In contrast to the situation with egg-larval and larval endoparasitic wasps, little is known about the effects of pupal endoparasitoids and their secretions on the hemocytes of their insect hosts. This study focuses on the pupal endoparasitoid, Pteromalus puparum, and its host, the small white butterfly, Pieris rapae. Parasitism by P. puparum, resulted in a significant increase in the total number of host hemocytes up to day five after parasitization. From day one to day four after parasitization, the percentage of plasmatocytes significantly decreased, and the proportion of granular cells increased. Moreover, from 12 h to day three after parasitization, hemocyte mortality in parasitized pupae was noticeably higher. When P. rapae pupae were parasitized by adult females of P. puparum irradiated by gamma-ray (pseudoparasitization), it was clear that the treated wasps could induce similar hemocyte changes. However, such phenomena did not occur in punctured host pupae (mimic-parasitization). After treatment with P. puparum venom, both the percentages of spreading plasmatocytes and encapsulated Sephadex G-25 beads were lessened significantly in vitro. Electron microscopy analysis and visualization of hemocyte F-actin with phalloidin-FITC showed that hemocytes treated with venom had a rounded configuration and neither spread nor extended pseudopods, while there was no marked alteration of hemocyte cytoskeletons after venom treatment. The results suggested that venom of P. puparum could actively suppress the hemocyte immune response of its host, but not by destroying the host hemocyte cytoskeleton.  相似文献

蝶蛹金小蜂是菜粉蝶蛹期的主要寄生蜂。在杭州,绝大多数以老熟幼虫在寄主蛹内越冬。田间养虫室内饲养结果:1979年发生11—12代;1981年早、中、迟三组分别为12—13代、10—11代以及9—10代,三组合计,非越冬代成虫自5月上旬至12月初始终不断,一年之内,前后(连续或不连续)两代成虫重叠发生的天数至少175天,前后(连续或不连续)三代成虫重叠发生的天数至少49天。 2、3月间小蜂即可羽化,远远早于田阔第一代菜粉蝶蛹的出现,惟不能寄生越冬后的越冬代菜粉蝶蛹,但越冬代蜂的羽化盛期仍处于田间第一代菜粉蝶蛹初见至化蛹始盛期之间,且喂食后寿命很长,故越冬代蜂与第一代菜粉蝶蛹之间的发生期基本上是吻合的。其后,两者代数既多(菜粉蝶一年发生9代),发生又都不整齐,小蜂成虫与菜粉蝶蛹均连绵不断,时间上彼此也是吻合的。这是此蜂自然寄生率高并成为其重要天敌的一个主要原因。 对发生代数多、世代重叠的重要天敌建议分期分批饲养,以深入掌握其生活史及与寄主发生的关系。  相似文献

Transgenic brassica crops producing insecticidal proteins from Bacillus thuringiensis (Bt) are being investigated as candidates for field release to control lepidopteran pests. Information on the potential impact of Bt brassica crops on pests and non-target natural enemies is needed as part of an environmental risk assessment prior to the commercial release. This first tier study provides insight into the tritrophic interactions among Bt broccoli plants, the herbivore Pieris rapae and its parasitoid Pteromalus puparum. We first evaluated the efficacy of three types of Bt broccoli plants, cry1Ac, cry1C and cry1Ac + cry1C, on different instars of P. rapae. Bt broccoli effectively controlled P. rapae larvae, although later instars were more tolerant. The efficacy of different Bt broccoli plants on P. rapae larvae was consistently cry1Ac > cry1Ac + cry1C > cry1C. When the parasitoid P. puparum developed in a P. rapae pupa (host) that had developed from Bt plant-fed older larvae, developmental time, total number and longevity of the P. puparum generated from the Bt plant-fed host were significantly affected compared with those generated from the non-Bt control plant-fed host. Simultaneously, negative effects on P. rapae pupae were found, i.e. pupal length, width and weight were significantly reduced after older P. rapae larvae fed on different Bt plants for 1 or 2 days. Cry1C toxin was detected using ELISA in P. rapae pupae after older larvae fed on cry1C broccoli. However, no Cry1C toxin was detected in newly emerged P. puparum adults developing in Bt-fed hosts. Only a trace amount of toxin was detected from entire P. puparum pupae dissected from the Bt plant-fed host. Moreover, no negative effect was found on the progeny of P. puparum developing from the Bt plant-fed host when subsequently supplied with a healthy host, P. rapae pupae. The reduced quality of the host appears to be the only reason for the observed deleterious effects on P. puparum. Our data suggest that the effects on P. puparum developing in Bt plant-fed P. rapae are mediated by host quality rather than by direct toxicity.  相似文献

