocap (0.019 %) were omitted, indicated that applications in the period from green cluster to early fruitlet protected the bourse buds of apple cv. Cox's Orange Pippin from infection, and so controlled the number of mildewed blossom trusses in the following year. This role of the sprays in preventing primary mildew declined after the early fruitlet stage. The existence of two phases of the disease was confirmed, and although dinocap at late blossom and early fruitlet contributed to the control of preliminary infections of secondary mildew on vegetative shoots, the latter phase did not become severe until after the peak invasion of bourse buds. Fewer than 20% of the fruit buds which produced primary-mildewed blossom in 1968 were succeeded by healthy fruit buds in the next year; this poor recovery was not improved by dinocap applied during the flowering period in 1968.
Fruit-set and crop of James Grieve and Cox's Orange Pippin were reduced in 1968 by dinocap applied at green cluster and pink bud, or at full blossom and petal-fall, but no deleterious effects were produced by equivalent sprays in 1969. The need for mildew control during flowering is discussed in relation to phytotoxic sensitivity.  相似文献

通过田间小区试验,设N0(0 kg·hm-2)、N1(112.5 kg·hm-2)、N2( 225 kg·hm-2)、N3( 337.5 kg·hm-2) 4个施氮水平,研究不同施氮水平下小麦与蚕豆间作对小麦白粉病发生、植株氮含量和氮素累积分配的影响,探讨间作系统氮肥调控下小麦植株氮素含量、氮素累积分配与白粉病发生的关系.结果表明: 无论单作还是间作,施氮(N1、N2和N3)均增加了小麦籽粒产量,以N2水平下产量最高,单、间作分别为4146和4679 kg·hm-2;施氮加重了小麦白粉病的发生与危害,N1、N2和N3水平下病害进展曲线下的面积(AUDPC)分别平均增加39.6%~55.6%(基于发病率DI)和92.5%~217.0%(基于病情指数DSI),病情指数受氮素调控的影响较发病率大;施氮显著提高小麦植株氮含量(8.4%~51.6%)和氮素累积量(19.7%~133.7%),对氮素分配比例无显著影响.与单作相比,间作小麦产量平均增加12.0%;AUDPC(DI)和AUDPC(DSI)分别平均降低11.5%和30.7%,间作对病情指数的控制效果优于发病率.间作显著降低发病盛期小麦氮含量、阶段累积量和叶片中氮素分配比例(降幅6.6%~12.5%、1.4%~6.9%和9.0%~15.5%).在本研究条件下,兼顾控病效果和产量效应,小麦施氮量不应超过225 kg·hm-2.  相似文献

Summary In the powdery mildew disease of barley,Erysiphe graminis f. sp.hordei forms an intimate relationship with compatible hosts, in which haustoria form in epidermal cells with no obvious detrimental effects on the host until late in the infection sequence. In incompatible interactions, by contrast, the deposition of papillae and localized host cell death have been correlated with the cessation of growth byE. g. hordei. With the advent of improved, low temperature methods of sample preparation, we felt that it was useful to reevaluate the structural details of interactions between barley andE. g. hordei by transmission electron microscopy. The haustoria that develop in susceptible barley lines appear highly metabolically active based on the occurrrence of abundant endoplasmic reticulum, Golgi-like cisternae, and vesicles. In comparison, haustoria found in the resistant barley line exhibited varying signs of degradation. A striking clearing of the matrix and loss of cristae were typical early changes in the haustorial mitochondria in incompatible interactions. The absence of distinct endoplasmic reticulum and Golgi-like cisternae, the formation of vacuoles, and the occurrence of a distended sheath were characteristic of intermediate stages of haustorial degeneration. At more advanced stages of degeneration, haustoria were dominated by large vacuoles containing membrane fragments. Here, we discuss findings made using Drosophila on Alzheimer's disease (AD) risk and progression. Recent studies have investigated the mechanisms underlying glia-mediated neuroprotection in AD. First, we discuss a novel mechanism of glial lipid droplet formation that occurs in response to elevated reactive oxygen species in neurons. The data suggest that disruptions to this process contribute to AD risk. We further discuss novel mechanistic insights into glia-mediated Aβ42-clearance made using the fly. Finally, we highlight work that provides evidence that the aberrant accumulation of reactive oxygen species in AD may not just be a consequence of disease but contribute to disease progression as well. Cumulatively, the discussed studies highlight recent, relevant discoveries in AD made using Drosophila.  相似文献

In the adult nervous system, chemical neurotransmission between neurons is essential for information processing. However, neurotransmission is also important for patterning circuits during development, but its precise roles have yet to be identified, and some remain highly debated. Here, we highlight viewpoints that have come to be widely accepted or still challenged. We discuss how distinct techniques and model systems employed to probe the developmental role of neurotransmission may reconcile disparate ideas. We underscore how the effects of perturbing neurotransmission during development vary with model systems, the stage of development when transmission is altered, the nature of the perturbation, and how connectivity is assessed. Based on findings in circuits with connectivity arranged in layers, we raise the possibility that there exist constraints in neuronal network design that limit the role of neurotransmission. We propose that activity-dependent mechanisms are effective in refining connectivity patterns only when inputs from different cells are close enough, spatially, to influence each other's outcome.  相似文献

Powdery mildew disease caused by Blumeria graminis f. sp. tritici (Bgt) is an economically important disease in wheat worldwide. The identification of germplasms resistant to the disease can not only facilitate the breeding of resistant cultivars, but can also broaden the diversity of resistance genes. The Mexican M53 is a synthetic hexaploid wheat line developed at the International Maize and Wheat Improvement Center (CIMMYT) from the cross between Triticum durum and Aegilops tauschii249. Infection of M53 with 15 different pathogen races revealed that the resistance in M53 was race-dependent and effective against the majority of the tested Bgt races, including the race 15 predominant in the Beijing wheat growing area. Inoculation of the parents of M53 with the race 15 demonstrated that M53 and Ae. tauschii249 were resistant, whereas T. durum was susceptible. The inoculation of three segregating F2 populations developed from the crosses between M53 and three susceptible Chinese wheat cultivars with the race 15 showed that the resistant gene in M53 segregated in a single dominant manner. Amplified fragment length polymorphism (AFLP) and simple sequence repeat (SSR) markers were used to map the gene in a segregating F2 population consisting of 213 lines developed from the cross Wan7107 × M53. Two closely linked AFLP markers, Apm109 and Apm161, were identified to flank the gene with genetic distances of 1.0 cM and 3.0 cM, respectively. The recognized gene was assigned to the long arm of chromosome 5D as determined by three linked SSR markers, Xwmc289b, Xgwm583, and Xgwm292, and by the physical mapping of Apm109 using Chinese Spring nullisomic–tetrasomic and ditelosomic stocks. The resistance gene identified in M53, temporarily designated as Pm-M53, could be used in local wheat-breeding programs to improve powdery mildew resistance.  相似文献
