The microbial degradation of chlorinated and nonchlorinated methanes, ethanes, and ethanes by a mixed methane-oxidizing culture grown under chemostat and batch conditions is evaluated and compared with that by two pure methanotrophic strains: CAC1 (isolated from the mixed culture) and Methylosinus trichosporium OB3b. With the exception of 1,1-dichloroethylene, the transformation capacity (Tc) for each chlorinated aliphatic hydrocarbon was generally found to be in inverse proportion to its chlorine content within each aliphatic group (i.e., methanes, ethanes, and ethenes), whereas similar trends were not observed for degradation rate constants. Tc trends were similar for all methane-oxidizing cultures tested. None of the cultures were able to degrade the fully chlorinated aliphatics such as perchloroethylene and carbon tetrachloride. Of the four cultures tested, the chemostat-grown mixed culture exhibited the highest Tc for trichloroethylene, cis-1,2-dichloroethylene, tetrachloroethane, 1,1,1-trichloroethane, and 1,2-dichloroethane, whereas the pure batch-grown OB3b culture exhibited the highest Tc for all other compounds tested. The product toxicity of chlorinated aliphatic hydrocarbons in a mixture containing multiple compounds was cumulative and predictable when using parameters measured from the degradation of individual compounds. The Tc for each chlorinated aliphatic hydrocarbon in a mixture (Tcmix) and the total Tc for the mixture (sigma Tcmix) are functions of the individual Tc, the initial substrate concentration (S0), and the first-order rate constant (k/Ks) of each compound in the mixture, indicating the importance of identifying the properties and compositions of all potentially degradable compounds in a contaminant mixture.  相似文献

A two-stage bioreactor has been developed to link dechlorination of halogenated methane compounds to the anaerobic processes of methanogenesis and denitrification. A digester methanogenic consortium was shown to dechlorinate chloroform (CF) and carbon tetrachloride (CT) to dichloromethane (DCM), and DCM was then mineralized by an acclimated denitrifying biological activated carbon consortium. Combining these two processes, a sequential methanogenic-denitrifying bioreactor (SMDB) system that completely degraded polychlorinated methanes including CT, CF, and DCM was developed. More than 95% of the added CT and CF was dechlorinated in the methanogenic bioreactor with methanol as the primary substrate, and the resultant DCM was biodegraded in the denitrifying bioreactor with nitrate as the electron acceptor. In the denitrifying bioreactor, the residual CF was completely removed, and the DCM removal efficiency was more than 95%. This novel bioreactor system eliminates the need for aeration and so avoids the air contamination associated with aerobic biotreatment of volatile chlorinated pollutants. This SMDB system provides an alternative to conventional biotreatment of wastewaters and other matrices contaminated with polychlorinated methanes and is, to our knowledge, the first report on such a sequential anoxic system. Kinetics were determined for methanogenic activity and chlorinated ethylene dehalogenation by a methanol-enriched, anaerobic sediment consortium. The culture reductively dechlorinated perchloroethylene (PCE) to trichloroethylene (TCE), 1,1-dichloroethylene (1,1-DCE), vinylchloride (VC), and ethylene and ethane. The absence : of methanol or the addition of 2-bromoethanesulfonic. acid in the presence of methanol suppressed both methanogenic activity and dechlorination. In contrast, acetate production continued in the presence of 2-bromoethanesulfonic acid. These results suggest that dechlorination was strongly linked to methane formation and not to acetate production. A kinetic model, developed to describe both methanogenesis and dechlorination, successfully predicted experimentally measured concentrations of biomass, methane, substrate, and chlorinated ethylenes. The average maximum specific dehalogenation rates for PCE, TCE, 1,1-DCE, and VC were 0.9 +/- 0.6, 0.4 +/- 0.1, 12 +/- 0.1, and 2.5 +/- 1.7 mumol contaminant/ g. DW/day, respectively. This pattern for dechlorination rates is distinctly different than that reported for transition metal cofactors, where rates drop by approximately one order of magnitude as each successive chlorine is removed. The experimental results and kinetic analysis suggest that it will be impractical to targeting methanol consuming methanogenic organisms for in situ ground-water restoration. Aerobic cometabolism of chloroform (CF) and 1,1,1-trichloroethane (1,1,1-TCA) was observed by subsurface microorganisms grown on butane. Studies performed in batch incubated microcosms were screened for CF transformation potential using the following cometabolic substrates: ammonia, methane, propane, butane, propene, octane, isoprene, and phenol. CF transformation was observed in microcosms fed ammonia, methane, propane, and butane. The butane microcosms achieved the most effective transformation. The transformation of CF and 1,1,1-TCA was strongly correlated with butane utilization and oxygen consumption. CF transformation ceased in the absence of butane or when oxygen was depleted to low concentrations in the microcosms. No transformation of carbon tetrachloride was observed. With successive additions of CF and butane to the microcosms, complete transformation of CF was achieved at solution concentrations as high as 1 mg/L. High CF concentrations appeared to inhibit butane utilization. Maximum transformation yield (Ty) of 0.01 mg CF trans-formed/mg of butane consumed, were achieved. The results indicate that a monooxygenase enzyme required for butane utilization is likely responsible for the transformation of CF. Chloride measurements demonstrated that CF was completely dechlorinated. Approximately 70% of the chloride in the transformed 1,1,1 -TCA was released into solution, indicating incomplete dechlorination of 1,1,1-TCA. The results indicate that butane is a promising cometabolic substrate for the transformation of chlorinated methanes, chlorinated ethanes, and potentially chlorinated ethenes.  相似文献