为揭示蝶蛹金小蜂Pteromalus puparum抑制寄主菜粉蝶Pieris rapae血细胞免疫反应的分子机制, 克隆了菜粉蝶肌动蛋白、肌动蛋白解聚因子及微管蛋白cDNA, 并研究了蝶蛹金小蜂寄生对其转录水平的影响。结果显示: 菜粉蝶肌动蛋白cDNA ORF为1 131 bp, 编码377 aa, 预测分子量为41.78 kDa, pI为5.29, 与其他昆虫肌动蛋白的相似性非常高, 在90%以上。氨基酸组成特点和系统进化分析表明, 克隆到的菜粉蝶肌动蛋白基因属细胞质型肌动蛋白。菜粉蝶肌动蛋白解聚因子cDNA全长为1 243 bp, ORF为 447 bp, 编码149 aa, 预测分子量为16.97 kDa, pI为7.11,与家蚕Bombyx mori、赤拟谷盗Tribolium castaneum、意大利蜜蜂Apis mellifera和桑粉介壳虫Maconellicoccus hirsutus肌动蛋白解聚因子的相似性分别为97%, 87%, 89%和72%。菜粉蝶微管蛋白cDNA全长为1 757 bp, ORF为1 344 bp, 编码448 aa, 预测分子量为50.38 kDa, pI为4.86, 与家蚕β型微管蛋白1, 2, 3和4的相似性分别为97%, 97%, 87%和93%。系统进化分析表明, 克隆到的菜粉蝶微管蛋白基因属β型微管蛋白。RT-PCR分析表明, 蝶蛹金小蜂寄生能抑制肌动蛋白、肌动蛋白解聚因子及微管蛋白基因在菜粉蝶蛹血细胞中的转录水平。由此推断蝶蛹金小蜂调控寄主血细胞骨架相关蛋白基因的转录是该蜂抑制寄主血细胞免疫反应的分子机制之一。  相似文献

Abstract Studies on the effect of parasitization by the endoparasitoid on host humoral immune reactions are carried out with the pupal endoparisitic wasp, Pteromalus puparum, and its host, Pieris rapae. Phenoloxidase (PO) activity of parasitized hosts hemolymph increased significantly at 12 h, day four and day five after parasitization. Hem‐agglutination activity of parasitized hosts hemolymph was always higher than that of wounded and unparasitized ones. Moreover, antibacterial activity of parasitized hosts hemolymph became more and more stronger, whilst wounded and unparasitized pupae only owned a weak antibacterial activity. It suggested that activities of humoral immune factors of Pieris rapae could be influenced to some degrees by P. puparum.  相似文献

