ABSTRACT. Leishmania differentiation in the gut of phlebotomine sand flies was evaluated based on five light and electron microscopic studies of natural (Leishmania panamensis/Lutzomyia gomezi, Leishmania chagasi/Lutzomyia longipalpis) and unnatural (Leishmania mexicana/Lutzomyia abonnenci, Leishmania panamensis/Phlebotomus papatasi, Leishmania major/Lutzomyia longipalpis) life cycles. In the bloodmeal, transformation of amastigotes into stumpy promastigotes occurred before or during division. Further division in pairs or rosettes resulted in the development of spatulate and/or elongate nectomonad (free-swimming) promastigotes. Elongate, short, and metacyclic nectomonad promastigotes, and nectomonad paramastigotes were present in the midgut lumen. Dividing short promastigotes predominated in the cardia, and appeared to generate metacyclic forms which were observed in three life cycles. Haptomonad (attached) forms of Leishmania panamensis in the hindgut were primarily spatulate promastigotes (natural host) or pear-shaped promastigotes (unnatural host); paramastigotes and dividing forms were rare. At the stomodeal valve, short haptomonad promastigotes predominated in unnatural hosts, while both short and pear-shaped haptomonads were abundant, along with paramastigotes in natural hosts. Haptomonad paramastigotes and pear-shaped promastigotes colonized the esophagus, while paramastigotes predominated in the pharynx. Metacyclics were free-swimming in the lumen of the foregut.  相似文献

The effects of a number of organic substrates on the autotrophic metabolism of Hydrogenomonas eutropha were examined. Dual substrate (mixotrophic) cultivation in the presence of hydrogen plus either fructose or alanine allowed autotrophic growth to begin immediately after the exhaustion of the organic substrate. On the other hand, the presence of acetate, pyruvate, or glutamate caused a lengthy lag to occur before autotrophic growth commenced. With acetate or pyruvate this lag (plateau) in the dicyclic growth curve was due to the repression of ribulose diphosphate carboxylase (RDPC) synthesis during mixotrophic growth. During heterotrophic growth with glutamate, RDPC was partially repressed; however, during mixotrophic growth, RDPC activity was high. Thus the delay of autotrophic growth was not due to a repression of RDPC by glutamate. The data suggest that glutamate interferes with autotrophic metabolism by repressing the incorporation of inorganic nitrogen. The repression of these vital autotrophic functions by acetate, pyruvate, and glutamate occurred both in the presence and absence of hydrogen, i.e., during both heterotrophic and mixotrophic cultivation. The derepression of the affected systems during the plateau phase of the dicyclic growth curves was demonstrated. Carbon dioxide assimilation by whole cells agreed well with the RDPC activity of extracts from cells grown under similar conditions.  相似文献

ABSTRACT. Attention, perhaps overdue, is drawn to the extent and significance of endosymbionts (xenosomes sensu lato) in the cytoplasm and nuclei of many protozoa from diverse taxonomic groups. Even more importantly, recent advances in the study of such intimate associations are reviewed and discussed and their impact on broader problems of cell biology and evolution are stressed. Workers inside and especially outside the fields of protozoology and parasitology have often neglected such data, failing to appreciate their relevance to significant problems in their own fields of investigation. The major topics covered by speakers in the Symposium (to which this paper serves only as an introduction) include the following, in order of their presentation: terminology for the symbiont-host relationship and a brief overview of the field; the evolutionary problem of the origin of contemporary associations, including cell organelles such as mitochondria and plastids; the adaptive value of endosymbionts to their protozoan hosts; mechanisms of establishment, maintenance, and integration of such foreign bodies/invaders in their unicellular eukaryotic host cells; and the extent of algal and bacterial endosymbioses in diverse protozoan groups. In all papers, the principal relatively well studied complexes used as examples are the following: various kinds of algae in the larger foraminifera and in ciliates, radiolarians, and acantharians; the several types of bacteria in the cytoplasm of Amoeba and of Pelomyxa; the endonuclear bacterial symbionts of Paramecium; the cytoplasmic prokaryotes in Paramecium and in Parauronema; and the methanogenic bacteria of certain ciliates.  相似文献

This study describes the occurrence, importance and seasonal patterns of picoplankton in two wetlands (TDNP and La Safor), and compares them to a system of fifteen interconnected lakes (Ruidera). In TDNP we performed a six‐year monthly study in three sites of the wetland. Bacterial abundance increased throughout time and the autotrophic picoplankton (APP) range was wide (up to 33 × 106 cells/ml). The annual averaged APP contribution to total picoplankton and phytoplankton biovolumes was 0.5–22% and 0.03–6% respectively. There were large differences among sites in terms of APP absolute and relative abundance and seasonal patterns. In La Safor, the APP relative contribution to picoplankton and phytoplankton biovolumes was 0–25% and 0–40%, respectively, while in the Ruidera lakes was 0–47% and 0–5%, respectively. In the three systems there was a significant correlation between bacterial abundance and chlorophyll a but the slopes of the linear regressions were different. No significant relationships were found of APP abundance and trophic status in the wetlands, but were noted in the lake system. There was no clear relationship of APP contribution to total phytoplankton biomass to the trophic gradient in wetlands. In the lakes, the higher contribution of APP was found in those with higher trophic levels.  相似文献

The nomenclature used for higher taxonomic categories of protista arose under the influence of the two-kingdom system, and is widely recognized as being evolutionarily misleading. The occurrence and promulgation of multiple, contradictory taxonomic systems have added to the confusion. Recently, we proposed a solution to this problem which involves identifying the largest taxa that are widely recognized to be monophyletic, and naming them using a commonly recognizable prefix and the suffix "protista". Thus, nomenclatural prejudice is eliminated, and, by keeping the system informal and thus circumventing the Linnaean system, our system remains flexible in accommodating new data yet should retain a great degree of stability. Here we discuss possible criticisms of our proposal, such as whether the introduction of more terms could lead to further taxonomic and nomenclatural instability, and conclude that our proposal remains a most reasonable solution to the problem of protistan nomenclature.  相似文献