The soil nitrifying bacterium Nitrosomonas europaea is capable of degrading trichloroethylene (TCE) and other halogenated hydrocarbons. TCE cometabolism by N. europaea resulted in an irreversible loss of TCE biodegradative capacity, ammonia-oxidizing activity, and ammonia-dependent O(2) uptake by the cells. Inactivation was not observed in the presence of allylthiourea, a specific inhibitor of the enzyme ammonia monooxygenase, or under anaerobic conditions, indicating that the TCE-mediated inactivation required ammonia monooxygenase activity. When N. europaea cells were incubated with [C]TCE under conditions which allowed turnover of ammonia monooxygenase, a number of cellular proteins were covalently labeled with C. Treatment of cells with allylthiourea or acetylene prior to incubation with [C]TCE prevented incorporation of C into proteins. The ammonia-oxidizing activity of cells inactivated in the presence of TCE could be recovered through a process requiring de novo protein synthesis. In addition to TCE, a series of chlorinated methanes, ethanes, and other ethylenes were screened as substrates for ammonia monooxygenase and for their ability to inactivate the ammonia-oxidizing system of N. europaea. The chlorocarbons could be divided into three classes depending on their biodegradability and inactivating potential: (i) compounds which were not biodegradable by N. europaea and which had no toxic effect on the cells; (ii) compounds which were cooxidized by N. europaea and had little or no toxic effect on the cells; and (iii) compounds which were cooxidized and produced a turnover-dependent inactivation of ammonia oxidation by N. europaea.  相似文献

A robust, naturally evolving methanotrophic community in landfill cover soil (LFCS) can be the simplest way to mitigate landfill methane emission. In this study, bacterial community composition in LFCS and methane oxidation potential of enriched methanotrophic consortium, in comparison to that of axenic Methylosinus sporium, was investigated. Growth and methane oxidation of the consortium was studied in liquid phase batch experiments under varying temperature (20–40°C), pH (5–10), headspace CO2, and in presence of porous adsorbent (1.3 cm3 sponge cubes). The 16S rRNA gene analysis revealed presence of both type-I and type-II methanotrophs along with few obligate methylotroph in LFCS. Though the optimal growth condition of the consortium was at 30°C and pH 7, it was more resilient in comparison to M. sporium. With increasing availability of porous adsorbent, methane consumption by the consortium was significantly improved (p < 0.001) reaching a maximum specific methane oxidation rate of 11.4 μmol mg?1 biomass h?1. Thus, inducing naturally thriving methanotrophs in LFCS is a better alternative to axenic methanotrophic culture in methane emission management.  相似文献