为了测明椰扁甲啮小蜂Tetrastichus brontispae寄生对寄主椰心叶甲Brontispa longissima蛹的血细胞和体液免疫反应的影响,开展了椰扁甲啮小蜂寄生对椰心叶甲蛹血细胞数量和延展性、血淋巴酚氧化酶活性、血淋巴黑化百分率和血细胞凝集素活性等影响的研究。结果表明:与同期未被寄生蛹相比,寄生蛹血细胞总量在寄生后2 d显著降低,但寄生后4 d显著升高; 寄生蛹的浆血细胞延展率在寄生后2 d显著降低,寄生后4 d显著升高;寄生蛹的血淋巴黑化百分率在寄生后0.5~2 d较高,寄生后3~4 d降低直至为0;寄生蛹的血淋巴酚氧化酶活性在寄生后0.5 d,1 d和4 d时显著升高;寄生蛹的血凝素活性在寄生后2 d较高,寄生后1 d和4 d较低。结果说明椰扁甲啮小蜂寄生使寄主椰心叶甲蛹血细胞和体液免疫反应呈现不规律的变化。  相似文献

Pteromalus puparum is a predominant endoparasitoid wasp of Pieris rapae. Its venom is the only active factor injected into host associated with oviposition. In this report, we explored whether the venom alone from this wasp affects the endocrine system of its host or not. We monitored the changes of hemolymph juvenile hormone (JH; only JH III detected), ecdysteroid, and juvenile hormone esterase activity (JHE) over 72 h in parasitized and venom‐microinjected P. rapae pupae. Non‐parasitized and PBS‐microinjected P. rapae served as controls. Results showed that JH titers were significantly higher in parasitized and venom‐microinjected pupae than that in control pupae during 24 to 72 h. After 12 h, JH titers were significantly promoted by parasitization and venom microinjection. JHE activities of non‐parasitized and PBS‐microinjected pupae were significantly higher than that of parasitized and venom‐microinjected pupae, which was with a peak at 12 h (parasitized pupae) or 24 h (venom‐microinjected pupae) during 6 to 48 and 12 to 36 h, respectively. The hemolymph titers of ecdysteroid in non‐parasitized and PBS‐microinjected pupae increased rapidly during 12 to 36 h with a peak at 36 h, and were higher than treatments before 48 h, while presenting a significant difference at 24 to 48 h between the treatments and controls. The results demonstrate that venom alone of this parasitoid wasp can disrupt its host's endocrine system. © 2009 Wiley Periodicals, Inc.  相似文献

在寄生蜂行为生态学研究中, 通常将寄主体型大小作为寄主品质的主要性状来探究寄生蜂的搜寻行为机理, 而忽略寄生蜂体型大小的意义。为揭示聚寄生蜂雌蜂体型大小对其产卵决策的影响, 在严格控制寄主菜粉蝶Pieris rapae蛹体型大小(体重)的情况下, 于室内观察了不同体型大小的蝶蛹金小蜂Pteromalus puparum雌蜂的产卵行为, 并调查了子代蜂数量(窝卵数)、 性比和体型大小的变化。结果表明: 雌蜂在寄主上的驻留时间随其自身体型增大而缩短, 但随寄主体重增大而延长。窝卵数和余卵量受到雌蜂体型大小的显著影响, 均随雌蜂体型增大而显著增加(P<0.05); 但子代蜂性比不受雌蜂体型大小的显著影响 (P>0.05)。子代雌、 雄性体型大小均与雌蜂体型大小无关, 但子代雌蜂体型随寄主体重增大而增大。结果证实, 雌性蝶蛹金小蜂体型大小影响其部分产卵决策。因此, 在建立聚寄生蜂产卵决策模型中应考虑雌蜂体型大小这一重要变量因素。  相似文献

SUMMARY. In Bere Stream three species of adult simuliids, S . ornatum, S. equinum and S. angustipes were parasitized by Sperchon setiger larvae. Trapped adult S. ornatum were more often parasitized than corresponding adults of S. equinum. The lower 95% confidence limits were used as an indication of the relative importance of the statistically significant differences and were high (10.4–33.6%). Laboratory experiments on the occupation of host pupae by pre-parasitic Sperchon setiger larvae also indicated that pupae of S. ornatum were more often occupied than S. equinum. This was confirmed by tests on the difference in percentage occupation of more than 4000 held-collected host pupae. In tests that indicated a significant difference in percentage parasitization between the sexes of the three host species, females were more often parasitized. However, the lower 95% confidence limits for the increase in percentage parasitization of females compared to males were low (0.9–10.2%). The majority of adult simuliids carried only one parasite. The mean number of larvae per trapped parasitized host was significantly greater for S. ornatum (2.69) than for S. equinum (1.77). There was no significant difference between the mean number of parasites carried by the sexes of S. ornatum for each year from 1975 through 1978 but females were significantly more heavily parasitized than males during 1979. The ventral neck region of all three host species is the main attachment site for Sperchon setiger larvae.
The morphology of the pupae of S. ornatum and S. equinum is related to differences in levels of percentage parasitization between these two species. It is suggested that pupal morphology may be the most important factor in the selection of host simuliids by Sperchon setiger larvae.  相似文献

A gregarious endoparasitoid wasp, Cotesia glomerata, parasitizes the cabbage butterfly, Pieris rapae. During wandering larval stage for pupal metamorphosis, the parasitoid larvae egress from the parasitized host to form cocoons thus eventually leading to death of the host. This study focused on the effect of C. glomerata parasitization on cellular immune response of P. rapae. For this purpose, an ideal anticoagulant buffer was formulated to procure the hemocytes in native form with morphological, behavioral, and functional characteristics. The hemocytes selectively encapsulated only DEAE beads under in vitro conditions and a quantitative study revealed about 70% of the beads being encapsulated. On the other hand, calyx fluid from C. glomerata injected to P. rapae markedly inhibited the spreading ability of the hemocytes in a dose-dependent manner and also attenuated the in vitro encapsulation response of the hemocytes against the cationic bead. The calyx fluid contained polydnavirus as well as ovarian proteins. The isolated polydnavirus genome consisted of variously sized-segments with their unequal amounts. The P. rapae injected with the calyx fluid expressed several polydnaviral genes within 2 h. These results suggest that the immunosuppression of the parasitized P. rapae may be induced by the polydnaviral gene products as well as ovarian proteins.  相似文献

Population densities of the endemic red admiral butterfly, Bassaris gonerilla, were monitored over two summers on Banks Peninsula, New Zealand. Egg-laying usually begins in September and ends in late May. Peaks in egg, larval and adult densities suggest that B. gonerilla completes two full generations per season and in favourable years, a third generation is started but not completed. Population density was lower in a low-rainfall season probably because of the lower survival and nutritional quality of the host plant, Urtica ferox. “Non-target” parasitism levels by Pteromalus puparum (introduced to manage populations of the small white butterfly Pieris rapae) were low at 3.5–16.9% of pupae collected from the field. However, parasitism by the self-introduced pupal parasitoid Echthromorpha intricatoria was very high at 67.5–82.3%. Echthromorpha intricatoria can overwinter in B. gonerilla pupae and is thus capable of attacking all generations of B. gonerilla. More long-term data are needed to determine the status of, and regulatory mechanisms affecting B. gonerilla populations.  相似文献

活体微注射测定结果表明,将0.5毒囊当量(venom reservoir equivalent, VRE)的蝶蛹金小蜂毒液注射于其寄主菜粉蝶蛹体内,注射后4~24 h寄主浆血细胞和颗粒血细胞的延展、存活和对Sephadex A50微珠的包囊能力显著下降;以0.002~0.02 VRE/μL的该蜂毒液处理其离体寄主血细胞均能产生同样的生理效应。该毒液抑制90%菜粉蝶蛹浆血细胞和颗粒血细胞延展的浓度各为0.00076 VRE/μL和0.00804 VRE/μL。可见,蝶蛹金小蜂毒液能显著抑制其寄主血细胞的延展和包囊作用,并导致血细胞的死亡。然而,同样条件下丽蝇蛹集金小蜂毒液对其非自然寄主菜粉蝶蛹的血细胞延展、存活和包囊作用则无任何效应。可见,寄生蜂毒液的生理作用具有明显的寄主特异性。  相似文献