The dechlorinating activity of a methanogenic granular sludge from a methanol-fed upflow anaerobic sludge blanket reactor was investigated with chlorinated ethanes. This unadapted methanogenic consortium degraded all chloroethanes tested. The product formation rates decreased with the number of chlorine substituents. The more highly chlorinated ethanes were also converted, although at a lower rate, in the presence of autoclaved (dead) sludge, indicating the involvement of reduced heat-stable cofactors like vitamin B12 and F430. Direct chemical dechlorination of hexa-, penta- and tetrachloroethanes was also observed in medium without sludge, although at a much lower rate. The results show the importance of cometabolic and abiotic (chemical) conversions for the transformation of chlorinated ethanes by the methanogenic consortium. The types of reaction and the products formed were correlated with the Gibbs free-energy change (ΔG 0′). Reductive hydrogenolysis and dichloroelimination were important dechlorinating mechanisms. Generally, these reactions have a higher ΔG 0′ value than dehydrochlorination reactions, which occurred less frequently during the transformation of chloroethanes by the methanogenic granular sludge. An area of cold methane seeps at the bottom of the Laptev Sea was investigated. High rates of methane oxidation were revealed in the sediments and in the water column. Anaerobic methane oxidation carried out by the ANME-2 a/b consortium was coupled to sulfate reduction. Bacteria of the genera Sulfurovum and Arcobacter were the agents of the sulfur cycle. Methane unconsumed in the sediments diffused into the near-bottom water, where it was oxidized by methanotrophic bacteria. Methanotrophic activity was essential for development of symbiotrophic tubeworms of the upper sediment layers, which were responsible for the process of bioturbation.  相似文献

Mixed cultures capable of dechlorinating chlorinated ethanes and ethenes were enriched from contaminated wetland sediment at Aberdeen Proving Ground (APG) Maryland. The “West Branch Consortium” (WBC-2) was capable of degrading 1,1,2,2-tetrachloroethane (TeCA), trichloroethene (TCE), cis and trans 1,2-dichloroethene (DCE), 1,1,2-trichloroethane (TCA), 1,2-dichloroethane, and vinyl chloride to nonchlorinated end products ethene and ethane. WBC-2 dechlorinated TeCA, TCA, and cisDCE rapidly and simultaneously. A Clostridium sp. phylogenetically closely related to an uncultured member of a TCE-degrading consortium was numerically dominant in the WBC-2 clone library after 11 months of enrichment in culture. Clostridiales, including Acetobacteria, comprised 65% of the bacterial clones in WBC-2, with Bacteroides (14%), and epsilon Proteobacteria (14%) also numerically important. Methanogens identified in the consortium were members of the class Methanomicrobia, which includes acetoclastic methanogens. Dehalococcoides did not become dominant in the culture, although it was present at about 1% in the microbial population. The WBC-2 consortium provides opportunities for the in situ bioremediation of sites contaminated with mixtures of chlorinated ethenes and ethanes.  相似文献

Reductive anaerobic dehalogenation is a useful method for remediation of sites contaminated by chlorinated ethylenes, where hydrogen concentration plays the key role. Under anaerobic conditions, dehalogenating bacteria compete best against methanogenic consortia when the hydrogen level is low; and methanogenic consortia outplay dehalogenating bacteria when the hydrogen level is high. Thus, in an anaerobic mixed culture, efficient use of hydrogen for dehalogenation can be achieved by strategies that maintain hydrogen at a certain low concentration. However, due to the role of acetate, expected dehalogenating results cannot be obtained and unexpected methane formation can be encountered in practice.  相似文献

Forest soils are thought to be an important sink for atmospheric methane. To evaluate methane consumption,14C-labeled methane was added to the headspace of intact soil cores collected from a mixed mesophytic forest and from a red spruce forest located in the central Appalachian Mountains. Both soils consumed the added methane at initially high rates that decreased as the methane mixing ratio of the air decreased. The mixed mesophytic forest soil consumed an average of 2 mg CH4 m–2 d–1 versus 1 mg CH, m–2 d–1 for the spruce forest soil. The addition of acetylene to the headspace completely suppressed methane consumption by the soils, suggesting that an aerobic methane-consuming microorganism mediated the process. At both forest sites, methane mixing ratios in soil air spaces were greater than that in the air overlying the soil surface, indicating that these soils had the ability to produce methane. Models of methane emission from forest soils to the atmosphere must represent methane flux as the balance between production and consumption of methane, which are controlled by very different factors  相似文献

Dental plaque samples collected from monkeys (Macaca mulatta) were found to contain a large amount of dissolved methane gas (0.6 nmol CH4/mg wet wt plaque). Enrichment cultures inoculated with dental plaque obtained from Macaca fascicularis produced methane when the medium contained ethanol, methanol, lactate, acetate or a hydrogen + CO2 atmosphere. Methane formation in the enrichments was inhibited by oxidation of the culture medium, autoclaving or the addition of 2-bromoethane sulfonic acid (BES). The methane producing enrichments were observed to contain fluorescent cocci occurring singly and in short chains. It was concluded that methane formation in the monkey dental plaque was the result of the presence of methanogenic bacteria.  相似文献

A methanogenic microbial consortium capable of reductively dechlorinating 1,2,4-trichlorobenzene (1,2,4-TCB) was enriched from a mixture of polluted sediments. 1,2,4-TCB was dechlorinated via 1,4-dichlorobenzene (1,4-DCB) to chlorobenzene (CB). Lactate, which was used as an electron donor during the enrichment, was converted via propionate and acetate to methane. Glucose, ethanol, methanol, propionate, acetate, and hydrogen were also suitable electron donors for dechlorination, whereas formate was not. The addition of 5% (wt/vol) sterile Rhine River sand was necessary to maintain the dechlorinating activity of the consortium. The addition of 2-bromoethanesulfonic acid (BrES) inhibited methanogenesis completely but had no effect on the dechlorination of 1,2,4-TCB. The consortium was also able to dechlorinate other chlorinated benzenes via various simultaneous pathways to 1,3,5-TCB, 1,2-DCB, 1,3-DCB, or CB as an end product. The addition of BrES inhibited several of the simultaneously occurring dechlorination pathways of 1,2,3,4- and 1,2,3,5-tetrachlorobenzene and of pentachlorobenzene, which resulted in the formation of CB as the only final product. Hexachlorobenzene and polychlorinated biphenyls (PCBs) were dechlorinated after a lag phase of ca. 15 days, showing a dechlorination pattern that is different from those observed for lower chlorinated benzenes: only chlorines with two adjacent chlorines were removed. The results show that the consortium possesses at least three distinct dechlorination activities toward chlorinated benzenes and PCBs.  相似文献

Abstract Contamination of ground water with halogenated aliphatic hydrocarbons threatens this source of drinking water. In order to study microbial processes that may enhance the removal of these compounds, Lincoln fine sand was exposed to an atmosphere containing methane (4%) to enrich microorganisms capable of growth on this gaseous hydrocarbon. The methane-enriched soil was then tested to determine whether the enriched microbes could remove seven halocarbons from aqueous solution. Removal of dichloromethane. trans -1,2-dichloroethylene, chloroform, 1,2-dichloroethane, trichloroethylene, and 1,1,1-trichloroethane was significantly different in methane-enriched soil compared to non-enriched soil (ANOVA, 95% significance level). Tetrachloroethylene was not removed. Autoclaving the methane-enriched soil inhibited completely the removal of all the compounds. Once the soil was enriched with methane, its presence in the headspace was not required for removal of several of the compounds but methane was required for their complete removal. These results suggest that methane stimulation of microbial communities may be an alternative treatment technology for bioremediation of contaminated subsurface soils and ground water.  相似文献

An anaerobic microbial consortium able to biodegrade saturation levels of perchloroethylene (PCE) in a column containing a source zone of PCE was examined phylogenetically to determine microbial community structure and spatial variation in relation to the PCE source. The consortium was comprised of at least 34 members with 7 organisms sharing affiliations with known respiratory or cometabolic dechlorinators. Seven other organisms had their closest phylogenetic relative detected in other environments containing chlorinated compounds. Based on denaturing gradient gel electrophoresis, significant Bacteria were Dehalococcoides ethenogenes, Shewanella putrefaciens, and an Acetobacterium species. Spatial variations in community structure of the consortium relative to the PCE source zone were observed. A Pseudomonas species was predominant in a zone 30 cm from the PCE source. A Methanothrix species was predominant at points over 85 cm from the source zone. A Trichlorobacter species was detected where PCE concentrations were highest, up to 85 cm from the PCE source, whereas D. ethenogenes was ubiquitous to over 128 cm from the PCE source.  相似文献

On equal parts of benzene, toluene and p-xylene (BTX), a stable bacterial consortium was enriched for removal of BTX vapours from air. As demonstrated by gas chromatographic monitoring, this consortium removed all three BTX components but was able to grow only on benzene and/or toluene. A Pseudomonas putida strain, PPO1, isolated from this consortium behaved in an identical manner. When immobilized on a porous peat/perlite column, both the consortium and the PPO1 isolated removed all three BTX components from metered air streams. However, due to the accumulation of products from the incompletely metabolized p-xylene, the removal rates were unsatisfactory and declined further with time. P. putida ATCC 33015 bearing the TOL plasmid was capable of growing on toluene, on para- and on meta- xylene isomers, but not on benzene. When the PPO1 and ATCC 33015 strains were immobilized, in equal parts, on peat/perlite columns a much improved and sustainable removal of all three BTX components was observed at the rate of 40–50 g/h. m3 filter bed. Due to the dominance of the ring-hydroxylating pathways over the TOL pathway, the classical enrichment approach did not result in a consortium capable of the sustained removal of all BTX components. However, a rationally formulated consortium consisting of members with complementary metabolic abilities was capable of this task and should be of use both in industrial emission control and in soil venting operations.  相似文献