分析昆虫被寄生蜂寄生后血淋巴蛋白质组分及组分含量变化,从蛋白质水平分析寄生蜂寄生引起寄主昆虫的生理变化,有利于探索寄生蜂调控寄主的机理。以椰心叶甲Brontispa longissima和椰扁甲啮小蜂Tetrastichus brontispae为研究材料,采用蛋白质双向电泳技术及图像分析技术分析了椰心叶甲蛹被椰扁甲啮小蜂寄生后不同时期血淋巴蛋白质图谱及其变化情况。结果表明:寄生后0.5, 1, 2, 3和4 d,被寄生蛹血淋巴共检测到的蛋白斑点数分别为982, 926, 712, 636和680个,其中特异性蛋白斑点的数量分别为650, 400, 336, 229和150个;同期未被寄生蛹血淋巴共检测到的到的蛋白斑点数分别为645, 817, 640, 873和940个,其中特异性蛋白斑点的数量分别为313, 291, 264, 466和410个;被寄生蛹和同期未被寄生蛹能匹配的蛋白斑点分别为332, 526, 376, 407和530对。被寄生蛹血淋巴蛋白斑点数量的异常变化表明寄主的生理功能受到了严重的影响。  相似文献

为了探讨蛹期寄生蜂对寄主蛋白代谢的寄生生理效应,利用Bradford蛋白含量测定法、Western免疫印迹法及酶联免疫吸附检测法研究了棕尾别麻蝇Boettcherisca peregrina蛹被丽蝇蛹集金小蜂Nasonia vitripennis寄生后其脂肪体和血淋巴中可溶性蛋白及芳基蛋白组成与含量的变化。结果表明:寄生蛹脂肪体和血淋巴中可溶性蛋白的组成与未寄生相比基本无明显差异; 不论寄生与否寄主蛹脂肪体和血淋巴中芳基蛋白亚基分子量均为80 kDa,该亚基在脂肪体中未出现降解现象,而在血淋巴中仅于寄生后12 h的寄主蛹中呈现2条分子量相近的Western免疫印迹带,说明其降解可能先于未寄生对照。就含量而言,寄生蛹脂肪体中可溶性蛋白含量除寄生后24 h外均显著低于未寄生对照,芳基蛋白含量除寄生后48 h外也均显著低于未寄生对照,其中寄生后12 h的含量仅为未寄生的32.0%。寄生蛹血淋巴中可溶性蛋白含量多低于未寄生蛹,且寄生后2,12,24 h的差异达显著水平;芳基蛋白的含量均有低于未寄生的趋势,其中寄生后12 h的含量为未寄生的17.0%。综合认为,丽蝇蛹集金小蜂的寄生可导致寄主脂肪体和血淋巴中可溶性蛋白及芳基蛋白含量下降。  相似文献

The interaction between the entomopathogenic fungusAschersonia aleyrodis and the parasitoidEncarsia formosa on greenhouse whitefly as a host organism was studied, in particular, the survival of the parasitoid after treatment of parasitized hosts with fungal spores. The mean number of parasitized black pupae per parasitoid produced at 25°C was significantly reduced after spore treatment in the first three days following parasitization. Spore treatment four, seven or ten days after parasitization resulted in a mean number of parasitized pupae not significantly different from the number of black pupae in the control. The rather sudden change from low to high survival of parasitized hosts when treated with spores four days after parasitization in spite of high numbers of infected unparasitized larvae, coincided with the hatching of the parasitoid larva from the egg inside the host. Possible reasons for this decrease in susceptibility to infection after parasitoid egg hatch, such as induced changes in host cuticle or haemolymph, are discussed. Parasitoids emerged from treated hosts did not show differences in reproduction compared with parasitoids emerging from untreated hosts. Both natural enemeies of whitefly are compatible to a great extent.  相似文献