为获得高效甲烷氧化微生物体系,从农业土壤中采样,以甲烷作为唯一碳源进行好氧选择性传代培养,得到生长性能稳定、生长优于Methylosinus trichosporium OB3b纯培养的具有甲烷单加氧酶(Methane Monooxygenase,MMO)活性甲烷氧化混合菌。利用MMO的共代谢特性,分别以苯酚和环氧丙烷作为目标对象,考察该混合菌对有机污染物的降解及用于生产有用化学物质的催化特性。结果表明,所得混合菌具有高效降解苯酚能力,对初始浓度为600mg/L的苯酚,经过11h培养,苯酚降解率可达99%。另外,以该混合菌为催化剂可以实现丙烯氧化生产环氧丙烷。通过降低磷酸盐浓度可以有效提高环氧丙烷的积累浓度,最大可至5mmol/L。此外,采用纯种分离方法结合PCR扩增、16SrRNA和MMO功能基因分析技术对混合菌群结构进行解析。结果表明,该混合菌群由Ⅱ型甲烷氧化菌及其它至少4种非甲烷氧化菌组成,它们分别属于Methylosinus trichosporium和Acinetobacter junii、Cupriavidusme tallidurans、Comamonas testosteroni和Stenotrophomonas maltophilia。采用PCR方法从混合菌及纯化菌株M.trichosporiums Y9总DNA中都能扩增得到mmoB、mmoX和pmoA基因片段,表明该甲烷氧化菌同时具有sMMO和pMMO两种形式的MMO。通过对从甲烷氧化混合菌中分离纯化得到的甲烷氧化菌进行PCR产物测序,结果发现其与Methylosinus trichosporium的同源性为99.9%。  相似文献

The degradation of trichloroethylene (TCE), chloroform (CF), and 1,2-dichloroethane (1,2-DCA) by four aerobic mixed cultures (methane, propane, toluene, and phenol oxidizers) grown under similar chemostat conditions was measured. Methane and propane oxidizers were capable of degrading both saturated and unsaturated chlorinated organics (TCE, CF, and 1,2-DCA). Toluene and phenol oxidizers degraded TCE but were not able to degrade CF, 1,2-DCA, or other saturated organics. None of the cultures tested were able to degrade perchloroethylene (PCE) or carbon tetrachloride (CC(4)). For the four cultures tested, degradation of each of the chlorinated organics resulted in cell inactivation due to product toxicity. In all cases, the toxic products were rapidly depleted, leaving no toxic residues in solution. Among the four tested cultures, the resting cells of methane oxidizers exhibited the highest transformation capacities (T(c)) for TCE, CF, and 1,2-DCA. The T(c) for each chlorinated organic was observed to be inversely proportional to the chlorine carbon ratio (Cl/C). The addition of low concentrations of growth substrate or some catabolic intermediates enhanced TCE transformation capacities and degradation rates, presumably due to the regeneration of reducing energy (NADH); however, addition of higher concentrations of most amendments reduced TCE transformation capacities and degradation rates. Reducing energy limitations and amendment toxicity may significantly affect T(c) measurements, causing a masking of the toxicity associated with chlorinated organic degradation. Tetrachloroethylene (PCE) and trichloroethylene (TCE), common industrial solvents, are among the most frequent contaminants found in groundwater supplies. Due to the potential toxicity and carcinogenicity of chlorinated ethylenes, knowledge about their transformation potential is important in evaluating their environmental fate. The results of this study confirm that PCE can be transformed by reductive dehalogenation to TCE, dichloroethylene, and vinyl chloride (VC) under anaerobic conditions. In addition, [14C]PCE was at least partially mineralized to CO2. Mineralization of 24% of the PCE occurred in a continuous-flow fixed-film methanogenic column with a liquid detention time of 4 days. TCE was the major intermediate formed, but traces of dichloroethylene isomers and VC were also found. In other column studies under a different set of methanogenic conditions, nearly quantitative conversion of PCE to VC was found. These studies clearly demonstrate that TCE and VC are major intermediates in PCE biotransformation under anaerobic conditions and suggest that potential exists for the complete mineralization of PCE to CO2 in soil and aquifer systems and in biological treatment processes.  相似文献
